Abstract
【解決手段】少なくとも1つのポリ(エチレングリコール)分子が、生物学的に活性なヒト成長ホルモン(hGH)ポリペプチドまたはそのアゴニスト変異体の少なくとも1つのアミノ酸残基に共有結合したものを含むコンジュゲート。該コンジュゲートは、非修飾hGHの持続的hGH活性よりもはるかに長い持続的活性を有し、投与量及び回数の軽減を可能にする。

本発明は化学的に修飾されたhGHコンジュゲートを提供し、それらは減少した不均一性、減少したクリアランス速度、増大した血漿中滞留時間、改善された溶解度、増大した安定性、低下した抗原性、またはそれらの組み合わせを有する。
本発明は、化学的に修飾されたhGHおよびそのアゴニスト変異体に関し、それらは減少したクリアランス速度、増大した血漿中滞留時間、増大した安定性、改善された溶解度、および低下した抗原性から選択されるがそれらに限定されない、少なくとも1つの改善された化学的または物理的性質を有する。したがって、より詳細に以下に記載のように、本発明は化学的に修飾しているhGHおよびそのアゴニスト変異体、ならびに多様なポリ(エチレングリコール)部分を使用した具体的な修飾に関する多数の側面を有する。
また、本発明は化学的に修飾されたhGHおよびそのアゴニスト変異体を含む組成物に関する。
米国特許第4,658,021号および米国特許第5,633,352号に記載のhGHおよびそのアゴニスト変異体は組換え蛋白質ファミリーのメンバーである。それらの組換え産物および使用の方法は、米国特許第4,342,832号、第4,601,980号;米国特許第4,898,830号;米国特許第5,424,199号;および米国特許第5,795,745号に記載される。
ペグ化と表現される結合反応は、歴史的にモル過剰ポリマーで、ポリマーが蛋白質のどこに結合するかにかかわりなく、溶液中で行われた。しかし、そのような一般的な技術は十分な生理活性を保持しながら非抗原性ポリマーに生理活性蛋白質を結合するためには概して不適切であることが証明されている。hGHまたはそのアゴニスト変異体の生理活性を維持するための一方法は、ポリマーカップリング過程において、受容体結合部位に関連したそれらのhGHまたはそのアゴニスト変異体の反応基の結合を実質的に避けることである。本発明の別の側面は、高レベルの保持活性を維持するhGHまたはそのアゴニスト変異体にポリ(エチレングリコール)を結合させる方法を提供することである。

実施例1に記載の手順を使用して、メトキシ直鎖30,000MW PEG−プロピオンアルデヒド試薬(Shearwater Corp)をhGHのN−末端に結合した。
実施例1に記載の手順を使用して、メトキシ直鎖5,000MW PEG−プロピオンアルデヒド試薬(Fluca)をhGHのN−末端に結合した。
アミンにつきPEG対hGHのモル比0.1〜0.5を使用し、実施例1に記載の手順を使用して、メトキシ分枝20,000MW PEG−アルデヒド(PEG2−ALD)試薬(Shearwater Corp)をhGHのN−末端に結合した。
実施例6に記載の手順を使用して、直鎖20,000MW SPA―PEG PEG試薬(Shearwater Corp.)をhGHのN−末端に結合した。

2以上の結合したPEGを有する(多重ペグ化)修飾PEGも実施例1および4から得られ、アニオン交換クロマトグラフィーを使用してモノ‐ペグ化種から分離した。また、2以上の結合したPEGを有する(多重ペグ化)修飾PEGはカチオン交換クロマトグラフィーを使用してモノ‐ペグ化種から分離する。さらに、2以上の結合したPEGを有する(多重ペグ化)修飾PEGが実施例2、3、5〜13で得られ、実施例1および4と同様の様式で精製する。
アニオン交換クロマトグラフィー
PEG hGH種は一回のアニオン交換クロマトグラフィー工程を使用して、反応混合物から>95%(SEC分析)に精製した。モノ‐ペグ化hGHは、非修飾hGHおよび多重ペグ化hGH種からアニオン交換クロマトグラフィーを使用して精製した。先に記載の代表的20K アルデヒドhGH反応混合物(5〜100mg蛋白質)は25mM HEPES、pH7.3(バッファーA)で平衡化したQ−Sepharose Hitrapカラム(1または5mL)(Amersham Pharmacia Biotech,Piscataway,NJ)またはQ−Sepharose fast flowカラム(26/20、70mL ベッド容積)(Amersham Pharmacia Biotech,Piscataway,NJ)上で精製した。反応混合物はバッファーAで5〜10倍に希釈し、流速2.5mL/分でカラムに流した。カラムは、カラム容積の8倍のバッファーAで洗浄した。次にカラム容積の80〜100倍のバッファーAおよび0〜100mMの直線NaCl勾配で各種hGH種をカラムから溶出させた。溶離液は280nm(A280)における吸光度でモニターし、5mL画分を集めた。画分は、ペグ化の程度、たとえば(実施例15で評価したように)、モノ、ジ、トリなどに応じてプールした。プールはその後Centriprep YM10濃縮器(Amicon,Technology Corporation,Northborough,MA)で0.5〜5mg/mLに濃縮した。この手順により精製したモノ20K PEG−アルデヒド hGHの総収率は25〜30%であった。
カチオン交換クロマトグラフィーは、10mM 酢酸ナトリウム pH4.0(バッファーB)で平衡化したSP Sepharose高速カラム(Pharmacia XK 26/20,70mL ベッド容積)上で行う。反応混合物はバッファーBで10倍に希釈し、流速5mL/分でカラムに流す。次に、カラムをカラム容積の5倍のバッファーB、続いてカラム容積の5倍の12%バッファーC(10mM 酢酸 pH4.5、1M NaCl)で洗浄する。次にカラム容積の20倍の12%〜27%バッファーCの直線勾配でPEG−hGH種を溶出する。溶離液は280nmでモニターし、10mL画分を集める。画分はペグ化の程度(モノ、ジ、トリなど)に応じてプールし、10mM 酢酸 pH4.5バッファーに交換し、Amicon YM10膜のついた撹拌セル中で1〜5mg/mLに濃縮する。プールの蛋白質濃度は吸光係数0.78を使用してA280nmで測定する。この手順から得られたモノペグ化hGHの総収率は10〜50%である。
精製したペグ化hGHプールは、還元および非還元SDS−PAGE、非変性および変性サイズ排除クロマトグラフィー、分析アニオン交換クロマトグラフィー、N−末端シークエンシング、疎水性相互作用クロマトグラフィー、および逆相HPLCにより性状を決定した。
非変性SEC−HPLC
非変性SEC−HPLCを使用して、各種結合化学、サイズ、リンカー、および形状のメトキシ‐PEGとhGHとの反応物、アニオン交換精製物プールおよび最終精製産物を評価した。分析非変性SEC−HPLCは、Tosohaas G4000PWXLカラム、7.8mmX30cm、(Tosohaas Amersham Bioscience,Piscataway,NL)またはSuperdex 200(Amersham Bioscience,Piscataway,NL)を使用して、20mM リン酸 pH7.2、150mM NaCl中、流速0.5mL/分で行った。ペグ化は蛋白質の流体力学的容積を非常に増し、より早い保持時間の方にシフトする。新しい種は非修飾hGHと共にPEG アルデヒド hGH反応混合物中に観察された。これらのペグ化および非ペグ化種はQ−Sepharoseクロマトグラフィー上で分離し、得られた精製モノPEG−アルデヒド hGH種はその後非変性SEC上の単一ピークとして溶出することが示された(>95%純度)。Q−Sepharoseクロマトグラフィー手順はモノ‐ペグ化hGHから遊離PEG、hGH、および多重ペグ化hGH種を効率的に除去した。非変性SEC−HPLCは、各種ペグ化hGHの効果的なサイズがそれらのそれぞれの理論的分子量よりかなり大きいことを示した(表1)。
変性SEC−HPLCを使用して、各種メトキシ‐PEGとhGHとの反応物、アニオン交換精製物、および最終精製産物を評価した。分析変性SEC−HPLCはTosohaas 3000SWXLカラム、7.8mmx30cm、(Tosohaas Pharmacia Biotech,Piscataway,NL)を使用して100mM リン酸pH6.8、0.1% SDS中、流速0.8mL/分で行った。ペグ化は蛋白質の流体力学的容積を非常に増し、より早い保持時間の方にシフトする。新しい種は非修飾hGHと共に20K PEG アルデヒド hGH反応混合物中に観察された。これらのペグ化および非ペグ化種はQ−Sepharoseクロマトグラフィー上で分離し、得られた精製モノ20K PEG−アルデヒド hGHはその後変性SEC上の単一ピークとして溶出することが示された(>95%純度)。Q−Sepharoseクロマトグラフィー手順はモノ‐ペグ化hGHから遊離PEG、hGH、および多重ペグ化hGH種を効率的に除去した。
SDS−PAGEを使用して、各種PEG試薬とhGHとの反応物、および最終精製産物を評価した。この技術の例としてはモノ20K 直鎖ならびに分枝20Kおよび40K PEG アルデヒド、および4X6 5K SPA PEGについて示す(図1&2)。SDS−PAGEは還元および非還元条件下、厚さ1mmの10〜20%Trisトリシンゲル上で行い、Novex Colloidal Coomassie(登録商標)G−250染色キット(Invitrogen,Carlsbad,CA)を使用して染色した。精製したモノPEG−アルデヒド hGH種はSDS−PAGE上で1つの主要バンドとして移動する。バンドはPVDF膜上にブロットし、その後N−末端配列決定を行った。
分析アニオン交換HPLCを使用して、各種mPEGとhGHとの反応物、アニオン交換精製画分および最終精製産物を評価した。分析アニオン交換HPLCは、Tosohaas Q5PWまたはDEAE−PWアニオン交換カラム、7.5mmx75mm、(Tosohaas Pharmacia Biotech,Piscataway,NL)を使用して、50mM Tris ph8.6中、流速1mL/分で行った。試料は5〜200mM NaClの直線勾配で溶出した。
PEG−GH反応混合物および精製したペグ化産物はRP HPLCを使用して分析し、hGH種、モノおよび多重ペグ化hGH種を評価し、そして酸化hGH型、および異なる部位(たとえばN−末端対リシンε‐アミノ基)に結合した単一PEGを有するPEG hGHイソ型をモニターした。RP−HPLCはZorbax SB−CN 150または250mmx4.6mm(3.5mmまたは5mm)逆相HPLCカラムを使用して行った。実験は周囲温度で、試料につき標準的な10mg蛋白質負荷で行った。バッファーAは水中0.1%トリフルオロ酢酸であり;バッファーBはアセトニトリル中0.1%トリフルオロ酢酸である。1分につきBに1/2%増加を生じるグラジエントは以下のとおりである:
自動Edman分解化学を使用して、NH2−末端蛋白質配列を決定した。分解には、Applied Biosystems Model 494 Procise sequencer(Perkin Elmer,Wellesley,MA)を使用した。それぞれのPTH−AA誘導体は、Perkin Elmer/Brownlee 2.1mm i.d.PTH−C18カラムが付いたApplied Biosystems Model 140C PTH analyzerを使用して、オンライン様式でRP−HPLC分析により同定した。PVDF膜に転写した20K 直鎖ならびに20および40K分枝PEG−ALD hGH蛋白質バンド、または精製した20K直鎖ならびに分枝20および40K PEG−ALD hGHの溶液を配列決定した。主要シグナル(約88%収率)を生じる精製した20K直鎖PEG−hGHが観察され、それはN−末端アミノ酸がないことを除いてhGHに予想された配列を有した。この結果はアルデヒド化学により蛋白質のN−末端がペグ化されたものに予想されたものと同じである。初めのサイクルの残留物は結合したPEG部分のために回収不能である。主要でないシグナル(約12% 収率)は正しいN−末端配列を有した。RP−HPLCから集めたピークが100%ペグ化されていることを考慮すると、これらのデータはPEG修飾の約88%はN−末端であり、残りは明らかにいくつかの可能性のあるリシン残基の1つに結合していることを示唆する。
脳下垂体切除ラットにおける有効性試験
Harlan Labsで脳下垂体切除した雌Sprague Dawleyラットは7〜10日間、成長速度をプレスクリーニングした。その後成長試験を11日間行った。ラットは6〜8の群に分割した。1群は毎日か、または0日目と6日目にベヒクルを皮下投与したラットからなっていた。2群には、GH(0.3mg/kg/投与)を毎日皮下投与した。3群には0日目と6日目にGH(1.8mg/kg/投与)を皮下投与した。4群には0日目と6日目にPEG−GH(1.8mg/kg/投与)を皮下投与した。脳下垂体切除ラットは試験中、少なくとも1日おきに体重を測定することにより体重増加をモニターした。週に1度投与した20K PEG−ALD hGH、20Kおよび40K 分枝PEG−ALD hGH、および4〜6x5PEG−SPA hGHに対する体重増加(平均+/−SEM)はhGHの毎日投与に対するものと同様であった(図3&4)。表3は、各種ペグ化hGH分子の毎日投与に比べたhGHの週1回投与に対する11日目の総体重増加(平均±SEM)を比較したものを要約する。
実験は上記の体重増加試験と同じように行ったが、血液試料は0、1、2、3、4、5日目および9日目の動物の致死時に採取した。IGF−1レベルはELISAにより測定した。図7は、hGHの毎日投与またはhGHの単回投与またはペグ化hGHの0、6日目投与のいずれかの後の血清中IGF−1レベル(平均+/−SEM)の上昇を比較する。
薬力学的試験は正常なSprague−Dawley雄ラット、マウス、およびカニクイザルで行った。1群につき6匹のラットおよび60匹までのマウスを使用し、ラットおよびマウスにおけるGHまたはPEG−GHの1.8mg/kgの単回ボーラス投与、または1.0mg/kgの単回静脈内投与として注射を行った。カニクイザルでは、1群につき2〜4頭のサルを使用し、単回皮下ボーラス投与および静脈内投与の両方に対して0.18mg/kgのGHまたはPEG−GH投与量を使用した。血液試料は、適切なPKパラメータの評価のために1〜5日目まで適宜採取した(表4)。(t1/2)=末期半減期(terminal halftime)、(Cl)=クリアランス、(Tmax)=最大濃度までの時間、Vss=定常状態における(みかけの)容積分布、および(Cmax)=最大濃度GHおよびPEG−GH血中レベルは、イムノアッセイを使用してそれぞれの採取毎にモニターした。
ラット、マウス、およびカニクイザル血漿中のhGHおよびペグ化hGH蛋白質濃度レベルは、hGH AutoDELFIA kit蛍光イムノアッセイ(PerkinElmer Life Sciences)を使用し、標準曲線を作製するために適切なPEG hGHを使用して測定した。

を有し、Rがヒト成長ホルモンポリペプチドであり、ポリ(エチレングリコール)が30kDaの分子量を有する、請求項1に記載のコンジュゲート。 - 前記ポリ(エチレングリコール)の少なくとも90%がアミノ末端α−アミノ基に結合している、請求項1〜3のいずれかに記載のコンジュゲート。
- 前記hGHポリペプチドがSEQ ID NO:1のアミノ酸配列を含む、請求項1〜4のいずれかに記載のコンジュゲート。
- 請求項1〜5のいずれかに記載のコンジュゲートおよび少なくとも1つの薬剤的に受容できるキャリアを含む組成物。
- 請求項1〜5のいずれかに記載のhGHコンジュゲートを含有する、成長または発育障害の治療のための医薬組成物。
- 前記成長または発育障害が成長ホルモン欠乏症(GHD)である、請求項7に記載の医薬組成物。
- 前記成長または発育障害がターナー(Turner)症候群である、請求項7に記載の医薬組成物。
- 前記成長または発育障害が慢性腎不全症である、請求項7に記載の医薬組成物。
- 前記成長または発育障害が胎内発育遅延(SGA)である、請求項7に記載の医薬組成物。
- 前記成長または発育障害が成人成長ホルモン欠乏症である、請求項7に記載の医薬組成物。
- 以下の工程を含む、高レベルの生物学的活性を持続する請求項1〜5のいずれかに記載のコンジュゲートの作製方法:
a)ヒト成長ホルモンポリペプチドまたはそのアゴニスト変異体の不活性化を避けるために、ヒト成長ホルモンポリペプチドまたはそのアゴニスト変異体と活性化ポリ(エチレングリコール)との間の結合反応を溶液中で温和な条件下で実施する工程;および
b)コンジュゲートを未反応種から分離する工程。 - 前記温和な条件が、反応溶液のpHを3〜10、且つ反応温度を0℃〜37℃に維持することを含む、請求項13に記載の方法。
