と毒性によって、実際の産生レベルがしばしば重大な制限を受け、発現量を増加
させるためのいくつかの試みがなされている。例えば、Knappik & Pluckthun(1
995)は、抗体フレームワークにおいて発現量に劇的な影響を及ぼす鍵となる残基
を同定した。同様に、Ullrich et al.(1995)は、CDRにおける点変異によっ
てペリプラズム抗体断片の発現量が増加することを見出した。それでもやはり、

1価の結合親和性と元のmAbの特異性を保持し、細菌にて効率的に生産される 14 。scFvは、ハイブリドーマ細胞に由来する目的の結合特性を示すmAbの
可変領域のクローニング、または免疫されたもしくはナイーブファージライブラ
リー15,16に由来する所望の特異性を有するscFv断片を直接選択することに
よって構築することができる。しばしば、ハイブリドーマからクローニングされ
たscFvは、低い産生量と低い熱力学的安定性を示し17、インビボでの適用に
対する有用性が制限されるのに対し、ファージライブラリーから選択されたsc
Fvはすでに、抗原結合だけでなくscFv形態における安定性と折り畳み特性
についても選択を受けている18。
ローナル抗体由来のマウス抗体断片に対する応答(HAMA応答)、を回避する
のが好ましい。そのような問題を解決するため、ヒト抗体断片をヒト抗体ライブ
リーのスクリーニングによって得ることができる(EP-A1 0 859 841;Vaughan e
t al., 1996)。別の解決手段は、CDR領域のヒトフレームワークへのグラフテ
ィングにより、非ヒトモノクローナル抗体の特異性を移すことである(EP-B1 0
239 400)。この方法の改善として、前記非ヒト抗体に由来するさらなる残基を導
入することによって、改善された結合様式を有するヒト化抗体または抗体断片を

ームワークであり、抗c−erbB2(p185Her2−ECD)4D5mAb(
HerceptinTM)のヒト化に使用された19。その後の研究によって4D5

定性と相関し(WornおよびPluckthun, 1999)ており、scFvのインビボ適用と

役割は提唱されているが(Simon et al., 1990)、膜貫通糖タンパク質EGP−
2の確かな機能は、わかっていない。最近の報告によって、EGP−2はホモフ
ィリックな細胞−細胞接着分子として同定されており5,6、EGP−2は、侵襲
性および化学感応性(chemoresponsiveness)の可能性のある修飾因子として同
定されている7。EGP−2を標的とした新しい免疫治療の可能性を評価する研
究において、mAbMOC31に化学的に融合したエキソトキシン−A(ETA
)が大きい腫瘍の成長を遅延させることが見出された8。
P−2は、腫瘍の造影や治療のための放射能標識された抗体の標的として役だっ
ている。分子量を小さくし、それによって組織浸透性とそのような抗体ベースの
構築物の血清クリアランスを高めることにより、標的効率を改善する努力がなさ
れている。組換え抗体法により得られるFab、(Fab)2、dsFvおよびs
cFv断片は、これに関して大きな潜在能力を有する9,10けれども、これまで、
安定性、分子量および親和性に関する最適な様式は決定されておらず、特定のイ
ンビボ系や適用目的によって、異なった抗体−エフェクター融合タンパク質につ
いて微調整がなされなければならない11。
影および治療剤に基づく新しい抗体断片の開発に関して、マウス抗EGP−2ハ
イブリドーマMOC31の可変領域が、一本鎖Fv断片形態にクローニングされ
た16。得られたscFvは、EGP−2に対する期待される結合親和性と特異性
を示し、これは他者によって免疫組織染色実験で組織片についても示された30が
、細菌のペリプラズムにおける発現は低かった。ヌードマウスにおいてこのsc
Fv断片を用いたインビボターゲティング実験は失敗した。scFvは腫瘍に蓄
積するだけでなく、フルオレセインに対する無関係な対照scFvよりもゆっく
りとしたクリアランス速度も示した。MOC31 scFvが高分子量の凝集体
を形成し、体温(37℃)にて血清中でインキュベーションしたときにその活性
を急速に失うことを示すことができた。高度に精製されたscFvの37℃での
PBS中でのインキュベーションにおいて、同様の沈殿と免疫活性の損失が観察
できたので、これは、主として、タンパク質加水分解よりもむしろ不十分な熱安
定性によるものである。

を、プログラムnaccess(S. HubbardおよびJ. Thornton, 1992, http:/
/sjh.bi.umist.ac.uk/naccess.html)を用いてリガンドの存在下および非存在下
での既知の抗体タンパク質複合体の側鎖溶媒接近可能表面を比較することにより
同定した。モデルを、ハイブリッドscFv 4D5MOC−Aにおいて生じる
、立体障害の可能性、可能性のある抗原接触およびCDRコンフォメーションで
間接的な影響を有するも知れない残基についてチェックした。第二の構築物sc
Fv 4D5MOC−Bにおいて、VHのコアにおける8個の鍵となる残基は、M
OC31VHフレームワークの構造サブタイプを保存するために、4D5配列に
対してこれら残基を変化させるのではなく、MOC31から保持されていた。

4D5MOC−Aおよび4D5MOC−Bのペリプラスム発現のために、pIG
6ベクターを使用し、一方scFv MOC31の発現にpAK400ベクター1 6 を使用した。大規模な生産のために、E. coli SB536株(Bass et al. 199
6)を使用した。5L容の振盪フラスコ中の1%gluおよびアンピシリン(30μ
g/mL)を含む1LのdYTを、30mLで接種し一晩培養した。培地のOD5 50nm が0.5になったら、最終濃度1mMのイソプロピル−D−ガラクトピラノ
シド(IPTG;Boehringer Mannheim)を用い、24℃にて、3〜4時間scFv産
生を誘導した。最終のODは4D5、4D5MOC−A 4D5MOC−Bにつ
いて5〜6、scFv MOC31については4であった。回収したペレットを
−80℃で保存した。

すべての一本鎖Fv断片を、以前に記載されたように(Pluckthun 1996, book c
hapter)、BIOCAD-System(Perseptive BioSystem-Inc.) にて直列に連結したN
i+−DNAカラムおよびHS/M−4.6/100イオン交換カラムを用いて精
製した。リゼートをNi+−DNAカラムに載せ、カラムを20mMHepes
( pH7.0)、500mM NaCl、および第2段階で20mM Hepes(
pH7.0)、40mMイミダゾールで洗浄したのち、結合したタンパク質を20
0mMイミダゾール( pH7.0)で溶出した。溶出液をHS/M−4.6/10
0イオン交換カラムに直接載せ、特異的に結合したタンパク質を、20mM H
epes( pH7.0)中の、0〜500mM NaClの塩グラジェントにより溶
出した。抗体断片を含む画分を過剰のPBSに対して透析し、10kDaカット
オフフィルター(Ultrafree-MC low protein binding, Millipore)を用い、4℃
にて4000gで遠心することにより、1mg/mLに濃縮した。抗EGP−2
scFv断片 MOC31については、第3の精製工程として、Superdex75 カラ
ム(Pharmacia)を用いて分離用ゲル濾過を行なうことが必要であったが、最終
的な収率は10分の1に落ちた。精製の結果を、還元的条件下で12.5%SD
S−PAGEによりチェックした。すべての精製されたscFvの分子量を質量
分析によりチェックした。

して使用した。マウスを注射後、1、4および24時間後に屠殺した。組織およ
び臓器を取り出し、ガンマカウンターを用いて活性を調べた。99mTc標識した
scFv 4D5による生体内分布の解析は最近記載された(Waibel et al., 19
99)。
Fv断片を構築し、その機能性を決定し、独立に構築したscFv MOC313 0 と配列および特性が一致したその抗原EGP−2(表1)に対するかなり高い親
和性を実証した。意外にも、このscFvのインビボでの存在は、EGP−2特
異性なしでは、対照のscFvとほとんど区別不可能であり、本質的にscFv
MOC31は、異種移植された腫瘍に局在しなかった(表2)。したがって、我
々は、このタンパク質は十分には安定でなく、最初によく特徴付られた安定なフ
レームワークをグラフティングすることによって2つのより安定な変異体を設計
すること、および、第2に、さらに可変領域のうちの1つの内部におけるいくつ
かの残基を変化させることとの仮説を立てた。レシピエントフレームワークとし
て、我々は、それ自身グラフティングの産物である、4D5のヒト化された形態 19 を選択した。このフレームワークは、本質的に、生殖細胞系IGHV 3−6
6(IMGT)由来の重鎖可変領域、VH3−18(Vbase)、Locus
DP 3−66(DP−86)および生殖細胞系IGKV 1−39(IMGT)由
来のκ軽鎖可変領域、VK 1−1(Vbase), locus DP O12から

した(Figure1A)。Kabat定義よりもむしろこの情報に基づいて、どの残基を4
D5フレームワークから選ぶかおよびどの残基をMOC31配列から選択するか
を決定した。得られたグラフトは、このように、Kabat et al.(1979)またはCho
tia(Allazikani et al., 1997を参照)のCDR定義には厳密には従わないが、
VLの「アウターループ」(残基L66-L71)のコンフォメーションを決定する2つ
の残基(L64およびL66)を含んでいる。このループの先端は、いくつかの複合体
において抗原と接触することが示され、CDRL1のコンフォメーションに対す
るこのループの影響は無視できなかった。残基L66は一般にκ軽鎖ではGlyであ
り、正のΦ角である。この残基がGly以外の残基で置換されれば(4D5のArg
)、他のループは異なるコンフォメーションをとり、その領域から離れて折れ曲
がる。VHでは、CDR H1に加えて、さらに残基H27−H30を含ませたが
、CDR定義(Kabat et al.(1979)に従えばCDR H2の一部であるにもかか
わらずCDR2の塩基のいくつかの残基を脱落させ(H62およびH63);All
azikani et al., 1997)、および、時としてCDR4と呼ぶ、VHの「アウタール
ープ」におけるいくつかの残基を含ませ(残基H69、H71、H75−H77
)、構築物4D5MOC−Aを得た(Figure1A)。

コンフォメーションの違いが分子に見つかっているが31、コンフォメーションの
違いは、フレームワーク1(FR1)、特にH7−H10の位置、においてもっ
とも顕著であった。これらのコンフォメーションの変化は、恐らくGluH6(4
D5におけるように)またはGln H6(MOC31におけるように) を完全に隠
しドメインのコアにおいて確立する、異なる水素結合パターンによって引き起こ
され、残基H9の性質(Pro、Glyまたは他の残基)32によってさらに影響を受
ける。SaulおよびPoljak(1993)は関連する構造上の変化によって、領域コアか
らCDR2のベースへ横切るFR1コンホメーションにおける変化の効果を伝え
る残基H9、H18、H82、H67およびH63に影響を及ぼし、抗原結合に
強力に影響を及ぼすことが可能である。
我々は、これらのフレームワークのクラスがどの程度、ループグラフトの機能に
影響を及ぼすのか知らなかったので、この問題を実験により試験することにした
。4D5MOC−A構築物において、VHドメインフレームワークは4D5サブ
タイプに変化しているので、4D5MOC−Bは完全にMOC31コアパッキン
グ、並びにコンフォメーションに重要な残基H6およびH9を保持している。こ
れを達成するために、抗EGP−2一本鎖Fv断片配列の8個のさらなるフレー
ムワーク残基(H6, H9, H18, H20, H38, H63, H82およびH
109)は、MOC31配列を保持しなければならなかった。これら変化のすべ
てはscFvの下半分(Figure1)に存在し、H9の位置におけるGlyからProの
置換を除いて、ドメインのコア内に隠れている。したがって、これらはこの構築
物の免疫原性には影響を及ぼしていないと思われる。

Fv 4D5MOC−Aを精製して400μgを得ることができ、4D5MOC
−Bは純度95%で1mgまで精製することができた。両一本鎖Fv断片は1m
g/mLまで濃縮することができ、還元SDS−PAGEで解析した(Figure2
)。両分子の質量分析は、scFv 4D5MOC−Aについて29.855Da
、scFv 4D5MOC−Bについては29.897Daの期待された分子量
を示した。
2結合特異性の移動は、EGP−2過剰発現SW2細胞に対する放射能標識した
グラフト変異体の結合競合により、両変異体4D5MOC−Aおよび4D5MO
C−Bについて成功したことが示された。モノクローナル抗体 MOC31のみ
がグラフト変異体の結合を阻害することができたにのに対し、無関係な対照抗体
は競合しなかった(Figure3A)。c−erbB2またはEFG−受容体(細胞
外領域)にてインキュベーションしたとき、グラフトされた分子の交差反応性は
みとめられなかった(Figure3B)。
ングのための抗体の最も重要な特性の1つであると考えられる。結合親和性がグ
ラフト実験において保存されることを確実にするために、細胞上での放射能標識
された一本鎖Fv断片解離定数をラジオイムノアッセイ(RIA)にて測定した
。グラフト変異体は、ナノモル濃度の範囲で、元の抗EGP−2 一本鎖Fv断
片に匹敵する特異的で類似の結合様式を示した(表1)。
BIAcore装置(Pharmacia)にて表面プラスモン共鳴により解析した(表1)。オフ
−レート(off-rate)の過小評価につながる再結合の効果を最小限にするために
、我々は、低密度のコーティングと速い流速を用いた。scFv MOC31は
、別の測定結果(半減期33分)30と一致して、半減期が約38分で、その標的
に対する安定な結合を示した。scFv 4D5MOC−Aおよび4D5MOC
−Bのkoff値は元のscFv MOC31と非常に類似しており(表1)、4D
5-フレームワークにおいてscFv MOC31の結合特性の完全な移動が成功
したことを示している。
高い温度でインキュベーションすることは重要である。これら分子の生物物理学
的挙動はしばしば生体内での適用の限界になる。したがって、我々は、高濃度お
よび高い温度でのscFvの凝集挙動を試験した。4D5、4D5MOC−Aお
よび4D5MOC−Bは、限外濾過によって1mg/mLまで濃縮することがで
きたのに対し、MOC31 scFvは、400μg/mLを越える濃度で沈殿し
た。この濃度では、Superdex 75 カラムを用いたSmart ゲル濾過システム(Phar
macia)による解析用ゲルろ過により、総タンパクの約10%が高分子量の凝集
体として溶出した。単量体については予想どおり溶出量1.27mLでほぼ90
%のタンパク質が溶出した。しかし、37℃で30分以内に既に約85%のタン
パク質が沈殿した。残りの15%の可溶性タンパク質は単量体として溶出した。

aの分子量に対応して、溶出量1.20mLで溶出し、両一本鎖Fv断片が1mg
/mLの濃度で単量体で存在していることを示した。4D5MOC−AはMOC3
1よりもゆっくりと沈殿したが、37℃でのPBS中での20時間の一晩のイン
キュベーションおよびその後のゲル濾過は、タンパク質がほとんど溶出しないこ
とを示した(Figure4A)。同じ条件下でインキュベーションすると、4D5M
OC−Bは依然として、溶出量1.20mLで対照的なピークとして溶出した(
Figure4B)、2つの変異体の間の、固有の(熱)安定性における大きな違いを示
した。最も重要なことに、4D5MOC−Bは、それによって、インビボ適用に
必要な生物物理学的特性を有することが示された。
末端ペンタまたはヘキサヒスチジンタグに安定に結合する新しい方法を用いて、
99mTcで標識した(Waibel et al, 1999)。元のscFv MOC31を除い
て、すべてのscFv断片を37℃にて1mg/mLのタンパク濃度で標識するこ
とができ、取りこまれた最初の99mTcの30〜40%において生じ、最終の比
活性は300〜400mCi/mLであった。凝集性のscFv MOC31につ
いては、インキュベーションの温度を30℃に下げなければならず、最大の可能
性のあるタンパク質濃度は400μg/mLであり、減少した取り込み量を示し
た(総Tcの25%、250mCi/mL)。
ベーション後も依然として活性なscFv分子の画分を測定した(Figure4c) 29 (Figure4D)。scFv MOC31については、我々は、30℃で標識反応
を行なえば67%±5.4のタンパク質がなおも活性であることを見出した。そ
の他の断片は、scFv 4D5MOC−Aについて47.25%±4.9、sc
Fv4D5MOC−Bについて74.5%±8.3、およびscFv 4D5につ
いて87.3%± 6.4活性であることを示し、すべて37℃で標識した。血清
安定性を試験するため、scFv断片(17μg/mL)をヒト血清中37℃にて
20時間インキュベーションし、得られた免疫活性を測定した。scFv MO
C31は一晩インキュベーションした後完全に不活性であることがわかったので
、より早い時点で測定した。1時間後には既に、活性は6.32%±0.17(最
初の免疫活性の9.4%)に落ちた。4時間後には、1.95%±0.175(2.
9%)が活性であるに過ぎなかった。対照的に、20時間で、4D5MOC−A
の活性は47.25%±4.9から8.1%±4.7(最初の活性の17.1%)に、
scFv 4d5MOC−Bは74.5%±8.3から36%±1.6(48.3%)
に、およびscFv 4D5は87.3%±6.4から40.45%±8.75(4
6.3%)に落ち、ゲル濾過アッセイにおいて異なる熱安定性がみとめられること
が確認された。
5MOC−Aおよび4D5MOC−Bについて行なった。scFv MOC31
については、我々は、1時間、4時間および24時間後に0.92よりも高い腫
瘍−血液比を得ることはできなかった(n=3、各時点)。24時間後、腫瘍に
対する総投与量は1.24%ID/g組織であったが、血液においても1.34%
ID/g血中であり、標識法と比較して血液中で通常認められる3〜5倍低い値
と比較して非常に高かった(表2)。対照的に、99mTc標識scFv 4D5の
生体内分布は、SK−OV−3細胞に対する24時間後の総投与量1.5%ID
/gで腫瘍対血液比が8.3であり(Waibel et al., 1999)、抗c−erbB2
scFcC6.5について同様の結果が報告されている34。4D5MOC−Aに
ついては、我々は、24時間後に、総投与量0.84%ID/gで弱い増加のみ
を認め、腫瘍対血液比は1.95であった(表3)が、scFv 4D5MOC−
BのSW2腫瘍を有するマウスにおけるインビボ適用は、腫瘍に対する24時間
後の総投与量1.47%ID/gで5.25の腫瘍対血液比をもたらした。4時間
後に測定した腫瘍に対する最大投与量は1.82%ID/gであり、これは抗原
に対するscFv 4D5MOC−Bの安定な結合を反映して非常にゆっくりと
減少した(表3)。比特異的フルオレセイン対照scFv FITC−E215 に
ついては、腫瘍における豊富化はみとめられなかった(表4)。

減少が効率を高めるであろうと提議された8。改善された組織浸透性およびもっ
と小さな抗EGP2scFv断片の速いクリアランス速度がより良好な結果をも
たらすか否かまたは正常なEGP−2発現上皮組織に接近し、mAbは150k
Daという分子量のために接近できない1その増大した能力がその方法の解決す
る力を制限するか否かについてなおも試験されている。改善されたscFvは、
大きさおよび結合活性の効果を最適化するための、他の組換え分子形態、二量体
化および多量体化されたscFv、Fabまたは(Fab)2 11等、のための構築
ブロックとして役に立つことができる。それらはまた、治療に基づいた抗体断片
の構築において、他のエフェクタ領域と融合することができる。scFv MO
C31−ETA融合は、SW2細胞に対し、ETAとのmab MOC31融合
物(Zimmermann, 非公開の結果)よりもインビトロで1万倍以上毒性であった。
元の修飾されていないscFv MOC31もまたT細胞再ターゲティングのた
めのCD3特異性を有するディアボディの構築に使用した。この形態では、sc
Fv MOC31は幾分安定化されたようであり、12時間の半減期が報告され
ているが、収量はscFv MOC31単独の場合と同じくらい低く37、インビ
ボ適用はこれまでのところ報告されていない。
Hドメインがループドナー MOC31とは異なる構造のサブクラスに属するこ
とに気づいた。単独のループグラフトが失敗した文献36がいくつか存在し、キメ
ラが複数のさらなる復帰突然変異により救済されなければならないので、我々は
MOC31の構造のサブクラスおよびコアパッキングが保存されている第2キメ
ラを直接設計した。これには、MOC31VHドメインの表面を付け替えること
に本質的に対応するよりも、マウスの配列に対する、主としてコア内に存在する
8つのさらなる残基の変化が関与する。これらのさらなる突然変異は抗原親和性
には何の効果もなかったが、キメラの安定性に対する有益な効果があった。4D
5MOC−Bに対するさらなる突然変異によって、非常によく振舞う4D5sc
Fvと非常によく似た振る舞いをする分子が得られた。このことは、4D5MO
C−Bが4D5MOC−Aよりもさらに4D5から配列においてさらに除去され
ており、全体で残基H6、H7およびH9によって定義され、これらの残基が相
互に関係づけられるのでフレームワークを混入しないフレームワーククラスを維

基と不適合なフレームワークコア残基との間の望ましくない相互作用によるので

gure1)、4D5およびMOC31の間で異なる低いコアに存在するそれらのフ

のインビボターゲティングを初めて報告した。処理を施されたscFv 4D5
MOC−Bは、scFv MOC31の制限を克服し、EGP−2を発現してい
