Description
- Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von 3,3-Diarylacrylsäureamiden.
- Die EP-A1-0 120 321, EP-A1-0 208 999 und EP-A1-0 219 756 beschreiben bestimmte 3,3-Diarylacrylsäureamide und verschiedene Verfahren zur Herstellung dieser Verbindungen. Diese Verbindungen zeigen eine fungizide Aktivität und sind besonders nützlich zur Bekämpfung van phytopathogenen Pilzen. Die EP 0 294 907 beschreibt ein anderes Verfahren zur Herstellung derartiger Verbindungen, welches die Umsetzung eines Benzophenons mit einem entsprechenden Acetamid, vorzugsweise in Gegenwart einer starken Base, wie Kalium-tert.butylat, eines Alkalimetallhydroxids oder -carbonats oder von tert.Butyllithium vorsieht. Die letztgenannte Methode führt im allgemeinen aber zu Ausbeuten von weniger als 50 %.
- Es wurde nun gefunden, daß eine überraschende Ausbeutesteigerung auf Ausmaße von über 90 % erzielt werden kann, wenn im Verfahren der EP 0 294 907 ein Natrium-tert.alkoholat als Base verwendet wird. Gemäß der vorliegenden Erfindung wird daher ein Verfahren zur Herstellung eines 3,3-Diarylacrylsäureamids mit der allgemeinen Formel I

- worin Q die oben angeführte Bedeutung aufweist, in Gegenwart einer Base geschaffen, welches Verfahren dadurch gekennzeichnet ist, daß die verwendete Base ein Natrium-tert.alkoholat ist, das im Überschuß vorliegt das Acetamid der Formel III im Überschuß zugegen ist, ein inertes Lösungsmittel verwendet wird und die Reaktionstemperatur im Bereich von 10ºC bis 150ºC liegt.
- Das Alkoholat kann von einem beliebigen tertiären Alkohol abgeleitet sein, gute Ergebnisse wurden jedoch mit einer Verbindung der Formel IV
- (R)&sub3;C-OH (IV)
- erhalten, worin jeder Rest R unabhängig voneinander eine Alkylgruppe mit 1 bis 4 Kohlenstoffatomen bedeutet. Vorzugsweise sind zwei der Gruppen R Methylgruppen und die restliche Gruppe R ist eine Methyl-, Ethyl-, Propyl- oder Butylgruppe. Hervorragende Ergebnisse hinsichtlich der Ausbeute an Verbindungen der Formel I wurden mit Alkoholaten erhalten, die von Amylalkohol und Butylalkohol abgeleitet sind. Das Alkoholat kann in situ hergestellt oder als solches zugesetzt werden und kann nach bekannten Methoden bereitet werden, beispielsweise durch die Umsetzung von Natriumhydrid mit dem tertiären Alkohol in Gegenwart eines inerten Lösungsmittels wie Toluol.
- Das Verfahren gemäß der Erfindung wird in Gegenwart von inerten Lösungsmitteln, wie Toluol, Benzol, Diethylether, Tetrahydrofuran, N,N-Dimethylformamid ausgeführt. Ebenso kann ein Überschuß einer der Reaktionskomponenten als Lösungsmittel dienen. In Abhängigkeit vom Reaktionsvermögen der Komponenten kann die Umsetzung unter Kühlen, bei Raumtemperatur oder bei erhöhter Temperatur bis zur Rückflußtemperatur des Reaktionsgemisches ausgeführt werden. Im allgemeinen erfolgt die Umsetzung bei einer Temperatur im Bereich von 10ºC bis 150ºC.
- Ein 1,5- bis 2-facher Überschuß des Natrium-tert.alkoholats wird bevorzugt. Überschüssiges Acetamid kann zurückgewonnen und wieder in die Umsetzung eingeführt werden.
- Das Verfahren gemäß der Erfindung kann zur Herstellung sämtlicher Verbindungen der allgemeinen Formel I angewendet werden, es ist aber besonders nützlich für Verbindungen der Formel I, worin
- in Gruppe A:
- R&sub1; für C&sub1;&submin;&sub4;Alkoxy, C&sub1;&submin;&sub4;Alkyl oder Amino steht;
- R&sub2; für C&sub1;&submin;&sub4;Alkoxy, C&sub1;&submin;&sub4;Alkyl oder Halogen steht;
- R&sub3; Wasserstoff oder Halogen bedeutet;
- in Gruppe B:
- R&sub4; Wasserstoff oder Halogen darstellt;
- R&sub5; Wasserstoff, Phenyl oder Halogenphenyl bedeutet;
- X eine Einfachbindung oder -0- bedeutet; und
- Q eine Morpholingruppe bezeichnet, worin R&sub8; und R&sub9; beide Wasserstoff sind. Das Halogen oder der Halogensubstituent kann ein Fluor-, Chlor-, Brom- oder Iodatom sein, ist aber vorzugsweise ein Chlor- oder Bromatom.
- In der Gruppe A liegt die Substitution im Phenylring vorzugsweise in den 3- und 4-Stellungen oder in den 3-, 4- und 5- Stellungen vor, wobei besonders bevorzugte Beispiele für eine derartige Substitution 3,4-Dimethoxy, 3-Ethoxy-4-methoxy, 3- Chlor-4-methoxy, 3-Brom-4-methoxy, 3-Methyl-4-methoxy, 3-Ethyl- 4-methoxy, 3-Propyl-4-methoxy, 3,4-Dimethyl, 3-Amino-4-methoxy, 3,5-Dichlor-4-amino und 3-Methoxy-4-methyl sind. Unter diesen wird eine 3,4-Dimethoxy-Substitution des Phenylrings besonders bevorzugt.
- Das Verfahren gemäß der Erfindung wird durch die nachfolgenden Beispiele weiter erläutert.
- 6,92 g (25 mMol) 4-Chlor-3',4'-dimethoxybenzophenon, 5,50 g (50 mMol) Natrium-tert.amylat, 23,90 g (185 mMol) Acetylmorpholin und 50 ml wasserfreies Toluol wurden unter Rückfluß 8 Stunden gerührt. Nach dem Kühlen wurde die Lösung zweimal mit Wasser gewaschen, getrocknet und das Toluol wurde auf einem Rotationsverdampfer abdestilliert. Der viskose Rückstand wurde bei 50ºC mit 40 ml Diisopropylether gerührt, worauf die Substanz erstarrte. Nach dem Abkühlen auf Raumtemperatur wurde das Rohprodukt zerstoßen, durch Absaugen filtriert, mit Diisopropylether gewaschen (2 x 5 ml) und getrocknet.
- Ausbeute: 8,1 g = 83,5 % der Theorie, dünnschichtchromatographisch reines Produkt.
- Natriumhydrid (mit 20 % Paraffinöl; 2,25 g, 75 mMol) wurde unter Rückfluß mit 50 ml absolutem Toluol gerührt und 7,25 g (82,5 mMol) tert.Amylalkohol wurden innerhalb 30 Minuten tropfenweise zugesetzt. Nach dem Akühlen der Lösung auf 100ºC wurden 9,22 g (25 mMol) 4-(4-Chlorphenoxy)-3',4'-dimethoxybenzophenon und 16,17 g (125 mMol) Acetylmorpholin zugesetzt und das Gemisch wurde 8 Stunden unter Rückfluß gerührt.
- Nach dem Kühlen auf Raumtemperatur wurde die Lösung mit Wasser gewaschen (2 x 100 ml), getrocknet und auf eine mit 50 g Silicagel gepackte Säule aufgetragen. Die Produkte wurden mit Toluol (200 ml) und Toluol-Aceton-Gemischen (jeweils 200 ml; 90:10, 80:20, 70:30) eluiert. Die die Substanz mit einem Rf- Wert von 0,47 (Toluol-Aceton 70:30) enthaltenden Fraktionen wurden vereinigt und auf einem Rotationsverdampfer eingeengt.
- Rohausbeute: 11 g viskoses Öl mit einem Gehalt an Toluol.Dieses Öl wurde mit 30 ml Diisopropylether gerührt, worauf die gewünschte Verbindung langsam kristallisierte.
- Ausbeute: 8,7 g = 72,5 % der Theorie
- F.: 115 - 127ºC
- Isomerenverhältnis E/Z: 45/55.
- Natriumhydrid (mit 20 % Paraffinöl; 3,0 g, 100 mMol) wurde mit 70 ml absolutem Toluol gerührt und eine Lösung von 8,15 g (110 mMol) tert.Butanol in 30 ml absolutem Toluol wurde tropfenweise zugesetzt. Sobald die Lösung klar war, wurden 13,84 g (50 mMol) 4-Chlor-3',4'-dimethoxybenzophenon und 19,37 g (150 mMol) Acetylmorpholin zugesetzt und das Reaktionsgemisch wurde 8 Stunden unter Rühren zum Rückfluß erhitzt. Nach dem Abkühlen der Lösung auf Raumtemperatur wurde sie zweimal mit Wasser gewaschen, getrocknet und auf einem Rotationsverdampfer eingeengt. In dieser Weise wurden 24,9 g viskoses Öl erhalten, das langsam beim Rühren in 50 ml Diisopropylether kristallisierte. Die Umkristallisation erfolgte aus Isopropanol/Diisopropylether.
- Ausbeute: 18,0 g = 92,8 % der Theorie
- F.:123 - 145ºC.
Claims (3)
1. Verfabren zur Herstellung eines 3,3-Diarylacrylsäureamids
mit der allgemeinen Formel I
worin A für
steht, B für
steht, Q für
steht, worin
R&sub1; die Bedeutung C&sub1;&submin;&sub4;Alkyl, C&sub1;&submin;&sub4;Alkoxy, NH&sub2;, NHC&sub1;&submin;&sub4;Alkyl,
N(C&sub1;&submin;&sub4;Alkyl)&sub2;, C&sub3;&submin;&sub4;Alkenyl, C&sub3;&submin;&sub4;Alkinyl, C&sub3;&submin;&sub4;-Alkenyloxy,
C&sub3;&submin;&sub4;Alkinyloxy oder C&sub3;&submin;&sub6;Cycloalkyl hat;
R&sub2; für C&sub1;&submin;&sub4;Alkyl, C&sub1;&submin;&sub4;Alkoxy oder Halogen steht;
R&sub3; Wasserstoff oder Halogen bedeutet;
R&sub4; Wasserstoff, Halogen, C&sub1;&submin;&sub4;Alkyl oder C&sub1;&submin;&sub4;Alkoxy bedeutet;
R&sub5; für Wasserstoff, eine gegebenenfalls durch einen oder
mehrere, unter C&sub1;&submin;&sub4;Alkyl-, C&sub1;&submin;&sub4;Alkoxy- und Halogenresten
ausgewählte Substituenten substituierte Phenylgruppe, eine
gegebenenfalls durch ein oder mehrere Halogenatome
substituierte C&sub1;&submin;&sub1;&sub2;Alkylgruppe, eine C&sub3;&submin;&sub7;Cycloalkyl-
Phenylphenyl- oder Phenoxyphenylgruppe, eine C&sub2;&submin;&sub6;Alkenylgruppe
oder C&sub2;&submin;&sub6;Alkinylgruppe die jeweils gegebenenfalls durch
eine Phenylgruppe substituiert sind, oder für eine
Naphthylgruppe oder C&sub5;&submin;&sub8;Cycloalkenylgruppe steht;
X eine einfache Bindung, -O-, S(O)p-, -N=N-, -CHR&sub9;O-,
-OCHR&sub9;-, -CHR&sub9;S(O)p- -S(O)pCHR&sub9;- -(CH&sub2;)n-, -HC=CH- oder
-C C- bedeutet;
R&sub6; für C&sub1;&submin;&sub4;Alkyl, C&sub3;&submin;&sub7;Cycloalkyl, Benzyl, C&sub3;&submin;&sub4;Alkenyl
oder C&sub3;&submin;&sub4;Alkinyl steht;
R&sub7; für C&sub1;&submin;&sub4;Alkyl steht;
R&sub8; Wasserstoff, C&sub1;&submin;&sub4;Alkyl oder C&sub1;&submin;&sub4;Alkoxy bedeutet;
R&sub9; Wasserstoff oder C&sub1;&submin;&sub4;Alkyl darstellt;
p den Wert 0, 1 oder 2 hat; und
n eine ganze Zahl von 1 bis 10 darstellt,
durch Umsetzen eines Benzophenons der Formel II
worin A und B die oben angegebenen Bedeutungen aufweisen,
mit einem Acetamid der Formel III
CH&sub3;COQ (III)
worin Q die oben angeführte Bedeutung aufweist, in
Gegenwart einer Base, dadurch gekennzeichnet, daß die
verwendete Base ein Natrium-tert.alkoholat ist, das im
Überschuß vorliegt, das Acetamid der Formel III im
Überschuß zugegen ist, ein inertes Lösungsmittel verwendet
wird und die Reaktionstemperatur im Bereich von 10ºC bis
50ºC liegt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß das
Natrium-tert.alkoholat von einem Alkohol mit der
allgemeinen Formel IV
(R)&sub3;C-OH (IV)
abgeleitet ist, worin jeder Rest R unabhängig voneinander
eine Alkylgruppe mit 1 bis 4 Kohlenstoffatomen bedeutet.
3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet,
daß das Natrium-tert.alkoholat von Amylalkohol oder
Butylalkohol abgeleitet ist.
