Abstract

Гематоэнцефалический барьер (ГЭБ) препятствует прохождению молекул массой более 500 Да из крови в мозг. Рецепторно-опосредованный трансцитоз (RMT) облегчает прохождение через ГЭБ специфических молекул, которые связывают рецепторы на эндотелиальных клетках мозга, образующих ГЭБ. Обнаружены антитело или его фрагмент, связывающие рецептор инсулиноподобного фактора роста 1, которые проходят ГЭБ посредством RMT. Антитело или фрагмент используют для доставки молекулы груза через ГЭБ, где молекула груза может представлять собой терапевтический или детектируемый агент. Антитело представляет собой верблюжий VH, полученный иммунизацией ламы полипептидом IGF1R, состоящим из 933 аминокислот. Гуманизированные формы верблюжьего VH также получены.
