Description
Cameron et al (1998)は、糖の1,2−プロパンジオールへの変換に関して代謝操作するためのプラットフォームとしての使用を検討した。彼らの理論分析は、現実的な生成物収率の上限(質量バランスと増殖のためのエネルギーの生産を考慮)は培養条件によって著しく異なる。嫌気性条件下では、減少した補因子を再循環させるために副生成物として酢酸が生成され、最良の収率はグルコース1モル当たり1,2−プロパンジオール1モル(0.42g/g)に限定されるはずである。好気性条件下では、補因子の再循環は末端電子受容体として酸素を用いる呼吸鎖によって確保されるはずであり、副生成物を生じずに1,2−プロパンジオールを生産することが可能となる。これらの条件下では、収率は良くて1.42mol/mol(0.6g/g)に達し得る。Cameron et alは、1,2−プロパンジオールの最大力価を考慮して、生成物および副生成物の毒性へのその依存を考察した。1,2−プロパンジオールは1,3−プロパンジオールよりも著しく毒性が低く、大腸菌(E. coli)は100g/l 1,2−プロパンジオールで、0.5/時の増殖速度の残存を示す。この増殖の阻害は、増殖阻害性が高いことが知られている副生成物の酢酸によるものである可能性が高い。高い力価および収率で1,2−プロパンジオールを製造するための嫌気性プロセスの開発では、この酢酸の問題に取り組まなければならない。酢酸の、阻害性がより低く、in situで容易に除去できるアセトンへの変換が提案されている(WO2005/073364)。
次に、1,2プロパンジオールの生産率が高まった(少なくとも20%高まった)進化株を選択および単離すること、
その後、その進化株において機能的tpiA遺伝子を再構築すること
を含む。
a)代謝がロックされた始原株を得るための微生物の改変(その始原株の細胞を適切な培地で増殖させた際に進化が所望の方向にのみ向くことができる)、
b)それを進化させるための、適切な培地での上記で得られた始原株の増殖(始原株は好気性、微好気性または嫌気性条件下で増殖される)、
c)所望の化合物に対して改良された生産特徴を示す、これらの特定の条件下で増殖可能な「進化株」の選択
を含んでなる。この進化プロセスは、同じ出願者らによる、2004年2月17日出願の特許出願WO2004/076659および2005年12月1日出願のWO2005/073364に包括的に記載されている。

・メチルグリオキサールからのラクトイルグルタチオンの合成を触媒するグリオキサラーゼをコードする遺伝子、例えばグリオキサラーゼIをコードするgloA遺伝子、
・ラクトアルデヒドデヒドロゲナーゼ((S)ラクトアルデヒドからの(S)乳酸の合成を触媒する)をコードするaldAおよびaldB遺伝子
である。
・内因性プロモーターのより強力なプロモーターでの置換
・目的遺伝子を有する発現ベクターの、微生物への導入
・目的遺伝子の付加的コピーの、染色体への導入
などの種々の方法を知っている。
・その遺伝子への、この遺伝子の発現レベルまたはコードされているタンパク質の活性のレベルを低下させる突然変異の導入
・その遺伝子の天然プロモーターの、より低い発現をもたらす強度の低いプロモーターでの置換
・対応するメッセンジャーRNAまたはタンパク質を脱安定化するエレメントの使用
・発現が全く望まれないならば、その遺伝子の欠失
がある。

2−前記始原株の進化

4−gapA遺伝子の減弱;pykAおよびpykF遺伝子の欠失;ppsA遺伝子の過剰発現
5−ackA−pta、poxB遺伝子の欠失
6−得られた数株の、好気性条件下での1,2−プロパンジオール生産に関する比較
7−最良の株を用いた流加培養での1,2−プロパンジオールの生産
a)改変大腸菌株MG1655 lpd*,ΔtpiA,ΔpflAB,ΔadhE,ldhA::km,ΔgloA,ΔaldA,ΔaldB,Δeddの構築
プロトコール1に従い、大腸菌株MG1655 lpd*,ΔtpiA,ΔpflAB,ΔadhE,ldhA::km,ΔgloA,ΔaldA,ΔaldB,Δedd::cm(WO2005073364参照)においてクロラムフェニコール耐性カセットを除去した。
クロラムフェニコールおよび/またはカナマイシン耐性カセットを以下の技術に従って除去した。クロラムフェニコールおよび/またはカナマイシン耐性カセットのFRT部位にFLPレコンビナーゼ作用を有するプラスミドpCP20をエレクトロポレーションによりこの株へ導入した。42℃で連続培養した後、これらの抗生物質耐性カセットの欠損を、表1に示されたオリゴヌクレオチドを用いたPCR分析により確認した。
カナマイシン耐性カセットを除去するため、およびldhA遺伝子を不活性化するために、プロトコール2に従い、関連遺伝子の大部分を欠失したldhA遺伝子にクロラムフェニコール耐性カセットを挿入した。
遺伝子または遺伝子間領域の置換のために選択され、表2に示されたオリゴヌクレオチドを用い、プラスミドpKD3由来のクロラムフェニコール耐性カセットまたはプラスミドpKD4由来のカナマイシン耐性カセットを増幅した(Datsenko, K.A. & Wanner, B.L. (2000))。次に、得られたPCR産物を、そこで発現したλ Red(γβ,exo)系が相同組換えに極めて有利である、プラスミドpKD46を担持するレシピエント株にエレクトロポレーションにより導入した。その後、抗生物質耐性形質転換体を選択し、その耐性カセットの挿入を、表1に示された適切なオリゴヌクレオチドを用いた分析により確認した。
大腸菌株MG1655において、プロトコール2に記載の技術を表2に示されたオリゴヌクレオチドとともに用い、カナマイシン抗生物質耐性カセットを挿入し、関連遺伝子の大部分を欠失させることにより、arcA遺伝子を不活性化した。得られた株を大腸菌MG1655 ΔarcA::kmと命名した。
レシピエント大腸菌における、その遺伝子を耐性カセット(カナマイシンまたはクロラムフェニコール)で置換することによる選択された遺伝子の欠失は、ファージP1を用いた形質導入技術によって行った。プロトコールは、(i)単一の遺伝子が欠失したMG1655株でのファージ溶解液の作製、および(ii)このファージ溶解液によるレシピエント株の形質導入の二段階であった。
・10mlのLB+Cm 30μg/ml+グルコース0.2%+CaCl2 5mMでの、単一の遺伝子が欠失したMG1655株の一晩培養物100μlを播種する。
・振盪しながら37℃で30分インキュベーション。
・野生株MG1655で作製したファージP1溶解液100μl(およそ1×109ファージ/ml)を加える。
・37℃で3時間、全ての細胞が溶解するまで振盪する。
・200μlのクロロホルムを加え、ボルテックスにかける・
・4500gで10分の遠心分離を行い、細胞残渣を除去する。
・上清を無菌試験管に移し、200μlのクロロホルムを加える。
・この溶解液を4℃で保存する。
形質導入
・LB培地中、大腸菌レシピエント株の一晩培養物5mlを1500gで10分遠心分離する。
・この細胞ペレットを2.5mlのMgSO4 10mM、CaCl2 5mM中に懸濁させる。
・対照試験管:細胞100μl
単一の遺伝子が欠失したMG1655株のファージP1 100μl
・試験管試験:細胞100μl+単一の遺伝子が欠失したMG1655株のファージP1 100μl
・振盪せずに30℃で30分インキュベーション。
・各試験管に1Mクエン酸ナトリウム100μlを加え、ボルテックスにかける。
・1mlのLBを加える。
・振盪しながら37℃で1時間インキュベーション。
・試験管を7000rpmで3分遠心分離した後、ディッシュ上、LB+Cm30μg/mlでプレーティング。
・37℃で一晩インキュベーション。
プロトコール2に記載の技術を表2に示されたオリゴヌクレオチドとともに用い、カナマイシン抗生物質耐性カセットを挿入し、関連遺伝子の大部分を欠失させることにより、ndh遺伝子を不活性化した。得られた株を大腸菌MG1655 Δndh::kmと命名した。
大腸菌株MG1655 lpd*,ΔtpiA,AplfAB,ΔadhE,ΔldhA,ΔgloA,ΔaldA,ΔaldB,Δedd,ΔarcAにおける、その遺伝子をカナマイシン耐性カセットで置換することによるndh遺伝子の欠失は、これまでのように、プロトコール3に記載のファージP1を用いた形質導入技術を用いて行った。
a)改変大腸菌株MG1655 tpiA::kmの構築
プロトコール2に記載の技術に従い、表2に示されたオリゴヌクレオチドを用い、tpiA遺伝子の上流にカナマイシン抗生物質耐性カセットを挿入した。得られた株を大腸菌MG1655 tpiA::kmと命名した。
a)天然gapAプロモーターの、合成ショートPtrc16プロモーターでの置換
「進化型大腸菌tpiArc」株における天然gapAプロモーターの、合成ショートPtrc16プロモーター(配列番号52:gagctgttgacgattaatcatccggctcgaataatgtgtggaa)での置換は、225pbの上流gapA配列をFRT−CmR−FRTおよび操作プロモーターで置換することにより行った。この技術はプロトコール2に記載され、表2に示されたオリゴヌクレオチドを用いる。得られた株を「進化型大腸菌tpiArc」Ptrc16−gapAと命名した。
pykA遺伝子は、プロトコール2に記載の技術を表2に示されたオリゴヌクレオチドとともに用い、カナマイシン抗生物質耐性カセットを挿入し、関連遺伝子の大部分を欠失させることにより不活性化した。得られた株を「進化型大腸菌tpiArc」Ptrc16−gapA ΔpykA::kmと命名した。
pykF遺伝子は、プロトコール2に記載の技術を表2に示されたオリゴヌクレオチドとともに用い、カナマイシン抗生物質耐性カセットを挿入し、関連遺伝子の大部分を欠失させることにより不活性化した。得られた株を「進化型大腸菌tpiArc」Ptrc16−gapA,ΔpykA,ΔpykF::kmと命名した。
ホスホエノールピルビン酸の生産を高めるために、ppsA遺伝子を、gapAプロモーターを用いてプラスミドpJB137から発現させた。プラスミドpJB137−P gapA−ppsAの構築のため、ppsA遺伝子を、以下のオリゴヌクレオチドを用いて大腸菌MG1655のゲノムDNAからPCR増幅した。

a)改変大腸菌株MG1655 ΔackA−pta::cmの構築
プロトコール2に記載の技術を表2に示されたオリゴヌクレオチドとともに用い、クロラムフェニコール抗生物質耐性カセットを挿入し、関連遺伝子の大部分を欠失させることにより、ackAおよびpta遺伝子を不活性化する。得られた株を大腸菌MG1655 ΔackA−pta::cmと命名する。
「進化型大腸菌tpiArc」Ptrc16−gapA,ΔpykA,ΔpykF株におけるackAおよびpta遺伝子の欠失を、前記のように、プロトコール3に記載のようなファージP1を用いた形質導入技術を用いて行う。
プロトコール2に記載の技術を表2に示されたオリゴヌクレオチドとともに用い、クロラムフェニコール抗生物質耐性カセットを挿入し、関連遺伝子の大部分を欠失させることにより、poxB遺伝子を不活性化する。
実施例4に記載のように得られた株と対照株(対照1:嫌気性条件下で進化したMG1655 lpd* ΔtpiA ΔpflAB ΔadhE ΔldhA::Cm ΔgloA Δald,ΔaldB Δeddおよび対照2:微好気性条件下で進化したMG1655 lpd* ΔtpiA ΔpflAB ΔadhE ΔldhA::Cm ΔgloA Δald,ΔaldB Δedd)を、エルレンマイヤーフラスコアッセイにて、好気性条件下、酵母抽出液および炭素源としてのグルコースを添加した最小培地で培養した。培養は34℃で行い、培養培地をMOPSで緩衝させることによりpHを維持した。培養の終了時に、発酵液中の1,2−プロパンジオール、アセトールおよび残留グルコースをHPLCにより分析し、グルコースに対する1,2−プロパンジオールおよびグルコースに対する1,2−プロパンジオール+アセトールの収率を算出した。その後、最良の株を発酵槽での流加培養のために選択した。
これまでの実験で選択された最良の株を、流加プロトコールを用い、21発酵槽で培養する。
Claims (5)
- 炭素源から1,2−プロパンジオールを製造するための進化した大腸菌株を製造する方法であって、
始原株を、増殖培地中で増殖させること、
次に、1,2−プロパンジオールの生産率が高まった進化株を選択および単離すること、
その後、その進化株に機能的tpiA遺伝子を導入すること
を含んでなり、前記始原株が、始原株において進化を促進するための、tpiA遺伝子の発現の減弱、およびメチルグリオキサールの乳酸への変換に関与する少なくとも1つの遺伝子の発現の減弱を含むものである、方法。 - メチルグリオキサールの乳酸への変換に関与する遺伝子が、gloA、aldA、aldBおよびそれらの組合せからなる群から選択される、請求項1に記載の方法。
- 始原株が、ldhA、pflA、pflB、adhE、eddおよびedaからなる群から選択される少なくとも1つの遺伝子の発現の減弱をさらに含む、請求項1または2に記載の方法。
- 始原株が、arcAおよびndhからなる群から選択される少なくとも1つの遺伝子の減弱をさらに含む、請求項1〜3のいずれか一項に記載の方法。
- 1,2−プロパンジオールを製造するための方法であって、請求項1〜4のいずれか一項に記載の方法によって得られた進化株が炭素源を含有する増殖培地で増殖され、生成した1,2−プロパンジオールが回収される、方法。
