がんの個別化治療が早期に実現可能な、多数の弾頭と爆薬配列の組み合わせからなるペプチドトキシンには短期間で入手可能な化学合成法が相応しいが、遺伝子組換えにより強制発現させ精製する方法も可能である。
治療しようとするがん細胞について、細胞表面に高発現しているタンパク質のプロファイルおよび爆薬ペプチドに対するがん細胞の傷害感受性を調べる。その結果をもとに弾頭と爆薬を選択し、そのがん細胞に最適なペプチドトキシンをデザインする。化学合成等により得られたオーダーメイドペプチドトキシンを必要に応じてアテロコラーゲン等のDDSと組み合わせ、局所投与または全身投与を行い治療する。

本発明のペプチド(たとえば、キメラペプチド)は、当該分野で周知の方法(例えば、化学合成、以下に考察される一般的な工学技術)により得られ、または産生され得る。例えば、所望の領域もしくはドメインを含むペプチドの一部と一致するペプチドか、またはインビトロで所望の活性を媒介するペプチドは、ペプチド合成機の使用により合成され得る。ペプチドはまた、ペプチドの疎水性領域および親水性領域を同定するために使用され得る親水性分析(例えば、HoppおよびWoods、1981.Proc.Natl.Acad.Sci.USA.78:3824〜3828を参照のこと)により分析され得、従って実験操作(例えば、結合実験、抗体合成)のための物質の設計において助けとなる。二次構造分析はまた、特定の構造的モチーフを確立するペプチドの領域を同定するために行われ得る(例えば、ChouおよびFasman、1974、Biochem. 13:222〜223を参照のこと)。操作、翻訳、二次構造の予想、親水性および疎水性プロファイル、オープンリーディングフレームの予想およびプロッティング、ならびに配列の相同性の決定は、当該分野で利用可能なコンピューターソフトプログラムを使用して達成され得る。構造分析の他の方法として、例えば、X線結晶解析(例えば、Engstrom、1974.Biochem.Exp.Biol.11:7〜13を参照のこと));質量分析およびガスクロマトグラフィー(例えば、METHODS IN PROTEIN SCIENCE、1997.J.WileyおよびSons、NewYork、NYを参照のこと)が挙げられ、そしてコンピュータモデリング(例えば、FletterickおよびZoller、編、1986.Computer Graphics and Molecular Modeling:CURRENT COMMUNICATION IN MOLECULAR BIOLOGY、Cold Spring HarborLaboratory Press、Cold Spring Harbor、NYを参照のこと)もまた使用され得る。
本発明の化合物またはその製薬上許容される塩は、そのまま単独で投与することも可能であるが、通常各種の医薬製剤として提供するのが好ましい。また、それら医薬製剤は、動物および人に使用される。
本明細書において「送達剤」または「送達媒体」とは、目的の物質の送達を媒介する担体(ビヒクル)をいう。送達される物質が薬物であれば、「薬物送達媒体」という。薬物送達システム(Drug Delivery System、DDS)とは、ドラッグデリバリーシステムとも呼ばれ、吸収制御型DDS、放出制御型DDS、標的指向型DDSに分類することもある。理想的なDDSは、薬物を「体内の必要な部位に」、「必要な量を」、「必要な時間だけ」送り込むシステムである。ターゲティングDDSは、パッシブ・ターゲティングDDSとアクティブ・ターゲティングDDSとに分類される。前者はキャリアー(薬物担体または薬物運搬体)の粒子径や親水性など物理化学的性質を利用して体内挙動を制御する方法である。後者はこれらに特殊な仕組みを付け加えて積極的に標的組織への指向性を制御しようとする方法であり、例えば、標的組織を構成する特定細胞の標的分子への特異的分子認識機能を有する抗体(たとえば、本発明のTPR結合ペプチド)などを結合したキャリアを利用する方法があり「ミサイルドラッグ」と呼ばれることもある。
本明細書において「スクリーニング」とは、目的とするある特定の性質をもつ生物または物質などの標的を、特定の操作/評価方法で多数を含む集団の中から選抜することをいう。スクリーニングのために、本発明の特異的部分を使用することができる。
[材料および方法]
(細胞株)
ヒト乳がん細胞株(BT−20およびT47D)、肺がん細胞株(H322およびH460)、膵臓がん細胞株(SU.86.86)、前立腺がん細胞株(LNCap)、脳腫瘍細胞株(U251)および肺線維芽細胞株(MRC−5およびWI−38)を、American Type Culture Collection(Manassas,VA)から購入した。ヒト膵臓がん細胞株(BxPC−3)および結腸がん細胞株(HCT116およびDLD−1)を、European Collection of Cell Cultures(ECACC;Salisbury,Wiltshire,UK)から購入した。ヒト胚性腎臓細胞株(HEK293)を、RIKEN Cell Bank(つくば市)から購入した。細胞は、10% FBS(BioWest,Miami,FL)、100μg/ml ペニシリンおよび100μg/ml ストレプトマイシン(ナカライテスク株式会社、京都市)を含有する、RPMI 1640(BT−20、T47D、H322、H460、SU.86.86、LNCap、U251、BxPC−3、DLD−1およびSW837)、MEM(MRC−5およびWI−38)、McCoy’s 5a(HCT116)またはD−MEM(HEK293)中で培養した。
以下のペプチドをInvitrogen,Carlsbad,CAから購入した:
1.がん細胞膜溶解性ペプチド:KLLLKLLKKLLKLLKKK(配列番号1:下線の文字はD−アミノ酸である)
2.EGFR結合(EB)−がん細胞膜溶解性キメラペプチド:YHWYGYTPQNVIGGGKLLLKLLKKLLKLLKKK(配列番号2)
3.オリジナル溶解性ペプチド:LKLLKKLLKKLLKLL−NH2(配列番号41)
4.EB−オリジナル溶解性キメラペプチド:YHWYGYTPQNVIGGGLKLLKKLLKKLLKLL−NH2(配列番号42)
これらのペプチドを、固相化学を用いて化学合成し、そして、高速液体クロマトグラフィーにより均質(すなわち、90%より高い純度)になるまで精製して、質量分析法により評価した。ペプチドを水中に溶解させ、そして、pH7.4に緩衝化させた。ペプチド溶液は、使用直前に毎回新たに調製し、使い回しを避けた。
ゲフィニチブ(Gefinitib)およびエルロチニブ(Erlotinib)は、Toronto research Chemicals(Ontario,Canada)より購入した。抗EGFRマウスモノクローナル抗体(クローン225)およびPD153035は、Calbiochem(La Jolla,CA)より購入した。
小さな単層状小胞(SUV)を、先に記載されたように調製した(Matsuzaki,K.& Horikiri,C.Interactions of Amiloid β−peptide(1−40)with Ganglioside−containing membranes.Biochemistry 38,4137−4142(1999))。簡単に述べると、所望の組成の脂質膜を水中またはTris緩衝液(10mM Tris/150mM NaCl/1mM EDTA、pH7.4)中に分散させた。得られたMLVを5回の凍結−溶解サイクルに供し、次いで、プローブ型の超音波処理装置(Tomy UD−201)を用いて、氷冷水中、窒素雰囲気下で15分間超音波処理した。プローブのチタン先端部からの金属片を遠心分離により除いた。リン解析により、脂質濃度を三連で決定した(Bartlett,G.R.Phosphorus assay in column chromatography.J.Biol.Chem.234,466−468(1959))。
CDスペクトルは、1mm光路長の石英セルを用いて、Jasco J−820にて測定し、緩衝液成分に起因する吸収を最小限にした。各サンプルにつき、8回の走査の平均をとった。平均化したブランクスペクトル(小胞懸濁物または溶媒)を差し引いた。ペプチドおよび脂質の濃度は、それぞれ、50μMおよび4mMであった。
1ウェルあたり合計3×103細胞を、96ウェルプレートに播種し、10% FBSを含有する培地中で24時間培養し、100μlにおいて漸増濃度のペプチドと共に、37℃にて48〜72時間インキュベートした。細胞の生存率を、WST−8溶液(Cell Count Reagent SF;ナカライテスク株式会社)を用いて測定した。
フローサイトメトリーによるEGFR発現を、FITCを結合したEGFRに対するヒトモノクローナル抗体(Santa Cruz)と共に、1×105細胞をインキュベートすることにより決定した。全ての染色は、室温にて40分間行った。細胞の蛍光を、フローサイトメトリー(FACSCalibur,Becton Dickinson,San Jose,CA)により測定した。EGFR陽性細胞の平均蛍光強度(MFI)を、CellQuestソフトウェア(Becton Dickinson)を用いて決定した。
BT−20細胞を、5μMのEB−Lyticキメラペプチドを用いて、または、これを用いずに、37℃にて2時間処理した。カスパーゼの活性化またはアネキシンV陽性化を評価するために、ペプチドで処理した培養物を、カルボキシフルオレセインFLICAカスパーゼ3&7アッセイ(Immunochemistry Technologies,Bloomington,MN)を用いてカスパーゼ活性およびヨウ化プロピジウム(PI)染色について、あるいは、マルチパラメトリックフローサイトメトリーによって、アネキシンV標識およびPI染色について、同時に解析した。また、製造業者の説明書に従い、共焦点レーザ走査顕微鏡を用いて、MEBSTAIN Apoptosis Kit Direct(MBL)により、ターミナル・デオキシヌクレオチジル・トランスフェラーゼ−dUTPニック末端標識(TUNEL)アッセイを行った。
BIACOREバイオセンサーシステム3000(BIACORE Inc,Uppsala,Sweden)を用いて、表面プラズモン共鳴(SPR)実験を行った。N−ヒドロキシスクシンイミドおよびN−エチル−N’−(ジメチルアミノプロピル)カルボジイミド活性化化学により約5000RUのストレプトアビジン(Sigma)をCM5センサーチップの表面に固定し、次いで、ビオチンを結合した2000〜3000RUのペプチドを、このストレプトアビジンを固定したセンサーチップの上に注入した。非特異的な結合のコントロールとして、ストレプトアビジンを固定していないセンサーチップの反応していないカルボキシメチル基を、エタノールアミンでブロックした。分析物として、Mem−PER真核生物膜タンパク質抽出試薬キット(Pierce)を用いて調製した細胞表面タンパク質を、25℃にて20μl/分の流速のフローセルに注入した。アッセイの間に非特異的な結合を防ぐために、HBS緩衝液(0.01M HEPES、0.15M NaCl、0.005% Tween 20、3mM EDTA[pH7.4])を泳動緩衝液として用いた。組換えヒトEGFレセプター(rhEGFR)のEB−Lyticキメラペプチドとの相互作用解析を、以下のように行った:上述のように、約5000RUのrhEGFR(配列番号3)をCM5のセンサーチップ上に固定し、次いで、いくつかの濃度のペプチドをこのセンサーチップの上に注入した。これらの実験において用いた全てのタンパク質濃度は、Bradford法(Bradford MM.A rapid and sensitive method for the quantitation of microgram quantities of protein utilizing the principle of protein−dye binding.Anal Biochem 1976;72:248−54)により決定した。データの解析は、BIA evaluation ver.3.2ソフトウェア(BIACORE)を用いて行った。
H322細胞に対するLyticペプチドまたはキメラペプチドのインビトロ細胞傷害活性を、コロニー形成アッセイにより決定した。10%FBSを含むRPMI−1640(3ml)を入れた6cmディッシュに細胞を播種し付着させた。細胞/ディッシュの数は、コントロール群において、100を超えるコロニーが得られるように設定した。細胞播種から24時間後に、種々の濃度のLyticペプチドまたはEB−Lyticキメラペプチド(0〜22.5μM)を細胞に曝露させ、10日間培養した。このディッシュをリン酸緩衝液で洗浄し、クリスタルバイオレット(25%アルコール中0.25%)で染色した。コントロール群および処理群において形成されたコロニーの数から、コロニーの生存率を決定した。
(二次構造に基づいた新規キメラペプチドの設計)
これまでに、PapoおよびShaiが、ロイシンおよびリジンのD型およびL型を含む15アミノ酸のジアステレオマー配列から構成される新規溶解型ペプチドを報告している(Papo,N.& Shai,Y.New Lytic peptides based on the D,L−amphipathic helix motif preferentially kill tumor cells compared to normal cells.Biochemistry 42,9346−9354(2003))。本研究において、本発明者らは、二次構造におけるその両親媒性に基づいて、EGFR結合(EB)ペプチドと組み合わせるのに適した新規溶解型ペプチドを設計した。図1Dに示すように、新たに設計した溶解性ペプチドの正に荷電したアミノ酸(Lys)のクラスターの位置は、EBペプチドと組み合わせた後、EB−Lyticペプチドのものと比較して保存されたままであった(図1D(a,b))。CDスペクトル解析は、EB−Lyticペプチドは、正常な細胞膜表面上で最も多い脂質種であるホスファチジルコリン(PC)から構成される小さな単層状小胞(SUV)への結合が弱く、PCリポソームではあまり上手く構造化されなかったが、このEB−Lyticペプチドが、がん細胞膜上で特異的に露出されるホスファチジルセリン(PS)を含むSUVに結合し得、そして、209〜210nmおよび222nmにおける2つの極小値によって特徴付けられる部分的ならせん構造をとっていたことを実証した(図1D(d))。一方、EB−オリジナル溶解性ペプチドは、PCリポソームおよびPC/PSリポソームの両方に強く結合し得、そして、らせん構造をとった(図1D(c))。これらの結果は、本研究において新たに設計したキメラペプチドが、PSを含む膜に対して選択性を有し、そして、細胞表面上に孔をつくるために必須と考えられているらせん構造をとることを示す(Papo,N.& Shai,Y.New Lytic peptides based on the D,L−amphipathic helix motif preferentially kill tumor cells compared to normal cells.Biochemistry 42,9346−9354(2003))。
7種のEGFRを発現するがん細胞株に対して、溶解性ペプチドの細胞傷害性を、EGFR結合(EB)−LyticキメラペプチドであるEGFR標的化ペプチドトキシンの細胞傷害性と比較した。図1Aに示すように、溶解性ペプチドまたはキメラペプチドでの処理は、試験した全てのがん細胞株において濃度依存性の細胞傷害性を生じた。キメラペプチドは、溶解性ペプチドのみの場合と比較して、がん細胞に対する細胞傷害活性のかなりの増強を示した。全ての細胞株において80%を超える細胞死を誘導するのに、キメラペプチドは15〜20μMの濃度で十分であった。対照的に、同じ濃度の溶解性ペプチドのみでは、がん細胞の十分な細胞殺傷を誘導できなかった。表1に示されるように、溶解性ペプチドはEGFR標的化することにより、がん細胞におけるIC50(コントロール細胞の増殖の50%阻害を誘導するペプチドの濃度)を1.6〜3.1倍増強させ、EGFR標的化により、溶解性ペプチドのみに比してEB−Lyticキメラペプチドに対するがん細胞の感受性が増強されたことが示唆された。本発明者らは、次いで、3種の正常細胞株におけるEB−Lyticキメラペプチドおよび溶解性ペプチドの細胞傷害性を評価した。図1Bに示されるように、MRC−5、WI−38およびHEK293を含む3種の正常細胞株は、溶解性ペプチドに対する感受性が低く、がん細胞株と比較してより低い細胞傷害性を示した。正常細胞株におけるキメラペプチドのIC50は、がん細胞と比較して正常細胞において3.6〜8.0倍高い(表1)。EB−Lyticキメラペプチドは正常細胞に対しても細胞傷害性が増強されたが、これはがん細胞における現象に類似する。これらの知見から、溶解性ペプチドは正常細胞よりもがん細胞において強い細胞傷害活性を有し、さらにEGFR標的化ペプチドトキシンはEGFR高発現がん細胞に対して優れた細胞傷害活性を有することを示唆する。興味深いことに、オリジナル溶解性ペプチドと組み合わせたEBペプチドは、細胞傷害活性の増強を示さず、そればかりか、EB−オリジナル溶解性ペプチドは、より低いペプチド濃度で正常細胞株(MRC−5)までも殺傷し、オリジナル溶解性ペプチドがEBペプチドとのキメラ化に適していないことが示唆された(図1C)。
k−ras変異を有するかまたは有さないがん細胞において、EB−lyticペプチドおよびTKIの細胞傷害活性を比較した。
k−ras変異の有無による各種がん細胞のエルロチニブまたは抗EGFR抗体に対する感受性を検討した。
k−ras変異を持たないがん細胞および正常細胞に対するTKIまたはEB−lyticキメラペプチドによる細胞傷害性を検討した。
k−ras変異を持つTKI耐性がん細胞に対するEB−lyticキメラペプチドによる細胞傷害性を検討した。
本発明者らは、次に、EB−Lyticキメラペプチドの細胞傷害性における増加が、細胞表面上のEGFRの発現レベルと相関するかどうかを調べた。7種のがん細胞株および3種の正常細胞株におけるEGFRの発現レベルを、FITC結合抗EGFRポリクローナル抗体を用い、フローサイトメトリーにより評価した。図2Aおよび表1に示されるように、EGFRの発現レベルは、EB−Lyticキメラペプチドまたは溶解性ペプチドのIC50とは相関しなかった(キメラペプチドにおいてr=−0.37(図2A左)、溶解性ペプチドにおいて、r=−0.14(図2A中央))。しかし、EGFRの発現レベルは、EB−Lyticキメラペプチドに対する溶解性ペプチドのIC50の比とは十分に相関しており、溶解性ペプチドのみに比較したEB−Lyticキメラペプチドによる細胞傷害活性の増強度合が、細胞表面上のEGFR発現レベルに依存する(r=0.89、図2A右)ことが示唆された。
EGFRに対するペプチドの結合特性を理解するために、EGFRタンパク質をセンサーチップ上に固定し、EB−Lyticキメラペプチドまたは溶解性ペプチドのみとの相互作用プロファイルを、BIACOREを用いて解析した。図3Aに示されるように、共鳴シグナル強度は、EB−Lyticキメラペプチドの濃度に応じて増加し、EGFRタンパク質に結合したEB−溶解性キメラペプチドの量は、EB−Lyticキメラペプチド濃度の増加に比例することが示された。対照的に、溶解性ペプチドのみによる共鳴シグナル強度は、濃度に応じて最低限に増加した。EGFRタンパク質に結合するEB−LyticキメラペプチドのKD値は、2.6×10−5(M)であった。次に、ペプチドの細胞への結合特性を理解するために、EB−Lyticキメラペプチドまたは溶解性ペプチドのみのいずれかをセンサーチップ上に固定し、そして、H322、BT−20またはMRC−5から抽出した細胞表面膜タンパク質との相互作用プロファイルを、BIACOREを用いて解析した。図3Bおよび3Cに示されるように、共鳴シグナル強度は、細胞膜タンパク質の濃度に応じて増加し、ペプチドに結合した細胞膜タンパク質の量は、細胞膜タンパク質濃度の増加と比例することが示された。溶解性ペプチドのみに対する細胞膜タンパク質の相互作用は、ペプチドの濃度を増加させても各細胞株において同程度の結合定数を示した。一方、H322がん細胞またはBT−20がん細胞の膜タンパク質に対するEB−Lyticキメラペプチドの結合定数は、溶解性ペプチドのみよりも4.2倍(H322)または4.4倍(BT−20)強かった(図3B、3Cおよび3D)。対照的に、MRC−5正常細胞の膜タンパク質に対する結合定数は、溶解性ペプチドのみと比較して有意に異ならなかった(図3D)。これらの結果は、WSTアッセイから得られたデータ(図1)と一致しており、EB−Lyticキメラペプチドの細胞傷害活性が、細胞膜に対する親和性と十分に相関したことが示された。
EB−キメラペプチドががん細胞を殺傷する適切な持続時間を評価するために、H322細胞またはBT−20細胞を、EB−キメラペプチドまたは溶解性ペプチドのみのいずれかで、10分間、30分間、1時間、または48時間処理した。図4に示されるように、H322細胞またはBT−20細胞の溶解性ペプチドのみでの処理は、時間依存性の様式で生存性の喪失をもたらした。対照的に、H322細胞またはBT−20細胞をほんの10分間EB−キメラペプチド(10μM)に曝露するだけで、がん細胞は十分に殺傷され、70%を超える殺細胞効果を示した。共焦点顕微鏡解析もまた、このキメラペプチドが細胞膜に透過して、がん細胞表面上に孔を形成することを示し、そして、20分以内にがん細胞の細胞質へのカルセイン標識した媒体の流入が観察された(図4B)。しかしながら、この迅速な透過は、溶解性ペプチドのみでは観察されなかった(図4C)。これらの結果から、EB−キメラペプチドが、溶解性ペプチドのみと比較して、かなり迅速に細胞を殺傷することが示唆される。インビトロコロニー形成アッセイもまた、このキメラペプチドが、濃度依存性の様式でH322がん細胞の細胞増殖を阻害することを示した(図4D)。
EB−キメラペプチドにより引き起こされる細胞死の作用機構を調べるために、マルチパラメトリックフローサイトメトリー解析を用いるアネキシンVアッセイおよびカスパーゼアッセイを行った。図5Aに示されるように、BT−20乳がん細胞に対するEB−Lyticキメラペプチド(5μM)の2時間の曝露は、カスパーゼ活性化およびアネキシンV陽性化を誘導した。この結果から、EB−Lyticキメラペプチドはがん細胞の膜を崩壊させ、アポトーシス性の機構による細胞死に誘導することが示唆された。
H322がん細胞に対するEB−Lyticキメラペプチドの細胞傷害活性をさらに確認するために、コロニー形成アッセイを行った(Kawakami K,Kawakami M,Leland P,Puri RK.Internalization property of interleukin−4 receptor alpha chain increases cytotoxic effect of interleukin−4 receptor−targeted cytotoxin in cancer cells.Clin Cancer Res 2002;8:258−66;Kawakami K,Joshi BH,Puri RK.Sensitization of cancer cells to interleukin 13−pseudomonas exotoxin−induced cell death by gene transfer of interleukin 13 receptor alpha chain.Hum Gene Ther 2000;11:1829−35)。図6に示されるように、H322細胞は、溶解性ペプチドのみ(IC50、14.2μM)に対して感受性であったが、この細胞は、キメラペプチドに対して少なくとも2倍高い感受性(IC50<7.5μM)を示した。コロニー形成アッセイによる2つのペプチドのIC50値は、WSTアッセイにより決定したIC50値と十分に相関する。
この研究において、本発明者らは、本発明の「ペプチドトキシン」と呼ばれる新規キメラペプチドを生成するために、アミノ酸の2つの機能性ドメインを連結した。この「ペプチドトキシン」は、EGFRを過剰発現するがん細胞を標的化するために、EGFRに結合して細胞膜を溶解する二機能性ペプチドとして設計した。本発明者らは、EB−Lyticキメラペプチドが、溶解性ペプチドのみと比較したとき、より迅速かつ効率的にがん細胞を殺傷することを見出した。一方で、正常細胞は、両方のペプチドに対し、あまり感受性は高くないことが見出された。
本実施例では、EGF受容体結合ペプチドのアナログ(アミノ酸配列X1X2X3X4X5X6X7X8X9X10X11X12(配列番号8)を有するものであって、ここでX1は、Yまたはそれに類似するS、HもしくはFであるアミノ酸であり;
X2は、Hまたはそれに類似するRもしくはKであるアミノ酸であり;
X3は、Wまたはそれに類似するY、FもしくはHであるアミノ酸であり;
X4は、Yまたはそれに類似するS、HもしくはFであるアミノ酸であり;

上記変異体ペプチドのうち、まず電荷を有する2番目のHに着目しKとRの変異体を化学合成し、実施例1に記載されるように、BIACOREにてリコンビナントヒトEGFRとの結合親和性を野生型EGFR結合ペプチドとの比較により評価した。その結果においてより効果が高いもののみがん細胞膜溶解配列とのキメラペプチドを合成し、ヒト肺がん細胞株H322に対する殺細胞効果を野生型EGFR結合キメラペプチドとがん細胞膜溶解単独との比較をした。
結果を図7に示した。野生型(wild−type)ペプチドに比べK置換ペプチドは若干バイオセンサー応答の増強がみられ、R置換体はwild−typeの2倍の応答がみられた(図7A)。H322に対する殺細胞効果は野生型EGFR結合キメラペプチド(三角)よりもR置換体キメラペプチド(黒丸)の方が増強された(図7B)。
本実施例ではKLLLKLLKKLLKLLKKK(すべてL体アミノ酸のみ;LyticL;配列番号27)のがん細胞膜溶解性配列とIL4R標的化配列とを組み合わせたキメラペプチドで実施例1と同様な殺細胞効果増強等の効果がみられるかどうかを調べた。そのキメラペプチド配列は以下の通りである。
IL4−LyticL:KQLIRFLKRLDRNGGGKLLLKLLKKLLKLLKKK(配列番号18)。
IL4RおよびIL−4との結合立体構造解析結果から結合に重要な部分ペプチド配列(KQLIRFLKRLDRN(配列番号26))を設計し化学合成し、BIACORE解析によりヒトリコンビナントIL4Rタンパク質との結合親和性を評価した。上記キメラペプチドを合成しヒト乳がん細胞株MDA−MB−231およびヒト膵臓がん細胞株BxPC−3に対するin vitro殺細胞効果を評価した。
結果を図8に示した。IL4R結合ペプチド配列(KQLIRFLKRLDRN(配列番号26))はBIACORE解析によりヒトリコンビナントIL4Rタンパク質との結合親和性が認められた(図8A)。IL4R標的化がん細胞膜溶解性キメラペプチド(IL4−LyticL)は2種類のがん細胞株いずれにおいても、がん細胞膜溶解性ペプチド単独(LyticL)よりも殺細胞効果が増強された(図8B)。キメラ化することにより、10分間の接触でがん細胞(BxPC−3)を十分に細胞死へ誘導することが示された(図8C)。
本実施例ではKLLLKLLKKLLKLLKKK(すべてL体アミノ酸のみ;LyticL;配列番号27)のがん細胞膜溶解性配列とIL13R標的化配列とを組み合わせたキメラペプチドで実施例1と同様な殺細胞効果増強等の効果がみられるかどうかを調べた。そのキメラペプチド配列は以下の通りである。
IL13−LyticL:KDLLLHLKKLFREGQFNGGGKLLLKLLKKLLKLLKKK(配列番号19)。
IL13RとIL−13との結合立体構造解析結果から結合に重要な部分ペプチド配列(KDLLLHLKKLFREGQFN(配列番号28))を設計し化学合成し、BIACORE解析によりヒトリコンビナントIL13Rタンパク質との結合親和性を評価した。上記キメラペプチドを合成しヒト脳腫瘍細胞株U251およびヒト頭頚部がん細胞株HN−12に対するin vitro殺細胞効果を評価した。
結果を図9に示した。IL13R結合ペプチド配列(KDLLLHLKKLFREGQFN(配列番号28))はBIACORE解析によりヒトリコンビナントIL13Rタンパク質との結合親和性が認められた(図9A)。IL13R標的化がん細胞膜溶解性キメラペプチド(IL13−LyticL)は2種類のがん細胞株いずれにおいても、がん細胞膜溶解性ペプチド単独(LyticL)よりも殺細胞効果が増強された(図9B)。キメラ化することにより、10分間の接触でがん細胞(U251)を十分に細胞死へ誘導することが示された(図9C)。
本実施例ではKLLLKLLKKLLKLLKKK(すべてL体アミノ酸のみ;LyticL;配列番号27)のがん細胞膜溶解性配列とNRP1標的化配列とを組み合わせたキメラペプチドで実施例1と同様な殺細胞効果増強等の効果がみられるかどうかを調べた。そのキメラペプチド配列は以下の通りである。

NRP1とそのリガンドSema3Aとの結合立体構造解析結果から結合に重要な部分ペプチド配列(NYQWVPYQGRVPYPR(配列番号29))を設計し化学合成し、BIACORE解析によりヒトリコンビナントNRP1タンパク質との結合親和性を評価した。上記キメラペプチドを合成しヒト膵臓がん細胞株SU86.86およびヒト乳がん細胞株SKBR3に対するin vitro殺細胞効果を評価した。
結果を図10に示した。NRP1結合ペプチド配列(NYQWVPYQGRVPYPR(配列番号29))はBIACORE解析によりヒトリコンビナントNRP1タンパク質との結合親和性が認められた(図10A)。NRP1標的化がん細胞膜溶解性キメラペプチド(Sema3A−LyticL)は2種類のがん細胞株いずれにおいても、がん細胞膜溶解性ペプチド単独(LyticL)よりも殺細胞効果が増強された(図10B)。
本実施例では細胞膜溶解・核酸結合配列(RLLRRLLRRLLRK(配列番号13)、以下bufと略)とEGFR標的化配列とを組み合わせたキメラペプチドで実施例1と同様な殺細胞効果増強等の効果がみられるかどうかを調べた。そのキメラペプチド配列は以下の通りである。
EGFbuf:YHWYGYTPQNVIGGGGGRLLRRLLRRLLRK(配列番号21)。
上記キメラペプチド(EGFbuf)および爆薬部ペプチド単独(buf)を合成しヒト肺がん細胞株H322、ヒト前立腺がん細胞株DU145およびヒト肺正常細胞株MRC5に対するin vitro殺細胞効果を評価した。
結果を図11に示した。EGFR標的化キメラペプチド(EGFbuf)は2種類のがん細胞株いずれにおいても、がん細胞膜溶解性ペプチド単独(buf)よりも殺細胞効果が増強された(図11A)。またEGFbufを肺がん細胞株H322と肺正常細胞株MRC−5で殺細胞効果を比較したところ、がん細胞に殺細胞感受性が高かった(図11B)。
本実施例ではKLLLKLLKKLLKLLKKK(下線はD体、他はL体アミノ酸;配列番号1)のがん細胞膜溶解性配列と3種類のがん細胞高発現受容体結合配列(her2、VEGF受容体、Transferrin受容体)とを組み合わせたキメラペプチドで実施例1と同様な殺細胞効果増強等の効果がみられるかどうかを調べた。そのキメラペプチド配列は以下の通りである(下線はD体、他はL体アミノ酸である。)。

文献検索により上記3つの受容体結合ペプチド配列を見出し、がん細胞膜溶解性配列(KLLLKLLKKLLKLLKKK、下線D体、以下Lyticと略;配列番号1)とのキメラペプチドを設計し化学合成した。上記キメラペプチドを合成しヒト肺がん細胞株H322およびヒト肺正常細胞株MRC−5に対するin vitro殺細胞効果を評価した。
結果を図12に示した。3種類の抗がん標的化がん細胞膜溶解性キメラペプチドはいずれもヒト肺がん細胞株H322において、Lyticペプチド単独よりも殺細胞効果が増強された(図12A)。またヒト肺正常細胞株MRC−5に対するin vitro殺細胞効果は2種類のがん細胞に比べマイルドであった(図12B)。
本実施例では、他のがん細胞高発現受容体ペプチドが使用可能かを調べる。

EGFRを使用した場合と同様、BIACORE解析によりそれぞれの受容体特異的に結合親和性を確認でき、それぞれの結合配列とがん細胞膜溶解性配列とのキメラペプチドはがん細胞膜溶解性ペプチド単独に比べ殺細胞効果が増強されることが期待できる。
実施例1において製造したキメラペプチドを用いて治療効果を確認した。がん細胞に高発現している受容体等の分子に結合する弾頭部ペプチドと、がん細胞に対して強力な殺細胞効果を示す爆薬部ペプチドを結合させた抗がん標的化キメラペプチド(ペプチドトキシン)を設計し、担がん動物モデルでの抗腫瘍効果を研究したものである。
ヌードマウスbalb/c−nu/nu、雌5週齢の皮下にヒト膵臓がん細胞株BxPC−3、5×106 cells/150μl リン酸緩衝液を注入し、移植から5日後より週3回3週間でEB−Lyticキメラペプチドを0mg/kg、0.3mg/kg、1mg/kg、50μl リン酸緩衝液/マウスで腫瘍内投与した。腫瘍径を経時的に電子ノギスで計測し、長径×短径×短径×0.5として腫瘍容積(mm3)を算出した。
結果を図13に示す。
前項と同様に、実施例1において製造したキメラペプチドを用いて治療効果を確認した。本実施例では全身投与による抗腫瘍効果を調べた。

結果を図14および図15Aの上側に示す。上述の記載から明らかなように、図14および図15Aは、それぞれ独立した実験系の結果を示す。
本実施例では、実際の治療への応用を調べる。
EB−Lyticキメラペプチドをアテロコラーゲンと混合させ、局所投与用のゲル化製剤により分解抑制および徐放効果を検討する。また、全身投与用にゲル化せずにペプチド分解抑制効果を示す製剤化を検討する。そのためにin vitroでのペプチドのアテロコラーゲン混合体からの遊離プロファイル解析を行い、最適なアテロコラーゲン含有率を決定する。
実施例9と同様にヌードマウス担がんモデルに局所投与用のゲル化混合物(EB−Lyticキメラペプチドとアテロコラーゲン)を腫瘍内投与し、抗腫瘍効果を評価する。また全身投与用の混合物を静脈内投与し、同様に抗腫瘍効果を評価する。両投与方法とも用法・用量設定試験を行う。
実施例1において製造したキメラペプチドを用いて用量設定について検討する。

ヌードマウスbalb/c−nu/nu、雌5週齢の皮下にヒト乳がん細胞株MDA−MB−231またはヒト前立腺がん細胞株DU145、5×106 cells/150μl リン酸緩衝液を注入し、移植から5日後より週3回3週間でEB−Lyticキメラペプチドを0.5mg/kg、50μl リン酸緩衝液/マウスで静脈内投与する。腫瘍径を経時的に電子ノギスで計測し、長径×短径×短径×0.5として腫瘍容積(mm3)を算出する。
ヒトのがんの異種移植モデルにおいてEB−Lyticキメラペプチドの抗腫瘍効果を評価するために、インビボでの腫瘍異種移植片におけるEB−Lyticキメラペプチドの抗腫瘍活性。乳がん細胞株MDA−MB−231(5×106細胞/150μl リン酸緩衝液)を、ヌードマウスbalb/c−nu/nu、雌7〜9週齢(体重17〜21g)の側腹部領域の皮下に注入した。腫瘍が20〜60mm3の体積に達した時点で、動物を無作為に3群に分け、そして、生理食塩水(コントロール)またはEB−Lyticペプチド(2mg/kgまたは5mg/kg)を、週3回3週間(合計9回の投薬)で静脈内投与(50μl/注射)した。腫瘍径を経時的に電子ノギスで計測し、長径×短径×短径×0.5として腫瘍容積(mm3)を算出した。処置の終了時に、マウスを屠殺して、腫瘍を摘出し、ヘマトキシリン染色の後に光学顕微鏡を用いて組織学検査を行った。全ての値は、平均±SDとして表し、統計的解析は、一元配置ANOVAおよびDunnet検定により行った。差はP<0.05で有意とみなした。
本実施例では、EB(H2R)−Lyticキメラペプチドの活性を調べた。
溶解性ペプチド、野生型EB−Lyticペプチド、およびEB(H2R)−Lyticペプチドを用いて、BT20細胞に対する細胞傷害性の比較を行った。1ウェルあたり合計3×103細胞を、96ウェルプレートに播種し、10% FBSを含有する培地中で24時間培養し、100μlにおいて漸増濃度のペプチドと共に、37℃にて48〜72時間インキュベートした。細胞の生存率を、WST−8溶液(Cell Count Reagent SF;ナカライテスク株式会社)を用いて測定した。結果を図16Cに示す。
がん細胞株H322、BT−20、U251、BxPC−3、SU.86.86およびLNCaPを、種々の濃度(0〜20μM)のEB−LyticキメラペプチドまたはEB(H2R)−Lyticキメラペプチドと共に72時間培養し、そして、WST−8試薬を用いて細胞傷害活性を評価した。結果を図17Aに示す。また、正常細胞株MRC−5およびHEK293を、種々の濃度(0〜20μM)の上記ペプチドと共に72時間培養し、細胞傷害活性を評価し、そして、上記のように細胞傷害性アッセイを行った。結果を図17Bに示す。以上の結果から、新たに設計した溶解性ペプチドが、がん細胞に対する細胞傷害活性を高めるためのキメラペプチドに適していることが示された。
H322細胞を、EB−Lyticキメラペプチド(白色の柱)またはEB(H2R)−Lyticキメラペプチド(黒色の柱)で、2分間、5分間、10分間、30分間、1時間、2時間または24時間処理し、その後、ペプチドを含む培地を新しい培地と交換し、さらに24時間細胞を培養した。WST−8を用いて、細胞生存率を解析した。結果を図18に示す。以上の結果から、EB−Lyticキメラペプチドが、原形質膜に透過して、がん細胞の迅速な殺傷を誘導することが示された。
EB(H2R)−LyticとEB−Lyticのがん細胞膜を破壊する能力について検討した。
EB(H2R)−LyticとEB−Lyticのがん細胞においてアネキシンV陽性化を誘導する能力について検討した。
ヒト乳がん細胞株MDA−MB−231担がんマウスモデルにおいて、EB−LyticキメラペプチドとEB(H2R)−Lyticキメラペプチドの抗腫瘍効果について検討した。
本実施例では、TfR−Lyticキメラペプチドの活性を調べた。
TfR−Lyticキメラペプチド(THRPPMWSPVWPGGGKLLLKLLKKLLKLLKKK;配列番号17、下線はD体アミノ酸)を用いて、がん細胞に対する細胞傷害活性を検討した。
TfR−Lyticペプチドが、原形質膜に透過して、T47Dがん細胞を迅速に殺傷することを示す。T47D細胞を、TfR−Lyticキメラペプチド(黒色の柱)または溶解性ペプチド(白色の柱)で、種々の時間(1〜180分)にわたり処理し、その後、ペプチドを含む培地を新しい培地と交換し、そして、さらに72時間細胞を培養し、WST−8を用いて細胞生存率を解析した。結果を図24Aに示す。
T47D細胞およびMDA−MB−231細胞を、siRNAまたはscRNAでトランスフェクトし、トランスフェクションから4日後に、フローサイトメトリー解析により細胞における標的遺伝子のレベルを解析した(データ示さず)。WST−8試薬を用いて阻害率を評価した。アッセイは3回繰り返し、結果は、三連の測定の平均±SD(バー)として表した。結果を図25Bに示す。
T47D細胞およびPE細胞を、TfR−Lyticキメラペプチド(10μM)および溶解性ペプチド(10μM)と共に2時間インキュベートした。アネキシンV(図26A)及びカスパーゼ3&7活性(図26B)を緑色チャネルで、ヨウ化プロピジウム染色を赤色チャネルで検出し、フローサイトメトリーで解析した。TfR−Lyticキメラペプチド処理したT47D細胞では、アネキシンV陽性及びカスパーゼ3&7活性細胞を示すパネル右半分の割合が増えていた。一方で、正常細胞PEでは、TfR−Lyticキメラペプチド処理を行ってもアネキシンV陽性及びカスパーゼ3&7活性細胞の割合が増えておらず、TfR−Lyticキメラペプチドががん細胞選択的にアポトーシス性の機構によりがん細胞の死を誘導することが示唆された。
ミトコンドリア膜貫通型電位依存性蛍光色素JC−1で標識したT47D細胞を、未処理のまま(処理なし:上左パネル)か、スタウロスポリン(Staurosporine)(コントロールミトコンドリア膜電位:上右パネル)もしくは、溶解性ペプチド(下左パネル)、もしくはTfR−Lyticキメラペプチド(下右パネル)で処理し、その2時間後に、フローサイトメトリーにより、膜貫通電位を示す赤色蛍光または緑色蛍光の変化における差示的な割合を解析した。結果を図26Cに示す。
MDA−MB−231乳がん細胞を、無胸腺ヌードマウスの皮下に移植した。矢印で示すように、5日目から、生理食塩水(コントロール)またはTfR−Lyticペプチド(0.3mg/kg、1mg/kgまたは3mg/kg)のいずれかを腫瘍内注射した。各群は3匹の動物(n=3)から構成された。経時的に腫瘍径を測定した結果を図27Aに示す。
本実施例では、IL4−Lyticキメラペプチドの活性を調べた。
L体のみのIL4−LyticL(KQLIRFLKRLDRNGGGKLLLKLLKKLLKLLKKK;配列番号18)およびDL混合IL4−Lytic(D,L)(KQLIRFLKRLDRNGGGKLLLKLLKKLLKLLKKK;配列番号44、下線はD体アミノ酸)を用いて、がん細胞に対する細胞傷害活性を検討した。
A172、BxPC−3および正常細胞株HEK293からの総RNAをcDNAに逆転写し、次いで、IL−4Rα特異的プライマーを用いた定量的PCRにより検出した。グリセルアルデヒド−3−リン酸デヒドロゲナーゼ(GAPDH)を内部標準として用いた。結果を図30に示す。
PE細胞(A)、KCCT873細胞、BxPC−3細胞(B)を、IL4−Lytic(L,D)キメラペプチド(黒色の柱)または溶解性(L,D)ペプチド(白色の柱)で、2分間、5分間、10分間、30分間、1時間、または2時間処理した。ペプチドを含む培地を新しい培地と交換し、さらに72時間細胞を培養した。WST−8試薬を用いて細胞生存率を決定した。結果を図31に示す。
正常細胞であるPE細胞、およびがん細胞であるKCCT873細胞を、IL4−Lytic(L,D)キメラペプチドまたは溶解性(L,D)ペプチド(10μM)と共に2時間インキュベートし、次いで、緑色チャネルにおいてアネキシンV標識について、そして、赤色チャネルにおけるPI染色について、二色フローサイトメトリーで解析した。結果を図32に示す。数値は各四分区画における細胞の割合%を示している。
MDA−MB−231乳がん細胞を、無胸腺ヌードマウスの皮下に移植した。矢印で示すように、5日目から、生理食塩水(コントロール)またはIL4−Lytic(L,D)ペプチド(0.5mg/kgまたは2mg/kg)のいずれかを腫瘍内注射した。各群は3匹の動物(n=3)から構成された。経時的に腫瘍径を測定した結果を図33Aに示す。
本実施例では、Sema3A−nLyticキメラペプチドの活性を調べた。
新たに設計したnLyticペプチド(LLKLLKKLLKKLLKL;配列番号45、下線はD体アミノ酸)、Sema3A(aa363−377)−nLyticペプチド(NYQWVPYQGRVPYPRGGLLKLLKKLLKKLLKL;配列番号46、下線はD体アミノ酸)およびSema3A(aa371−377)−nLyticペプチド(D,L)(GRVPYPRGGLLKLLKKLLKKLLKL;配列番号47、下線はD体アミノ酸)を用いて、がん細胞に対する細胞傷害活性を検討した。
乳がん細胞を、種々の濃度(0〜20μM)のSema3A(371−377)−nLyticまたはSema3A(363−377)−nLyticと共に48時間培養し、そして、WST−8試薬を用いて細胞傷害活性を評価した。結果を図34Bおよび表6に示す。
種々の細胞を、種々の濃度(0〜20μM)のSema3A−nLytic(363−377)またはSema3A−nLytic(371−377)と共に48時間培養し、そして、WST−8試薬を用いて細胞傷害活性を評価した。アッセイは3回繰り返し、結果は、三連の測定の平均±SD(バー)として表した。結果を図34Cおよび表7に示す。
いくつかの膵臓がん細胞株(BxPC−3、CFPAC−1、Panc−1およびSU8686)および膵臓上皮細胞、ならびに乳がん細胞株(BT−20、MDA−MB−231、SKBR−3、およびT47D)におけるニューロピリン−1の発現を、RT−PCR解析により見積もった。全てのRT−PCR解析において、β−アクチンをポジティブコントロールとして用いた。結果を図36に示す。
PE細胞(上パネル)およびBxPC−3細胞(下パネル)を、Sema3A−Lyticペプチド(5μM)と共に37℃で3時間インキュベートし、6時間後に、アネキシンV標識について二色フローサイトメトリーにより解析した。結果を図37に示す。図から明らかなように、Sema3A−nLytic処理したBxPC−3細胞では、アネキシンV陽性細胞を示すパネル右半分の割合が増えていた。一方で、正常細胞PEでは、Sema3A−nLytic処理を行ってもアネキシンV陽性細胞の割合が増えておらず、Sema3A−nLyticががん細胞選択的にアポトーシス性の機構によりがん細胞の死を誘導することが示唆された。
本実施例では、新たに設計したkLyticペプチド(KLLLKLLKKLLKLLKKK;下線はD体アミノ酸、配列番号49)と、Sema3A(363−377)−kLyticペプチド(NYQWVPYQGRVPYPRGGGKLLLKLLKKLLKLLKKK;下線はD体アミノ酸、配列番号50)とを用いて、がん細胞に対する細胞傷害活性を検討した。
いくつかの膵臓がん細胞株(BxPC−3、Panc−1、SU8686およびCFPAC−1)および膵臓上皮細胞におけるニューロピリン−1の発現を、RT−PCR解析により見積もった。全てのRT−PCR解析において、GAPDHをポジティブコントロールとして用いた。結果を図39Aに示す。
NRP−1を高発現するヒト膵臓がん細胞株SU8686細胞において、Sema3A(363−377)−kLyticペプチドがアネキシンV陽性化を誘導するかどうか検討した。

実施例1において製造したキメラペプチドの安定性を検討する。
−20℃、4℃、25℃で、生理食塩中、もしくは血清中にペプチドトキシンを短期あるいは長期保存し、HPLCによる定量解析および培養がん細胞への殺細胞効果等によって、化学的、生物学的活性を評価する。
本実施例では、EGFR高発現がん細胞におけるEGFRとそのがん細胞膜の両方をターゲットとするアミノ酸配列を用いる、医薬/抗がん剤のスクリーニング方法を実証する。
EGFR高発現がん細胞(ヒト肺がん細胞株H322等)の細胞膜標品を調製し、以下のようにBIACORE解析を行う。スクリーニングされる被験薬物はビオチン化しておく必要がある。

Claims (12)
- 上皮増殖因子(EGF)受容体結合ペプチドと、細胞殺傷性ペプチドとを含むキメラペプチドであって、該EGF受容体結合ペプチドは、YRWYGYTPQNVI(配列番号9)またはYKWYGYTPQNVI(配列番号10)を有し、該細胞殺傷性ペプチドは、KおよびLのみからなる10〜20個連なったアミノ酸配列を有し、該アミノ酸は、L体、D体もしくはDL混合である細胞膜溶解性ペプチドである、キメラペプチド。
- 前記細胞膜溶解性ペプチドは、KLLLKLLKKLLKLLKKK(配列番号48)であり、該アミノ酸は、L体、D体もしくはDL混合である、請求項1に記載のキメラペプチド。
- 前記細胞殺傷性ペプチドは、KLLLKLLKKLLKLLKKK(配列番号1)であり、ここで、各アミノ酸はL体またはD体であり、下線は、D体を示す、請求項1または2に記載のキメラペプチド。
- スペーサーペプチドをさらに有する、請求項1〜3のいずれか1項に記載のキメラペプチド。
- 前記スペーサーペプチドはグリシンまたはプロリンまたはその混合物が0〜5個連なった配列である、請求項4に記載のキメラペプチド。
- 前記スペーサーペプチドはGGGである、請求項4または5に記載のキメラペプチド。
- YRWYGYTPQNVIGGGKLLLKLLKKLLKLLKKK(配列番号43)の配列を有するものであり、下線は、D体を示す、請求項1に記載のキメラペプチド。
- 請求項1、2または4〜6のいずれかに記載のキメラペプチドをコードする核酸であって、前記アミノ酸はL体である核酸。
- 請求項1、2または4〜6のいずれかに記載のキメラペプチドをコードする核酸を含むベクターであって、前記アミノ酸はL体であるベクター。
- 請求項1、2または4〜6のいずれかに記載のキメラペプチドをコードする核酸を含む細胞であって、前記アミノ酸はL体である細胞。
- 請求項1〜7のいずれかに記載のキメラペプチドを含む医薬組成物。
- 請求項1〜7のいずれかに記載のキメラペプチドを含む抗がん剤。
