本発明は、概して遺伝子療法、より詳細にはウイルス媒介性およびその他の形態の遺伝子療法に関する。より具体的には、本発明は血管新生を促進する際に有用なアデノウイルス媒介性遺伝子送達に関する。

血管新生とは既存血管からの発芽プロセスによる新規毛細血管の形成であり、発達中並びに数多くの生理的および病理的プロセスにおいて発生する。血管新生は、内皮細胞の増殖、細胞外マトリックスの分解、血管の分岐、および、それに続く、細胞接着事象を含む生理学的に複雑なプロセスである。成体では、血管新生は厳密に制御されており、正常な状況下では女性生殖器系に限定されている。しかし、血管新生は、組織損傷に応答してスイッチが入れられる可能性がある。固型腫瘍もまた周囲組織において血管新生を誘発し得、これによって、腫瘍増殖を維持し、転移の形成を容易にする(フォークマン,ジェイ(Folkman J.),Nature Med. 1:27−31(1995))。複雑な血管新生プロセスの基礎にある分子機序は理解されていると言うには程遠い。リンパ系においても類似のプロセスが発生するがこのプロセスについての研究ははるかに進んでおらず、これはしばしば、リンパ管新生と称されている。

組織損傷に応答して、または女性生殖器内で発生する血管新生を除いて、健常成体内で内皮細胞の代謝回転が発生する可能性は極めて少ない。内皮静止期が維持されるのは外因性の負の調節因子が存在するためであると考えられるが、それは明らかに血管新生が発生しない成体組織内では正の調節因子が頻回に検出されるためである。その逆もまた真である、つまり内皮細胞の代謝回転が増加している組織内では正および負の調節因子がしばしば共存している。これは、内皮細胞活性化状態が正および負の調節因子間のバランスによって決定される「血管新生スイッチ」の概念をもたらしてきた。

活性化された(血管新生性)内皮では正の調節因子が優勢であるが、他方、内皮の静止期は負の調節因子の優勢によって達成かつ維持される[ハナハン(Hanahan)ら,Cell, 86:353−364(1996)]。「スイッチ」の概念は、最初は腫瘍進行の状況において早期血管相から血管相への移行を説明するために使用されたが、発達的、生理的並びに病理的血管新生の状況においても適用できる。依然としてin vivoでは明確には証明できてはいないが、「スイッチ」には正の調節因子の誘導および/または負の調節因子の消失が含まれているというのが最新の研究仮説である。

血管新生は極めて多数の生理的および病理的プロセスにおいて決定的に重要な役割を有しているので、血管新生の制御に関与する因子が鋭意研究されてきた。血管新生の調節には多数の増殖因子が関与していることが証明されている;これらには繊維芽細胞増殖因子(FGF)、血小板由来増殖因子(PDGF)、トランスフォーミング増殖因子α(TGF−α)、および肝細胞増殖因子(HGF)が含まれる。概観については例えば、フォークマン(Folkman)ら,J.Biol.Chem.,267:10931−10934(1992)を参照。

VEGF−D遺伝子はヒト成体において広範囲に発現するが、もちろん、至るところに発現する訳ではない。VEGF−Dは心臓、肺および骨格筋で強度に発現する。中間レベルのVEGF−Dは脾臓、卵巣、小腸および結腸で発現し、より低レベルの発現は腎臓、膵臓、胸腺、前立腺および精巣で発生する。脳、胎盤、肝臓もしくは末梢血白血球からのRNA中ではVEGF−D mRNAは全く検出されなかった。

今までに次の2種のサブクラスに属する5種の内皮細胞特異的レセプターチロシンキナーゼが同定されている:3種の血管内皮細胞増殖因子レセプターであるVEGFR−1(Flt−1)、VEGFR−2(KDR/Flk−1)、VEGFR−3(Flt4)、およびTieファミリーであるTieおよびTie−2(Tek)の2種のレセプター。これらのレセプターは特異性および親和性が相違する。これらはすべて、シグナル伝達のために必要な内因性チロシンキナーゼ活性を有している。

VEGFR−1欠損マウスは、結果として異常な血管路(vascular channel)の形成を生じさせる血管内への内皮細胞の非効率的な構築を原因として妊娠中期に子宮内で死亡する[フォン(Fong)ら,Nature 376:66−70(1995)]。VEGFR−2欠損マウスは***8.5および9.5日後に子宮内で死亡し、VEGFR−1とは対照的に、これは内皮細胞前駆物質の発達不全が原因であると思われる[シャラビー(Shalaby)ら,Nature 376:62−66(1995)]。腫瘍血管新生におけるVEGFR−2の重要性は優性阻害アプローチ(dominant−negative approach)を使用することによっても明白に証明されている[ミラウアー(Millauer)ら,Nature 367:576−579(1994);ミラウアー(Millauer)ら,Cancer Res.56:1615−1620(1996)]。VEGFR−3欠損マウスの表現型についてはデュモン(Dumont)ら,Cardiovascular Failure in Mouse Embryos Deficient in VEGF Receptor−3(VEGFレセプター3欠損マウス胎児における心血管不全),Science,282:946−949(1998)の中で報告されている。VEGFR−3欠損マウスは欠陥のある血管発達を原因として妊娠12〜14日に子宮内で死亡する。

発明の概要
本発明は、血管新生の刺激において有用な遺伝子療法アプローチに向けられている。本発明の1つの目的は、少なくとも、VEGF−B167に対するポリヌクレオチド配列を有する第1ベクターコンストラクトおよび別のVEGFに対するポリヌクレオチド配列を含有する第2ベクターコンストラクト、好ましくは非増殖性アデノウイルスコンストラクトを用いる、他のVEGFタンパク質およびペプチドに加えてVEGF−B167タンパク質またはペプチドが血管新生を刺激するために持続的な期間に渡って治療的に有意な程度まで産生させられる、血管新生を刺激するための方法を提供することである。

本発明の1つの実施形態に従うと、血管新生を刺激する方法は、VEGF−B167またはそのフラグメントもしくは保守的置換物をコードするポリヌクレオチド配列を備える第1の非増殖性アデノウイルスベクターを生体に導入するステップ、および、VEGF−A、または、そのフラグメント、もしくは、保守的置換物をコードするポリヌクレオチド配列を備える第2の非増殖性アデノウイルスベクターを生体に導入するステップを備えている。この実施形態では、非増殖性アデノウイルスベクターは生体の少なくとも1個の細胞に送達されるが、標的細胞は内皮細胞またはその内皮細胞に近接する細胞のいずれかである。

非増殖性アデノウイルスコンストラクトを使用する理由は、それが導入遺伝子の有効なin vivo発現を許容することにある。このウイルスは高力価へ産生されるので、多数の細胞型をアデノウイルスで感染させることができる。これはさらにまた組換えアデノウイルスとの同時感染により2種以上の導入遺伝子の同時発現も許容する(例、VEGF−B + VEGF−A)。発現はさらにまた極めて迅速で、たいていは24〜48時間以内である。

好ましい実施形態の詳細な説明
実施例1 VEGF−B167もしくはVEGF−Aを有する組換えアデノウイルスの産生
AdVEGF−B167ベクターを作製するために、PCR(フォワードプライマー:5’ CGA TCT GGC CAT ACA CTT 3’(配列番号1);リバースプライマー:5’ CTA TGG ATC CTC ACC TTC GCA GCT T 3’(配列番号2)、BamHI部位を含有する)によってプラスミドから全長VEGF−B167 cDNAを増幅させ、その後TA−プラスミドpCR2.1ベクター内へクローニングした(インビトロジェン[Invitrogen]社製)。

E.coli内での増幅およびプラスミド精製に引き続いて、BamHIを用いてインサートを切り出し、プラスミドpQBI−AdCMV5−IRES−GFP(クワンタム・バイオテクノロジーズ[Quantum Biotechnologies], ケベック、カナダ;トランスファープラスミド)のBamHI部位内へクローニングした。このプラスミド内では、cDNA発現はCMVプロモーターによって推進される。このプラスミドはさらにグリーン蛍光タンパク質(GFP)の同時発現を許容するIRES−エレメントも含有している。VEGF−Bの発現は、プラスミドをCOS−1細胞内へトランスフェクトすることにより検証した。

FseIを用いて上記のトランスファープラスミドを直線化させ、その後に環状アデノウイルスプラスミドであるpJM17(マイクロビックス・バイオシステムズ社[Microbix Biosystems Inc.]、オンタリオ州、カナダ)を用いて293細胞内へコトランスフェクトした。トランスフェクションはリポフェクタミン・プラス(Lipofectamin Plus)を用いて実施した。14日後、細胞を採取し、冷凍−解凍によって破壊し、低速遠心分離から採取した上清を使用して新しい293細胞を感染させた。増幅ステップを1回繰返した。インサートの発現は、インジケーターとしてGFPを使用して検証した。この後、標準プロトコールを使用して組み換えウイルスを293細胞中でプラーク精製した。GFP陽性プラークを増幅し、引き続いてこれを使用してCsCl遠心分離により精製されるストックを調製した。

AdVEGF−Aベクターを作製するために、全長VEGF−Aをヒルトゥネン(Hiltunen)ら,Circulation,102:2262−2268(2000)に記載されている第1世代アデノウイルスベクター内へクローニングした。この参考文献は明示的に参照してここに組み込まれる。あるいはまた、VEGF−Aの活性フラグメントまたは保守的置換を含有する全長配列もしくはフラグメント配列を使用できよう。当分野において知られている他のベクター類の調製および様々なベクター調製法の使用は本発明の範囲内である。さらに、VEGF−B167もしくはVEGF−Aアデノウイルスベクターを作製するために使用した方法は、VEGF−CもしくはVEGF−Dアデノウイルスベクターを作製するためにも使用できよう。

293EBNA細胞は、VEGF−B167の2x107pfuを含有する無血清培地中で1時間インキュベートした。さらに、リン酸カルシウム法を使用してVEGF−BをコードするpREP7(インビトロジェン[Invitrogen]社製)プラスミド発現ベクターもしくはpREP7ベクターコントロールを用いて細胞をトランスフェクトした。細胞は、翌日にS35メチオニンおよびシステイン(プロミックス[Promix]、アマシャム社製)を用いて6時間かけて代謝的に標識した。培地を収集し、VEGF−Bに対して産生したポリクローナル抗体を使用して標識VEGF−Bタンパク質を沈降させた[オロフソン,ビー(Olofsson B.)ら,Proc.Natl.Acad.Sci.USA,93:2576−2581(1996)参照]。プロテインAセファロースに結合した沈降タンパク質をリン酸緩衝生理食塩液中で3回洗浄し、Laemmliサンプル緩衝液中に溶解させ、12.5%もしくは15%SDS−PAGEゲル上で分析した。その後ゲルを乾燥させ、ホスホイメージャーおよびオートラジオグラフィーによって分析した。図1に示した通り、AdVEGF−B167を用いて感染させた、またはVEGF−B167をコードするpREC7プラスミドを用いてトランスフェクトした細胞は約21kDの主要ポリペプチドを産生した。

実施例2 組換えアデノウイルスに対する血管新生応答の評価
約2x108pfu(プラーク形成単位)のVEGF−B167およびVEGF−A組換えアデノウイルス各々を3匹のNMRI nu/nuマウス(ハーラン[Harlan]社、オランダ)の耳5個に皮下注射した。注射後の様々な時点でマウスを屠殺し、注射部位からの皮膚を4%パラホルムアルデヒド中で固定し、パラフィン内に包埋した。7μm切片をVEGFR−2に対するモノクローナル抗体(カタオカ,エイチ(Kataoka H.)ら,Development Growth & Differentiation,39:729−740(1997))または血小板内皮細胞接着分子1(PECAM−1)(BDファーミンジェン[Pharmingen]社製、サンディエゴ、CA、米国、製品番号01951D)に対するモノクローナル抗体を用いて染色した。

PECAM−1は、PECAM−1分子が106まで濃縮されている内皮細胞細胞間接合部の主要構成分である免疫グロブリン(Ig)スーパーファミリーの130kDメンバーである。PECAM−1はさらに循環血小板単球、好中球および選択したT細胞サブセットの表面上でも発現する。チラミドシグナル増幅法(TSA)キット(NENライフサイエンス[Life Sciences]社製、ボストン、MA、米国)を使用して染色を増強した。陰性対照は一次抗体を除外することによって産生させた。結果は、オリンパス(Olympus)AX80顕微鏡を用いて視認し、写真を撮影した。

図2A〜2Fに示されているPECAM−1染色から見て取ることができるように、AdVEGF−B167およびAdVEGF−Aの複合トランスファーはAdVEGF−A単独(図2Bおよび2E)より強力なPECAM−1陽性血管(図2Cおよび2F)の形成を誘導したが、他方、AdLacZ(図2Aおよび2D)は血管構造に何の作用ももたらさなかった。バーの目盛は100μmを表している。

定量のために、正方形グリッド(面積=0.16mm2、倍率20倍)を使用して、PECAM−1陽性血管を倍率200倍で計数した。平均値およびp値は対応のスチューデントt検定を使用して計算した。Lacウイルス感染対照サンプル中の血管数の標準偏差の2倍を超える平均値だけを考慮に入れた。

図3から読み取ることができるように、AdVEGF−B167およびAdVEGF−Aの複合遺伝子トランスファーは血管密度のほぼ3倍の増加を誘発し、VEGF−AはAdVEGF−B167もしくはAdLacZ単独を注射した耳と比較してp<0.001で血管密度の2倍の増加を誘発した。このように、VEGF−Bは、VEGF−Aの血管新生効果を増強する。VEGF−Aに対して、VEGF−CもしくはVEGF−Dの全長、活性フラグメント、もしくは保守的に置換した配列を置換し、上記と同様の結果を達成することは本発明の範囲内である。

実施例3 本発明に従ったアデノウイルスの投与
VEGF−B167配列の全長もしくは活性フラグメントを備える第1組換えアデノウイルスを上記の通りに、または当分野において許容し得る他の方法に従って調製する。この配列に保守的置換を行うことができる。VEGF−A、VEGF−CもしくはVEGF−D配列の全長もしくは活性フラグメントを備える第2組換えアデノウイルスを上記の通りに、または、当分野において許容し得る他の方法に従って調製する。この配列に保守的置換を行うことができる。

2種の組換えアデノウイルスベクターを投与するために標的細胞を同定する。例えば、微小血管内皮細胞を標的とすることができる。2種のベクターを用いて標的細胞をトランスフェクトすることのできる方法によって2種のベクター類を標的細胞へ投与することができる。例えば、微小血管構造内へのベクターの注入のような。所望であれば、血管新生の刺激の程度を測定するために標的細胞を備える生体を監視もしくは評価することができる。

組換えアデノウイルスベクターを内皮細胞に近接する細胞へ導入することが望ましい場合がある。トランスフェクションに感受性であるあらゆる細胞型およびあらゆるトランスフェクション法を使用できる。これらのベクターでコードされたVEGFはトランスフェクトされた細胞から分泌され、近位内皮細胞内での所望の血管新生刺激を惹起するであろう。例えば、虚血性脚モデル、骨格筋細胞が本発明に従ったアデノウイルスベクター投与の標的とできよう。トランスフェクションに引き続いて、標的とされる骨格筋細胞はトランスフェクトされたGEGFを分泌し、近位内皮細胞中での細胞増殖および血管形成を誘発するであろう。

- 血管新生を刺激するための、下記を備える方法:
VEGF−B167または、そのフラグメント、もしくは、保守的置換物をコードするポリヌクレオチド配列を備える、第1の非増殖性アデノウイルスベクターを生体に導入するステップ;および
VEGF−A、または、そのフラグメント、もしくは、保守的置換物をコードするポリヌクレオチド配列を備える第2の非増殖性アデノウイルスベクターを前記生体に導入するステップ、
このとき前記第1の非増殖性アデノウイルスベクターおよび前記第2の非増殖性アデノウイルスベクターは前記生体の内皮細胞および内皮細胞に近接する細胞からなる群から選択される少なくとも1個の生細胞へ送達される。 - 前記少なくとも1個の生細胞が血管細胞である請求項1記載の方法。
- 前記内皮細胞が微小血管内皮細胞である請求項1記載の方法。
- 前記内皮細胞が大動脈内皮細胞である請求項1記載の方法。
