本発明は、核酸がファージディスプレイのために原核細胞に形質転換された場合には一のFab融合タンパク質、核酸が発現及び精製のために真核細胞にトランスフェクトされた場合には別個又は同一のFab融合タンパク質の発現及び分泌のための、発現及び分泌系及びその使用方法に関する。また、本明細書で提供されるのは、核酸分子、ベクター及びそのようなベクター及び核酸分子を含む宿主細胞である。
繊維状ファージ粒子上のペプチド又はタンパク質のファージディスプレイは、変異体ペプチド又はタンパク質の大きなプールから所望の特性を有するペプチド又はタンパク質の選択を可能にするインビトロの技術である(McCafferty et al., Nature, 348: 552-554 (1990); Sidhu et al., Current Opinion in Biotechnology, 11: 610-616 (2000); Smith et al., Science, 228: 1315-1317 (1985))。ファージディスプレイは、抗体の検出の場において、Fabのような抗体断片を含む、ペプチド又はタンパク質の多様なライブラリーを、後に目的の特定の抗原への結合のために選択される、糸状ファージ粒子の表面上に提示するために使用することができる。抗体断片は、抗体断片の遺伝子をファージコートタンパク質の遺伝子に対して融合することによって、繊維状ファージ粒子の表面上に提示することができ、コードされた抗体断片を表面上に表示するファージ粒子を生じる。この技術は、大きなファージライブラリーから、多数の抗原に対して所望の親和性を有する抗体断片の単離を可能にする。
一態様において、本発明は、(1)原核細胞における非原核生物起源の機能のシグナル配列及び(2)異なったFab融合タンパク質は、mRNAプロセシングが原核細胞でなく真核細胞において起きる場合に、宿主細胞依存的に同一の核酸分子から発現される(原核細胞中のFabファージ融合タンパク質及び真核細胞中のFab−Fc融合タンパク質)ことを実証する実験的発見に一部基づいている。従って、本明細書中に記載されるのは、サブクローニングを必要とせずに、核酸が、原核生物宿主細胞(例えば大腸菌)に形質転換された場合に、細菌におけるファージディスプレイのための、ファージ粒子タンパク質、コートタンパク質又はアダプタータンパク質に融合したFab断片、及び核酸が真核細胞(例えば、哺乳動物細胞)に形質転換された場合に、Fcに融合したFab断片の発現及び分泌のための核酸分子、及びその使用の方法である。

I.定義

ファージに基づく抗体の検出プロセスは、個々のファージクローン1−3の大規模なプールから所望の結合特異性を有するFab断片を選択するファージディスプレイ技術を利用する。このアプローチでは、直接的又は間接的に主要コートタンパク質の一つ介してM13繊維状ファージ粒子に融合し、多様な相補性決定領域(CDR)を含むFab断片からなるファージライブラリーは、確立された分子生物学技術及び特殊なファージディスプレイベクターを使用して生成される(Tohidkia et al., Journal of drug targeting, 20: 195-208 (2012); Bradbury et al., Nature biotechnology, 29: 245-254 (2011); Qi et al., Journal of molecular biology, 417: 129-143 (2012))。そうしたライブラリーの理論的多様性は、1025のユニークな配列を容易に超過ことができるが、ファージプールの構築における実際的な限界は、実際の多様性を、特定のライブラリにおいて≦1011のクローンへ制限する(Sidhu et al., Methods in enzymology, 328: 333-363 (2000))。
本発明の実施は、特に明記しない限り、当分野の技術の範囲内である、分子生物学(組換え技術を含む)、微生物学、細胞生物学、生化学及び免疫学の従来の技術を使用する。このような技術は、「分子クローニング:実験室マニュアル(Molecular Cloning: A Laboratory Manual)」,第2版(Sambrook et al., 1989);「オリゴヌクレオチド合成(Oligonucleotide Synthesis)」(M.J. 原核細胞及び真核細胞のための発現及び分泌系は、目的のタンパク質(例えば、IgG分子の重鎖又は軽鎖)の調節配列及びコード化配列を含むベクターを含み、ここで原核生物プロモーター及び真核生物プロモーター(例えば、CMV(真核生物)及びPhoA(原核生物))は、目的の遺伝子の上流にタンデムに配置され、単一のシグナル配列が、原核細胞及び真核細胞における目的のタンパク質の発現を駆動する。本発明は、宿主細胞依存的に、IgG1のVH/CH1とヒンジ−Fc領域との間に位置する合成イントロンであって、真核細胞におけるmRNAプロセシング中にスプライスされる合成イントロンを使用することにより、目的のタンパク質の2つの異なる融合形態を生成するこのベクターのための手段を提供する。
原核生物(大腸菌)細胞及び真核生物(哺乳動物)細胞で目的のタンパク質を発現することが可能なベクターの構築における一つの問題は、これらの細胞型に見い出されるシグナル配列の違いから生じる。シグナル配列の特定の機能は、一般的に原核細胞及び真核細胞の両方で保存されているが(例えば配列の中央に位置する疎水性残基のパッチ及び成熟ポリペプチドのN末端における切断部位に隣接する極性/荷電残基)、他の機能は、他方よりも一細胞型でより特徴的なものである。また、配列が全て哺乳動物起源であっても、別のシグナル配列は、哺乳動物細胞における発現レベルに大きな影響を与えることができることが当技術分野で知られている(Hall et al., J of Biological Chemistry, 265: 19996-19999 (1990); Humphreys et al., Protein Expression and Purification, 20: 252-264 (2000))。例えば、細菌のシグナル配列は、典型的には正に帯電した残基(最も一般的にリジン)を開始メチオニンの直後に有するが、一方哺乳動物のシグナル配列には必ずしも存在しない。両方の細胞型において分泌を指示することができるシグナル配列が知られているが、そのようなシグナル配列は、典型的には、一方の細胞型又は他方においてのみ、タンパク質分泌の高レベルを指向する。
本発明は、異なるFab融合タンパク質は、原核細胞でなく真核細胞においてmRNAのプロセシング中に起きるイントロンスプライシングの天然のプロセスを利用することにより、宿主細胞依存的に同一の核酸分子から発現され得るという発見に一部基づいている。
他の例としては、スプライスドナーの8つの位置のうち、位置1と5で置換を有する変異体スプライスドナーを含む。
原核細胞及び真核細胞におけるFab融合タンパク質の発現及び分泌のための発現及び分泌系は、当該分野の技術範囲内である様々な技術を用いて構築することができる。
本発明は、abベースのライブラリのファージ又は細菌ディスプレイによって目的のタンパク質に対する抗体をスクリーニングし選択する方法、又は同様の方法により、既存の抗体を最適化する方法を提供する。上述の二重ベクターの使用は、原核細胞におけるFab断片のスクリーニング及び選択、及びサブクローニングを必要としない更なる試験用に、完全長IgG分子として容易に発現することができるFabの選択のために使用することができる。
本発明の更なる態様では、上記実施態様のいずれかに記載の抗体は、キメラ、ヒト化又はヒト抗体を含むモノクローナル抗体である。一実施態様において、抗体は、抗体断片、例えば、Fv、Fab、Fab’、scFv、ダイアボディ、又はF(ab’)2断片である。その他の態様において、抗体は、全長抗体、例えば、インタクトなIgG1、IgG2、IgG3又はIgG4抗体、又は本明細書において定義された他の抗体クラス又はアイソタイプである。

ある実施態様において、本明細書で提供される抗体は、多重特異性抗体、例えば二重特異性抗体である。多重特異性抗体は、少なくとも二つの異なる部位に対して結合特異性を有するモノクローナル抗体である。ある実施態様において、結合特異性の一つは第一抗原に対してであり、他は、任意の他の抗原に対してである。所定の実施態様において、二重特異性抗体は、第一抗原の2つの異なるエピトープに結合することができる。二重特異性抗体はまた第一抗原を発現する細胞に細胞傷害性薬物を局所化するために用いることができる。二重特異性抗体は、全長抗体又は抗体断片として調製することができる。
ある実施態様において、本明細書で提供される抗体のアミノ酸配列変異体が企図される。例えば、抗体の結合親和性及び/又は他の生物学的特性を改善することが望まれ得る。抗体のアミノ酸配列変異体は、抗体をコードするヌクレオチド配列に適切な改変を導入することにより、又はペプチド合成によって調製することができる。このような改変は、例えば、抗体のアミノ酸配列内における、残基の欠失、及び/又は挿入及び/又は置換を含む。最終コンストラクトが所望の特性、例えば、抗原結合を有していることを条件として、欠失、挿入、及び置換の任意の組み合わせが、最終コンストラクトに到達させるために作成され得る。

ある実施態様において、1つ又は複数のアミノ酸改変を、本明細書で提供される抗体のFc領域に導入することができ、それによってFc領域変異体を生成する。Fc領域の変異体は、1つ又は複数のアミノ酸位置においてアミノ酸修飾(例えば置換)を含むヒトFc領域の配列(例えば、ヒトIgG1、IgG2、IgG3又はIgG4のFc領域)を含んでもよい。

一態様において、本発明の抗体は、例えばELISA、ウエスタンブロット法など公知の方法により、その抗原結合活性について試験される。
一態様において、アッセイは、生物学的活性を有するその抗体を同定するために提供される。インビボ及び/又はインビトロでこのような生物学的活性を有する抗体もまた提供される。
ベクターが、大腸菌及び真核(哺乳動物)細胞の両方において目的のタンパク質を発現することができるかどうかを検討するために、哺乳動物細胞中で機能することができるとする考えを支持する事例証拠が存在する非細菌起源の4つのシグナル配列が選択された。我々は、これらのシグナル配列を、ファージELISAを使用してM13ファージ上で抗Her2(h4D5)Fabディスプレイを駆動するその能力について試験した(図1A)。ディスプレイのレベルは、細菌の熱安定性エンテロトキシンII(STII)のシグナル配列と比較して評価した。Fabファージディスプレイを駆動するシグナル配列の能力は大きく変化し、マウス結合性免疫グロブリンタンパク質(mBiP)からの、一つのシグナル配列が、Fabディスプレイの効率的なレベルを恐らくは可能としうるディスプレイのレベルを駆動した。
宿主細胞依存的に異なるFab融合タンパク質を生成するために、我々は、原核細胞でなく真核生物においてmRNAのプロセシング中に生じるイントロンスプライシングの天然のプロセスを活用しようとした。hIgG1のHC定常領域のゲノム配列は、3つの天然イントロンが含まれる(図3A)。これらのうち最初(イントロン1)は、HC可変ドメイン(VH)及びヒンジ領域との間に位置する384塩基対のイントロンである。RT−PCR及び転写産物の配列決定によって評価されるように、イントロン1及び最適化されたスプライスドナー配列を含むHC発現ベクターは、完全にスプライシングされたmRNAを発現し、LCベクターを同時導入されると、イントロンを含まないベクターに匹敵するレベルでIgG1を発現した(図5)。
二重ベクタープラスミドの生成のために、我々はファージディスプレイに現在使用したpBR322由来のファージミドベクター、pRSを使用した。このバイシストロン性ベクターは、その関連するSTIIシグナル配列を有する抗体の軽鎖カセット、続いて関連するSTIIシグナル配列を有する抗体の重鎖カセットの発現を駆動する細菌のPhoAプロモーターからなる。軽鎖配列の最後に、ファージ粒子上のFabディスプレイを検出するためのgDエピトープタグがある。従来のファージミドでは、重鎖配列は、hIgGのVH及びCH1ドメインのみからなり、ファージ融合タンパク質、ほとんどの場合ファージコートタンパク質pIII又はアダプターペプチドをコードする、遺伝子IIIなどのユーティリティペプチドに対してヌクレオチドレベルで融合される。軽鎖及び重鎖カセットの3’末端は、大腸菌において転写を停止するためのラムダ転写ターミネーター配列を含む。このベクターは、単一のmRNA転写産物からの軽鎖及び重鎖のpIIIを生成するので、LC配列及びHC配列間に転写調節エレメントは存在しない。ベクターは、アンピシリン耐性を付与するβ−ラクタマーゼ(BLAの)遺伝子、大腸菌における複製のためのpMB1起点、及びM13ファージによる感染を可能にする細菌表面上での線毛の発現のためのf1起点を含む。このベクターの別の形態はまた、哺乳動物細胞の適切な菌株におけるプラスミドのエピソーム複製のためのSV40複製起点を含む。
M13ファージ上のpIIIに融合したタンパク質のディスプレイを増強するため、我々は部位特異的突然変異誘発を使用して、変異体ヘルパーファージ、アンバーKO7を生成させた。アンバーKO7は部位特異的変異誘発によってM13KO7ヘルパーファージゲノムに導入されたアンバーコドンを有する。pIIIのヌクレオチド配列(変異体ヘルパーファージアンバーKO7のヌクレオチド1579から2853)が図8に示されている。
実施例3に記載のHC及びLCの両方で、原核生物プロモーター及び真核生物プロモーターを特徴づける直接的及び間接的な融合ベクターに加えて(pDV5.0とpDV6.0)、我々は、改変された直接融合二重ベクターコンストラクトを生成し(図14に示すpDV.6.5)、ここではFab LCはSTIIシグナル配列に融合され、細菌のPhoAプロモーターによってのみ駆動されるが、一方Fab HC(哺乳動物細胞における完全長hIgG1 HCの発現のための遺伝子III−合成イントロン配列及びhIgG Fc配列を含有する)は、真核生物のCMVプロモーター、及び原核生物のPhoAプロモーターの両方によって駆動された。このコンストラクトは、多様性がHCのみに導入される、前述の合成ヒトFabライブラリーを再現するために使用された(Lee, et al., Journal of Molecular Biology, 340. 1073-1093 (2004))。このベクターからの完全長IgGの発現は、LCをコードする哺乳動物発現ベクターの同時導入を必要とする。
