Description
R1は、水素原子または任意に置換されたアルキルもしくはシクロアルキル基を表し、
R2は、それぞれ同一または異なっていてもよく、水素原子または任意に置換されたアルキル基を表し、

実施例1−カルボキシメチル5−アミノ−4−オキソペンタノエートヒドロハライド(塩酸塩および臭化水素酸塩)の調製

2bの生成物(4.92g;11.5mmol)、10%のパラジウム坦持活性炭(100mg)、水素ガス、および2−プロパノール(15mL)およびジオキサン(15mL)中のジエチルエーテル(10mL;20mmol)中2.0Mの塩化水素の攪拌した混合物を、周囲温度で2日間、約6バールで水素添加した。混合物を、Celite(登録商標)545パッドを通して濾過し、2−プロパノール(2×5mL)で残渣を洗浄した。合わせた濾液を蒸発させ、得られた油状物をジエチルエーテル(4×10mL)で粉砕し、次いで、冷凍庫で保管した。油状物は固化しなかった。一晩、減圧乾燥(0.005mm Hgで)させた後、1.97g(71%)の生成物を得た(黄褐色、粘稠な油状物)。

この化合物は、実施例2aに従い、ブロモジフルオロ酢酸(9.82g;68.7mmol)、ベンジルアルコール(7.0g;65mmol)、およびシクロヘキサン(120mL)中のp−トルエンスルホン酸一水和物(50mg)から調製された。蒸発させた後に得た粗生成物は、3bでさらに処理することなくそのまま使用された。

この化合物は、実施例2cに記載されるように、3bの生成物(4.0g;8.9mmol)、10%のパラジウム坦持炭素(0.20g)、水素ガス、および2−プロパノール(75mL)およびテトラヒドロフラン(25mL)中のジエチルエーテル(10mL;20mmol)中2MのHClから生成された。Celite(登録商標)を通して濾過した後、ひだ付き濾紙を通して溶液を再濾過した。蒸発後、淡琥珀色の油状物を得て、これは、ジエチルエーテル(4×10mL)で粉砕され、1.6g(80%)の生成物が得られた(淡黄褐色のゴム状固体)。得られた生成物は、主にカルボキシジフルオロメチル5−アミノ−4−オキソペンタノエート塩酸塩、多少のイソプロピル5−アミノ−4−オキソペンタノエート塩酸塩、ならびに多少の1−(イソプロピルカルボキシ)ジフルオロメチル5−アミノ−4−オキソペンタノエート塩酸塩を含有した混合物であった。カルボキシジフルオロメチル5−アミノ−4−オキソペンタノエート塩酸塩は、標準的な方法、例えば、分取HPLCにより、混合物から単離され得る。

シリンジを用いて、アルゴン下で、周囲温度のN,N’−ジシクロヘキシルカルボジイミド(DCC,3.34g;16.2mmol)の溶液を、ジクロロメタン(40mL)中の5−(Boc−アミノ)−4−オキソペンタン酸(3.70g;16.0mmol)、t−ブチル3−ヒドロキシプロパノエート(2.31g;15.8mmol)、および乾燥ジクロロメタン(20mL)中の4−ジメチルアミノピリジン(DMAP,50mg)の攪拌した混合物に添加した。適度の発熱反応は、氷水浴で抑えられた。周囲温度で2日間攪拌した後、混合物をガラス−焼結フィルタで吸引濾過した。残渣をジクロロメタン(3×20mL)で洗浄した。合わせた濾液を蒸発させ、残渣を、酢酸エチル−ヘキサン(1:1)(500mL)で溶出した45×55mmシリカゲル60カラムフラッシュクロマトグラフィーにより精製し、7×50mlの画分を収集した。画分2〜4を蒸発させた後、5.45g(96%)の生成物を得た(淡黄色の油状物)。

この化合物は、4aの生成物(5.43g;15.1mmol)および実施例1cによる酢酸(15mL)中30%のHBrから調製された。3.38g(79%)の生成物を得た(固化しなかった琥珀色のゴム)。

この化合物は、実施例1aに記載される、ベンジル4−ブロモブタノエート(7.2g;28.0mmol)、5−(Boc−アミノ)−4−オキソペンタノエート(6.7g;29.0mmol)、トリエチルアミン(2.95mmol)、および酢酸エチル(100mL)から調製された。粗生成物を、酢酸エチル−ヘキサン(1:2)(750mL)で溶出した60×55mmシリカゲル60カラムフラッシュクロマトグラフィーにより精製し、6×100mLの画分を収集した。画分3および4を蒸発させた後、5.43g(46%)の生成物を得た(黄色の油状物)。

この化合物は、5aからの生成物(5.40g;13.2mmol)、実施例2cに記載される、10%のパラジウム坦持活性炭(250mg)、水素ガス、および96%のエタノール(100mL)から調製された。4.30gの生成物を得(帯黄色の油状物)、これは、さらに処理することなく5cで使用された。
この化合物は、5bの生成物(4.20g;13.2mmol)、実施例1bに記載される、ジエチルエーテル(7.5mL;15mmol)中2MのHCl、およびジエチルエーテル(50mL)から調製された。溶媒を蒸発させた後、油状の残渣を、ジエチルエーテル(4×10mL)で粉砕し、淡黄褐色の固体に固化するまで冷凍庫で一晩維持した。12mm Hgおよび周囲温度で乾燥させた後、1.90g(57%)の生成物を得た。

濃縮した硫酸(0.55mL;10mmol)を、周囲温度の乾燥ジクロロメタン(40mL)中の無水MgSO4(4.8g;40mmol)の攪拌懸濁液に添加した。15分間攪拌した後、α−ブロモフェニル酢酸(2.15g;10.0mmol)、続いてt−ブチルアルコール(4.8mL;50mmol)を添加した。栓で反応フラスコを閉じ、混合物を周囲温度で7日間攪拌した。飽和NaHCO3溶液(75mL)および水(75mL)を反応混合物に添加した。分離した後、水層をジクロロメタン(2×15mL)で抽出した。混合有機層を水で洗浄し、乾燥させた(MgSO4)。濾過および蒸発後、2.39g(88%)の生成物を得た(無色の油状物)。

実施例8〜15の化合物の調製に使用された一般手順:
手順A カルボジイミドを用いたベンジルエステルの調製
シリンジを介して、N,N’−ジシクロヘキシルカルボジイミド(DCC)または乾燥ジクロロメタン(10mL)中のN,N’−ジイソプロピルカルボジイミドの溶液を、アルゴン下で、カルボン酸、ベンジルアルコール、または2,2,2−トリクロロエタノール、および乾燥ジクロロメタン(40mL)中の4−ピロリジニルピリジンの攪拌溶液に滴下して添加した。周囲温度で1日または2日間攪拌した後、混合物を減圧濾過し、残渣を少量のジクロロメタンで洗浄した。溶媒を蒸発させ、クーゲルロール装置を用いて残渣を50℃(浴温)および0.014mm Hgで減圧乾燥させ、過剰なベンジルアルコールを除去した。
カルボン酸、ベンジルアルコール、およびトルエン中のp−トルエンスルホン酸の混合物を、ディーンスタークトラップを通して一晩還流させた。周囲温度に冷却した後、混合物を、飽和NaHCO3溶液(1×15mL)および飽和NaCl(1×15mL)で洗浄した。乾燥(MgSO4)、濾過、および蒸発させた後、クーゲルロール装置を用いて、残渣を50℃(浴温)および0.014mm Hgで減圧乾燥させた。

アルゴン下で、乾燥ジクロロメタン(15mL)中のN,N’−ジシクロヘキシルカルボジイミド(DCC,2.41g;14.1mmol)の溶液を、2,2,2−トリクロロエチルω−ヒドロキシアルカノエート、5−(Boc−アミノ)−4−オキソペンタン酸、および0℃(浴温)に冷却した乾燥ジクロロメタン中のピリジンの攪拌溶液に滴下して添加した。0℃で1時間攪拌した後、混合物を周囲温度で一晩攪拌した。反応混合物を減圧濾過し、酢酸(3mL)を濾液に添加した。30分間放置した後、混合物を再濾過し、濾液をジエチルエーテル(150mL)で希釈した。溶液を、1MのHCl(3×25mL)、水(3×25mL)、飽和NaHCO3溶液(2×25mL)、および飽和NaCl溶液(1×25mL)で洗浄した。乾燥させ(MgSO4)、濾過し、蒸発させた後、残渣を、酢酸エチル−ヘキサンで溶出するシリカゲル60カラムフラッシュクロマトグラフィーにより精製した。生成物を含有する画分を蒸発させて、生成物を得た。

この化合物は、実施例12aの手順を用いて、12−ヒドロキシドデカン酸(2.0g;9.2mmol)、2,2,2−トリクロロエタノール(6.0g;40mmol)、N,N’−ジシクロヘキシルカルボジイミド(DCC,4.1g;20mmol)、および乾燥ジクロロメタン(50mL)中のピリジン(5.0mL)から調製された。3.63g(66%)の生成物を得た(無色の油状物)。

この化合物は、手順Eを用いて、5−(Boc−アミノ)−4−オキソペンタン酸(2.0g;8.6mmol)、14aの生成物(3.0g;8.6mmol)、N,N’−ジシクロヘキシルカルボジイミド(DCC,2.1g;10.0mmol)およびジクロロメタン(75mL)中のピリジン(4.0mL)から調製された。粗生成物を、酢酸エチル−ヘキサン(1:3)で溶出した90×50mmシリカゲルカラムで精製し、13×50mLの画分を収集した。画分6〜11を蒸発させた後、1.44g(30%)の生成物を得た(帯黄色の油状物)。

この化合物は、実施例12cの手順に従い、同量の1M KH2PO4溶液および亜鉛粉末を用いて、テトラヒドロフラン(25mL)中の14bの生成物(1.38g;2.5mmol)から調製された。処理後、1.0g(93%)の生成物を得た(淡黄色の油状物)。
13C NMR(50MHz;CDCl3):δ 24.7、25.8、28.3、28.5、29.0、29.2、29.3、29.4、32.6、34.0、50.3、65.0、172.4、178.9、204.3 ppm。
アルゴン下で、周囲温度のジエチルエーテル中のHClの1.0M溶液(5.0mL;5.0mmol)を、酢酸エチル(10mL)中の14cからの生成物(0.95g;2.2mmol)の攪拌溶液に添加した。5日後、混合物を濾過し、残渣をジエチルエーテル(2×5mL)で粉砕した。残渣を、40℃および15mm Hgで一晩乾燥させ、0.11gの白色固体を得た。合わせたエーテル抽出物を蒸発させた。LC−MS分析は、生成物が約25%の5−アミノ−4−オキソペンタン酸を含有したことを示し、混合収穫物は、アセトニトリル(1000mL)中3%のメタノール、アセトニトリル(1000mL)中6%のメタノール、次いで、アセトニトリル(2000mL)中9%のメタノールで連続的に溶出させる150×25mmシリカゲル60カラムフラッシュクロマトグラフィーにより精製され、56×50mLの画分を収集した。生成物を含有する画分を蒸発させ、残渣を水(10mL)に溶解した。溶液を一晩凍結乾燥させ、0.06g(7.5%)の生成物を得た(白色固体,mp110〜115℃(軟化、鋭いmpなし))。

この化合物は、手順Bを用いて、16−ヒドロキシヘキサデカン酸(2.45g;9.0mmol)、ベンジルアルコール(4.9g;45mmol)、およびトルエン(100mL)中のp−トルエンスルホン酸(100mg)から調製された。2.82gの粗生成物を得て、これは、予測された生成物と16−ヒドロキシヘキサデカン酸の2つの分子間のエステルの1:1の混合物であった。ベンジルアルコール(10g)中のNaH(約100mg)の溶液(前に乾燥ヘキサンで3回洗浄された)を、粗生成物に添加し、アルゴン下で3時間、混合物を100℃(浴温)で攪拌した。周囲温度に冷却した後、混合物をジクロロメタン(75mL)で希釈し、10%の水性クエン酸溶液(1×10mL)で洗浄し、乾燥させた(Na2SO4)。濾過し、蒸発させた後、クーゲルロール装置を用いて、残渣を50℃(浴温)および0.014mm Hgで減圧乾燥させた。2.56g(76%)の生成物を得た(白色固体)。プロトンNMRは、約10%の二量体エステルが残存していたことを示した。
13C NMR(50MHz;CDCl3):δ 24.9、25.7、28.9、29.1、29.2、29.4、29.6、32.8、34.3、63.0、66.0、128.0、128.2、128.4、136.0、173.6 ppm。
この化合物は、手順Dにより、15aの生成物(1.2g;3.2mmol)、5−(Cbz−アミノ)−4−オキソペンタン酸(0.84g;3.2mmol)、4−ピロリジニルピリジン(50mg)、およびジクロロメタン(35mL)中のN,N’−ジイソプロピルカルボジイミド(0.50g;4.0mmol)から調製された。反応時間は3日であった。粗生成物を、酢酸エチル−ヘキサン(1:1)(1000mL)で溶出した85×55mmシリカゲル60カラムにより精製し、13×50mLの画分を収集した。生成物(3〜6)を含有する画分を蒸発させ、0.57g(29%)の生成物を得た(白色固体,mp78〜81℃)。

この化合物は、実施例8dの手順を用いて、15bの生成物(0.45g;0.74mmol)、12MのHCl(0.062mL;0.74mmol)、10%のPd/C(100mg)、水素ガス、および2−プロパノール(25mL)から調製された。0.06g(19%)の生成物を得た(白色固体)。

皮膚用途のために光増感剤の前駆体としての本発明の化合物の可能性を評価するために、研究が行われた。そのため、本発明の化合物は、皮膚に浸透し、皮膚の細胞によって取り込まれ、光増感剤に変換される必要がある。このプロセスの結果は、光増感剤分子を励起する光に皮膚を曝露し、励起された光増感剤分子がエネルギー的により低い状態に緩和されるときに蛍光の形態で放出されるエネルギーの量を決定することによって可視化される。

上記の化合物2、4、および7は、Unguentum Merckにおいてクリームとして製剤化された。化合物は、等モル量で試験された。

19%(190mg/g)の化合物7。
化合物1c(カルボキシメチル5−アミノ−4−オキソペンタノエート臭化水素酸塩、実施例1cに従い調製)は、Unguentum Merckにおいてクリームとして製剤化された(15%,150mg/g;0.56mmol)。
図1に見られるように、皮膚蛍光は時間と共に増加した。
クリームは上述のように施された。t=0(すなわち、クリームを施す前)、1.5、5、および12時間で、10mmの生検パンチ(AcuPunch 10mm,Acuderm,USA)を用いて生検試料を採取した。試料を採取し、最小限に照明された部屋で取り扱った。各試料はいかなる皮下脂肪組織も含まないように縁取りされ、その後半分に切断された。次いで、各試料を、アルミニウム箔の袋または類似物の中で、液体窒素で急速凍結し、−80℃の冷凍庫に移した。
1.5時間後、蛍光は脂腺において既に見ることができ、5時間後、蛍光は表皮においても見られた。化合物1cは、よって、脂腺に対して選択性を示し、したがって、座瘡、毛孔性角化症、皮脂過形成、脂腺癌、または脂腺腫等の脂腺に影響を及ぼす疾患の光線力学的治療に有用であり得る。
UV損傷皮膚において選択的蓄積を示す化合物6の可能性を評価するために実験を行った。評価のために、化合物の適用後に皮膚蛍光が健康なマウスの皮膚およびUV損傷マウスの皮膚においてインビボで測定された。UV損傷皮膚(日射損傷)を有するヌードマウスモデルは、K.Togsverd−Bo et al.,Exp.Dermatol.2012,21,260−264に記載される通りに確立された。
UV損傷マウスの皮膚の皮膚蛍光[a.u]に対する健康なマウスの皮膚における皮膚蛍光[a.u.]の比率が決定された。
光線力学的反応を介して細菌を死滅させるための、本発明に従う様々な化合物の有効性は、グラム陽性菌スタヒロコッカス・アウレウスにおいて検証された。
化合物1c:カルボキシメチル5−アミノ−4−オキソペンタノエート臭化水素酸塩(実施例1cに従い調製)
化合物4:2−カルボキシエチル5−アミノ−4−オキソペンタノエート臭化水素酸塩(実施例4に従い調製)
細菌株:S.アウレウス株DSM 20231(ATCC 12600)。実験を行う前に細菌株を、35〜37℃で、心臓浸出液寒天(Difco)上で24時間成長させ、McFarland 0.5標準液に対応する濃度(約1−5×108CFU/mL)の20mMのPIPES,pH7.2〜7.4に再懸濁した。生存細胞培養を確実にし、接種サイズおよび純度をチェックするために、細菌のストックは、実験の直前に作製され、CFU/mLは、標準コロニーアッセイにおいて決定された。
暗所毒性は、プレートが照明されなかったことを除き、以下の「PDT治療」の段落に記載されるように決定される。続いて、マイクロプレートアッセイが以下に記載されるように行われた。
結果は図2に示され、両化合物が、S.アウレウス菌を死滅させるのに有効であり、細菌細胞の再成長を効果的に遅延させることができることがこの図から見られる。
エームズ試験(B.N.Ames et al,Methods for detecting carcinogens and mutagens with the Salmonella/mammalian−microsome mutagenicity test.Mutation Research 31,347−364,1975を参照)は、試験化学化合物がある特定の細菌株において変異を誘発する能力を検出するための、規制機関によって承認された標準的な試験法である。試験化学化合物がそのような変異を誘発する場合、ヒトにおいて使用するための医薬化合物に使用することができない。
光増感剤の前駆体としての本発明の化合物の可能性を評価するために、研究が行われた。そのため、本発明の化合物は、細胞によって取り込まれる必要があり、光増感剤、すなわち、PpIXに変換される必要がある。このプロセスの結果は、PpIX分子を励起する光に細胞を曝露し、励起されたPpIX分子がエネルギー的により低い状態に緩和されるときに蛍光の形態で放出されるエネルギーの量を決定することによって可視化される。

化合物14(実施例14に従い調製された11−カルボキシウンデシル5−アミノ4−オキソペンタノエート塩酸塩)
化合物15(実施例15に従い調製された15−カルボキシペンタデシル5−アミノ4−オキソペンタノエート塩酸塩)
以下の化合物は、参照化合物として使用された。
5−ALA塩酸塩(ALA)
5−ALAヘキシルエステル塩酸塩(HAL)
上記の化合物のストック液は、100mMの濃度にそれらをDMSOに溶解することにより調製された。実験に使用された濃度は、ストック液をPBSまたは細胞培養培地で希釈することにより得られた。
直腸S状結腸の原発性腺癌に由来するWiDr細胞を、10%のウシ胎児血清、100U/mlのペニシリン、100μg/mlのストレプトマイシン、および1%のグルタミンを含有するRPMI 1640培地(Gibco)中で継代培養した。1週間に2回、細胞を1:100に分け、湿潤環境において、37℃および5%のCO2で維持した。実験を行うために、上述の2mlの培地中の5×105WiDr細胞を、6−ウェルプラスチック製組織培養プレート(Nunc)の各ウェルに添加し、基質に適切に付着させるために、湿潤環境において、37℃および5%のCO2で48時間放置した。次いで、細胞を、血清を含まないRPMI 1640培地で2回洗浄し、続いて、2つ組で、2mlの新しい培養培地中の上記の化合物および参照物を0.001、0.003、0.01、0.03、0.1、0.3、および1mMの最終濃度に適切に希釈したウェルに添加した。2つのウェルには、2mlの新しい培養培地のみが添加された。これらの未処理の細胞は、対照として機能する。細胞を、37℃で4時間、暗所でインキュベートした。
暗所毒性、すなわち、光の不在下での細胞毒性は、様々な濃度の化合物および参照物を用いた上記の4時間のインキュベーション直後に、MTSアッセイにより測定された。ALA塩酸塩および化合物5、9、10、12、および14に関して、細胞毒性は、試験をした濃度のいずれにおいても観察されなかった。ヘキシルエステル塩酸塩に関して、弱い細胞毒性が85%の最小細胞生存率で0.3〜1mMの範囲で観察された。化合物15は、0.1mMを超える濃度で毒性であった。
上述の本発明の化合物および参照化合物で処理した後、細胞をPBSで2回洗浄し、Costar細胞スクレーパで基質を50%のメタノール中の1M HClO4の溶液中にこすり落とした。細胞残屑を遠心分離により除去した。PpIXは、この手順で細胞から定量的に抽出された。各試料中のPpIX含量は、Perkin Elmer LS50B分光蛍光計を用いて蛍光定量的に決定された。PpIXは407nmで励起され、放出された蛍光は、ロングパスカットオフフィルタ(530nm)を用いて、放出側で、606nmで測定された。合計蛍光を50〜100%増加させる濃度で、既知の標準PpIX濃度が試料に添加された。各試料中のPpIX濃度は、ビシンコニン酸タンパク質アッセイにより測定される、対照細胞中のタンパク質含量に対して計算された。
5−ALAヘキシルエステル塩酸塩(HAL)は、Hexvix(登録商標)の活性成分であり、膀胱における癌の光線力学的検出のための市販の薬物である。調査研究は、膀胱癌の光線力学的治療のためのHexvix(登録商標)の使用も示す。HALは、膀胱癌細胞においてPpIXに変換され、その特有の蛍光によって検出され得る。Hexvix(登録商標)の開発における初期研究は、ヒト膀胱細胞および組織のモデルとして、ラットの膀胱細胞および組織において行われた。この研究は、膀胱において使用することができる光増感剤の前駆体としての本発明の化合物の可能性を評価する第1のステップとして行われた。そのような第1のステップでは、化合物の適切な濃度は、化合物のPpIX蛍光をHALのものと比較することにより、インビトロで確立された。その結果に基づき、さらなるインビボ研究の濃度が確立される。
化合物4:2−カルボキシエチル5−アミノ−4−オキソペンタノエート臭化水素酸塩(実施例4に従い調製)
化合物5:(実施例5に従い調製された3−カルボキシプロピル5−アミノ−4−オキソペンタノエート塩酸塩)
化合物8:実施例8に従い調製された2−カルボキシブチル5−アミノ−4−オキソペンタノエート塩酸塩)
5−ALAヘキシルエステル塩酸塩(HAL)は、参照化合物として使用された。
ラットの膀胱細胞(AY−27)を、培養培地(9%のウシ胎児血清、1%のL−グルタミン(200mM)、および1%のペニシリン(10 000UI)、ストレプトマイシン(10000μg.mL−1)で補足されたRPMI 1640)中で成長させた。指数関数的に成長する細胞(AY−27)を、新しく調製した0.8mMのHAL溶液(ヒト患者において膀胱癌の検出に使用される濃度の10分の1)および3〜15mMの濃度で(下の表を参照)新しく調製した化合物4、5、および8の溶液と共に2時間インキュベートした。対照細胞は、いずれの試験または参照化合物を含まない無血清培地を受けた。インキュベーション後、溶液または培地を廃棄し、細胞を氷冷PBSで2回すすいだ。エタノール/DMSO/酢酸(80/20/1,v/v/v)からなる氷***解抽出混合物を使用して、PpIXを細胞から抽出した。細胞可溶化物を遠心分離し(400g、10分、2℃)、ペレットを粉砕し、遠心分離する前に懸濁液を15分間音波処理した。PpIXの蛍光シグナルは、Perkin−Elmer LS 55分光計(Perkin−Elmer,Beaconsfield,UK)で測定され、タンパク質含量の画分として表された。各試料における635nmでの蛍光強度は、上述の抽出混合物中に希釈されたPpIX(Sigma−Aldrich,France)を用いて確立された較正曲線について報告された。PpIX濃度は、タンパク質含量に対して表された。簡潔に、溶解緩衝混合物(1mMのEDTA、1%のTriton X−100、および10mMのTris−HCl;pH7.4)を、DMSO中500mMのPMSF(抗プロテアーゼ)溶液と共に凍結細胞皿に添加された。可溶化物を氷上に30分間維持し、4℃で20分間、15000gで遠心分離した。次いで、Lowryによって開発された方法に基づくBiorad DCタンパク質アッセイキットBio−Rad,France)により、BSA較正曲線に対してタンパク質含量を測定した。
膀胱は低pHに感受性であり、一方、本発明の化合物(および一般的にALA−エステル)は、高pHでピラジンを形成する。よって、本発明の化合物の溶液は、膀胱が容認できないpHとなってはならない。培地のpHに対する試験化合物の作用は、1、5、および25mMの濃度で化合物をRPMI 1640培地に添加し、続いてpHを測定することにより試験された。1mMの化合物は、培地のpHを変えないが、5および25mMの化合物は、pHを、それぞれ、7.5から7.1および5.5に変えたことが分かった。データをプロットすることにより、15mMの化合物が補間によりpH6.0となるであろうことが分かった。pH6未満で、膀胱における有害作用(収縮など)が生じる可能性がある。15mMは、インビボ研究の10mMの予測された濃度より十分に高い(HALでの実験、およびインビトロ濃度の10倍がインビボで使用されるという過程に基づく)。

- X基が、1〜4の炭素原子からなる直鎖状のアルキレン基である請求項1に記載の化合物。
- 請求項1〜6のいずれか一項に記載の式Iの化合物、またはその薬学的に許容される塩を含む、組成物。
- 少なくとも1つの薬学的に許容されるか、または美容的に許容される担体または賦形剤をさらに含む請求項7に記載の組成物。
- 式Iの化合物が、X基が、1〜4の炭素原子からなる直鎖状のアルキレン基である化合物である請求項7または8に記載の組成物。
- 光線力学的治療または診断の方法において使用するための、請求項7〜12のいずれか一項に記載の組成物。
- 前記方法が、癌;癌に関連した感染;ウイルス、細菌、もしくは真菌感染;または非癌性状態の治療または診断のためである、請求項13に記載の使用のための組成物。
- 前記非癌性状態が、炎症である請求項14に記載の使用のための組成物。
- 前記方法が、癌の治療または診断のためである請求項13に記載の使用のための組成物。
- 前記方法が、膀胱癌、結腸癌、もしくは胃癌;ヒトパピローマウイルス、B型肝炎、もしくはエプスタインバーウイルス;ヘリコバクターピロリ感染もしくは座瘡;または炎症性座瘡、大腸炎、もしくは感染性皮膚炎の治療または診断のためである、請求項13に記載の使用のための組成物。
- 光線力学的治療または診断の方法において使用するための、請求項1〜6のいずれか一項に記載の化合物。
- 前記方法が、癌;癌に関連した感染;ウイルス、細菌、もしくは真菌感染;または非癌性状態の治療または診断のためである、請求項18に記載の使用のための化合物。
- 前記非癌性状態が、炎症である請求項19に記載の使用のための化合物。
- 前記方法が、癌の治療または診断のためである請求項18に記載の使用のための化合物。
- 前記方法が、膀胱癌、結腸癌、もしくは胃癌;ヒトパピローマウイルス、B型肝炎、もしくはエプスタインバーウイルス;ヘリコバクターピロリ感染もしくは座瘡;または炎症性座瘡、大腸炎、もしくは感染性皮膚炎の治療または診断のためである、請求項18に記載の使用のための化合物。
