Description
(a)hES細胞とEND−2細胞との共培養
ヒト胚性幹細胞(hES細胞)(Reubinoffら著、Nature Biotech.第16巻:399〜404頁)を、END−2細胞(Mummeryら著、1985年、Dev Biol.第109巻:402〜410頁)と共培養した。1:1比のダルベッコ最少必須培地(DMEM)とハムF12培地(DF)(FCS7.5%含む)とにおいてEND−2細胞を増殖させた。次に、細胞を、1:5のトリプシン/EDTA(0.125%w/v;50mM resp)中で1週間に2回、継代培養した。hES細胞を、DMEMにおいて、20%のFCS、0.1mMのβ−メルカプトエタノール、1%の非必須アミノ酸、2mMのグルタミン+抗生物質(pen/strep)と共に、マイトマイシン(10μg/ml)処理胚のフィーダー細胞上で培養した。HESを、ジスパーゼ(dispase)で処理し、個々のコロニーを6〜10枚の断片に機械的にスライスし、続いて断片を新しいフィーダー細胞に移すことにより二次培養した。
内臓内胚葉細胞を、ジスパーゼを用いて前述のようにE7.5(E0.5は、膣栓の日正午である)にて、原腸形成しているマウス胚の3つの胚葉から単離した(Roelenら著、1994年、Dev.Biol.第166巻:716〜728頁)。分離した胚葉を、M16培地におけるポリ−L−リシン被覆培養皿上に乗せ、一晩付着させた。翌日、M16をhES完全培地に置換し、胚細胞単離後3日目に、未分化細胞塊「移植片」を、マウス胚からの付着した内胚葉および外胚葉細胞上で培養した。次に、培地を2〜3週間増殖させ、培地を5〜6日毎に新しくした。
前記(a)および(b)に記載した培養を、10日後の収縮筋の領域の存在について評価した。
次に、収縮筋の領域が明らかになった後、培養を、2%パラホルムアルデヒド中で30分間固定し、3回洗い、使用するまでPBS中に貯蔵した。次に、培地を、α−アクチニン抗体(モノクローナル抗体−α−アクチニン(筋節)クローンEA−53、1:1000希釈、Sigma製;二次抗体:ヤギ抗マウス−IgG−Cy3)を用いて、筋肉表現型を確認するために染色した。
(i)hES−END−2共培養
共培養の最初の週の間、細胞の塊を徐々に広げ分化させて、混合形状を有するが上皮様細胞の割合が比較的高い細胞を得る。第2週までに、これらは膨張して流体を満たした嚢胞になる(図1A参照)。これらの間に、細胞の異なるパッチが明らかになり、数日後に収縮するようになる。12〜21日の間に、これらの収縮しているパッチが次第に多く現れる。ガラス基質と組織培養基質との間に明らかな相違はなく、いずれも収縮している筋肉を産生し、これは、3つの独立した実験において示され、収縮筋肉の1または2以上の領域を含むウェルが15〜20%あることが示される。収縮速度は1分当たり約60回であり、高度に温度感受性である。これらの細胞は、α−アクチニンで陽性に染まり、実際に筋細胞であることが確認される(図2)。
培養の第1週中、hES断片を内胚葉の上で培養し、これは徐々に広がり平らになり、12日目に、収縮している筋細胞の第1の領域が明らかになる。これは、END−2共培養において観察される強度の嚢胞形成を伴わないが、血管内皮細胞ネットワークに似ている領域は、培養の端部に現れない。
実施例1で用いたヒト幹細胞(hES)を、E7.5日胚(E0.5は栓の日)から単離された外胚葉上に乗せた。鋭いピンセットとタングステン針を用いて、脱落膜から胚を取り出し、10%FCSを含むHEPES緩衝DMEM中で氷上に維持した。ライヘルト膜を除去した後、受胎産物の胚と胚体外部分を、タングステン針で分離した。結節と原始線条を取り出し、胚部分を、氷上のPBS中の2.5%パンクレアチンおよび0.5%トリプシン中で8分間インキュベートした。インキュベーション後、胚を、氷上の10%FCSを含むHEPES緩衝DMEMに移した。次に、外胚葉、内胚葉および中胚葉を、タングステン針を用いてきれいに単離することができた。
実施例1で用いたヒト幹細胞(hES)を、外胚葉および/または内胚葉細胞上に乗せた。外胚葉または内胚葉とのhESの共培養条件は前述と同じであった。ネットワークにおける血管内皮細胞は、他の体細胞型への分化を伴った。
a)共培養
END−2細胞、P19EC、hECおよびhES細胞を、前述のように培養した(Mummeryら著、1985年、1991年;vanden Eijnden−van Raaijら著、1991年;Slagerら著、1993年;Reubinofら著、2000年)。全ての実験において、ESI(Reubinofら著、2000年)からのhES2細胞株を用いた。共培養を開始するために、実施例1に記載のようにマイトマイシンC(10μg/ml)で1時間処理した有糸***不活性のEND−2細胞培養で、hEC、mESおよびhES用のフィーダーであるマウス胚性線維芽細胞(MEF)を置き換えた。フィーダーに依存しないP19ECとの共培養を、先に記載(Mummeryら著、1991年)のように開始し維持した。次に、培地を2〜3週間成長させ、5日目からの収縮筋肉の領域の存在について評価した。
手術生検からのヒト心房細胞が、抗体染色、電気生理学およびRT−PCRによるイオンチャンネルの特徴付け用の対照として使用された。心臓手術を受ける患者からの同意を得た心臓組織を得た。手術中に定法で除去された心房付属器を、直ちに、氷冷クレープス−リンガー(KR)食塩水に移した。過剰結合組織および脂肪組織を切り取り、滅菌KR溶液で2回洗った。心筋組織を、滅菌鋏で切り刻み、次に、公開された方法(Peetersら著、(1995年)、Am.J.Physiol.第268巻、H1757〜H1764)を用いて3段階酵素的単離手順により分離して個々の細胞を放出した。第1の段階は、37℃で4.0U/mLプロテアーゼ型XXIV(Sigma製、St Louis、MO、USA)を用いて15分間インキュベートすることを含む。次に、組織を、1.0mg/mLコラーゲナーゼおよび0.5mg/mLヒアルロニダーゼを含む溶液に移し、続いて、コラーゲナーゼ(1.0mg/mL)を用いてさらに3回、各々37℃で20分間インキュベートした。組織抽出物を一緒にし、カルシウム濃度を1.79mmol/Lに回復させた。心筋細胞を、10%のFBS、ペニシリン(100U/mL)ストレプトマイシン(100μg/mL)、2.0mmol/LのL−カルニチン、5.0mmol/のクレアチン、5.0mmol/Lのタウリンを加えた組織培養培地M199に移し、50μg/mLのポリL−リシンを被覆したガラスカバースリップ上に直接接種し、一晩培養した。
付着した一次心筋細胞、mES(E14およびR1)およびhES誘導心筋細胞を、Ca2+およびMg2+を含むPBS中の3.0%パラホルムアルデヒドで室温にて30分間固定し、次に、PBS中の0.1%トライトンX100で4分間透過性にした。α−アクチニンおよびトロポミオシン(Sigma製)を含む筋節タンパク質に対するモノクローナル抗体を用いて標準的方法により免疫細胞化学を実施した。ミオシン軽鎖(MLC2a/2v)のアイソフォームに特異的な抗体を用いて、心房細胞と心室細胞(Dr Ken Chienから戴いた)とを区別した(表1)。二次抗体は、Jackson Immunoresearch Laboratoriesから得た。培養した心臓線維芽細胞は、筋節タンパク質用のネガティブコントロールとして役立ち、Zeiss Axiovert 135Mエピ蛍光顕微鏡(Carl Zeiss製、Jena GmbH、Germany)を用いて細胞を可視化した。画像処理ソフトウェアを用いて画像を擬似着色した。
1%ジメチルスルホキシド(RudnickiおよびMcBurney著、1987年)の存在下、凝集プロトコールによりP19EC細胞を収縮筋に分化させた。これらの培養条件で16日後、収縮心房を切除し、RNAを、Trizol(Gibco製)を用いて単離し、M−MLV−RT(Gibco製)を用いて逆転写した。心臓アクチン(Lansonら著、(1992年)Circulation、第85巻、1835〜1841頁)、MLC2v(Meyerら著、(2000年)FEBS Lett,第478巻、151〜158頁)、ERG(Lees−Millerら著、(1997年)、Circ.Res.第81巻、719〜726頁)およびKir2.1(Vandorpeら著、(1998年)、J.Biol.Chem.第273巻、21542〜21533頁)用のプライマーを、前述のように用いた。マウスL型カルシウムチャンネルサブユニットα1c(センス5−CCAGATGAGACCCGCAGCGTAA(配列番号1);アンチセンス5'−GTCTGCGGCGTTCTCCATCTC(配列番号2);GenBank登録番号L01776;生成物寸法745bp)、Scn5a(センス5'−CTTGGCCAAGATCAACCTGCTCT(配列番号3);アンチセンス5'−CGGACAGGGCCAAATACTCAATG(配列番号4);AJ271477、770bp)およびβ−チューブリン(センス5−TCACTGTGCCTGAACTTACC(配列番号5);アンチセンス5'−GAACATAGCCGTAAACTGC(配列番号6);X04663、319bp)用のプライマーを、Vector NTIソフトウェア(InforMax製、North Bethesda、MD、USA)を用いて設計した。
パッチ−クランプ技術の全細胞電圧クランプ構造を用いて33℃で実験を行った。封鎖達成後、電流固定式に活動電位を測定した。Axopatch 200B増幅器(Axon Instruments Inc.製、Foster City、CA、USA)を用いて、自発収縮領域における細胞からのデータを記録した。出力信号を、AD/DAC LAB PC+収集ボード(acquisition board)(National Instruments製、Austin,TX,USA)を備えたペンティアム(登録商標)IIIを用いて2kHzでデジタル化した。抵抗2〜4MΩのパッチピペットを用いた。浴槽培地の組成は、140mMのNaCl、5mMのKCl、2mMのCaCl2、10mMのHEPESであり、NaOHでpH7.45に調節されている。ピペット組成:145mMのKCl、5mMのNaCl、2mMのCaCl2、10mMのEGTA、2mMのMgCl2、10mMのHEPESであり、KOHによりpH7.30に調節されている。
(i)mEC−END−2共培養
END−2細胞との共培養の開始から2日後に、P19EC細胞は自然に凝集し、7〜10日後、凝集塊の多くが、先に記載(Mummeryら著、1991年)のように収縮細胞の領域を含んでいた(図3a)。電気生理学およびRT−PCRは、胚心筋細胞の機能的イオンチャンネル特性がこれらの細胞において発現されたことを示した(図4、表2)。
半定量的RT−PCRにより決められる心臓マーカーとイオンチャンネルmRNA発現との相対水準。示された遺伝子産物について、未分化P19(EC)、分化P19心筋細胞(CMC)および成体マウス心臓(心臓)の同一量のcDNAをPCR増幅した。各産物についての相対水準を示す。
2つの独立したマウスES細胞株(E14およびR1)を、それらの共培養条件への反応について試験した。培養は単一細胞懸濁液として開始されなかったが、3日以内に、最初に接種したよりも大きな凝集塊が、両方の細胞株について明らかとなった(図3b、c)。殆ど同時に、自発的収縮心筋細胞の著しい領域がR1ES細胞培地において明らかとなったが、僅か7日後には、収縮筋肉の(より小さな)領域がE14ES細胞中において見つかった。収縮領域中の細胞は、α−アクチニンで染色した場合に、筋細胞の特徴的筋節バンドパターンを示した(図6d参照)。
ヒトEC細胞株GCT27Xは、フィーダーに依存する全能EC細胞株であり、ヒトES細胞(Peraら著、(1989年)、Differentiation、第42巻、10〜23頁)に類似の特徴を有する。END−2細胞との共培養において、大きな凝集塊の形成が観察された(図3d)。しかしながら、3週間後でも、収縮筋肉の証拠は無かった。
共培養の第1週中に、細胞の小さな塊が徐々に広がり分化して、混合形状を有するが上皮様細胞の割合が比較的高い細胞を得る。第2週までに、これらは膨張して流体を満たした嚢胞になる(図示せず)。これらの間に、細胞の異なる区画が明らかになり、数日後に収縮するようになる。12〜21日の間に、これらの収縮している区画が次第に多く現れる(例えば、図3e)。全体では、15〜20%のウェルが収縮筋肉の1または2以上の領域を含む。収縮速度は1分当たり約60回であり、マウスES誘導心筋細胞と比べて高度に温度感受性である。これらの細胞は、α−アクチニンで陽性に染まり、その筋肉表現型が確認される(図6e)。しかしながら、mESおよびP19EC誘導心筋細胞と対照的に、筋節バンドパターンはあまり定まらないが、培養中で2日しか増殖していない一次ヒト心筋細胞と充分に匹敵している(図5および6a〜c)。一次ヒト心筋細胞は初期に筋節構造を維持するが、標準的培養条件により、劣化が迅速になることが明らかである(図5)。hES培養条件は、心筋細胞に最適ではなく、そのために、hES誘導心筋細胞はその特徴的表現型において同様に劣化を示すことが推定される。培地中の幹細胞から充分に機能的な心筋細胞を得るために、これらの条件を最適化することが必要である。筋節構造における劣化にも拘わらず、hES誘導心筋細胞は数週間にわたって周期的に収縮を続け、活動電位は、図4(c)に示すように電極を凝集塊に導入することにより行われる電流固定電気生理学により検出することができる(図4b)。しかしながら、このように電気生理学を行う、すなわち、単一細胞よりも凝集塊において電気生理学を行うことにより、細胞の群の蓄積された結果である活動電位が得られる。従って、これらは理解が困難であり、心室細胞、心房細胞またはペースメーカー細胞のいずれに帰することが困難である。単一細胞について決めるために凝集塊を分離し再培養する作業が現在進行中である。
心臓発育中に、成体活動電位のその後の相の原因となるイオン電流が生じる順番が、電気生理学的研究において確立された(Dviesら著、(1996年)、Circ.Res.第78巻、15〜25頁)。内向きL型Ca2+電流は、初期心臓胚形成において重要な役割を果たすが、内向きNa2+電流は、出産直前にしか上昇しない(Daviesら著、1996年)。マウスESおよびP19EC細胞は、イオン電流発現において同様のタイミングを示す(Wobusら著、(1994年)、In Vitro Cell.De.Biol.第30A巻、425〜434頁)。P19EC細胞の分化中の分子水準でのイオンチャンネル発現の順序を解明するために、我々は、未分化かつ16日齢の、−40〜−60mVの比較的陽性の膜電位(IK1が無いまたは少し)であるP19誘導心筋細胞の収縮塊から単離されたRNAについてRT−PCRを行った。これらの結果は、mES誘導心筋細胞について先に記載(Doevendansら著、(1998年)、cardiovasc.Res.第39巻、34〜39頁)したように、16日齢のP19心筋細胞がイオンチャンネル発現に関して胎児心筋細胞に似ていることを示している。
(a)方法
i)細胞培養
END−2細胞およびhESを、実施例1に記載のように培養した。全ての実験において、ES Cell International Pte LtdからのhES2細胞株を用いた。共培養を開始するために、マイトマイシンC(10μg/ml)で1時間処理した有糸***不活性のEND−2細胞培養で、hEC細胞用のフィーダーとして、マウス胚性線維芽細胞(MEF)を置き換えた。次に、共培地を6週間まで成長させ、5日目からの収縮筋肉の領域の存在について評価した。肝臓実質細胞に似ている癌細胞株であるHepG2細胞(Knowlesら著、(1980年)Science第29巻:497〜9頁)を、10%FCSを含むDMEM中で培養し、1週間に2回継代培養した。END−2細胞のように共培養を開始したが、HepG2は集密性単層として成長せず、hES細胞がHepG2に選択的に付着した。P19EC細胞の懸滴培養を開始し、先に記載(Mummeryら著、1991年)のように通常のFCSを用いて、DMEM/ハムF12(1:1)中に維持し、一方、ノーザンブロット用のRNAを単離するためのEND−2調整培地の存在または不存在下におけるP19胚体のバルク生成を、前述(Mummeryら著、1991年)のように活性炭処理したFCSの存在下に行って、FCS中にレチノイド様活性を有する可能性のある親油性物質により引き起こされる心筋細胞分化の背景水準を低下させた。電気生理学のために、コラーゲナーゼを用いて収縮性凝集塊を分解し、ゼラチン被覆オーバースリップ上に乗せた。
付着した一次心筋細胞hES誘導心筋細胞を、Ca2+およびMg2+を含むPBS中の3.0%パラホルムアルデヒドで室温にて30分間固定し、次に、PBS中の0.1%トライトンX100で4分間透過性にした。α−アクチニンおよびトロポミオシン(Sigma製)を含む筋節タンパク質に対するモノクローナル抗体を用いて標準的方法により免疫細胞化学を実施した。ミオシン軽鎖(MLC2a/2v)のアイソフォームに特異的な抗体を用いて、心房細胞と心室細胞(Dr Ken Chienから戴いた)とを区別した。Oct−4およびα1c抗体はSigma製である。二次抗体は、Jackson Immunoresearch Labs.製である。
一次組織は、標準的インフォームドコンセント手順に従いUniversity Medical Centre,Utrechtの倫理委員会の承認を受けた患者から得た。成体心筋細胞を前述(Mummeryら著、(2002年)、J.Anat.第200巻:233〜242頁)のように単離し、培養した。胎児心筋細胞を、Langendorffにより灌流された胎児心臓から単離し、穏やかに遠心分離した後に残っている上澄みが、ラミニン被覆オーバースリップに付着した生活力のある心筋細胞を主要量で含んでいたことを除いて同様の方法で培養した。電気生理学のために、細胞を、低カルシウム含量のタイロード緩衝液(Sipidoら著、1998年、Cardiovasc.Res.第37巻:478〜88頁)中に集め、ガラスウェル中で直接パッチクランプを置換した。
RNAを、Trizol(Gibco製)を用いて単離し、M−MLV−RT(Gibco製)を用いて逆転写した。α1c(Van Gelderら著、1999年)、Kv4.3(Calmelsら著、2001年、Ref)およびANF(Kehatら著、2001年、J.Clin.Invest.第108巻、407〜414頁)様のプライマーは前述の如くである。ヒトKvLQT1(センス5'−TTCTTGGCTCGGGGTTTGCC(配列番号7);アンチセンス5'−TGTTGCTGCCGCGATCCTTG(配列番号8);GenBank登録番号AF000571;生成物寸法723bp)およびβ−チューブリン(センス5'−TGGCTTTGCCCCTCTCACCA(配列番号9);アンチセンス5'−CGGCGGAACATGGCAGTGAA(配列番号10);AF141349、369bp)用のプライマーを、Vector NTIソフトウェア(InforMax製、North Bethesda、MD、USA)を用いて設計した。アニーリング温度および増幅サイクル数:ANF、58℃、35サイクル;α1c、56℃、38サイクル;Kv4.3、55℃、35サイクル;KvLQT1、58℃、32サイクル;β−チューブリン、61℃、30サイクル。エチジウムブロミド1.5%染色アガロースゲル上で生成物を分析した。
RNAは、先に記載(Mummeryら著、1990年、Dev.Biol.第142巻:406〜413頁)のようにP19凝集塊から単離され、ポリA+RNAが選択された。

本明細書で記載の実験の大部分は、hES2細胞株(Reubinoffら著、2000年)を用いて行った。細胞を、前述(Mummeryら著、2002年)のように血清含有培地においてマイトマイシンC処理MEF「フィーダー細胞」と共培養することにより未分化状態に維持した。これらの条件下において、培養の塊中の全ての細胞が、oct−4について核染色を示すが、培養の端部における平坦細胞は陰性であった。Oct−4発現は、未分化細胞の表現型特性に関連する。未分化細胞の小さい塊を、新しいマイトマイシンC処理MEFに、またはマイトマイシンC処理END−2細胞の集密性培養に移すことによりhESを二次培養した。これらの条件下で約5日後、上皮細胞が明白になり、次の数日で、流体で満たされた嚢胞になった(図1A)。これらはαフェトプロテインを染色し、胚体外内臓内胚葉であることが分かる。さらに、10日までに、より固体の凝集塊中で周期的に収縮している種々の全体形状(図1B、3e)の細胞の領域が明らかになった(図1A、3e)。12ウェルプレートのウェルの16〜35%が収縮領域を含み、各々を分解し置き換えることにより、3次元構造よりも2次元構造を有する収縮細胞の12個までの新しいコロニーが産生され;これにより、パッチ−クランプ電気生理学によるさらなる特徴付けのために細胞に接触することが容易になる(図4、図9;以下参照)。分解の前後において、hES誘導心筋細胞は1分当たり45〜60回収縮し、時には不規則に収縮し;収縮は、カルバコール、イソプレナリンおよびフェニレフリンのような薬理学的作動薬に反応してアップレギュレーションされる。
心臓特異的イオンチャンネルの発現を、未分化hES細胞および、END−2細胞との共培養の9日および14日後の分化細胞において判定した(図8)。先に他で示されている(Kehatら著、2001年)ように、収縮しているhESに由来する心筋細胞の領域はANFを発現する。心臓特異的L型カルシウムチャンネル(α1c)および一時的外向きカルシウムチャンネル(Kv4.3)のα−サブユニットの発現も検出され、Kv4.3の発現は、収縮の開始より数日間早い。遅延整流カリウムチャンネルKvLQT1用のRNAは、未分化細胞において見られ、これは、初期分解中に消滅し、やや後の段階において再び現れる。
hES心筋細胞の解離し再培養した凝集塊のパッチクランプ電気生理学は、胎児起源の一次ヒト心筋細胞(図9B)に匹敵する範囲の(電気的)表現型が培養中に存在する(図9A)ことを示した。心室様活動電位が主要であった(31決定のうち28)が、心房細胞とペースメーカー様細胞の両方共が存在した(それぞれ、31決定のうち2および1)。注目すべきは、比較的遅い上昇速度(7.0+/−0.8V/s)および低い膜電位(図9A)であり、これは、細胞が、妊娠17週の胎児ヒト心筋細胞と比較しても比較的未熟であったことを示している。細胞が収縮していないが収縮領域から区別できない形状を有する共培養の領域において、電流注入は、繰り返し活動電位および持続する規則的収縮を誘導するのに充分であった。さらに、hES細胞とHepG2細胞との共培養により、hES−END−2共培養におけるものと類似の活動電位を有する心筋細胞が得られた。
我々は、先に(Mummeryら著、1991年)、END−2細胞により24時間調節した培地が、その分化を誘導することができ、それにより、懸滴が収縮筋を含むので、3日間成長後の殆ど全ての凝集塊が10日以内に再び培養されることを示した。正規FCSの存在下に、分化が、背景水準を越えて著しく高められる。背景分化水準は、FCSが活性炭カラム(DCC−FCS)上を通過して残留レチノイドが除去される場合、実質的に0に低下し;さらに、アクチビンにより分化が阻害される(van den Eijnden−van Raaijら著、1991年、Mech.Dev.第33巻:157〜166頁)。END−2CMを、MEF上で培養したhES細胞とmES細胞の両方について試験した(MEFがないと、細胞は間接的に死ぬ/分化する)が、MEFにより分泌された分化阻害活性を超えることはできなかった。P19ECはアッセイ系が好ましい、それは、これらの細胞が、未分化成長についてフィーダーに依存しないからである。これらの実験は、P19EC細胞の2つの独立したクローンにおいて、再培養した凝集塊における心筋細胞の外観の促進において投与量依存的効果を有することを示している(図10A)。より重要なことは、効果が、発生期の中胚葉の分化への初期の効果に関連すると思われ;ノーザンブロット分析は、最初の3日間の凝集相における初期中胚葉マーカーBrachyury T(Hermann著、1991年)をEND−2 CMがアップレギュレーションし(図10B)、その水準が培養直後は維持されるが、9日後には、Brachyury Tがもはや検出不可能であることを示している。さらに、Brachyury Tの誘導が、アクチビンのさらなる存在により阻害(図10B)、これは、凝集塊中の収縮筋肉の形成を阻害する性能に一致している。Brachyury Tの一時的発現が、マウス胚の原腸形成中の初期の中胚葉分化中に発見される発現に類似している。
幹細胞誘導心筋細胞が心臓機能を回復する性能を試験するために、マウスにおいてMIモデルを開発した。ペントバルビタール麻酔した成体マウスにおいて、胸部を胸骨中央から開く。前下降枝を確認し結さつする。首尾良い手順により、遠位心筋の変色を誘導する。胸部を、3回の縫合により閉じ、動物を回復させる。合計で、17匹の動物を手術した。7匹は、縫合の位置決めを含む偽手順を行い、10匹は結さつした。MIの4週間後、同じ薬剤を用いて腹腔内注射によりマウスを再び麻酔した。血行力学的研究のために、動物をインキュベートし、げっ歯類の人工呼吸器に接続した(Hugo Sachs Electronics、March−Hugstetten Germany)。機器を使用して胸部を閉じてカテーテルを頸静脈に導入した。
Claims (6)
- 未分化のhES細胞を心筋細胞へと分化させる方法であって、
hES細胞の未成熟心筋細胞への分化を誘導するために、マウス胚に由来する内胚葉細胞又は内胚葉様END−2細胞の存在下に、hES細胞を培養すること
を含む方法。 - 未成熟心筋細胞を含む細胞集団を得る方法であって、前記未成熟心筋細胞は細胞集団における未分化のhES細胞に由来するものであり、前記方法は、請求項1の未分化hES細胞の分化を誘導することを含む、方法。
- 前記内胚葉細胞が、マウス原腸形成胚に由来する請求項1の方法。
- 前記内胚葉細胞が、マウス7.5日胚に由来する請求項1〜3のいずれか1項の方法。
- マウス胚に由来する内胚葉細胞又は内胚葉様END−2細胞を実質的に集密な単層にまで予備培養することと、
前記マウス胚に由来する内胚葉細胞の単層又は前記内胚葉様END−2細胞の単層の存在下で、hES細胞を共培養することと
をさらに含む請求項1〜4のいずれか1項の方法。 - 細胞集団が未成熟心筋細胞からなる請求項2〜5のいずれか1項の方法。
