脂質プロファイリングの結果を図11及び図12に示す。図11は、正常肝臓とNASH(非アルコール性脂肪肝炎:nonalcoholic steato−hepatitis)マウスの肝臓で脂質プロファイリングを行った結果を示す。NASHでは、ほとんどの脂質クラスにおいて脂質量が上昇している。また、脂肪酸側鎖が1つしか結合していないリゾフォスファチジルコリン(LPC)とリゾフォスファチジルエタノールアミン(LPE)の定量値も参考として掲載した。LPCとLPEでは、脂肪酸側鎖はポジティブイオンモードでMRM測定をしている。注目すべきはPGであり、PG全体として増加しているが、パルミチン酸(16:0)はまったく増加していない(ゆえに構成比率としては減少している)。NASHが肝臓のミトコンドリア不全である事を考えると、ミトコンドリア内にのみ存在するPGの変化はNASHを研究する上では重要かもしれない。またLPCとLPEは6種類の脂肪酸側鎖のみの合計値であるが,他のリン脂質は増加しているにもかかわらず減少している。

<脂質プロファイリング結果の考察>
各脂質の脂肪酸側鎖のイオン強度比の補正係数の結果(図10)から、DGの定量用トランジションにおける飽和脂肪酸((16:0),(18:0))のニュートラルロスと、高度不飽和脂肪酸((20:4),(22:6))のニュートラルロスを比較すると、5〜10倍以上のイオン強度差となっている。この結果からアラキドン酸、DHAともに他の脂肪酸側鎖と比較すると非常に脱離しやすい化合物と言える。PSの定量用トランジションにおける飽和脂肪酸のイオン強度と高度不飽和脂肪酸のイオン強度とでは、6〜20倍以上のイオン強度差が存在する。また、他のグリセロリン脂質においても、PSと同様に脂肪酸側鎖のイオン強度を測定しているが、PCとPEは比較的類似の傾向を示しており、ともに(18:2)が脱離しやすい。グリセロリン脂質の種類によって同じ脂肪酸側鎖であっても、そのイオン強度比がかなり異なる事が分かった。全体的にはグリセロリン脂質のトランジションは、DGのニュートラルロスと逆の傾向となり、高度不飽和脂肪酸のイオン強度は他の脂肪酸と比較すると特にDHAは低くなっている。また、あらかじめsn−1,sn−2の分かっている標品を測定した結果(表1)から、同じ脂肪酸でもsn−1,sn−2と位置異性体でイオン強度が逆転していることが伺える。この事実から、脂肪酸側鎖を2本有する脂質に関して、質量分析を用いる場合はsn−2に結合している脂肪酸側鎖は非常に脱離しやすい傾向がある。すなわち、高度不飽和脂肪酸は通常の生体内ではsn−2に結合しているため、脂肪酸側鎖の種類以外の部分でもプロダクトイオン強度が高くなったと考えられる。この事実は、通常、生体内に存在している脂質は飽和脂肪酸がsn−1に、不飽和脂肪酸はsn−2に結合している事実と合致する。実際に(18:0)/(18:1)と(16:0)/(18:1)を脂肪酸側鎖として結合しているPCの標準品のイオン強度比と、生体サンプルのイオン強度比とを比較すると、sn−1,sn−2の結合様式が明確に分かる。そのイオン強度比から飽和脂肪酸がsn−1に、不飽和脂肪酸がsn−2に結合していることが証明された。PC(16:0/18:0)に関しては、標準品と生体サンプルのイオン強度比に開きがあることから、生体サンプルのPC(16:0/18:0)は必ずしもsn−1が16:0とはなっておらず、半数程度がsn−1に18:0が結合しているものと思われる。このように、生体サンプルのみでもsn−1,sn−2のどちらに脂肪酸側鎖が結合しているかは不明のまま定量を行うことになるが、位置異性体の標準品が存在する場合はそのイオン強度比をつかってsn−1,sn−2のどちらにどの程度の脂肪酸が結合しているかを求めることも可能である。
