Description
Claims (4)
- 核酸の混合物を含有する試料から標的核酸を固定化及び増幅する方法であって、
そこに固定された第一オリゴヌクレオチドおよび第二オリゴヌクレオチドを有する固体支持体に前記試料を適用する工程であって、前記第一オリゴヌクレオチドは、3'−末端からの伸長を防ぐためにブロックされており、前記標的核酸の第一部分に相補的な配列を有し、前記第二オリゴヌクレオチドは、前記標的核酸の第二部分と同一の配列を有し、前記の標的核酸が第一オリゴヌクレオチドに結合し、そして必要に応じて結合していない核酸を除去するために前記固体支持体を洗浄する工程;
標的結合領域およびプロモーター配列を有する第一プライマーを前記標的核酸にハイブリダイズし、第一二本鎖核酸を生成するためにポリメラーゼにより前記第一プライマーを伸長し、そして必要に応じて前記標的核酸を前記二本鎖核酸から取り除き第一核酸を生成する工程;
前記第一核酸を前記第二オリゴヌクレオチドにアニーリングし、前記第二オリゴヌクレオチドをポリメラーゼで伸長し、第一核酸と第二核酸を含む第二二本鎖核酸を生成する工程;ならびに
プロモーター依存ポリメラーゼにより第三核酸の複数の複製物を生成し、それにより前記標的核酸を増幅する工程
を含む方法。 - 前記増幅された第三核酸を前記第二オリゴヌクレオチドにアニーリングし、前記第二オリゴヌクレオチドを伸長して第三二本鎖核酸を生成する工程;
必要に応じて、前記増幅された第三核酸を取り除き、固体支持体に結合された第四核酸を生成する工程;
前記プライマー配列を前記第四核酸にアニーリングし、前記プライマーおよび前記第四核酸をポリメラーゼにより伸長し、第五核酸と追加の第二核酸を含む第四二本鎖核酸を生成する工程;および
ポリメラーゼにより前記第三核酸の複数のコピーを生成する工程であって、前記ポリメラーゼが前記第四二本鎖核酸上のプロモーターに特異的であり、それにより前記標的核酸を増幅する工程
をさらに含む請求項1記載の方法。 - 前記プライマーが、1つまたはそれ以上の非天然ヌクレオチドを含むタグ配列をさらに含む請求項1記載の方法。
- 前記第二オリゴヌクレオチドが、3'−末端に前記第一核酸の4つの異なるSNPに特異的な別々のヌクレオチドをそれぞれ有する4つのオリゴヌクレオチドである請求項1記載の方法。
