本発明で提供される核酸及び/又はアミノ酸配列は、37C.F.R.1.822に定義されるように、ヌクレオチド塩基についての標準的な文字の略語、及びアミノ酸についての3文字のコードを用いて示される。各核酸配列について一方の鎖のみが示されているが、相補鎖は表示された鎖を参考することによって理解され得る。
配列番号3及び4は、cIAP1のsiRNA干渉断片2のそれぞれのセンス鎖及びアンチセンス鎖である。
配列番号5及び6は、cIAP1のsiRNA干渉断片3のそれぞれのセンス鎖及びアンチセンス鎖である。
配列番号7及び8は、cIAP2のsiRNA干渉断片1のそれぞれのセンス鎖及びアンチセンス鎖である。
配列番号9及び10は、cIAP2のsiRNA干渉断片2のそれぞれのセンス鎖及びアンチセンス鎖である。

本明細書に記載されているのは、驚くべき抗腫瘍効果をもたらす、カスパーゼ活性化剤とアルファウイルスの組み合わせを用いた抗腫瘍療法の発見である。カスパーゼ活性化は細胞アポトーシスを引き起こすことが知られている。カスパーゼファミリーのプロテイナーゼは、3つの亜群に分けられる。カスパーゼ−1、4、5及び11は第1亜群、カスパーゼ−2及び9は第2亜群、カスパーゼ−3、7、8,及び10は第3亜群に属する。最初のカスパーゼは、外来タンパク質シグナルの作用下で切断され活性化され、活性化されたカスパーゼは、さらに及びエグゼキューターカスパーゼを切断し活性化し、活性化されたエグゼキューターカスパーゼは、さらにカスパーゼ標的タンパク質を加水分解することによってプログラム細胞死(即ち、アポトーシス)をもたらす。
ヒト大腸がん細胞株HCT116(ATCCから購入)、M1ウイルス(CTCC V201423から取得)、高グルコースDMEM培地(4.5g/lグルコース)(Corningから購入)、倒立位相差顕微鏡
a)細胞培養
ヒト大腸がん細胞株HCT116を10%FBS、100U/mlのペニシリン及び0.1mg/mlのストレプトマイシンを含むDMEM完全培地中で増殖させた。全ての細胞を5%CO2、37℃の恒温密閉インキュベーター(相対湿度95%)に入れて継代培養した。培養した細胞の増殖を倒立顕微鏡で観察した。細胞を約2〜3日毎に継代し、対数増殖期にある細胞をアッセイに使用した。
図1に示すように、位相差顕微鏡により細胞の形態を観察した。HCT116細胞は、単一層で接着増殖し、同一な表現型で緊密に並んでいた。LCL161(5μM)及びM1ウイルス(MOI=1)で72時間処理した後、細胞の形態は顕著に変化した。対照群の細胞、M1単独処理群の細胞、及びLCL161単独処理群の細胞と比較すると、併用処理群の細胞数は顕著に減少し、細胞体は球形に収縮し、屈折率は顕著に増加し、細胞は細胞死を示す病理学的変化を示した。
ヒト大腸がん細胞株HCT116、SW480、ヒト肝臓がん細胞株Huh7、PLC、M1ウイルス、高グルコースDMEM培地、酵素免疫測定法のための自動マイクロプレートリーダー。細胞及びウイルスは実施例1と同じ供給源に由来する。
a)細胞の培養及び投与処理
対数増殖期にある細胞を選択してDMEM完全培地(10%ウシ胎児血清及び1%ペニシリン/ストレプトマイシンを含む(Life Technologies))に加え、細胞懸濁液を調製した。細胞を4×103/ウェルの密度で96ウェル培養プレートに接種した。12時間後、細胞はウェルに完全に付着した。試験を対照群、単剤群(5μMLCL161単独又はビリナパント単独)、M1感染群、及びLCL161/M1併用群又はビリナパント/M1併用群に分けた。M1含有アッセイでは、ウイルスは様々なMOIで提供された。LCL161又はビリナパントの濃度は5μMであった。
培養72時間後、20μlのMTT(5mg/ml)を各ウェルに加え、さらに4時間インキュベートした。インキュベートした後、生細胞中に形成された粒状の青紫色ホルマザン結晶を顕微鏡検査により観察した。
ホルマザン結晶を有する細胞を含む培養物を以下のようにさらに処理した。上澄みを注意深く吸い取って捨てた後、100μl/ウェルでDMSOを加えて形成された結晶を溶解し、マイクロ振動機で5分間振盪した。次いで、酵素結合検出器を用いて570nmの波長で各ウェルの光学密度(OD値)を測定した。実験は各群につき3回繰り返した。細胞の生存率=薬物処理群のOD値/対照群のOD値×100%
図2aに示すように、M1ウイルス単独処理は、腫瘍細胞株HCT116の生存率を減少させた(84.0%)。5μMのLCL161で処理した腫瘍細胞の生存率は92.6%と高かった。一方、M1/LCL161の組み合わせ(M1+LCL161)で処理した腫瘍細胞は、生存率が33.6%に急激に低下した。同様に、M1ウイルス単独処理群又はLCL161単独処理群に比べて、様々な用量のLCL161/M1の組み合わせで処理した腫瘍細胞は、生存率が大幅に低下した。ビリナパント/M1併用処理群では、HCT116について同様な結果が観察された(図2b)。さらに、LCL161又はビリナパントとM1との組み合わせも腫瘍細胞SW620(図2c及びd)、Huh7細胞(図2e及びf)及びPLC細胞(図2g及びh)の生存率を顕著に低下させた。
M1ウイルス、ヒト大腸がん細胞株HCT116、肝臓がん細胞株Huh7、cIAP1及びcIAP2のRNA干渉断片、MTT(メチルチアゾリルテトラゾリウム)(MPbioから購入)、位相差顕微鏡。細胞及びウイルスは実施例1と同じ供給源に由来する。
対数増殖期にある細胞を選択してDMEM完全培地に加え、細胞懸濁液を調製した。細胞を1×105/ウェルの密度で6ウェルプレートに接種した。24時間後、リポソームで包まれたSiRNA標的遺伝子断片を加えた(図3a−3d、配列番号1−12)。48時間後、細胞をM1ウイルスに感染させた。感染の48時間後、試料を以下のように処理した。
(1)細胞の一部を採取し、タンパク質を単離した。ウエスタンブロットによりRNA干渉効率を検出した。
(2)MTT法により細胞の生存率を計算した。
異なる干渉断片でcIAP1及びcIAP2をそれぞれ干渉した後、IAPタンパク質のウエスタンブロット検出により測定した。少なくとも2つのsiRNA断片がcIAP1及びcIAP2タンパク質(図3a及び3b)の発現を顕著に低減できることが発見された。これは、RNAi干渉断片が翻訳のレベルでcIAP1及びcIAP2タンパク質の発現を干渉できることを示している。
図3c−3dには、controlは対照群、即ち、未処理群を示し、mockはトランスフェクション試薬RNAiMAX)単独使用群を示し、NCはスクランブルsiRNA干渉断片群を示し、最後の3つのコラムは発現がsiRNA断片によって阻害された細胞を示す。
処理細胞のMTT法により、cIAP1及びcIAP2単独のRNA干渉群は、非干渉群(対照群)又はスクランブル干渉(NC)群と比較して、腫瘍細胞の生存率の変化を引き起こさず、M1ウイルス単独による干渉も細胞生存率の変化を引き起こさない一方、cIAP1又はcIAP2のRNA干渉とM1感染との組み合わせは、腫瘍細胞の生存率を顕著に減少させることが実証された(図3c及び3d)。データをANOVA統計にかけたところ(*はp<0.05を示し、**はp<0.01を示し、***はp<0.001を示す)、細胞の生存率の減少に統計的有意性があることを示した。
これらの結果は、cIAP1及びcIAP2の阻害とM1ウイルスの使用との組み合わせは、抗腫瘍効果を生じることを示している。
M1ウイルス、大腸がん細胞株HCT116、肝臓がん細胞株Huh7、4週齢の雌BALB/cヌードマウス(南京大学モデル動物研究センター)。細胞及びウイルスは実施例1と同じ供給源に由来する。
無作為化単盲検試験を実施した。5×106個のHCT116又は1×107個のHuh7細胞を4週齢のBALB/cヌードマウスの背部に皮下注射した。腫瘍の大きさが50mm3に達した後、マウスを未処理対照群(対照)、LCL161単独処理群(i.p. 20mg/kg/d)、M1単独感染群(HCT116がん株の移植可能な腫瘍をM1ウイルスと共に尾静脈注射した(2×106PFU/回)、Huh7がん株の移植可能な腫瘍をM1ウイルスと共に尾静脈注射した(5×105PFU/回))、及びLCL161/M1併用群(LCL161及びM1ウイルスを同じ方法及び同じ用量で投与する)に分け、注射を4回連続して行った(HCT116は、5日目、6日目、7日目及び8日目に注射を行った。Huh7は、10日目、11日目、12日目及び13日目に注射を行った)。腫瘍の重量、長さ及び幅を2日ごとに測定し、腫瘍体積を式:(長さ×幅2)/2に従って計算した。腫瘍体積を測定した後、一元配置分散分析統計を行った。ここで、***はp<0.001、**はp<0.01、*はp<0.05を示す。
腫瘍移植動物についての病理学研究を実施して、腫瘍体積を測定した。結果は、対照群と比較して、LCL161単独処理群及びM1単独処理群は、腫瘍体積のわずかな減少しかもたらさなかった(図4a及び4b。左パネル) .
HCT116腫瘍を接種した動物において、LCL61処理群は、対照群と比較して、わずか29.15%の腫瘍体積の減少を示した。M1処理群は、対照群と比較して、わずか27.93%の腫瘍体積の減少を示した。一方、LCL161/M1併用処理群は、腫瘍体積の顕著な減少を誘導した(図4a及び4b、左パネル)。対照群と比較して、併用処理群は80.42%の腫瘍体積の減少をもたらした。
同様に、Huh7腫瘍を接種した動物において、LCL161処理群は、対照群と比較して、わずか18.1%の腫瘍体積の減少を示した。M1処理群は、対照群と比較して、わずか4.7%の腫瘍体積の減少を示した。一方、LCL161/M1併用処理群は、腫瘍体積の顕著な減少を誘導した(図4a及び4b、右パネル)。対照群と比較して、併用処理群は70.3%の腫瘍体積の減少をもたらした。図に示すように、一元配置分散分析統計は提示されたデータの統計的有意性を示す。
M1ウイルス、LCL161、カスパーゼ3/7/8/9の活性を検出するキット (Promegaから購入;カスパーゼ3/7 G8091;カスパーゼ8 G8201;カスパーゼ9 G8211)、DMEM完全培地(10%ウシ胎児血清、1%ペニシリン/ストレプトマイシンを含む(Life Technologies))、大腸がん細胞株HCT116、肝臓がん細胞株Huh7、酵素免疫測定法のための自動マイクロプレートリーダー。細胞及びウイルスは実施例1と同じ供給源に由来する。
対数増殖期にある細胞を選択してDMEM完全培地(10%ウシ胎児血清及び1%ペニシリン/ストレプトマイシンを含む(Life Technologies))に加え、細胞懸濁液を調製した。細胞を4×103/ウェルの密度で96ウェル培養プレートに接種した。12時間後、細胞は培養プレートの壁に完全に付着した。この試験において、培養物を対照群、単剤群(LCL161単独)、M1単独感染群、LCL161/M1併用処理群に分けた。用量は以下の通りである。細胞を1MOI(HCT161細胞)又は0.1MOI(Huh7細胞)でM1に感染させ、LCL161を5μMの濃度で加えた。MTTとカスパーゼ3/7、8及び9の活性を検出するために、上記の各処理群を4つに分けた。処理の72時間後、体積に基づいてカスパーゼ3/7、8及び9を検出するための試薬を各ウェルに添加し、得られたウェルをインキュベーターに入れてさらに30分間培養した。液体を不透明な96ウェルプレートに吸い込んで、マイクロプレートリーダーにより化学発光強度を測定した。一方、MTTを検出して細胞の生存率を計算した。カスパーゼの活性は、MTT値に対する化学発光値の比によって標準化された。
図5a−5fに示すように、検出されたカスパーゼ3/7、8及び9の活性は、HCT116細胞及びHuh7細胞において、対照群(Ctrl)と比較して、LCL161又はM1(M)単独による処理は、カスパーゼ3/7、8及び9の活性をわずかに向上させることを示した。一方、LCL161/M1併用処理群(LM)は、カスパーゼ3/7、8及び9において顕著な向上を示した(図5a−f)。これらの観察は、カスパーゼ活性剤、即ち、LCL161とM1の組み合わせがカスパーゼ3/7、8、及び9の活性を顕著に活性化できることを実証する。

前記IAPタンパク質のレベルを低下させる遺伝的手段は、RNA干渉、マイクロRN A、遺伝子編集又は遺伝子ノックアウト材料である、IAP阻害剤を含むアルファウイル スの抗腫瘍相乗剤。 - 前記組成物は、腫瘍を治療する組成物である、請求項1に記載の医薬組成物又はキット 又は相乗剤。
- 前記少なくとも1つのIAP阻害剤及び前記少なくとも1つのアルファウイルスは、別 々の成分として存在する、請求項1または3に記載のキット。
- 前記アルファウイルスは、寄託番号CCTCC V201423で寄託されているM1 ウイルスのゲノムに対して少なくとも97.8%のヌクレオチド配列同一性を有する、請 求項1から4のいずれか1項に記載の医薬組成物又はキット又は相乗剤。
- 前記アルファウイルスは、寄託番号CCTCC V201423で寄託されているM1 ウイルスのゲノムに対して少なくとも100%のヌクレオチド配列同一性を有する、請求 項1から5のいずれか1項に記載の医薬組成物又はキット又は相乗剤。
- 前記腫瘍は、固形腫瘍又は血液腫瘍である、請求項2から6のいずれか1項に記載の医 薬組成物又はキット又は相乗剤。
- 前記固形腫瘍は、肝臓がん、大腸がん、膀胱がん、乳がん、子宮頸がん、前立腺がん、 神経膠腫、黒色腫、膵がん、鼻咽頭がん、肺がん、又は胃がんである、請求項7に記載の 医薬組成物又はキット又は相乗剤。
- 前記腫瘍は、腫瘍溶解性ウイルスに非感受性の腫瘍である、請求項2から8のいずれか 1項に記載の医薬組成物又はキット又は相乗剤。
