Abstract

PROCEDIMIENTO PARA PRODUCIR INTERFERON DE FIBROBLASTO HUMANO. CONSISTE EN OBTENER LA PURIFICACION DEL INTERFERON MEDIANTE LA COMBINACION DE ETAPAS DE CROMATOGRAFIA DE AFINIDAD Y CROMATOGRAFIA DE LIQUIDO DE ALTA PRESION. EN LA PRIMERA ETAPA SE HACE PASAR UNA SOLUCION ACUOSA DE INTERFERON DE FIBROBLASTO HUMANO EN UN ESTADO IMPURO A TRAVES DE UNA COLUMNA DE CROMATOGRAFIA DE AFINIDAD, QUE ABSORBE EL INTERFERON Y SE ELUYE DE LA MISMA EN FRACCIONES SELECCIONADAS DE MAS PUREZA. ESTAS FRACCIONES SE HACEN PASAR EN UNA SEGUNDA ETAPA A TRAVES DE UNA O MAS COLUMNAS DE MATRIZ DE SILICE ENLAZADO A HIDROCARBURO CON TAMPON BAJO CONDICIONES DE CROMATOGRAFIA DE LIQUIDO DE ALTA PRESION, PARA ABSORVER Y ELUIR A CONTINUACION EL INTERFERON, OBTENIENDOLO COMO UNA SOLA CRESTA DISTINTIVA EN FRACCIONES SELECCIONADAS DE DICHO ELUATO EN EL ESTADO DE UNA PROTEINA HOMOGENEA.
