Description
本発明はスキンケアの分野であり、具体的には、ブドウ種子、並びに葉酸、ビオチン、ビフィドバクテリウムロンガム(Bifidobacterium longum)抽出物及びエキナセアプルプレア(Echinacea purpurea)のうちの少なくとも数種類を含む局所用製品に関する。
ヒトに対する環境的損傷の効果は十分に文書化されている。環境における日光や様々な汚染因子への暴露は、皮膚の急性及び慢性老化に寄与する反応性酸素中間体の発生の原因となる。結果として、皮膚における反応性酸素種(ROS)を捕捉する抗酸化剤を含有する局所用製剤が一般的になっている。一般に、抗酸化剤を含有する製品の局所適用は、依然としてスキンケア分野における重要な研究領域である。
もちろん、種々の活性及び効果をスキンケア製品に与える非常に多くの物質が存在する。これらの物質は、単一の製品中に複数の利益を与えるために組み合わされることが多い。しかしながら、活性のある化学薬剤であると、これらの物質が互いに干渉し合い、意図される効果の一部又は全部を打ち消すことも珍しくない。したがって、商業的に実行可能な費用で、安全で、安定な、効果のある製品を処方することは大きな挑戦である。
局所用製剤におけるブドウ種子(ヴィティスヴィニフェラ(Vitis vinifera))抽出物の抗酸化作用が知られている。ブドウ種子抽出物はまた、軟化薬、乳化剤及び皮膚調整剤として使用される。
葉酸(CAS 59-30-3)は、式(C19H19N7O6)：
を有する有機化合物である。
葉酸は、局所用製剤において、皮膚調整剤として使用される。
ビオチン(CAS 58-85-5)は、局所用製剤、例えばシャンプー、コンディショナー、石鹸、バスオイル及びソルト、並びに化粧品において、髪及び皮膚の調整剤として使用されることが知られている。ビオチンは、式(C10H16N2O3S)：
で表されるカルボン酸である。
ビフィズス(Bifidus)又はビフィダ(Bifida)発酵抽出物は、ビフィドバクテリウムロンガムの発酵から得られる抽出物である。これは、皮膚調整剤及び保湿剤として使用されている。これはまた、皮膚細胞のDNA修復過程を促進することが報告されている。例えば米国特許第4,464,362号明細書は、光損傷を治療するための、ビフィドバクテリウムロンガム(ロイター(Reuter))種の局所的使用について開示する。
エキナセアプルプレア(コーンフラワー(Coneflower))抽出物は、髪用製品を含む局所用組成物において使用されることが知られており、これは、抗細菌性及び抗炎症性の作用を有することが報告されている。エキナセアプルプレアは、エキナセアアングスティフォリア(Echinacea angustifolia)、エキナセアパリダ(Echinacea pallid)及び他の種類のエキナセア(Echinacea)と混同するべきではない。
我々は、ブドウ種子抽出物、並びに葉酸、ビオチン、ビフィズス、及びエキナセアプルプレアのうちの1種類又は複数種類を含む商業的に実行可能な組成物が非常に様々な抗酸化作用を有することを発見した。これらの成分の各々は、環境的に誘発された皮膚損傷に対する対策において有用な利益を提供する。我々は、ベース製剤において、これらの5種類の成分が複雑に干渉し合い、時には有用な抗酸化作用を有する局所用組成物を与え、時には与えないのではないかと考えた。したがって、課題は、安全で、安定な局所用スキンケア製剤において、これら5種類の成分の1つ又は複数の最適な混合物を発見することであった。
様々な局所用製剤の抗酸化作用は、以下のアッセイにより測定された。

「対照」と表示されたベース製剤は、0％の抗酸化作用を示す。ベース製剤に0.05％のブドウ種子抽出物(公知の抗酸化剤)を加えた製剤8は、約17.5％の酸化抑制を示す。
この時点では、我々は、ビフィズス抽出物、葉酸、エキナセアプルプレア及びビオチンを、それらの公知の利益のために、製剤中に含ませることを望んでいた。我々がこれ（製剤1）を実施したところ、予期しないことが起こった。意外なことに、抗酸化作用は、(約5％の酸化抑制まで)著しく減少した。活性成分の様々な組み合わせを試験して、どの製剤が著しい抗酸化作用を与えるかについて測定した。
ブドウ種子抽出物＋3種類の組み合わせ
製剤3を製剤1と比較すると、葉酸はブドウ種子抽出物の抗酸化作用を減少させるようである。

ビフィズス抽出物、葉酸、エキナセアプルプレア及びビオチンの1つ1つは、他の3種類も存在する場合、ブドウ種子抽出物の抗酸化作用を減少させるようである。言い換えれば、ブドウ種子抽出物の抗酸化作用に関して、4種類全てよりも、ビフィズス抽出物、葉酸、エキナセアプルプレア及びビオチンのうちの任意の3種類を有する製剤の方が良好である。これは、全く予想外の結果であり、「ブドウ種子抽出物、並びにビフィズス抽出物、葉酸、エキナセアプルプレア及びビオチンのうちのちょうど3種類を含む局所用組成物」等の請求項をサポートする。

製剤7を製剤8と比較すると、葉酸及びエキナセアプルプレアの組み合わせは、ブドウ種子抽出物の抗酸化作用を著しく減少(この場合、消滅)させる。しかしながら、製剤10を製剤1と比較すると、葉酸及びエキナセアプルプレアの組み合わせは、ビオチン及びビフィズスも存在する場合、ブドウ種子抽出物の抗酸化作用にほとんど効果を有しない(わずかに高める可能性がある)ようである。

製剤11を製剤8と比較すると、ビオチン及びエキナセアプルプレアの組み合わせは、ブドウ種子抽出物の抗酸化作用を著しく減少させる。しかしながら、製剤6を製剤1と比較すると、ビオチン及びエキナセアプルプレアの組み合わせは、葉酸及びビフィズスも存在する場合、ブドウ種子抽出物の抗酸化作用を高めるようである。

製剤10を製剤8と比較すると、ビオチン及びビフィズスの組み合わせは、ブドウ種子抽出物の抗酸化作用を著しく減少させる。しかしながら、製剤7を製剤1と比較すると、ビオチン及びビフィズスの組み合わせは、葉酸及びエキナセアプルプレアも存在する場合、ブドウ種子抽出物の抗酸化作用を高めるようである。
したがって、以下の組み合わせ:葉酸/ビフィズス、ビオチン/エキナセアプルプレア、ビオチン/ビフィズス、及び葉酸/エキナセアプルプレアは、他の2種類が存在しない場合、ブドウ種子抽出物の抗酸化作用を著しく減少させ、他の2種類が存在する場合、ブドウ種子抽出物の抗酸化作用を高めるが、活性剤がちょうど3種類存在する場合ほどではない。したがって、データは以下の請求項をサポートする。
ブドウ種子抽出物、並びに葉酸及びビフィズスの組み合わせを含むが、ビオチン及びエキナセアプルプレアの組み合わせを含まない局所用組成物。

考察
これらの結果について興味深いことは、異なるブドウ種子製剤の抗酸化作用が同じ4種類の活性剤の組み合わせの間で異なる相乗的挙動を示すことである。したがって、ブドウ種子抽出物、ビフィズス抽出物、葉酸、エキナセアプルプレア及びビオチンの公知の利益を組み入れる抗酸化性スキンケア組成物を製剤化する際は、抗酸化作用の全てを破壊しない組み合わせで活性剤を混合するように注意しなければならない。
これらの結果について興味深いことは、異なるブドウ種子製剤の抗酸化作用が同じ4種類の活性剤の組み合わせの間で異なる相乗的挙動を示すことである。したがって、ブドウ種子抽出物、ビフィズス抽出物、葉酸、エキナセアプルプレア及びビオチンの公知の利益を組み入れる抗酸化性スキンケア組成物を製剤化する際は、抗酸化作用の全てを破壊しない組み合わせで活性剤を混合するように注意しなければならない。
我々は、ブドウ種子抽出物を含む組成物に関して、葉酸、ビオチン、ビフィズス及びエキナセアプルプレアの組み合わせが有用であり、葉酸、ビフィズス及びエキナセアプルプレアの組み合わせ(ビオチンなし)と同じように、ビオチン、ビフィズス及びエキナセアプルプレアの組み合わせ(葉酸なし)が好ましく、葉酸、ビオチン及びビフィズスの組み合わせ(エキナセアプルプレアなし)がより好ましく、葉酸、ビオチン及びエキナセアプルプレアの組み合わせ(ビフィズスなし、例えば、製剤5)が最も好ましいことを決定した。
相対濃度
本発明の組成物において、特定の成分の相対濃度は以下に従い選択される:
ブドウ種子と葉酸の比は、50:0〜50:1で変わり得る。
ブドウ種子とビオチンの比は、50:0〜50:1で変わり得る。
ブドウ種子とエキナセアプルプレアの比は、50:0〜50:100で変わり得る。
ブドウ種子とビフィズスの比は、50:0〜50:10,000で変わり得る。
本発明の組成物において、特定の成分の相対濃度は以下に従い選択される:
ブドウ種子と葉酸の比は、50:0〜50:1で変わり得る。
ブドウ種子とビオチンの比は、50:0〜50:1で変わり得る。
ブドウ種子とエキナセアプルプレアの比は、50:0〜50:100で変わり得る。
ブドウ種子とビフィズスの比は、50:0〜50:10,000で変わり得る。
本発明による組成物中に、5種類の成分全てが存在する場合、葉酸と、ビオチンと、ブドウ種子と、エキナセアプルプレアと、ビフィズス抽出物の最も好ましい濃度比は、1 : 1 : 50 : 100 : 10,000であり、成分のいずれもが、10％以下の変動である。これは、0.9〜1.1 : 0.9〜1.1 : 45〜55 : 90〜110 : 9,000〜11,000 (葉酸:ビオチン:ブドウ種子:エキナセアプルプレア:ビフィズス抽出物)と記載することができる。したがって、1つの例示的な比は、0.001 gの葉酸と、0.001 gのビオチンと、0.05 gのブドウ種子抽出物と、0.10 gのエキナセアプルプレアと、10.0 gのビフィズス抽出物である。
本発明による組成物は、スキンケア製剤、ヘアケア製剤として、又はカラー化粧品として使用され得る。カラー化粧品は、肌の外観を変えるための、ブラッシュ、アイシャドウ、リップスティック、リップグロス、ファンデーション、コンシーラー等を含む。本発明による組成物は、ローション、クリームトナー、ジェル、ソリッド又はセミソリッドの製剤として使用され得る。スキンケア製剤は、顔、手、及び頭皮用の製剤を含む。ヘアケア製剤は、頭髪、又はまつ毛用の製剤を含む。組成物は、エマルション、懸濁液、又は混合物の形態であり得る。
組成物は、実質的に、任意の化粧品に許容される成分を含有し得る。多くの髪及び/又は皮膚の調整剤を挙げることができるが、頭皮製品用の以下のものがとりわけ有用である：アデノシンホスフェート、アロエベラ(aloe barbadensis)、シロイヌナズナ(Arabidopsis thaliana)抽出物、アルギニン、ブチレングリコール、カフェイン、カプリリルグリコール、カラギーナン、エチルヘキシルグリセリン、ダイズ抽出物、加水分解コムギタンパク質、ラクトバチルス発酵物(lactobacillus ferment)、カラフトコンブ(laminaria saccharina)抽出物、パンテチン(panthenine)、プルラン、キノア種子抽出物、ローズマリー葉抽出物、サッカロミセス溶解質抽出物、リボ核酸ナトリウム断片、酵母抽出物。これらは、典型的に、最終組成物の0.0001〜5.0重量％の濃度で使用され得る。効果的な頭皮の処置のためには、これらのうちの少なくとも3種類を使用することが好ましい。頭皮のニーズに対処するためには、これらのうちの少なくとも5種類がより好ましい。
有用な溶剤は、水及び/又はアルコールを含む。典型的には、溶剤全部で、組成物の60重量％〜90重量％になり得る。典型的には、アルコールは、組成物の0重量％〜20重量％の範囲であり得る。有用な防腐剤としては、安息香酸、クロルフェネシン、フェノキシエタノール、ソルビン酸カリウム、安息香酸ナトリウム、デヒドロ酢酸ナトリウム及びソルビン酸が挙げられる。防腐剤の総量は、組成物の0.01重量％〜5.0重量％の範囲であり得る。組成物は、エマルション促進剤、例えばアクリロイルジメチルタウリンアンモニウム/メタクリル酸ベヘネス(Benheneth)-25クロスポリマー及びレシチン;pH調整剤、例えばステアリン酸アンモニウム;保湿剤、例えばパンテノール及び異性化糖;結合剤、例えば水添ヒマシ油;充填剤、例えばシリカ;吸着剤、例えばマルトデキストリン;抗酸化剤(ブドウ種子抽出物以外)、例えばトコフェロールを含み得る。
以下は、本発明による頭皮調整製品の非限定例である。成分は、最終組成物の重量％として与えられる。
実施例1〜4は、ブドウ種子＋4種類の活性剤、葉酸、ビオチン、ビフィズス及びエキナセアプルプレアを有する。追加の実施例は、実施例1〜4から葉酸、ビオチン、ビフィズス又はエキナセアプルプレアのうちのいずれか1種類を、他の3種類を保持したまま、取り除くことにより、調製することができる。追加の実施例はまた、ブドウ種子、葉酸、ビオチン、ビフィズス及びエキナセアプルプレアの相対濃度を上記の相対濃度に関する指標内で変更することにより調製することができる。
Claims (12)
- ブドウ種子抽出物、葉酸、ビオチン、ビフィズス発酵抽出物及びエキナセアプルプレアを含む局所用組成物。
- ブドウ種子抽出物、並びに以下：葉酸、ビオチン、ビフィズス発酵抽出物及びエキナセアプルプレアのうちのちょうど3種類を含む局所用組成物。
- 葉酸、ビオチン及びビフィズス発酵抽出物を含む、請求項２に記載の組成物。
- 以下の相対濃度：
ブドウ種子と葉酸の比が、50:0〜50:1で変わり得る；
ブドウ種子とビオチンの比が、50:0〜50:1で変わり得る；且つ
ブドウ種子とビフィズス抽出物の比が、50:0〜50:10,000で変わり得る
を示す、請求項３に記載の組成物。 - 葉酸、ビオチン及びエキナセアプルプレアを含む、請求項２に記載の組成物。
- 以下の相対濃度：
ブドウ種子と葉酸の比が、50:0〜50:1で変わり得る；
ブドウ種子とビオチンの比が、50:0〜50:1で変わり得る；且つ
ブドウ種子とエキナセアプルプレアの比が、50:0〜50:100で変わり得る
を示す、請求項５に記載の組成物。 - 葉酸、ビフィズス発酵抽出物及びエキナセアプルプレアを含む、請求項２に記載の組成物。
- 以下の相対濃度：
ブドウ種子と葉酸の比が、50:0〜50:1で変わり得る；
ブドウ種子とエキナセアプルプレアの比が、50:0〜50:100で変わり得る；且つ
ブドウ種子とビフィズス抽出物の比が、50:0〜50:10,000で変わり得る
を示す、請求項７に記載の組成物。 - ビオチン、ビフィズス発酵抽出物及びエキナセアプルプレアを含む、請求項２に記載の組成物。
- 以下の相対濃度：
ブドウ種子とビオチンの比が、50:0〜50:1で変わり得る；
ブドウ種子とエキナセアプルプレアの比が、50:0〜50:100で変わり得る；且つ
ブドウ種子とビフィズス抽出物の比が、50:0〜50:10,000で変わり得る
を示す、請求項９に記載の組成物。 - ブドウ種子抽出物；
以下：葉酸、ビオチン、ビフィズス発酵抽出物及びエキナセアプルプレアのうちの少なくとも3種類：並びに
最終組成物の0.001〜5.0重量％の濃度の、以下の調整剤：アデノシンホスフェート、アロエベラ、シロイヌナズナ抽出物、アルギニン、ブチレングリコール、カフェイン、カプリリルグリコール、カラギーナン、エチルヘキシルグリセリン、ダイズ抽出物、加水分解コムギタンパク質、ラクトバチルス発酵物、カラフトコンブ抽出物、パンテチン、プルラン、キノア種子抽出物、ローズマリー葉抽出物、サッカロミセス溶解質抽出物、リボ核酸ナトリウム断片、酵母抽出物、のうちの少なくとも3種類
を含む局所用組成物。 - 前記調整剤のうちの少なくとも5種類を含む、請求項１１に記載の局所用組成物。
