Claims (1)
- 【特許請求の範囲】 1.化合物のライブラリーに選択された興味ある性状を持っている化合物を同定 する方法であって、前記化合物のそれぞれは固相担体に結合しており、N反応工 程で構成される独自の反応系列によって生成され、そこで各化合物が構成要素か ら調製され、Nは少なくとも1から約100までの整数で、 a)CO2H,OH,SH,NH2,NHR,CH2Cl,CH2BrおよびCHN2よりなる群から選択さ れた前記固相担体表面の少なくとも1のタイプの第一の官能基を持つ1群をMバ ッチに分割すること、ここでRはC1-C1の直鎖アルキル基でMは少なくとも2から 約25の整数であり; b)前記第一の官能基を通じて固相担体との結合を形成するために、それぞれM の異なった構成要素と少なくとも1セットの1反応において固相担体のMバッチ とをカップリングすることで、前記構成要素は独立任意に保護されて、 c)任意にカップリング工程b)の前に、それとも同時に、または工程b)の後に 、約0.001から0.5モル当量のスペクトルで区別可能な各構成要素と独自に関連し たフルオロホア標識を各バッチへ付加すること、前記標識はその独特の励起波長 、放射波長、励起状態寿命および放射強度によって同定され、構成要素に結合し た第一の官能基と同じか、または異なるかもしれない任意に保護された、第二の 官能基を任意に経て固相担体表面へ直接的結合か、または少なくとも1の酸素か 窒素原子またはカルボニル、(C=O)NHまたはNH(C=O)基によって任意に中断される C1-C9の直鎖または分枝アルキル・リンカー基を経て間接的結合を形成できるよ うに前記標識が活性化され、ここで前記第二の官能基が保護されている場合、前 記直接または間接的結合を形成する以前に、前記官能基は脱保護され、前記リン カーは固相担体の表面の第二の官能基に結合し、 d)任意にすべてのMバッチを再結合すること、前記再結合工程は工程e)以降で あり e)ライブラリーの何れの化合物が、固相担体に結合または脱離している間に 、興味のある性状を有することを示すことができる分析法を実施すること、 f)そこで結合したフルオロホア標識のそれぞれの相対量を測定するために、 それぞれの固相担体のスペクトル蛍光データを集めること、 g)化合物の独自の反応系列を決定し、それによって興味のある性状をもつ化 合物を同定するため、工程f)で測定されるフルオロホア標識のそれぞれの相対量 を比較することによって、収集されたスペクトル蛍光データを分析すること、 を含むことを特徴とする化合物を同定する方法。 2.構成要素が、アミノ酸、ヒドロキシ酸、オリゴアミノ酸、オリゴペプチド、 サッカリド、オリゴサッカリド、ジアミン、ジカルボン酸、アミンで置換された スルフィドリル、スルフィドリルで置換されたカルボン酸、脂環式、脂肪族、異 種脂肪族、芳香族、および異種環状化合物からなる群から独自に選択されている ことを特徴とする請求項1記載の方法。 3.サッカリドは適当に保護されたD-またはL-ブドウ糖、フルクトース、イノシ トール、マンノース、リボース、デオキシリボース、またはフコースであること を特徴とする請求項2記載の方法。 4.オリゴペプチドはエンケファリン、バソプレシン、オキシトシン、心房ナト リウム増加性因子、ボンベシン、カルシトニン、上皮小体ホルモン、ニューロペ プチドY、またはエンドルフィン、またはそれについてそれの構成要素の少なく ともその20%を構成する断片、またはそれの等配性アナログであり、ここでは独 自にNH(C=O)がNH(C=O)NH、NH(C=O)O、CH2(C=O)またはCH2Oに置換される;NH2はO H、SH、NO2またはCH3に置換され;CH3SはCH3(S=O)またはCH3CH2に置換され;イ ンドールはナフチルまたはインデンに置換され;ヒドロキシフェニルはトリル、 メルカプトフェニルまたはニトロフェニルに置換され;および/または、芳香環 の水素は塩素、臭素、沃素またはフッ素に置換され;C1-C4アルキルは部分的ま たは完全にフッ素化されたC1-C4アルキルに置換されることを特徴とする請求項 2記載の方法。 5.オリゴペプチドはACE阻害剤、HIVプロテアーゼ阻害剤、細胞溶解性オリゴペ プチド、または抗菌性オリゴペプチドであることを特徴とする請求項2記載の 方法。 6.芳香族はパラ位で2基置換されたベンゼン、ビフェニル、ナフタリンまたは アンスラセン、直鎖または分枝低級アルキル、アルコキシ、ハロゲン元素、ヒド ロキシ、シアノまたはニトロで任意に置換されたものであることを特徴とする請 求項2記載の方法。 7.異種環状化合物は2,6位で2基置換されたビリジン、チオフェン、3,7位で2 基置換されてN-保護されたインドールまたは2,4位で2基置換されたイミダゾル 、直鎖または分枝低級アルキル、アルコキシ、ハロゲン元素、ヒドロキシ、シア ノまたはニトロによって任意に置換されたものであることを特徴とする請求項2 記載の方法。 8.固相担体がミクロスフェア、ビーズ、樹脂または粒子であり、ポリスチレン 、ポリエチレン、セルロース、ポリアクリレート、ポリアクリルアミド、または 望ましくはシリカまたはガラスビーズから成る群から選択される物質で構成され ることを特徴とする請求項1記載の方法。 9.固相担体が任意に置換されたオリゴ-またはポリエチレングリコールの共有 結合付加によって化学的に修飾され、任意にヒドロキシメチル、クロロメチル、 アミノメチルまたはメルカプトメチルによって置換されたアミンにより終止し、 65.ビーズアレイのスペクトル蛍光データは、電極と電解質溶液との界面に懸 濁されたビーズの空間的にコード化されたアレイを最初に形成することにより収 集され、 i)電極と電解質溶液を提供すること; ii)それぞれのタイプが複数の貯蔵器の1つに化学的または物理的に区別可能 な粒子の特性に従って貯蔵され、各貯蔵器は前記電解質溶液に懸濁された複数の 似た粒子を含む多重タイプの粒子を供給すること; iii)mxn格子配置の様式での前記貯蔵器を供給すること; iv)貯蔵器の前記mxn格子に対応するmxn構成要素を定義するために前記電極を パターン化すること; v)前記mxn貯蔵器から前記対応するmxn構成要素にmxn小滴を堆積させること、 それぞれの前記小滴は1つ前記貯蔵器から由来して1つの前記mxn構成要素に閉 じ込められるままになり、前記小滴それぞれが少なくとも1つの粒子を含む; vi)それぞれの前記小滴に同時に接触するため、先端電極を前記小滴上に配置 すること; vii)前記先端電極と前記rnxn小滴の間に電場を発生すること; viii)それぞれの前記mxn構成要素のビーズ・アレイを形成するために前記電 場を使用すること、各ビーズ・アレイは1つの前記rnxn小滴へ空間的限定された ままになる; ix)前記所定の光パターンとmxn構成要素のパターンに従って前記ビーズ・ア レイの配置を維持するために、所定の光パターンで前記パターン化された電極で 前記mxn構成要素を照射すること; x)1つの対応するmxn構成要素にそれぞれの前記mxnビーズ・アレイを維持し ている間、前記先端電極を前記電極により近くへ配置し、それによって連続した 液相に前記mxn小滴を融合すること; を含むことを特徴とする請求項62記載の方法。 66.前記構成要素が親水性であり、前記電極表面の残りは疎水性であることを 特徴とする請求項65記載の方法。 67.Nは少なくとも3から約12までの整数であることを特徴とする請求項37記載 の方法。 68.HとM.(N)は独自に少なくとも4から約12までの整数であることを特徴とす る請求項37記載の方法。 69.約0.01から約0.05モル当量までのスペクトル上で区別可能なフルオロホア 標識が工程c)で加えられることを特徴とする請求項37記載の方法。 70.請求項37に従って同定されて選択された興味のある特性持っている化合物 。 71.請求項37に従って調製した化学ライブラリー。 72.ライブラリー化合物における選択された興味のある特性を持っている化合 物を同定する装置であって、各前記化合物はそれぞれの固相担体に結合しており 、Nカップリングおよび反応工程で構成される独自の反応系列によって生産され 、そこで、それぞれの前記化合物が独自に同一か異なる1セットの構成要素から 準備され、Nは整数で少なくとも1からほほ100までで、前記固相担体が少なくと も1つの粒子アレイで、前記装置は a)その間に界面を持つ電極と電解質溶液と、 b)電極と電解質溶液との界面で電場を発生させる電場発生器と、 c)前記電極の電気化学的性質を修飾するパターン化された前記電極と、 d)前記所定の光パターンと前記電極の電気化学的性質に従って前記粒子の動 きを制御するために所定の光パターンで前記界面を照射する照明源と、 e)前記化学ライブラリーを準備する手段であって、 i)CO2H,OH,SH,NH2,NHR,CH2Cl,CH2BrおよびCHN2よりなる群から選択さ れた前記固相担体表面で少なくとも1タイプの第一の官能基を持つ1群の固相担 体をMバッチに分割する手段で、ここではRはC1-C9の直鎖アルキル基でMは少なく とも2から約50の整数であり、 ii)前記第一の官能基を通じて固相担体との結合を形成するために、それぞれ Mの異なった最初の構成要素と少なくとも1セットの1反応において固相担体のM バッチとのカップリングを行う手段、前記構成要素はさらなるカップリング工程 に参加することになっている群で任意に保護されていて、非参加の群で直交的に 保護されていて、 iii)任意のカップリング工程ii)の前に、それとも同時に、または工程ii)の 後に約0.001から約0.5モル当量のスペクトルで区別可能な各最初の構成要素とユ ニークに関連したフルオロホア標識を各バッチへ付加する手段、前記標識はその 間接的結合を形成できるように前記標識が活性化され、前記リンカーは固相担体 の表面の前記第二の官能基に結合し、ここで前記第二の官能基が保護されている 場合、前記直接または問接的結合を形成する以前に、前記官能基は脱保護され、 iv)任意にすべてのMバッチを再結合し、さらなるカップリング工程に参加す る群に存在する如何なる保護基をも脱離する手段と、 v)N構成要素を持っている化合物を形成するように、 (1) 1群の固相担体をM(N)バッチへ分割すること、ここでM(N)はNに依存し、 少なくとも2から約25までの整数であり、 (2) それぞれH(N)の異なった構成要素とH(N)バッチの固相担体のカップリン グをすること、ここでM(N)はN番目の工程中のバッチの数であり、前記構成要素 はさらなるカップリング工程に参加することができる群で任意に保護され、不 参加群で直交的に保護されていて、 (3) 任意のカップリング工程(2)の前に、それとも同時に、または工程(2)の 後に約0.001から約0.1モル当量のスペクトルで区別可能なN番目カップリング工 程中各構成要素と独自に関連した異なったフルオロホア標識を各バッチへ付加す ること、前記標識はその励起波長、放射波長、励起状態寿命または放射強度によ ってユニークに同定され、ここで、任意に保護されて、第一の官能基と同じか、 識またはリンカーはさらなるカップリング工程に参加する群を経て結合する、こ こで前記官能基が保護されている場合、前記直接または間接的結合を形成する以 前に、前記官能基は脱保護され、 (4) すべてのM(N)バッチ再結合し、さらなるカップリング工程に参加する群 に存在する場合存在するすべての保護群の脱離すること、 をN-1回反復する手段と、 vi)固相担体に結合またはそれから脱離されたライブラリー化合物が興味ある 特性を持つことを示すことができる分析法の実施する手段と、 vii)その結合したフルオロホア標識のそれぞれの相対量を測定するためにそ れぞれの固相担体のスペクトル蛍光データ収集する手段と、 viii)化合物のユニークな反応系列でカップルしたN構成要素を測定するため 、工程vii)で測定されるフルオロホア標識のそれぞれの相対量を比較することに よって収集されたスペクトル蛍光データを分析し、それによって、選択された興 味ある特性を持っている化合物の同定する手段と、 を備えることを特徴とする装置。
