合成は、サイクロトロン標的から移送されたアニオン交換カラム(トラップアンドリリースカラム(DW−TRC) D and W, Inc., Oakdale, TN, USA)由来の[18F]フルオリドではじめられる。カラムは水溶性カリウムカルボネート(0.3mlに2.3mgが溶解している)で、Kryptofixを含む5ccバイアルへと溶出される。Kryptofix、ホスホニウムカルボネート、[18F]フルオリド混合物は120℃で乾燥され、0.5mlのアセトニトリル中に7mgのプロピルジトシレート(アルドリッチ)が添加される。80℃で5分間加熱した後、0.5mlのトルエン中の21mgトリフェニルホスフィン(アルドリッチ)が加えられる。アセトニトリルは留去され、トルエン混合物は3−5分沸騰するように熱せられる。トルエンを留去させてバイアルを冷やした後、0.5mlの高圧液体クロマトグラフィー(HPLC)溶液[アセトニトリル:水が35:65(0.1Mの蟻酸アンモニウム)]がバイアルへ加えられる。混合物は0.45μmのテフロンHPLCフィルター(アルテック13mm)を通じてろ過され、調整されたHPLCカラム(ウオーターズノバパックC−18 6μM、7.8x300mm)へ精製のために7ml/分で添加される。生成物は、溶液の添加と除去ができるように変えられた回転エバポレーターで集められ、HPLC溶剤は留去され、滅菌された通常の生理食塩水に放射性標識されたホスホニウム塩が溶解される。原料の[18F]フルオリドから算出される[18F]FPTPの集められた全体の消失放射化学的収率は、12パーセントである。滅菌したバイアルへの滅菌したろ過(ポールゲルマン 0.2μm トフリン)の後、溶液は分析用HPLC[アセトニトリル:水が40:60(0.1Mの蟻酸アンモニウム)、ウオーターノバパックC−18 60A 4μm、3.9x150mm]にて3ml/分の速度で処理し、、3―フルオロプロピルトリフェニル−ホスホニウムブロミド[物理学的データ:融点313−316、C;1H NMR(CDCl3、δ)1.81−2.17(m、2H)、4.01−4.11(m、2H)、4.72−4.75(m、1H)、4.87−4.90(m、1H)、7.69−7.88(m、15H)]の冷たい標準的な特徴をもつ溶液の既知の濃度で、放射化学、化学的純度、及び特異的な活性がチェックされる。本合成は式1において要約される。
[18F]FETPは、上述の実施例1において開示された[18F]FPTPの調製に使用された方法に従って調製された。
[18F]FBTPは、上述の実施例1において開示された[18F]FPTPの調製に使用された方法に従って調製された。
カリウムカルボネート/クリプトフィックスにおいて[18F]フルオリドを集めて乾燥した後(実施例1に記載されたように)、0.2mlのジメチルスルフォキシド(使用に先立ってバリウムオキシドから真空蒸留された)中のトリメチルアンモニウムベンザルデヒドトリフレート塩(7mg)が、混合物へ添加された。120℃で5分加熱した後、アルデヒドは5mlのHPLC水で希釈され、C−18固相抽出カートリッジ(ウオーターズC−18プラスSep−Pak)上で集められ、10mlのHPLC水で洗浄され、カートリッジを3分間流れる不活性ガスで乾燥される。アルデヒドは2mlのジエチルエーテル(アルドリッチ)でカートリッジから溶出され、それをアルコールへと還元するために、基本的なアルミナ(アルドリッチ−200−400mg)上で10%のナトリウムホウ素ヒドリドを通過させられる。アルコールは1mlのメチレンクロライド内のトリフェニルホスフィンジブロマイド(75−100mg)と5分間混合され、続いて[18F]フルオロベンジルブロマイドへと変換される。シリカ固相抽出カートリッジ(ウオーターシリカクラシックSep−Pak)を通過させ、1mlのメチレンクロライドで洗浄後、[18F]フルオリネーテドベンジルブロマイドは、5mlバイアル内で0.5mlのトルエンに溶解された21mgのトリフェニルホスフィン(又はその類似体)へと加えられる。メチレンクロライド/エーテル溶液は、不活性ガス流で低温下留去され、バイアルはふたをして3−5分温められる。トルエンを留去してバイアルを冷却した後、0.5mlの高圧液体クロマトグラフィー(HPLC)溶媒[アセトニトリル:水が50:50(0.1Mの蟻酸アンモニウム)]がバイアルへ加えられる。混合物は0.45μmのテフロンHPLCフィルター(アルテック13mm)でろ過され、精製のために調整されたHPLCカラム(ウオーターノバパックC−18 6μm、7.8x300mm)へ7ml/分で注入される。生成物は溶液の添加及び除去をできるように変えられた回転エバポレーター上で集められ、HPLC溶媒は留去され、放射性標識されたホスホニウム塩が滅菌された通常の生理食塩水に溶解される。滅菌したバイアルへの滅菌したろ過(ポールゲルマン 0.2μm トフリン)の後、溶液は分析用HPLC[アセトニトリル:水が40:60(0.1Mの蟻酸アンモニウム)、ウオーターノバパックC−18 60A 4μm、3.9x150mm]で、実施例1同様にp―フルオロベンジルトリフェニルホスホニウムイオンの公知の標準物質と比較して、3ml/分の速度で、放射化学、化学的純度、及び特異的な活性がチェックされる。原料の[18F]フルオリドから算出される[18F]フルオロベンジルトリフェニルホスホニウムイオンの集められた全体の消失放射化学的生産率は、14パーセントである。[物理学的データ:融点313−316、C;1H NMR(D6−dmso、δ)5.17−5.21(d、2H)、6.99−7.08(m、4H)、7.67−7.94(m、15H)]。本合成は式2において要約される。
放射性標識されたハロアルキルホスホニウムイオン誘導体は、実施例4に開示されたように18F−フルオオベンジルハライドの合成により調節される。放射性標識されたベンジルハライドは、放射性標識されたリンを製造するためにジフェニルホスフィンへと付加され、次に放射性標識されたリンは適切なハロアルキルイオジドとの反応によりハロアルキルホスホニウムイオンへと変換されるであろう。放射性標識されたクロロアルキルホスホニウムイオンの合成は式3に要約される。様々な長さ、分岐及びハライド置換部位を有するアルキル鎖に付加されたクロロ、ブロモ、ヨウ素を含む他のハロアルキル種もまた調製されるであろう。
ピリジル環を含む放射性標識されたホスホニウムイオンは、式4に示される一般化された反応によって調製される。放射性合成には18F−フルオロアルキル(実施例1)と上記においてトリピリジルリンと共に記載されたベンジル部分(実施例4)の反応を含む。
放射性標識されたホスホニウムイオンの調製のための実施例1乃至6に記載された工程はまた、式5で説明される四級アンモニウムイオンの調製においても適用可能である。四級アンモニウムイオンは、ホスホニウムイオンに匹敵する生体内分布性を有する。
R=フェニル及び/またはアルキル部分
本発明の塩の分配に適する一つのイメージングプロトコールには、本塩を静脈投与し、ベッドポジションあたり数分静止した(static)スキャンを得ることが含まれる。正確なスキャン時間は、患者の大きさ、塩の投与量、イメージされる組織の性質により様々であろう。しかしながら霊長類、特にヒトのイメージングパラメータは放射線医学分野で他の放射性医薬と共にPET及び/またはSPECTイメージングに精通した当業者により必要に応じて修正されるであろう。
18F−FBnTPの放射化学的純度と生体内における安定性がクロマトグラフィーにより測定された。
方法:
18F−フルオロベンジルトリフェニルホスホニウム(18F−FBnTP)の調製:オルト位がF−18で標識されたFBnTPの合成は以下に概要を開示する。F−18フルオリドをクリプトフィックス内で(フルオロアルキル誘導体と共に)集めた後、アセトニトリル中のニトロベンズアルデヒドが混合物へ添加された。加熱後、アルデヒドはアルコールへと還元され、続いて放射性標識されたベンジルハライドへと変換される。フルオリネートされたベンジルハライドは、トリフェニルリン又はその類似体とトルエン中で反応させられる。混合物は精製され、上述されたように品質の調整が行われる。
これらの放射性クロマトグラムはF−18−FBnTPが高い放射化学的純度で標識され、生体内におけるマウスの追跡子の静脈注射後30分以上安定であることのよい証明となる。
方法:マウスは尾の血管を通じて25μCiのF−18−PhCを5及び30分p.i.で注入され、右及び左の大腿骨が除去され、骨の放射活性が標準(注入された投与量の1:100)とともにγカウンターで計測された。並行して他のマウスの集団はフリーのフルオリド(F−18)を注入された。それぞれの集団のマウス3匹がそれぞれの時間ポイントにおいて調べられた。放射活性は注入された投与量のパーセンテージ(%ID)として表され、総骨の取り込みは大腿骨x20における活性として計算された。
結果:図2は5及び30分p.i.にける総骨取り込みを表す。F−18のみを注入したマウスにおける骨取り込みと比較した、F−18−PhCのわずかな骨取り込みは、ホスホニウム化合物のフルオリネーションの安定性を示している。骨におけるF−18−FBnTPの最少骨取り込みは自由なフルオリドが欠乏していることを示し、フルオリネーションの安定性を意味している。
F−18−FBnTPの細胞取り込みのMMP−依存性が、MMPを選択的にだめにする公知のプロトノフォアであるCCCPを用いて調べられた。
方法:ヒト肺がんH549細胞(10/ml)は0.1μCi/mlのF−18−FBnTPと共に約30分間インキュベートされた。懸濁液サンプル(1ml)はエッペンドルフチューブへ移され、370℃バス上におかれた。様々な濃度のCCCP(30,60,90,120μM)が懸濁液へ加えられた。CCCPとの30分のインキュベーション後、エッペンドルフチューブは1分間遠心され、ペレットと上澄み液の活性が即座に測定された。
結果:図3は様々な濃度のCCCPの存在下でのF−18−FBnTPの細胞への取り込みを表す。CCCPはF−18−FBnTP細胞性取り込みの投入量に対する減少依存性を誘導した。F−18−FBnTP細胞性取り込みの非常に多くの部分(86%)は、MMP−依存性である。
方法:新規なホスホニウム化合物F−18−フルオルベンジルトリフェニルホスホニウム(FBnTP)とF−18−フルオプロピルトリフェニルホスホニウムカチオン(FPTP)の生体内分布が成人マウスにおいて調べられ、C−11−トリフェニルメチルホスホニウム(TPMP)、テトラフェニルホスホニウム(TPP)そしてTc−99m−セスタミビル(MIBI)追跡子と比較された。F−18−FBnTPは前述のように調製された。FPTPの調製は以下に開示される。TPMPは既に開示されているように調製された(Madar I, J Nucl Med. 1999; 40:1180-5)。TPPとMIBIは(NENおよびデュポンから各々)から購入された。マウス五匹のうちの三匹が、個々の追跡子の生体内分布を調べるために使用された。無麻酔下で動物は追跡子溶液を静脈内投与され(FBnTP又はFPTP、TPMP25μCi、MIBI40μCi、TPP2μCi、全て容量0.2mLの生理食塩水中)、注入の後60分後に頚椎脱臼で殺された。興味深い器官と組織は取り除かれ、標準物質と共に(1:1000)放射活性カウンターにより計測された(LKB Wallac, 1282 Compugamma CS)。
発ガン物質ニトロソメチルウレア(NMU)は、メスのラットの乳腺においてのみ、ガン性腫瘍を誘導する。それゆえ同所性NMU***腫瘍は、がん細胞を有する乳腺と、健常乳腺における放射活性の蓄積を対比することにより腫瘍選択的追跡子を評価するための優れたモデルである。フリュンドコンプリートアジュバント(FCA)はラットにおいてよく研究された炎症剤である。NMU−ベアリングラットにおけるFCA炎症の誘導は、炎症からガンを区別化する追跡子能力を直接的に定量できるようにする。
方法:2x106ヒト肺ガンA549細胞が12匹のヌードマウスに皮下(s.q.)接種する。腫瘍の大きさが約5−10mmに達したときに、6匹のマウスにはタキセターが静脈内投与され、他の6匹のマウスは対照とされた。取り込みアッセイはそれから48時間後に行われた。F−18−FBnTP25μCiは、静脈内投与され、腫瘍及び筋肉組織は60分後に取り出された。
結果:図5は筋肉に対してノーマライズされた腫瘍活性を示す。タキセターは非処置のマウスに比べて、腫瘍におけるF−18−FBnTPを約50%減少させた。
方法:オスのラットは精巣除去され、4日後に取り込みアッセイが行われた。前立腺の腹面部(VP)、前面の(AP)、背面の(DLP)裂片が後ろ足の筋肉と共に取り出された。
組織サンプルにおいて追跡子活性を計測後、TUNEL染色が行われ、腹面部及び前の裂片におけるアポトーシス細胞画分が測定された。
結果:図6のデータは、筋肉に対してノーマライズされた、9匹の処置された及び8匹の対照ラットの平均である。精巣除去は腹面において裂片特異的減少を誘導したが、前の及び背面の裂片においては誘導しなかった。これらの知見は染色ヒストロジカルセクションにおいて計測されたアポトーシス細胞のフラクションと一致している。AP裂片では細胞のたった3.3%±1.7%がDNAラダリングを示した(図8)のに対して、VP裂片においては、細胞の12.4%±3.8%がDNAラダリングを示した(図7)。
上述したデータはF−18−PhCが動物体におけるアポトーシス過程を検出できることを示す。さらに前立腺においては、F−18−PhC蓄積における変化が、標的組織におけるアポトーシスの程度と相関している。これらのデータは、F−18−PhCが前立腺ガンの、そしておそらく同様に他のタイプのガンにおける化学療法の有効性を測定するためのPET追跡子として適していることを示唆する。しかしながら、アポトーシスの程度を正確に報告できる追跡子の能力は、腫瘍に対する追跡子の選択性に依存するだろう。この問に答えるために、F−18−FBnTPの腫瘍選択性が前立腺ガンの同所性モデルにおいて調べられた。
我々は、PET(Kraws、1994)を用いてイヌにおける局所的心筋流を評価するためのC−11−トリフェニルホスホニウムカチオン(TPMP)の性能を調べた。
方法:C−11−TPMP4mCiは静脈投与され、増加する持続性(15秒から20分)のダイナミックなイメージが、85分全時間を通じて得られた。イメージはGE 4096+PETスキャナー(15スライス、6.5mmのスライス厚さ)において得られた。イメージはバックプロジェクションを使用して再構築され、減衰を修正された。方法の詳細な説明は、Kraus,1994において開示される。
結果:投与後5,30、60分での心臓の軸上の部位が図10に示される。これらのイメージは、周囲の肺組織と非常に対照的に心筋をよく視覚化する。安定水準に達した間、心臓/肺の比は>14:1であり、心臓/血液は>100:1であった。
方法:モングレル犬(BW=35kg)は、F−18−FBnTPを3−4mCi投与された。イメージはGE4096+スキャナー(15スライス、6.5mmスライス厚さ)で得られた。増加継続(1秒から20分)のPETスキャンは、投与後85分の全時間を通じて得られた。最初の3分間は、数秒ごとに動脈の血液サンプル(容量0.5ml)が集められ、イメージング調査の残りの時間では徐々に間隔が広げられた(1乃至10分)。F−18−FBnTPの血管および心筋の反応速度論はROI法を用いて分析された。
結果:F−18−FBnTPは血液プールから迅速に消えることを示した(図13)。F−18−FBnTPは心筋において迅速に蓄積し、数分で平衡に達し、それはスキャン時間の間、維持された(図14)。迅速な取り込みが、左及び右の心室の両方で見られた(図15)。対照的に、F−18−FBnTPは、隣接した肺と同様に心房から迅速に消失した(図16)。注入後60−85分での、心筋の心房及び肺に対する比は、>15:1であった。結果的に、F−18−FBnTPは、優れた視覚的鮮明さを有する高い対比の心臓のイメージを提供した。
4週間にわたるモングレル犬の心臓の早い速度(210bpm)のペーシングは、心臓疾患のよく確立されたモデルである。このモデルの利点は、心筋障害が、冠動脈の狭窄や関連した虚血を伴わないアポトーシスのみを含むことである。それゆえ、このモデルでは、FBnTP取り込みにおけるアポトーシスの影響を取り出すことができる。
方法:犬の調製及びデータ取得は上述したように行われた。モングレル犬は肋骨内(case)にペースメーカーを装着され、FBnTP PETスキャン(上述した)に供された。ベースラインスキャンに続いて、犬の心臓は4週間210bpmのペースにされ、二番目のスキャンが得られた。
結果:図17は、4週間のペーシングの前及び後でのFBnTPの短軸イメージを示す。ペーシングはFBnTP取り込みにおいて劣化した壁のほとんどを通じて、40−60%の顕著な取り込み減少をもたらした(図18−19、表3)。PETスキャンがゲートされなかったにも関わらず、FBnTPの優れた潅流能力とその結果の心筋と左心室壁のすばらしい鮮明さのおかげで、左心室壁が薄くなることと、左心室の膨張を含む心臓疾患に典型的な心筋層のペース誘導された再モデリングが鮮明に見られるであろう。
心筋ページングは劣化した壁において40乃至60%の顕著な(p<0.001)減少を誘導した(表3参照)。
Claims (28)
- 少なくとも一つの薬学的に許容されるアニオン及び、少なくとも一つの化学式I、
Eはリンであり、
X1、及びX2は、それぞれ独立して、任意に置換されたアリール、任意に置換されたヘテロアリール、又は任意に置換されたアラルキルであり、
X3は、任意に置換されたハロアルキル、任意に置換されたアリール、任意に置換されたヘテロアリール、又は任意に置換されたアラルキルであり、
X4は、任意に置換されたアルキル、任意に置換されたアルケニル、任意に置換されたアルキニル、任意に置換されたハロアルキル、任意に置換されたシクロアルキル、任意に置換されたアラルキルであり、X4は少なくとも一つの18Fを含むものである。)
で表されるカチオンからなる化合物。 - X4が、一つ若しくはそれ以上の18Fで置換されたC2−6アルキル、又は一つ若しくはそれ以上の18Fで置換されたベンジルである、請求項1に記載の化合物。
- カチオンが次の化学式III、

p、q、rはそれぞれ0乃至5から選択された整数であり、
R1は任意に置換されたアルキル、任意に置換されたアルケニル、任意に置換されたアルキニル、任意に置換されたハロアルキル、任意に置換されたシクロアルキル、又は任意に置換されたアラルキルであり、R1は、少なくとも1つの18Fで置換されており、
R2、R3、R4はそれぞれ独立して、水素、ハロゲン、シアノ、ニトロ、任意に置換されたアルキル、任意に置換されたアルケニル、任意に置換されたアルキニル、任意に置換されたアルコキシ、任意に置換された(シクロアルキル)アルキル、任意に置換されたアルキルチオ、任意に置換されたアルキルスルフィニル、任意に置換されたアルキルスルホニル、及び任意に置換されたモノ又はジアルキルカルボキサミドからなる基から選択されたものである。)
で表されるものである、請求項1又は2に記載の化合物。 - R1が、少なくとも1つの18Fで置換されたアラルキルである、請求項3に記載の化合物。
- R1が、ω−18F−C2−6アルキルである、請求項3に記載の化合物。
- Ar1、Ar2及びAr3の少なくとも1つが、アリールである、請求項6に記載の化合物。
- Ar1、Ar2及びAr3の少なくとも1つが、フェニルである、請求項6又は7に記載の化合物。

18F−3−フルオロベンジル−トリフェニルホスホニウム。 - 薬学的に許容されるアニオンと、以下からなるグループから選択されるカチオンからなる、請求項1〜10のいずれかに記載された化合物;
18F−2−フルオロエチル−トリフェニルホスホニウム、
18F−2−フルオロエチル−トリ−オルト−トリルホスホニウム、
18F−2−フルオロエチル−トリ−メタ−トリルホスホニウム、
18F−2−フルオロエチル−トリ−パラ−トリルホスホニウム、
18F−3−フルオロプロピル−トリフェニルホスホニウム、

- 請求項1〜12のいずれかに記載の化合物、及び薬学的に許容される担体を含む、患者への薬剤投与プロトコールの治療上の有効性を評価するための医薬組成物。
- 請求項1〜12のいずれかに記載の化合物、及び薬学的に許容される担体を含む、炎症の存在下で、悪性細胞及び腫瘍を選択的にイメ−ジするための医薬組成物。
- 請求項1〜12のいずれかに記載の化合物、及び薬学的に許容される担体を含む、ミトコンドリア膜電位(Δφm)を測定するための医薬組成物。
- 請求項1〜12のいずれかの化合物を含んでなる、放射線によってイメージングをするためのイメージング剤。
- イメージング剤が、ミトコンドリア膜電位(Δφm)を測定するものである、請求項17に記載のイメージング剤。
- イメージング剤が、ミトコンドリアの機能低下をイメージングするものである、請求項17又は18に記載のイメージング剤。
- イメージング剤が、ガンをイメージングするものである、請求項17〜19のいずれかに記載のイメージング剤。
- イメージング剤が、ガン細胞のミトコンドリアに選択的に蓄積するものである、請求項20に記載のイメージング剤。
