上記で説明したように、本発明者らは、約+5℃以上、液体状態でポックスウイルス、特に、ワクシニアウイルスの安定性を維持するのに好適な液体処方物を特定した。このような処方物は、薬学上許容可能なバッファー、一価の塩、薬学上許容可能な二糖または糖アルコール、薬学上許容可能なキレート剤を含んでなり;およびpHは6.5〜8.5の間であるべきである。
a)ポックスウイルス、特に、ワクシニアウイルス、
b)薬学上許容可能なバッファー、
c)一価の塩、
d)薬学上許容可能な二糖または糖アルコール、および
e)薬学上許容可能なキレート剤
を含んでなるか、これらから本質的になるか、またはこれらからなる、そのpHが6.5〜8.5である液体処方物に関する。
本発明による処方物は液体であり、コストと時間のかかる凍結乾燥工程を必要としない、かつ、事前の再構成の必要なくそのまま投与できるという利点を有する。
・本発明による液体処方物中の感染性ポックスウイルス、特に、ワクシニアウイルス力価の損失は、+37℃で少なくとも1週間、好ましくは少なくとも2週間、少なくとも3週間、またはさらには少なくとも4週間の保存中に、好ましくは、感染性ウイルス1log10未満である、
・本発明による液体処方物中の感染性ポックスウイルス、特に、ワクシニアウイルス力価の損失は、+25℃で少なくとも4週間、好ましくは少なくとも3か月、または少なくとも6か月の保存中に、好ましくは、感染性ウイルス1log10未満である、かつ/または
・本発明による液体処方物中の感染性ポックスウイルス、特に、ワクシニアウイルス力価の損失は、約+5℃(すなわち、+5℃±3℃)で少なくとも12か月、好ましくは少なくとも18か月、少なくとも24か月、少なくとも30か月、またはさらには少なくとも36か月の保存中に、好ましくは、感染性ウイルス0.3log10(感染力の50%の損失に相当する)未満である。
本発明による処方物は、ポックスウイルスを含んでなる。
−MVA−FCU 1(WO99/54481参照)、TG4023とも呼ばれる;
−MVA−FCU 1−8(WO2005/007857参照);および
−VV−FCU 1、前記ワクシニアウイルス(VV)は、より詳しくは、欠陥型I4Lおよび/またはF4L遺伝子、欠陥型J2R遺伝子(WO2009/065546およびWO2009/065547参照)を含んでなる、

本発明による液体処方物は、一価の塩を含んでなる。この一価の塩は、適当な浸透圧を保証すると考えられる。加えて、前記一価の塩はまた、処方物中に存在し得るプロテアーゼの阻害特性を有し、従って、安定性を改善するとも考えられる。このようなプロテアーゼには、パッケージング細胞を破壊する場合およびまた、処方物中に存在するワクシニアウイルスがプロテアーゼの使用を含む方法により精製され、前記の付加されたプロテアーゼの痕跡が残留する場合に遊離する細胞プロテアーゼが含まれる。プロテアーゼに対する有意な一価の塩濃度の阻害作用が当技術分野で報告されている(TOUGU V et al. Eur J Biochem. 1994 Jun , 222(2):475-81)。例えば、Pierceトリプシンプロテアーゼ(Thermo Scientific)に関して、製造者は説明書で、>100mM NaClのような高い一価の塩濃度はトリプシン活性を妨げることを示している。
本発明による液体処方物は、薬学上許容可能な二糖または糖アルコールを含んでなる。
本発明による液体処方物は、薬学上許容可能なキレート剤、特に、薬剤キレートジカチオンを含んでなる。
本発明による液体処方物は、ワクシニアウイルスに対して安定化効果を持つ付加的化合物をさらに含んでなってもよい。
6.5〜8.5の間のpHを有することが可能な薬学上許容可能なバッファー、一価の塩、薬学上許容可能な二糖または糖アルコール、および薬学上許容可能なキレート剤の存在は液体状態のワクシニアウイルスに対する安定化に不可欠であることが判明したが、本発明者らはまた、低濃度のC2−C3アルコールの付加的存在が、ワクシニアウイルスの安定化には必要でないが、液体状態のワクシニアウイルスの安定性をさらに改善するためにキレート剤の存在と相乗作用することを見出した。これに対して、同じC2−C3アルコールの濃度が高すぎると液体状態のワクシニアウイルスの安定性に有害な影響を持つ。よって、本発明による液体処方物は好ましくは、0.05〜5%(容量/容量またはv/v)濃度のC2−C3アルコールをさらに含んでなる。アデノウイルスなどの非エンベロープウイルスの場合とは対照的に、ポックスウイルス、特に、ワクシニアウイルスはエンベロープウイルスであって、極性溶媒の添加がエンベロープを変化させないと思われることから、この所見は全く予期されないものであった。

界面活性剤
非イオン性界面活性剤は、液体状態の種々のウイルスの安定化を誘導することが示されている(EVANS et al. J Pharm Sci. 2004 Oct, 93(10):2458-75、米国特許第456,009号、SHI et al. J Pharm Sci. 2005 Jul, 94(7):1538-51、米国特許出願第2007/0161085号参照)。
MgCl2またはCaCl2などの二価の塩は、液体状態の種々のウイルスの安定化を誘導することが見出されている(EVANS et al. J Pharm Sci. 2004 Oct, 93(10):2458-75および米国特許第7,456,009号参照)。このような場合、二価の陽イオンは液体処方物中に少なくとも0.5mM、好ましくは少なくとも1mMの濃度で存在した。
アミノ酸、特に、ヒスチジン、アルギニンまたはメチオニンは、液体状態の種々のウイルスの安定化を誘導することが見出されている(EVANS et al. J Pharm Sci. 2004 Oct, 93(10):2458-75、米国特許第7,456,009号、米国特許出願第2007/0161085号、米国特許第7,914,979号、WO2014/029702、WO2014/053571参照)。安定性を高めるために液体処方物中にヒスチジンが存在する場合、ヒスチジンは一般に少なくとも5mM、好ましくは少なくとも10mMの濃度で存在する(EVANS et al. J Pharm Sci. 2004 Oct, 93(10):2458-75、米国特許第7,456,009号、WO2014/029702参照)。安定性を高めるために液体処方物中にアルギニンが存在する場合、アルギニンは一般に少なくとも50mM(米国特許出願第2007/0161085号、少なくとも1%w/vアルギニンは少なくとも約57.4mMに相当する)、場合によって好ましくは少なくとも150mM、特に、約300mM(WO2014/029702参照)の濃度で存在する。安定性を高めるために液体処方物中にメチオニンが存在する場合、メチオニンは一般に少なくとも25mM、好ましくは約67mMの濃度で存在する(WO2014/029702参照)。
本発明者らはまた、尿素がワクシニアウイルスの安定性に有益な効果を持たないことをも見出した(実施例1参照)。
凍結乾燥させたウイルス処方物は一般に、デキストランまたはポリビニルピロリドン(PVP)などの高分子量ポリマーを含有し、これらは凍結乾燥中に塊状物の形成を助ける(EP1418942およびWO2014/053571参照)。
血清またはゼラチンなどの動物またはヒト由来安定剤は、生ウイルスの安定化のために長い間使用されてきた(米国特許出願第2007/0161085号参照)。しかしながら、動物またはヒト起源のこのような動物またはヒト由来安定剤は、ウイルスまたは非従来型因子の潜在的混入のために潜在的に健康リスクを含む。
本発明による処方物の各必須要素または任意選択要素の系列に属す種々の具体的化合物を、上記でこの要素に特に関連する節に記載した。本発明に関して、特定の要素に関する適当な化合物の各リストおよび特定の要素に関して開示されている各具体的化合物は、一般的な他の任意の要素、前記の他の要素に関する適当な化合物のリストまたは前記の他の要素に関して開示されている任意の具体的化合物と組み合わせてもよい。
ワクシニアウイルスはUV損傷を特に受けやすい(LYTLE et al. J. Virol. 2005, 79(22):14244参照)。
液体処方物中、+5℃でのMVAウイルスを安定化させるための種々の候補化合物の効果は、他の種類のウイルスの安定化効果を有することが従来技術から知られている化合物に基づいて試験した。
ウイルス
以下のMVAウイルスを使用した:
・1〜4 108PFU/mLの初期目標濃度に希釈したMUC1腫瘍関連抗原およびインターロイキン2を発現する組換えMVAウイルスであるMVA−MUC1(TG4010)(WO92/07000およびWO95/09241参照)。
・1〜4 107PFU/mLの初期目標濃度に希釈したHCV遺伝子型1b株ja由来の非構造HCVタンパク質(NS3、NS4およびNS5B)を発現する組換えMVAウイルスであるMVA−HCV(TG4040)(WO2004/111082参照)。
・0.5〜2 108PFU/mLの初期目標濃度に希釈したヒト乳頭腫ウイルスE6およびE7抗原およびインターロイキン2を発現する組換えMVAウイルスであるMVA−HPV(TG4001)(WO90/10459、WO95/09241、WO98/04705、WO99/03885、WO2007/121894参照)。
供試処方物を以下の表2〜11に示す。

安定性は+37℃(±2℃)、+25℃(±2℃)、および/または+5℃(±3℃)で分析した(結果の節を参照)。
所与の時点での感染力価は、mL当たりの感染ゲノム(IG)の数(IG/mL)を測定することまたはBHK−21細胞でのプラークアッセイを使用すること(その後、感染性ワクシニアウイルス力価はmL当たりのプラーク形成単位(PFU)(PFU/mL)で表される)のいずれかによって測定され得る。ここでは、mL当たりの感染ゲノムの数(IG/mL)の測定が、この方法がより迅速かつより正確であることから好ましかった。しかしながら、BHK−21細胞でのプラークアッセイを用いた場合に特異なデータが示されるわけではなく、BHK−21細胞でのプラークアッセイを用いてもいくつかの時点で感染力価が測定され、それらの結果は感染ゲノム法を用いて得られた結果と一致することが常に判明したことを記載しておくべきである。
・D日:感染および細胞播種
ウイルスサンプルを、96ウェル培養プレート(100μL/ウェル)にて、5%ウシ胎仔血清(FCS)を添加したDMEM培養培地を連続希釈する。
BHK−21細胞を培養培地(DMEM+5%FCS)に採取し、ウイルス希釈液を含有する96ウェルプレートに1:100の比率で播種する(100μL/ウェル)。
次に、培養プレートを+37℃、5% CO2で24時間+/−4時間インキュベートする。
感染20〜28時間後に、上清を廃棄し、細胞単層をPBSで2回洗浄する。プロテイナーゼKを含有する100μLの溶解バッファーを各ウェルに加える。このプレートを+56°で少なくとも30分(420分まで)インキュベートした後、プロテイナーゼKを不活化するために+95℃で5分間加熱する。
細胞溶解液を水で49倍希釈し、ワクシニアウイルスゲノムの分泌ケモカイン結合タンパク質(SCBP)領域を標的とする特異的プライマーおよびプローブセットを用いてqPCRを行う。
1.細胞の伸展
宿主細胞BHK−21をDMEM中、単層として増殖させた。コンフルエント時に細胞を10mL PBSで洗浄した後、トリプシンで処理した。トリプシンを除去した後、細胞を37℃で10%SFVを含む10mL DMEMに再懸濁させた。
細胞伸展の約1日後に、工程1のBHK−21細胞を含んでなる各ウェルにウイルス懸濁液のアリコートを加えた。必要に応じて、前記懸濁液をまず、当業者に周知の方法に従って、PBS、陽イオン100×および1%ウシ胎仔血清(FCS)で連続希釈した。場合に応じて、工程1のBHK−21細胞に加えたウイルス懸濁液は、凍結乾燥の前の液体ウイルス含有組成物か再構成したウイルス含有組成物(すなわち、凍結乾燥後、種々の時点および温度)かのいずれかであった。
培地を除去した後、細胞をPBS(約1mL/ウェル)で洗浄した。その後、1mLのメタノール/アセトン(50/50)溶液を加え、得られた混合物を室温で穏やかに撹拌した。
ウイルス力価の決定は、周知のペルオキシダーゼ反応に従い、ペルオキシダーゼと組み合わせた抗ワクチン抗体および抗ウサギ抗体を用いて行った。より厳密には、反応前に、抗ワクチン抗体をPBS+2%FCSで100倍希釈した。その後、500μLの前記抗体を各ウェルに加え、37℃で約30分間インキュベートした後、1mL PBS+1%トリトンX−100で3回洗浄した。
[ウイルスプラーク数の平均×4]×希釈倍率=PFU数/mL
を用いて感染力価をPFU/mLで計算した。
一価の塩の必要性
MVA−MUC1の安定性を、トリス−HCl、グルタミン酸Na、スクロース、pH8.0を含有し、かつ、0mMまたは50mMいずれかのNaClを含有する処方物(上記表2を参照)中で試験した。
トリス−HCl、グルタミン酸Na、スクロース、NaClを含有するpH7.5の処方物中、+37℃および+5℃でのMVA−HCVの安定性に対するEDTAの影響を、種々の濃度のEDTAを用いて試験した(表3参照)。結果を図2に示す。
トリス−HCl、グルタミン酸Na、スクロース、NaClを含有するpH7.5の処方物中、+37℃および+25℃でのMVA−HCVの安定性に対するグルタミン酸Naの影響を、0〜10mMの種々の濃度のグルタミン酸Naの含有または不含で試験した(表5参照)。結果をそれぞれ図5A、5Bおよび5Cに示す。
トリス−HCl、グルタミン酸Na、スクロース、NaClを含有するpH8.0の処方物中、+37℃でのMVA−MUC1の安定性に対するスクロースの影響を、種々の濃度のスクロースを用いて試験した(表6参照)。結果を図6に示し、スクロース濃度は安定性レベルに有意な影響を及ぼさないことを示す。
トリス−HCl、グルタミン酸Na、スクロース、NaClを含有するpH7.5の処方物中、+25℃および+5℃でのMVA−HPVの安定性に対するポリソルベート80またはポリソルベート40の影響(表7参照)を、他のウイルスに対して安定化効果を持つことが示されている(EVANS et al. J Pharm Sci. 2004 Oct, 93(10):2458-75、米国特許第7,456,009号、SHI et al. J Pharm Sci. 2005 Jul, 94(7):1538-51、米国特許出願第2007/0161085号参照)種々の濃度のポリソルベート80またはポリソルベート40を用いて試験した。結果を図7Aおよび7Bに示す。
トリス−HCl、グルタミン酸Na、スクロース、NaClを含有するpH8.0の処方物中、14日間、+37℃でのMVA−MUC1の安定性に対するMgCl2の影響(表8参照)を、他のウイルスに対して安定化効果を持つことが示されている(EVANS et al. J Pharm Sci. 2004 Oct, 93(10):2458-75および米国特許第7,456,009号参照)種々の濃度のMgCl2を用いてまたは用いずに試験した。結果を図8に示し、MgCl2はMVA安定性に対して有益でない影響を持ち、少なくとも0.5Mの濃度でMVAの安定性に対して有害な影響さえ持つことを明らかに示す。
トリス−HCl、グルタミン酸Na、スクロース、NaClを含有するpH8.0の処方物中、+37℃でのMVA−MUC1の安定性に対するアルギニンの影響(表9参照)を、他のウイルスに対して安定化効果を持つことが示されている(米国特許出願第2007/0161085参照)種々の濃度のアルギニンを用いてまたは用いずに試験した。結果を図9に示し、アルギニンはMVAの安定性に対して有益でない影響を持つことを明らかに示す。これに対して、37℃で28日後では、アルギニンの存在は、特に高濃度では、むしろ有害である。
トリス−HCl、グルタミン酸Na、スクロース、NaClを含有するpH8.0の処方物中、+37℃および+25℃でのMVA−MUC1の安定性に対するアミノ酸混合物の影響(表10参照)を、前記アミノ酸混合物を用いてまたは用いずに試験した。結果を図10に示し、アミノ酸混合物の存在はMVAの安定性に有意な効果を持たないことを明らかに示す。
トリス−HCl、グルタミン酸Na、スクロース、NaClを含有するpH7.5の処方物中、+25℃および+5℃でのMVA−HPVの安定性に対するヒスチジンの影響を、他のウイルスに対して安定化効果を持つことが示されている濃度(EVANS et al. J Pharm Sci. 2004 Oct, 93(10):2458-75、米国特許第7,456,009号、米国特許出願第2007/0161085号、米国特許第7,914,979号、WO2014/029702、WO2014/053571参照)である10mMのヒスチジンを用いてまたは用いずに試験した(表11参照)。結果を図11Aおよび11Bに示す。
上記の結果は、以下のことを明らかに示す:
・以下の化合物は液体処方物中でのワクシニアウイルスの安定性に対して有意な有益効果を持つ:
○一価の塩、例えば、NaCl。
○低パーセンテージの二糖、例えば、スクロース。このような成分は低温保護性があり、従って、約+5℃などの低い保存温度でワクシニアウイルスを保護すると考えられる。加えて、このような化合物は液体処方物の粘度を高め、これによりワクシニアウイルスと潜在的に有害な化合物の間の相互作用を制限し得る。
○EDTA、強い安定化効果を有する。
○エタノール、EDTAよりは小さいが有意な安定化効果を有する。
○EDTAとエタノールの組合せ、この組合せは、各化合物単独に比べてさらなる安定化効果を提供する。
○グルタミン酸Na。この化合物は安定性に不可欠ではなく、低濃度はMVAに対して小さな安定化効果を持ち得る。
○アミノ酸混合物。この化合物は安定性に不可欠ではなく、低濃度はMVAに対して小さな安定化効果を持ち得る。
○界面活性剤、例えば、ポリソルベート。低濃度では効果は見られない。しかしながら、少なくとも0.02%v/vの濃度では、ポリソルベート濃度とともに増大する脱安定化効果が見られる。
○ヒスチジン:10mMで、弱い脱安定化効果が見られる場合がある。
○MgCl2:0.5または1M、またはさらには75mMで(以下の実施例5を参照)、脱安定化効果が見られる。
○アルギニン:少なくとも30mMの濃度で弱い脱安定化効果が見られ、脱安定効果はアルギニン濃度とともに増大する。
安定な液体処方物のための好適なpHを決定するために、ワクシニアウイルスの安定性に対する液体処方物のpHの影響を試験した。
MVAウイルス
使用したMVAウイルスは、HCV遺伝子型1b株ja由来の非構造HCVタンパク質(NS3、NS4およびNS5B)を発現する組換えMVAウイルスであるMVA−HCV(TG4040)(WO2004/111082参照)であり、初期目標濃度4〜8 107 IG/mLに希釈した。
供試処方物を以下の表12に示す。
安定性の分析は実施例1に記載のとおりに行った。
感染力価の測定は実施例1に記載のとおりに行った。
トリス−HCl、グルタミン酸Na、スクロース、NaCl、EDTAおよびエタノールを含有する処方物中、+37℃および+5℃でのMVA−HCVの安定性に対するpHの影響を種々のpH値で試験した。結果を図12Aおよび12Bに示す。
上記の結果は、ワクシニアウイルス液体処方物のpHは好ましくは6を越え9未満の間に含まれるべきであることを示す。特に、pHが7〜8の間に含まれる場合に良好な結果が得られる。従って、6.5〜8.5の間に含まれるpHが許容可能である。
次に、液体処方物中でのその後の安定性に対するワクシニアウイルス初期力価の影響も試験した。
MVAウイルス
使用したMVAウイルスは、HCV遺伝子型1b株ja由来の非構造HCVタンパク質(NS3、NS4およびNS5B)を発現する組換えMVAウイルスであるMVA−HCV(TG4040)(WO2004/111082参照)であり、種々の初期目標濃度:1.0− 108PFU/mL、5.0 107PFU/mL、1.0 107PFU/mL、および5.0 106PFU/mLに希釈した。
供試処方物を以下の表13に示す:
安定性の分析は実施例1に記載のとおりに行った。
感染力価の測定は実施例1に記載のとおりに行った。
+37℃、+25℃および+5℃、種々の初期力価でのMVA−HCVの感染低下の評価をそれぞれ図13A、13Bおよび13Cに示す。
上記の結果から、MVAの液体処方物の保証安定性には少なくとも1.0 107PFU/mLの初期力価が極めて好ましいと思われる。
これまでの実施例で定義された最適化処方物の安定性を確認するために、このような最適化処方物を、2つの異なる実験で種々のワクシニアウイルス株に対して試験した。
ウイルス
以下のワクシニアウイルスを使用した:
・実験1:
○MUC1腫瘍関連抗原およびインターロイキン2を発現する組換えMVAウイルスであるMVA−MUC1(TG4010)(WO92/07000およびWO95/09241参照)、5〜8 108 IG/mLの初期目標濃度に希釈した。
○HCV 遺伝子型1b株ja由来の非構造HCVタンパク質(NS3、NS4およびNS5B)を発現する組換えMVAウイルスであるMVA−HCV(TG4040)(WO2004/111082参照)、4〜6 107 IG/mLの初期目標濃度に希釈した。
○ヒト乳頭腫ウイルスE6およびE7抗原ならびにインターロイキン2を発現する組換えMVAウイルスであるMVA−HPV(TG4001)(WO90/10459、WO95/09241、WO98/04705、WO99/03885、WO2007/121894参照)、2〜3 108 IG/mLの初期目標濃度に希釈した。
○以下:
・ニワトリ胚細胞(MVA−HCV/CEC)(初期目標力価2.5〜3 107 IG/mL)、または
・不死化カモ胚細胞株(MVA−HCV/カモ細胞株)(初期目標力価3〜4 107 IG/mL)
で生産した、HCV遺伝子型1b株ja由来の非構造HCVタンパク質(NS3、NS4およびNS5B)を発現する組換えMVAウイルスであるMVA−HCV(TG4040)(WO2004/111082参照)、
○ニワトリ胚細胞(MVA−FCU1/CEC)で生産された、シトシンデアミナーゼ活性1を有する融合タンパク質を発現する組換えMVAウイルスであるMVA−FCU1(TG4023)(WO99/54481参照)(初期目標力価1〜1.5 108 IG/mL)、
○ニワトリ胚細胞で生産された、シトシンデアミナーゼ活性を有するFCU1融合タンパク質を発現し、かつ、欠陥型I4L遺伝子および欠陥型J2R遺伝子を含んでなる組換えワクシニアウイルスコペンハーゲン株であるコペンハーゲン−FCU1(TG6002)(WO2009/065546およびWO2009/065547参照)(コペンハーゲン−FCU1/CEC)(初期目標力価2〜3.0 108 IG/mL)。
○ヒト連続細胞株で生産され、少なくとも1種類のプロテアーゼで処理する少なくとも1つの工程を含む方法により精製されたワイス株の組換えワクシニアウイルス(VVワイス)も初期目標力価5〜8 108PFU/mLで使用した。
実験1:
MVAウイルスに対する供試処方物を以下の表14に示す:
MVA−HCV/CEC、MVA−HCV/カモ細胞株、MVA−FCU1/CEC、およびコペンハーゲン−FCU1/CECウイルスに対する供試処方物を以下の表15Aに示す:

実験1:
+37℃、+25℃および+5℃、本発明の最適化処方物中での3種類のMVAウイルスの安定性をそれぞれ図14A、14Bおよび14Cに示す。
この第2の実験では、MVAに対してこれまでに指摘かされた同じ処方物を、種々の方法により得た数種のMVAウイルス、および別のワクシニアウイルス株:コペンハーゲン(ベクターTG6002)に対して試験した。
この第3の実験では、MVAに対して従前に最適化された同じ処方物を、ヒト連続細胞株で生産され、少なくとも1種類のプロテアーゼで処理する少なくとも1つの工程を含む方法により精製された別のワクシニアウイルス株(ワイス株)に対して試験した。
上記の結果は、本発明者らにより設計された最適化処方物が種々のワクシニアウイルス株に適用可能であること、およびこのようなウイルスに挿入され得る異種構築物は本発明による最適化処方物より提供される安定化に有意な影響を及ぼさないことを明らかに実証する。
これまでに試験した個々の化合物が同じ系列または他の系列の他の化合物に置き換え可能であるかどうかを確認するために、従前に供試した最適化処方物の個々の化合物のそれぞれを同じ系列または他の系列の化合物に置き換える実験を行った:
・バッファー:従前に供試したトリス−HClバッファーを、同じ20mM濃度のトリシンまたはHEPESバッファーに置き換えた。両置換バッファーもpH7〜pH8の間で緩衝能を有する。
・塩:従前に供試した一価の塩NaCl、を同じ75mM濃度の別の一価の塩(KCl)または二価の塩(MgCl2)のいずれかに置き換えた。
・二糖または糖アルコール:従前に供試した二糖スクロースを、同じ10%w/v濃度の別の二糖(トレハロース)または糖アルコール(マンニトール)のいずれかに置き換えた。
・キレート剤:従前に供試したキレート剤EDTAを、同じ150μM濃度の2種類の他のキレート剤EGTAまたはDTPAに置き換えた。
・C2−C3アルコール:従前に供試したC2−C3アルコールエタノールを、同じ0.5%濃度のイソプロパノールに置き換えた。
ウイルス
使用したMVAウイルスは、MUC1腫瘍関連抗原およびインターロイキン2を発現する組換えMVAウイルスであるMVA−MUC1(TG4010)(WO92/07000およびWO95/09241参照)であり、8.0 107〜2 108IG/mLの間の初期目標力価に希釈した。
供試処方物を以下の表16に示す:
安定性の分析は実施例1に記載のとおりに行った。
感染力価の測定は実施例1に記載のとおりに行った。
+37℃での種々の供試処方物の感染低下を図16Aに示す。
・バッファー:21日目では、トリス−HClをトリシンまたはHEPESバッファーに置き換えても、MVAウイルスの安定性に有意に影響を及ぼさないが、トリス−HClはMVA−MUC1の安定化にやや良好であると思われる。28日目では、トリス−HClをトリシンに置き換えてもMVAウイルスの安定性に有意に影響を及ぼさないが、トリス−HClの方がやや良好であると思われ、HEPESバッファーはトリス−HClおよびトリシンより効果が低い。
・塩:NaClをKClに置き換えても、21日後または28日後にMVAウイルスの安定性に有意に影響を及ぼさない。これに対し、NaClまたはKClをMgCl2に置き換えることは、21日目と28日目の両方でMVAの安定性に有意に有害な効果を持ち、MgCl2は75mMといった低濃度で有害であり得ることがさらに確認される。
・二糖または糖アルコール:スクロースをマンニトールに置き換えても、MVAウイルスの安定性に有意に影響を及ぼさない。スクロースをトレハロースに置き換えると、MVAの安定性の改善に向かう小さな効果が21日目および28日目に認められる。
・キレート剤:EDTAをDTPAまたはEGTAに置き換えても、21日目および28日目にMVAウイルスの安定性に有意に影響を及ぼさない。
・C2−C3アルコール:エタノール(EtOH)をイソプロパノールに置き換えても、21日目および28日目にMVAウイルスの安定性に有意に影響を及ぼさない。
上記の結果は、本発明による最適化処方物では、最初に試験した化合物は、ワクシニアウイルスの安定性を有意に変更することなく同じ系列の他の化合物に置換され得る(トリス−HClをpH7〜8の間で緩衝能を有する別のバッファーに、NaClを別の一価の塩に、スクロースを別の二糖または糖アルコールに、EDTAを別のキレート剤に、EtOHを別のC2−C3アルコールに)ことを明らかに証明する。
本発明による安定化液体MVA処方物が、12か月保存後に得られたばかりのMVA処方物と同等のin vivo免疫原性を持っていたかどうかをさらに確認した。
MVAウイルス
使用したMVAウイルスは、MUC1腫瘍関連抗原およびインターロイキン2を発現する組換えMVAウイルスであるMVA−MUC1(TG4010)(WO92/07000およびWO95/09241参照)であり、1〜3 108PFU/mLの初期目標力価に希釈した。
MVA−MUC1を、トリス−HCl 20mM、スクロース10%w/v、NaCl 75mM、EDTA 150μM、EtOH 0;5%v/v、およびグルタミン酸Na 5mMを含有する液体処方物中に処方した。
使用したモデルは、MUC1抗原を発現する腫瘍細胞をさらに投与する前にMVA−MUC1ベクターがマウスに注入されるという、予防モデルである。さらなる詳細を以下に示す:
第1の実験:
・マウス:C57BL/6マウスを使用した。各群は20個体の動物から構成された:
○群1:処方物単独、
○群2〜4:MVA−MUC1 T=0(104、105、または106PFU)、
○群5〜7:MVA−MUC1 T=12か月(104、105、または106PFU)。
・試験する生成物の投与:各条件につき(T=0/T=12か月)、各生成物を1週間間隔で3回(D0/D7/D14)皮下注射した。各生成物につき、3種類の用量(104、105、または106PFU)を試験した。加えて、陰性対照として、処方物単独(MVA−MUC1ウイルス不含)を同じプロトコールで投与した。
・腫瘍細胞の投与:ウイルス/処方物の最後の注射から1週間後、すなわち、21日目(D21)に、RMA−MUC1細胞を皮下注射した。
・マウスの生存、腫瘍の存在および大きさを107日目(D107)までまたは腫瘍体積が2000mm3となるまでモニタリングし、この時点でマウスを犠牲にした)。
T=0およびT=24か月、104PFUの用量に関して、処方物単独または処方物中のMVA−MUC1の免疫原性を比較するために同じモデルを使用した。
第1の実験:
以下の表17Aは生成物(0または12か月保存後の処方物単独またはMVA−MUC1)および注射用量に応じた、86日目に腫瘍不含であったマウスのパーセンテージを表す。
・供試した液体処方物は、そのままでは、RMA−MUC1腫瘍細胞からマウスを保護する免疫応答を誘導しない、および
・供試処方物中、MVA−MUC1の+5℃±3℃で12か月の保存は、RMA−MUC1腫瘍細胞からマウスを保護する免疫応答を誘導するMVA−MUC1の能力に有意に影響を及ぼさない。
以下の表17Bは、生成物(0または24か月保存後の処方物単独またはMVA−MUC1)に応じた、65日目に腫瘍不含であったマウスのパーセンテージを表す。
・供試した液体処方物は、そのままでは、RMA−MUC1腫瘍細胞投与からマウスを保護する免疫応答を誘導しない、および
・供試処方物中、MVA−MUC1の+5℃±3℃で24か月の保存は、RMA−MUC1腫瘍細胞投与からマウスを保護する免疫応答を誘導するMVA−MUC1の能力に有意に影響を及ぼさない。
上記のデータから、本発明による最適化処方物は2年の保存中に感染性ウイルス力価を維持するだけでなく、in vivoで防御免疫応答を誘導する能力も維持することが確認される。
ワクシニアウイルスは、UV損傷を受けやすいことが知られている(LYTLE et al. J. Virol. 2005, 79(22):14244参照)。UV損傷からワクシニアウイルスを保護する種々の処方物の能力を2種類の独立した実験で調べた。
MVAウイルス
使用したMVAウイルスは、HCV遺伝子型1b株ja由来の非構造HCVタンパク質(NS3、NS4およびNS5B)を発現する組換えMVAウイルスであるMVA−HCV(TG4040)(WO2004/111082参照)であり、実験1では5 107〜7 107IG/mLの間、実験2では3 108〜5 108IG/mLの間の初期目標力価に希釈した。
実験1のMVA−HCVに対する供試処方物を以下の表18に示す:
サンプルを以下の光条件で保存した:
・光無し、
・室温、PSM下(可視出力と紫外(UV)出力を組み合わせた人工日光蛍光灯、キセノン灯、またはメタルハリド灯などの、D65/ID65放射基準と同様の出力を作り出すように設計された任意の光源。D65は、ISO 10977(1993)に定義される屋外日光として国際的に認知されている基準である。ID65は、等価な屋内間接的日光基準である。320nmより下方域の有意な放射線を発する光源としては、そのような放射線を発するように適当なフィルターを取り付ければよい)、または
・ICH光下(320nm〜400nmのスペクトル分布を有し、最大エネルギー放射が350nm〜370nmの間である近UV蛍光灯;有意な割合のUVが320〜360nmおよび360〜400nmの両バンド内になければならない。ICH Q1B Photostability Testing of New Drug Substances and Products参照)。
安定性は28日間、+25℃(±2℃)で分析した。
感染力価の測定は実施例1に記載のとおりに行った。
実験1および2に関する結果をそれぞれ図17Aおよび17Bに示す。
上記の結果は、本発明による処方物は光の不在下でワクシニアウイルスを安定化させるだけでなく、さらにUV損傷による分解からワクシニアウイルスを保護することを明らかに示す。これは液体処方物中でのワクシニアウイルスの保存時の制約の限定に非常に役立ち得る。
本発明による処方物の安定化効果のロバストネスを調べるために、種々の濃度の異なる成分を含有する数種の処方物を、MVAベクターを安定化させるそれらの能力に関して試験した。
ウイルス
使用したMVAウイルスは、MUC1腫瘍関連抗原およびインターロイキン2を発現する組換えMVAウイルスであるMVA−MUC1(TG4010)(WO92/07000およびWO95/09241参照)であり、1〜4 108PFU/mLの初期目標力価に希釈した。
供試処方物を以下の表20に示す:
安定性の分析は実施例1に記載のとおりに行った。
感染力価の測定は実施例1に記載のとおりに行った。
+37℃での種々の供試処方物の感染低下を図18に示す。
上記のデータから、最適化本発明による処方物は、処方物中に含まれる成分の一定範囲の濃度にわたってロバストな安定化効果を有することが確認される。
材料および方法
ウイルス
非組換え偽牛痘ウイルス(パラポックスウイルス科)を1〜3 108PFU/mLの初期目標力価で使用した。
供試処方物を以下の表21に示す:
安定性の分析は37℃で実施例1に記載のとおりに行った。
感染力価の測定は実施例1に記載のとおりに行った。
結果を図19に示し、本発明による供試処方物は偽牛痘ウイルスを大幅に安定化させることを示す。特に、トリスおよびスクロース単独中の対照偽牛痘ウイルスでは、37℃で28日後の感染低下が2.5log10を越え、処方の偽牛痘ウイルスでは、37℃で28日後の感染低下がわずか約0.5log10である。
上記の結果は、本発明による処方物がパラポックスウイルス科のポックスウイルスである偽牛痘ウイルスを安定化させるためにも好適であることを明らかに示す。
Claims (18)
- a)10 7 〜10 12 PFU/mLの力価のポックスウイルス、
b)10〜50mMの濃度で存在する、トリス−HCl、トリスおよびトリシンから選択される薬学上許容可能なバッファー、
c)10〜1000mMの濃度で存在する、NaClおよびKClから選択される一価の塩、
d)5〜20%(w/v)の濃度で存在する、スクロース、マンニトールおよびトレハロースから選択される薬学上許容可能な二糖または糖アルコール、および
e)50〜250μMの濃度で存在する、エチレンジアミン四酢酸(EDTA)、ジエチレントリアミン五酢酸(DTPA)およびエチレングリコール四酢酸(EGTA)から選択される薬学上許容可能なキレート剤
を含んでなり、そのpHが7〜8である、液体処方物。 - a)界面活性剤を含まないか、または0.005%(w/v)未満の濃度の界面活性剤を含む、

- 前記ポックスウイルスがワクシニアウイルスである、請求項14に記載の液体処方物。
- 前記ワクシニアウイルスが、改変ワクシニアウイルスアンカラ(MVA)株、ワイス株、およびコペンハーゲン株から選択される、請求項15に記載の液体処方物。
- 前記ワクシニアウイルスが、
・直接的または間接的細胞傷害機能を有する分子をコードする外因性配列、
・腫瘍関連抗原(TAA)をコードする外因性配列、
・ウイルス抗原をコードする外因性配列、ならびに
・細菌抗原をコードする外因性配列
から選択される少なくとも1つの外因性配列を含んでなる組換えワクシニアウイルスである、請求項1〜16のいずれか一項に記載の液体処方物。 - 前記ワクシニアウイルスが、
・シトシンデアミナーゼ(CDase)遺伝子、
・MUC1およびインターロイキン2(IL−2)をコードする外因性配列、
・ヒト乳頭腫ウイルス抗原、B型肝炎抗原、またはC型肝炎抗原をコードする外因性配列、ならびに
・マイコバクテリア抗原をコードする外因性配列
から選択される少なくとも1つの外因性配列を含んでなる組換えワクシニアウイルスである、請求項17に記載の液体処方物。
