Description
- Die Erfindung betrifft Zusammensetzungen, die natürliche Pflanzenprodukt-Isolate umfassen, die geeignet sind als Sonnenschutz-Verbindungen für die topische Anwendung auf menschlicher Haut und auf menschlichem Haar, um einen verbesserten Schutz vor den schädigenden Einwirkungen von Sonnenlicht zu liefern.
- Die schädigenden Wirkungen von Sonnenlicht auf menschlicher Haut wurden seit undenklichen Zeiten beobachtet und viele Heilmittel wurden vorgeschlagen, um die Haut vor dieser Schädigung zu schützen.
- Allgemein können die schädlichen Ultraviolett-(UV)-Strahlen, insbesondere solche, die vom Sonnenlicht stammen, die die äußere Atmosphäre durchdringen und die Erdoberfläche erreichen, klassifiziert werden in:
- i. die energiereichen UV-B-Strahlen (290 bis 320 nm Wellenlänge), die eine intensive physiopathologische Aktivität auf der Haut besitzen; diese werden direkt oberhalb der Dermis absorbiert und sind verantwortlich für Erytheme und Hautpigmentierung und
- ii. UV-A-Strahlen (320 bis 400 nm Wellenlänge), die tiefer in die Haut eindringen (zur Dermis und darunter). Deren Energie ist viel geringer und die photobiologischen Effekte, die sie verursachen, dauern in der Natur viel länger, z. B. beschleunigen sie die Alterung der Haut.
- Bestimmte synthetische organische Substanzen (Sonnenschutzmittel), deren Moleküle die schädlichen Ultraviolettstrahlen absorbieren, wurden zur Verwendung vorgeschlagen, um die schädlichen Wirkungen der Ultraviolettstrahlung abzuschwächen. Beispiele schließen ein:
- 2-Hydroxy-4-methoxy-4-methyl-benzophenon, auch bekannt als Benzophenon 10 (CTFA) und erhältlich von Ward Blenkinsop unter dem Handelsnamen UVISTAT 2211;
- 2-Hydroxy-4-n-octyloxybenzophenon, auch bekannt als Benzophenon 12 (CTFA) und erhältlich von American Cyanamid unter dem Handelsnamen CYASORB UV 531;
- 2,2',4,4'-Tetraheptoxybenzophenon, auch bekannt als Benzophenon 2 (CTFA) und erhältlich von BASF Chemical Co. unter dem Handelsnamen UVINUL D50;
- 4-t-Butyl-4'-methoxydibenzoylmethan, auch bekannt als Butyl-methoxy-dibenzoyl-methan (CTFA) und erhältlich von Givaudan Corporation unter dem Handelsnamen PARSOL 1789; und
- 2-Ethylhexyl-p-methoxycinnamat, auch bekannt als Octylmethoxycinnamat (CTFA) und erhältlich von Givaudan Corporation unter dem Handelsnamen PARSOL MCX.
- FR-A-2285853 offenbart als Sonnenschutzmittel 1,3-Diketone, die Di(para-alkoxybenzoyl)methane sind. EP-A2-416564, die nur gemäß Artikel 54(3)EPC relevant ist, offenbart auch 1,3-Diketone als Sonnenschutzmittel.
- Einige dieser Substanzen absorbieren im UV-A-Bereich effektiver, wodurch ein Filtern der UV-Strahlung in diesem Bereich stattfindet, während andere im UV-B-Bereich effektiver sind.
- Ein allgemeines Problem existiert jedoch, welches synthetische organische Sonnenschutzmittel auch immer gewählt wird, zum Schutz vor irgendeiner Wellenlänge der Ultraviolett-Strahlung und dies besteht darin, daß eine physiologische Schädigung des Körpers nach dem topischen Auftragen dieser Sonnenschutzmittel in Mengen, die notwendig sind, um ein wirksames Filtern der schädlichen Ultraviolettstrahlung zu liefern, auftreten kann. Sogar die synthetischen organischen Sonnenschutzmittel, von denen angenommen wird, daß ihre Verwendung sicher ist, haben notwendigerweise Sicherheitsgrenzen, die auf der auf die Haut aufgetragenen Menge beruhen, die zu einem nur mäßigen oder schlechten Schutz vor schädlicher Ultraviolettstrahlung führen.
- Natürliche Hydroxyanthacenpolyglykoside wurden als Sonnenschutzmittel beschrieben (Bader et al., Cosmetics & Toiletries (1981), 96 (10) 67) und die Verwendung von Kaffeeöl als Sonnenschutzmittel wurde berichtet in dem Französischen Patent 2 479 688. Ebenso wird angegeben, daß Extrakte von Brustbeerenfrüchten (JP 84-227813) und Aloeextrakte (US 4,656,029) geeignet sind für Anwendungen auf der Haut.
- Natürliche Extrakte aus Aloe, Faulbaumrinde, Kassie und Kaskara, die Anthrachinon-Derivate enthalten, wurden auch als nützliche Sonnenschutzmittel angegeben (Fox, Cosmetics & Toiletries (1987), 102 (3), (41).
- Diese früheren Beobachtungen, die auf die Verwendung natürlicher Materialien als Sonnenschutzmittel hinweisen, haben nur begrenzte, kommerzielle Nutzung gefunden und keines der Mittel ist so wirksam, wie die vorher erwähnten synthetischen Sonnenschutzmittel.
- EP-A-20274 offenbart die Extraktion eines natürlichen Farbstoffs aus verschiedenen Kurkumaarten. JP-A-61/291515 offenbart entzündungshemmende Kosmetika, die Extrakte aus verschiedenen Pflanzenarten enthalten.
- Es wurde nun eine Reihe substituierter 1,3-Diketone gefunden, die aus natürlichen Quellen isoliert werden können, die Absorptions-Eigenschaften im UV-A- und/oder UV-B-Bereich haben und die als Sonnenschutzmittel in kosmetischen Zusammensetzungen angewendet werden können, die insbesondere geeignet sind für die topische oder äußere Anwendung auf Haut oder Haar. Diese natürlich vorkommenden substituierten 1,3-Diketone können somit anstelle von synthetischen UV-A- und/oder UV-B-Sonnenschutzverbindungen in Hautzusammensetzungen verwendet werden, wie z. B. solchen, auf die vorher Bezug genommen wurde, insbesondere PARSOL 1789.
- Somit liefert gemäß einem Aspekt die Erfindung die Verwendung eines substituierten 1,3-Diketons als Sonnenschutzmittel mit einer Ultraviolett-Absorptionsbande im Bereich von 250 bis 500 nm und einem Extinktions-Koeffizienten ε von 5.000 bis 70.000, worin das 1,3-Diketon die Struktur (1) hat:
- oder dessen Tautomer mit der Struktur (2) ist:
- worin R¹ und R² verschieden sind, wobei die Gruppe R¹ ausgewählt ist aus Benzyliden-, substituiertem Benzyliden-, Phenyl- oder substituierten Phenylgruppen und die Gruppe R² ausgewählt ist aus
- oder worin R¹ und R² identisch sind und beide Gruppen aus den Strukturen (3), (6) oder (7) ausgewählt sind.
- Gemäß einem Aspekt liefert die Erfindung die Verwendung von mindestens 0,01 Gew.-% eines solchen Diketons in einer topischen Zusammensetzung, die einen größeren Anteil eines physiologisch annehmbaren Trägers umfaßt. Gemäß einem weiteren Aspekt liefert die Erfindung eine Zusammensetzung, die das Diketon und einen physiologisch annehmbaren Träger umfaßt.
- Das Sonnenschutzmittel hat ein basisches 1,3-Diketon als Chromophor und vorzugsweise liefern seine auxochromen Gruppen mindestens eine Absorptionsbande in dem Bereich von 280 bis 450 nm und einen Extinktions-Koeffizienten ε von 10.000 bis 60.000.
- Bevorzugte Beispiele für die Gruppe R¹ sind die folgenden Strukturen (3) bis (7):
- Spezifische Beispiele für natürliche Verbindungen zur Verwendung in der erfindungsgemäßen Zusammensetzung sind Kurkumin und seine Analoge Demethoxy- und Didemethoxy-Kurkumin, die aus Kurkuma longa-Wurzeln isoliert werden können und Pongamol, das auch Pongamia glabra-Samen isoliert werden kann.
- Kurkumin ist auch bekannt als Di-4-hydroxy-3-methoxycinnamoyl-methan und hat die Struktur (8):
- Pongamol ist auch bekannt als 1,3-Propandion, 1-(4-Methoxy- 5-benzofuranyl)-3-phenyl und hat die Struktur (9):
- Die in den erfindungsgemäßen Zusammensetzungen verwendeten Verbindungen sind wirksam als Sonnenschutzmittel entweder als reine Verbindungen, als Konzentrat eines geeigneten natürlichen Isolats, das die substituierte 1,3-Diketon-Verbindung umfaßt oder als Rohextrakt des natürlichen Materials oder sogar als Material aus natürlichen Quellen selbst. Zum Beispiel kann Karanjaöl auf diese Weise als Quelle für Pongamol verwendet werden.
- Die erfindungsgemäße Zusammensetzung umfaßt vorzugsweise 0,01 bis 15 Gew.-% des substituierten 1,3-Diketons.
- Die erfindungsgemäße Zusammensetzung umfaßt auch einen physiologisch und kosmetisch annehmbaren Träger, der als Verdünnungsmittel, Dispersionsmittel oder Träger für andere Materialien, die in der Zusammensetzung vorhanden sind, dient, um so die Verteilung zu erleichtern, wenn die Zusammensetzung auf die Haut und/oder das Haar aufgetragen wird.
- Andere Träger als Wasser können flüssige oder feste Weichmacher, Lösungsmittel, Feuchthaltemittel, Verdickungsmittel und Pulver einschließen. Beispiele für jedes dieser Arten von Träger, die einzeln oder als Mischung eines oder mehrerer Träger verwendet werden können, sind die folgenden:
- Weichmacher wie Stearylalkohol, Glycerinmonoricinoleat, Nerzöl, Cetylalkohol, Isopropylisostearat, Stearinsäure, Isobutylpalmitat, Isocetylstearat, Oleylalkohol, Isopropyllaurat, Hexyllaurat, Decyloleat, Octadecan-2-ol, Isocetylalkohol, Eicosanylalkohol, Behenylalkohol, Cetylpalmitat, Silikonöle wie Dimethylpolysiloxan, Di-n-butylsebacat, Isopropylmyristat, Isopropylpalmitat, Isopropylstearat, Butylstearat, Polyethylenglykol, Triethylenglykol, Lanolin, Kakaobutter, Maisöl, Baumwollsaatöl, Olivenöl, Palmkernöl, Rapsöl, Safloröl, Nachtkerzenöl, Sojabohnenöl, Sonnenblumenöl, Avocadoöl, Sesamöl, Kokosnußöl, Erdnußöl, Rizinusöl, Reisschalenöl, Karanjaöl, acetylierte Lanolinalkohole, Rohvaseline, Mineralöl, Butylmyristat, Isostearinsäure, Palmitinsäure, Isopropyllinoleat, Lauryllactat, Myristyllactat, Decyloleat, Myristylmyristat;
- Treibmittel wie Propan, Butan, Isobutan, Dimethylether, Kohlendioxid, Stickoxide;
- Lösungsmittel wie Ethylalkohol, Methylenchlorid, Isopropanol, Aceton, Ethylenglykolmonoethylether, Diethylenglykolmonobutylether, Diethylenglykolmonoethylether, Dimethylsulfoxid, Dimethylformamid, Tetrahydrofuran;
- Pulver wie Kreide, Talkum, Bleicherde, Kaolin, Stärke, Gummi, kolloidales Siliciumdioxid, Natriumpolyacrylat, Tetraalkyl- und/oder Trialkylarylammoniumsmektite, chemisch modifiziertes Magnesiumaluminiumsilikat, organisch modifizierter Montmorillonitton, Aluminiumsilikathydrat, gebranntes Siliciumdioxid, Carboxyvinylpolymer, Natriumcarboxymethylcellulose, Ethylenglykolmonostearat.
- Der physiologisch annehmbare Träger bildet normalerweise 10 bis 99,9 Gew.-%, vorzugsweise 50 bis 99 Gew.-% der Zusammensetzung und kann in Abwesenheit anderer kosmetischer Hilfsmittel den Ausgleich der Emulsion bilden.
- Fakultative, für die Haut nützliche Materialien und kosmetische Hilfsmittel
- Eine besonders geeignete Form der erfindungsgemäßen Zusammensetzung ist eine Emulsion, wobei in diesem Fall normalerweise ein Öl oder öliges Material vorhanden ist zusammen mit einem Emulgator, um entweder eine Wasser-in- Öl-Emulsion oder eine Öl-in-Wasser-Emulsion zu liefern, hauptsächlich abhängig von dem durchschnittlichen hydrophilen-lipophilen Gleichgewicht (hydrophilic lipophilic balance) (HLB) des angewendeten Emulgators.
- Die erfindungsgemäße Zusammensetzung kann gegebenenfalls ein oder mehrere Öle oder andere Materialien mit den Eigenschaften eines Öls umfassen.
- Beispiele für geeignete Öle schließen Mineralöl und Pflanzenöle und Ölmaterialien ein, wie solche, die bereits als Weichmacher vorgeschlagen wurden. Andere Öle oder ölige Materialien schließen Silikonöle, sowohl flüchtige als auch nicht flüchtige, ein.
- Geeignete nicht flüchtige Silikone schließen Polyalkylsiloxane, Polyalkylarylsiloxane oder Mischungen davon ein.
- Beispiele für Polyalkylsiloxane schließen Polydimethylsiloxane mit einer Viskosität von 5 bis 100.000 Centistoke bei 25ºC ein. Diese Siloxane sind im Handel erhältlich von General Electric Company als Viscasil-Reihe und von Dow Corning als 244, 245, 344, 345, 200/5 und 200-Flüssigkeiten. Andere Beispiele schließen die Silikone 7202 und 7518 von Union Carbide und SWS 03314 von Stauffer Chemical ein.
- Die Viskosität kann mit Hilfe eines Glaskapillar-Viskosimeters gemessen werden, wie in dem Testverfahren von Dow Corning Corporate CTM004 vom 20. Juli 1970 ausgeführt.
- Auch geeignet ist Polydiethylsiloxan.
- Beispiele für Polyalkylarylsiloxane schließen Polymethyl- phenylpolysiloxane mit einer Viskosität von 15 bis 65 Centistokes bei 25ºC ein. Diese Siloxane sind im Handel erhältlich von General Electric Company als SF 1075-Methyl- phenyl-Flüssigkeit oder von Dow Corning als 556-Flüssigkeit mit kosmetischer Qualität.
- Geeignete flüchtige Silikone sind cyclische oder lineare Polydimethylsiloxane. Die Anzahl der Siliciumatome in den cyclischen Silikonen ist 3 bis 7, am meisten bevorzugt 4 oder 5. Die allgemeine Formel für cyclische Silikone ist:
- worin n eine ganze Zahl von 3 bis 7 ist. Die Viskositäten sind allgemein geringer als 10 Centipoise (cP) bei 25ºC.
- Lineare Polydimethylsiloxane sind solche mit einer Viskosität von weniger als 5 cP bei 25ºC. Die linearen flüchtigen Silikone enthalten gewöhnlich 3 bis 9 Siliciumatome und haben die allgemeine Formel:
- (CH&sub3;)&sub3; Si-O-[Si (CH&sub3;)&sub2;O]mSi (CH&sub3;)&sub3;
- worin m eine ganze Zahl von 1 bis 7 ist.
- Silikone der oben beschriebenen Arten sind weit verbreitet und Beispiele schließen solche ein, die von Dow Corning als 244-, 245-, 344-, 345-, 200/5- und 200-Flüssigkeiten, von Union Carbide als Silikon 7202 und 7158 und von Stauffer Chemical als SWS-03314 erhältlich sind.
- Das Öl oder ölige Material bildet, falls für die Zwecke zur Bildung einer Emulsion vorhanden, normalerweise bis zu 90 Vol.-%, vorzugsweise 10 bis 80 Vol.-% der Zusammensetzung.
- Es versteht sich, daß das Öl oder ölige Material, wenn es vorhanden ist, auch weichmachende Eigenschaften haben kann und somit auch als ein physiologisch annehmbarer Träger, wie vorher definiert, wirken kann.
- Die erfindungsgemäße Zusammensetzung kann gegebenenfalls auch einen oder mehrere Emulgatoren umfassen, deren Auswahl normalerweise davon bestimmt wird, ob eine Wasser-in-Öl- oder Öl-in-Wasser-Emulsion gebildet wird.
- Wenn eine Wasser-in-Öl-Emulsion erforderlich ist, sollte/n der ausgewählte Emulgator oder die ausgewählten Emulgatoren normalerweise einen durchschnittlichen HLB-Wert von 1 bis 6 haben. Wenn eine Öl-in-Wasser-Emulsion erforderlich ist, sollte/n der ausgewählte Emulgator oder die ausgewählten Emulgatoren einen durchschnittlichen HLB-Wert von mehr als 6 haben.
- Beispiele für geeignete Emulgatoren sind in Tabelle 1 unten angegeben, worin der chemische Name des Emulgators zusammen mit einem Namen, z. B. einem Handelsnamen, unter dem er im Handel erhältlich ist, und der durchschnittliche HLB-Wert angegeben ist. - Die vorhergehende Liste von Emulgatoren gibt nur beispielhaft ausgewählte Emulgatoren an, die zur Verwendung für die Erfindung geeignet sind.
- Es versteht sich, daß zwei oder mehr Emulgatoren angewendet werden können, falls erwünscht.
- Die Menge an Emulgator oder Mischungen davon, die in die erfindungsgemäße Zusammensetzung eingearbeitet wird, ist gegebenenfalls 1 bis 50, vorzugsweise 2 bis 20 und am meisten bevorzugt 2 bis 10 Gew.-%, bezogen auf die Zusammensetzung.
- Die erfindungsgemäße Zusammensetzung kann auch Wasser umfassen, gewöhnlich bis zu 80 %, vorzugsweise 5 bis 80 Vol.-%.
- Die erfindungsgemäße Zusammensetzung kann gegebenenfalls auch ein Silikontensid mit hohem Molekulargewicht enthalten, das auch als Emulgator dienen kann anstelle von oder zusätzlich zu den vorher erwähnten fakultativen Emulgatoren.
- Das Silikontensid ist ein Polymer mit hohem Molekulargewicht von Dimethylpolysiloxan mit Polyoxyethylen- und/oder Polyoxypropylen-Seitenketten mit einem Molekulargewicht von 10.000 bis 50.000 und der Struktur:
- worin die Gruppen R' und R'' jeweils ausgewählt sind aus -H, C&sub1;&submin;&sub1;&sub8;-Alkylgruppen und
- worin
- a einen Wert von 9 bis 115 hat,
- b einen Wert von 0 bis 50 hat,
- x einen Wert von 133 bis 673 hat und
- y einen Wert von 25 bis 0,25 hat.
- Vorzugsweise ist das Dimethylpolysiloxan-Polymer eines, bei dem
- a einen Wert von 10 bis 114 hat,
- b einen Wert von 0 bis 49 hat,
- x einen Wert von 388 bis 402 hat und
- y einen Wert von 15 bis 0,75 hat,
- bei dem eine der Gruppen R' und R'' ein Laurylrest ist und die andere Gruppe ein Molekulargewicht von 1.000 bis 5.000 hat.
- Ein besonders bevorzugtes Dimethylpolysiloxan-Polymer ist eines, bei dem:
- a den Wert 14 hat,
- b den Wert 13 hat,
- x den Wert 249 hat und
- y den Wert 1,25 hat.
- Das Dimethylpolysiloxan-Polymer wird geeigneterweise als Dispersion in einem flüchtigen Siloxan zur Verfügung gestellt, wobei die Dispersion z. B. 1 bis 20 Vol.-% Polymer und 80 bis 99 Vol.-% des flüchtigen Siloxans umfaßt. Idealerweise besteht die Dispersion aus 10 Vol.-% des Polymers, dispergiert in dem flüchtigen Siloxan.
- Beispiele für flüchtige Siloxane, in denen das Polysiloxan- Polymer dispergiert werden kann, schließen Polydimethylsiloxan (Pentamer und/oder Hexamer) ein.
- Ein besonders bevorzugtes Silikontensid ist Cyclomethicone- und Dimethicone-Copolyol, z. B. DC 3225C- Formulierungs- Hilfsmittel, erhältlich von Dow Corning. Ein weiteres ist Laurylmethicone-Copolyol, wie z. B. DC Q2-5200, auch erhältlich von Dow Corning.
- Die Menge an Silikontensid in der Zusammensetzung, falls vorhanden, ist normalerweise bis zu 25, vorzugsweise 0,5 bis 15 Gew.-%, bezogen auf die Emulsion.
- Die erfindungsgemäße Zusammensetzung kann gegebenenfalls ein synthetisches, organisches Sonnenschutzmittel enthalten, zusätzlich zu dem aus den natürlichen Pflanzenquellen erhältlichen substituierten 1,3-Diketon, das vorher definiert wurde, um den Nutzen der Zusammensetzung weiter zu verbessern, indem ein Schutz vor den schädlichen Wirkungen eines übermäßigen Kontaktes mit Sonnenlicht geliefert wird.
- Wie bereits angegeben, können einige synthetische organische Sonnenschutzmittel gesundheitsschädlich sein, wenn sie topisch auf die Haut in einer Konzentration aufgetragen werden, die ausreicht, um wirksam die Strahlung entweder im UV-A-Bereich oder im UV-B-Bereich auszufiltern. Die Gegenwart eines natürlich vorkommenden substituierten 1,3-Diketons, das Schutz im UV-A- und/oder UV-B-Bereich liefern kann, ermöglicht jedoch, eine geringere Menge an synthetischen organischen Sonnenschutzmaterialien als gewöhnlich zu verwenden, um den Gesamt-Sonnenschutzfaktor der Zusammensetzung auf einen außergewöhnlich hohen Wert zu bringen, ohne das Risiko, die Art von Hautschädigung oder anderen Gesundheitsproblemen zu verursachen, die mit der Verwendung von höheren Gehalten an synthetischen, organischen Sonnenschutzmitteln alleine verbunden sein können.
- Im Hinblick darauf kann eine relativ geringe Menge eines synthetischen organischen Sonnenschutzmittels gegebenenfalls in die erfindungsgemäße Zusammensetzung eingearbeitet werden.
- Beispiele für geeignete synthetische, organische Sonnenschutzmittel, falls erforderlich, schließen solche ein, die in Tabelle 2 unten angegeben sind, und Mischungen davon. - Die erfindungsgemäße Zusammensetzung kann somit gegebenenfalls 0,1 bis 10, vorzugsweise 1 bis 5 Gew.-% an synthetischen organischen Sonnenschutzmaterialien enthalten.
- Die erfindungsgemäße Zusammensetzung kann gegebenenfalls auch ein anorganisches Sonnenschutzmittel enthalten.
- Beispiele für anorganische Sonnenschutzmittel schließen ein:
- Titandioxid, mit einer durchschnittlichen Teilchengröße von < 100 nm (auch bekannt als ultrafeines Titandioxid),
- Zinkoxid, mit einer durchschnittlichen Teilchengröße von 1 bis 300 nm,
- Eisenoxid, mit einer durchschnittlichen Teilchengröße von 1 bis 300 nm,
- Siliciumdioxid, z. B. pyrogenes Siliciumdioxid, mit einer durchschnittlichen Teilchengröße von 1 bis 100 nm.
- Es ist anzumerken, daß Siliciumdioxid, wenn es als Inhaltsstoff für die erfindungsgemäße Zusammensetzung verwendet wird, Schutz vor Infrarotstrahlung bieten kann.
- Beispiele für übliche Hilfsmittel, die gegebenenfalls angewendet werden können, schließen Konservierungsmittel, wie z. B. para-Hydroxybenzoatester; Antioxidantien, wie z. B. Butylhydroxytoluol; Feuchthaltemittel, wie z. B. Glycerin, Sorbit, 2-Pyrrolidon-5-carboxylat, Dibutylphthalat, Gelatine, Polyethylenglykol, wie z. B. PEG 200-600; Puffer wie z. B. Milchsäure zusammen mit einer Base, wie z.B. Triethanolamin oder Natriumhydroxid; Wachse, wie z.B. Bienenwachs, Ozokeritwachs, Paraffinwachs; Pflanzenextrakte, zusätzlich zu denen, die ein substituiertes 1,3-Diketon enthalten, die von Pflanzen stammen können wie z. B. Aloe vera, Kornblume, Zaubernuß, Holunder, Gurke, Verdickungsmittel; Aktivitätsverbesserer, Farbstoffe und Parfums ein. Kosmetische Hilfsstoffe können den Ausgleich der Zusammensetzung bilden.
- Die erfindungsgemäße Zusammensetzung ist hauptsächlich vorgesehen als Sonnenschutzprodukt für die topische Anwendung auf menschlicher Haut oder menschlichem Haar, um einen Schutz vor den schädlichen Wirkungen eines übermäßigen Kontaktes mit Sonnenlicht zu liefern.
- Bei der Verwendung wird eine geringe Menge der Zusammensetzung, z. B. 1 bis 5 ml, auf die exponierten Flächen der Haut oder des Haares aus einem geeigneten Behälter oder Applikator aufgetragen und falls notwendig dann über die Haut oder das Haar verteilt und/oder auf der Haut oder dem Haar verrieben unter Verwendung der Hand oder der Finger als geeignete Vorrichtung.
- Die topische Haut- und/oder Haarbehandlungs-Zusammensetzung der Erfindung kann als Lotion formuliert werden mit einer Viskosität von 4.000 bis 10.000 mPa.s, als fließfähige Creme mit einer Viskosität von 10.000 bis 20.000 mPa.s oder als Creme mit einer Viskosität von 20.000 bis 100.000 mPa.s oder darüber. Die Zusammensetzung kann in einem geeigneten Behälter, der der Viskosität angemessen ist und zur Verwendung durch den Verbraucher vorgesehen ist, verpackt werden. Zum Beispiel kann eine Lotion oder fließfähige Creme in eine Flasche oder einen Kugelapplikator oder eine Treibmittel getriebene Aerosol-Vorrichtung oder einen Behälter, der mit einer Pumpe, die zum Betrieb mit dem Finger geeignet ist, verpackt werden. Wenn die Zusammensetzung eine Creme ist, kann sie einfach in einer nicht verformbaren Flasche oder einem zusammendrückbaren Behälter, z. B. einer Tube oder einem Behälter mit Deckel, gelagert werden.
- Die Erfindung liefert somit einen geschlossenen Behälter, der eine kosmetisch annehmbare Zusammensetzung, wie hier definiert, enthält.
- Das Verfahren zur Bestimmung des SPF in vitro für die erfindungsgemäße Zusammensetzung betrifft die spektrophotometrische Abtastung von Stratum corneum zwischen 400 nm und 290 nm unter Verwendung eines Spektrophotometers von Perkin Elmer Modell Lambda 17, das mit einem Detektionssystem für gestreute Transmission ausgestattet ist.
- Stratum corneum von Meerschweinchen wird verwendet anstelle von menschlicher baut und das folgende Verfahren wird durchgeführt:
- i. Stratum corneum von Meerschweinchen wird isoliert in Form von feinen Scheiben aus der Haut von Meerschweinchen und luftgetrocknet.
- ii. Ein Stück des Stratum corneums wird auf die äußere Oberfläche einer 0,5 cm Quarzküvette aufgebracht unter Verwendung eines Tropfens destillierten Wassers, um das Stratum corneum einheitlich auf der Quarzoberfläche abzudichten.
- iii. Die Quarzküvette, die das Stück von Stratum corneum trägt, wird in den Lichtstrahl des Spektrophotometers gestellt, der für diesen Zweck mit einem Fluoreszenz- Kantenfilter ausgestattet ist. Dieser Filter eliminiert die Autofluoreszenz des Stratum corneum und filtert alle Transmissionen oberhalb 400 nm aus.
- iv. Das Stratum corneum wird von 290 bis 400 nm abgetastet und das erhaltene Spektrum wird als Kontrolle aufbewahrt.
- v. Die Küvette mit dem Stratum corneum wird aus dem Spektrophotometer entfernt und das Testmaterial (d.h. Sonnenschutz) wird auf das Stratum corneum mit einer Rate von 1,5 ul/cm² aufgetragen, gemäß dem Deutschen DIN-Protokoll und einheitlich über die gesamte Oberfläche der Haut verrieben, unter Verwendung der Finger, die mit einem Fingerling versehen sind.
- vi. Das aufgebrachte Sonnenschutzmaterial wird fünf Minuten bei Raumtemperatur (20ºC) stehengelassen, um es trocknen zu lassen und dann wird die Probe wieder im Spektrophotometer wie vorher von 290 bis 400 nm abgetastet. Dieses Spektrum wird als Testspektrum aufbewahrt. Es wurden keine Änderungen der Spektralabsorption beobachtet bei Trocknungszeiten zwischen 2 und 15 Minuten; die Trocknungszeit von 5 Minuten wurde daher als Standard übernommen.
- vii. Das Kontrollspektrum wird von dem Testspektrum abgezogen, was die Spektralextinktion der Testprobe des Sonnenschutzmaterials liefert und diese Extinktion wird in die Durchlässigkeit umgewandelt.
- viii. Der in vitro Sonnenschutzfaktor (SPF) wird schließlich berechnet aus den Messungen der Durchlässigkeit wie beschrieben von Diffey et al., in einem Artikel mit dem Titel "A new substrate to measure sunscreen protection factors throughout the ultra-violet spectrum" in J. Soc. Cosmet. Chem 40, 127-133 (Mai/Juni 1989); siehe insbesondere Seite 130.
- Der folgende Versuch bestätigt die Wirksamkeit von zwei natürlich vorkommenden substituierten 1,3-Diketonen als Sonnenschutzmittel im Vergleich mit zwei im Handel erhältlichen synthetischen Sonnenschutzmitteln.
- Bei diesem Versuch wurden Albino-Meerschweinchen am Rücken enthaart. 24 Stunden später wurde eine Harzschablone, die 3 2 cm x 2 cm große Öffnungen enthielt, auf den Rücken der Tiere angelegt und die Öffnungen markiert. Eine Lösung von Pongamol oder Kurkumin mit Konzentrationen im Bereich von 0,25 bis 2 mg/cm² in Alkohol wurde auf die markierten Bereiche aufgetragen. Ein benachbarter Flecken wurde identisch behandelt mit Alkohol allein. 15 Minuten nach dem Aufbringen des Produktes wurde die Schablone wieder angelegt und das Tier wurde mit einer Wotan Ultravitalux UV- Lampe (300 W) aus einer Entfernung von 15 cm 6 Minuten lang bestrahlt. Die Schablone wurde entfernt und das Erythem wurde 4 Stunden nach der Bestrahlung ausgewertet und auf einer Fünfpunktskala eingeteilt. Zum Vergleich wurden die synthetischen Standard-Sonnenschutzmittel, Parsol MCX und 1789 von Givaudan verwendet. Die in diesen Untersuchungen verwendeten Stoffe Kurkumin und Pongamol waren natürlichen Ursprungs und wurden erhalten aus Kurkumawurzel bzw. Karanjasamen. Verbindung Erythem Leerwert Parsol MCX (0,3 mg/cm²) Parsol 1789 (0,5 mg/cm²) Kurkumin (2,0 mg/cm²) Pongamol (0,7 mg/cm²) Pongamol (0,8 mg/cm²)
- Es wurde geschlossen, daß sowohl Kurkumin als auch Pongamol bezüglich des Schutzes vor Erythemen bei Meerschweinchen wirksam waren.
- Die Erfindung wird weiter erläutert unter Bezugnahme auf die folgenden Beispiele.
- Dieses Beispiel zeigt eine Zusammensetzung gemäß der Erfindung, die Pongamol als natürliches substituiertes 1,3-Diketon als Sonnenschutzmittel enthält. Die Ergebnisse sind auch angegeben, die die Wirksamkeit der Zusammensetzung bei Verwendung an einer Reihe von Freiwilligen zeigen.
- Pongamol wurde mit 1,2 Gew.-% und 0,6 Gew.-% in einen Tagescreme-Grundstoff, der 18 Gew.-% Stearinsäure, 0,5 Gew.-% Kaliumhydroxid, 1 Gew.-% Glycerin, 0,2 Gew.-% Konservierungsmittel enthielt, eingearbeitet. Eine Kontrollcreme enthielt kein Pongamol. Eine Harzschablone, die drei 1 cm x 1 cm große Öffnungen enthielt, wurde an den Armen der menschlichen Freiwilligen angelegt und die Öffnungen wurden markiert. Die Creme (5 mg/cm²) wurde auf die markierten Bereiche aufgetragen. 15 Minuten später wurde die Schablone wieder angelegt und bis zu 6 Minuten (abhängig von der Hautfarbe der Freiwilligen) mit einer Wotan Ultravitalux UV-Lampe (300 W) aus einer Entfernung von 15 cm bestrahlt. Die Schablone wurde entfernt und die Bräunung wurde visuell täglich 10 Tage lang ausgewertet. Die Ergebnisse der Auswertung am Ende von 5 Tagen sind in Tabelle 3 unten gezeigt: Tabelle 3 Testperson Bedingungen (Dauer der Bestrahlung) Ergebnisse (abnehmende Sonnenschutz-Wirksamkeit, zunehmendes Bräunen) Minuten linker Arm rechter Arm Kontrolle
- Es wurde gefunden, daß Cremes, die Pongamol enthielten, Schutz vor dem Bräunen bei diesem Versuch lieferten.
- Dieses Beispiel zeigt die Verwendung eines natürlichen substituierten 1,3-Diketons, wenn es zusammen mit einem synthetischen Sonnenschutzmittel in eine Tagescrem-Basiszusammensetzung eingearbeitet wird.
- Pongamol wurde zusammen mit einem bekannten UV-B-Sonnenschutz (Parsol MCX von Givaudan) in einen Tagescreme-Grundstoff eingearbeitet, der 18 Gew.-% Stearinsäure, 0,5 Gew.-% Kaliumhydroxid, 1 Gew.-% Glycerin, 0,2 Gew.-% Konservierungsstoff enthielt. Die Gehalte an Parsol MCX und Pongamol waren 1,0 und 0,7; 1,5 und 0,4; 2,0 und 0,3; 2,5 und 0,3 %. Cremes, die Parsol MCX und Parsol 1789 (von Givaudan) in einem Tagescreme-Grundstoff enthielten, der 18 % Stearinsäure, 0,5 % Kaliumhydroxid, 1,0 % Glycerin, 0,2 % Konservierungsstoff enthielt, wurden formuliert. Die Gehalte an Parsol MCX und Parsol 1789 waren 1,0 und 0,2; 1,0 und 0,5; 1,25 und 0,4; bzw. 2,5 und 0,2 %.
- Der Unterarm von jedem aus einer Gruppe von Freiwilligen wurde mit Seife und Wasser gewaschen und trockengetupft. Eine Harzschablone, die fünf 1 cm x 1 cm große Öffnungen enthielt, wurde an den Unterarmen der Freiwilligen angelegt und die Öffnungen wurden markiert. Das Reflexionsvermögen von der Haut in den markierten Bereichen wurde bestimmt unter Verwendung eines Instruments, das das Reflexionsvermögen quantifizierte aus einem runden Flecken der Haut mit 1 cm Durchmesser. Die verschiedenen Cremes wurden auf die markierten Bereiche aufgetragen (4 mg). 15 Minuten später wurde die Schablone wieder angelegt und die Arme mit einer Wotan Ultravitalux UV-Lampe (300 W) aus einer Entfernung von 15 cm bestrahlt. Die Schablonen wurden entfernt und das Bräunen visuell ausgewertet (subjektive Auswertung) auf einer Punktskala mit 0 bis 8 Punkten und durch Messen des Reflexionsvermögens. Die Ergebnisse am Ende des 5. Tages sind in Tabelle 4 unten angegeben. Alle Werte sind Mittelwerte von verschiedenen unabhängigen Bestimmungen (Anzahl in Klammern). - Aus den obigen Angaben wurde geschlossen, daß Pongamol die Haut vor den Wirkungen der UV-Strahlung in analoger Weise wie der wohlbekannte UV-A-Sonnenschutz Parsol 1789 schützt. Beispiel 3 Dieses Beispiel zeigt eine erfindungsgemäße Lotion. Claims (9)
1. Sonnenschutz-Zusammensetzung, geeignet für die
topische Anwendung auf menschlicher Haut oder
menschlichem Haar zum Schutz vor übermäßigem Kontakt mit
Ultraviolettstrahlen, umfassend:
i. eine wirksame Menge eines substituierten
1,3-Diketons mit einer Ultraviolett-Absorptionsbande
im Bereich von 250 bis 500 nm und einem
Extinktions-Koeffizienten ε von 5.000 bis 70.000 und
ii. einen physiologisch annehmbaren Träger für das
substituierte 1,3-Diketon;
wobei das 1,3-Diketon die Struktur (1):
hat oder dessen Tautomer mit der Struktur (2) ist:
worin R¹ und R² verschieden sind, wobei die Gruppe R¹
ausgewählt ist aus Benzyliden-, substituierten
Benzyliden-, Phenyl- oder substituierten Phenylgruppen und
die Gruppe R² ausgewählt ist aus
oder worin R¹ und R² identisch sind und beide Gruppen
eine Struktur haben, ausgewählt aus den Strukturen
(6) oder (7).
2. Zusammensetzung nach Anspruch 1, worin die Gruppe R¹
ausgewählt ist aus:

der Zusammensetzung bildet.
5. Zusammensetzung nach einem der vorhergehenden
Ansprüche, die in Form einer Creme oder Lotion ist.
6. Verwendung von mindestens 0,01 Gew.-% eines
substituierten 1,3-Diketons mit einer
Ultraviolett-Absorptionsbande im Bereich von 250 bis 500 nm und
einem Extinktions-Koeffizienten ε von 5.000 bis
70.000 als Sonnenschutzmittel in einer topischen
Zusammensetzung, die als Hauptanteil einen
physiologisch annehmbaren Träger für das substituierte 1,3-
Diketon umfaßt, worin das 1,3-Diketon die Struktur

hat oder dessen Tautomer mit der Struktur (2) ist:
worin R¹ und R² verschieden sind, wobei die Gruppe R¹
ausgewählt ist aus Benzyliden-, substituierten
Benzyliden-, Phenyl- oder substituierten Phenylgruppen und
die Gruppe R² ausgewählt ist aus
oder worin R¹ und R² identisch sind und beide Gruppen
ausgewählt sind aus den Strukturen (3), (6) oder (7).
7. Verwendung nach Anspruch 6, worin die Gruppe R¹
ausgewählt ist aus
8. Verwendung nach Anspruch 6, worin das substituierte
Diketon Di-4-hydroxy-3-methoxycinnamoylmethan
