Description
【発明の属する技術分野】
本発明は、安定化した変性蛋白質及びこの製造方法に関する。より詳しくは、β−1,3-グルカン主糖鎖に規則的なβ−1,6-分枝を有するグルカンと結合され、その活性を維持しつつ、改善された安定性を有する蛋白質及びこの製造方法並びにこの安定化した蛋白質を含有する外用剤組成物に関する。

【従来の技術】
蛋白質は、すべての生体機序において直接的な触媒作用をするか、情報伝達をする物質であって、その種類及び効果が多様であるので、医薬、食品、化粧品等様々の分野において広く使用されてきた。しかしながら、このような基本的な機能性にも拘わらず、構造的不安定性により使用するに多くの制約を受けており、特に化粧品等のような外用剤分野では、長期間保存の問題、皮膚刺激の問題、皮膚への作用性の問題、製造の容易さの問題等のような問題点により今まで充分に活用されていないことが実情である。

従って、このような問題解決の一環として、産業的に利用される固定化方法又は化学的変形方法の適用及び改善が進行されたが、化粧品等のような外用剤に適用するには未だ足りないことが現状であり、これを改善するための適切な材料及び結合方法の選定が持続的に要求されている。

蛋白質を固定化又は安定化させて、安定化した蛋白質を製造する方法として知られた例は、次のようである。
USP4,556,554 号には、酵素を機能性ポリマーに固定及び結合させる方法により固定化された酵素を含有する皮脂抑制用化粧料組成物が開示されているが、かかる固定化方法は、一般的な固定化方法であって、実際問題を解決するための特定的な方法を提示していない。

また、T. Masunaga 等(IFSCC, Yokohama, A205, p483-501) には、蛋白分解酵素にポリエチレングリコール等を化学結合させて、安定度又は刺激度を改善する方法が開示されているが、この方法は、長期間保存するに問題があり、製造方法も複雑である。

また、USP5,230,891 号には、デキストラン、アルギン酸、カラゲニン(carrageenin) 等のポリサカライドと三塩化シアヌルとを反応させて、トリアジン−リングが結合されたポリサカライドを得、これをプロテアーゼと反応させて、プロテアーゼを安定化させる方法が開示されているが、依然として製造方法が複雑である。

さらに、EP0,803,257A2 号とJP平4-141097号には、各々多糖類酸化による蛋白質結合方法で酵素を安定化させ、これを医薬用器具の改善、反応器表面の吸着抑制等に包括的に応用したものが開示されているが、安定度の提示が十分でないので、化粧品等のような外用剤としての直接的な応用には、問題がある。

【発明が解決しようとする課題】
これより、前記問題を解消するため、本発明者らは、安定化した蛋白質を製造して、化粧品等のような外用剤として使用することについて鋭意研究し、その結果、β−1,3-グルカン主糖鎖に規則的なβ−1,6-分枝を有するグルカンを安定化剤として使用して、安定化した蛋白質を製造する場合、安定度及び作用性に優れ、皮膚刺激のような不作用が少なく、製造方法が容易であることを知見し、本発明を完成することに至った。

従って、本発明の目的は、安定化した変性蛋白質を提供することにある。
また、本発明の他の目的は、安定化した変性蛋白質を製造する方法を提供することにある。
本発明のさらに他の目的は、前記方法により製造された安定化蛋白質を含有する外用剤組成物を提供することにある。

【課題を解決するための手段】
前記目的を達成するため、本発明による変性蛋白質は、その安定度を向上させるために変性された蛋白質であって、前記変性蛋白質は、β−1,3-グルカン主糖鎖に規則的なβ−1,6-残基を有するグルカンと結合することを特徴とする。

また、本発明による変性蛋白質の製造方法は、(1)β−1,3-グルカン主糖鎖に規則的なβ−1,6-残基を有するグルカンを、過ヨウ素酸塩類と反応させることにより、β−1,6-残基をアルデヒドに酸化させる段階と、(2)前記段階(1)で得られた反応液から未反応の過ヨウ素酸塩類を除去する段階と、(3)前記段階(2)で得られた溶液に、蛋白質を0.00001 〜10.0重量%の量で添加して、前記蛋白質を酸化したグルカンと接触させる段階と、(4)前記段階(3)で得られた溶液に、0.0001〜1.0 重量%の還元剤を添加する段階とを含むことを特徴とする。

そして、前記製造方法は、前記段階(4)の生成物を洗浄する段階(5)をさらに含むことができる。
さらに、本発明による外用剤組成物は、活性成分として前記変性蛋白質を含むことを特徴とする。

【発明の実施の形態】
以下、本発明をより詳しく説明する。
本発明において、安定化蛋白質の製造に使用されるグルカンは、β−1,3-グルカン主糖鎖に規則的なβ−1,6-残基を有するグルカン(以下、グルカンという)であって、このグルカンは、水相で中性であり、三重らせん構造を有する安定的な形態を有し、化学反応方法によりβ−1,6-残基のみに反応基を作ることができる。即ち、デキストランのような一般的なポリサカライドが1つの線形主糖鎖に残基毎に又は不規則的に反応基を作ることとは別に、β−1,6-残基のみに反応基を作ることができるので、一般的なポリサカライドを用いて蛋白質を安定化させる方法とは、結合度、安定度及び作用性等において異なる特性を示すことができる。

本発明に使用されるグルカンは、その種類が特に限定されないが、例えば、スエヒロタケ(Schizophyllum commune) により生産されたスキゾフィラン(schizophyllan) 、スクレロチニア属(Sclerotinia sp.) 菌株により生産されたスクレログルカン(scleroglucan)、椎茸(Lentinus edodes) により生産されたレンチナン(lentinan)等がある。

前記グルカンは、数十万から数百万の分子量を有するものを加工することなく、そのまま使用するか、マイクロフルイダイザ(microfluidizer)、超音波粉砕器(sonicator) 又はβ−グルカナーゼ (β-glucanase) 等を用いて50、000乃至数十万に分子量を調節したものを使用することができ、0.001 〜20.0重量%、好ましくは 0.1〜5.0 重量%水溶液の形態で使用する。

本発明においてグルカンにより安定化されることができる蛋白質は、特に限定されないが、例えば、パパイン(papain)、ブロメライン(bromelein) 、フィシン(ficin) 、トリプシン(trypsin) 、キモトリプシン(chymotrypsin)、コラゲナーゼ(collagenase) 、エラスターゼ(elastase)、プラスミン(plasmin) 又は細菌性プロテアーゼ(bacterial protease)等の蛋白分解酵素類;α−アミラーゼ(amylase) 、グルコアミラーゼ(glucoamylase)、セルラーゼ(cellulase) 、ペクチナーゼ(pectinase) 、キシラナーゼ(xylanase)、α−グルコシダーゼ(glucosidase) 、β−グルコシダーゼ、α−ガラクトシダーゼ(galactosidase) 、β−ガラクトシダー、β−グルカナーゼ(glucanase) 、キチナーゼ(chitinase) 又はマンナン分解酵素(mannannase)等の炭水化物分解酵素類;ホスホリパーゼ(phospholipase) 又はトリアシルグリセロール分解酵素(triacylglycerol hydrolase) 等の脂肪分解酵素類;デオキシリボ核酸分解酵素(DNase) 又はリボ核酸分解酵素(RNase) 等の核酸分解酵素類;リン酸分解酵素(phosphatase) ;リゾチーム(lysozyme);カタラーゼ(catalase);スーパーオキシドジスムターゼ(superoxide dismutase);トランスグルタミナーゼ(transglutaminase)、ペルオキシダーゼ(peroxidase);ホトリアーゼ(photolyase);微生物由来複合酵素類;サイトカイン類(cytokines) ;細胞成長因子類(growth factors);ホルモン類(hormones);抗原(antigen) ;抗体(antibody);免疫グロブリン(immunoglobulins) ;ラクトフェリン(lactoferrin) ;メタロチオネイン(metallothionein) ;チオレドキシン(thioredoxin) ;抗菌性蛋白質;抗酸化蛋白質;これらの活性ペプチド又は糖及び脂肪結合体よりなる群から選ばれ、単独又は併用して使用することができる。

すなわち、本発明の製造方法は、(1)β−1,3-グルカン主糖鎖に規則的なβ−1,6-残基を有するグルカンを、過ヨウ素酸塩類と反応させることにより、β−1,6-残基をアルデヒドに酸化させる段階と、(2)前記段階(1)で得られた反応液から未反応の過ヨウ素酸塩類を除去する段階と、(3)前記段階(2)で得られた溶液に、蛋白質を0.00001 〜10.0重量%の量で添加して、前記蛋白質を酸化したグルカンと接触させる段階と、(4)前記段階(3)で得られた溶液に、0.0001〜1.0 重量%の還元剤を添加する段階とを含む。また、本発明の製造方法は、前記段階(4)の生成物を洗浄する段階(5)をさらに含むことができる。

上記の方法において、前記段階(1)は、グルカン中のβ−1,6-残基をアルデヒドに酸化して、蛋白質と結合させるための段階であって、グルカンに対して過ヨウ素酸塩類を0.1 〜50.0モル当量、好ましくは2 〜5 モル当量で添加した後、暗所で攪拌しつつ反応させる。この際、過ヨウ素酸塩類としては、特に限定されないが、例えば、過ヨウ素酸(HIO4 、H5IO6)、過ヨウ素酸ナトリウム(NaIO4、Na3H2IO4) 、過ヨウ素酸カリウム(KIO4)等を使用することができる。一方、上記の方法において、グルカンを過ヨウ素酸塩類と反応させた後、酸化反応を停止させるため、エチレングリコールを0.01〜10.0重量%の量でさらに添加することができる。

前記段階(2)は、前記段階(1)の反応液中に反応せず残存している過ヨウ素酸塩類等を除去するための段階であって、除去方法は、分子量限界(molecular cut off) が1000〜500000、好ましくは10000 〜100000の膜を用いて透析(dialysis)又は限外濾過(ultrafiltration) して除去する。

前記段階(3)は、グルカンのβ−1,6-残基のアルデヒドと蛋白質アミノ基とが結合してシッフ塩基(schiff's base) が形成されるようにする段階であって、前記段階(2)で洗浄されたグルカン溶液に蛋白質を0.00001 〜10.0重量%、好ましくは0.001 〜5.0 重量%の量で添加する。

前記段階(4)は、シッフ塩基を還元させる段階であって、前記グルカン−酵素(蛋白質)溶液(シッフ塩基溶液)に還元剤を0.001 〜1.0 重量%、好ましくは0.01〜0.1 重量%の量で添加する。この際、還元剤としては、特に限定されないが、例えば、ほう水素化ナトリウム(NaBH4) 、ソジウムシアノボロハイドライド(NaBH3CN) 、アミンボラン(amine borane)等を使用することができる。一方、反応しないアルデヒド反応基をなくすため、還元されたグルカン−酵素(蛋白質)溶液に0.01〜10重量%、好ましくは0.1 〜1.0 重量%のリシン(lysine)をさらに添加することができる。

前記段階(5)は、最終的に安定化した酵素(蛋白質)溶液を洗浄する段階であって、前記段階(2)のように、分子量限界が1000〜500000、好ましくは10000 〜100000の膜を用いて透析又は限外濾過して洗浄する。

以上の各段階は、蛋白質の変性や活性の損失が起こらない条件、例えば、0℃〜室温の温度で穏やかな攪拌下に実施することができるが、必ずこれに限定されるものではない。

本発明のグルカンを用いて安定化蛋白質を製造するにあたり、製造しようとする蛋白質が2つ以上である場合、蛋白質をグルカンと同時に反応させることができるが、まず一つをグルカンと反応させ、次いでもう一つを反応させて安定化させることもできる。

本発明の方法により製造された安定化蛋白質は、使用される蛋白質の特性によって角質除去、皮脂調節、抗炎症、刺激緩和、ニキビ(痕)緩和、抗酸化、毒性物質の除去、金属イオン伝達、弾力増進、しわ除去、老化防止、美白、タンニング(tanning) 、脱毛防止、除毛、有害細菌及び真菌抑制、臭い除去、日光損傷治癒及び傷つけ治癒等の効果を有する外用剤組成物に使用される。その使用量は、その処方によって適当量で選定されることができ、好ましくは外用剤組成物の総重量に対して0.00001 〜100 重量%の量で含有される。

本発明の外用剤組成物は、その処方において特に限定されるものではなく、例えば、クレンジングローション、クレンジングクリーム、柔軟化粧水、栄養化粧水、マッサージクリーム、栄養クリーム、メークアップベース、口紅、フェイシャルパック(facial pack) 、フェイシャルゲル(facial gel)、シャンプー、リンス、ヘアトニック又は石鹸等の処方を有する化粧料組成物であることができ、ローション、軟膏、ゲル、クリーム、パッチ又はスプレーの処方を有する皮膚組成物であることができる。

また、各処方の外用剤組成物において、前記安定化した蛋白質以外の他の成分は、外用剤の処方又は使用目的等によって当業者が適宜選定して配合することができる。

以下、製造例及び試験例を例にして本発明をより詳しく説明する。しかしながら、本発明がこれらの例のみに限定されるものではない。

〈製造例1〉
(1)分子量が200 万である0.5 重量%のスキゾフィラン水溶液50mlに過ヨウ素酸ナトリウム(NaIO4) 2 gを添加し、4 ℃の暗所で1時間攪拌した。
(2)反応されたスキゾフィラン溶液を分子量限界10000 の透析膜に入れ、4 ℃の暗所で2500mlの水中に12時間毎に水を交換しつつ48時間透析した。

(3)100ml 程度に量が増加したグルカン溶液に0.1 重量%のパパインを入れ、2 時間4℃の暗所で攪拌した。
(4)スキゾフィラン−パパイン溶液にほう水素化ナトリウム(NaBH4) 0.05gを入れ、4 時間攪拌した後、リシン0.1gを入れ、4 ℃の暗所で2 時間攪拌した。

(5)スキゾフィラン−パパイン溶液を、前記(2)と同様の方法で透析して、スキゾフィラン−パパイン結合体溶液であるGE−1を得た。
前記方法により製造されたGE−1の収率は、酵素活性度基準で95%である。

〈製造例2〉
(1)マイクロフルイダイザにより分子量を30万に調整した1 重量%のスクレログルカン水溶液50mlに過ヨウ素酸カリウム2 gを添加し、4 ℃の暗所で1時間攪拌した。
(2)反応されたスクレログルカン溶液を分子量限界30000 の限外濾過膜を用いて常温、暗所で10000ml の水を徐々に加えつつ、限外濾過した。

(3)50ml程度の量で洗浄されたスクレログルカン溶液に0.5重量%のリゾチームを入れ、1時間4℃の暗所で攪拌した。
(4)スクレログルカン−リゾチーム溶液にほう水素化ナトリウム0.05gを入れ、4℃の暗所で2時間攪拌した。

(5)スクレログルカン−リゾチーム溶液を、前記(2)と同様の方法で限外濾過して、スクレログルカン−リゾチーム結合体溶液であるGE−2を得た。
前記方法により製造されたGE−2の収率は、酵素活性度基準で93%である。

〈製造例3〉
(1)分子量が100 万である0.5 重量%のスキゾフィラン水溶液50mlに過ヨウ素酸ナトリウム2 gを添加し、常温で1時間攪拌した。
(2)反応されたスキゾフィラン溶液を分子量限界10000 の透析膜に入れ、4 ℃の暗所で2500mlの水中に12時間毎に水を交換しつつ48時間透析した。

(3)100ml 程度に量が増加したスキゾフィラン溶液に0.1 重量%のパパインを入れ、2 時間常温で攪拌した。
(4)スキゾフィラン−パパイン溶液にほう水素化ナトリウム0.01gを入れ、常温で10分間攪拌した後、0.1 重量%のリゾチーム(0.1g)を入れ、さらに2 時間反応した後、ほう水素化ナトリウム0.04gとリシン0.1 gを入れ、24時間攪拌した。

(5)スキゾフィラン−パパイン及びスキゾフィラン−リゾチームの混合溶液を、前記(2)と同様の方法で透析して、グルカン−酵素結合体溶液であるGE−3を得た。
前記方法により製造されたGE−3の収率は、酵素活性度基準でパパインは94%であり、リゾチームは90%である。

〈製造例4〉
(1)分子量が200 万である0.1 重量%のスキゾフィラン水溶液5ml に過ヨウ素酸ナトリウム0.05gを添加し、4 ℃の暗所で1時間攪拌した。
(2)反応されたスキゾフィラン溶液を分子量限界1000の透析膜に入れ、4 ℃の暗所で2500mlの水中に12時間毎に水を交換しつつ48時間透析した。

(3)透析されたグルカン溶液に0.01重量%の人体再組合上皮細胞成長因子(human recombinant epidermal growth factor)(Sigma Aldrich Co. 製) を入れ、2 時間4 ℃の暗所で攪拌した。
(4)スキゾフィラン−上皮細胞成長因子溶液にほう水素化ナトリウム0.002 gを入れ、4 時間攪拌した。

〈比較製造例1〉
(1)分子量が50万である1 重量%のデキストラン水溶液50mlに過ヨウ素酸ナトリウム2 gを添加し、4 ℃の暗所で1 時間攪拌した。

すなわち、D−板状細胞(squame)を用いて正常人の下膊で角質を採取した各ディスクを、予め同一の活性度に調整した各酵素試料に浸漬し、37℃で反応させた。次いで、時間によって回収したディスクを0.2 %ナイル青A染色色素(Nile blue A) 溶液に10分間浸漬した後、蒸留水で洗浄し、クロマメーター(chromameter) を用いて角質の分解度を測定して百分率で換算し、試験結果は表3に示した。

前記表3から明らかなように、本発明により製造された安定化酵素は、角質に対する作用性が自然状態の酵素と類似するが、安定度面において自然状態の酵素より優れるので、最終的な角質に対する作用性は、本発明の酵素が優れることがわかる。さらに、本発明の方法により製造された安定化酵素が、公知の方法により製造された安定化酵素より角質に対する作用性面において非常に優れることがわかる。

[試験例4]
前記製造例1と3で製造した安定化酵素の皮膚刺激度を、同一の活性度の自然状態の酵素と比較した。

すなわち、各酵素試料による皮膚刺激度を紅斑形成度により測定した。測定方法は、皮膚色素測定器(Chromameter CM2002)を用いて試料塗布部位の皮膚色初期値を測定し、各酵素試料を各々20μl ずつ1 日2 回、7 日間塗布しつつ、皮膚色素測定器を用いて前記試料による紅斑を測定し、対照区(control) と比較して皮膚刺激度を評価した。この際、紅斑指数は、最初皮膚色素測定値を100 に換算した値であって、紅斑指数はアノバテスト(Anova test)により統計的有意性(significance)を検証したところ、p<0.05範囲で統計的に有意なものと判定された。その結果は、下記表4に示した。

前記表4から明らかなように、対照区と比べて、自然状態の酵素は、1/100 に希釈した状態でも刺激を示す反面、本発明の方法により安定化した製造例1と3の酵素は、希釈しない状態でも全然皮膚刺激を示さないことがわかる。

[試験例5]
前記製造例4で製造したグルカン−上皮細胞成長因子結合体と自然状態の上皮細胞成長因子の安定度を繊維芽細胞増殖能により比較した。

すなわち2.5 %の牛胎児血清が含有されたDMEM(Dulbecco's Modified Eagle's Media) 培地で培養したヒトの繊維芽細胞を、96孔平板培養器(96-well microtiter plate)で5000細胞/wellとなるように分注した。試料として、製造例4で得られたグルカン−上皮細胞成長因子結合体と、同一濃度に蒸留水に溶解した自然状態の上皮細胞成長因子を用いて、前処理として各々37℃の温度で7 日間保存した。前処理したグルカン−上皮細胞成長因子結合体と、前処理した自然状態の上皮細胞成長因子及び新たに溶解した自然状態の上皮細胞成長因子を繊維芽細胞に処理し、さらに4 日間培養した。試料の培養を完了した後、0.2 %MTT(3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide)溶液を、各ウエル(well)当たり50μl ずつ添加し、さらに37℃で4 時間培養した後、生成されたフォルマザン(formazan)をDMSO(dimethyl sulfoxide)を用いて溶解した。溶解されたフォルマザンの吸光度を平板培養測定器(microplate reader) を用いて570nm で測定した。これを無処理群と比較して繊維芽細胞の増殖如何を判定した。その結果を下記表5に示した。

前記表5から明らかなように、自然状態の上皮細胞成長因子と比べて、前処理として蛋白質活性損失を誘導した処理群の場合、製造例4の試料は、細胞増殖機能をそのまま維持する反面、自然状態の上皮細胞成長因子は、蛋白質の機能をほとんど消失した。従って、本発明の方法により製造された蛋白質の場合、安定的に機能を維持することができることがわかる。

【発明の効果】
以上説明したように、β−1,3-グルカン主糖鎖に規則的なβ−1,6-分枝を有するグルカンを用いて、安定化した蛋白質を製造することにより、蛋白質の安定度を著しく向上させることができ、従って、このような安定化蛋白質を含有する外用剤組成物は、各々の蛋白質の有する角質除去、皮脂調節、抗炎症、刺激緩和、ニキビ(痕)緩和、抗酸化、毒性物質の除去、金属イオン伝達、弾力増進、しわ除去、老化防止、美白、タンニング(tanning) 、脱毛防止、除毛、有害細菌及び真菌抑制、臭い除去、日光損傷治癒及び傷つけ治癒等の効果を長時間安定に 提供することができる。
Claims (13)
- その安定度を向上させるために変性された蛋白質であって、前記変性蛋白質は、β−1,3-グルカン主糖鎖に規則的なβ−1,6-残基を有するグルカンと結合することを特徴とする変性蛋白質。
- 前記β−1,3-グルカン主糖鎖に規則的なβ−1,6-残基を有するグルカンは、スキゾフィラン、スクレログルカン又はレンチナンであることを特徴とする請求項1に記載の変性蛋白質。
- 前記蛋白質は、パパイン、ブロメライン、フィシン、トリプシン、キモトリプシン、コラゲナーゼ、エラスターゼ、プラスミン又は細菌性プロテアーゼ;α−アミラーゼ、グルコアミラーゼ、セルラーゼ、ペクチナーゼ、キシラナーゼ、α−グルコシダーゼ、β−グルコシダーゼ、α−ガラクトシダーゼ、β−ガラクトシダーゼ、β−グルカナーゼ、キチナーゼ又はマンナン分解酵素;ホスホリパーゼ又はトリアシルグリセロール分解酵素;デオキシリボ核酸分解酵素又はリボ核酸分解酵素;リン酸分解酵素;リゾチーム;カタラーゼ;スーパーオキシドジスムターゼ;トランスグルタミナーゼ、ペルオキシダーゼ;ホトリアーゼ;微生物由来複合酵素類;サイトカイン類;細胞成長因子類;ホルモン類;抗原;抗体;免疫グロブリン;ラクトフェリン;メタロチオネイン;チオレドキシン;抗菌性蛋白質;抗酸化蛋白質;これらの活性ペプチド又は糖及び脂肪結合体よりなる群から選ばれ、単独又は併用して使用することを特徴とする請求項1に記載の変性蛋白質。
- 安定度を向上させるために蛋白質を変性する方法であって、前記蛋白質とβ−1,3-グルカン主糖鎖に規則的なβ−1,6-残基を有するグルカンとを結合させる段階を含むことを特徴とする変性蛋白質の製造方法。
- (1)β−1,3-グルカン主糖鎖に規則的なβ−1,6-残基を有するグルカンを、過ヨウ素酸塩類と反応させることにより、β−1,6-残基をアルデヒドに酸化させる段階と、
(2)前記段階(1)で得られた反応液から未反応の過ヨウ素酸塩類を除去する段階と、
(3)前記段階(2)で得られた溶液に、蛋白質を0.00001 〜10.0重量%の量で添加して、前記蛋白質を酸化したグルカンと接触させる段階と、
(4)前記段階(3)で得られた溶液に、0.0001〜1.0 重量%の還元剤を添加する段階とを含むことを特徴とする請求項4に記載の製造方法。 - 前記段階(4)の生成物を洗浄する段階(5)をさらに含むことを特徴とする請求項5に記載の製造方法。

- 前記β−1,3-グルカン主糖鎖に規則的なβ−1,6-残基を有するグルカンは、スキゾフィラン、スクレログルカン又はレンチナンであることを特徴とする請求項4に記載の製造方法。
- 前記β−1,3-グルカン主糖鎖に規則的なβ−1,6-残基を有するグルカンは、マイクロフルイダイザ、超音波粉砕器又はβ−グルカナーゼを用いて分子量を調整したものであることを特徴とする請求項8に記載の製造方法。
- 2種以上の蛋白質を変性する場合、まず一方の蛋白質を前記段階(3)で添加した後、他方の蛋白質を前記段階(4)で添加することを特徴とする請求項5に記載の製造方法。
- 請求項1の変性蛋白質を、組成物の総重量に対して0.00001 〜100 重量%の量で含有することを特徴とする外用剤組成物。
- 前記外用剤組成物は、クレンジングローション、クレンジングクリーム、柔軟化粧水、栄養化粧水、マッサージクリーム、栄養クリーム、メークアップベース、口紅、フェシャルパック、フェシャルゲル、シャンプー、リンス、ヘアトニック又は石鹸よりなる群から選ばれる化粧料組成物であることを特徴とする請求項11に記載の外用剤組成物。
- 前記外用剤組成物は、ローション、軟膏、ゲル、クリーム、パッチ又はスプレーよりなる群から選ばれる皮膚組成物であることを特徴とする請求項11に記載の外用剤組成物。
