Description
本出願は、2008年10月10日に出願された米国特許仮出願第61/195,761号の利益を主張するものである。
本明細書で使用されるとき、「a」という用語は、別途記載のない限り1つ以上を指す。
種々の態様において、本発明は、生体外活性に与える影響を最小限に抑える一方で、FGF21分子の薬物動態特性を向上させることができる、FGF21ポリペプチドおよびFGF21変異体ポリペプチドの部位特異的PEG化のための一連の方法を開示する。これらのPEG化されたFGF21分子の薬物動態プロファイルの向上は、治療剤への曝露を増加することによって、その分子の生体内での有効性に影響を与える。また、本明細書に記載されるストラテジーは複数のPEG化部位の作製に対応しており、それによって、分子の薬物動態特性をさらに向上させると同時に、空胞を形成する可能性を低減することができる。本明細書に記載されるように、2つの主要なストラテジーが、これを達成するために用いられる。

FGF21の成熟した形態(すなわち、181残基の形態)が生体内分解を受けることが明らかになっており、それは最終的にタンパク質分解攻撃から生じることが分かった。成熟FGF21の生体内分解は、有効半減期の短縮を引き起こすことが明らかになっており、それは分子の治療可能性に有害な影響を与える可能性がある。したがって、タンパク質分解に対する耐性を示すFGF21変異体を同定するために、該当する試験を行った。この調査の結果として、特にタンパク質分解を受け易いことが明らかになっている成熟FGF21ポリペプチドの部位は、4〜5位、20〜21位、151〜152、および171〜172位でアミノ酸残基間にペプチド結合を含む。
本発明の種々の態様において、FGF21変異体ポリペプチドが開示される。別の態様において、本発明のFGF21変異体ポリペプチドは、非自然発生ポリマー(例えば、PEG化)結合部位(複数化)が導入されたFGF21ポリペプチドを含む。本発明のさらに別の態様において、非自然発生ポリマー(例えば、PEG)結合部位(複数化)が導入されたFGF21変異体ポリペプチドの切断された形態が開示される。非自然発生ポリマー結合部位と、野生型FGF21の生物学的活性を維持することができるがその必要はない、1つ以上の保存的または非保存的な置換と、を含むFGF21変異体ポリペプチドが、本発明の別の態様を形成する。本発明の種々のFGF21ポリペプチド変異体は、本明細書に記載するように、また本明細書に提供される参考文献に記載されるように調製することができる。
本発明の一実施形態は、1つ以上の非自然発生ポリマー結合部位を含む変異体FGF21ポリペプチドの切断された形態を対象とする。そのような切断された変異体ポリペプチドは、化学修飾されてもよいが、その必要はない。
別の実施形態において、本発明は、ポリペプチドのC末端に別の1つ以上の残基を付加して、アミノ酸配列を野生型タンパク質よりも長くすることによってキャッピングされた、1つ以上の非自然発生ポリマー結合部位を含む変異体FGF21ポリペプチドを対象とする。さらに別の実施形態において、本発明は、1つ以上のC末端突然変異をさらに含む1つ以上の非自然発生ポリマー結合部位を含むFGF21変異体ポリペプチドを対象とする。そのようなキャッピングされたC末端突然変異させたFGF21変異体ポリペプチドは、化学修飾されていてもよいが、その必要はない。
本発明のさらなる態様において、野生型FGF21ポリペプチド配列の両方のシステイン残基が、ジスフィルド結合を形成せず、またポリマー結合部位としての役割も果たさないアラニンまたはセリン等の残基と置き換えられた、FGF21変異体ポリペプチドを調製することができる。続いて、チオール含有残基(例えば、システイン残基、またはチオール基を有する非自然発生アミノ酸)あるいは遊離アミノ基(例えば、リジン残基もしくはアルギニン残基、または遊離アミノ基を有する非自然発生アミノ酸)の形態で非自然発生ポリマー結合部位を導入するFGF21変異体ポリペプチド配列において、置換が行われてもよい。次いで、結合するためにチオール基または遊離アミノ基に依存するPEG等のポリマーは、既知の位置でFGF21変異体ポリペプチド配列に導入されたシステイン、リジン、またはアルギニン残基に標的化されてもよい。このストラテジーは、より効率的かつ制御されたポリマーの配置を促進することができる。
本発明のさらに別の態様において、本明細書に記載されるように「連結分子」を調製することができる。「連結分子」は、リンカー分子によって連結された2つのFGF21ポリペプチドを含む分子である。2つのFGF21ポリペプチドを結合させることにより、連結分子の有効半減期および強度を、単一のFGF21ポリペプチドの有効半減期および強度を超えて拡張することができる。
連結分子が2つのFGF21変異体ポリペプチドを含む場合、FGF21変異体ポリペプチドは、配列に導入された1つ以上の突然変異を含むことができるが、その突然変異は、FGF21変異体ポリペプチドのそれぞれにおける同じアミノ酸位置にある必要はない。例として、連結分子が2つのFGF21変異体ポリペプチドを含む場合、一方のFGF21変異体ポリペプチドはH125C突然変異を含有することができ、それがリンカー分子のための結合部位を形成してもよい。対照的に、他方のFGF21変異体ポリペプチドはH125以外の位置で突然変異を含有することができ、それが2つのFGF21変異体ポリペプチドを一緒に連結するリンカーの結合点としての役割を果たすことができる。例え1つまたは2つのFGF21変異体ポリペプチドが用いられたとしても、リンカーはFGF21変異体ポリペプチドのN末端に結合することができ、必ずしも導入された結合点が用いられなくてもよい。

本発明のいくつかの実施形態において、FGF21変異体ポリペプチドは、PEGサブユニットで共有結合的に修飾される。いくつかの実施形態において、1つ以上の水溶性ポリマーが、FGF21変異体の1つ以上の特異的な位置(例えば、N末端)で結合される。いくつかの実施形態において、1つ以上の水溶性ポリマーがFGF21変異体の1つ以上の側鎖に結合し、これらの側鎖は自然発生であってもよいか、または遺伝子操作されたポリマー結合部位の構成要素を形成することができる。いくつかの実施形態において、PEGは、FGF21変異体ポリペプチドの治療能力を向上するために使用される。特定のそのような方法は、例えば、任意の目的のために、参照により本明細書に組み込まれる米国特許第6,133,426号において考察される。
多糖類ポリマーは、タンパク質の修飾に使用することのできる別の種類の水溶性ポリマーである。したがって、本発明のFGF21変異体ポリペプチドは、本発明の実施形態を形成するように多糖類ポリマーに結合されてもよい。したがって、FGF21変異体ポリペプチドの遺伝子操作した非自然発生ポリマー結合部位は、多糖類が結合される残基を含むことができる。デキストランは、主にα1〜6結合によって結合されるグルコースの個別サブユニットを含む多糖類ポリマーである。デキストラン自体は多くの分子量の範囲で使用可能であり、約1kD〜約70kDの分子量で容易に使用可能である。デキストランは、それ自体がビヒクルとして使用される好適な水溶性ポリマーであるか、または、または別のビヒクルと合わせて(例えば、Fc)使用される好適な水溶性ポリマーである。例えば、国際公開第96/11953号を参照のこと。治療用または診断用の免疫グロブリンと共役されたデキストランの使用が報告されている。例えば、参照により本明細書に組み込まれる欧州特許公開第0315456号を参照のこと。また本発明は、約1kDa〜約20kDaのデキストランの使用を包含する。
一般に、化学修飾(例えば、PEG化またはグリコシル化)は、タンパク質を活性化されたポリマー分子と反応させるために用いられる任意の好適な条件下で行うことができる。化学修飾されたFGF21変異体ポリペプチドを調製するための方法は、通常、以下の工程:(a)FGF21変異体ポリペプチドが1つ以上のポリマー分子に結合する条件下で、ポリペプチドを活性化されたポリマー分子(反応性エステル、マレイミド、またはポリマー分子のアルデヒド誘導体等)と反応させる工程と、(b)反応産物を得る工程、を含む。最適な反応条件は、既知のパラメータおよび所望の結果に基づいて決定される。例えば、タンパク質に対するポリマー分子の比率が高いほど、結合するポリマー分子の割合が大きい。本発明の一実施形態において、化学修飾されたFGF21変異体ポリペプチドは、アミノ末端に単一のポリマー分子を有することができ、他の実施形態において、FGF21変異体ポリペプチドは、例えば、ポリペプチドのN末端に1つのポリマー、そしてポリペプチドの別の残基に第2のポリマーのように、一次配列に会合する2つ以上のポリマーを有することができる。代替として、FGF21変異体は、一次配列の2つの異なる残基と会合する2つ以上のポリマーを有することができるが、N末端にはポリマーを有することはできない。
FGF21変異体ポリペプチドを含む治療用組成物は、本発明の範囲内であり、特に、強化された特性を示す連結分子、FGF21変異体ポリペプチド、および化学修飾されたFGF21変異体ポリペプチドの同定を考慮して企図される。そのような連結分子、FGF21変異体ポリペプチド、および化学修飾されたFGF21変異体ポリペプチドの治療用組成物は、投与様式との適合性に合わせて選択される薬学的にまたは生理学的に許容される製剤用物質との混合物中で化学修飾することのできる、治療的有効量の連結分子、FGF21変異体ポリペプチド、または化学修飾されたFGF21変異体ポリペプチドを含むことができる。
本発明の連結分子、FGF21変異体ポリペプチド(切断されているか、キャッピングされているか、またはC末端突然変異されていてもよい)、および化学修飾されたFGF21変異体ポリペプチドは、これに限定されないが、代謝異常を含む多数の疾患、障害、または状態を治療、診断、寛解、または予防するために使用されてもよい。一実施形態において、治療される代謝異常は糖尿病である。別の実施形態において、代謝異常は肥満である。他の実施形態は、脂質異常症等の代謝疾患または代謝異常、高血圧、非アルコール性脂肪性肝炎等(NASH)等の脂肪肝、粥状動脈硬化等の循環器疾患、および加齢を含む。
本発明の連結分子、FGF21変異体ポリペプチド、および化学修飾されたFGF21変異体ポリペプチドには特異的に結合するが、野生型FGF21ポリペプチドには特異的に結合しない抗体および抗体断片が企図され、本発明の範囲内に含まれる。抗体は、単一特異的なポリクローナルを含むポリクローナル;モノクローナル(MAbs);組換え;キメラ;相補性決定領域(CDR)グラフト化等のヒト化;ヒト;単鎖;および/または二重特異性、ならびにそれらの断片、変異体、あるいは化学修飾された分子であってもよい。抗体断片は、FGF21変異体ポリペプチドのエピトープに特異的に結合する抗体のそれらの部分を含む。そのような断片の例は、全長抗体の酵素的切断によって作製されるFabおよびF(ab’)断片を含む。他の結合断片は、抗体可変領域をコードする核酸配列を含有する組換えプラスミドの発現等の、組換えDNA技術によって作製されるものを含む。
FGF21ポリペプチド発現コンストラクトの調製
成熟FGF21配列の5’および3’末端に対応するヌクレオチド配列を有するプライマーを用いて、ポリメラーゼ連鎖反応(PCR)増幅により成熟FGF21ポリペプチドをコードする核酸配列を得た。表4に、成熟FGF21配列を増幅するために使用したプライマーを列挙する。
細菌からの野生型FGF21ポリペプチドの精製
以下の実施例では、細菌発現系において野生型FGF21ポリペプチドを発現させた。以下に記載される発現の後、野生型FGF21ポリペプチドを、別途記載のない限り、本実施例に記載するように精製した。
FGF21変異体の同定
野生型FGF21は比較的短い半減期を有するため、ある場合においては、治療剤として野生型FGF21を使用することが望ましくない可能性がある。また、ポリペプチドのPEG化等、半減期を延長する従来方法は、FGF21配列の好適なPEG化部位の数および場所によって限定される。野生型FGF21ポリペプチド配列には、7つの自然発生PEG化部位、すなわち、αアミノ基、4つのリジン残基、および2つシステイン残基が存在する。PEG分子は、FGF21がその天然構造を得る能力に有害な影響を与える可能性があるか、または、FGF21とその受容体もしくはβ−klothoとの間の相互作用に有害な影響を与える可能性があるため、これらの部位はPEG化には理想的ではない。また、αアミノ基を除いて、これらの反応性残基は、単一の標的の場所での部位特異的なPEG化を許容しない。したがって、化学修飾に好適な残基に突然変異させることができる野生型FGF21ポリペプチド内の残基を同定するために該当する集中的な試験を開始した。試験の検討事項には、FGF21配列中の残基の場所、およびタンパク質表面上での残基の位置が含まれた。
相同性モデルを使用して同定された突然変異候補
第1のストラテジーでは、系統的な合理的タンパク質工学アプローチにおいて、相同性モデルを用いて、FGF21の活性を妨害することなくPEG化に耐える可能性のある表面に露出した側鎖を有する確率の高い残基を同定した。そのような残基を同定するために使用することのできるFGF21のX線構造またはNMR構造が発表されていないため、Protain Databank(PDB)から取得したFGF19(1PWA)の高解像度(1.3Å)X線結晶構造を用いて、MOE(Molecular Operating Environment;Chemical Computing Group;Montreal,Quebec,Canada)モデリングソフトウェアを使用して、FGF21の3D相同性モデルを作製した。PDBに寄託されたタンパク質の中で、アミノ酸配列相同性の点において、FGF19がFGF21のタンパク質に最も密接に関連していたため、FGF19をテンプレートとして選択した。
望ましくない自然発生ポリマー結合部位を除去することによって作製される
FGF21変異体
第2のストラテジーは、PEGをFGF21に結合させるためにアミノ特異的共役化学を用いた。潜在的に空胞を形成する共役の部位選択的な二重PEG化は、それらが空胞を形成する可能性を有意に減少させることが可能である(例えば、米国特許第6,420,339号を参照のこと)。したがって、本発明の一態様において、FGF21の部位選択的な二重PEG化は、2つの一次アミノ基のみがPEG化のために残るようにタンパク質を突然変異させることによって達成される。
単一の突然変異を含むFGF21変異体の同定
実施例3Aおよび3.Bで上述した合理的タンパク質工学アプローチによって作製した、単一の突然変異を含むFGF21変異体の概要を表3に提供する。これらの単一の変異体は、非自然発生ポリマー結合部位、とりわけシステイン残基またはリジン残基を提供し、また組み込む。これらの残基の側鎖は、PEG分子等のポリマーの結合による化学修飾に特に好適である。所望の場合は、導入された非自然発生ポリマー結合部位に加えて、ポリマーが任意でタンパク質のN末端に結合できることが確認された。
2つの突然変異を含むFGF21変異体の同定
2つの非自然発生ポリマー結合部位を提供するであろう変異体の組み合わせを同定するために、相同性モデルと単一のシステイン変異体から得られたデータとのさらなる分析を行った。化学修飾後、これらのFGF21変異体は、所望の場所でポリペプチドに結合する2つのポリマー(例えば、PEG分子)を有する。本発明のすべてのFGF21変異体ポリペプチドと同様に、ポリマー自体の性質に依存して、ポリマーはポリペプチドのN末端にも結合することができる。二重にPEG化されたFGF21変異体を表すイラストを図1に図式化して示す。
3つの突然変異を含むFGF21変異体の同定
上記の通り、非自然発生ポリマー結合部位を野生型FGF21ポリペプチド配列に組み込むことができる。このことは、ポリペプチドの所望の場所における部位選択的な化学修飾の機会を提供する。また、野生型FGF21配列の残基P171はタンパク質分解を受け易いことが事前に確認されている。したがって、上記修飾の組み合わせ、さらにP171位の突然変異を導入することにより、強化されたタンパク質分解に対する安定性と部位特異的ポリマー結合部位の両方を有するFGF21分子を作製することができる。
FGF21変異体ポリペプチドの調製および発現
表3(単一の突然変異)、表4(2つの突然変異)、および表5(3つの突然変異、すなわち、2つの突然変異+P171G)に列挙したFGF21変異体(この実施例では集合的に「FGF21変異体」)をコードするコンストラクトを、以下に記載するように野生型FGF21発現ベクターのPCR増幅によって調製した(野生型FGF21発現ベクターの構築は実施例1に記載)。これらの実験の目的は、1つ以上の非自然発生ポリマー結合部位を含み、またある場合にはタンパク質分解に耐性であるFGF21変異体を作製することであった。これらのFGF21変異体の化学修飾された形態は、より長い半減期を示し、またある場合にはタンパク質分解に耐性でもあることが予測される。
細菌からのFGF21およびFGF21変異体ポリペプチドの精製
以下の実施例では、細菌発現系において野生型FGF21ポリペプチドを発現させた。実施例7に記載される発現後、FGF21変異体ポリペプチドを、別途記載のない限り、本実施例に記載するように精製した。
FGF21変異体の化学修飾

FGF21変異体ポリペプチドおよび
化学修飾されたFGF21変異体ポリペプチドの生体外活性
次いで、FGF21変異体ポリペプチドの非PEG化形態およびN末端PEG化FGF21と比較して活性を評価するために、PEG化FGF21変異体ポリペプチドを生体外アッセイに供した。
1つの突然変異を含むPEG化FGF21変異体ポリペプチドのEC50値
下の表9は、特定の既知の場所に1つの非自然発生ポリマー結合部位を導入する2つの突然変異を含む種々のFGF21変異体ポリペプチドのEC50値をまとめたものである。
選択されたArg/Lys FGF21変異体のEC50値
実施例3に記載したように、突然変異誘発により自然発生ポリマー結合部位(例えば、PEG化部位)が除去された一連のFGF21変異体ポリペプチドを作製した。これらのFGF21変異体において、最初に反応性Lys基をアルギニンに突然変異させ、N末端のαアミノ基のみをPEG化に使用可能とした。次に、天然または変異のいずれかである選択アルギニン残基を1つずつリジンに変換し、そうすることで、FGF21表面上の規定の位置に第2のPEG化部位を導入した。このストラテジーの1つの目的は、ポリマー(例えば、PEG分子)が1つ以上の特定の既知の場所で共役されるであろうFGF21変異体ポリペプチドを作製することであった。
2つの突然変異を含む選択されたPEG化FGF21変異体ポリペプチドの
EC50値
2つの突然変異を含む多くのFGF21変異体ポリペプチドの活性も、本明細書に記載されるERK−ルシフェラーゼアッセイにおいて調べた。これらの変異体を、野生型FGF21配列に、2つの非自然発生ポリマー結合部位(システイン残基の形態)と、強化されたタンパク質分解に対する安定性を提供する突然変異(P171G)とを導入するように遺伝子操作した。
3つの突然変異を含む選択された化学修飾FGF21変異体ポリペプチドのEC50値
3つの突然変異を含む多くのFGF21変異体ポリペプチドの活性も、本明細書に記載されるERK−ルシフェラーゼアッセイにおいて調べた。これらの変異体を、野生型FGF21配列に、2つの非自然発生ポリマー結合部位(システイン残基の形態)と、強化されたタンパク質分解に対する安定性を提供する突然変異(P171G)とを導入するように遺伝子操作した。
選択されたFGF21の連結分子のEC50値
本発明の連結分子は、リンカー分子によって一緒に連結された2つのFGF21ポリペプチド配列を含む。図7は、連結分子の例を図式化して示す。本明細書に記載するように、これらの連結分子は、望ましいレベルの生物学的活性を保持する一方で、より長い半減期を提供するであろうと予想された。
化学修飾されたFGF21変異体ポリペプチドの生体内活性
FGF21は、血糖値、インスリン、トリグリセリド、もしくはコレステロールレベルの低下、体重の減少、または耐糖能、エネルギー消費量、もしくはインスリン感受性の向上を含む、多くの生物学的活性を保持する。実施例10に記載した最初の生体外評価の後、PEG化FGF21変異体ポリペプチドを生体内でのFGF21の活性についてさらに分析した。PEG化FGF21ポリペプチドをインスリン耐性ob/obマウスに導入し、特定のPEG化FGF21ポリペプチドが血糖値を低下させる能力を測定した。生体内研究のための手順は以下の通りである。
N末端PEG化野生型FGF21の生体内活性
ob/obマウスモデルにおいて、ポリペプチドのN末端PEG化により化学修飾した野生型FGF21を調べた。同じ実験で非PEG化野生型FGF21についても調べ、PBSを対照として用いた。単一の20kDaのメトキシPEGマレイミド分子を用いて野生型FGF21をN末端でPEG化した。図8は、この実験の結果を示す。
N末端および導入されたポリマー結合部位の両方で共役された、
選択された化学修飾FGF21変異体ポリペプチドの生体内活性
N末端および第2の遺伝子操作部位の両方で部位選択的にPEG化されたFGF21のいくつかの化学修飾した変異体をマウスob/obモデルにおいて調べた。これらの二重PEG化FGF21コンストラクトはすべて、N末端と、実施例4に記載したような遺伝子操作したリジンまたは実施例6に記載したような遺伝子操作したシステインのいずれかを含む第2の部位とでPEG化した。
2つまたは3つの突然変異を含む、
選択された化学修飾FGF21変異体ポリペプチドの生体内活性
マウスob/obモデルにおいて、多くの化学修飾されたFGF21変異体ポリペプチドについて調べた。これらの変異体は、導入されたシステイン残基の各々に1つずつ、また、単一の突然変異を含むFGF21変異体ポリペプチドの場合は、ポリペプチドのN末端に、2つの20kDaメトキシPEGマレイミド分子を付加することによって化学修飾した。突然変異は、1つ以上のPEG分子に別個の既知の結合点を提供するように選択した。いくつかのFGF21変異体も、タンパク質分解耐性を強化するためにP171G突然変異を含んだ。PEG化FGF21変異体ポリペプチドをマウスに注射し、9日間にわたる試験の間に間隔を空けてマウスから採血した。
グルコースレベルに及ぼす影響
さらに別のグループのPEG化FGF21変異体ポリペプチドを用いて実験を行った。図11は、ビヒクル(PBS)か、または突然変異(すなわち、E91/H125C、E91C/R175C、E37C/G120C、E37C/H125C、E37C/R175C)を含むFGF21変異体の二重PEG化形態を注射したマウスの血糖値レベルにおける変化率を示すプロットである(非PEG化Fc−G170E FGF21についても調べた)。20kDaのメトキシPEGマレイミド分子を用いた。この実験の結果は、二重にPEG化されたFGF21変異体が、野生型FGF21タンパク質と比較して強化された薬動力学を示すことを再度確認するものであった。
体重に及ぼす影響
2型糖尿病は、体脂肪および体重の増加を伴うことが多い。したがって、治療分子は、体重を減少させる望ましい効果とともに、血糖値レベルを低下させる望ましい効果も有することが好ましい。したがって、本明細書に記載されるPEG化FGF21変異体の多くをマウスの体重に与える影響について評価した。
マウス腎臓空胞試験
治療タンパク質の半減期を延長するためにPEGおよび他のポリマーを用いる際に考慮するべき1つの点は、PEG化分子が腎臓に空胞を形成する可能性である。PEGのこの特性は十分に裏付けされており(例えば、Bendele,et al 1998,“Short Communication:Renal Tubular Vacuolation in Animals Treated with Polyethylene‐glycol‐conjugated Proteins,”Toxicological Sciences,42:152〜157およびConover et al.,1996,“Transitional Vacuole Formation Following a Bolus Infusion of PEG‐hemoglobin in the Rat,”Art.Cells,Blood Subs.,and Immob.Biotech.24:599〜611を参照のこと)、予測不可能なことがある。腎臓の空胞形成を誘発するPEG化分子は、典型的には、必ずではないが、分子を治療タンパク質としてあまり望ましくない状態にする。いくつかの状況において、腎臓の空胞形成はそれほど懸念されるものではなく、許容されてもよい。したがって、マウスにおいて長期の腎臓空胞試験を行った。

- 前記第1のポリペプチド、前記第2のポリペプチド、または両方のポリペプチドは、前記リンカーに加えて、1つのポリマーにさらに共有結合している、請求項60に記載の組成物。
- 前記ポリマーは、水溶性ポリマーである、請求項66に記載の組成物。
