Description
- Die vorliegende Erfindung betrifft neue Methoden zur Herstellung von Anilinofumaraten, um Chinolin-2,3-dicarbonsäuren herzustellen. Diese Säuren sind brauchbare Zwischenprodukte bei der Herstellung von herbiziden Pyridin- und Chinolinimidazolinonverbindungen.
- Die herbiziden Pyridin- und Chinolinimidazolinonverbindungen, die aus den Verbindungen der vorliegenden Erfindung hergestellt werden, umfassen 2-(4-Isopropyl-4-methyl-5-oxo-2-imidazoln-2-yl)chinolin-3-carbonsäure und deren Ester und Salze und sind in U.S.Patent 4 638 068 beschrieben, auf das hiermit Bezug genommen wird. Diese herbiziden Imidazolinylchinolincarbonsäuren können hergestellt werden nach dem Verfahren, das in U.S.Patent 4 518 780 beschrieben ist (auf das hiermit Bezug genommen wird), und zwar durch Cyclisierung, unter basischen Bedingungen, mit einer zweckentsprechend substituierten 2-Carbamoyl-chinolin-3-carbonsäure, die wiederum hergestellt wurde durch die Umsetzung eines substituierten Chinolin-2,3-dicarbonsäureanhydrids und zweckentsprechend substituiertem Aminocarboxamid oder Aminothiocarboxamid. Chinolin- 2,3-dicarbonsäureanhydride werder leicht auf den Disäuren mittels an sich bekannter Verfahrensweisen erhalten. Die Disäuren selbst sind jedoch nicht leicht erhältlich.
- US-A-4656283, angemeldet am 4. Februar 1985, beschreibt eine Methode, die zur Herstellung von Chinolin-2,3-dicarbonsäuren und deren Estern brauchbar ist, und zwar durch Umsetzung eines β-Anilino-α, β-ungesättigten Esters mit einem Immoniumsatz (allgemein als Vilsmeier-Reagenz bezeichnet). De β-α,β- unge-sättigten Ester werden erhalten durch die Umsetzung von zweck-entsprechend substituierten Anilinen mit Ketoestern oder Di-alkylacetylendicarboxylaten. Diese Gesamtreaktion zur Herstel-lung von Chinolin-2,3-dicarboxylaten ist in dem Fließdiagramm I dargestellt. Fließdiagramm I

- Falls R'' für CH&sub3; steht, wird die Disäure erhalten durch gleichzeitige Oxidation und Hydrolyse des Produkts unter wässrigen basischen Bedingungen in Anwesenheit von Nickelperoxid, und zwar wie in U.S.Patent 4 459 409 beschrieben (auf das hiermit Bezug genommen wird).
- Unglücklicherweise ist jedoch die Verfügbarkeit der Ketoester und Dialkylacetylendicarboxylate, wie beispielsweise Diethyloxalacetat und Diethylacetylendicarboxylat, limitiert, was die Mengen an Anilinofumarat und Chinolin-2,3-dicarbonsäuren einschränkt, die als Zwischenprodukte zur Herstellung der herbiziden 2-(4-Isopropyl-4-methyl-5-oxo-2-imidazolin-2-yl)- chinolin-3-carbonsäure, deren Estern und Salzen, erforderlich sind.
- In der anhängigen U.S.Patentanmeldung von D.Maulding, US-A- 4766 218, angemeldet am 29. August 1986, ist ein Verfahren zur Herstellung von Anilinofumaraten beschrieben durch die Umsetzung von Dichlorsuccinaten mit speziellen Aminen und anschließendem Austausch des Amins durch Anilin in Anwesenheit einer organischen Säure.
- In U.S.Patent 4 675 432 ist eine Methode zur Herstellung von Anlinofumaraten beschrieben, bei der Dichlorsuccinate mit Anilin in einem organischen Lösungsmittel in Anwesenheit von wässriger Base und einem Phasentransferkatalysator bei einer Temperatur in einem Bereich von 20 bis 90 ºC während einer bis 24 Stunden umgesetzt werden.
- Die vorliegende Erfindung überwindet nicht nur die Beschränkungen hinsichtlich der Bereitstellung einer leicht verfügbaren Quelle von Chinolin-2,3-dicarbonsäure, Estern und Salzen derselben, sondern schafft auch eine Verbesserung bei der Reaktion von Dichlorsuccinaten mit Anilin, die zu einer signifikant gesteigerten Produktivität und zu einer signifkanten Reduzierung der Reaktionszeiten führt.
- Es ist somit Aufgabe der vorliegenden Erfindung, ein verbessertes Verfahren zur Herstellung von Anilinofumaraten zur Verfügung zu stellen, bei dem Dichlorsuccinate genutzt werden, die aus leicht erhältlichen Dialkylmaleaten erhalten werden können. Es soll somit ein verbessertes Verfahren zur Herstellung von großen Mengen Chinolin-2,3-dicarbonsäure und deren Estern und, daran anschließend, von herbiziden 2-(4- Isopropyl-4-methyl-5-oxo-2-imidazolin-2-yl)-chinolin-2-carbonsäuren, deren Estern und Salzen, vorgeschlagen werden. Diese und andere Aufgaben der vorliegenden Erfindung werden durch die folgende Beschreibung der Erfindung noch deutlicher.
- Erfindungsgemäß wird ein verbessertes Verfahren zur Herstellung von Anilinofumaraten und chinolin-2,3-dicarbonsäuren und deren Estern geschaffen. Das Verfahren umfaßt die Umsetzung eines die Umsetzung eines Dichlorsuccinats (Formel I)
- mit einem molaren Äquivalent von Anilin und einer minimalen Menge von 2 Äquivalenten wässriger Base in Anwesenheit eines Tri- oder Tetraalkylamoniumphasentransferkatalysators bei einer Temperatur in einem Bereich von 85 ºC bis 100 ºC während etwa einer bis 4 Stunden in Abwesenheit eines Lösungsmittels und Isolierung des so gebildeten Anilinofumarats.
- Chinolin-2,3-dicarbonsäure wird anschließend aus dem so gebildeten Anilinofumarat hergestellt durch Umsetzung des Anilinofumarats mit einer etwa äquimolaren Menge eines Vilsmeier Reagenz (Immoniumsalz) in Anwesenheit eines Kohlenwasserstofflösungsmittels, wie beispielsweise Toluol, oder einem chlorierten Kohlerwasserstofflösungsmittel, wie beispielsweise Methylenchlorid, Dichlorethan, Orthodichlorbenzol, Chlorbenzol oder Mischungen derselben bei einer Temperatur von etwa 40 ºC bis 130 ºC während einer Zeitspanne, die ausreicht, um die Reaktion im wesentlichen zu vervollständigen und ein Dialkylchinolin-2,3-dicarboxylat zu liefern. Dieses Chinolin-2,3-dicarboxylat wird hydrolysiert, und zwar entweder unter sauren oder basischen Bedingungen, um Chinolin-2,3-dicarbonsäure zu erhalten. Dieses Verfahren ist in der anhängigen U.S.Patentanmeldung von R. Doehner, US-A-465 6283, angemeldet am 4. Februar 1985, beschrieben. Die obigen Reaktionen werden graphisch in den Fließdiagrammen IIA und IIB erläutert. - wobei R wie bei Formel I beschrieben ist.
- Es hat sich überraschenderweise herausgestellt, daß die Reaktion von Anilin mit Dialkylchlorsuccinaten nach dem erfindungsyemäßen Verfahren, d.h. ohne ein Lösungsmittel und vorzugsweise in Gegenwart eines Tri- oder Tetra-butyl-ammoniumsalz-Phasentransferkatalysator, verringerte Reaktionszeiten zur Folge hat und eine verbesserte Produktivität bei der Bildung von Anilinofumarat.
- Mit dem neuen Verfahren der vorliegenden Erfindung wird somit eine Einfache und effizientere Methode zur Herstellung von Anilinofumarat geschaffen. Diese Methode verringert die Handhabung, die Verarbeitung, das Abwasser und die Lösungsmittel und es resultiert somit ein Verfahren, das sauberer, sicherer, billiger und effizienter ist als die bisher bekannten Verfahren.
- Erfindungsgemäß wird Diethyldichlorsuccinat (0,053 Mol), welches nach der Methode hergestellt werden kann, die im Japanischen Patent 71 21 564 beschrieben ist, mit Anilin (0,053 Mol) umgesetzt, indem man das Gemisch in Gegenwart von 0,08 molaren Äquivalenten wässrigem Natriumcarbonat (20 % Na&sub2;CO&sub3; Gew./Gew.) und einer katalytischen Menge (5 Mol-%) Tetrabutylammoniumbromid bei 95 ºC während 2 Stunden und 30 Minuten rührt. Das so gebildete Anilinofumarat wird leicht isoliert, indem man die organische Phase von der wässrigen Schlammphase abtrennt.
- Wässrige Basen, die für die Herstellung von Anilinofumarat mit dem vorliegenden Verfahren geeignet sind, umfassen Natrium- und Kaliumhydroxid, -carbonat und -bicarbonat mit Konzentrationen von 15 bis 50 Gew.% in Mengen, die ausreichen, um etwa 2 Äquivalente zu schaffen. Die Carbonate werden in Mengen von 1 bis 3 molaren Äquivalenten eingesetzt und die Hydroxide und Bicarbonate werden in Mengen von 2 bis 3 molaren Äquivalenten eingesetzt.
- Die quaternären Ammoniumphasentransferkatalysatoren N-methyl- tri-n-butylammonium- oder Tetra-n-butylammoniumchlorid, -bromid oder -bisulfat haben gezeigt, daß sie in Mengen von so wenig wie 0,025 molarem Äquivalent vergleichbare Ergebnisse liefern und werden für das erfindungsgemäße Verfahen bevorzugt.
- Das Verfahren der vorliegenden Erfindung wird durch die folgenden Beispiele näher erläutert.
- Chlorgas wird in eine Ethylendichloridlösung von Diethylmaleat mit einem Gehalt an Ethanol (0,1 molares Äquivalent) eingesprudelt. Nachdem das Gemisch bei Zimmertemperatur 8 h gerührt wurde, wird mit Stickstoffgas 5 Minuten gespült und das Lösungsmittel wird unter verringertem Druck entfernt. Man erhält das Dichlorsuccinat mit 94 % Ausbeute.
- Tetra-n-butylammoniumbromid (0,9 g, 0,002 Mol-%) wird zu einer 25 %igen wässrigen Lösung von Natriumcarbonat gegeben (8,5 g, 0,08 Mol). Diethyldichlorsuccinat (12,9 g real, 0,053 Mol) wird bei 20 ºC bis 25 ºC zugesetzt, gefolgt von Anilin (4,5 g, 0,0483 Mol). Das Reaktionsgemisch wird bei 95 ºC 30 Minuten erhitzt und bei dieser Temperatur während 1,7 Stunden gehalten. Die Phasen werden bei 50 ºC bis 75 ºC getrennt. Eine Portion des Lösungsmittels, das für die nächste Stufe erforderlich ist, kann zugesetzt werden, um die Geschwindigkeit dieser Phasentrennung zu steigern.
- Die organische Produktphase wird auf 25 ºC abgekühlt und mit 8 bis 10 g 5 %iger Chlorwasserstoffsäure gewaschen, um überschüssiges Anilin zu entfernen. Die Anilinofumarat-Produktschicht wird durch azeotrope Destillation getrocknet.
- Die Rohausbeute (14.7 g) zeigt 63,1 % Anilinofumarat. Dieses bedeutet eine 73 %ige Ausbeute, bezogen auf Anilin als das limitierende Reagenz, oder eine 66,5 %ige Ausbeute, bezogen auf das real eingesetzte Diethyldichlorsuccinat.
- Unter Anwendung der obigen Verfahrensweisen wird Anilin mit Diethyldichlorsuccinat in Gegenwart von Natriumcarbonat mit verschiedenen Phasentransferkatalyatoren umgesetzt und zwar sowohl in Gegenwart als auch in Abwesenheit eines organischen Lösungsmittels bei eetwa 80 bis 95 ºC. Das Produkt wird isoliert und durch Gaschromatographie (GC) analysiert, um die Verbesserung zu demonstrieren die mit dem erfindungsgemäßen Verfahren gegenüber dem bekannten Verfahren erzielt wird. Die Ergebnisse dieser Experimente sind in der Tabelle I zusammengefaßt und zeigen die Verbesserung, die unter Nutzung des erfindungsgemäßen Verfahrens erhalten wird. Claims (3)
1. Verbessertes Verfahren zur Herstellung von
Anilinofumarat wobei Dichlorsuccinat der Formel I
wobei R für C&sub1;&submin;&sub4;-Alkyl steht, umgesetzt wird mit einem
molaren Äquivalent von Anilin unn einem Minimum von 2
Äquivalenten einer wässrigen Base, dadurch gekennzeichnet, daß man die
obige Reaktion in Anwesenheit eines Tri- oder
Tetraalkylammoniumsalz-Phasentransferkatalysators bei einer Temperatur von
etwa 85 ºC bis 100 ºC während 1 bis 4 Stunden in Abwesenheit
eines Lösungsmittels durchführt.
2. Verfahren gemäß Anspruch 1, wobei die wässrige Base 15
bis 50 Gew.%-ig an Natriumhyaroxid, -carbonat oder
-bicarbonat oder Kaliumhydroxid, -carbonat oder -bicarbonat oder
Mischungen derselben, ist.
3. Verfahren gemäß Anspruch 2, wobei der
Phasentransferkatalysator Tetra-n-butylammoniumbromid, -chlorid oder
-bisulfat oder N-Methyl-tri-n-butylammoniumbromid, -chlorid oder
-bisulfat ist.
