Abstract

本発明は、一般に、特定の抗体及び小分子薬物の組み合わせを使用することによるメラノーマの治療に関する。
メラノーマは、発生率が驚くべき増加をたどっている皮膚癌の高悪性度の形態である(Thompson JF et al., Cutaneous melanoma in the era of molecular profiling. Lancet 2009;374:362−5)。治癒は、局所化した病変の外科的切除によって達成することができるが、メラノーマの進行期では現在承認され治療法にはわずかに応答するだけである。ステージIVの転移性メラノーマの5年生存率は約10%である(Thompson, Lancet 2009)。Bcl2に対するアンチセンス、CTLA4に対する抗体、小分子RAFキナーゼ阻害剤、及び養子免疫療法を含む新しい治療アプローチは、転移性メラノーマの臨床試験に入っている(Ascierto PA et al., Melanoma: a model for testing new agents in combination therapies. J Transl Med 2010;8:38-45)。これらの最近の研究の幾つかからの結果は有望であるように見えるが、全生存に対する永続的効果のためには、追加の新しい薬剤を含む治療の組み合わせを必要とし得る。
本発明は、被験体へ抗エンドセリンB受容体(ETBR)抗体薬剤コンジュゲートの有効量をMAPキナーゼ阻害剤の有効量と組み合わせて投与することを含む、メラノーマに罹患している被験体における腫瘍増殖阻害(TGI)を意図する。
I.定義
本明細書における目的のための「アクセプターヒトフレームワーク」とは、下記に定義されるように、ヒト免疫グロブリンフレームワーク又はヒトコンセンサスフレームワークから得られる軽鎖可変ドメイン(VL)フレームワーク又は重鎖可変ドメイン(VH)フレームワークのアミノ酸配列を含有するフレームワークである。ヒト免疫グロブリンのフレームワーク又はヒトコンセンサスフレームワーク「に由来する」アクセプターヒトフレームワークは、その同一のアミノ酸配列を含んでもよく、又はそれはアミノ酸配列の変化を含み得る。幾つかの実施態様において、アミノ酸変化の数は、10以下、9以下、8以下、7以下、6以下、5以下、4以下、3以下、又は2以下である。幾つかの実施態様において、VLアクセプターヒトフレームワークは、Vはヒト免疫グロブリンのフレームワーク配列又はヒトコンセンサスフレームワーク配列に、配列が同一である。
分率X/Yの100倍
ここで、Xは配列アラインメントプログラムALIGN−2により、AとBのそのプログラムのアラインメントにおいて同一と一致したスコアのアミノ酸残基の数であり、YはBの全アミノ酸残基数である。アミノ酸配列Aの長さがアミノ酸配列Bの長さと異なる場合、AのBに対する%アミノ酸配列同一性は、BのAに対する%アミノ酸配列同一性とは異なることは理解されるであろう。特に断らない限り、本明細書で使用されるすべての%アミノ酸配列同一性値が、ALIGN−2コンピュータプログラムを使用し、直前の段落で説明したように得られる。
一態様において、本発明は、一部が、ETBRに結合する抗体に基づく。本発明の抗体は、例えば、メラノーマの治療のために有用である。
一態様において、本発明は、ETBRに結合する単離された抗体を提供する。ある実施態様において、抗ETBR抗体は、(a)CDR−L1(KSSQSLLDSDGKTYLN,配列番号7)、(b)CDR−L2(LVSKLDS、配列番号8)、(c)CDR−L3(WQGTHFPYT;配列番号9)、(d)CDR−H1(GYTFTSYWMQ;配列番号1)、(e)CDR−H2(TIYPGDGDTSYAQKFKG;配列番号2)、及び(f)CDR−H3(WGYAYDIDN;配列番号3)から選択される少なくとも一、二、三、四、五、又は六のCDRを含む。
所定の実施態様において、本明細書において与えられる抗体は、解離定数(Kd)が、≦1μM、≦100nM、≦10nM、≦1nM、≦0.1nM、≦0.01nM、又は≦0.001nM(例えば、10−8M未満、例えば、10−8Mから10−13M、例えば、10−9Mから10−13M)。
ある実施態様において、本明細書で提供される抗体は、抗体断片である。抗体断片は、限定されないが、Fab、Fab’、Fab’−SH、F(ab’)2、Fv,及びscFv断片、及び下記の他の断片を含む。所定の抗体断片の総説については、Hudson et al. Nat. ある実施態様において、本明細書で提供される抗体は、キメラ抗体である。所定のキメラ抗体は、例えば、米国特許第4,816,567号、及びMorrison et al., Proc. Natl. Acad. Sci. USA, 81:6851-6855 (1984))に記載されている。一例において、キメラ抗体は、非ヒト可変領域(例えば、マウス、ラット、ハムスター、ウサギ、又はサル等の非ヒト霊長類由来の可変領域)及びヒト定常領域を含む。更なる例において、キメラ抗体は、クラスまたはサブクラスが親抗体のものから変更された「クラススイッチ」抗体である。キメラ抗体は、その抗原結合断片を含む。

本発明の抗体は、所望の活性または活性(複数)を有する抗体についてコンビナトリアルライブラリーをスクリーニングすることによって単離することができる。例えば、様々な方法が、ファージディスプレイライブラリーを生成し、所望の結合特性を有する抗体についてのライブラリーをスクリーニングするために、当該技術分野で知られている。そのような方法は、例えば、Hoogenboom et al. in Methods in Molecular Biology 178:1-37 (O'Brien et al., ed., Human Press, Totowa, NJ, 2001)に総説され、更に、例えば、McCafferty et al., Nature 348:552-554; Clackson et al., Nature 352: 624-628 (1991); Marks et al., J. ある実施態様において、本明細書で提供される抗体は、多重特異性抗体、例えば二重特異性抗体である。多重特異性抗体は、少なくとも二つの異なる部位に対して結合特異性を有するモノクローナル抗体である。ある実施態様において、結合特異性の一つはETBRに対してであり、他は、任意の他の抗原に対してである。ある実施態様において、二重特異性抗体は、ETBRの2つの異なるエピトープに結合することができる。二重特異性抗体はまたETBRを発現する細胞に対して細胞傷害性薬物を局在化させるために用いることができる。二重特異性抗体は、全長抗体又は抗体断片として調製することができる。
ある実施態様において、本明細書で提供される抗体のアミノ酸配列変異体が企図される。例えば、抗体の結合親和性及び/又は他の生物学的特性を改善することが望まれ得る。抗体のアミノ酸配列変異体は、抗体をコードするヌクレオチド配列に適切な改変を導入することにより、またはペプチド合成によって調製することができる。このような改変は、例えば、抗体のアミノ酸配列内における、残基の欠失、及び/又は挿入及び/又は置換を含む。最終コンストラクトが所望の特性、例えば、抗原結合を有していることを条件として、欠失、挿入、及び置換の任意の組み合わせが、最終コンストラクトに到達させるために作成され得る。
ある実施態様において、一つ以上のアミノ酸置換を有する抗体変異体が提供される。置換型変異誘発の対象となる部位は、HVRとFRを含む。保存的置換は、表1の「保存的置換」の見出しの下に示されている。より実質的な変更が、表1の「典型的な置換」の見出しの下に与えられ、アミノ酸側鎖のクラスを参照して以下に更に説明される。アミノ酸置換は、目的の抗体に導入することができ、その産物は、所望の活性、例えば、抗原結合の保持/改善、免疫原性の減少、又はADCC又はCDCの改善についてスクリーニングされる。

所定の実施態様において、本明細書で提供される抗体は、抗体がグリコシル化される程度を増加または減少するように改変される。抗体へのグリコシル化部位の付加または欠失は、一以上のグリコシル化部位が作成または削除されるようにアミノ酸配列を変えることによって簡便に達成することができる。
ある実施態様において、1つまたは複数のアミノ酸改変を、本明細書で提供される抗体のFc領域に導入することができ、それによってFc領域変異体を生成する。Fc領域の変異体は、1つまたは複数のアミノ酸位置においてアミノ酸修飾(例えば置換)を含むヒトFc領域の配列(例えば、ヒトIgG1、IgG2、IgG3又はIgG4のFc領域)を含んでもよい。
ある実施態様において、抗体の1つ以上の残基がシステイン残基で置換されている、システイン操作抗体、例えば、「thioMAb(複数)」を作成することが望まれ得る。特定の実施態様において、置換された残基は、抗体のアクセス可能な部位で生じる。それらの残基をシステインで置換することにより、反応性チオール基は、それによって抗体のアクセス可能な部位に配置され、本明細書中でさらに記載されるように、イムノコンジュゲーを作成するために、例えば薬物部分またはリンカー−薬剤部分などの他の部分に抗体をコンジュゲートするために使用することができる。ある実施態様において、一以上の以下の残基がシステインで置換され得る:軽鎖のV205(Kabatの番号付け);重鎖のA118(EU番号付け);及び重鎖Fc領域のS400(EU番号付け)。システイン改変抗体は、例えば、米国特許第7521541号に記載のように生成され得る。
ある実施態様において、本明細書で提供される抗体は、当技術分野で知られ、容易に入手される追加の非タンパク質部分を含むように更に改変することができる。抗体の誘導体化に適した部分としては、限定されないが、水溶性ポリマーを含む。水溶性ポリマーの非限定的な例は、限定されないが、ポリエチレングリコール(PEG)、エチレングリコール/プロピレングリコールの共重合体、カルボキシメチルセルロース、デキストラン、ポリビニルアルコール、ポリビニルピロリドン、ポリ−1,3−ジオキソラン、ポリ−1,3,6−トリオキソラン、エチレン/無水マレイン酸共重合体、ポリアミノ酸(単独重合体又はランダム共重合体の何れか)及びデキストラン又はポリ(n−ビニルピロリドン)ポリエチレングリコール、プロピレングリコール単独重合体、プロピレンオキシド/エチレンオキシド共重合体、ポリオキシエチル化ポリオール(例えばグリセロール)、ポリビニルアルコール及びこれらの混合物を包含する。ポリエチレングリコールプロピオンアルデヒドはその水中での安定性に起因して製造における利点を有し得る。ポリマーは何れかの分子量のものであってよく、そして分枝鎖又は未分枝鎖であってよい。抗体に結合するポリマーの数は変動してよく、そして、一以上の重合体が結合する場合は、それらは同じか又は異なる分子であってよい。一般的に、誘導体化に使用するポリマーの数及び/又は種類は、限定されないが、向上させるべき抗体の特定の特性又は機能、抗体誘導体が限定された条件下で治療に使用されるのか等を含む考慮に基づいて決定することができる。
抗体は、例えば米国特許第4,816,567号で説明したように、組換えの方法および組成物を用いて製造することができる。一実施態様において、本明細書に記載される抗ETBR抗体をコードする単離された核酸が提供される。このような核酸は、抗体のVLを含むアミノ酸配列、及び/又は抗体のVHを含むアミノ酸配列(例えば、抗体の軽鎖及び/又は重鎖)をコードし得る。更なる実施態様において、そのような核酸を含む1つ以上のベクター(例えば、発現ベクター)が提供される。更なる実施態様において、そのような核酸を含む宿主細胞が提供される。一実施態様において、宿主細胞は以下を含む(例えば、以下で形質転換される):(1)抗体のVLを含むアミノ酸配列、及び抗体のVHを含むアミノ酸配列をコードする核酸を含むベクター、又は(2)抗体のVLを含むアミノ酸配列をコードする核酸を含む第一ベクター、及び抗体のVHを含むアミノ酸配列をコードする核酸を含む第二ベクター。一実施態様において、宿主細胞は、真核生物、例えばチャイニーズハムスター卵巣(CHO)細胞、またはリンパ系細胞(例えば、Y0、NS0、Sp20細胞)である。一実施態様において、抗ETBR抗体を作る方法が提供され、その方法は、上記のように、抗体の発現に適した条件下で、抗体をコードする核酸を含む宿主細胞を培養することを含み、および必要に応じて、宿主細胞(または宿主細胞培養培地)から抗体を回収することを含む。
本明細書で提供される抗ETBR抗体は、同定され、当技術分野で公知の様々なアッセイによってその物理的/化学的性質及び/又は生物学的活性についてスクリーニングされ、または特徴づけることができる。
一態様において、本発明の抗体は、例えばELISA、ウエスタンブロット法など公知の方法により、その抗原結合活性について試験される。
一態様において、アッセイは、抗ETBR抗体及び/又はBRAFi化合物は、生物学的活性を有するか否かを同定するために与えられる。生物学的活性は、実施例に記載されるもの、例えば、インビトロメラノーマ細胞生存アッセイ、又はメラノーマ細胞株がヌードマウスに移植され、腫瘍増殖阻害(TGI)が評価されるインビボ異種移植モデルを含むことができる。
本発明はまた、化学療法剤又は薬剤、成長抑制剤、毒素(例えば、タンパク質毒素、細菌、真菌、植物、または動物起源の酵素活性毒素、又はそれらの断片)、又は放射性同位元素など、1つ以上の細胞傷害性薬物にコンジュゲートした本明細書中の抗ETBR抗体を含むイムノコンジュゲートを提供する。
本発明のイムノコンジュゲート(又は「抗体−薬剤コンジュゲート」(「ADC」))は、下記式Iであって良く、ここで抗体は任意のリンカー(L)を通じて一又は複数の薬剤部分(D)にコンジュゲートしている。
リンカーは、一以上のリンカー成分を含んでもよい。例示的なリンカー成分は、6−マレイミドカプロイル(「MC」)、マレイミドプロパノイル(「MP」)、バリン−シトルリン(「val−cit」又は「vc」)、アラニン−フェニルアラニン(「ala−phe」)、p−アミノベンジルオキシカルボンイル(「PAB」)、N−スクシンイミジル4(2−ピリジルチオ)ペンタノエート(「SPP」)、N−スクシンイミジル4−(N−マレイミドメチル)シクロヘキサン−1カルボキシレート(「SMCC」)、及びN−スクシンイミジル(4−イオド−アセチル)アミノ安息香酸エステル(「SIAB」)を含む。様々なリンカー成分は当分野で公知であり、そのうち幾つかは以下において記述される。
メイタンシン及びメイタンシノイド
ここで、Abは抗体であり、nは0、1又は2であり、そして、pは1、2、3又は4である。
幾つかの実施態様では、イムノコンジュゲートは、ドラスタチン又はドロスタチンペプチド性類似体又は誘導体、例えばアウリスタチン(auristatin)(米国特許第5635483号;同第5780588号)にコンジュゲートした抗体を含んでなる。ドラスタチン及びアウリスタチンは、微小管動態、GTP加水分解及び核と細胞の***を妨げ(Woyke 等 (2001) Antimicrob. Agents and Chemother. 45(12): 3580-3584)、抗癌活性(米国特許第5663149号)及び抗真菌活性(Pettit 等 (1998) Antimicrob. Agents Chemother. 42:2961-2965)を有することが示されている。ドラスタチン又はアウリスタチン薬剤成分は、ペプチジル薬剤分子のN(アミノ)末端又はC(カルボキシル)末端により抗体に接着しうる(国際公開公報02/088172)。
ある例示的な実施態様では、R10は−フェニールである。例示的な実施態様では、Zが−O−の場合、R11は−H、メチル又はt−ブチルである。一実施態様では、Zが−NHである場合に、R11は−CH(R15)2であり、ここでR15は−(CH2)n−N(R16)2であり、R16は−C1−C8アルキル又は−(CH2)n−COOHである。
他の実施態様では、Zが−NHである場合に、R11は−CH(R15)2であり、ここでR15は−(CH2)n−SO3Hである。
他の実施態様では、イムノコンジュゲートは、一又は複数のカリケアマイシン分子と結合した抗体を含んでなる。抗生物質のカリケアマイシンファミリーはサブ−ピコモルの濃度で二重鎖DNA切断を生じることができる。カリケアマイシンファミリーのコンジュゲートの調製については、米国特許第5712374号、同5714586号、同5739116号、同5767285号、同5770701号、同5770710号、同5773001号、同5877296号(全て、American Cyanamid Company)を参照のこと。使用可能なカリケアマイシンの構造類似体には、限定するものではないが、γ1 I、α2 I、α3 I、N−アセチル−γ1 I、PSAG及びθI 1(Hinman等、Cancer Research, 53:3336-3342(1993)、Lode等 Cancer Research, 58:2925-2928(1998)及び上述したAmerican Cyanamidの米国特許)が含まれる。抗体が結合可能な他の抗腫瘍剤は、葉酸代謝拮抗薬であるQFAである。カリケアマイシン及びQFAは双方共、細胞内に作用部位を有し、原形質膜を容易には通過しない。従って、抗体媒介性インターナリゼーションによるこれらの薬剤の細胞への取込により、細胞障害効果が大きく向上する。
抗体と結合可能な他の抗腫瘍剤には、BCNU、ストレプトゾイシン、ビンクリスチン及び5−フルオロウラシル、米国特許第5053394号、同第5770710号に記載されており、集合的にLL−E33288複合体として公知の薬剤のファミリー、並びにエスペラマイシン(esperamicine)(米国特許第5877296号)が含まれる。
薬剤ローディングは、式Iの分子において、抗体当たりの薬剤部分の平均数であるpによって表される。薬剤ローディングは抗体当たり1〜20の薬剤部分(D)の範囲であってもよい。式IのADCには、1から20の薬剤部分の範囲でコンジュゲートされる抗体の集まりが含まれる。コンジュゲート反応からのADCの調製において抗体当たりの薬剤部分の平均数は、質量分析、ELISAアッセイ及びHPLCなどの従来の方法によって特徴付けられうる。pの観点からのADCの定量的分布も決定されうる。いくつかの場合では、pが他の薬剤ローディングを有するADCの一定の値である場合の均質なADCの分離、精製及び特徴づけは、逆相HPLC又は電気泳動などの手段によって達成されうる。
式IのADCは、当業者に公知の有機化学的反応、条件及び試薬を用いた様々な手段、例えば、(1)共有結合によってAb−Lを形成するための二価のリンカーと抗体の求核基との反応とその後の薬剤部分Dとの反応、及び(2)共有結合によってD−Lを形成するための二価のリンカーと薬剤部分の求核基との反応とその後の抗体の求核基との反応などによって調製されてもよい。後者の経路による式IのADCを調製するための例示的な方法は米国特許公開第2005−0238649号A1に記載されており、出典明記によって本明細書中に明確に援用される。
一態様において、本発明は、本発明の少なくとも一抗体及び/又はその少なくとも一のイムノコンジュゲートを含む薬学的製剤を更に提供する。いくつかの実施態様では、薬学的製剤は、1)本発明の抗体及び/又はこのイムノコンジュゲートと、2)薬学的許容性のある担体とを含有する。いくつかの実施態様では、薬学的製剤は、1)本発明の抗体及び/又はこのイムノコンジュゲートと、場合によって2)少なくとも一の付加的治療剤とを含有する。更なる治療剤には以下に記載のものが含まれるが、これらに限定されるものではない。
本明細書に記載の抗ETBR抗体の薬学的処方物は、所望の程度の純度を有するその抗体と任意の薬学的に許容される担体(Remington's Pharmaceutical Sciences 16th edition, Osol, A. Ed.: Williams and Wilkins PA, USA (1980))とを、凍結乾燥製剤または水性溶液の形態で混合することによって調製される。薬学的に許容される担体は、使用される投薬量および濃度でレシピエントに毒性でなく、そしてこれには、限定しないが、リン酸塩、クエン酸塩および他の有機酸のような緩衝液;アスコルビン酸およびメチオニンを含む抗酸化剤;防腐剤(例えば、オクタデシルジメチオルベンジルアンモニウムクロライド;ヘキサメトニウムクロライド;塩化ベンザルコニウム、塩化ベンゼトニウム、フェノール、ブチルまたはベンジルアルコール;アルキルパラベン、例えば、メチルまたはプロピルパラベン;カテコール;レゾルシノール;シクロヘキサノール;3−ペンタノール;およびm−クレゾール);低分子量(約10残基未満)ポリペプチド;タンパク質、例えば、血清アルブミン、ゼラチン、または免疫グロブリン;親水性ポリマー、例えば、ポリビニルピロリドン;アミノ酸、例えば、グリシン、グルタミン、アスパラギン、ヒスチジン、アルギニンまたはリジン;マンノサッカライド、ジサッカライド、およびグルコース、マンノースまたはデキストリンを含む他の炭水化物;キレート剤、例えば、EDTA;糖、例えば、スクロース、マンニトール、トレハロースまたはソルビトール;塩形成対イオン、例えば、ナトリウム、金属錯体(例えば、Zn−タンパク質錯体);及び/又はポリエチレングリコール(PEG)等の非イオン性界面活性剤が挙げられる。本明細書における典型的な薬学的に許容される担体は、介在性薬物分散剤、例えば、水溶性の中性アクティブヒアルロニダーゼ糖タンパク質(sHASEGP)、例えば、rHuPH20(HYLENEX(登録商標)、Baxter International, Inc.)などのヒト可溶性PH−20ヒアルロニダーゼ糖タンパク質を更に含む。所定の典型的なsHASEGP及び使用法は、rHuPH20を含み、米国特許出願公開第2005/0260186号及び第2006/0104968に開示されている。一態様において、sHASEGPは、コンドロイチナーゼなどの1つまたは複数の追加のグルコサミノグリカンと組み合わされる。
本明細書で提供される抗ETBR抗体のいずれかを、治療方法で使用することができる。
本発明の他の実施態様において、上述した障害の治療、予防、及び/又は診断に有用な物質を含む製造品が提供される。製造品は、容器とラベルまたは容器上にある又は容器に付属するパッケージ挿入物を含む。好適な容器は、例としてボトル、バイアル、シリンジ、IV輸液バッグ等を含む。容器はガラス又はプラスチックなどの様々な物質から形成されうる。容器は、疾患の治療、予防、及び/又は診断に有効である、それ自体か、又はその他の組成物と併用される化合物を収容し、無菌のアクセスポートを有し得る(例えば、容器は皮下注射針による穴あきストッパーを有する静脈内溶液バッグ又はバイアルであってよい)。組成物中の少なくとも一の活性剤は本発明の抗体である。ラベルまたはパッケージ挿入物は、組成物が特定の症状の治療のために使用されることを示している。更に、製造品は、(a)組成物が本発明の抗体を包含する組成物を含む第一の容器;および(b)組成物が更なる細胞障害性又はその他の治療的薬剤を包含する組成物を含む第2の容器を含み得る。本発明の本実施態様における製造品は、組成物が特定の疾患を治療することに用いることができることを示すパッケージ挿入物をさらに含んでいてもよい。別法として、または加えて、製造品は、薬学的に許容されるバッファー、例えば注射用静菌水(BWFI)、リン酸緩衝化塩水、リンガー溶液およびデキストロース溶液を含む第二(または第三)の容器をさらに含んでもよい。これは、他のバッファー、希釈剤、フィルター、針、およびシリンジを含む、商業的およびユーザーの立場から望まれる他の物質をさらに含んでもよい。
抗ETBR抗体−ADCの候補であるHu5E9v1−ADCは、細胞株A2058(アメリカン・タイプ・カルチャー・コレクションから入手)の場合、ETBRの比較的低いコピー数、又は細胞株UACC−257X2.2の場合、ETBRの高コピー数のどちらかを発現するメラノーマ細胞株においてインビトロで評価された。UACC−257X2.2細胞株は、インビボでの増殖に最適化された親のUACC−257細胞株の誘導体である。親のUACC−257は雌のNCrヌードマウスの右脇腹に皮下注射され、1腫瘍を採取して分離し、インビトロで増殖させ、UACC−257X1.2細胞株を得た。UACC−257X1.2株は、細胞株の増殖を改善する目的のため、再び雌のNCrヌードマウスの右脇腹に皮下注射された。この試験からの腫瘍は収集され、UACC−257X2.2細胞株を生成するために、再びインビトロでの増殖に適応させた。フローサイトメトリーによって決定されるように、この細胞株はETBRを高レベルで発現する。これらの細胞株におけるHu5E9v1−vc−MMAEの細胞殺傷に対する受容体レベルの関係を以下のように評価した。
上記実施例1に記載した試験に基づいて、メラノーマ細胞株A2058およびUACC−257X2.2を、広範囲のETBRの発現を意味するインビボにおける抗腫瘍活性の研究に適したモデルとして選択した。UACC−257X2.2メラノーマ細胞株は、インビボでの増殖に最適化された親のUACC−257メラノーマ細胞株(国立癌研究所(NCI))の誘導体である。具体的には、親のUACC−257は雌のNCrヌードマウスの右脇腹に皮下注射され、1腫瘍を採取し、インビトロで増殖させ、UACC−257X1.2細胞株を得た。UACC−257X1.2株は、細胞株の増殖を改善する目的のため、再び雌のNCrヌードマウスの右脇腹に皮下注射された。この試験からの腫瘍は収集され、UACC−257X2.2細胞株を生成するために、再びインビトロでの増殖に適応させた。この細胞株及びこの株に由来する腫瘍は、親細胞株UACC−257に匹敵するETBRを発現する(データ非表示)。
ETBR転写物およびタンパク質(全タンパク質及び細胞表面タンパク質)の発現レベルに対するBRAF阻害薬物の作用は、例えば、BRAF(V600E)の変異体、BRAFの野生型及びRAS(Q61L)の変異体など、メラノーマの様々な遺伝的背景を表す様々なメラノーマ細胞で評価した。
上の実施例3に示される結果を考慮し、抗ETBR ADCのインビボ効力に対するBRAF阻害薬物の影響を試験した。これを行うにため、Hu5E9v1−ADCとBRAFi−945の様々な組み合わせためにおいてインビボでの有効性が、前述したUACC−257X2.2メラノーマモデルに対して評価された。腫瘍は、約200mm3の平均サイズまで増殖させ、その上で動物を10匹の群に無作為に分けた。適切なビヒクル対照(Klucel LF)又はBRAFi−945は、試験日0日目に始まり21日間1日1回、投与量が1mpk、6mpk、又は20mpkで経口投与された。単回の1mpk又は3mpk投与量のHu5E9v1−ADC又は対照、ヒスチジン緩衝液#8が、試験日1日目に尾静脈を介して(945の2回投与量後に)静脈投与された。
上記実施例に記載される試験は、BRAF阻害薬物RG7204.とのHu5E9v1−ADCの組み合わせによるインビボでの有効性の評価の精密化を可能とする。薬間間の拮抗作用の欠如が予想され、従って、薬物の組み合わせによる有効性は低用量で試験した。COLO829異種移植モデルが、組み合わせ試験のストリンジェンシーを更に増加させる、ETBR発現の中庸なレベルの代表として選択された。腫瘍は、約200mm3の平均サイズまで増殖させ、その上で動物を9匹の群に無作為に分けた。適切なビヒクル対照(Klucel LF)又はG00044364.1−12(RG7204)は、試験日0日目に開始し21日間1日1回、投与量が10mpk又は30mpkで経口投与された。Hu5E9v1−ADCの1mpk或いは3mpkの何れかの、又は対照のヒスチジン緩衝液#8の単一用量が1日目に静脈内投与された。結果を図7A、7B、7C、及び7Dに示す。
A2058異種移植モデルにおけるHu5E9v1−ADC及びRG7204の有効性を試験した。このモデルは、それが示す高レベルのストリンジェンシーのため特に重要である。A2058異種移植モデルは、メラノーマ患者に見いだされるETBRの発現スペクトルの下限を示し、それによって、それを抗ETBR ADCを実現するための挑戦的なモデルとしている。更に、そのBRAF V600E変異状態にも関わらず、このモデルは、インビトロでの致死効果が>20μMを有し、RG7204に対して非応答性であることを示している。
ETBR転写物及びタンパク質(総タンパク質及び細胞表面タンパク質)の発現レベルへのMEK阻害薬物の作用が、BRAF野生型或いは変異型及び/又はRAS野生型或いは変異型の何れかである様々なメラノーマ細胞:COLO829(BRAFV600E)、A2058(BRAFV600E)、SK23−MEL(BRAFWT/RASWT)、又はIPC−298(BRAFWT/RASC61L)で評価された。
上の実施例7に示される結果を考慮し、抗ETBR ADCのインビボ効力に対する本明細書に記載されるMEK阻害剤の影響を試験した。これを行うために、Hu5E9v1−ADC及びMEKi−623及び/又はMEKi−973の様々な組み合わせについてのインビボ有効性を、インビボモデルにおけるA2058およびSK−MEL−23およびIPC−298に対して、実施例4に上述されるように実施して評価した。適切なメチルセルローストゥイーンビヒクル対照(0.5%メチルセルロース、0.2%のTween−80(MCT)又はMEK阻害剤は、試験日0日目に始まり21日間1日1回、投与量が1mpk、3mpk、又は7.5mpkで経口投与された。単回の3mpk又は6mpk投与量のHu5E9v1−ADC又は対照、ヒスチジン緩衝液#8が、試験日1日目に尾静脈を介して(MEK阻害剤の2回投与量後に)静脈投与された。
図7に示される試験の終了時に収集した腫瘍(COLO829対抗ETBR−ADCとBRAFi RG7204の組み合わせ)はETBRの増加を示さなかった。これは、腫瘍収集のタイミングが投与されたBRAFi薬物のウオッシュ・アウト期間を十分過ぎていたことに起因するであろう。細胞株においてBRAFi/MEKiのインビトロでの作用(即ちETBRの増加及びPerkの減少)はインビボでも生じるか、そしてそれゆえ、抗ETBR ADCとBRAFi/MEKiの組み合わせにおいてより高い有効性を可能にするかどうかを評価するため、以下の実験を実施した。
Claims (77)
- 被験体へ抗エンドセリンB受容体(ETBR)抗体の有効量をMAPキナーゼ阻害剤の有効量と組み合わせて投与することを含む、メラノーマに罹患している被験体における腫瘍増殖阻害(TGI)の方法。
- 前記組み合わせは相乗的である、請求項1に記載の方法。
- 前記TGIは、抗ETBR抗体単独を使用して観察されるTGIよりも大きい、請求項1に記載の方法。
- 前記TGIは、MAPキナーゼ阻害剤単独を使用して観察されるTGIよりも大きい、請求項1に記載の方法。
- TGIは、抗ETBR抗体単独の使用よりも約10%を超えて、又は約15%を超えて、又は約20%を超えて、又は約25%を超えて、又は約30%を超えて、又は約35%を超えて、又は約40%を超えて、又は約45%を超えて、又は約50%を超えて、又は約55%を超えて、又は約60%を超えて、又は約65%を超えて、又は約70%を超えて大きい、請求項3に記載の方法。
- TGIは、MAPキナーゼ阻害剤単独の使用よりも約10%を超えて、又は約15%を超えて、又は約20%を超えて、又は約25%を超えて、又は約30%を超えて、又は約35%を超えて、又は約40%を超えて、又は約45%を超えて、又は約50%を超えて、又は約55%を超えて、又は約60%を超えて、又は約65%を超えて、又は約70%を超えて大きい、請求項4に記載の方法。
- 前記抗ETBR抗体は、配列番号10のアミノ酸番号64から101からなるETBRエピトープに特異的に結合する、請求項1に記載の方法。
- 前記抗ETBR抗体は3つの可変重鎖CDR及び3つの可変経鎖CDRを有し、ここでVH CDR1は配列番号1であり、VH CDR2は配列番号2であり、VH CDR3は配列番号3であり、及びVL CDR1は配列番号4であり、VL CDR2は配列番号5であり、VL CDR3は配列番号6である、請求項1に記載の方法。
- 前記抗ETBR抗体は可変重鎖及び可変経鎖を有し、ここで前記VHは配列番号7又は9である、請求項1に記載の方法。
- 前記VLは配列番号8である、請求項9に記載の方法。
- 前記抗ETBR抗体は細胞毒素にコンジュゲートする、請求項1に記載の方法。
- 前記細胞毒素は、毒素、抗生物質、放射性同位体及び核酸分解酵素からなる群から選択される細胞傷害性薬物である、請求項11に記載の方法。
