Description
ギャップ結合は、直接的な細胞−細胞連絡を容易にする細胞膜構造である。ギャップ結合チャンネルが、それぞれが6個のコネキシンサブユニットから構成される2つの半チャンネル(コネキソン)から形成されている。これらのコネキシンは、その分子量に従って一般に名付けられているか、あるいは系統発生的な基準に基づいて、すなわち、αクラスおよびβクラスに分類されるタンパク質のファミリーである。
従って、第1の局面において、本発明は、治療的処置および/または整形的処置において使用される配合物であって、コネキシンタンパク質に対する少なくとも1つのアンチセンスポリヌクレオチドと薬学的に受容可能なキャリアまたはビヒクルとともに含む配合物を提供する。
上記に広く規定されているように、本発明の中心は、コネキシン発現の部位特異的なダウンレギュレーションにある。これは、コネキシン発現をダウンレギュレーションさせる部位での直接的な細胞−細胞連絡を低下させるという作用を有し、これにより、下記に記載されているように多数の治療的/整形的な適用がもたらされる。
本発明のアンチセンスポリヌクレオチド(ODNを含む)(一般に本明細書で述べる調合物の形で)はしたがって、本明細書で述べる疾病または症状のいずれかを持つヒトまたは動物などの、治療の必要なヒトまたは動物に投与される。すると、ヒトまたは動物の症状を改善することができる。ポリヌクレオチドおよび調合物はそのため、治療によるヒトまたは動物の体の処置に使用できる。これらは本明細書で述べる症状すべてを治療するための薬剤の製造に使用される。
本明細書では相同および相同体について述べる(たとえば該ポリヌクレオチドはコネキシンmRNAにおける配列の相同体でもよい)。このようなポリヌクレオチドは一般に、たとえば(相同性配列の)少なくとも15、20、40、100以上の近接ヌクレオチドの領域に渡って、関連配列と少なくとも70%は相同性であり、少なくとも80%、90%、95%、97%または99%は相同性であることが好ましい。
実験1
物質および方法
アンチセンスの適用
リン酸緩衝生理食塩水(分子級水)中の30%PluronicF−127ゲル(BASF Corp)を用いて、未修飾a1コネキシン(コネキシン43)特異性アンチセンスODNを発生中のニワトリ胚に送達した(Simons, et al., 1992)。ニワトリ胚は38℃でインキュベートし、HamiltonおよびHamburgerのステージに従って実施した。卵に窓を開け、治療する領域のvitlelineおよび羊膜は細い鉗子を用いて開いた。アンチセンスの適用の後、卵をテープで密封し、大半の実験が解析される48時間の間、インキュベータ内に置いた。ただし、a1コネキシン「ノックダウン」および回復の時間経過解析の場合は除く。
DB1は、a1コネキシン遺伝子の塩基1094−1123の対して相補性の、マウスのアンチセンス配列である。ニワトリa1コネキシン配列とは4つのミスマッチがある。CGはニワトリa1コネキシン塩基720−749に対して相補性である。このプローブの有効性は、1%のジミメチルスフホキシド(DMSO)をゲルに添加すると向上した。DMSOは他のアンチセンスODNまたは対照の結果に、追加の影響を与えなかった。
DB1(ニワトリ)は、ニワトリa1コネキシン塩基954−983に一致する、DB1のニワトリa1コネキシン相当物である。しかし解析は、ステムループ構造(G=−7.0kcal/mol,ループTm=92°)の形成およびホモ二量体化(Tm=1.5°)が高い確率で生じることを示しているため、対照配列として作用する。一部のセンスオリゴヌクレオチドが、転写を阻害する安定なDNAトリプレットを形成可能であることが報告されている(Neckers et al., 1993)。しかし、これはDB1(センス)では明確ではなかった。安定な第2構造(G=1.4kcal/mol)を持たず、不安定なホモニ量体化が生じるランダム対照配列も使用され、CV3と呼ばれた。DB1の追加の対照適用同濃度混合物およびDB1(センス)は、欠失のバックグラウンドレベルを与えた。
細胞−細胞界面におけるalコネキシンギャップ結合タンパク質の免疫組織化学局在化は、アンチセンス効果の直接測定値を与える。アンチペプチドa1コネキシン特異性抗体プローブを用いて、ホールマウント胚を染色し、コネキシン分布は、確立された手順(Green et al.,1995)に従って、共焦レーザー走査顕微鏡法を用いて解析した。発生ニワトリ胚で発現される他の2つのコネキシン(コネキシンb1およびb2)の対照標識付けも、配列特異性抗体を使用して同様に実施した(Becker et al.,1995)。
a1コネキシン発現の低下
未修飾a1コネキシン特異性アンチセンスODNを発生ニワトリ胚に送達するためにPluronic F−127ゲルを使用すると、タンパク質発現が選択された時点で阻害され、ニワトリ胚の特定領域を標的としたアンチセンス治療が可能となる。比較的低濃度でアンチセンスを含むゲルの小滴を各胚に正確に滴下した。ゲルは凝固し、少なくとも12時間は定着したままであるため、この領域では低用量のアンチセンスが持続した。アンチセンス適用は、肢、神経管および顔における上昇発現の前に結合を抑止するために、標的化および計時された。これらの時間は、標的組織の細胞膜中にすでに存在するタンパク質のターンオーバーに依存するのではなく、新たなタンパク質の発現を低下させることによってアンチセンスの効果を最適化するために選択した。DB1およびCG1 ODNの両者が、2時間以内に神経管および肢芽内のa1コネキシンタンパク質の発現を低下させ、4〜8時間内に劇的に低下させ、18〜24時間および48時間では一部の組織で持続していた(データは示さない)。a1コネキシンタンパク質のダウンレギュレーションは、使用したいずれの対照においても明確ではなかった。同様に、ニワトリ胚で発現したコネキシン族の2つの要素、b1コネキシンおよびb2コネキシンは、a1コネキシン特異性アンチセンスODNによって影響を受けなかった。
概要
星状細胞は、哺乳類の脳において最も豊富な細胞種を構成している。これらは主としてコネキシン43より成るギャップ結合によって、相互におよびニューロンに幅広く結合する(Giaurme and McCarthy(1996))。誘発された虚血または物理的脳損傷の後、これらのチャネルは開いたままで、(上昇した組織内カリウムならびにグルタミン酸およびアポトーシス信号によって開始される)機能低下の拡張波が伝播される(Cotrina et al.,(1998);Lin et al(1998))。上昇したサイトゾルカルシウムの波およびIP3などの第2メッセンジャー分子は、ギャップ結合チャネル経由で損傷領域の核を越えてゆっくりとニューロンに広がり、攻撃の24〜48時間後に損傷への拡張が起きる。このような方法で、損傷していない隣接細胞は破壊され(Lin et al., 1998)、これがいわゆるバイスタンダー効果である。
オリゴヌクレオチドは以下の配列によって調製した。
オリゴヌクレオチド(ODN)
未修飾ODNは、リン酸緩衝生理食塩水(PBS)中のPluronic F−127ゲル(BASF)によって送達した。Pluronicゲルは低温(0−4℃)にて液体であり、生理温度にて凝固し、弱い界面活性剤でもある。未修飾ODNは通常、細胞内での半減期が約20分であるが(Wagner 1994)、Pluronicゲル装填法によって連続拡散源が与えられ、ゲルがリザーバーとして作用する(Becker et al.,(1999))。コネキシン43に対して特異性のODNまたは同様の塩基組成の対照ランダムODNは、2mMの最終濃度にて適用した。ゲルのみの対照も実施された。ODNは30merであり、解析によってヘアーピンループまたはホモ二量体化が生じないことが示された。
脳障害は250〜300gのオスのウィスターラットで実施した。動物は酸素中の1〜2%ハロセンによって麻酔をかけ、頭は定位固定クランプで保持した。障害部位周辺の領域は剪毛し、頭骨上の皮膚はメスを用いて矢状断面で切開し、頭骨板を露出させておくために引き戻した。Arlecエングレーバーを用いて前頂の右側に直径0.5mmの穴を頭骨板3mmに開け、マイクロメータステージに結合された19G1 1/2ゲージシリンジを用いて、脳の皮質内に窓外を作成した。ステージによって正確な方向制御と、障害を脳梁のはるか上の皮質内に維持する、正確な2mmの貫入深さが可能となる。
動物はNembutal(ペントバルビトンナトリウム、Virbac)を用いて屠殺し、断頭した。脳を損なわないように取り出し、ドライアイスでただちに凍結させ、切断準備ができるまで−80℃で保存した。連続凍結切片(30mm切片)を前部から後部(冠状面)まで採取し、乾燥した切片をクロム明礬処理スライドに装着し、組織化学または免疫組織化学用に−80℃で保存した。障害の中間切片が明確に識別されるように、各障害の最初と最後の切片を記録した。
ヘマトキシリンおよびエオシン染色するため、切片を順次濃度を下げたアルコール(無水、2×95%、1×70%、水)で水和させ、Gillのヘマトキシリンで4分間染色した。次に切片を水で洗浄し、Scott水に浸漬し、再度水で洗浄した。次にMooreの緩衝エオシンで30秒間染色した。切片を再度水で洗浄してから、一連のアルコール(2×95%、1×無水)、50:50のアルコール:キシロールで脱水し、キシレンに浸漬した。さらに切片をHistomounta封入剤を用いて標本とした。
最初に、凍結切片をPBS中で室温に戻した。次にメタノール中で2分間透過化処理し、PBSですすいで、0.1Mリジンおよび0.1%Triton−XのPBS溶液中に移して、30分以上ブロッキングした。次にPBSで2分間、2回洗浄した。PBSを除去し、切片当たり50mlの一次抗体を加えた。

Leica TCS 4D共焦レーザー走査顕微鏡を用いて、免疫蛍光標識を実施した。次いで、Leica Combine機能を用いて、またはAdobe Photoshopによって二重標識画像を組み合わせた。ヘマトキシリンおよびエオシン、Nissl染色サンプル、またはNeuronal−N標識切片は、Kontron(Zeiss)Progress 3008デジタルカメラを用いてキャプチャーし、障害領域はMetaMorph(Universal Imaging Corp)を用いて解析した。障害領域は各障害の中間切片で解析した。
我々の対照ゲル実験、すべての障害、対照およびアンチセンス処理において、外傷発生から最初の24〜48時間における詳細に記録された脳障害の拡大は、ゲルの装填後、ゲルが停止しにくい外縁付近で広がる傾向を見せた。しかし、対照障害は脳梁に向かって下方および側面に広がって、でこぼこに広がった縁を形成した(図1および2)。Neuronal−N抗体標識組織を調べると、障害縁の十分後ろからニューロンの死が起こっており、Nissl染色領域では生存しているニューロンが残っていないことが明らかになった。この拡大は主として24時間以内に発生し(図1および2)、障害後48時間まで続いた。このことは、図2で特に明確である。図2では、24時間以内でニューロンの死が、障害縁の十分後ろから別の正常に見える組織にかけて明らかであり、障害はすぐ下の脳梁へ広がっている。これに対して、コネキシン43アンチセンス処理した、より良い障害は、元の障害部位に限定されたままであり、基準面が明確に限定されている(図3および4)。Neuronal−N標識はNissl染色組織と共存し、いずれのコネキシン43アンチセンス処理した障害も脳梁へは広がらない。Neuronal−N標識は、元の針路障害のすぐ縁までニューロンが生存していることを示している。これらの障害周囲の生存ニューロンは、針路障害の縁を示す明確な境界を規定することが多い(図3および5)。アンチセンス処理後は、障害自体の内部でさらに多くの組織が生存している;対照障害では、細胞死は障害区域内の組織損失を引き起こす(図2の24時間後の対照障害と、図3および4の48時間後のアンチセンス処理障害を比較)。
Pluronicゲル栓−アンチセンスODN法を用いて、哺乳動物の脳の大脳皮質における障害後に発生する星状細胞増加時の、コネキシン43ノックダウンの効果を調査した。脳において、瀕死のニューロンによる毒素放出によって、バイスタンダー効果として知られる影響が生じ、毒素がギャップ結合チャネルを通じて近隣細胞に広がる(Lin et al.,(1998))。神経変性条件下で、毒素をゆっくりと放出させると、明らかに星状細胞におけるコネキシン43チャネルのアップレギュレーションが生じ、血流への毒素の輸送および血流からの毒素の除去が可能となる。しかし重篤な外傷の場合は、このアップレギュレーションが高濃度の毒素が近隣のニューロンに広がるのを促進するため、近隣のニューロンは死に至る。コネキシン43アップレギュレーションおよびコネキシン43チャネルのノックダウンを阻止すると、この広がりが阻害され、障害が断面積で最大50%に縮小する。このことは、虚血性脳卒中の処置、神経変性疾病の治療、外科的介入による副作用の調整に多大な意味を持つ。
概要
損傷ニューロンが放出する毒素による神経組織におけるバイスタンダー効果が近隣細胞に広がり、それを死に至らしめることは詳細に記録されている。実験2は、ギャップ結合タンパク質コネキシン43をノックダウンするアンチセンスオリゴデオキシヌクレオチドの持続性放出手法を用いて、脳におけるこの効果を低下させられることを示している。
オリゴヌクレオチドは以下の配列を用いて調製した。
ウィスターラットに麻酔をかけ、脊髄を露出させた。脊髄に標準二等分障害を作成し、コネキシン43に対するアンチセンスまたはセンスODN(5mM)のいずれかを含む冷却Pluronicゲル5mlを障害中に配置した。適用は盲目的に行った。次に露出した脊髄を元に戻し、ラットをケージに戻した。一部の動物は24時間後に屠殺したが、残りの動物は12日間および2ヶ月間維持し、ニューロン再生の程度と障害の最終サイズを判定した。軸索再生実験では、ラットに麻酔をかけ、脊髄への入口部位の前で軸索を切断した。セイヨウワサビペルオキシダーゼ(HRP)のペレットを切断部に配置して、軸索を24時間に渡って逆標識した。翌日ラットを屠殺し、その脊髄を取り出し、2%パラホルムアルデヒドで固定した。次に脊髄を細胞切断用に処理し、索軸から連続した縦方向8mmの切片を取り出した。次いで切片を、コネキシンまたはGFAP用にヨウ化プロピジウムを核マーカーとして用いて免疫染色するか、HRPを示すように処理した。
障害の24時間後、コネキシン43およびアンチセンスODNで処理した脊髄には著しい相違が見られた。センス障害は未処理対照との相違は示さなかったが、これに対してアンチセンス処理障害はより小さく、炎症が少ないように見えた(図9)。
本発明の調合物を使用すると、コネキシン43のアンチセンスオリゴデオキシヌクレオチドノックダウンによって、脊髄損傷の最初の24〜48時間後に発生する障害拡張が著しく低減される。コネキシン43のノックダウンも炎症を低減し、さらに神経保護効果を促進するが、ニューロンが障害部位で再増殖する能力は変化しなかった。したがって、コネキシン43特異性オリゴヌクレオチドによるアンチセンス処理は、損傷ニューロンの再増殖を促進できないが、損傷の拡大を低減する著しい神経保護効果を備えている。
概要
皮膚創傷を修復するために、繊維芽細胞、内皮細胞およびケラチノサイトなどの多数の細胞種が活性化され、細胞外基質を増殖、移動および固着させて創傷を満たす。
オリゴヌクレオチドは以下の配列を用いて調製した:
CD1系統の新生マウスにスプレーによる局所麻酔で麻酔をかけた。次に長さ2mmの清潔な切開創傷を前脚に沿って虹彩刀で作成した。解剖顕微鏡の下で該創傷を作成することによって、大きさが非常に再現しやすくなる。創傷は通常3〜6日で治癒する。後の時点で創傷部位を引き続き確認するための印として、炭素粉を創傷に振りかけた−どちらにしても、炭素粉は治癒には影響を与えない。次に、センスまたはアンチセンスODNを含む、冷却した5mlのPluronicゲルを創傷に塗布した。Pluronicゲルは0〜4℃では液体であるが、それ以上の温度では凝固する。創傷に塗布すると、ゲルはその場で凝固し、ODNの徐放リザーバーとしてはもちろん弱い界面活性剤として作用し、ODNが組織に浸透するのを助ける。センスODNの塗布は片脚に行い、もう一方にはアンチセンスを塗布し、同腹仔間で左右交互にする。新生マウスはランプの下で温めた後、母親に返した。創傷は毎日検査し、治癒品質について評価した。手術の1日後、5日後および8日後に代表となる新生マウスを選択し、その前肢の写真を撮ってから、麻酔をかけ、2%パラホルムアルデヒドを振りかけた。前肢を取り外し、2%パラホルムアルデヒド中で一晩浸漬固定し、その後、樹脂(1日)またはワックス(その後2日間)組織学用に処理した。
肉芽は治癒結合組織の特徴であり、多数の毛細血管の浸潤によって囲まれる。マクロファージは、VEGFなどの強力な血管形成因子を発現することで知られている。血管新生の程度は、VEGFレセプターに対する抗体である、抗−PCAMおよび抗−flt−1を用いて監視される。この組織の収縮は、創傷繊維芽細胞が収縮性筋線維芽細胞に分化することによって引き起こされる。創傷を引き合わせた後、アポトーシスによって死に、マクロファージによって除去される。これらの細胞は、平滑筋アクチン特異性抗体によって明らかにされ、次にその形成および除去が行われる。
知覚神経は、損傷時に放出される信号に非常に敏感であり、成人の創傷部位では一過性発芽を示す。しかし新生創傷では、この発芽はさらにおびただしく、永久の過神経支配を生じる。これらの信号が何であるか明らかではないが、炎症性マクロファージから放出されると思われる。過神経支配は創傷7日後に極大であり、神経分布は神経フィラメントに対するPGP9.5抗体を用いて明らかになる。
1日
創傷の24時間後、センスおよびアンチセンス処置肢で著しい相違が明らかであった。センス処理創傷は、未処理と違いがないように見え、正常な治癒グレードおよび速度のスペクトルが得られた(図10)。アンチセンス処理肢は、対照とは著しく異なり、炎症が少ないように思われ、治癒速度が一般に速かった。
創傷の数日後には、かさぶたが落ち始めた。この段階では、アンチセンス処理創傷の大半がセンス処理よりも小さく見え、かさぶたが小さいか、瘢痕がより目立たないかのどちらかであった(図12)。
創傷の8日後、肢に体毛が生えた。センス処理創傷はまだ見えており、創傷部位周辺に体毛jがないために境界が定められていた。アンチセンス処理創傷はほとんど見えず、正常な体毛成長によって覆われていた。体毛成長におけるこのような相違は、アンチセンス処理創傷では瘢痕の発生が減少することを示している(図13)。
コネキシン43アンチセンスODNを創傷に塗布すると、治癒プロセスに著しい影響が見られる。最初の顕著な効果は、好中球のレベルによってはるかに低い炎症反応を示す部分にて顕著である創傷の炎症が減少することである。治癒が進むにつれ、アンチセンス処理創傷はより速く治癒し、対照障害よりも瘢痕が少ない。
概要
火傷後の炎症および第2細胞死は、主な関心事である。身体の高い割合を占める重篤な火傷の犠牲者は、外傷後1または2日で死に至ることが多い。本実験は、本発明の調合物の火傷回復プロセスに有利な影響を及ぼす能力を調査する。
オリゴヌクレオチドは以下の配列を用いて調製した:
再現性がよい火傷は湿った皮膚にもたらされ、アンチセンスODNを含むPluronicゲルは火傷に皮下注射した。一連の火傷は、6匹の新生マウスの頭蓋の左側と右側にはんだごてを用いて作成した。頭の片側の火傷はPluronicゲルに含まれるコネキシン43−特異性ODNによって、反対側はPluronicゲルに含まれるセンス対照ODNによって処理した。
24時間後、6匹すべてのコネキシン43ODN処置火傷は、対照火傷と比較して低レベルの炎症を示した。これらの相違は顕著であった(データは示さない)。
そのため本発明により、細胞−細胞伝達が一時的で、部位特異的な方法でダウンレギュレート可能な調合物が与えられる。したがって、該調合物は治療方法における用途と化粧処置における用途がある。
Claims (14)
- ヒトまたは動物の創傷を治療する医薬の製造方法であって、薬学的に許容可能なキャリアとコネキシン43アンチセンスポリヌクレオチドを配合して前記創傷の治癒を促進する、前記製造方法。
- 前記薬学的に許容可能なキャリアが、ゲル、ラノリン、硬パラフィン、軟黄色パラフィンまたは軟白色パラフィン、白蜜ろう、黄蜜ろう、セトステアリルアルコール、セチルアルコール、ジメチコーン、乳化ワックス、ミリスチン酸イソプロピル、微結晶ワックス、オレイルアルコールおよびステアリルアルコールから選択される、請求項1に記載の方法。
- 前記コネキシン43アンチセンスポリヌクレオチドが、配列番号1の配列、配列番号1の配列と少なくとも90%の同一性を有する配列、配列番号2の配列、配列番号2の配列と少なくとも90%の同一性を有する配列、配列番号3の配列及び配列番号3の配列と少なくとも90%の同一性を有する配列からなる群から選択される配列を含む請求項1または2に記載の方法。
- 前記ゲルが持続放出ゲルである請求項2または3に記載の方法。
- 前記ゲルがポリオキシエチレン−ポリオキシプロピレン共重合体ゲルである請求項4に記載の方法。
- 前記ゲルがプルロニックゲルである請求項5に記載の方法。
- 前記ゲルがプルロニックF−127である請求項6に記載の方法。
- 前記ポリヌクレオチドが未修飾ホスホジエステルオリゴマーである請求項1〜7のいずれか一項に記載の方法。
用。 - 前記ポリヌクレオチドが修飾ホスホジエステルオリゴマーである請求項1〜7のいずれか一項に記載の方法。
- 前記ポリヌクレオチドが、ホスホロチオ酸オリゴヌクレオチド、メチルホスホン酸、ホスホルアミド酸、ホスホロジチオ酸、N3’P5’−ホスホルアミド酸、オリゴリボヌクレオチドホスホロチオ酸、2’−O−アルキル類似体、2’−O−メチルリボヌクレオチドメチルホスホン酸および混合主鎖オリゴヌクレオチド(MBO)のいずれかで化学修飾されている、請求項9に記載の方法。
- 前記コネキシン43アンチセンスポリヌクレオチドを注射で投与する請求項1〜10のいずれか一項に記載の方法。
- 前記医薬が前記アンチセンスポリヌクレオチドの継続的な放出が得られるように配合されている、請求項1〜11のいずれか一項に記載の方法。
- 前記創傷が外傷、手術または火傷の結果である、請求項1〜12のいずれか一項に記載の方法。
- 前記創傷が脳、脊髄または視神経の損傷である、請求項1〜13のいずれか一項に記載の方法。
