Claims (20)
1. Molekula nukleinske kiseline koja obuhvaća sekvencu nukleinskih kiselina koja kodira β lanac humanog folikulo stimulirajućeg hormona (FSH), a to je kodirajuća regija sekvence nukleinskih kiselina prema SEQ ID No. 1.
2. Rekombinantna molekula nukleinske kiseline koja ima prvu sekvencu nukleinskih kiselina prema zahtjevu 1 pod kontrolom promotora koji je aktivan u stanici domaćinu.
3. Rekombinantna molekula nukleinske kiseline prema zahtjevu 2, dalje ima drugu sekvencu nukleinskih kiselina koja kodira α lanac humanog folikulo stimulirajućeg hormona (FSH), a izabire se iz grupe koja obuhvaća kodirajuću regiju sekvence nukleinskih kiselina prema SEQ ID No. 2 i sekvencu nukleinskih kiselina čija je jednakost sekvence najmanje 85% u odnosu na kodirajuću regiju sekvence nukleinskih kiselina kako je opisano za SEQ ID No. 2.
4. Rekombinantna molekula nukleinske kiseline prema zahtjevu 2, dalje ima drugu sekvencu nukleinskih kiselina koja se izabire iz grupe koja obuhvaća kodirajuću regiju sekvence nukleinskih kiselina prema SEQ ID No. 3 i sekvencu nukleinskih kiselina čija je jednakost sekvence najmanje 70% u odnosu na kodirajuću regiju sekvence nukleinskih kiselina kako je opisano za SEQ ID No. 3.
5. Rekombinantna molekula nukleinske kiseline prema zahtjevu 3 ili 4, gdje je druga sekvenca nukleinskih kiselina pod kontrolom posebnog promotora.
6. Rekombinantna molekula nukleinske kiseline prema bilo kojem od zahtjeva 2 do 5, gdje je prva sekvenca nukleinskih kiselina i/ili druga sekvenca nukleinskih kiselina pod kontrolom virusnog promotora.
7. Rekombinantna molekula nukleinske kiseline prema zahtjevu 6, gdje je prva sekvenca nukleinskih kiselina pod kontrolom SV40 promotora.
8. Rekombinantna molekula nukleinske kiseline prema zahtjevu 6, gdje je druga sekvenca nukleinskih kiselina pod kontrolom CMV promotora.
9. Rekombinantna molekula nukleinske kiseline prema bilo kojem od zahtjeva 2 do 8, ima sekvencu nukleinskih kiselina kako je prikazano za SEQ ID No. 7.
10. Stanica domaćin koja sadrži rekombinantnu molekulu nukleinske kiseline prema bilo kojem od zahtjeva 3 do 9.
11. Stanica domaćin prema zahtjevu 10, koja je stanica sisavaca.
12. Stanica domaćin prema zahtjevu 10 ili 11, koja je stanica jajnika kineskog hrčka (CHO).
13. Stanica domaćin prema bilo kojem od zahtjeva 10 do 12, ima depozitni broj DSM ACC2833.
14. Stanica domaćin koja sadrži prvu rekombinantnu molekulu nukleinske kiseline prema zahtjevu 2 i drugu rekombinantnu molekulu nukleinske kiseline sa sekvencom nukleinskih kiselina koja kodira α lanac humanog folikulo stimulirajućeg hormona (FSH), a izabire se iz grupe koja obuhvaća kodirajuću regiju sekvence nukleinskih kiselina prema SEQ ID No. 2, sekvencu nukleinskih kiselina čija je jednakost sekvence najmanje 85% u odnosu na kodirajuću regiju sekvence nukleinskih kiselina kako je opisano za SEQ ID No. 2 i sekvencu nukleinskih kiselina kako je opisano za SEQ ID No. 3.
15. Stanična kultura koja sadrži stanicu domaćina prema bilo kojem od zahtjeva 10 do 14 u prikladnom kulturnom mediju.
16. Postupak proizvodnje rekombinantnog humanog FSH, koji obuhvaća korake:
- uzgoj kulture stanica domaćina prema bilo kojem od zahtjeva 10 do 14 u prikladnom kulturnom mediju; i
- ubiranje supernatanta stanične kulture.
17. Postupak prema zahtjevu 16, dalje obuhvaća korak pročišćavanja rekombinantnog humaog FSH iz supernatanta stanične kulture.
18. Postupak proizvodnje stanica domaćina prema bilo kojem od zahtjeva 10 do 13, obuhvaća transfekciju stanica, u kulturi suspenzije pod uvjetima bez prisutnosti seruma, rekombinantnom molekulom nukleinske kiseline prema bilo kojem od zahtjeva 3 do 9.
19. Postupak proizvodnje stanica domaćina prema zahtjevu 14, obuhvaća transfekciju stanica, u kulturi suspenzije pod uvjetima bez prisutnosti seruma, prvom rekombinantnom molekulom nukleinske kiseline prema zahtjevu 2 i drugom rekombinantnom molekulom nukleinske kiseline koja obuhvaća sekvencu nukleinskih kiselina koja kodira α lanac humanog folikulo stimulirajućeg hormona (FSH), a izabire se iz grupe koja obuhvaća kodirajuću regiju sekvence nukleinskih kiselina prema SEQ ID No. 2, sekvencu nukleinskih kiselina čija je jednakost sekvence najmanje 85% u odnosu na kodirajuću regiju sekvence nukleinskih kiselina kako je opisano za SEQ ID No. 2 i sekvencu nukleinskih kiselina kako je opisano za SEQ ID No. 3.
20. Uporaba sekvence nukleinskih kiselina prema SEQ ID No. 1 ili obje sekvence nukleinskih kiselina prema SEQ ID No. 1 i 2 za in vitro proizvodnju humanog rekombinantnog FSH.
