Abstract
Description
− 162位のバリンが、別の中性アミノ酸残基で置換されている;
− 2011位のセリンが、別の親水性のアミノ酸残基で置換されている;
− 2223位のバリンが、酸性アミノ酸残基で置換されている;
− 740位のアルギニンから1640位のグルタミン酸までのB領域が、10〜25個の、好ましくは14〜20個のアミノ酸残基を含むアルギニンに富んだリンカーペプチドで置換されている。
ここで示す代表的実験において、2つのドナープールから得られた約106個の臍帯血由来CD34陽性細胞を、ゼラチン被覆プレート上のEDM中で培養した。その後、接着細胞が老化するまでEGM−2中で継代培養した。黒四角(WT;■)で定義されたグラフは、最初の継代後の時間に対してプロットした内皮細胞の数を表す。37日後(A)および23日後(B)にそれぞれコンストラクトcPPT−C(FVIIIΔB)IGWS(C(F8)IGWSと略記;白丸;○)およびコンストラクトpHR’SIN.cPPT−SEW(SEWと略記;白三角;△)を使用して、104個の細胞を48時間、形質導入のために培養した。これらの細胞もまた、増殖が止まるまでEGM−2中に維持された。
4つの異なるバッチからのCBECを、培養中の様々な時点においてフローサイトメトリーにより、Dil−Ac−LDLの取り込みおよび細胞表面マーカーの発現を分析した。X軸は、分析された細胞の蛍光強度である。Dil−Ac−LDLまたは抗体で標識された細胞は太線で表され、細線は適当な陰性対照を表す。CBECは一様にDil−Ac−LDLの取り込みおよび内皮マーカーVE−カドヘリン、CD146、およびCD31に対して陽性であることをパネルは示している。細胞のサブセットはCD34を発現し、非常に弱くではあるが、KDRを発現した。すべてのCBECが、CD133、HLA−DR、CD45、およびCD14に対しては陰性であった。
4つの独立した実験で、継体数3〜8のCBECは、レンチウイルスコンストラクトpHR’SIN.cPPT−SEW(SEWと略する)およびcPPT−C(FVIIIΔB)IGWS(C(F8)IGWSと略する)により形質導入される。(A)それぞれのベクターに対して、感染多重度(MOI)10および100で該コンストラクトにより形質導入を行い、陰性対照として形質導入していない細胞のEGFP発現の分析を行うことによって、形質導入効率を測定した。(B)ヒストグラムは、FACSにより検出されたEGFP発現のレベルを表す。線図は、4つの実験の代表例の結果を示す。FACS分析は、C(F8)IGWSによる形質導入(MOI 10;曲線下が黒い領域)、およびSEWによる形質導入(MOI 10;曲線下が灰色の領域)して30日後に行った。形質導入していない細胞は、陰性対照として用いた(曲線下が白い領域)。
(AおよびB)pHR’SIN.cPPT−SEWによる形質導入(MOI 100)の5週間後、細胞をDil−Ac−LDLと共に1時間インキュベートし、蛍光顕微鏡法により(A)EGFP発現および(B)Dil−Ac−LDL取り込みを測定した。(CおよびD)管腔形成アッセイのために、CBECをトリプシン/EDTA処理により剥離し、マトリゲル(登録商標)基底膜マトリックス上で培養した。該細胞を、8〜10時間37℃でインキュベートし、顕微鏡法で血管の形成を測定した(C:相の比較; D:EGFP発現)。
3つのドナープール由来のCBECを、MOI 10でBDD FVIIIをコードするコンストラクトcPPT−C(FVIIIΔB)IGWSにより形質導入した。形質導入後様々な時点でのFVIII:Cの分泌を定量化するために、5×104個の細胞を1mlのEGM−2中で培養し、37℃でインキュベートした。48時間後、上清を採り、短時間の遠心分離により細胞残屑を除き、分析するまで−80℃で貯蔵した。どのアッセイにも、pHR’SIN.cPPT−SEWにより形質導入した細胞由来の上清、および形質導入していない細胞由来の上清が含んでいた。FVIII:Cは、これらの対照のいずれでも検出できなかった(検出限界:0.01 IU/ml)。
形質導入していないCBEC由来、およびcPPT−C(FVIIIΔB)IGWSにより形質導入したCBEC由来の無血清培養上清を、限外ろ過により200〜400倍に濃縮した。試料を、SDS−PAGEにより分離し、免疫ブロットを行った。レーン1および2は、対照として市販のFVIII濃縮物を示す。レーン3および4はCBEC由来の濃縮上清を示す。様々な強度のシグナルを検出するために、オートラジオグラフィー・フィルムを異なる時間で曝露した(1a〜4a:1分間;1b〜4b:20分間)。レーン1:Oct、Octanate(登録商標)(血漿由来完全長FVIII);レーン2:ReF、ReFacto(登録商標)(組換えBDD FVIII);レーン3:cPPT−C(FVIIIΔB)IGWSにより形質導入したCBEC由来上清(BDDFVIII);レーン4:形質導入していないCBEC由来上清。
図7はヒト第VIII因子のアミノ酸配列を示す(20位〜2351位のアミノ酸は成熟第VIII因子に対応する)。
単核細胞を、フィコールによる比重分離(d=1.077g/ml)により健康なドナーの臍帯血から単離し、免疫蛍光染色法および蛍光活性化細胞選別法(FACS)分析により測定すると、該単核細胞はCD34陽性細胞を2%未満含んでいた。2mM EDTA(シグマ社(Sigma)、タウフキルヒェン、ドイツ)および0.5%ヒト血清アルブミン(HSA;ドイツ赤十字血液センター(DRK−Blutspendedienst) ニーダーザクセン、シュプリンゲ、ドイツ)を含むダルベッコ(Dulbecco)PBS(リン酸緩衝液;バイオウィッタカー社(BioWhittaker)、ベルビエ、ベルギー)で、該細胞を2回洗浄し、直接CD34陽性前駆細胞単離キット(Direct CD34 Progenitor Cell Isolation Kit)(ミルテニーバイオテク社(Myltenyi Biotec)、ベルギッシュグラッドバッハ、ドイツ)を用いて製造者の指示書に従い、CD34陽性細胞を磁気活性化細胞選別法(MACS)により富化した。CD34陽性細胞の純度は、80%から95%の範囲内であった。
CD34陽性細胞を、次から成る内皮細胞分化培地に再懸濁させた:80%基礎Iscove培地(バイオクロム社(Biochrome)、ベルリン、ドイツ)、10%ウマ血清(パンバイオテック社(PAN Biotech)、アイデンバッハ、ドイツ;選択されたロット)、10%加熱不活化ウシ胎仔血清(FCS;バイオクロム社;選択されたロット)、L−アラニル−L−グルタミン(最終濃度 2mM;バイオウィッタカー社)およびペニシリン−ストレプトマイシン(最終濃度 それぞれ100U/mlおよび100μg/ml;バイオウィッタカー社)。培地を濾過し(孔径0.22μm)、その後、組換えヒト(rh)VEGF(血管内皮成長因子;50ng/ml)、rh塩基性FGF(線維芽細胞成長因子−2;20ng/ml)、rhSCF(幹細胞因子;50ng/ml;以上全てアール・アンド・ディー・システムズ社(R&D Systems)、ウィーズバーデン、ドイツ)、およびrhSCGF−β(幹細胞成長因子−β;20ng/ml;ペプロテック社(PeproTech)、フランクフルト・アム・マイン、ドイツ)を添加した。成長因子を含むこの培地は、内皮細胞分化培地(EDM)と称する。
1%FCS(バイオクロム社)および0.1%アジ化ナトリウム(シグマ社)を含むPBSで洗浄することにより、およそ105個の細胞をフローサイトメトリー用に調製した。造血細胞表面マーカーおよび内皮細胞表面マーカーの発現を特徴づけるために、次のモノクローナル抗体(mAb)を用いた:抗CD14−PE抗体、抗CD34−PE抗体、抗CD45−FITC抗体、抗HLA−DR−FITC抗体(これら全てビー・ディー・バイオサイエンス社(BD Bioscience)、ハイデルベルク、ドイツ)、抗CD133−PE抗体(ミルテニーバイオテク社)、抗CD146抗体、抗CD146−FITC抗体、抗VE−カドヘリン抗体(これら全てケミコン社(Chemicon)、テメキュラ、カリフォルニア州)、抗CD34−PC5抗体(イミューノテック社(Immunotech)、マルセイユ、フランス)、抗CD31−FITC抗体および抗KDR抗体(両者ともシグマ社)。細胞は、それぞれのmAbに対して20分間、20℃でインキュベーション(抗CD31−FITC抗体、抗KDR抗体の場合)、あるいは4℃で(他の全ての抗体の場合)インキュベーションした。第2ステップとして、ウサギ抗マウス免疫グロブリン(ダコ社(Dako)、グロストラップ、デンマーク)のRPEでコンジュゲートしたF(ab’)2断片をインキュベーションすることにより、コンジュゲートしていない1次モノクローナル抗体が検出された。細胞のサンプルは、アイソタイプが適合した対照抗体(ビー・ディー・バイオサイエンス社とイミューノテック社から購入)でも染色した。FACS分析は、FACScanフロー・サイトメーター(ビー・ディー・バイオサイエンス社)およびセル・クエスト(Cell Quest)ソフトウェア(ビー・ディー・バイオサイエンス社)を用いて行った。それぞれの分析は、少なくとも10,000回行った。死細胞は、その前方散乱特性および側方散乱特性に基づいて、除去した。
アセチル化低密度リポタンパク質(Ac−LDL)の取り込みによる内皮細胞の蛍光標識は、EGM−2中60分間37℃で、2μg/mlのDil−Ac−LDL(ハーバー・バイオ・プロダクツ社(Harbor Bio−Products)、ノーウッド、マサチューセッツ州)により該細胞をインキュベートすることで行った。その後、培地をEGM−2と交換し、該細胞のDil−Ac−LDL取り込みを、蛍光顕微鏡法(ニコン社、Eclipse TE300)およびフローサイトメトリーにより分析した。
2つのHIV−1型由来自己不活化レンチウイルス遺伝子導入コンストラクトを、この実施例で用いた:pHR’SIN.cPPT−SEWベクター(Demaison et al.,2002)は、脾限局巣形成ウイルス(U3−LTR)のエンハンサー領域、強化緑色蛍光タンパク質(EGFP)のcDNA、およびウッドチャック肝炎ウイルス転写後調節因子(WPRE)から成る遺伝子発現カセットを包含する。
レンチウイルス上清の産生
レンチウイルス粒子を生成するために、gag遺伝子、pol遺伝子、rev遺伝子、およびtat遺伝子をコードするパッケージングコンストラクトpCMVΔR8.91(Zufferey et al.,1997)、および水疱性口内炎ウイルス糖タンパク質(VSV−G)をコードするシュードタイピングコンストラクトpMD2.VSVG(Follenzi and Naldini,2002)のコトランスフェクションを3回行うことにより、遺伝子導入ベクターのDNAが一時的に、293T細胞(Tonnet al.,(2002)に記述のように培養した)に導入された。
内皮細胞を、EGM−2中で1ウェルあたり1×104個、ゼラチン被覆6ウェルプレート上で培養し、37℃で48時間インキュベートした。形質導入のために、4μg/mlの硫酸プロタミン(シグマ社)および50μMのdNTPs(ニュー・イングランド・バイオラブズ(New England Biolabs)社、フランクフルト・アム・マイン、ドイツ)の存在下、EGM−2中で濃縮されていないウイルス上清または濃縮されたウイルス上清と、培地を交換した。スピノキュレーション(spinoculation)(1250g、90分、32℃)後、該細胞を37℃でさらに16時間インキュベートした。それから、ウイルス上清を除去し、該細胞に新鮮なEGM−2を与えた。感染多重度(MOI)は、標的細胞に対する293T−TU/mlの割合として計算した。EGFP発現は、様々な時点でFACS分析および蛍光顕微鏡法により分析した。
分泌したFVIIIの量を評価するために、形質導入した内皮細胞と形質導入していない対照細胞を、ゼラチンコートした12ウェルプレートに、総体積1mlのFGM−2中に5×104個の細胞で播種した。48時間後、遠心分離(700g、3分、4℃)により上清から細胞残屑を除去し、分析まで−80℃で複数のアリコート中に貯蔵した。FVIII抗原(FVIII:Ag)およびFVIII活性(FVIII:C)の測定は、それぞれ市販のImmunozym FVIII:Ag ELISAおよびImmunochrom FVIII:C発色検出アッセイ(イミューノ社(Immuno)、ハイデルベルク、ドイツ)により製造者の指示書に従い行った。FVIII濃度は国際単位(IU)/mlで与えられ、150ng/mlが1.0IU/mlに対応する。両アッセイにおけるFVIII標準品は、WHO血漿標準に対して製造者が計算したものである。
コンフルエントな内皮細胞層をPBSで洗浄し、15〜24時間FCS無しのEGM−2で培養した。培養上清を濾過し(0.22μm)、細胞残屑を除去し、ビバスピン(Vivaspin)20濃縮器(ザルトリウス社(Sartorius)、ゲッチンゲン、ドイツ)での超遠心分離(排除分子量(MWCO)30kDa)により、400倍にまで濃縮した。濃縮物をラエムリ(Laemmli)緩衝液(RotiLoad 1;ロート社(Roth)、カールスルーエ、ドイツ)中で5分間煮沸し、10%ポリアクリルアミドゲル上でSDS−PAGEにより分離し、ポリビニリデンジフルオライド膜(ロート社)に移した。その膜を5%粉末スキムミルク中20℃で3時間ブロックし、ポリクローナルヒツジ抗ヒト第VIII因子:C抗体(エンザイム・リサーチ・ラボラトリーズ社(Enzyme Research Laboratories)、スワンシー、イギリス)とともに4℃で15〜20時間インキュベートした。十分に洗浄した後、ペルオキシダーゼでコンジュゲートしたロバ抗ヒツジIgG二次抗体(シグマ社)とともに、ブロットを室温で40分間インキュベートした。さらに洗浄した後、ECL法(enhanced chemiluminescence:高感度化学発光法)(ピアス(Pierce)社、ボン、ドイツ)で、タンパク質を視覚化した。陽性対照として、組換えB領域欠損FVIIIであるReFacto(登録商標)(ファルマシア&アップジョン社、マルチンスリード、ドイツ)および血漿由来ヒトFVIII(Octanate(登録商標);オクタファーマ(Octapharma)、フランクフルト・アム・マイン、ドイツのLothar Biesert氏が快く提供)を用いた。
予め冷蔵した24ウェルプレートを、1ウェルあたり500μlのマトリゲル(登録商標)基底膜マトリックス(ビー・ディー・バイオサイエンス社)で被覆し、37℃で1時間インキュベートした。内皮細胞はトリプシン/EDTA処理により採取し、EGM−2中に再懸濁し、ゲル化したマトリックス上に400μl中6×104〜1×105個、該細胞を播種した。培養物を37℃でインキュベートした。8〜10時間後、位相差顕微鏡法および蛍光顕微鏡法(ニコン社 Eclipse TE300)により管腔形成を確認した。
データは平均±標準偏差で示される。1対のスチューデントt検定を用いて、異なる感染多重度で形質導入した細胞の形質導入効率およびFVIII分泌レベルを比較した。統計的分析は、GraphPad Prism3.0ソフトウェアを用いて行った。
臍帯血由来内皮細胞(CBEC)の分化および培養
臍帯血単核細胞の単離後、MACS免疫磁気ビーズを用いて、CD34陽性細胞分画を純度80%〜95%で得た。rhVEGF、rh塩基性FGF、rhSCFおよびrhSCGF−β(「器具および方法」参照)を含むEDM中で、単一のドナーからの細胞あるいは数人のドナーからの細胞プールを約3週間培養した。内皮形態を有する増殖している接着細胞が検出できた場合、それらの細胞はコンフルエントに達する前に継体培養される。105〜107個のCD34陽性細胞の植え付けで、8週間の総培養時間で接着細胞が109倍増殖した。図1に、ドナープールからの細胞(「WT」と表す)を用いた2つの代表実験の累積増殖曲線を示す。この実施例では、約30〜35時間の倍化時間で細胞は増殖した。細胞が増殖を止め、培養物が老化する108倍を超えるまで増殖させた。
該接着細胞は、紡錘状扁平細胞の単層として増殖する(図2)。該細胞は、蛍光顕微鏡法およびフローサイトメトリーで検出されるDil−Ac−LDLを取り込むことができ、マトリゲルを用いたアッセイで管腔の形成を示した(以下参照)。該細胞は、様々な内皮細胞表面マーカーおよび造血細胞表面マーカーの発現に関して、フローサイトメトリーによりさらに特徴づけられ、内皮細胞について典型的な、VE−カドヘリン(CD144)、CD146、およびCD31に対して一様に陽性であることが判明した。該細胞は、造血細胞表面マーカーであるCD45、CD14、CD133、およびHLA−DRに対しては一様に陰性であった(図2)。ECの5バッチをFACS分析することにより、このプロトコルで得られた細胞がCD34およびKDR/VEGF−R2の発現に関して不均一であることが示された(図2)。CD34の発現は細胞の5〜45%のサブセットで検出され、一方KDRはCBECの5%未満で弱く発現した。上記細胞表面マーカーの発現は、細胞培養の間、不変であるように見えた。
CBECがレンチウイルスベクターにより効率的に形質導入されることができるかを調べるために、3〜8の継体数でかつ10および100の感染多重度(MOI)で、EGFPをコードするコンストラクトpHR’SIN.cPPT−SEW、およびヒトB領域欠損第VIII因子とEGFPをコードするコンストラクトcPPT−C(FVIIIΔB)IGWSを用いた最初の一連の実験において、4つの異なるドナープール由来のCBECが形質導入された。形質導入効率は、形質導入後8日以降の少なくとも2つの異なる時点で、EGFP陽性細胞の検出により測定した。
形質導入間/後の(特により高いMOIにおける)細胞死、および図1に示した形質導入していない対照細胞の増殖速度に細胞が戻る前の誘導期から明らかなように、形質導入したECへのいくつかのベクター媒介性毒性を観察した。したがって、このことが形質導入細胞と対照細胞間の表現型の変化に付随して起こるのかどうかを検討したいと考えた。形質導入して数週間後に、フローサイトメトリーによるCD146、CD34、KDR、およびCD133発現に対して、あるいはDil−Ac−LDLを取り込む能力に対して、細胞を分析した。上記細胞表面マーカーの発現は、Dil−Ac−LDLの取り込みと同様、形質導入していない細胞と比較して変化が無かった(図4AおよびB)。また、CBECは、マトリゲルを用いたアッセイ(図4CおよびD)において、成熟した機能性ECの特徴である管腔形成能力を保持した。
ヒトB領域欠損FVIIIを発現するCBECの能力を、7つの独立した実験で調べた。細胞は、ヒトB領域欠損FVIIIおよびEGFPをコードするレンチウイルスコンストラクトcPPT−C(FVIIIΔB)IGWSにより形質導入され、老化するまで上記のように培養された。12ウェルプレートに、1mlのEGM−2中5×104個の細胞を播種し、48時間後に細胞培養上清を回収することにより、様々な時点でFVIIIの分泌を定量した。検出抗体としての軽鎖に対するモノクローナル抗体による発色検出アッセイおよびELISAを用いた分析を行うまで、−80℃でアリコートを貯蔵した。細胞は、増殖を止めるまで形質導入を維持した。
造血細胞株、HUVEC、およびCBEC(臍帯血由来内皮細胞)を、同じFVIII発現カセットを含むレンチウイルスベクターにより形質導入した。比活性は、これら細胞の上清におけるFVIII:AgとFVIII:Cの比率として計算され、少なくとも3つの実験の平均値として与えられる。
ECの分泌が正しく進み、それにより活性な凝血源FVIIIが産生しているという仮定を確認するために、排除分子量30kDaのビバスピン濃縮器を用いた限外濾過によって、細胞培養上清からFVIIIを富化した。EGM−2の血清含量が有効な濃度にならなかったので、ECを血清なしで終夜培養した。試料は400倍にまで濃縮され、ELISA、FVIIIウエスタンブロット、および発色検出アッセイにより分析した。

Claims (23)
- 工程:
a)ヒト臍帯血由来内皮前駆細胞および/または骨髄から得られる内皮前駆細胞を、内皮前駆細胞が成熟内皮細胞へ分化するのを促進する少なくとも1つの成長因子とin vitroで接触させること;および
b)タンパク質をコードするDNAにより成熟内皮細胞に形質導入する、または、工程a)に先立って、タンパク質をコードするDNAにより内皮前駆細胞に形質導入すること、を含むことを特徴とする、該タンパク質を発現するヒト内皮細胞の産生方法。 - 該ヒト臍帯血由来内皮前駆細胞が、CD34、AC133、CD146、およびFGF1−Rから成る群より選択される細胞表面マーカーを少なくとも1つ発現することを特徴とする、請求項1に記載の方法。
- 工程a)およびb)に先立って、臍帯血からCD34陽性細胞、AC133陽性細胞、CD146陽性細胞、および/またはFGF1−R陽性細胞を富化させることをさらに含む、請求項1または2に記載の方法。
- レトロウイルスベクター、好ましくはレンチウイルスベクターが該細胞に形質導入するために用いられることを特徴とする、請求項1から3のいずれかに記載の方法。
- 該タンパク質が血液凝固因子であることを特徴とする、請求項1から4のいずれかに記載の方法。
- 該血液凝固因子がヒト第VIII因子であることを特徴とする、請求項5に記載の方法。
- 請求項1から6のいずれかに記載の方法によって得ることができるヒト内皮細胞群。
- 少なくとも細胞の10%が、タンパク質をコードする組換えDNAを含むことを特徴とする、臍帯血由来のヒト内皮細胞群。
- 該タンパク質が、ヒト血液凝固第VIII因子または第IX因子であることを特徴とする、請求項7または8に記載のヒト内皮細胞群。
- タンパク質をコードする組換えDNAがヒト血液凝固第VIII因子の突然変異タンパク質をコードすることを特徴とする、請求項7から9のいずれかに記載のヒト内皮細胞群。
- ヒト第VIII因子の該突然変異タンパク質が、野生型第VIII因子のB領域を少なくとも部分的に欠損していることを特徴とする、請求項10に記載のヒト内皮細胞群。
- タンパク質をコードする組換えDNAが修飾第VIII因子の相補的DNAであり、
(i)野生型第VIII因子の相補的DNAのうち、少なくともB領域部分が欠損されている、
(ii)少なくとも1つのイントロンが、第VIII因子の相補的DNAの少なくとも1つの位置に挿入されている、および/または
(iii)ヒト第VIII因子の野生型相補的DNA配列の少なくとも1つのヌクレオチドが置換されている、
ことを特徴とする、請求項7から11のいずれかに記載のヒト内皮細胞群。 - 該細胞の少なくとも75%が、CD144、CD31、CD146、およびLDL受容体から成る群より選択されるマーカーの少なくとも1つを発現することを特徴とする、請求項7から12のいずれかに記載のヒト内皮細胞群。
- 該細胞の少なくとも75%がフォン・ビルブラント因子(VWF)を内因的に発現することを特徴とする、請求項7から13のいずれかに記載のヒト内皮細胞群。
- 請求項7から14のいずれかに記載のヒト内皮細胞群から生成した不死化細胞株。
- a)適当な条件下、請求項7から14のいずれかに記載のヒト内皮細胞群、または請求項15に記載の細胞株を培養する、およびb)該細胞培養培地から、タンパク質、好ましくは血液凝固因子を単離する、工程からなることを特徴とする該タンパク質、好ましくは該血液凝固因子を産生する方法。
- 該タンパク質が血液凝固第VIII因子であり、かつ該単離工程がフォン・ビルブラント因子の存在下での血液凝固第VIII因子の精製を含むことを特徴とする、請求項16に記載の方法。
- 該タンパク質、好ましくは該血液凝固因子をウイルス不活化処理することを特徴とする、請求項16または請求項17に記載の方法。
- 血友病Aまたは血友病Bの治療用医薬の製造のための、請求項7から14のいずれかに記載のヒト内皮細胞群の使用。
- ヒト血液凝固第VIII因子を分泌する細胞を、血友病Aを患う個体に投与することを特徴とする、請求項19に記載の使用。
- 循環器系疾患の治療用医薬の製造のための、請求項7から14のいずれかに記載のヒト内皮細胞群の使用。
- 該疾患が心不全、および急性もしくは慢性心疾患から選ばれることを特徴とする、請求項21に記載の使用。
- 請求項7から14のいずれかに記載のヒト内皮細胞群である医薬組成物。
