Claims (13)
- ENZYMBEHANDLING AF CHLOROPHYLLI PLANTEOLIER1. Fremgangsmåde til behandling af en planteolie, hvilken fremgangsmåde omfatter et trin med etablering af kontakt mellem olien og et enzym, hvor enzymet er i stand til at hydrolysere en a’ eller b’ stereoisomer af chlorophyll eller et chlorophyllderivat, og hvor enzymet omfatter et polypeptid med mindst 90 % sekvensidentitet med SEQ ID NO: 13.
- 2. Fremgangsmåde ifølge krav 1, hvor a’ eller b’ stereoisomeren omfatter chlorophyll a \ pheophytin a \ chlorophyll b’ eller pheophytin b’.
- 3. Fremgangsmåde ifølge krav i eller krav 2, hvor enzymet er i stand til at hydrolysere en a’ stereoisomer af chlorophyll eller chlorophyllderivatet.
- 4. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor enzymet har et aktivitetsforhold for (a) en a stereoisomer af chlorophyll eller et chlorophyllderivat sammenlignet med en a' stereoisomer af chlorophyll eller chlorophyllderivatet; eller (b) en b stereoisomer af chlorophyll eller et chlorophyllderivat sammenlignet med en b' stereoisomer af chlorophyll eller chlorophyllderivatet; på mindre end 10.

Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor olien efter behandling med enzymet omfatter (a) mindst 50 % a stereoisomerer af chlorophyll eller chlorophyllderivatet, baseret på den totale mængde af a og a’ stereoisomerer af chlorophyll eller chlorophyllderivatet i olien; eller (b) mindst 50 % b stereoisomerer af chlorophyll eller chlorophyllderivatet, baseret på den totale mængde af b og b' stereoisomerer af chlorophyll eller chlorophyllderivatet i olien.
- 6. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor enzymet har et aktivitetsforhold for pheophytin sammenlignet med pyropheophytin på mindre end 10.
- 7. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor enzymet omfatter en chlorophyllase-, pheophytinase- og/eller pyropheophytinaseaktivitet.
- 8. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor enzymet omfatter aminosekvensen GHSRG (SEQ ID NO: 32).
- 9. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor enzymet er afledt af Ricinus communis.
- 10. Anvendelse af et enzym, der er i stand til at hydrolysere chlorophyll eller et chlorophyllderivat, for fjernelse af en a’ eller b’ stereoisomer af chlorophyll eller chlorophyllderivatet fra en planteolie, hvor enzymet har en hydrolytisk aktivitet på mindst 1000 E/g, baseret på aktivitetsenhederne pr. gram af det oprensede enzym, på en prime-stereoisomer af chlorophyll eller et chlorophyllderivat, og hvor en enzymaktivitetsenhed defineres som mængden af enzym, der hydrolyserer en mikromol substrat pr. minut ved 40 °C, hvor assayet til måling af hydrolytisk aktivitet er følgende: a) 170 μΐ 50 mM HEPES, pH 17 tilsættes til 20 μΐ 0,3 mM chlorophyll, pheophytin eller pyropheophytin, der er opløst i acetone; b) enzymet opløses i 50 mM HEPES, pH 7; c) 10 μ 1 enz.ymopløsning tilsættes til 190 μΐ substratopløsning for at initiere reaktionen og inkuberes ved 40 °C i forskellige tidsrum; d) reaktionen stoppes ved tilsætning af 350 μΐ acetone; e) efter centrifugering i 2 min ved 18.000 g analyseres den ovenstående væske ved hjælp af HPLC og mængderne af (i) chlorophyll og chlorophyllid (ii) pheophytin og pheophorbid eller (iii) pyropheophytin og pyropheophorbid bestemmes; hvor enzymet omfatter et polypeptid med mindst 90 % sekvensidentitet med SEQ ID NO: 2 eller SEQ ID NO: 13.
- 11. Anvendelse ifølge krav 10, hvor enzymet har et aktivitetsforhold for (a) en a stereoisomer af chlorophyll eller et chlorophyllderivat sammenlignet med en a’ stereoisomer af chlorophyll eller chlorophyllderivatet; eller (b) en b stereoisomer af chlorophyll eller et chlorophyllderivat sammenlignet med en b’ stereoisomer af chlorophyll eller chlorophyllderivatet; på mindre end 10.
- 12. Anvendelse ifølge krav 10 eller krav 11, hvor enzymet har et aktivitetsforhold for pheophytin sammenlignet med pyropheophytin på mindre end 10.
- 13. Anvendelse ifølge et hvilket som helst af kravene 10 til 12, hvor a’ eller b’ stereoisomeren omfatter chlorophyll a\ pheophytin a\ chlorophyll b’ eller pheophytin b’.
