Abstract
Description
Claims (1)
- 【特許請求の範囲】 1.複合磁気粒子を生成するためのプロセスであり、前記プロセスが、 (i) 親水性分枝状有機ポリマー含有水溶性媒質内に磁気粒子を形成し、 (ii) 前記ポリマーを開裂させ、その際前記複合粒子を遊離する ことを含むプロセス。 2.前記媒質がゲルである、請求項1に記載のプロセス。 3.前記ゲルが陰イオン部位を含む、請求項2に記載のプロセス。 4.前記磁気粒子が酸化鉄粒子である、請求項1に記載のプロセス。 5.前記磁気粒子が超常磁性である、請求項1に記載のプロセス。 6.前記分枝状ポリマーがデンプンである、請求項1に記載のプロセス。 7.前記ポリマーが酸化処理により開裂する、請求項1に記載のプロセス。 8.前記ポリマーの開裂が、前記ポリマーのマトリックス内に複数の前記磁気粒 子を含む粒子を処理することにより起こる、請求項1に記載のプロセス。 9.前記複合粒子を更に処理し、親水性血液残留延長ポリマーの皮膜を付着させ る、請求項1に記載のプロセス。 10.前記親水性血液残留延長ポリマーが、ステップ(ii)以前に前記媒質中に 存在する、請求項9に記載のプロセス。 11.前記血液残留延長ポリマーが、官能化ポリアルキレンオキシドである、請 求項9及び10のいずれかに記載のプロセス。 12.前記水溶性媒質が、更に直鎖ポリマーを含有する、請求項1に記載のプロ セス。 13.前記ポリマーが、磁気粒子−ポリマー複合粒子の形成後、開裂する請求項 1に記載のプロセス。 14.請求項1に記載のプロセスであり、以下の連続段階を含む: (i)加熱された水溶液中に、デンプン、第一及び第二鉄塩、並びに塩基を化合し ; (ii)オプションとして、前記溶液を15℃未満まで冷却し、ゲルを凝固差せ; (iii)pHを6.0から8.5の範囲内まで下げ、この段階は、随意に段階(ii) の前に行ってもよく; (iv)酸化物で処理し、前記デンプンを開裂させ、前記粒子を遊離し; (v) 前記の遊離した粒子を洗浄し、濾過し; (vi)オプションとして、前記の遊離した粒子を、官能化ポリアルキレンオキシド 誘導体と反応させ、前記誘導体を前記粒子に結合させ;そして (vii)オプションとして、前記の遊離した粒子をオートクレーブ殺菌する。 15.前記複合粒子の大部分が遊離するように、単一磁気粒子を含む、請求項1 に記載のプロセス。 16.前記ステップ(ii)の開裂が、二段階に行われ、第二の開裂ステップでは 、前記磁気粒子から実質的に全ての前記ポリマーを除去し、前記第二の開裂ステ ップを、前記磁気粒子に結合する安定剤の存在下で、又は添加後に行う、請求項 1に記載のプロセス。 17.プロセスステップ(i)及び(ii)を、同一反応容器において行う、請求項 1に記載のプロセス。 18.平均全粒径が4〜30nmであり、開裂した親水性ポリマー皮膜物質と共 に、超常磁性無機コア粒子を含有する複合粒子。 19.前記皮膜物質が、酸化炭水化物である、請求項18に記載の粒子。 20.前記皮膜物質が、開裂したデンプンである、請求項18及び19のいずれ かに記載の粒子。 21.請求項18に記載の粒子であって、更に親水性血液残留延長ポリマーの皮 膜を含有する粒子。 22.平均全粒径が4〜30nmの、超常磁性無機コア粒子の懸濁液であって、 前記粒子が、有機皮膜物質を実質的に含まず、表面結合無機静電安定剤を含有す る懸濁液。 23.前記安定剤がオリゴリン酸塩又はポリリン酸塩である、請求項22に記載 の懸濁液。 24.請求項18に記載の粒子であって、平均全粒径が4〜30nmであり、超 常磁性無機コア粒子を含有し、前記コア粒子表面に結合された生体分布調節剤を 有し、複数の鉄イオン結合基により、又はオリゴリン酸塩基又はポリリン酸塩基 、或いは前記調節剤に付着したホスホン酸塩基により結合されている粒子。 25.前記体内分布調節剤が、ポリアルキレンオキシド成分を含有する請求項2 4に記載の粒子。 26.請求項24及び25のどちらかに記載の粒子であり、更に開裂した親水性 ポリマー皮膜物質を含有する粒子。 27.請求項18〜26のいずれか一つに記載の粒子で、画像診断法において検 出可能な前記コア粒子表面に結合されたレポーター成分を含有する粒子。 28.前記請求項27に記載の粒子であり、前記コア粒子表面に結合されたトリ ヨードベンゼン系有機燐化合物を含有する粒子。 29.注射可能な複合粒子剤であり、無機粒子核を含有し、その表面に化学的に 又は物理的に結合された親水性血液残留延長ポリマーを含み、親水性有機ポリマ ー皮膜を遮蔽する結合部位を備えている注射可能な複合粒子剤。 30.前記遮蔽ポリマー皮膜が、開裂した分枝状炭水化物である、請求項29に 記載の注射可能な複合粒子剤。 31.前記親水性血液残留延長ポリマーが、官能化ポリアルキェンオキシドであ る請求項29及び30のいずれかに記載の注射可能な複合粒子剤。 32.少なくとも生理学的に許容可能な一担体又は一賦形剤とともに、請求項1 〜31のいづれか一つに記載の方法により生成されるか、又は請求項1〜31の いづれか一つに記載の粒子を含有する診断用組成物。 33.造影剤組成物であり、超常磁性無機金属酸化物核結晶及び有機皮膜を含む 、コントラスト有効量の複合粒子を含有し、前記コア粒子が平均粒径2〜10n mであり、前記粒子の平均粒径は、最高30nmで、前記皮膜が、酸化により開 裂したデンプンを含む造影剤組成物。 34.前記皮膜が、更に官能化ポリアルキレンオキシドを含有する、請求項33 に記載の組成物。 35.前記コア結晶の平均粒径が4〜8nmであり、前記粒子の平均粒径が最高 15nmである請求項33及び34のいづれかに記載の組成物。 36.人体又は非人体内に、コントラスト増強画像を形成する方法であり、前記 方法が、請求項32〜35のいずれかに記載の組成物を前記体に投与し、前記コ ントラスト剤が分布している前記体の最小部の画像を形成することを含む。 37.人体又は非人体内にコントラスト剤の分布測定方法であり、前記方法は、 請求項32〜35のいずれか一つに記載の組成物を、前記体に投与し、前記体か ら放出されたシグナル又は、前記粒子により調節されたシグナルを検出すること を含む。 38.診断用コントラスト剤組成物を生成するための、請求項1〜31のいずれ か一つに記載の又は、請求項1〜31のいずれか一つにより生成される複合磁気 粒子の使用法であり、前記コントラスト剤組成物は、人体又は非人動物体への前 記組成物の投与を含む診断方法において使用される。
