Description
The present invention relates to fluorescent nanoparticles and a method for observing biological materials using the same.
It is known that silicon nanoparticles having a nano-sized crystal have a crystal size-dependent fluorescence / luminescence property in the visible region. This fluorescent light emission is characterized by being less susceptible to light degradation and fading very slowly compared to conventional fluorescent probes such as organic fluorescent dyes and fluorescent proteins. It is known that such fluorescent silicon nanoparticles can be produced by an electrochemical technique (Patent Document 1).
However, in the conventional manufacturing method, the yield of silicon nanoparticles is not sufficient, the operation is complicated, and the storage period can be stored in an aqueous solution for only a few days. Therefore, an object of the present invention is to produce fluorescent silicon nanoparticles with a high yield by a simpler method, having high fluorescence intensity and hydrophilicity and capable of being stored for a long time even in an aqueous solution. Provided is a method for observing a biological material using the produced fluorescent silicon nanoparticles.

Exposing to the vapor of the third mixed solution.
The method for producing fluorescent silicon nanoparticles according to the invention includes a step of applying a first mixed solution of hydrofluoric acid solution and nitric acid to a silicon substrate, and a silicon substrate having the first mixed solution applied thereto. A step of applying a second mixed solution of hydrofluoric acid solution , nitric acid and water , a silicon substrate on which the second mixed solution is applied , and a third base of hydrofluoric acid solution and water. 1.
1. Production of Porous Silicon <Step 1> First, a
The mixing ratio of HF and HNO 3 in the mixed solution used here is in the range of HF: HNO 3 = 2: 1 to 1: 2 in the volume ratio. If the ratio of HNO 3 in the mixed solution is too small, the etching of the silicon substrate does not proceed sufficiently, and if it is too large, the reaction rate is too fast and uniform etching cannot be performed.
Further, if the silicon substrate is completely immersed in the mixed solution at this stage, the etching rate becomes excessively high and porous silicon is not uniformly formed. Therefore, it is preferable to immerse a part of the silicon substrate in the mixed solution as described above.
The time for immersing one end of the silicon substrate in the mixed solution is in the range of 0.5 to 5 seconds. If the immersion time is short, the etching does not proceed uniformly and sufficient porous silicon is not formed. On the other hand, when the immersion time is too long, the formed porous silicon is eluted. This treatment can be performed at room temperature.
<Step 2> Next, as shown in FIG. 2, the
The time for dipping in the mixed solution can be appropriately selected depending on the intended porous silicon and the composition of the mixed solution to be used, but is usually in the range of 2 to 10 minutes. This treatment can be performed at room temperature.
In addition, you may perform the process which immerses a silicon base | substrate in a mixed solution using the several mixed solution from which composition ratio was changed in multiple steps.

The ultrasonic treatment time of the
Further, this treatment can be performed by putting several silicon substrates in a container at a time. The number of silicon substrates that can be processed at one time is not limited because it depends on the container used.
Moreover, a high concentration silicon nanoparticle suspension can be obtained by performing the operation of
3. Surface Modification of Silicon Nanoparticles with Allyl Compound <Step 1> The surface modification of silicon nanoparticles obtained by the above method can be performed by allowing an allyl compound to act. Thereby, the functional group of the allyl compound can be introduced onto the surface of the silicon nanoparticle. Examples of the functional group possessed by the allyl compound include an amino group, a thiol group, a methyl group, and an ethyl group, and among them, an amino group and a thiol group are preferable.
First, the solvent in the silicon nanoparticle suspension is completely distilled off with an evaporator or the like, and allylamine or allyl mercaptan is added and suspended therein to obtain a
The concentration of the platinum catalyst in the reaction solution depends on the type of allyl compound, but is usually in the range of 0.1 to 100 mM. If the concentration is low, the modification with the allyl compound does not proceed sufficiently. If the concentration is too high, the platinum catalyst cannot be completely removed in the post-treatment operation described later. The kind of platinum catalyst is appropriately selected depending on the reaction system.
The reaction time depends on the type of allyl compound, but is usually in the range of 1 to 36 hours. If the reaction time is short, the modification with the allyl compound does not proceed sufficiently, and if it is too long, the modification is performed non-specifically.
The allyl compound is not limited to those described above, and is appropriately selected from allyl alcohol, allyl bromide, allyl chloride, allyl isothiocyanate, 1-butene, and the like.
<Step 2> The platinum catalyst and the unreacted allyl compound are removed from the reaction solution obtained in Step 1 above. Along with the solvent in the reaction solution, the unreacted allyl compound is completely distilled off with an evaporator or the like, and an organic solvent such as hexane is added to perform ultrasonic treatment. Thereby, since the silicon nanoparticle surface-modified with the allyl compound and the platinum catalyst are separated, it is possible to recover only the supernatant silicon nanoparticle.
By repeating this operation about 2 to 10 times, the allyl compound and the platinum catalyst can be completely removed. The number of repetitions is not limited because it depends on the type of allyl compound, but the amount of silicon nanoparticles recovered decreases as the number of repetitions increases. The absorption and fluorescence spectrum of the recovered silicon nanoparticle suspension can be measured with an absorption spectrophotometer and a fluorescence spectrophotometer.
The organic solvent is not limited to hexane, and cyclobenzene, cyclohexane, dichloromethane, heptane, octane, nonane, decane, or the like may be used.
使用目的によるため限定するものではないが、通常はPBS(phosphate-buffered saline)等のリン酸緩衝液が使用される。

4). Purification of silicon nanoparticles When silicon nanoparticles modified with high-purity allyl compounds are required, for example, silicon nanoparticles recovered by filtration can be purified by gel filtration high-speed chromatography (HPLC). . Although it does not limit for the purpose of use, a phosphate buffer solution such as PBS (phosphate-buffered saline) is usually used.
5. Biomolecule labeling using silicon nanoparticles Silicon nanoparticles modified with an allyl compound can label biomolecules by a known method. For example, the functional group of the silicon nanoparticles modified with the allyl compound and the functional group of the biomolecule may be covalently bonded in the presence of another compound, if necessary, via a compound such as a crosslinking agent, or Ion bond.
より具体的には、例えば、粒子表面にアミノ基をもつシリコンナノ粒子を、カルボキシル基を持つ蛋白質やペプチドに、架橋試薬EDC(1-ethyl-3-[3-dimethyl-aminopropyl]carbodiimide HCl)を用いて標識することが可能である。 More specifically, for example, a silicon nanoparticle having an amino group on the particle surface, a protein or peptide having a carboxyl group, and a crosslinking reagent EDC (1-ethyl-3- [3-dimethyl-aminopropyl] carbodiimide HCl). Can be used to label.
また、SPDP(N-succinimidyl 3-[2-pyridyldithio]propionate)や、2−イミノチオラン塩酸塩などを用いて導入した、チオール基とアミノ基を持つ蛋白質やペプチドに、EMCS(N-[ε-Maleimidocaproyloxy]Succinimide Ester)のような架橋剤でマレイミド基を導入し、そこへ粒子表面にアミノ基をもつシリコンナノ粒子を反応させて、標識しても良い。 In addition, EMCS (N- [ε-Maleimidocaproyloxy) was introduced into proteins and peptides having a thiol group and an amino group introduced using SPDP (N-succinimidyl 3- [2-pyridyldithio] propionate) or 2-iminothiolane hydrochloride. A maleimide group may be introduced with a cross-linking agent such as [Succinimide Ester], and silicon nanoparticles having amino groups on the particle surface may be reacted therewith for labeling.

In the observation with the fluorescence microscope, the labeled cells (+) phagocytosed with the silicon nanoparticles showed fluorescence emission of 535/50 nm with excitation light of 470/40 nm. Further, the fluorescence emission could be observed over a long period of 3 days or longer. In addition, visually, the fluorescent silicon nanoparticles of the present invention were observed to have a
According to the results of analysis by the flow cytometer, there is a clear difference in fluorescence intensity between unlabeled cells (−) that do not phagocytose silicon nanoparticles and labeled cells (+) that have phagocytosed. The particles were able to label all cells.
As described above, the present invention has been described in detail by way of examples.
Since the fluorescent silicon nanoparticles of the present invention can be produced by a very simple operation of etching by a chemical method, that is, only a surface treatment in HF and a recovery treatment using ultrasonic waves, the yield is high. It can be manufactured in large quantities. Moreover, by modifying the surface with an allyl compound, the silicon nanoparticles exhibit hydrophilicity, can be stably stored for a long time in an aqueous solution, and can also be bound to biomolecules.
In addition, according to the thus produced quantum size fluorescent silicon nanoparticles, biomolecules such as proteins do not cause cross-linking with other molecules, and in the living cell or on the cell surface, 1 cell can be labeled, and observation on a very small scale can be realized. In addition, due to the slow discoloration, the behavior of cells within an individual can be observed over a long period of time. Furthermore, compared to conventional methods such as quantum dots, it is safer for the living body, elucidates the functions of molecules, searches for various diseases caused by abnormal functions of these molecules, and develops treatments. Can be greatly expected.
The above embodiment and experimental examples can be variously modified within the scope of the technical idea of the present invention.
For example, porous silicon is produced by the above method, and the silicon substrate surface is modified with an allyl compound. Thereafter, the amorphous silicon film may be removed with an aqueous HF solution, and the silicon nanoparticles may be separated (collected) by ultrasonic treatment. In this way, silicon nanoparticles modified with a larger amount of allylamine can be produced.
The labeling target is not limited to the above. Considering the hydrophobicity and functional group of the fluorescent silicon nanoparticles of the present invention, biomolecules such as receptor ligands including nucleic acids, proteins, peptides, lipids, sugar chains, vitamins, agonists / antagonists, bioactive molecules, It is obvious that the drug molecule can be labeled and observed. Further, by using these, it is possible to label and observe individuals, tissues, organs, and intracellular organelles.
