Description
- Gegenstand der vorliegenden Erfindung sind bioverträgliche Fikne und Gele, die aus chemisch modifizierter Hvaluronsäure gebildet werden.
- Hyaluronsäure ("HA") ist ein natürlich vorkommendes Mucqpolysaccharid, das z.B. in Synovialflüssigkeit, in Glaskörperflüssigkeit, in Blutgefäßwänden und in der Nabelschnur und in anderen Bindegeweben vorkormt. Das Polysaccharid bestent aus abwechselnden N-Acetyl-D-glucosamin und D- Glucuronsäureresten, die durch abwechselnde β-1-3-glucuronidische und β- 1-4-glucosaminidische Binnungen verknüpft sind, so daß die Wiederholungseinheit -(1T4)-β-D-GlcA-(1T3)-β-GlcNAc- ist. In Wasser löst sich Hyaluronsäure unter Bildung einer hodiviskosen Flüssigkeit. Das Molekulargewicht von Hyaluronsäure, die von natürlichen Quellen isoliert wird, fällt im allgemeinen in den Bereich von 5 x 10&sup4; bis zu 1 x 10&sup7; Dalton.
- Der hier gebrauchte Begriff "HA" bedeutet Hyaluronsäure und jegliche Hyaluronatsalze einschließlich z.B. Natriumhyaluronat (das Natriumsalz), Kaliumhyaluronat, Magnesiumhyaluronat und Calciumhyaluranat.
- HA in chemisch modifizierter ("derivatisierter") Form ist als ein chirurgisches Hilfsmittel nützlich, um die Verklebung oder die Verwachsung von Körpergeweben während der postoperativen Phase zu verhindern. Das derivatisierte HA-Gel oder der derivatisierte HA-Film wird in den Raum zwischen den Geweben, die getrennt gehalten werden sollen, injiziert oder eingefügt, um deren wechselseitige Verklebung zu verhindern. Um wirksam zu sein, muß das Gel an Ort und Stelle verbleiben und einen Gewebekontakt für einen ausreichend langen Zeitraum verhindern, so daß, wenn das Gel letztlich zerfällt und die Gewebe in Kontakt kommen, sie nicht mehr eine Neigung haben, zu verkleben.
- Chemisch modifizierte HA kann außerdem für die kontrollierte Freisetzung verabreichter Medikamente nützlich sein. Balasz et al., 1986, US- Patent Nr. 4 582 865, stellen fest, daß "quervernetzte HA-Gele die Freisetzung einer Substanz mit niedrigem Molekulargewicht, die im Gel dispergiert ist, allerdings nicht kovalent an die makromolekulare Gelmatrix gebunden ist, erniedrigen können." R.V. Sparer et al., 1983, Kap. 6, S. 107-119, in T.J. Roseman et al., Controlled Release Delivery Systems, Marcel Dekker, Inc., New York, beschreiben eine lang dauernde Freisetzung von Chloramphenicol, das entweder direkt oder in einem Esterkomplex, der eine Alaninbrucke als eine verbindende Bindungsgruppe enthält, kovalent über eine Esterbindung an Hyaluronsäure gebunden ist.
- I. Danishefsky et al., 1971, Carbohydrate Res., Bd. 16, S. 199-205, beschreiben die Modifizierung eines Mucopolysaccharides durch Umwandlung der Carboxylgruppen des Mucopolysaccharides in substituierte Amide durch die Reaktion des Mucopolysaccharides mit einem Aminosäureester in Anwesenheit von 1-Ethyl-3-(3-dimethylaminopropyl)carbodiimidhydrochlorid ("EDC") in wäßriger Lösung. Sie brachten Glycinmethylester mit einer Vielzahl von Polysacchariden einschließlich HA zur Reaktion. Die entstehenden Produkte sind wasserlöslich, d.h. sie dispergieren schnell in Wasser oder in einer wäßrigen Umgebung, so wie sie zwischen Hörpergeweben anzutreffen ist.
- Vorschläge, HA-Zusammensetzungen weniger wasserlöslich werden zu lassen, schließen die Quervernetzung der HA ein. R.V. Sparer et al., 1983, Kap. 6, S. 107-119, in T.J. Roseman et al., Controlled Release Delivery Systems, Marcel Dekker, Inc., New York, beschreiben die Modifikation von HA durch Anheften von Cysteinresten an die HA über Amidbindungen, gefolgt von der Quervernetzung der Cystein-modifizierten HA durch Ausbildung von Disulfidbindungen zwischen den angefügten Cysteinresten. Die Cystein-modifizierte HA selbst war wasserlöslich und wurde erst durch die Quervernetzung durch die Oxidation in die Disulfidform unlöslich.
- De Belder et al., POT-Publikation Nr. WO 86/00912, beschreiben ein langsam abbaubares Gel zur Verhinderung von Gewebeverklebungen infolge von Operationen, das dadurch hergestellt wird, daß ein Carboxyl enthaltendes Polysaccharid mit einem bi- oder polyfunktionalen Epoxid quervernetzt wird. Andere Reaktive bi- oder polyfunktionale Reaktionspartner, die zur Herstellung von quervernetzten HA-Gelen mit verringerter Wasserlöslichkeit vorgeschlagen wurden, beinhalten: 1,2,3,4-Diepoxybutan in alkalischem Medium bei 50ºC (T.C. Laurent et al., 1964, Acta Chem. Seand., Bd. 18, S. 274); Divinylsulfon in alkalischem Medium (E.A. Balasz et al., US-Patent Nr. 4 582 865 (1986); und eine Vielzahl anderer Reagentien einschließlich Formaldehyd, Dimethylolharnstoff, Dimethylolethylenharnstoff, Ethylenoxid, ein Polyaziridin und ein Polyisocyanat (E.A. Balasz et al., GB-Patentanm. Nr. 84 20 560 (1984). T. Mälson et al., 1986, PCT-Publikation Nr. WO 86/00079 beschreiben die Herstellung quervernetzter HA-Gele zur Verwendung als Ersatzstoff für Glaskörperflüssigkeit durch Reaktion von HA mit einem bi- oder polyfunktionalen quervernetzenden Reaktionspartner, wie z.B. einem di- oder polyfunktionalen Epoxid. T. Mälson et al., 1986, EP-A 0 193 510 beschreiben die Herstellung eines geformten Artikels durch Vakuumtrocknen oder Komprimierung eines quervernetzten HA-Gels.
- Erfindungsgemäß wird unter einem ersten Gesichtspunkt ein Verfahren zur Herstellung eines wasserlöslichen bioverträglichen Gels bereitgestellt, umfassend Kombinieren in wäßriger lösung von HA, in einer Konzentration im Bereich zwischen 0,4 % und 2,6 Gew.-%, eines Mittels (im folgenden als "Aktivierungsmittel" bezeichnet) in einer Menge, die wirksam ist, um die Carboxylgruppen von HA gegenüber nucleophilem Angriff in wäßriger Lösung verletzlich zu machen oder ein wasserunlösliches Gel mit einem polyanionischen Polysaccharid zu bilden und eines polyanionischen Polysaccharides unter Bedingungen, die ausreichen, ein Gel zu bilden.
- Erfindungsgemäß wird unter einem zweiten und alternativen Gesichtspunkt ein wasserunlösliches bioverträgliches Gel bereitgestellt, umfassend das Reaktionsprodukt von Hk, ein polyanionisches Polysaccharid und ein Mittel (das im folgenden als "Aktivierungsmittel" bezeichnet wird), das in wäßriger lösung wirksam ist, um die Carboxylgruppen von HA gegenüber nucleophilem Angriff verletzlich zu machen oder ein wasserunlösliches Gel mit polyanionischem Polysaccharid zu bilden.
- Bevorzugte polyanionische Polysaccharide schließen Carboxymethylcellulose ("CMC"), Carboxymethylamylose ("CMA"), Chondroitin-6-sulfat, Dermatinsulfat, Heparin und Heparinsulfat ein; CMC und CMA sind besonders bevorzugt. Die HA und das polyanionische Polysaccharid können zusammen zugegeben werden, gefolgt von der Zugabe des Aktivierungsmittels, oder das polyanionische Polysaccharid kann mit dem Aktivierungsmittel vereinigt werden, gefolgt von der HA-Zugabe. Eine andere Möglichkeit ist die Vereinigung des Aktivierungsmittels und der HA, gefolgt von der Zugabe des polyanionischen Polysaccharids.
- Der bevorzugte pH-Wert, um die Reaktion durchzuführen, ist 4,0 bis 5,0. Die bevorzugte Konzentration des Polysaccharides beträgt 0,005 bis 0,1 M, bevorzugter 0,01 bis 0,02 M. Das molare Verhältnis von Polysaccharid zu Aktivierungsmittel ist vorzugsweise wenigstens 1:1, bevorzugter etwa 1:4. Das bevorzugte Aktivierungemittel ist ein Carbodiimid, z.B. 1- Ethyl-3-(3-dimethylaminopropyl)carbodiimid oder 1-Ethyl-3-(3-dimethylaminopropyl)carbodiimidmethiodid.
- Das Gel kann in Form einer verklebungsverhindernden Zusammensetzung bereitgestellt werden, z.B. in Form einer Membran oder einer Zusammensetzung, die zur Inkorporation in einer Spritze geeignet ist. Es kann auch eine pharmazeutisch aktive Verbindung beinhalten, die im Gel verteilt ist; in einem derartigen Fall ist das Gel als ein System zur Verabreichung von Medikamenten nützlich. Geeignete Substanzen schließen Wachstumsfaktoren, Enzyme, Medikamente, Biopolymere und biologisch kompatible synthetische Polymere ein.
- Der hier verwendete Begriff "Film" bedeutet eine Substanz, die dadurch gebildet wird, daß ein Gel komprimiert wird oder daß man ein Gel dehydrieren läßt oder daß man ein Gel dazu bringt, zu dehydrieren. Aus jedem erfindungsgemäßen Gel kann ein solcher Film gebildet werden.
- Eine "biokompatible" Substanz in der Bedeutung, in der sie hier verwendet wird, ist eine, die keine medizinisch unannehrtibaren toxischen oder verletzende Wirkungen auf biologische Funktionen hat.
- Ein "polyanionisches Polysaccharid" ist ein Polysaccharid, das mehr als eine negativ geladene Gruppe, z.B. Carboxylgruppen, bei pH-Werten über etwa 4,0 enthält.
- Die Erfinder haben gefunden, daß durch Behandiung von HA mit einem geeigneten Aktivierungsmittel und einem polyanionischen Polysaccharid ein Gel oder ein Film hergestellt werden kann, der eine verringerte Wasserlöslichkeit hat, ohne daß irgendein getrennt zugefügtes bi- oder polyfunktionales quervernetzendes Reagens verwendet wird.
- Ein "wasserlösliches" Gel oder ein "wasserlöslicher" Film in der hier verwendeten Bedeutung ist ein Gel/Film, das/der durch Trocknen einer 1 % Gewicht/Gewicht ("Gew./Gew.") Natriumhyaluronat-in-Wasser-lösung entsteht und die Ausmäße 3 cm x 3 cm x 0,3 mm hat und das/der, wenn es/er in ein Becherglas mit 50 ml destilliertem Wasser bei 20ºC gegeben wird und ohne Rühren stehengelassen wird, seine strukturelle Integrität als Film nach 3 min verliert und innerhalb 20 min vollständig dispergiert wird. Ein "wasserunlöslicher" Film in der Bedeutung, in der der Begriff und ähniiche Begriffe hier verwendet werden, wird unter Verwendung einer 1%igen wäßrigen lösung an HA, wie im folgenden genau beschrieben, hergestellt, hat die gleichen Ausmäße und ist nach 20 min, wenn man ihn in ähnlicher Weise ohne Rühren in einem Becherglas mit 50 ml destilliertem Wasser bei 20ºC stehenläßt, strukturell intakt; die Filmgrenzen und Kanten sind auch nach 24 h noch vorhanden, obwohl der Film angeschwollen ist.
- HA wird als "aktiviert" bezeichnet, in der Bedeutung, in der der Begriff hier verwendet wird, wenn sie in einem wäßrigen Gemisch auf eine Art und Weise behandelt wird, bei der die Carboxylgruppen von HA gegenüber nucleophilem Angriff verletzlich gemacht werden oder bei der ein wasserunlösliches Gel mit einem polyanionischen Polysaccharid gebildet wird; und ein "Aktivierungsmittel" ist eine Substanz, die in einem wäßrigen Gemisch, das HA beinhaltet, HA dazu bringt, derart aktiviert zu werden.
- Da die Gele und Filme wasserunlöslich sind, können sie vor der Verwendung gründlich mit Wasser gewaschen werden, um nichtreagierte Substanzen zu entfernen.
- Erfindungsgemäß hergestellte Filme und Gele können auch in gefärbter Form hergestellt werden, dadurch, daß ein Farbstoff oder ein Färbemittel in das Reaktionsgemisch eingeschlossen wird. Derartige gefärbte Filme und Gele können einfacher gesehen werden, während sie an Ort und Stelle gebracht werden, was dazu führt, daß sie während chirurgischer Verfahren einfacher zu handhaben sind als farblose Gele und Filme.
- Die Polysacchsrid-modifizierten Filme und Gele behalten ihre Festigkeit selbst dann bei, wenn sie hydratisiert sind. Da sie an biologische Gewebe anhaften, ohne daß Nähte notwendig sind, sind sie als postoperative verklebungsverhindernde Membranen nützlich. Sie können selbst beim Vorliegen von Blutungen an Gewebe angewendet werden.
- Andere Merkmale und Vorteile der Erfindung werden von der folgenden Beschreibung der bevorzugten Ausführungsformen offensichtlich.
- Polyanionische Polysaccharid-modifizierte HA-Gele und Filme werden im allgemeinen durch Mischen von HA (wie oben beschrieben) mit einem polyanionischen Polysaccharid und einem Aktivierungsmittel unter Bildung eines wasserunlöslichen Materials hergestellt. Der Niederschlag kann in dünne Membranen geformt werden, die zur Verhinderung der postoperativen Verklebung nützlich sind. Es kann ebenso, wie oben beschrieben, gefärbt werden. Um die Festigkeit der Filme, die aus dem Niederschlag geformt wurden, zu erhöhen, können die Filme einer dehydrothermalen Behandlung unterzogen werden, bei der sie im Vakuum (etwa 30 mm Hg) bei etwa 105ºC wäbrend 24 h erhitzt werden.
- Das Polysaccharid und die HA können zusammengemischt werden, wonach das Aktivierungsmittel zugegeben wird. Alternativ kann das Polysaccharid mit dem Aktivierungsmittel zur Reaktion gebracht werden, gefolgt von der Zugabe von HA. Eine dritte Möglichkeit ist es, HA mit dem Aktivierungsmittel zu kombinieren, gefolgt von der Zugabe des Polysaccharids. Bevorzugte Aktivierungemittel wurden oben beschrieben und schließen die Carbodiimide EDC und ETC ein. Die Reaktion wird vorzugsweise bei einem pH zwischen 4 und 5 durchgeführt. Die bevorzugte Polysaccharidkonzentration liegt im Bereich von 0,005 bis 0,1 M und liegt mehr bevorzugt im Bereich von 0,01 bis 0,02 M. Das bevorzugte molare Verhältnis von Polysaccharid zu Aktivierungsmittel ist wenigstens 1:1, mehr bevorzugt etwa 1:4.
- Wird ein gefärbtes Produkt erwünscht, kann dem Reaktionsgemisch eine Farbstoff- oder Färbemittellösung, wie z.B. der blaue Farbstoff "Brillant Blue R", auch als "Coomassie -Brillant Blue R-250" bekannt, und von Serva als "Serva Blue" vertrieben, beigemischt werden. Das entstehende Produkt hat eine blaue Farbe und führt zu einem guten Kontrast im Vergleich zur Farbe der Körpergewebe, was dazu führt, daß der Film oder das Gel während er/es während der Operation gehandhabt wird und wenn er/es schließlich an der richtigen Stelle liegt, leicht zu sehen ist.
- Sobald die Reagentien (und ggf. der Farbstoff oder das Färbemittel) zusairmengemischt wurden, kann das Reaktionsgemisch einfach während einer Zeitspanne stehengelassen werden oder es kann kontinuierlich oder gelegentlich gerührt oder geschüttelt werden.
- Die erfindungsgemäß modifizierte HA kann in unkottplizierter Art und Weise als Film geformt werden. In typischer Weise wird das Reaktionsgemisch in ein Gefäß, welches die gewunschte Größe und Form hat, gegossen, und man läßt es an der Luft trocknen. Im allgemeinen besitzen Filme, die durch Trocknen eines Gemisches, das dick gegossen wurde, hergestellt wurden, so daß sie ein niedrigeres Oberflächen/Volumen-Verhältnis haben, eine größere Festigkeit als Filme, die durch Trocknen dünnerer Gemische mit höherem Oberflächen/Volumenverhaltnis entstanden sind.
- Alternativ kann ein Film dadurch geformt werden, daß ein Gel unter Bedingungen komprimiert wird, die das Entweichen von Wasser erlauben, z.B. durch Komprimieren des Gels zwischen zwei Oberflächen, von denen wenigstens eine porös ist, wie z.B. in EP-A 0 193 510 beschrieben.
- Falls gewünscht, kann ein Gel oder ein Film vor der Verwendung, z.B. durch Perfusion mit Wasser oder 1 M wäßrigem Natriumchlorid, Gewaschen werden. Alternativ kann das Reaktionsgemisch dialysiert werden, um vor dem Gießen des Films restliche Reagentien zu entfernen. Das Waschen zur Entfernung von restlichen Reagentien oder von Reagentien abgeleitetem Material, wie z.B. substituierte Harnstoffe, ist wünschenswert, wenn der Film oder das Gel für therapeutische Anwendungen verwendet werden soll. Gele oder Filme, die, wie oben beschrieben, mit Brillant Blue R blaugefärbt wurden, verlieren ihre Färbung während eines solchen Waschens nicht. Die Entfernung von Reagentien oder Reaktionsprodukten kann durch Hochdruckflüssigchromatographie überwacht werden.
- In den folgenden Beispielen wird die Erfindung detaillierter beschrieben. Diese Beispiele werden zum Zweck der Darstellung gegeben und dienen nicht dazu, die Erfindung einzuschränken.
- Dieses Beispiel illustriert die Herstellung von CMC-modifizierter HA.
- HA (0,4 % Gew. /Gew., 0,01 M) und Aqualon-Typ CMC, das ein Molekulargewicht von 250.000 und einen Substitutionsgrad im Bereich von 0,65 bis 0,90 (0,19 % Gew./Gew., 0,01 M) hat, wurden in einer wäßrigen lösung bei Raumtemperatur zusammengemischt. Der pH-Wert des Gemisches wurde durch Zugabe von 1 M HCl auf einen pH-Wert von 4,7 bis 4,8 eingestellt und auf diesem Wert gehalten. Zu je 100 ml dieser lösung wurden 0,67 g (0,04 M) EDP zugegeben. Während der Reaktion mit EDC wurde der pH der Lösung auf 4,7 bis 4,8 durch Zugabe von 0,1 M HCl gehalten, und man ließ die Reaktion während 1 h stattfinden. Während dieser Zeitspanne entstand ein Präzipitat. Das nichtumgesetzte EDC wurde vom Präzipitat durch Dialyse gegen acidifiziertes Wasser (pH 4,0) während 24 h mit zweimaligem Dialysatwechsel nach 3 und 19 h entfernt. Die HA/CMC-Aufschlämmung wurde dann in flache Gießformen gegossen und während 24 h bei Raumtemperatur luftgetrocknet.
- Es wurde gezeigt, daß HA/CMC-Membranen das Auftreten postoperativer Verklebungsbildung in experimentellen Tiermodellen verminderte. In Experimenten unter Verwendung des Rattenblinddarm-Abschürfungsmodelles wurden HA/CMC-Membranen um operativ abgeschürfte Rattenblinddärme plaziert; vorherige Studien zeigten, daß an Blinddärmen von Ratten, die auf kontrollierte Art und Weise abgeschürft wurden, leicht Verklebungen entstanden. Blinddarmverklebungen in Tiergruppen, die entweder HA/CMC-Membranen oder ORC-Membranen (Interceed TC7-Membranen, von Johnson & Johnson zur Verklebungsvermeidung vermarktet) erhielten, wurden mit Verklebungskontrollen in Tieren, deren Blinddärme abgeschürft wurden, die jedoch keinerlei Membranen erhielten, verglichen. Die Ergebnisse dieser Experimente zeigten, daß die HA/CMC-Membranen in konsistenter Weise die Bildung von Verklebungen im Vergleich zu Kontrolltieren und im Vergleich zu Tieren, die den Interceed TC7-Film erhielten, verringerten.
- In diesem Beispiel wurde die HA-Konzentration um die Hälfte reduziert, um die entstehenden Geleigenschaften zu vergleichen.
- Das Verfähren war wie in Beispiel 1 mit der Ausnahme, daß HA (400 mg; 1,0 mmol Carboxylgruppen) in 30 mi Wasser statt in 15 ml (1-1/3 % Gew./Gew. HA) Wasser gelöst wurde. Ein Hydrogel wurde gebildet, obwohl es schwächer war als das, welches in Beispiel 1 erhalten wurde.
- In diesem Beispiel wurden verschiedene HA-Konzentrationen zur Herstellung von Filmen verwendet, um die entstehenden Filmeigenschaften zu vergleichen.
- Das in Beispiel 2 beschriebene Verfähren wurde unter der Verwendung dreier verschledener anfänglicher HA-Konzentrationen, die durch Lösen der HA (400 mg; 1,0 mmol Carboxylgruppen) in 30 ml, 40 mi oder 100 ml destilliertem Wasser hergestellt wurden, wiederholt. Die Filme, die unter Verwendung jeder dieser Anfangskonzentrationen an HA hergestellt wurden, waren fest und in Wasser und 1 M wäßrigem NaCl unlöslich. Dadurch wurde gezeigt, daß ein Konzentrationsbereich an HA verwendet werden kann. Jeder dieser Filme konnte mit Wasser oder wäßrigem NaCl ohne Verlust seiner physikalischen Eigenschaften gewaschen werden.
- In diesem Beispiel wurden Filme durch Trocknen von dick gegossenen Reaktionsgemischen gebildet, um die Eigenschaften von Filmen zu vergleichen, die durch Trocknen von Gemischen mit unterschledlichem Oberflächen/Volumen hergestellt wurden.
- Ein wie in Beispiel 2 erhaltenes Reaktionsgemisch (40 ml Reaktionsvolumen) wurde in eine kleine Petri-Schale (Fläche 3330 mm²) gegossen. Der so erhaltene Film war in 1 M wäßrigem NaCl und in Wasser (100ºC; 1 h) unlöslich.
- Dieses Beispiel zeigt die Gewebebioverträglichkeit eines Films aus chemisch modifizierter HA.
- Vier Filmstreifen, die gemäß dem in Beispiel 2 beschriebenen Verfähren hergestellt wurden, und zwei USP-negative Kontrollstreifen wurden chirurgisch in den paravertebralen Muskel von White New Zealand-Rabbits (zwei pro Test) implantiert. Die Teststellen wurden entweder makroskopisch nach 72 h oder mit vollständiger Histopathologie nach 7 Tagen ausgewertet. In Übereinstimmung mit USP XXI, S. 1237, erfüllte das Testmaterial die Anforderungen des USP-Implantationstests zur Auswertung von Plastikmaterialien.
- Filme oder Gele, die nach dem erfindungsgemäßen Verfähren hergestellt wurden, können als chlrurgisches Hilfsmittel verwendet werden, um Verklebungen oder Verwachsungen von Körpergeweben infolge von Verfahren, die dem Fachmann auf dem Gebiet der Chirurgie bekannt sind und die z.B. in Debelder et al., PCT-Publikation Nr. WO 86/00912 beschrieben worden sind, während einer postoperativen oder Heilungsphase zu verhindern. Während der Operation werden je nach Bedarf ein oder mehrere Gel- oder Filmstücke zwischen oder über die Gewebe, die getrennt gehalten werden sollen, gelegt oder initiiert.
- Die Filme oder Gele können auch zur Medikamentenverabreichung mit langdauernder Freisetzung verwendet werden. Das zu verabreichende Medikament kann an das Gel oder den Film kovalent gebunden sein, wie z.B. in R.V. Sparer et al., 1983, Kapitel 6, S. 107-119, in T.J. Roseman et al., Controlled Release Delivery Systems, Marcel Dekker, Inc., New York, beschrieben; und das Gel oder der Film kann an dem Ort, an dem die Verabreichung gewünscht ist, implantiert oder injiziert werden.
Claims (16)
1. Verfahren zur Herstellung eines wasserunlöslichen,
bioverträglichen Gels, umfassend Vermischen in wäßriger
Lösung von Hyaluronsäure (HA) in einer Konzentration im
Bereich zwischen 0,4 und 2,6% Gew./Gew. eines Mittels (im
folgenden als "Aktivierungsmittel" bezeichnet) in einer
Menge, die wirksam ist, um die Carboxylgruppen von HA
gegenüber nukleophilem Angriff in wäßriger Lösung verletzlich
zu machen oder ein wasserunlösliches Gel mit einem
polyanionischen Polysaccharid zu bilden, und eines
polyanionischen Polysaccharids unter Bedingungen, die ausreichen, ein
Gel zu bilden.
2. Verfahren nach Anspruch 1, dadurch
gekennzeichnet, daß das polyanionische Polysaccharid
ausgewählt wird aus Carboxymethylcellulose,
Carboxymethylamylose, Chondroitin-6-sulfat, Dermatinsulfat, Heparin und
Heparinsulfat und bevorzugt Carboxymethylcellulose oder
Carboxymethylamylose ist.
3. Verfahren nach Anspruch 1, dadurch
gekennzeichnet, daß die HA und das polyanionische
Polysaccharid zusammengegeben werden und daß darauf die Zugabe
des Aktivierungsmittels folgt.
4. Verfahren nach Anspruch 1, dadurch
gekennzeichnet, daß das polyanionische Polysaccharid mit
dem Aktivierungsmittel vermischt wird und daß darauf die
Zugabe von HA folgt.
5. Verfahren nach Anspruch 1, dadurch
gekennzeichnet, daß die HA mit dem Aktivierungsmittel
vermischt wird und daß darauf die Zugabe des polyanionischen
Polysaccharids folgt.
6. Verfahren nach Anspruch 1, dadurch
gekennzeichnet, daß das Aktivierungsmittel Carbodiimid,
bevorzugt 1-Ethyl-3-(3-dimethylaminopropyl)carbodiimid oder
1-Ethyl-3-(3-dimethylaminopropyl)carbodiimidmethiodid, um-
7. Verfahren nach Anspruch 1, dadurch
gekennzeichnet, daß das polyanionische Polysaccharid in
einer Konzentration von 0,005 bis 0,1M, bevorzugt in einer
Konzentration von 0,01 bis 0,02M, vorhanden ist.
8. Verfahren nach Anspruch 1, dadurch
gekennzeichnet, daß das Verfahren bei einem pH-Wert von
4,0 bis 5,0 durchgeführt wird.
9. Verfahren nach Anspruch 1, dadurch
gekennzeichnet, daß das Molverhältnis von polyanionischem
Polysaccharid zu Aktivierungsmittel mindestens 1:1 und
bevorzugt etwa 1:4 beträgt.
10. Wasserunlösliches, bioverträgliches Gel, umfassend das
Reaktionsprodukt von HA, ein polyanionisches Polysaccharid
und ein Mittel (das im folgenden als "Aktivierungsmittel"
bezeichnet wird), das in wäßriger Lösung wirksam ist, um die
Carboxylgruppen von HA gegenüber nukleophilem Angriff
verletzlich zu machen oder ein wasserunlösliches Gel mit
polyanionischem Polysaccharid zu bilden.
11. Gel nach Anspruch 10, dadurch
gekennzeichnet, daß das Aktivierungsmittel Carbodiimid, bevorzugt 1-
Ethyl-3-(3-dimethylaminopropyl)carbodiimid oder 1-Ethyl-3-(3-
dimethylaminopropyl)carbodiimidmethiodid, umfaßt.
12. Gel nach Anspruch 10, dadurch
gekennzeichnet, daß es weiter eine pharmazeutisch aktive Substanz,
die in dem Gel dispergiert ist, umfaßt.
13. Gel nach Anspruch 12, dadurch
gekennzeichnet, daß die pharmazeutisch aktive Substanz ausgewählt
wird aus Wachstumsfaktoren, Enzymen, Arzneimitteln,
Biopolymeren und biologisch verträglichen synthetischen Polymeren.
14. Gel nach Anspruch 10, dadurch
gekennzeichnet, daß das polyanionische Polysaccharid ausgewählt wird
aus Carboxymethylcellulose, Carboxymethylamylose,
Chondroitin-6-sulfat, Dermatinsulfat, Heparin und Heparinsulfat und
bevorzugt Carboxymethylcellulose oder Carboxymethylamylose
ist.
15. Gel nach Anspruch 10, dadurch
gekennzeichnet, daß das Gel in Form einer Zusammensetzung vorliegt,
die die Adhäsion verhindert.
16. Gel nach Anspruch 15, dadurch
gekennzeichnet, daß die Zusammensetzung in Form einer Membran
vorliegt.
