发明背景
临床制造治疗性蛋白质是一项成本高的大规模的尝试。对更大量的治疗性重组蛋白的需求已驱使在产生显著提高的产物滴度的细胞培养物处理方面取得了长足的进展。高滴度细胞培养过程典型地是通过在更长的培养持续时间内维持高的活细胞密度而产生的。同样还会观测到生物质固体(活细胞和非活细胞)以及亚微米细胞碎片颗粒相应增多。固体和亚微米细胞碎片颗粒的更高负荷可能对哺乳动物细胞培养物收获方法提出挑战,会使得收获方法在不使产物产量大幅损失的情况下去除碎片方面的有效性较低。
阳离子型聚合物絮凝剂被用于以下范围内的多种应用:饮用水纯化、废水处理、在石油、采矿以及造纸行业中使用、化妆品和医疗用途,并且还已经被用于封装哺乳动物细胞和酶以及用于使微生物细胞培养物絮凝。然而,对于在商业规模的哺乳动物细胞收获方法中使用来说,漫长的絮凝沉降时间可能成问题,会使得收获方法耗时并且与标准收获操作规程相比效率更低。
持续需要对哺乳动物细胞培养物收获方法,特别是商业规模的方法进行改进。任何使得回收时间更快和/或回收率更大的改进都可以使得与制造蛋白质治疗剂相关的成本降低。本发明通过提供一种快速并且高效的细胞培养物收获方法来满足这一需要。
发明概述
本发明提供了一种哺乳动物细胞培养物收获方法,所述方法包括将表达重组蛋白的哺乳动物细胞在细胞培养基中培养预定的时间或直到达到所需的细胞密度和/或细胞压积为止,将阳离子型聚合物和非离子型聚合物添加至细胞培养基中以引发絮凝,在絮凝期间混合细胞培养基,使得絮凝体沉降,以及回收澄清了的上清液。

发明详述
本发明提供了一种简单的收获絮凝技术,所述技术旨在使得高细胞量的细胞培养过程的回收操作达到最大限度。提供了一种哺乳动物细胞培养物收获方法,所述方法在使细胞培养肉汤絮凝时将阳离子型聚合物与非离子型聚合物组合使用。还提供了将阳离子型聚合物与非离子型聚合物和非离子型表面活性剂这两者组合使用。
本发明是基于以下的发现:将非离子型聚合物或非离子型聚合物和非离子型表面活性剂与阳离子型聚合物组合用于使哺乳动物细胞培养肉汤絮凝会使得絮凝体沉降时间从24小时或更长时间缩短至不到1小时,在一些情况下,缩短至15分钟,而与细胞培养过程的密度(高达44%的细胞压积)或乳酸盐水平(10g/L)无关。使用非离子型聚合物还能够将与所需的重组产物共同纯化的高级聚集体和宿主细胞蛋白去除。这种简单的收获方法使得细胞培养过程,特别是高细胞量的细胞培养商业水平过程的回收操作达到最大限度。
絮凝是使悬浮液中的颗粒形成更大尺寸的聚集体或簇集体的过程。在絮凝中,通过添加絮凝剂(flocculation agent/flocculent)使颗粒以絮状物形式从悬浮液中离开。絮凝剂可以是阴离子型聚合物或阳离子型聚合物。存在天然絮凝剂,如藻酸盐或壳聚糖;矿物絮凝剂,如胶态粘土和活性二氧化硅;以及合成絮凝剂,如聚丙烯酰胺和聚二烯丙基二甲基氯化铵。合成絮凝剂可以被制造成具有特定的分子量(基于链长)和分子分布。
阳离子型聚合物与诸如有机物质之类的带负电荷的颗粒相互作用。在细胞培养肉汤中,阳离子型聚合物与诸如活细胞和非活细胞、细胞代谢物以及细胞碎片(如核酸、蛋白以及脂质体)之类的带负电荷的颗粒相互作用。使用阳离子型聚合物使细胞培养肉汤中存在的带负电荷的化合物絮凝是经由以下方式发生离子相互作用而进行的:带负电荷的颗粒的架桥;阳离子型聚合物的补缀结合(patch binding),从而引起絮凝;或对较大的带负电荷的颗粒的电荷中和,从而使得所中和的颗粒从溶液中沉降出。通过阳离子型聚合物对带负电荷的颗粒的架桥作用所形成的絮状物产生更大的絮状物并且具有增加的剪切敏感性,从而引起絮状物破坏,进而产生更高水平的更缓慢沉降的更小颗粒。在补缀或电荷中和的情况下,具有高电荷密度的阳离子型聚合物与悬浮液中的颗粒上的阴离子补缀物相互作用,并且中和颗粒上的电荷或形成更大颗粒而从溶液中沉降出。以这种方式形成的絮状物具有更小的絮状物颗粒体积并且更不易于由于剪切而被破坏。举例来说,将聚二烯丙基二甲基氯化铵(PDADMAC)添加至细胞培养肉汤中会经由静电补缀机制使带负电荷的细胞和细胞碎片絮凝成更大的颗粒(Ramsden等(1998),Biotechnology Techniques,12(8):599-603)。PDADMAC还使带负电荷的亚微米颗粒絮凝以产生与典型的以离心方式收获的进料流相比具有显著更高的收获过滤器组通量的进料流。经由离子相互作用所实现的絮凝作用可以通过提高盐浓度或改变pH值而被破坏。
将诸如聚二烯丙基二甲基氯化铵之类的阳离子型聚合物添加至含有或已经含有表达重组蛋白的细胞的哺乳动物细胞培养基中会使得包括细胞(活细胞和非活细胞)、细胞代谢物以及细胞碎片在内的带负电荷的颗粒絮凝。可以通过离心或通过重力沉降将这些大的絮凝颗粒去除。絮凝的细胞和细胞碎片沉降出所需的沉降速率或时间取决于细胞、细胞碎片以及细胞代谢物的密度。在对于分批式细胞培养过程典型的低细胞密度下,絮凝的物质典型地在4小时至24小时,典型地约20-24小时内的某个时间点沉降出(无进一步沉降)。沉降速率在产生高生物质细胞密度(>10%的细胞压积)、亚微米细胞碎片和/或高乳酸盐水平(>2-3g/L)的细胞培养过程的情况下显著降低。产生高细胞密度或具有升高的乳酸盐水平的细胞培养过程需要显著量的细胞肉汤稀释以使得絮状物在24小时内沉降出。虽然使用诸如PDADMAC之类的阳离子型聚合物作为传统收获方法的替代方案是非常方便的,但是长时间的沉降时间可能对于商业规模来说不太合乎需要。
本发明提供了经过阳离子型聚合物絮凝的物质的絮凝颗粒尺寸以及颗粒尺寸的增长率在非离子型聚合物和非离子型表面活性剂的存在下会得到极大地提高。在将PDADMAC与非离子型聚合物(如聚乙二醇)和非离子型表面活性剂(如Triton X100)组合使用时,通常观测到少于2小时的絮状物重力沉降时间,尽管高生物质/细胞密度会显著地延长单独PDADMAC絮凝的沉降时间。在添加非离子型聚合物和非离子型表面活性剂的情况下能够耐受可能破坏絮状物和/或降低絮凝速率的剪切力。在使得宿主DNA显著减少的情况下始终如一地实现80%-90%或更大的收获物回收产率。添加非离子型聚合物还会减少宿主细胞蛋白质和一些高分子量物质。
如本文所用的“阳离子型聚合物”是与带负电荷的悬浮颗粒结合的带正电荷的聚合物。阳离子型聚合物包括但不限于二烯丙基二甲基氯化铵(DADMAC)的聚合物。在一个优选的实施方案中,使DADMAC聚合形成N-取代的吡咯烷结构,即PDADMAC(图1)。阳离子型聚合物还包括聚乙烯亚胺(PEI)、聚丙烯酰胺(PAA)以及壳聚糖。
等于或约20皮克至等于或约90皮克的PDADMAC/总细胞密度的浓度会产生低的上清液浊度和良好的絮状物沉降作用。“总细胞密度”是如通过台盼蓝拒染(Trypan Blueexclusion),使用Cedex细胞计数器和分析器所测量的活细胞加上非活细胞的总数。在用于小细胞系(如二倍体细胞系)的一个实施方案中,添加等于或约25皮克/总细胞密度的PDADMAC。在另一个实施方案中,对于更大的细胞系,如四倍体细胞系,添加等于或约43皮克/总细胞密度至等于或约57皮克/总细胞密度的PDADMAC。
与更低分子量形式相比,分子量为200,000-500,000的PDADMAC通过提高沉降速率和上清液澄明度来影响絮凝性能。在一个实施方案中,PDADMAC的分子量在400,000至500,000的范围内。在一个实施方案中,以22皮克/总细胞密度的最终浓度使用具有在400,000-500,000范围内的分子量的PDADMAC。在另一个实施方案中,以25皮克/总细胞密度的最终浓度使用具有在400,000-500,000范围内的分子量的PDADMAC。在另一个实施方案中,以45皮克/总细胞密度的最终浓度使用具有在400,000-500,000范围内的分子量的PDADMAC。
如本文所用的“沉降速率”、“重力沉降速率”以及“絮凝并且压紧的沉降速率”可互换使用。沉降速率可以通过本领域已知的以及本文所述的方法来测定。举例来说,在1g下进行重力沉降。通过将絮状物体积除以在0.5L或1L玻璃量筒中所测量的总体积来确定沉降速率。总体积是包括所有絮凝剂/沉降剂在内的细胞肉汤体积。
“沉降时间”是絮状物沉降所要花费的时间。在絮状物的沉降速率小于或等于每小时1%时实现沉降时间。本文描述了在将PDADMAC絮凝与给予非离子型聚合物或非离子型聚合物连同非离子型表面活性剂组合的情况下沉降时间短达15分钟。沉降时间快速,尽管高生物质/细胞密度会显著地延长单独PDADMAC絮凝的沉降时间。在添加非离子型聚合物或非离子型表面活性剂的情况下能够耐受会破坏絮状物和/或降低絮凝速率的剪切力。
上清液澄明度与沉降速率无关,但是取决于阳离子型聚合物的给予水平。诸如温度、细胞培养流体密度以及粘度之类的其它因素对沉降速率或上清液澄明度的影响非常小。PDADMAC的给予量尤其是随细胞体积、总细胞(活细胞和非活细胞)密度以及亚微米细胞碎片颗粒的浓度而变的。
如本文所用的“非离子型聚合物”指的是增强分子之间的相互作用,从而增强沉淀作用的亲水性聚合物。非离子型聚合物包括但不限于聚乙二醇(PEG)、麦芽糖糊精、淀粉、甲基纤维素以及葡聚糖。
在添加阳离子型聚合物絮凝剂同时或之后将PEG或葡聚糖添加至哺乳动物细胞培养基中时会使得沉降速率有所提高。产物回收率取决于非离子型聚合物浓度、PEG分子量、非离子型聚合物和PDADMAC的添加顺序(同时添加或首先添加PDADMAC,继而添加非离子型聚合物)以及细胞培养持续时间或细胞培养肉汤中的碎片水平。
可使用在等于或约3%至等于或约4.5%(w/v)范围内的PEG 3,000。可使用在等于或约2.5%至等于或约3.5%(w/v)范围内的PEG 6,000。在一个实施方案中,以3%(w/v)的最终浓度使用PEG 3,000。在另一个实施方案中,以15%(w/v)的最终浓度使用PEG 3,000。在另一个实施方案中,以25%(w/v)的最终浓度使用PEG 3,000。
如本文所用的“非离子型表面活性剂”指的是具有两亲性的有机化合物,意指它们含有疏水性基团和亲水性基团,并且包括但不限于Sapoin和Triton X100。在一个实施方案中,以0.05%(w/v)的最终浓度使用Triton X-100。
可以单独添加或与非离子型表面活性剂组合添加非离子型聚合物。任一者均可以与阳离子型聚合物同时或在添加阳离子型聚合物之后添加。非离子型聚合物和非离子型表面活性剂两者均可以在1分钟或更短的添加时间内快速添加。
在絮凝体沉降(初级沉降)后,可以收获澄清了的上清液。为了提高重组产物回收率,可以将絮状物洗涤或重悬以便移去任何残留的重组产物。合适的洗涤稀释剂包括蔗糖、PEG、细胞培养基以及缓冲盐水溶液。在一个实施方案中,洗涤稀释剂是9%的蔗糖。将絮状物与洗涤稀释剂混合<1分钟至60分钟并且允许沉降约1小时至24小时。在絮状物沉降(二次沉降)后,可以收获二次澄清了的上清液。可以将来自初级沉降的上清液与来自二次沉降的上清液合并或单独地纯化。
可以通过抽吸或倾析取出上清液,继而经由容纳硅藻土的深度过滤器,继而0.2μ截留膜式过滤器过滤,或仅经由0.2μ截留过滤器过滤来收获澄清了的上清液。
阳离子型聚合物清除率可以通过本领域已知的方法,如用于监测哺乳动物细胞毒性的测定法;用于测定对通过DNA聚合酶或逆转录进行的DNA或RNA转录的抑制作用的测定法;以及测定mRNA的蛋白质翻译的测定法;以及本文所述的方法来监测。举例来说,重组蛋白纯化过程中间体的PDADMAC清除率可以通过在使用定量聚合酶链式反应(QPCR)中对DNA扩增的抑制作用来监测。
可以直接使用来自生物反应器的细胞培养肉汤或可以在絮凝之前将它冷却。在一个优选的实施方案中,细胞培养肉汤的温度范围是等于或约36℃至等于或约20℃。在另一个实施方案中,将细胞培养肉汤冷却至20℃或冷却至约20℃。
本发明提供了一种从哺乳动物细胞培养物中收获重组蛋白的方法。用于通过哺乳动物细胞培养物产生重组蛋白的商业过程中所使用的典型方法包括分批式培养法、分批补料培养法以及灌注培养法。分批式培养法是一种不连续的方法,其中将细胞在固定体积的培养基中培养较短的一段时间,然后完全收获。典型地在达到最高细胞密度(典型地是5-10×106个细胞/毫升)时进行收获。分批补料培养法提供了一次性或连续的培养基进料以便补充已经被消耗的那些培养基组分。由于分批补料培养物在整个操作期间接受额外的营养素,所以在与分批法相比时,它们有可能达到更高的细胞密度(>10至30×106个细胞/毫升)以及增加的产物滴度。在灌注法的情况下,典型的大规模商业细胞培养策略力图达到高细胞密度,即60-90(+)×106个细胞/毫升,其中反应器容积的差不多五十到超过一半是生物质。在灌注培养的情况下,已经达到了>1×108个细胞/毫升的极限细胞密度并且预期达到甚至更高的密度。

为了实现本发明的目的,细胞培养基是在体外细胞培养中适于动物细胞,如哺乳动物细胞生长的培养基。细胞培养基制剂是本领域熟知的。典型地,细胞培养基包含缓冲剂、盐、碳水化合物、氨基酸、维生素以及痕量的必需元素。细胞培养基可以含有或可以不含血清、蛋白胨和/或蛋白质。包括无血清培养基和成分明确培养基在内的各种组织培养基是可商购获得的,例如尤其可以使用以下细胞培养基中的任一种或组合:RPMI-1640培养基、RPMI-1641培养基、杜氏改良伊格氏培养基(Dulbecco's Modified Eagle's Medium,DMEM)、伊格氏最低必需培养基(Minimum Essential Medium Eagle)、F-12K培养基、汉姆氏F12培养基(Ham's F12 Medium)、伊斯科夫改良杜氏培养基(Iscove's ModifiedDulbecco's Medium)、麦考伊氏5A培养基(McCoy's 5A Medium)、莱氏L-15培养基(Leibovitz's L-15 Medium),以及无血清培养基,如EX-CELLTM 300系列(JRHBiosciences,Lenexa,Kansas)。细胞培养基可以补充有另外的组分或增加浓度的组分,如氨基酸、盐、糖、维生素、激素、生长因子、缓冲剂、抗生素、脂质、痕量元素等,这取决于所要培养的细胞的需要和/或所需的细胞培养参数。
细胞培养基可以是无血清、无蛋白质和/或无蛋白胨的。“无血清”适用于不含动物血清,如胎牛血清的细胞培养基。“无蛋白质”适用于不含外源添加的蛋白质,如转铁蛋白、蛋白质生长因子IGF-1或胰岛素的细胞培养基。无蛋白质培养基可以含有或可以不含蛋白胨。“无蛋白胨”适用于不含外源性蛋白质水解产物,如动物和/或植物蛋白水解产物的细胞培养基。细胞培养肉汤或类似术语指的是尤其含有活哺乳动物细胞和非活哺乳动物细胞、细胞代谢物以及细胞碎片(如核酸、蛋白质以及脂质体)的细胞培养基。
细胞培养或“培养”的意思是使细胞在多细胞生物体或组织外部生长和繁殖。适用于哺乳动物细胞的培养条件是本领域已知的。参见例如Animal cell culture:APractical Approach,D.Rickwood编著,Oxford University Press,New York(1992)。哺乳动物细胞可以悬浮培养或在贴壁于固体基质上时进行培养。具有或不具有微载体并且以分批、分批补料、连续、半连续或灌注模式操作的流化床生物反应器、中空纤维生物反应器、滚瓶、摇瓶或搅拌釜式生物反应器可供哺乳动物细胞培养使用。
可以在小规模培养,例如像100ml至大规模细胞培养,如具有成千和上万毫升的培养尺寸的***中培养哺乳动物细胞(如CHO细胞),以便在临床和商业上制造蛋白质治疗剂。
对细胞系(也被称为“宿主细胞”)进行遗传工程化以使其表达具有商业或科学价值的多肽。细胞系典型地源自于由原代培养物所产生的谱系,所述原代培养物可以在培养中维持无限的时间。对细胞系进行遗传工程化包括使用重组多核苷酸分子对细胞进行转染、转化或转导,和/或以另外的方式发生改变(例如通过使重组细胞与非重组细胞进行同源重组和基因活化或融合)以便产生表达所需重组多肽的宿主细胞。用于对细胞和/或细胞系进行遗传工程化以使其表达所关注的多肽的方法和载体是为本领域技术人员所熟知的;例如,各种技术说明于Current Protocols in Molecular Biology,Ausubel等编著(Wiley&Sons,New York,1988以及每季更新的内容);Sambrook等,Molecular Cloning:ALaboratory Manual(Cold Spring Laboratory Press,1989);Kaufman,R.J.,Large ScaleMammalian Cell Culture,1990,第15-69页。
适于在培养中生长的多种多样的哺乳动物细胞系可获自美国典型培养物保藏中心(American Type Culture Collection)(Manassas,Va.)和商业供应商。常用于工业中的哺乳动物细胞系的实例包括VERO、BHK、HeLa、CV1(包括Cos)、MDCK、293、3T3、骨髓瘤细胞系(例如NSO、NS1)、PC12、WI38细胞以及中国仓鼠卵巢(CHO)细胞。CHO细胞被广泛用于产生复杂的重组蛋白,例如细胞因子、凝血因子以及抗体(Brasel等(1996),Blood 88:2004-2012;Kaufman等(1988),J.Biol Chem 263:6352-6362;McKinnon等(1991),J Mol Endocrinol6:231-239;Wood等(1990),J.Immunol.145:3011-3016)。二氢叶酸还原酶(DHFR)缺陷型突变细胞系(Urlaub等(1980),Proc Natl Acad Sci USA 77:4216-4220)、DXB11以及DG-44是合乎需要的CHO宿主细胞系,这是因为高效的DHFR选择和可扩增基因表达***允许在这些细胞中进行高水平的重组蛋白表达(Kaufman R.J.(1990),Meth Enzymol 185:537-566)。另外,这些细胞作为贴壁或悬浮培养物是易于操作的并且展现出相对良好的遗传稳定性。CHO细胞和在它们当中重组表达的蛋白质已被广泛地表征并且已由监管机构批准用于临床商业制造。

实施例
实施例1
该实验对二烯丙基二甲基氯化铵(PDADMAC)的不同分子量制备物进行比较,使用这些制备物使哺乳动物细胞培养肉汤絮凝并且比较它们的沉降时间。
以分批补料培养法将表达重组单克隆抗体的CHO细胞在2,000L的生物反应器中培养15天。在测试之前将细胞培养肉汤冷却至10℃。设置一系列旋转烧瓶,其中每一个烧瓶中存在1L的细胞培养肉汤。PDADMAC以20%(w/v)液体的形式提供(Sigma Aldrich,St.Louis,MO),并且通过用纯水稀释至10%(w/v)来制备所有这些实验中所用的操作储备溶液。将分子量为100,000-200,000、200,000-350,000以及400,000-500,000的PDADMAC添加至每一个烧瓶中达到介于29皮克至86皮克的PDADMAC/总细胞密度之间的最终浓度。将PDADMAC溶液连续添加约1分钟并且在以70rpm-80rpm搅拌下在10℃下孵育15分钟。使絮状物在环境温度下沉降。将这一物质用于沉降时间测定。
在1,000L一次性反应器中培养第二批分批补料培养物,持续15天。将细胞培养肉汤维持在约36℃下。设置一系列旋转烧瓶,其中每一个烧瓶中存在1L的细胞培养肉汤。将分子量为<100,000、100,000-200,000、200,000-350,000以及400,000-500,000的PDADMAC添加至每一个烧瓶中达到介于25皮克至76皮克的PDADMAC/总细胞密度之间的最终浓度。将PDADMAC溶液连续添加约1分钟并且在以70rpm-80rpm搅拌下在约36℃下孵育15分钟。使絮状物在环境温度下沉降。将这一物质用于浊度测定。
通过将如使用Cedex细胞计数器和分析器(Roche Innovatis AG,Indianapolis,IN)由台盼蓝拒染所测量的活细胞总数与非活细胞总数相加来确定总细胞密度。然后将絮凝了的溶液转移到1L玻璃量筒中以测定絮凝并且压紧的沉降速率。以15分钟的时间间隔获得读数,持续90分钟并且将相对絮凝体积计算为沉降了的絮状物体积/总体积。
通过倾析,继而通过0.2μ过滤器将上清液从沉降并且絮凝了的细胞物质中取出。使用2100P浊度计(Hach,Loveland,CO)测量浊度。
图2A和2B示出了与平均分子量小于200,000的PDADMAC相比,使用具有大于200,000但是小于500,000的平均分子量的PDADMAC进行絮凝会产生最佳的沉降时间和澄明度。
实施例2
该实验比较使由小细胞系(如二倍体细胞系)与大细胞系(如四倍体细胞系)表达重组抗体的哺乳动物细胞培养肉汤絮凝所需的PDADMAC的量。
如上文所述培养二倍体细胞系和四倍体细胞系。设置一系列旋转烧瓶,其中每一个烧瓶中存在1L来自二倍体培养物和四倍体培养物中的每一种的细胞培养肉汤。在该实验和以下所有实验中,除非另外指出,否则使用具有400,000至500,000的平均分子量的PDADMAC。将PDADMAC添加至每一个烧瓶中达到如表2中所示的最终浓度。如上文所述测定总细胞密度。表2:用于二倍体培养物和四倍体培养物的PDADMAC的最终浓度

图3A示出了使用25皮克/总细胞的浓度的PDADMAC进行絮凝具有最快的沉降时间。
图3B示出了使用57皮克/总细胞的浓度的PDADMAC进行絮凝具有最快的沉降时间。
实施例3
该实验考察PDADMAC絮凝作用对产生高乳酸盐水平和/或具有高细胞密度的细胞培养物的影响。
将表达重组单克隆抗体的CHO细胞在1,000L一次性生物反应器中培养14天。细胞培养肉汤处于环境温度下。设置四个旋转烧瓶,其中每一个烧瓶中存在1L的细胞培养肉汤。在添加PDADMAC之前,在每一个烧瓶中搀入3g/L、6g/L或9g/L的60%(w/w)DL-乳酸钠(SigmaAldrich,St.Louis,MO),或不搀入乳酸盐作为对照组。然后将PDADMAC添加至每一个烧瓶中达到25皮克/总细胞密度的最终浓度(如上文所述的PDADMAC储液)。如上文所述测定总细胞密度。
如上文所述将PDADMAC在环境温度下连续地添加并且搅拌。然后使絮状物在环境温度下沉降。
然后将絮凝了的溶液转移到1L玻璃量筒中以测定絮凝并且压紧的沉降速率。以不同的时间间隔获得读数,持续1500分钟并且如上文所述计算相对絮凝体积。
以灌注培养法将第二批CHO细胞培养物在1,000L一次性生物反应器中培养20天。将细胞肉汤冷却至环境温度。在添加PDADMAC之前,使用细胞培养基将细胞肉汤稀释25%、50%以及75%。在添加PDADMAC之前不稀释细胞压积为44%的细胞肉汤。PDADMAC的最终浓度是25至26皮克/总细胞密度。PDADMAC的添加速率是约1分钟,在搅拌下进行添加并且如上文所述在环境温度下沉降。
然后将絮凝了的溶液转移到1L玻璃量筒中以测定絮凝并且压紧的沉降速率。以不同的时间间隔获得读数,持续1400分钟并且如上文所述计算相对絮凝体积。
图4A和4B示出了沉降速率在具有高乳酸盐水平(>2-3g/L)和/或产生高生物质细胞密度(>10%的细胞压积)的细胞培养过程的情况下显著降低。
实施例4
该实验比较了在将不同的稀释剂与PDADMAC组合使用时的沉降时间。
以灌注培养法将表达重组单克隆抗体的CHO细胞在1,000L一次性生物反应器中培养18天。在36℃下将来自第16天的细胞肉汤递送供测试使用并且在絮凝之前冷却至环境温度,持续2.5小时。在环境温度下进行添加和沉降。在添加PDADMAC之前,使用不同的稀释剂将细胞肉汤稀释至67%。乳酸盐水平是5g/L。PDADMAC的添加速率是约1分钟,在75rpm-85rpm下孵育15分钟的时间段。PDADMAC储备溶液是10%(w/v),是从20(w/v)的原始储备溶液制备而来(使用纯水)。PDADMAC的最终浓度是25皮克/总细胞密度。使絮状物在环境温度下沉降。
制备一系列稀释剂;浓度示于表3中。

然后将絮凝了的溶液转移到1L玻璃量筒中以测定絮凝并且压紧的沉降速率。以不同的时间间隔获得读数,持续1600分钟并且如上文所述计算相对絮凝体积。
如图5中所示,PDADMAC与PEG 6,000的组合具有最快的沉降时间。与单独的PDADMAC相比,PDADMAC与PEG 6,000、PEG 1,000或葡聚糖70/甜菜碱的组合具有提高了的沉降速率。添加非离子型聚合物会显著地提高絮状物的沉降速率。与单独的PDADMAC相比,PDADMAC与PEG或葡聚糖的组合也使沉降时间缩短,而与培养物中的乳酸盐水平无关。
实施例5
该实验考察PDADMAC与PEG 3,000的组合对具有高细胞密度的细胞培养物的沉降时间的影响。
以灌注培养法将表达重组单克隆抗体的CHO细胞在80L的生物反应器中培养20天。在测试之前将细胞培养肉汤冷却至环境温度。在添加PDADMAC之前,使用36%(w/v)的蔗糖或25%(w/v)的PEG 3,000(这两者均是纯水溶液)将细胞肉汤稀释至10%。最终细胞肉汤蔗糖浓度是3.6%(w/v),并且最终PEG 3,000浓度是2.5%(w/v)。PDADMAC的添加速率是约1分钟,在75rpm-85rpm下孵育15分钟的时间段。PDADMAC储备溶液是10%(w/v),是从20(w/v)的原始储备溶液制备而来(使用纯水)。对于PEG 3,000来说,PDADMAC的最终浓度是22皮克/总细胞密度,并且对于蔗糖和对照组或未稀释的细胞肉汤来说,PDADMAC的最终浓度是25皮克/总细胞密度。对照组或未稀释细胞肉汤的PCV是48%。稀释过的细胞肉汤的PCV=(对照组PCV X稀释因数)=43.2%。
然后将絮凝了的溶液转移到1L玻璃量筒中以测定絮凝并且压紧的沉降速率。以不同的时间间隔获得读数,持续240分钟并且如上文所述计算相对絮凝体积。
如图6中所示,与单独的PDADMAC相比,PDADMAC与PEG 3,000的组合引起更快速的沉降。如在实施例3中所看到,单独PDADMAC的沉降速率随细胞密度增加而显著降低。与单独的PDADMAC相比,PEG 3,000与PDADMAC的组合使絮凝体沉降时间缩短,而与细胞培养过程的密度无关(在此情况下,是43%的细胞压积)。
实施例6
该实验考察添加PEG与PDADMAC的时序对沉降时间的影响。
以灌注培养方法将表达重组单克隆抗体的CHO细胞在80L的生物反应器中培养19天。将细胞培养肉汤冷却至21℃。设置三个旋转烧瓶,其中每一个烧瓶中存在1L的细胞培养肉汤。向一个烧瓶中添加45皮克/总细胞密度的浓度的PDADMAC(分子量:400,000-500,000)。向另一个烧瓶中以一次性添加法(单步法)添加45皮克/总细胞密度的PDADMAC和15%(w/v)的PEG 3,000。向第三个烧瓶中,首先以45皮克/总细胞密度的浓度添加PDADMAC,并且在添加PDADMAC之后,添加PEG 3,000达到15%(w/v)的最终浓度(两步法)。PDADMAC的添加速率是约1分钟。PDADMAC/PEG的添加速率是约5分钟。所有添加均在环境温度下进行。在75rpm-85rpm下将所有烧瓶孵育15分钟。使絮状物在环境温度下沉降。
然后将絮凝了的溶液转移到1L玻璃量筒中以测定絮凝并且压紧的沉降速率。以不同的时间间隔获得读数,持续240分钟并且如上文所述计算相对絮凝体积。
如图7中所示,与单独的PDADMAC相比,同时添加或依次添加的PDADMAC与PEG 3,000的组合使絮凝体沉降时间缩短。
随后制备大规模的培养物。以灌注培养法将表达重组单克隆抗体的CHO细胞在80L的生物反应器中培养19天。将细胞培养肉汤冷却至21℃。在环境温度下同时添加45皮克/总细胞密度的浓度的PDADMAC与15%(w/v)的最终浓度的PEG 3,000,添加速率是21分钟,继而在100rpm下孵育5分钟。使絮状物在环境温度下沉降。
在絮凝体已沉降(初级沉降)后,通过从生物反应器中抽吸流体,继而经由容纳硅藻土的深度过滤器过滤,继而经由0.2μ截留膜式过滤器过滤来收获澄清了的上清液。
将絮状物在等体积的9%蔗糖溶液中洗涤以移去任何残留的重组蛋白并且允许沉降16小时。在絮状物沉降(二次沉降)后,如上文所述收获二次澄清了的上清液。
使用蛋白质A色谱法对来自上述絮凝(小规模和大规模)的所收获的澄清了的细胞培养上清液进行纯化,继而进行产物质量测定。在进行产物质量测定之前,不对蛋白质A洗脱物进行中和。所测量的蛋白质A洗脱物的产物质量属性是如通过SEC所测量的分子变体、如通过ELISA所测定的宿主细胞蛋白。
然后使经过蛋白质A纯化的物质穿过CEX柱(pH 7.5)。

对于两种规模来说,对照组与初级絮凝收获之间的产物质量是相似的。PDADMAC/PEG初级收获倾向于去除高级聚集体,这反映在蛋白质A池中HMW的水平较低。观测到PDADMAC/PEG收获物中的宿主细胞蛋白质水平降低。与初级PDADMAC/PEG收获相比,使用蔗糖进行重悬会使得CHOP和HMW的水平略微更高并且据信是这些杂质被重新溶解。
实施例7
该实验考察连同PDADMAC和PEG一起添加表面活性剂对沉降时间的影响。
以灌注细胞培养法将表达重组单克隆抗体的CHO细胞在80L的生物反应器中培养15天。将来自第14天的细胞肉汤冷却至30℃以用于测试。设置两个旋转烧瓶,其中每一个烧瓶中存在1L的细胞培养肉汤。向一个烧瓶中同时添加PDADMAC和PEG 3,000,以25皮克/总细胞密度的浓度添加PDADMAC(分子量:400,000-500,000),并且添加PEG 3,000达到3%(w/v)的最终浓度。向另一个烧瓶中,除了添加上述浓度的PDADMAC和PEG之外,还添加Triton X-100达到0.05%(v/v)的最终浓度。(Triton X-100储备溶液是10%(v/v),由20%(v/v)的原始储备溶液制得,Sigma Aldrich,St.Louis,Mo.)同时添加这三种组分。添加速率是约1分钟,在75-85rpm下孵育15分钟。如之前的实施例中所述将所有烧瓶旋转。使絮状物在环境温度下沉降。
然后将絮凝了的溶液转移到1L玻璃量筒中以测定絮凝并且压紧的沉降速率。以不同的时间间隔获得读数,持续240分钟并且如上文所述计算相对絮凝体积。
如图8中所示,与单独的PDADMAC和PEG 3,000相比,连同PDADMAC和PEG 3,000一起添加Triton X-100会使絮凝体沉降时间缩短。
Claims (18)
1.一种哺乳动物细胞培养物收获方法,所述方法包括
将表达重组蛋白的哺乳动物细胞在细胞培养基中培养预定的时间或直到达到所需的细胞密度和/或细胞压积为止,
将阳离子型聚合物和非离子型聚合物以及非离子型表面活性剂添加至所述细胞培养基中,从而引发絮凝,所述阳离子型聚合物选自二烯丙基二甲基氯化铵的聚合物和聚二烯丙基二甲基氯化铵的聚合物,所述非离子型聚合物选自聚乙二醇和葡聚糖,
在絮凝期间混合所述细胞培养基,
使絮凝体沉降以进行初级沉降,以及
回收初级澄清了的上清液。
2.根据权利要求1所述的哺乳动物细胞培养物收获方法,其中所述非离子型聚合物选自PEG 3,000和PEG 6,000。
3.根据权利要求2所述的方法,其中PEG 3,000的浓度是3%至4.5%。
4.根据权利要求2所述的方法,其中PEG 6,000的浓度是2.5%至3.5%。
5.根据权利要求1所述的方法,其中所述非离子型表面活性剂是Sapoin或TritonX100。
6.根据权利要求1所述的哺乳动物细胞培养物收获方法,其中所述非离子型表面活性剂是Triton X-100。
7.根据权利要求6所述的方法,其中Triton X100的浓度是0.05%(w/v)。
8.根据权利要求1所述的哺乳动物细胞培养物收获方法,所述方法进一步包括
对所述初级沉降的絮凝体进行洗涤,
使经过洗涤的絮凝体沉降以进行二次沉降,以及
回收所述二次澄清了的上清液。
9.根据权利要求8所述的方法,其中在9%蔗糖溶液中对来自所述初级沉降的所述絮凝体进行洗涤。
10.根据权利要求1所述的方法,其中同时添加所述阳离子型聚合物和所述非离子型聚合物。
11.根据权利要求1所述的方法,其中首先添加所述阳离子型聚合物并且混合至少30秒,继而添加所述非离子型聚合物。
12.根据权利要求1所述的方法,其中同时添加所述阳离子型聚合物、所述非离子型聚合物以及一种非离子型表面活性剂。
13.根据权利要求1所述的方法,其中首先添加所述阳离子型聚合物并且混合至少30秒,继而添加所述非离子型聚合物和一种非离子型表面活性剂。
14.根据权利要求1所述的方法,其中以20皮克/总细胞密度至90皮克/总细胞密度的浓度添加所述聚二烯丙基二甲基氯化铵。
15.根据权利要求1所述的方法,其中以25皮克/总细胞密度的浓度添加所述聚二烯丙基二甲基氯化铵,其中所述哺乳动物细胞来源于二倍体细胞系。
16.根据权利要求1所述的方法,其中添加介于43皮克/总细胞密度与57皮克/总细胞密度之间的所述聚二烯丙基二甲基氯化铵,其中所述哺乳动物细胞来源于四倍体细胞系。
17.根据权利要求1所述的方法,其中所述哺乳动物细胞培养基在36℃与20℃之间。
18.根据权利要求1所述的方法,其中所述哺乳动物细胞培养基处于20℃或高于20℃。
