Den foreliggende opfindelse angår en antigen, immunogen, proteinholdig vaccine, som er ejendommelig ved det i den kendetegnende del af krav 1 angivne.
I den her omhandlede vaccine indgår et antigen for 5 monoklonale antistoffer, der reagerer specifikt mod mero-zoitter og sporozoitter af parasitten Eimeria tenella, og eventuelt tillige en stabilisator og/eller et pharmaceutisk acceptabelt adjuvans herfor. Disse antistoffer er opnået ved hjælp af hybridomateknologi, nemlig ved hjælp af hybri-10 domakulturer, der frembringer antistoffer mod E. tenella, samt merozoitantigener. Disse antigener er sammen med visse, monoklonale antistoffer effektive til forebyggelse og behandling af coccidiose. Vaccinen ifølge opfindelsen er derfor anvendelig mod coccidiose.
15 Coccidiose er en sygdom hos dyr, der fremkaldes af en række forskellige parasitiske protozoer. Fuglecoccidiose er en ødelæggende fjerkræsygdom, der forårsages af arter af slægten Eimeria. Denne sygdom har en kompliceret cyklus, der omfatter både kønnede og ukønnede stadier. Kyllinger 20 inficeres til at begynde med med lidelsen efter indtagelse af fritlevende oocyster, der i reglen findes i forbindelse med fækalier. Oocyster udvikler sig til invaderende, ukønnede sporozoitter i kyllingens mavetarmkanal. Sporozoitterne inficerer epithelceller og udvikles til flerkernestrukturer, 25 der kaldes schizonter. Hver schizont modnes og frigør efterhånden mange invaderende, ukønnede strukturer, der kaldes merozoitter. Disse merozoitter forlader de inficerede celler og invaderer igen andre epithelceller. De mangfoldige, invaderende, ukønnede stadier, der omfatter sporozoitter og 30 merozoitter, er ansvarlige for meget af coccidiosens patologi. Coccidioses kønnede cyklus begynder, når merozoitterne diffenrentierer til gametocytter. Der sker befrugtning, og produkterne heraf, der kaldes oocyster, frigøres i fæces. Parasittens livscyklus er således sluttet. Hos kyllinger 35 afsluttes cyklussen for Eimeria tenella, der er en repræsentativ art, i løbet af ca. 7-9 døgn.

2 På grund af de kolossale økonomiske tab, som fjerkræindustrien påføres af Eimeriaarter, er en vaccine yderst ønskværdig. Imidlertid er det på grund af parasittens komplekse livscyklus og på grund af den 5 varierende mængde antigener, der er til stede i hvert stadium, ikke tidligere blevet påvist, at deaktiverede eller dra±>te parasitter har danne tilsvarende immunitet.
En løsning på dette problem er at isolere og karakterisere særlige antigener fra parasitten og indgive 10 dem i tilstrækkelig mængde til, at de kan fungere som immuniseringsmiddel. Sådanne antigener vil fortrinsvis yde beskyttelse mod infektion med alle vigtige arter.
Man ved, at forskellige Eimeria-arter samt forskellige stadier i denne arts livscyklus har både fælles og spe-15 cifikke antigener [Z. Cerna, Folia Parasitologica (Prag), 17, 135-140 (1970), Davis m.fl., Immunol. 34, 879-888 (1978), M.E. Rose, Immunol. 2, 112-122 (1959),

Sådanne Eime-20 ria-immunogener vil sandsynligvis yde en vellykket immunisering mod coccidiose.
Udviklingen af lymphocyt-hybridomteknologi udgør et værktøj til frembringelse af forholdsvis store mængder specifikke antistoffer mod en række forskelli-35 ge Eimeria-antigener. Ved at sammensmelte specifikt-an-tistof-producerende celler (miltceller) med celler fra 0 3

en myelomtumor, er det muligt at fremstille hybridomcel-ler, der udskiller monoklone antistoffer rettet specifikt mod det oprindelige sensibiliseringsantigen (Kohier & Milstein, Nature (London) 256, 495-497 (1975)]. Hvis 5 der fås monoklone antistoffer mod parasitten, ville det være muligt at fremstille sådanne antistoffer mod inficeret eller modtageligt fjerkræ og således bibringe værtsorganismen et vist omfang af passiv immunitet.
Når først sådanne hybridomkulturer, der producerer mo-10 noklone antistoffer, er opnået, er det muligt ved hjælp af forskellige metoder at benytte sådanne antistoffer til at isolere og identificere specifikke antigener, der igen kan benyttes som vaccine, så at værtsorganismer får et system af aktiv immunitet. Der kendes en række 15 patentskrifter vedrørende hybridomkulturer og monoklone antistoffer, nemlig USA patentskrifterne nr. 4.172.124, 4.196.265, 4.271.145, 4.361.549, 4.631.550, 4.364.932, 4.364.933, 4.364.934, 4,364,935, 4.364.936, 4.381.292 og 4.381.295.
20 På baggrund af den ovennævnte omtale af coc- cidiosens økonomiske virkninger inden for dyrehold og især fjerkræindustrien, er en bekæmpelse af protozoparasitten yderst påkrævet. Det er således opfindelsens formål at angive nye og anvendelige monoklone antistof-25 fer mod merozoitter af parasitten E. tenella. Endvidere er det opfindelsens formål at isolere og identificere specifikkke E. tenella-antigener, der kan anvendes som vaccine til bekæmpelse af fjerkræcoccidiose. Disse formål fremgår klart af den følgende bskrivelse og 30 de medfølgende krav.
Eimeria tenella, den art, der inficerer coe-cum hos kyllinger, er en særlig ødelæggende parasit, der forårsager svære blødende læsioner hos inficerede dyr.
E. tenella har to merozoit-stadier i sin livscyklus, 35 der betegnes hhv. første og anden generationsmerozoit-ter. Immunitet over for E. tenella, ved man, udvikles 0 4

Der anvendes et præparat af E. Teneralla-mero- 5 zoitter til at immunisere mus, for at de efterhånden kan danne monoklone antistoffer ifølge den nedenfor beskrevne metode. De monoklone antistoffer anvendes til at identificere parasittens antigener. Monoklone antistoffer bedømmes endvidere for at fastslå deres evne til at neu-10 tralisere væksten af parasitten in vivo. Antigener, der fremkalder monoklone antistoffer, der reagerer med mindst én Eimeria-art eller med enten merozoitter eller sporo-zoitter, og udviser neutralisering af parasitvækst, anses for at være beskyttende antigener. Beskyttende anti- ' 15 gener anses som mulige kandidater ved udviklingen af en vaccine mod fuglecoccidiose.
Opfindelsen vil i det følgende blive nærmere beskrevet ved hjælp af eksempler.

Eksempel 1
Kvantitativ radioimmunanalyse
Til at bedømme antistoffers kvalitet udvikles en kvantitativ radioimmunanalyse. Glutaradehyd-fik- serede sporozoitter eller merozoitter (i reglen 2-5 x 10^ 25 pr. fordybning) centrifugeres på polyvinylchlorid med 96 fordybninger eller "Removawell'^-plader (Dynatech) ved 1400 x G i 10 minutter for at binde organismen til bunden af fordybningerne. Sporozoitter eller merozoitter, der hænger fast på pladens bund, omsættes med serumprø-30 ver af mus immuniseret med parasit- eller monoklont antistof i overpå flydende hybridomkultur. Inkubering med antistoffer sker i 16-18 timer ved 4°C eller i to timer ved stuetemperatur. Det bundne antistof påvises med et radioaktivt-mærket andet antistof, 187,1, der er et mo-35 noklont rotteantistof, der er specifikt for muse-k-let-kæde. Radioimmunanalysemetoden anvendes derpå på overflade membraner, der fås fra E. tenella-spo-5 rozoitter, for at sikre antistoffernes reaktivitet i immune musesera med membranproteiner.
Eksempel 2
Opbygning af hybridomkulturer 10 Primære kulturer af kyllingenyreceller infi ceres med frisk excysterede sporozoitter ved hjælp af kendte metoder. Merozoitter, der er frigjort i mediet fem dage efter infektion, høstes ved centrifugering ved 350 x G i 10 minutter. 18-Uger gamle BALB/c-hunmus 7 15 immuniseres intraperitonealt (i.p.) med 1 x 10 intakte eller framenterede E. tenella-merozoitter i komplet Freund's adjuvans. Dyrene igangsættes med to i.p.-injektioner af merozoitter i medium 199 (Gibco) med to mellemrum på 5 uger efter indledende immunisering. Det 20 serum, der fås tre dage efter hver igangsætning, kontrolleres for tilstedeværelse af antistoffer mod E. tenella-merozoitter ved indirekte immunfluorescensanalyse (IFA) og radioimmunanalyse (RIA). Begge disse analyser udføres på glutaraldehyd-fikserede parasitter.

Miltceller fra to mus, der er immuniseret med E. tenella-merozoitter, sammensmeltes 30 med et ikke-sekreterende klon af P3-myelom, P3-x 63.Ag-8,653 i nærvær af 30% polyethylenglycol (PEG 1000 fra Baker Chemical) i 8 minutter. Sammensmeltede celler fordeles i vævskulturskåle med 96 fordybninger (Linbro) og holdes i HAT selektionsmedium [J.W. Littlefield, 35 Science, 145, 709-710 (1964)]. Kendetegnede på 8 anti-merozoit-monoklone an 4 tistoffer er vist i tabel I. Størstedelen af de mono 5 klone antistoffer reagerer med merozoitter samt med spo- 6 rozoitter ved IFA og RIA. Monoklone antistoffer 1,90 7 og 4,76 reagerer ikke med sporozoitter ved IFA og lej 8 lighedsvis ved RIA (jfr. * i tabel I). Monoklont an 9 tistof 15.84 reagerer ikke med merozoitter ved IFA, men 10 reagerer lige godt med sporozoitter og merozoitter ved 11 RIA. De data, der er anført i tabel I, viser, at alle de monoklone antistoffer, der er afledt ud fra anti-merozoit-fusionen er kryds-reaktive med sporozoitter og merozoitter af E. tenella.

De monoklone antistoffer analyseres ved IFA 20 mod sporozoitter afledt ud fra fem fugle-coccidia-para-sitarter, der har økonomisk interesse. Data fra disse forsøg findes i tabel II. 6 monoklone antistoffer, der i IFA omsættes med sporozoitter afledt ud fra E. tenella, anvendes til at undersøges arternes kryds-re-25 aktivitet. En in-vivo-metode til analysering af monoklone antistoffer anvendes til at bedømme de monoklone antistoffer. Tre monoklone antistoffer, der reagerer med E. tenella-sporozoitter, bedømmes ved denne prøve.
20 Frisk isolerede sporozoitter inkuberes under sterile betingelser med varme-inaktiveret ascitesvæske afledt ud fra tre forskellige hybridomer. Inkubationstiden er 1 time ved stuetemperatur. Sporotozitterne indføres derpå i 3-uger gamle kyllingers coecum ad kirurgisk vej.
25 Parasitudviklingen får lov at forløbe i 5 dage efter in-okulering. Ved afslutningen af dette tidsrum gives læsionerne points for at bedømme infektionens omfang. Resultaterne af disse forsøg udtrykkes som procent beskyttelse, som det monoklone antistof yder, og anføres i ta- 30 bel III. Disse data viser, at hvert antistof er i det mindste delvis beskyttende under visse betingelser i dette prøvesystem.

10 o sin-inhiberende enheder/ml). Denne lysemetode omfatter inkubation i 30 minutter ved 0°C efterfulgt af centrifugering ved 2.500 x G i 30 minutter. Pellet'en kasseres, og det opløseliggjorte materiale anvendes derpå 5 til gel-elektrophorese. SDS-Polyacrylamid-gelelektro-phorse af prøver, reduceret med 2-mercaptoethanol udføres i et diskontinuerligt tris-glycinsystem på 7-15% polyacrvlamid-gradientgeler.
De ved SDS PAGE fraskilte proteiner overføres 10 til et nitrocellulose filter elektrophoretisk i 45 minutter. Nitrocellulosefilteres omsættes derpå enten med ascitesvæsken (1-100 fortynding) eller brugt dyrkningsvæske fra hybridomer i 16-18 timer ved 4°C. Normal kaninserum medtages i en koncentration på 10% i al- 15 le inkubationer med antistoffer. Det bundne monoklone 125 antistof påvises med omsætning med et I-mærket kanin--antimuse-IgG-antistof (New England Nuclear). Reaktionen med det andet antistof varer i reglen 3-4 timer ved stuetemperatur. Det ubundne andet antistof fjernes 20 ved vask. Nitrocellulosefiltrene eksponeres derpå med Kodak røntgenfilm XAR-5 eller SB-5.
Alternativt udvikles pletterne ved en ELISA-metode, hvorved der anvendes peberrodsperoxidase-koblet kanin-antimuse-IgG (Miles) og chlornaphthol (Aldrich) 25 og #2®2 som substrater. Proteinantigenernes mo lekylvægt bestemmes i forhold til molekylvægtstandarder.
De antigener, der genkendes af det monoklone antistof 15,84 er illustrerende. Monoklont antistof 15,84 reagerer med en dublet med en omtrentlig molekyl-30 vægt på 130 + 20 kD. Imidlertid er beregningen af proteiners molekylvægt i størrelsesordenen 100 kD underkastet variation afhængigt af de anvendte molekylvægt-standarder. Foruden 130 + 20 kD-arterne reagerer 15,84 også med et bånd, der har en tilsyneladende molekylvægt 35 på 300 + 50 kD. Det største molekylevægt-bånd udgør ca. 15-20% af antigenet og er måske enten et aggregat

Et kontrolforsøg med et monoklont antistof, som man ved reagerer med proteinantigener, anvendes til at bevise anti-merozoit-antistofernes specificitet. Desuden vi-5 ser andre kontrolforsøg, at de monoklone antistoffer er parasit-specifikke og ikke reagerer med det værts--specifikke protein.
I tabel IV findes anført molekvlevægtdata for antigener, der genkendes af forskellige antistoffer.
10 Det signifikante træk er tilstedeværelsen af immungenetiske antigener med en molekylvægt i enten ^T20 kD- eller 100 kD-interval i E. tenerella-merozoitter.

1. Antigen, immunogen, proteinholdig vaccine, kendetegnet ved, at den er opløselig i nærvær af en detergentholdig puffer og indeholder et antigen for Eime- 5 ria tenella-merozoitter, hvilket antigen har en omtrentlig molekylvægt på 300 ± 59 kD og er specifikt reaktivt med antimerozoit-monoklonale antistoffer, der udskilles af hy-bridomakloner 1.90 (ATCC HB8336) og 4.76, eller har en omtrentlig molekylvægt på 130 ± 20 kD og er specifikt reaktivt 10 med anti-merozoit-monoklonale antistoffer, der udskilles af hybridomakloner 10.84 (ATCC HB8387) og 15.84 (ATCC HB8335), eller har en omtrentlig molekylvægt på 18 ± 3 kD og er specifikt reaktivt med anti-merozoit-monoklonale antistoffer udskilt af hybridomakloner nr. 10.08, 8.03 (ATCC HB8388), 15 2.03 (ATCC HB8389), 13.90 (ATCC HB 8337).
