標準的な方法は時間がかかるだけでなく細胞培養のメンテナンスに高度な努力とそれによる経費がかかっていた。
この観測の観点から、ここで説明されたモノクローン抗体細胞株の生産性の為の目新しいファストトラック(fast-track)品質評価は新規生産細胞株の樹立の為の信頼できる手順を確立するのに必要である開発時間を抜本的に減少させるだろう。
本発明は従来技術から自明なものではない。
しかしながら、HTRF(登録商標)アッセイに関するいかなる先行文献も例えば組み替えタンパクのようなタンパクの産生宿主細胞株のスクリーニング又は細胞クローンの選択方法における適用は示唆していない。

FACSによりデポジットされた単一細胞が96ウェル内で細胞群に成長する間に新規モノクロナール細胞株の生産性の為の可能な限り最も早期のスクリーンが行われる。この概念は自動化 96ウェルインキュベーターの自動化力価測定を規則正しい間隔で行う無菌ユニットへの統合を必要とする。
IgG型抗体を産生するCHO DG44 モノクロナール細胞株は既知組成である無血清培地内で96ウェルプレートで培養される。上清を収集し、培地中の抗体濃度を、96ウェル形式におけるサンドイッチ型抗IgG ELISAにより、同時に96ウェル及び384ウェルアッセイ形式におけるHTRFにより測定する。ELISA及びHTRF(登録商標)形式において使用する2つの抗体は同じ供給源由来である。
単一細胞を含む96ウェルプレートをFACSユニットから自動化インキュベーターへ移動する。ソフトウェアが単一のプレートのインキュベーターからエアロックを介した無菌環境への移動をスケジュールする。細胞がインキュベーターに戻される間に培養体積の2.5% (v/v)未満の試料を各上清から取り除き、ピペッティングユニットで希釈する。ピペッティングユニットは次に試料及びHTRF(登録商標)試薬を384ウェルプレート内で混合し、それをインキュベートのために貯蔵ホテル(storage hotel)に移動する。2時間後にプレートを665 nm及び620 nmにおける測定のために読み取り装置に移動する。試料の追跡はバーコードプレート及びバーコードリーダーで保証する。

IgG4型治療用抗体を発現する安定なCHO細胞プールはFACSによって96ウェル内に単一細胞デポジットした。細胞を自動化インキュベーターに移動し、自動化力価測定プログラムは単一細胞スクリーニングの15日後に開始する。抗体力価は各ウェルにつき3日ごとに計測する。
A) IgG産生 CHOクローンを96ウェルプレートにデポジットし、HTRF(登録商標)アッセイでクローニングの10、13、15及び17日後に測定した。
IgGタイプ1治療用抗体を発現する安定なCHO細胞プールはFACSによって96ウェル内に単一細胞デポジットされた。細胞を自動化インキュベーターに移動し、自動化力価測定プログラムは単一細胞スクリーニングの10日後に開始した。抗体力価をHTRF(登録商標)スクリーニング プラットホームにより各ウェルにつき2〜3日ごとに4回測定した。
各個々の列(異なる灰色の陰影)はそれぞれモノクロナール細胞株を表す単一の96ウェルの力価曲線を表す。
B) 力価によるクローンの等級付け
続いて、クローンは単一細胞デポジションの17日後に採集し、6ウェルプレートに拡張し3継代の間に力価測定を行う。最初期クローンスクリーニング(IECS)によって選択されたクローンの力価をMAT6スケールによって得た力価と比較した。IECSにおいて高い力価を有するクローンは又MAT6スケールにおいても高力価である。
具体的には、IECSによって上位クローンとして同定されたクローンの5つのうち4つが続いてMAT6スケール スクリーニングによって同様に上位クローンとして同定されたものと同じである。
表のダーク/グレーで埋められたセルは首位のクローンを表す。
インキュベーターは好ましくは細胞培養バイアルを自動化プラットホームに連続的な又は予定された提示又は移動を可能とする自動化インキュベーターである。
HTRF(登録商標)(均一時間分解蛍光測定法)はTR-FRET、つまりFRET化学現象と長い発光半減期を有するフルオロフォアの使用の組み合わせを基礎とする技術である。一方HTRF(登録商標)は他のTR-FRET産物から分離する多くの特性を有するTR-FRET化学現象を基礎とする。これらは、非常に長い半減期を有するランタニド(ユーロピウム)の使用、Eu3+のクリプテートへの結合、アッセイの安定性の増加及び消光及び試料の干渉を補正することができるレシオ測定(ratiometric measurement)の使用を付与する実体を含む。他のHTRF(登録商標)法の特徴には均一なアッセイ形式、低バックグラウンド、アッセイの小型化の容易、DMSO及びEDTAのような添加剤への耐性、偽陽性/偽陰性の少なさ、細胞ベースの機能的アッセイが含まれる。
「選択増幅マーカー遺伝子」は通常、これらの条件下で真核細胞が成長するのに必要な酵素をコードする。例えば、選択増幅マーカー遺伝子は、トランスフェクトされた宿主細胞を選択薬剤メトトレキセート(MTX)の存在下で育成した際に増幅するDHFRをコードする。非限定的な例である表3に示した選択遺伝子も本発明の実施に使用することができる増幅マーカー遺伝子である。表3に列挙された選択増幅マーカー遺伝子については参照として組み込まれる Kaufman, Methods in Enzymology, 185:537-566 (1990)を参照されたい。従って、本明細書に記載された任意の方法で遺伝子的に改変された宿主細胞は本発明に含まれ、そのなかで選択増幅 マーカー遺伝子はジヒドロ葉酸還元酵素(DHFR)、グルタミン合成酵素、CAD、アデノシン脱アミノ酵素、アデニレート脱アミノ酵素、UMP合成酵素、IMP 5'-脱水素酵素,キサンチングアニンホスホリボシルトランスフェラーゼ、HGPRTase、チミジンキナーゼ、チミジル酸合成酵素、P-糖タンパク 170、リボヌクレオチド還元酵素、アスパラギン合成酵素、アルギニノコハク酸合成酵素、オルニチン脱炭酸酵素、HMG CoA還元酵素、アセチルグルコサミン転移酵素、スレオニル-tRNA 合成酵素又はNa+K+-ATPaseの機能を有するポリペプチドをコードする。
対象生産物アンチセンスRNAであっても良い。
a)対象タンパクを発現している単一細胞を個々の容器の培地中に入れる工程、
b)少なくとも1日細胞を培養する工程、
c)細胞が最初の継代をする前に各容器から培養の一定分量を取り除く工程、
d)各一定分量中の対象タンパクの量を測定する工程、
e)各一定分量中のタンパク測定量に従ってクローンを選択する工程、
を特徴とする前記方法である。
a. 対象タンパクを発現している単一細胞をマルチウェル容器の培地中にデポジットする工程、
b. 誘導した細胞培養を10回継代する工程、
c. 前記マルチウェル容器を自動化インキュベーターに移動する工程、
d. 続いて前記マルチウェル容器をインキュベーターからエアロックを介してクラスAの粒子数が立方メートルあたり100未満である無菌環境に移動する工程
e. 各容器から培養の一定分量を取り除く工程、
f. 細胞がインキュベーターに戻される間に試料をピペッティングユニットを用いて希釈する工程
g. 希釈した試料とアッセイ反応液を他のマルチウェル容器内で混合する工程、
h. 工程g)のマルチウェル容器をインキュベーションのために貯蔵ホテルに移動する工程、:
i. 工程h)のマルチウェルプレートを読み取り装置に移動する工程、
j. 各容器中の対象タンパクの量を測定する工程であって、これによりスループットが少なくとも12時間以内に250測定であり、好ましくは12時間以内に500測定であり、より好ましくは12時間以内に2000測定であり、最も好ましくは4000測定又は一定分量である工程
を特徴とする、前記方法に関する。
発明の方法の好ましい実施態様は試料の追跡はバーコードプレート及びバーコードリーダーで保証されている方法である。
他の好ましい実施態様は工程b)における継代の数が0であり、工程e)が細胞が最初に継代する前に行われる方法である。
a. IgG型抗体のFc部分及び
b. IgG型抗体の軽鎖
に対する抗体の検出を含む方法によって行われる任意の発明の方法である。
本発明は真核細胞、例えば哺乳類細胞における、無血清培養条件化でタンパクを産生する方法に関し、以下の工程:
a. 対象タンパクをコードする対象遺伝子を含む真核細胞を産生する工程、
b. 細胞の増殖を許容する無血清条件化での細胞の培養工程
c. 単一細胞を96ウェルプレートのようなマルチウェル容器へデポジットする工程
d. 前記単一細胞の培養であって、任意に自家フィーダー細胞の存在下の前記培養工程
e. 前述の任意の1つの発明の方法によるクローン細胞のスクリーニング工程
f. 多量の対象タンパクを発現すると測定された細胞の上位30%、好ましくは上位20%及び最も好ましくは上位10%の選択細胞を培養する工程
g. 例えば細胞を上清から分離することによる対象タンパクの収集工程及び
h. 対象タンパクの精製工程
に従うこと特徴とする、前記方法に関する。
本発明はさらに記載された方法のうち任意の1つの方法により選択された産生宿主細胞株に関する。
以下の工程:
a)対象タンパクを発現するように遺伝子的に改変されたトランスフェクトされた細胞
の単一細胞クローニングを行う工程
b)自動化プラットホームを用いてマルチウェルプレート方式において育成されたモノ
クロナール細胞の初代培養における前記対象タンパクを検出するのに適したタンパ
ク検出アッセイを行う工程
c)初代培養の無菌環境を維持しながら行う工程
を特徴とする、前記方法に関する。
a.単一細胞を含む96ウェルプレートがFACSユニットを自動化インキュベータ
ーに移動する工程
b.ソフトウェアが単一のプレートのインキュベーターからエアロックを介した
無菌環境への移動を連続敵意スケジュールする工程
c.細胞がインキュベーターに戻される間に上清がピペッティングユニットによ
り除去され希釈される工程
d.ピペッティングユニットが次に試料及びアッセイ試薬をマルチウェルプレー
ト内で混合しそれらをインキュベーションのために貯蔵ホテルに移動する工程
e.2時間後プレートを読み取り装置に移動し予想された波長において測定する
工程
f.試料の追跡はバーコードプレート及びバーコードリーダーで保証した。
が行われる前記方法に関する。
力価ポテンシャルは培養が最初の継代前に達する最終的なタンパク濃度を意味する。
材料及び方法
細胞培養
産生及び開発スケールにおいて使用する全ての細胞株は、系列の種培養として、表面通気されたT-フラスコ (Nunc社、デンマーク)でインキュベーター内(Thermo社、ドイツ)において又は空気と5% CO2の混合物を散布されたスピナーフラスコ(Wheaton社、米国)で特別に設計されたインキュベータールーム内において37℃の温度で維持した。
パルスプロセッシング、ソートエンハンスメントモジュール及び自動化細胞ポジションユニットを備えたFACS Vantage (Coulter EPICS ALTRA HyPerSort System))フローサイトメーター装置を分析及び細胞sortingに用いた。488 nmに調整したアルゴンレーザー(Coherent)を使用した。レーザー出力は220 mWであった。生存細胞は前方散乱光(forward scatter (FSC))対、側方散乱光(side scatter (SSC))のドットプロットに従った全ての単一細胞を含むゲートを設定することでソートした。ソートした細胞は200μlの成長培地を含む96ウェル マイクロタイタープレートにウェルあたり2個の細胞を自動化細胞でポジションユニットを用いてデポジットした。無菌ソーティングの為に、セルソーターの管状部品はシース液(sheath fluid)として1時間それぞれ以下の溶液70%エタノール、滅菌H2Oを流すことで洗浄し滅菌した。
HTRFアッセイはドナー及びアクセプター分子間のFRETによるシグナルを発生する均一時間分解蛍光測定法である。
ドナーは、多環式クリプテート内にケージ化されたEu3+(Eu-クリプテート)であり、一方アクセプターは改変されたアロフィコシアニンタンパクである。ドナーが337 nmにおけるレーザーによる励起により、アクセプターに、それらが非常に近接(690Å)している際に620 nmでエネルギーが転移し、ミリセカンドの長期にわたる665nmにおける発光が起こる。発光の記録における50-lsの遅延及び665-及び620-nmの発光の比を分析することが、培地及び不対のフルオロフォアからの蛍光の干渉を最小限にする。培地中のIgG型抗体の内容量を検出するために、Eu-クリプテートをFc領域に特異的に結合し、抗体がIgG 産物に結合した際に提示される抗ヒトIgG 抗体に結合し、一方κ軽鎖に特異的に結合する抗ヒトIgG 抗体は複合体を完成するためにD2アクセプターとして標識される。このアッセイ形式は培地中のIgG型抗体の検出を1 mg/lより低い濃度で可能にする。
HTRF(登録商標)アッセイは完全自動化ピペッティングプラットホームで無菌条件下行われる。単一細胞を含む96ウェルプレートはFACSユニットから自動化インキュベーターに移動される。ソフトウェアが単一のプレートのインキュベーターからエアロックを介した無菌環境への移動をスケジュールする。細胞がインキュベーターに戻される間に培養体積の2.5% (v/v)未満の試料が各上清から除去され、ピペッティングユニットにより希釈される。ピペッティングユニットは続いて試料及びHTRF(登録商標)試薬を384ウェルプレート内で混合しそれらをインキュベーションのために貯蔵ホテルに移動する。2時間後プレートを665 nm及び620 nmにおける測定のために読み取り装置に移動する。試料の追跡はバーコードプレート及びバーコードリーダーで保証した。
抗体は最初にリン酸バッファー(50mM pH8)中で透析しMillipore社のBiomax チップ(30 000 M.Wで除外)を用いて1mg/mLまで濃縮する。抗体は次にN-ヒドロキシ-スクシンイミドで活性化したクリプテートと30分間、室温で、分子比15(クリプテート/抗体)で反応させた。抗体クリプテート複合体は最終的に未反応のフルオロフォアからG25スーパーファインゲルで精製した。
抗体は最初にリン酸バッファー(50mM pH8.5)中で透析しMillipore社のBiomax チップ(30 000 M.Wで除外)を用いて1mg/mLまで濃縮する。抗体は次にN-ヒドロキシ-スクシンイミドで活性化したD2と1時間、室温で、分子比5(D2 / 抗体)で反応させた。抗体D2複合体は最終的に未反応のフルオロフォアからG25スーパーファインゲルで精製した。
無菌環境においてCHO細胞の培養上清中のIgG 抗体のHRTF-ベース測定を実行するロボットプラットホーム
図1Bは使用した最初期スクリーン設備の概要を示す。96ウェルで育成し、つづいてFACS-ベース 単一細胞デポジションした細胞の培養上清の生産物力価を測定した。さらに384ウェル 形式内における完全自動化方式で行う力価測定により高産生クローンの為のハイスループット初代スクリーニングが可能になった。
IgG-4 型抗体産生のモノクロナールCHO細胞自動化最初期スクリーニング
IgG4型抗体をコードする遺伝子を既知組成の無血清培地内で育成したCHO DG44 細胞内にトランスフェクトし、ネオマイシンで選択することにより安定な細胞プールを作製した。細胞は、自家フィーダー細胞を上記のように使用することを含むFACS-ベース単一細胞クローニングに供した。単一細胞クローニングの15日後に42個のプレートを自動化インキュベーター内に移動し、最初期クローンスクリーニングプログラムを開始した。すべてのクローン培養の上清は3日毎に採取し抗体濃度は記載されたHTRFアッセイによって測定した。
いくつかの培養は測定の最後の時点において初期の指数増殖期に入ったばかりである(9及び13に示したクローンのように)。
採取するタンパクの数及び/又は選択工程の時間枠の制限を望む場合は、所望の設定によりそれらのクローンの力価ポテンシャルを評価するのに使用する典型的な力価特性を作製することが可能である (数学的モデル化アプローチ)。力価ポテンシャルは培養が最初の継代前に達する最終的なタンパク濃度を意味する。
IgG-1型抗体産生モノクロナールCHO細胞の自動化最初期スクリーニングならびにMAT6ウェル内におけるさらなる継代培養及び最初期クローンスクリーニングとMAT6スケールにおける生産性の比較
IgG1型抗体をコードする遺伝子を既知組成の無血清培地内で育成したCHO DG44 細胞内にトランスフェクトし、安定な細胞プールをネオマイシンで選択することで作製した。細胞は、自家フィーダー細胞を上記のように使用することを含むFACS-ベース単一細胞クローニングに供した。単一細胞クローニングの10日後にプレートを自動化インキュベーター内に移動し、最初期クローンスクリーニングプログラムを開始した。すべてのクローン培養の上清は2〜3日毎に4回採取し抗体濃度は記載されたHTRFアッセイによって測定した。
IgG-1型抗体産生モノクロナールNS0細胞の自動化最初期スクリーニング
IgG1 型抗体をコードしている遺伝子を既知成分の無血清培地中で培養するNS0細胞にトランスフェクトし、ネオマイシン及び ピューロマイシンを用いた選択により安定な細胞プールを作製した。細胞は、材料及び方法の項に記載するようにFACS-ベース単一細胞クローニングに供した。単一細胞クローニングの15日後に42プレートを自動化インキュベーターに移動し最初期クローンスクリーニングプログラムを開始した。全てのクローン培養の上清を3日ごとに採取し、記載されたHTRFアッセイで抗体濃度を測定した。クローンはこれらのデータに従って序列化し、続いて選択したクローンを6ウェルプレートに拡張し、以前に得たデータを検証するために3継代の間、力価定量を行った。
Claims (43)
- 細胞クローンの選択方法であって、以下の工程
a. 対象タンパクを発現している単一細胞を個々の容器の培地中にデポジットする工程、
b. 少なくとも1日細胞を培養する工程、
c. 細胞が最初の継代をする前に各容器から培養の一定分量を取り除く工程、
d. 各一定分量中の対象タンパクの量を測定する工程、
e. 一定分量中のタンパク測定量に従ってクローンを選択する工程、
によることを特徴とする前記細胞クローンの選択方法。 - スループットが少なくとも12時間以内に250測定であり、好ましくは12時間以内に500測定であり、より好ましくは12時間以内に2000測定であり、最も好ましくは4000測定である、請求項1記載の方法。
- 工程c)がクラスAの粒子数が立方メートルあたり100未満である無菌環境下で行われる、請求項1又は2記載の方法。
- 少なくとも1つの工程がマルチウェルプレート内で行われる、請求項1〜3いずれか1項記載の方法。
- 少なくとも工程d)がマルチウェルプレート内で行われる、請求項4記載の方法。
- マルチウェルプレートが96ウェルプレート又は384ウェルプレートであり、好ましくは384ウェルプレートである、請求項4又は5記載の方法。

- 細胞クローンの選択方法であって、以下の工程

- 一定分量が 細胞培養体積の2.5% (v/v)未満であり、タンパク測定の検出感度が少なくとも1mg/lである、請求項16記載の方法。

に対する抗体を検出することを含む、請求項17記載の方法 - 抗体の検出が抗 h IgG (Fc)とユーロピウムクリプテートドナーの複合体及び抗 h κ 軽鎖とD2 アクセプターの複合体による、請求項21記載の方法。
- 培地が無血清及び/又は動物性成分無添加及び/又はタンパク無添加及び/又は既知組成である、請求項1〜22いずれか1項記載の方法。
- 細胞が懸濁培養中で育成される、請求項1〜23いずれか1項記載の方法。
- 選択されたクローンが多量の対象タンパクを発現すると測定された細胞の上位30%、好ましくは上位20%及び最も好ましくは上位10%である、請求項1〜24いずれか1項記載の方法。
- 方法がさらに自家フィーダー細胞を使用することを特徴とする、請求項1〜25いずれか1項記載の方法。
- デポジットした細胞がCH-又はBHK-細胞である場合に使用されるフィーダー細胞がハムスター細胞であり、デポジットした細胞がNSO細胞である場合に、マウス骨髄腫細胞がフィーダー細胞として使用される、請求項26記載の方法。
- デポジットした細胞が1mLの培地あたり100〜200,000個のフィーダー細胞の存在下育成される請求項26又は27記載の方法。
- 対象タンパクが治療用タンパクである、請求項1〜28いずれか一項記載の方法。
- タンパクが抗体、特に治療用抗体である、請求項1〜29いずれか1項記載の方法。
- デポジットした細胞がハムスター細胞、例えばCHO又はBHK細胞である、請求項1〜30いずれか1項記載の方法。
