本発明は、概して、免疫応答(抗原に対する免疫応答など)を生じさせるかまたは増強する方法、該方法を実施するための組成物、ならびにこのような方法に有用な改変免疫細胞に関する。
免疫欠損を特徴とする、あるいは、より攻撃的な免疫応答により消散可能な疾患および症状に対処すべく宿主の免疫応答を増強するためのアプローチが記載されている。このようなアプローチが有利となりうる疾患または症状の例として、癌、インフルエンザおよびヒト免疫不全ウイルス(HIV)などが挙げられる。

本発明は、抗原、例えば、TAA、またはヒトおよびトリインフルエンザもしくはHIVのような感染因子に関連する抗原に対する免疫応答などの免疫応答を生じさせるかまたは増強する方法および組成物を提供する。本発明はまた、抗原に対する免疫応答を生じさせるかまたは増強するのに有用な改変免疫細胞も提供する。本発明はまた、免疫療法も提供し、このような方法として、腫瘍サイズを縮小する方法および腫瘍細胞の増殖を阻害する方法のような癌免疫療法、ならびに感染症を治療または予防する方法などが挙げられる。
免疫共刺激分子は、ナイーブT細胞と抗原提示細胞との自然相互作用に関与し、その結果、それらの相互活性化が起こり、これによって、免疫応答の開始、維持および長期記憶に寄与する様々な細胞表面リガンドおよび受容体、ならびに可溶性タンパク質の発現が促進される。前述のように、ナイーブT細胞の初期活性化には、少なくとも3つのシグナルが必要である。シグナル1は、T細胞受容体(TCR)と、プロフェショナルAPC、例えば、樹状細胞(DC)の表面に主要組織適合複合体(MHC)により提示されるノミナルペプチドとの相互作用により生じる。シグナル2は、数個の異なる分子により媒介されると考えられ、持続的免疫応答に重要である。シグナル3は、活性化T細胞およびAPCにより合成されるサイトカインを介して伝達され、エフェクター免疫応答の維持に重要である。
本発明の具体的な一実施形態では、免疫共刺激ポリペプチドは4-1BBLポリペプチドである。4-1BBL(4-BB-L、4-BBリガンド、TNFSF9、ILAリガンドとしても知られる)は、活性化から2〜3日後、活性化B細胞、マクロファージ、およびDC上に発現されるII型タンパク質である。例えば、Aldersonら、Eur. J. Immunol. 1994, 24: 2219-27;Goodwinら、Eur. J. Immunol. 1993, 23: 2631-41;Pollokら、Eur. J. Immunol. 1994, 24: 367-74;DeBenedetteら、J. CD80(B7.1、CD28LG、LAB7としても知られる)とCD86(B7.2、CD28LG2、LAB72としても知られる)は共刺激ポリペプチドの例であり、いずれも、T細胞が発現したCD28/CTLA4共受容体に結合する。CD80は288個のアミノ酸(33048 Da)を含む。Freemanら、J. Immunol. 143 (8), 2714-2722(1989)を参照。ヒトCD80の全アミノ酸配列は、Swiss-Protデータベースにおいて登録番号P33681で見いだすことができる。CD80は、分泌シグナルを形成する残基1〜34と、潜在的細胞外ドメインを形成する残基35〜242と、潜在的膜貫通ドメインを形成する残基243〜263と、潜在的細胞質ドメインを形成する残基264〜288とを含むI型糖タンパク質である。従って、その分泌シグナル配列を含まない成熟CD80分子は、アミノ酸35〜288を呈示する。CD80をコードするヒトヌクレオチド配列は、GenBank登録番号NM_005191に見いだすことができる。
免疫活性化の初期段階後、「二次」受容体/リガンド対、例えば、4-1BBL/4-1BB(前述)およびLIGHT/HVEMはT細胞およびAPCの表面でアップレギュレートされることになる。これらの受容体/リガンド対は、初期活性化後事象の維持、免疫ホメオスタシス、および免疫記憶の発生に関与している。
OX40Lは、樹状細胞およびその他のAPCにより発現され、活性化T細胞上に存在するOX40に結合する。OX40Lは、183個のアミノ酸(21950 Da)を含む。Miuraら、Mol. Cell. Biol. 11:1313-1325(1991)を参照。OX40Lの全アミノ酸配列は、Swiss-Protデータベースにおいて登録番号P23510で見いだすことができる。OX40Lは、残基1〜23で細胞質ドメインを、残基24〜50で膜貫通ドメインを、そして残基51〜183で細胞外ドメインを含むII型糖タンパク質である。OX40Lのヌクレオチド配列は3510bpであり、コード配列は157-708である(GenBank登録番号NM_003326.2参照)。OX40Lのコグネイト受容体OX40に結合することができるOX40Lの残基51〜183またはその断片は、本発明で使用する結合対メンバーとのC末端融合物として結合または発現させることができる。
CD40Lは、活性化T細胞により発現され、タンパク質分解プロセシングにより膜形態から誘導される細胞外可溶性形態としても存在する。CD40L(a.k.a. PD-L1は、活性化TおよびB細胞、樹状細胞、ケラチノサイトおよび単球上に発現される。PD-L1(a.k.a. B7-H;B7H1;PDL1;PDCD1L1)は290個のアミノ酸(33275 Da)を含む。Dongら、Nat. GL50イソ型1は広範に発現され(脳、心臓、腎臓、肝臓、肺、膵臓、胎盤、骨格筋、骨髄、結腸、卵巣、前立腺、精巣、リンパ節、白血球、脾臓、胸腺および扁桃);GL50イソ型2(swissprot O75144)はリンパ節、白血球および脾臓において、また、活性化単球および樹状細胞上で発現される。GL50(a.k.a. 本発明の方法および組成物は、任意の抗原または感染因子(例えば、TAA、感染因子関連抗原、および感染因子そのものなど)に対する免疫応答を生じさせるかまたは増強するのに有用である。本発明によれば、標的腫瘍もしくは感染因子に関連する抗原(または感染因子そのもの)を免疫細胞に提示することによって、免疫応答を生じさせるかまたは増強する。
一実施形態では、前記抗原はTAAであり、本発明は、腫瘍に対する患者の免疫応答を生じさせるかまたは増強するのに有効な癌免疫療法を提供する。この実施形態によれば、本発明は、腫瘍サイズを縮小する方法と、腫瘍細胞の増殖を阻害する方法とを提供する。
結合対の例としてビオチンおよびストレプトアビジン(SA)またはアビジンが挙げられる。また、ビオチンに対する実質的な結合活性(例えば、それぞれ天然SAまたはアビジンの結合親和性の少なくとも50%またはそれ以上)を保持するSAまたはアビジン断片を用いてもよい。このような断片として、「コアストレプトアビジン(CSA)」、すなわち、全長ストレプトアビジンポリペプチドのトランケート型があり、これはストレプトアビジン残基13〜138、14〜138、13〜139及び14〜139を含みうる。例えば、Pahlerら、J. 免疫共刺激ポリペプチド、抗原または感染因子および結合対のメンバーを含むコンジュゲートは、当該分野で周知の方法により作製することができる。例えば、ポリペプチド/抗原/感染因子および結合対メンバーを互いに共有結合させる、あるいは、これらを直接またはリンカーを介して互いにコンジュゲートさせることができる。一実施形態によれば、ポリペプチド/抗原/感染因子および結合対メンバーは、融合タンパク質の成分である。当分野で周知の多数の異なる方法のいずれかにより融合タンパク質を製造することができる。例えば、融合タンパク質の1以上の成分ポリペプチドを化学的に合成する、または周知の組換え核酸技術を用いて作製することができる(本明細書で用いる「核酸」とはRNAまたはDNAを指す)。本発明において有用な核酸配列は、例えば、ポリメラーゼ連鎖反応(PCR)を用いて取得することができる。様々なPCR方法、例えば、PCR Primer: A Laboratory Manual, Dieffenbach 7 Dveksler編、Cold Spring Harbor Laboratory Press, 1995にが記載されている。
本発明の一実施形態は、(a)(i)第1免疫共刺激ポリペプチドを含むコンジュゲートメンバーと(ii)結合対の第1メンバーを含むコンジュゲートメンバーとを含む、第1コンジュゲート、および(b)(i)第1抗原または感染因子を含むコンジュゲートメンバーと(ii)結合対の第2メンバーを含むコンジュゲートメンバーとを含む、第2コンジュゲートを、それを必要とする患者に投与することにより、第1抗原または感染因子に対する免疫応答を生じさせるかまたは増強する方法を提供する。別の実施形態では、免疫細胞を第1および第2コンジュゲートで処理した後、患者に投与する。既述したように、いずれの免疫共刺激ポリペプチド、ならびに、腫瘍もしくは感染因子に関連するいずれの抗原(または感染因子そのもの)を用いてもよく、いずれの結合対を用いてもよい。
本発明はまた、前述したように、免疫療法に有用な改変免疫細胞、ならびにこれらを作製する方法を提供する。本発明のこの態様によれば、免疫細胞を改変することにより、第1腫瘍関連抗原を発現する腫瘍または感染因子に対する免疫応答を生じさせるかまたは増強する方法が提供される。本方法は、(a)(i)第1免疫共刺激ポリペプチドを含むコンジュゲートメンバーと(ii)結合対の第1メンバーを含むコンジュゲートメンバーとを含む、第1コンジュゲート;および(b)(i)腫瘍もしくは感染因子に関連する抗原または感染因子を含むコンジュゲートメンバーと(ii)結合対の第2メンバーを含むコンジュゲートメンバーとを含む、第2コンジュゲートに、第1免疫共刺激ポリペプチドの受容体を発現する免疫細胞を接触させることを含む。この方法によれば、第1コンジュゲートは、免疫共刺激ポリペプチドと受容体との間の結合により上記免疫細胞にコンジュゲートし、また、第2コンジュゲートは、第1および第2結合対メンバー間の結合を介して上記免疫細胞にコンジュゲートする。
本発明はまた、免疫刺激活性を有する免疫刺激部分を提供する。免疫刺激部分は、単独で投与するとき、または抗原もしくはその他の免疫刺激物質の投与と一緒にアジュバントとして使用するとき有用である。例えば、免疫刺激部分は、ワクチン、癌免疫療法、ならびに免疫に基づく障害の治療に有用である。免疫刺激部分は、動物への投与に適した組成物として製剤化することができ、免疫刺激を必要とする動物、例えば、ワクチン、癌免疫療法を受けている動物、または免疫に基づく障害の治療中である動物に投与することができる。
動物.Jackson Laboratories(Bar Harbor、メイン州)から成体の近交系BALB/c(H-2d)およびC57BL/6マウスを購入する。TCRトランスジェニックOT-I、DO11.10、C57BL/6.SJL動物をTaconics(Germantown、ニューヨーク州)から購入し、NIH指針に従って維持する。
既述のように、4-1BBLは適応および先天性免疫応答の調節に重要な役割を果たす。4-1BBLは、CD4+およびCD8+T細胞、NK細胞およびDCの活性化のための共刺激分子として働き、Treg細胞の抑制機能を阻害する。従って、この分子は、癌治療に有効な腫瘍応答の発生のための特異的アジュバントとして働くことができる。
4-1BBに対する4-1BBL融合タンパク質対モノクローナル抗体の相対活性を評価するために、フローサイトメトリーにより選別したCD4+およびCD8+T細胞を、増殖アッセイにおいて、様々な量の4-1BBL融合タンパク質及び抗体の存在下で、準最適濃度のポリクローナル抗CD3抗体で刺激した。T細胞の増殖に対する融合タンパク質の活性は前記抗体の70倍であった(図9)。
市販のキット(Pierce Biotechnology、イリノイ州ロックフォード)を用いて、オボアルブミン(OVA)をビオチン化した。ビオチン化OVAを様々な比でコンジュゲート用のCSA-4-1BBL融合タンパク質とin vitroで前混合し、百万個のOT-IT細胞を養子免疫的に移入したナイーブC57BL/6.SJLマウスに腹腔内注射した。具体的には、百万個のOT-I CD8+T細胞をCFSEで標識し、これを、ビオチン化オボアルブミン(10μg/注射)(「OVA」)、およびビオチン化OVAと混合したCSA-4-1BBL(1μg/注射)(「41BBL+OVA」)またはコンジュゲート型OVA-ビオチン/CSA-4-1BBL(41BBL-OVA)で免疫したB/6.SJLマウスに移入した(図10)。図10の最後のパネル(「41BBL-OVA*」)は、10μgビオチン化OVAにコンジュゲートさせた5μgのCSA-4-1BBLに対する応答を示す。対照として、CSA-4-1BBLと等モルレベルのコアストレプトアビジン(「SA」)を用いた。
この実施例は、CSA-41BBLがDCに抗原を有効に送達することを明らかにする。蛍光抗原としてビオチン化PEを用いた。ビオチン化PE(250 ng)を氷上で250 ngのCSA-41BBLと30分間コンジュゲートさせた。Jaws II樹状細胞(5×105/ウェル)をビオチン化PE(250 ng/ml)またはビオチン化PE/CSA-41BBLコンジュゲートと一緒に16時間培養した。フローサイトメトリーを用いてPEのレベルを検出した。図11Aは、PE+細胞を示すヒストグラムである。グレーで塗りつぶした部分は非処理細胞を、黒い点線はビオチン化PEで処理した細胞を、また、黒い線はビオチン化PE/CSA-41BBLコンジュゲートで処理した細胞を示す。図11Bは、各処理についてのPEの平均蛍光強度(MFI)を示すが、これにより、コンジュゲートで処理した細胞が有意に高い応答を提示することが明らかにされた。
この実施例では、4-1BBLが樹状細胞を活性化することを明らかにする。Jaws II樹状細胞(5×105/ウェル)は未処理のままか、または5ng/mlのGM-CSFの存在下、5μg/mlのCSA-41BBLコンジュゲートもしくは5μg/mlのリポ多糖(LPS)で24ウェルプレート中で48時間処理した。図12Aに示すように、フローサイトメトリーを用いて、CD86およびMHCクラスIIレベルを分析した。薄いグレーで塗りつぶした部分はイソタイプ処理細胞を示し、濃いグレーで塗りつぶした部分は未処理細胞を、黒い線はCSA-4-1BBL処理細胞を、また点線はLPSを示す。図12Bは、CD86およびMHCクラスIIの平均蛍光強度(MFI)を示すが、これにより、CSA-4-1BBLで処理した細胞が有意に高い応答を呈示することが明らかにされた。
この実施例では、CSA-4-1BBLがビオチン化抗原を樹状細胞に送達し、これらの細胞をin vivoで活性化へと駆動することを明らかにする。ビオチン化OVAをCSA-41BBLと接触させることにより、ビオチン化OVA/CSA-4-1BBLコンジュゲートを取得した。該コンジュゲートまたはビオチン化OVA/CSAコンジュゲートをナイーブC57BL/6マウスに静脈内注射した。24時間後、マウスを安楽死させ、脾細胞を採取した。CD11c+細胞集団においてフローサイトメトリーを用いて樹状細胞活性化を分析した。図13に示すように、ナイーブビオチン化OVA-SA処理、およびビオチン化OVA/CSA-41-BBL処理マウス由来の樹状細胞上でのCD40、CD86およびMHCクラスII発現の平均蛍光強度(MFI)を測定した。この図から、ビオチン化OVA/CSA-41-BBL処理マウスが有意に高い応答を呈示することがわかる。
前述のように、天然に存在するCD4+CD25+FoxP3+Treg細胞は、4-1BB受容体を構成的に発現し、それ自体4-1BBL刺激に応答する。以下の実施例により、Treg細胞に対する4-1BBL融合タンパク質の刺激活性を明らかにする。
Treg機能の調節における4-1BB/4-1BBL媒介シグナル伝達の役割は、2つの近年の研究の主題であり、これらはそれぞれ相反する知見を有する。一方の研究では、4-1BBシグナル伝達がTreg細胞の抑制機能を中和することを明らかにした(Choiら、2004, J. Leukoc. Biol. 75: 785-91)のに対し、他方は、4-1BBシグナル伝達が、抑制機能に大きな作用をもたらすことなく、Treg増殖を媒介すると報告した(Zhengら、2004, J. Immonul. 173: 2428-34)。この矛盾を解明するため、CSA-4-1BBL融合タンパク質を用いて、Treg機能における4-1BBシグナル伝達の役割を調べた。
(a)可溶性タンパク質としてOVAを発現するA20形質転換体の作製
LipofectamineTM2000キットを、製造者(Invitrogen)プロトコルに従って用い、前述したOVA構築物をA20にトランスフェクトする。G418選択培地において安定な形質転換体を選択し、単細胞レベルでクローニングし、ウェスタンブロットを用いてOVAの発現について試験する。有意なレベルのOVA発現を有するクローンを、図10を参照にして説明したCFSE増殖アッセイにおいてOVAに特異的なDO11.10 CD4+T細胞のスティミュレーターとして用いる。OVA発現について陽性であると確認したら、百万個のA20生細胞をBALB/cマウスの右側腹部に皮下注射する。腫瘍発達および生存についてマウスを一日おきにモニターし、腫瘍が直径20 mmのサイズに達したら安楽死させる。親A20細胞を接種したマウスを腫瘍増殖の対照として用いる。OVAを発現するA20細胞形質転換体は、同系BALB/cマウスに注射すると、腫瘍を形成する。
DES発現系(Invitrogen)と、本発明者らが確立したプロトコルを用いて、CSA-4-1BBLタンパク質を作製する。例えば、Singhら、2003, Cancer Res. 63: 4067-73; Yolcuら、2002, Immunity 17: 795-808を参照されたい。CSA-4-1BBL融合タンパク質に施した6XHisタグを利用した固定化金属ベースのアフィニティークロマトグラフィーを用いて、融合タンパク質を精製する。タンパク質を脱塩し、超ろ過により濃縮し、SDS-PAGEにより純度について分析する。ビシンコニン酸(BCA)アッセイ(Pierce)を用いて、タンパク質調製物を濃度について評価し、CambrexのQCL-1000(登録商標)Chromogenic LALエンドポイントアッセイを用いて、内毒素の存在について試験する。
DSB-X Biotin Labeling Kitを製造者(Molecular Probes、カリフォルニア州サンディエゴ)のプロトコルに従って用いて、マレイミド活性型の、内毒素を含まないニワトリOVA(Pierce)をビオチン化する。PBSでの十分な透析後、ウェスタンブロット、およびプロービングのためのアルカリホスファターゼコンジュゲート型ストレプトアビジンを用いて、濃度、内毒素レベル、およびビオチン化について、ビオチン化OVAを評価する。必要であれば、Detoxi-Gel Endotoxin Removingキット(Pierce)を用いて、内毒素を除去する。ビオチン化OVAをCSA-4-1BBL融合タンパク質とコンジュゲートさせる。タンパク質コンジュゲートをアリコートに分け、使用まで−80℃で冷凍する。
CSA-4-1BBLとビオチン化OVAをPBS中で様々な4-1BBL:OVAのモル比、例えば、1:1、1:5、1:10、5:1、および10:1で前混合し、3週間毎に様々な用量(例えば、10、50および100μgのOVA)でBALB/cマウスの群に腹腔内注射する。ビオチン化OVAにコンジュゲートしたストレプトアビジン、ビオチン化OVAのみ、またはCSA-4-1BBLと混合した非ビオチン化OVAを注射したマウスを対照として用いる。
既述のように、腫瘍は、腫瘍増殖の過程で発達する多様な機構により、免疫系を回避する。腫瘍進行の早期における本発明のコンジュゲートの効能は、免疫回避機機構が確立されていないとき、腫瘍チャレンジと同時に動物にワクチン接種することにより証明される。樹立された腫瘍に対する本発明のコンジュゲートの効能については、いったん腫瘍が樹立され、免疫回避機構が完全に発達した動物をワクチン接種することにより証明される。
BALB/cマウスに百万個のA20生細胞を右側腹部にチャレンジし、同時に、ビオチン化OVA/CSA-4-1BBLコンジュゲートを腹腔内にワクチン接種する。抗原/4-1BBLコンジュゲートによるワクチン接種を4週間の間(この時点までに対照動物における腫瘍は直径10〜15mmのサイズに達する予定である)に週に1回繰り返す。非操作マウスと、CSA-OVAコンジュゲートをワクチン接種したマウスを対照として用いる。
百万個のA20腫瘍生細胞をBALB/cマウスの右側腹部に皮下接種する。腫瘍発達についてマウスをモニターし、腫瘍が直径4〜6mmのサイズに達したとき、ビオチン化OVA/CSA-4-1BBLコンジュゲートをワクチン接種する。ワクチン接種プロトコルはまず、腫瘍が消失するか、直径20 mmのサイズに達するまで、毎週腹腔内注射することを含む。
以下の実施例では、ビオチン化OVA/CSA-4-1BBLコンジュゲートが、バイスタンダー効果またはエピトープ拡散のいずれかにより、不定のA20腫瘍抗原(OVA以外)に対する免疫応答を生じさせることを明らかにする。
いくつかの状況では、本発明のワクチンの抗原成分として用いる目的で、遺伝子的にビオチン化した抗原を作製するのが有利な場合もある。これに関して、Avidity, Inc.(コロラド州デンバー)のBiotin AviTag技術を用いることができる。Biotin AviTagは、ビオチンリガーゼであるBirA(ペプチド配列内のリシン残基にビオチンを結合させる)により認識される固有の15個のアミノ酸ペプチドから構成される。Biotin AviTagは、目的とする任意のタンパク質に遺伝子的に融合させ、該タンパク質をビオチン分子でタグ付けすることもできる。
ビオチン化OVA/CSA-4-1BBLを伴う上で例示したビオチン化抗原/4-1BBLコンジュゲートは、任意の抗原を含むあらゆるワクチン設定で用いることができる。癌ワクチンに関しては、2つの万能ヒトTAAであるテロメラーゼ逆転写酵素とサバイビンが、本発明のビオチン化抗原/CSA-4-1BBLコンジュゲートの有利な抗原成分となりうる。
前述したように、抗原成分としてヒトパピローマウイルスE7を含む本発明のコンジュゲートは子宮頚癌に対して有用である。この実施例は、本発明のこの具体的実施形態に関する。
HPVワクチンとして有用な本発明のコンジュゲートの抗原成分としてビオチン化E7を用いる。一実施形態では、全長E7タンパク質を用いて、最大数のエピトープを与える。TC-1細胞由来の全RNAを用いたRT-PCRにより、全長HPV-16E7をコードするcDNAをクローニングする。配列確認後、6X-Hisタグを含むpMIB/V5-Hisベクター(Invitrogen)にcDNAをインフレームでサブクローニングして、構成的発現およびDES系での分泌を達成する。前述のように金属親和性樹脂を用いて分泌タンパク質を精製する。EZ-Link Sulfo-NHS-LC-Biotinを製造者のプロトコル(Pierce)に従って用い、精製したE7をin vitroでビオチン化する。手短には、精製、濃縮したE7をリン酸緩衝化生理食塩水(PBS)でバッファー交換し、EZ-Link Sulfo-NHS-LC-Biotinと一緒に室温で1時間インキュベートする。接線流濾過(Spectrum Labs、ニュージャージー州)を用いて、非結合ビオチンを除去する。
前述の概略的手順に従い、ビオチン化E7およびCSA-4-1BBL融合タンパク質を用いて、E7と4-1BBLとを含むコンジュゲートを作製する。比較のために、以下のようにE7/4-1BBL融合タンパク質を作製する。E7および4-1BBLをコードするcDNAを、6X-Hisタグを含むpMIB/V5-Hisベクター(Invitrogen)にインフレームでサブクローニングして、構成的発現およびDES系での分泌を達成し、前記のようにタンパク質を発現させ、精製する。
E7/4-1BBLコンジュゲートのビオチン結合および4-1BB受容体結合活性を以下のように評価する。
(a)用量の最適化
E7/4-1BBLコンジュゲートを含むワクチンの最適用量は次のように評価することができる。1、10もしくは50μgのビオチン化E7を用いて、2つの比(例えば、1:4および1:8のCSA-4-1BBL:E7)でビオチン化E7とCSA-4-1BBLとを混合することにより、ビオチン化E7とCSA-4-1BBLとを含むコンジュゲートを形成する。また、同程度の量の対照非ビオチン化E7も用いる(E7の上記用量は、50μgのE7がTC-1細胞に対する防御免疫応答を生じさせるのに有効であることを証明した研究に基づく)。4-1BBL:E7の最適比および抗原の最適量は、以下に説明するような様々なプロトコルに従ってワクチンに対する免疫応答を評価することにより、実験的に決定および調整することができる。
四量体染色により、CD8+T細胞の増殖に関するワクチン効能を評価することができる。C57BL/6雌マウスに対し、PBS中の前記ワクチン調製物を腹腔内注射する。PBS、CSA-4-1BBL、CSA-4-1BBL+非ビオチン化E7、もしくはE7-4-1BBL融合タンパク質を注射したマウスを対照として用いる。10日後、同等用量で2回目の腹腔内投与を行い、最後のワクチン接種から3日後に、脾細胞を採取し、四量体技術およびフローサイトメトリーを用いて、E7特異的CD8+T細胞数を定量する。手短には、FITC-抗-CD8抗体と、E7の免疫優性エピトープであるペプチド49〜57(RAHYNIVTF)をロードしたMHCクラスIH-2Db分子のPE-四量体で、免疫化マウス由来の脾細胞を標識する(この四量体は、National Institutes of Health Tetramer Facility(ジョージア州アトランタ)から入手することができる)。センダイウイルス核タンパク質324〜332ペプチド(FAPGNYPAL)をロードしたクラスIH-2Db分子を陰性対照として用いる。染色後、フローサイトメトリーにより細胞を分析して、四量体についてCD8+T細胞陽性の%を求める。
エフェクターCD8+T細胞のシグネチャーサイトカインである、IFN-γの発現についてワクチン誘導CD8+T細胞を特性決定することにより、T細胞の機能の評価が可能になる。雌C57BL/6マウスに対し、最適用量のビオチン化E7/CSA-4-1BBLコンジュゲートワクチン(前述のように決定)を腹腔内注射し、10日後、免疫化マウス由来の脾細胞を採取し、E7を発現する放射線照射済TC-1細胞と一緒に5日間共培養した後、ゴルジ輸送阻害因子のブレフェルジンAを一晩補充する。フィコール勾配を用いて生細胞を採取し、抗マウスFcγ受容体抗体(アメリカンタイプカルチャーコレクションから2.4G2)と一緒に1時間インキュベートした後、FITC標識抗CD8抗体で染色する。次に細胞を固定、透過性にし、PE-標識抗IFN-γ抗体(Pharmingen)について染色し、フローサイトメトリーにより分析する。イソタイプ抗体で染色した細胞を対照として用いる。PBS、E7-4-1BBL融合タンパク質、もしくはCSa-4-1BBL+非ビオチン化E7で免疫したマウス由来の脾細胞を対照として用いる。
ワクチン誘導CD8+T細胞がE7分子を発現するTC-1細胞を溶解する能力を以下のように評価する。前述のようにワクチン接種したマウスから採取した脾細胞を100μg/ml E7タンパク質の存在下で5日間共培養する。培養物に50U/mlgの外因性IL-2を補充して、CD8+T細胞の増殖を支持する。フィコール勾配を用いて、生存脾細胞を回収し、JAMアッセイにおいて様々なエフェクター:標的比(例えば、1:1、10:1、20:1、40:1、および80:1)でTC-1標的細胞に対するエフェクター細胞として用いる。例えば、Singhら、2003, Cancer Res. 63: 4067-73を参照。癌免疫療法には、CD8+T細胞による腫瘍細胞の直接死滅が重要であるため、このアッセイでの効能の証明は、子宮頚癌に対するワクチンの効能をさらに支持することになる。
CD4+T細胞応答の誘導におけるビオチン化E7/CSA-4-1BBLの効能を以下のように評価する。免疫化マウスからの脾細胞をCFSEで標識し、IL-2を培養物に添加しない以外は、前記と同じ培養条件下で組換えE7タンパク質と一緒に共培養する。培養中、様々な日に細胞を回収し、APC-CD4抗体で染色してから、フローサイトメトリーを用いて増殖について分析する。E7タンパク質を含まない、またはOVAタンパク質を含む培養物を対照として用いる。CD4+T細胞応答はCD8+T細胞およびB細胞応答に重要であることについて一般に見解が一致しているため、このアッセイにおける効能の証明は子宮頚癌に対するワクチンの効能をさらに支持することになる。
ビオチン化E7/CSA-4-1BBLワクチンによるin vivo処理が液性応答を生じさせる能力を次のように評価する。ワクチン接種したマウス由来の血液を採取し、血清を単離して、E7タンパク質をコーティングしたMaxisorb ELISAプレート(Nalgene Nunc International)をスクリーニングする。セイヨウワサビペルオキシダーゼコンジュゲート型ヤギ抗マウスIgGおよびヤギ抗マウスIgM抗体を用いて、抗E7 IgGおよびIgMを検出する。対照は、PBSおよび前記のような対照タンパク質で免疫したマウスから採取した血清を含む。
TC-1移植性腫瘍モデルにおいて、腫瘍形成の阻止および根絶における本発明のE7/4-1BBLコンジュゲートを含むワクチンの効能を2つの異なる設定で評価する。第1の設定は、免疫回避機構がまだ発達していない、腫瘍注射前のワクチン接種を含む。第2の設定は、免疫回避機構が十分に発達した樹立腫瘍に対するワクチン接種を含む。C57BL/6マウスにTC-1細胞を注射して、腫瘍形成を誘発し、TC-1注射の前および後に、ビオチン化E7/CSA-4-1BBLコンジュゲートを免疫する。前記のように免疫応答を評価する。腫瘍発達および生存についても1日おきにマウスをモニターし、腫瘍が直径20 mmのサイズに達したら安楽死させる。
以下の実施例では、本発明のE7/4-1BBLコンジュゲートによる免疫が後の腫瘍チャレンジに対する防御免疫を生じさせることを証明する。
既存の腫瘍に対する本発明のE7/4-1BBLコンジュゲートの治療効果を以下のように証明する。
A型インフルエンザRNAからの逆転写酵素−ポリメラーゼ連鎖反応により、目的のインフルエンザタンパク質(例えば、H1、N1、NPおよび/またはMP2)のcDNAを作製する。cDNAをpCSAベクターにサブクローニングし、ショウジョウバエ(Drosophila)昆虫細胞にトランスフェクトすることにより、安定な形質転換体を確立する。
(a)用量最適化
前記A型インフルエンザ抗原/4-1BBLコンジュゲートを様々な用量でC57BL/6マウスにワクチン接種する。既述のように、四量体技術、サイトカイン染色、細胞傷害性アッセイ、および液性応答の測定を含む標準的な免疫学的技術を用いて、マウスにおける免疫応答を測定する。初期結果を用いて、ワクチンの最適な投与計画を決定する。
以下のようにA型インフルエンザでチャレンジしたマウスにおいて、A型インフルエンザ抗原/4-1BBLコンジュゲートワクチンの予防および治療効果を明らかにする。ヒトインフルエンザウイルスに感染したマウスをA型インフルエンザ抗原/4-1BBLコンジュゲートワクチンで感染前および感染後に治療し、ウイルス力価を測定することにより、治療の効果を判定した。ワクチン接種した感染マウスおよび対照の感染マウス由来の肺を感染から1、3、5、7および9日後に採取する。罹患率の間接的測定として体重減少を毎日測定する。
この実施例で用いるヒト単球性白血病THP-1およびマウスA20B細胞リンパ腫株は、アメリカンタイプカルチャーコレクション(ATCC、米国メリーランド州ロックビル)から購入した。A20細胞は、10%熱不活性化ウシ胎仔血清(FBS;Valley Biomedical、米国バージニア州ウィンチェスター)、12 mM L-グルタミン、100 U/mlペニシリン、100μg/mlストレプトマイシン(すべてGIBCO製)および50μM 2-メルカプトエタノール(Sigma、米国ミズーリ州セントルイス)を補充したDMEM(GIBCO、米国メリーランド州ゲイサーズバーグ)中で培養した。THP-1細胞は、加湿5%CO2インキュベーターにおいて37℃で、5%FBS、100 U/mlペニシリンおよび0.1 mM Hepesバッファー(GIBCO)を補充したRPMI中で培養した。
PCRにおいて、鋳型としてのストレプトミセス・アビジニ(Streptomyces avidinii)から単離したゲノムDNAと、特異的プライマーとを用いて、CSAをコードする遺伝子をクローニングした(図16のaおよびb)。PCRにおいて、鋳型としてフィトヘマグルチニン(PHA)活性化ヒト末梢血液リンパ球から単離した全RNAから作製した第1鎖cDNAを用い、CD40L特異的プライマーを使用して、ヒトCD40Lの細胞外ドメインをクローニングした(図16のcおよびd)。コンカナバリンA(ConA)で活性化したマウス脾細胞から単離した全RNAを用いて、CSA-hCD40Lと同様にマウスCD40Lをクローニングした。
Quantikine CD40Lイムノアッセイ(製品説明書(R&D Systems)に記載されているように、マイクロプレートに予めコーティングしたCD40Lに特異的なポリクローナルAbを用いる)を用いて、CSA-hCD40Lの発現を検出および定量した。ウェスタンブロット分析のために、まず、天然および変性条件下で、ドデシル硫酸ナトリウム−ポリアクリルアミドゲル電気泳動によりCSA-hCD40LおよびCSA-mCD40Lの上清を分画し、次に、セミドライブロット装置(BioRad、米国カリフォルニア州ハークリーズ)を用いて、二フッ化ポリビニリデン膜に移した。最初にブロッキングバッファー中で、次に、1:1,000希釈でヤギ抗SA Ab(Pierce、米国イリノイ州Rockford)を含むブロッキングバッファー中で、室温で1時間膜をインキュベートした。その後、膜を十分に洗浄し、1:4,000希釈のセイヨウワサビペルオキシダーゼコンジュゲート型抗ヤギ抗体と一緒に1時間インキュベートした。最後に、化学発光基質を製品説明書(ECL、Amercham Bioscience、英国)に従って用いて、タンパク質を検出した。
百万個のCD40陽性マウスA20 B細胞リンパ腫またはヒトマクロファージTHP-1細胞を200 ng/mlのCSA-CD40L(ヒトもしくはマウス)部分または対照CSAタンパク質と一緒に4℃で30分インキュベートした。PBSで数回洗浄した後、フローサイトメトリーにおいてFITCコンジュゲート型抗ストレプトアビジン抗体(Vector Laboratories、米国カリフォルニア州バーリンガム)を用いて、結合タンパク質を検出した。陰性対照としてCSAを用いて非特異的結合を検出した。CD80およびMHCクラスII分子の発現に対するCSA-CD40Lでの刺激の効果について、0.5x106 THP-1細胞を100 ng/mlのCSA-hCD40LもしくはCSAと一緒に培養するかまたは膜形態のCD40Lでトランスフェクトした0.5x106 CHO細胞と一緒に共培養することにより(48時間)判定した。次に、細胞を洗浄し、飽和濃度のFITCコンジュゲート型抗CD80(L307.4)およびHLAクラスII(TU36)抗体(BD-Pharmingen、米国カリフォルニア州サンディエゴ)で染色した後、フローサイトメトリーで分析した。
生後6〜8週間のマウスの大腿骨から骨髄を洗い流し、ピペット操作により単細胞に分散させ、塩化カリウムアンモニウム(ACK)溶液で赤血球を溶解させた。次に、氷上で30分、TIB 105、TIB 146およびクローンRL-172ハイブリドーマ細胞培養物飽和上清のカクテルを用いて、TおよびB細胞について単細胞懸濁液を枯渇させた(培養上清は、Tatiana Zoria博士(ピッツバーグ大学、ペンシルバニア州)から贈与されたものである)。ウサギ補体と一緒に単細を37℃で30分インキュベートし、6ウェルプレート中の完全培地(RPMI 1640、2mM L-グルタミン、100μg/mlペニシリンおよびストレプトマイシン、10%FBS、0.1 mM非必須アミノ酸、1mMピルビン酸ナトリウム、1μg/mlインドメタシンおよび50μM N-メチル-L-アルギニン)(Sigma))中、106細胞/mlの濃度で一晩(37℃、5%CO2)培養した。非接着細胞を穏やかなピペット操作により回収し、計数し、組換えマウス顆粒球−マクロファージコロニー刺激因子(5ng/ml)およびrmIL-4(5ng/ml)(すべてUS Biological(米国マサチューセッツ州スワンプスコット)製)を補充した完全培地において105細胞/mlの濃度で再懸濁させた。6ウェルプレート(4ml/ウェル)で細胞を5日間培養した。
ヒト単球を96ウェルマイクロタイタープレートに塗布し、1μg/mlの市販の三量体組換えヒトCD40L(rhsCD40L)+1μg/mlのエンハンサー(Alexis Biochemicals、米国カリフォルニア州サンディエゴ)と100 ng/mlのCSA-hCD40L、CSA-mCD40L、もしくはCSAとを用いて刺激した。18時間のインキュベーション後、上清を回収し、全サイトカイン用のOptEIATMセット(PharMingen)を用いたELISAによりアッセイした。E-max Precisionマイクロプレートリーダー(Molecular Devices、米国カリフォルニア州サニーベール)を用いて分析を実施した。
サイトカインmRNA合成の分析をRNAse保護アッセイにより実施した。細胞を6ウェルプレートに塗布し、CSA-CD40L部分を用いて、CD40を介して3〜4時間刺激した。エンハンサーと共にCD40LおよびrhsCD40Lを発現するCHO形質転換体を対照として用いた。Trizolを製品説明書(Invitrogen)に記載されるとおりに使用してRNAを抽出した。ヒトサイトカイン/RNA鋳型セットから作製された放射線標識プローブであるmCK-3b(RiboQuant、BD-PharMingen、米国カリフォルニア州サンディエゴ)と共にRNA(5μg)を55℃で一晩ハイブリダイズした。RNAse処理を37℃で45分実施した後、保護されたプローブを精製し、TBEバッファー中の5%ポリアクリルアミドゲル(BioRad)を用いた電気泳動により分離した。ゲルを乾燥させ、Kodak Biomax XL X線フィルム(Eastman Kodak、米国ニューヨーク州ロチェスター)に暴露した。非消化プローブをマーカーとして、片対数紙に、泳動距離対ヌクレオチド長の標準曲線をプロットした。次に、サンプルにおけるRNAse保護バンドの同一性をグラフから推定した。
IFN-γ初回免疫マクロファージのCD40ライゲーションにより、一酸化窒素産生の刺激が起こるが、これは、マクロファージの殺菌および細胞傷害活性に重要な役割を果たす。誘導性一酸化窒素シンターゼ(iNOS)は、L-アルギニンからの一酸化窒素の合成を触媒する一酸化窒素シンターゼのファミリーに属する。Th1およびTh2ヘルパー細胞は、マクロファージにおけるアルギニン代謝を示差的に調節することができる。Th1細胞はマクロファージによるiNOS産生を誘導するのに対し、Th2細胞はマクロファージを誘導して、抗炎症機能に関連するアルギナーゼを産生させる。従って、マクロファージiNOS産生は、Th1型免疫応答の特質である。
抗原として50μgのオボアルブミン(OVA)を、そしてアジュバントとして2用量(それぞれ、12.5μgと25μg)のCSA-4-1BBLまたはLPSをナイーブC57BL/6マウスに静脈内免疫した。ナイーブ動物を対照として用いた。
HPV16 E7エピトープP1(アミノ酸配列RAHYNIVTFを有する)のCD8+T細胞エピトープの投与に基づくワクチン接種プロトコルにおいて、ヒトパピローマウイルス−16 E7タンパク質を安定に発現する1x105個のTC-1生細胞をナイーブB6マウスの右側腹部に皮下チャレンジした。図23は、このTC-1腫瘍モデルにおけるマウスの生存率を示す。10日後、マウスに対し、(i)PBS(黒ひし形、n=20);(ii)50μg P1+12.5μg CSA(黒四角、n=6)(iii)25μg CSA-4-1BBL(黒三角、n=10);(iv)50μg P1+25μg CSA-4-1BBL(白三角、n=13);または(v)50μg P1+10μg CpG(白四角、n=7)のいずれかを1回皮下注射した。
25μg CSA-4-1BBLにコンジュゲートさせた50μg OVAまたは50μgビオチン化OVAでナイーブC57BL/6マウスを免疫した。対照として、何匹かのマウスは非処理のままにした。7日後、1x105個のOVA発現性EG.7腫瘍細胞をマウスの右側腹部に皮下チャレンジした。カリパスを用いて腫瘍増殖を週に3回モニターした。結果(腫瘍無含生存率)を図24に示す。図示するように、対照マウスと、OVAをワクチン接種したマウスはすべて腫瘍を発達させたのに対し、ビオチン化OVA/CSA-4-1BBLをワクチン接種したマウスはすべて腫瘍を発達させなかった。これは、ビオチン化OVA/CSA-4-1BBLによるワクチン接種により、甲状腺腫瘍の増殖が100%阻止されたことを証明するものである。
ナイーブC57BL/6マウスに(i)50μg OVA、(ii)50μg OVAと25μg CSA-4-1BBL、(iii)50μg OVAと25μg 抗CD137抗体または(iv)50μg OVAおよび25μg LPSを静脈内に免疫した。ナイーブマウスを対照として用いた。7日後、全マウスにCFSE標識標的細胞を投与した。手短には、ナイーブC57BL/6由来の脾細胞を2つの集団に分けた。第1の集団を0.25μM CFSEで標識した(CFSElow)。第2の集団は2.5μM CFSEで標識した後、2μg/ml OVA257-264 SIINFEKLペプチドで1時間パルスした(CFSEhi)。細胞を1:1の比で混合し、総数1x107個の細胞をレシピエントマウスに静脈内注射した。48時間後に脾臓を採取し、フローサイトメトリーにより、CFSE蛍光強度を分析したところ、図25に示す結果が得られた。結果は、ナイーブマウスに標準化した参照CFSElowピークと比較したペプチドパルスCFSEhiピークの溶解%として各パネルの隅に表示する。このアッセイから、4-1BBLが、抗原(OVA)のみ(24.2%)、または抗原とLPS(35%)と比較して、高レベル(95%)まで抗原特異的CTL応答を増強することができ、その結果、大部分の標的細胞を死滅させることを明らかにした。
ナイーブB6-SJL(CD45.1+)マウスに(i)10μg OVA、(ii)10μg OVAおよび5μg 4-1BBLを静脈内免疫するか、または(iii)非処理のままにした。2日後、1x106個のCFSE標識OT-1細胞(CD45.2+)を静脈内注射により投与した。3日後脾臓を採取し、図26に示すように、フローサイトメトリーを用いてOT-1細胞の増殖を分析した。抗原と一緒に4-1BBLを投与すると、大部分のOT-1細胞の増殖からわかるように、CD8+T細胞に対する抗原提示が増大した(OVA+4-1BBLは83.2%;OVAは13.7%;非処理は8.8%)。
ナイーブBALB/cマウスに、25μg OVA-FITC、25μg OVA-FITCおよび10μg CSA、または25μg OVA-FITCおよび25μg CSA-4-1BBLを皮下注射した。3時間後、注射部位の鼠径リンパ節を採取した。フローサイトメトリーを用いて、CD11c+集団におけるFITC+細胞を分析し、図27に示すように、in vivo蛍光標識抗原取込みを測定した。図からわかるように、4-1BBLシグナル伝達は、CD11c+DCによる抗原取込みを増大したが、対照CSAタンパク質は何の作用も与えなかった。

その際、第2免疫共刺激ポリペプチドは、第1免疫共刺激ポリペプチドと同じであるかまたは違うものであり;第2抗原は第1抗原と同じであるかまたは違うものであり;第3コンジュゲートの第1および第2結合対メンバーは、第1および第2コンジュゲートの第1および第2結合対メンバーと同じであるかまたは違うものであり、
第1コンジュゲートメンバーは、第1および第2結合対メンバー間の結合を介して前記第2コンジュゲートメンバーに結合している、実施形態1に記載の組合せ。
(b)(i)感染因子を含むコンジュゲートメンバーと(ii)結合対の第2メンバーを含むコンジュゲートメンバーとを含む、第2コンジュゲート
を含む組合せ。
(a)(i)第1免疫共刺激ポリペプチドを含むコンジュゲートメンバーと(ii)結合対の第1メンバーを含むコンジュゲートメンバーとを含む、第1コンジュゲート;および(i)第1腫瘍関連抗原を含むコンジュゲートメンバーと(ii)結合対の第2メンバーを含むコンジュゲートメンバーとを含む、第2コンジュゲート;または
(b)第1および第2コンジュゲートでin vitro処理した免疫細胞
を投与することを含む、上記方法。
その際、第2免疫共刺激ポリペプチドは、第1免疫共刺激ポリペプチドと同じであるかまたは違うものであり;第2抗原は第1抗原と同じであるかまたは違うものであり;第3コンジュゲートの第1および第2結合対メンバーは、第1および第2コンジュゲートの第1および第2結合対メンバーと同じであるかまたは違うものであり、
第1コンジュゲートメンバーは、第1および第2結合対メンバー間の結合を介して第2コンジュゲートメンバーに結合している、実施形態20に記載の方法。
(a)(i)第1免疫共刺激ポリペプチドを含むコンジュゲートメンバーと(ii)結合対の第1メンバーを含むコンジュゲートメンバーとを含む、第1コンジュゲート;および
(b)(i)前記腫瘍もしくは感染因子に関連する抗原または該感染因子を含むコンジュゲートメンバーと(ii)結合対の第2メンバーを含むコンジュゲートメンバーとを含む、第2コンジュゲート、
と接触させることを含み、
その際、第1コンジュゲートは、免疫共刺激ポリペプチドと受容体との間の結合により免疫細胞にコンジュゲートされ、かつ、第2コンジュゲートは、第1および第2結合対メンバー間の結合を介して免疫細胞にコンジュゲートされる、上記方法。
第2免疫共刺激ポリペプチドは、第1免疫共刺激ポリペプチドと同じであるかまたは違うものであり;第2抗原は、存在する場合には、第1抗原が存在する場合にこれと同じであるかまたは違うものであり;第3コンジュゲートの第1および第2結合対メンバーは、第1および第2コンジュゲートの第1および第2結合対メンバーと同じであるかまたは違うものであり、
第1コンジュゲートメンバーは、第1および第2結合対メンバー間の結合を介して第2コンジュゲートメンバーに結合している、実施形態42に記載の方法。
(a)(i)第1免疫共刺激ポリペプチドを含むコンジュゲートメンバーと(ii)結合対の第1メンバーを含むコンジュゲートメンバーとを含む、第1コンジュゲート;および
(b)(i)第1抗原または感染因子を含むコンジュゲートメンバーと(ii)上記結合対の第2メンバーを含むコンジュゲートメンバーとを含む、第2コンジュゲートを含み、
その際、第1コンジュゲートは、免疫共刺激ポリペプチドと受容体との間の結合を介して免疫細胞にコンジュゲートされ、かつ、第2コンジュゲートは、第1および第2結合対メンバー間の結合を介して免疫細胞にコンジュゲートされている、上記改変免疫細胞。
(a)(i)第1免疫共刺激ポリペプチドを含むコンジュゲートメンバーと(ii)結合対の第1メンバーを含むコンジュゲートメンバーとを含む、第1コンジュゲート;および
(b)(i)前記感染因子に関連する第1抗原を含むかまたは該感染因子を含むコンジュゲートメンバーと(ii)結合対の第2メンバーを含むコンジュゲートメンバーとを含む、第2コンジュゲート
を投与することを含む、上記方法。
第2免疫共刺激ポリペプチドは、第1免疫共刺激ポリペプチドと同じであるかまたは違うものであり;第2抗原は、存在する場合には、第1抗原が存在する場合にはこれと同じであるかまたは違うものであり;第3コンジュゲートの第1および第2結合対メンバーは、第1および第2コンジュゲートの第1および第2結合対メンバーと同じであるかまたは違うものであり、
第1コンジュゲートメンバーは、第1および第2結合対メンバー間の結合を介して第2コンジュゲートメンバーに結合している、実施形態82に記載の方法。
Claims (6)
- (a)4-1BBL免疫共刺激ポリペプチドとアビジン又はストレプトアビジンとを含むコンジュゲート、及び
(b)抗原
を含む組合せ。 - 前記抗原は(a)腫瘍関連抗原、及び(b)感染因子又は感染因子に関連する抗原よりなる群から選択される、請求項1に記載の組合せ。
- 前記抗原は前記4-1BBLコンジュゲートに結合する、請求項1に記載の組合せ。
- 前記抗原はビオチン部分とコンジュゲートすることにより抗原-ビオチンコンジュゲートを形成し、さらに該抗原-ビオチンコンジュゲートは該ビオチン部分を介して前記4-1BBLコンジュゲートに結合する、請求項1に記載の組合せ。
- 前記抗原は前記4-1BBLコンジュゲートと結合しない、請求項1に記載の組合せ。
- 配列番号8のアミノ酸配列からなる、4-1BBL免疫共刺激ポリペプチドとストレプトアビジンとを含むコンジュゲート。
