- - aktivierter Ester wie Methylthioester, Phenylthioester, p-Nitrophenylthioester, p-Kresylthioester, Carboxymethylthioester, Pyranylester, Pyridylester, Piperidylester, 8-Chinolylthioester oder Ester mit N,N-Dimethylhydroxylamin, 1-Hydroxy-2-(1H)-pyridon, N-Hydroxysuccinimid, N-Hydroxyphthalimid oder 1-Hydroxy-6-chlorbenzotriazol.
- (1) 61,4 g N-(p-Methoxybenzyloxy)-p-toluolsulfonamid wurden zu einer Lösung von Natriumethoxid in absolutem Ethanol (Na: 4,6 g; absoluter C₂H₅OH: 540 ml) hinzugegeben und 1,5 Stunden lang bei 70°C gerührt. Nach Abkühlen auf Raumtemperatur wurde die Mischung mit 121,2 g 1,3-Dibrompropan versetzt und dann 2 Stunden unter Rühren auf Rückflußbedingungen gebracht und filtriert. Das Filtrat wurde unter vermindertem Druck eingeengt und der Rest mit einer Mischung von Ethylacetat und Wasser versetzt. Die organische Schicht wurde abgetrennt, über Magnesiumsulfat getrocknet und unter vermindertem Druck zur Trockne eingedampft unter Erzielung eines Öls, das aus einer Mischung von Ethylacetat und n-Hexan kristallisiert wurde unter Erzielung von 75,1 g N-(3-Brompropyl)- N-(p-methoxybenzyloxy)-p-toluolsulfonamid mit einem Fp von 89,5∼91,5°C.

- (1) 5,76 g einer 50%igen Natriumhydriddispersion
in Mineralöl wurden zweimal mit 200 ml trockenem
Petrolether gewaschen und in 400 ml trockenem Benzol
suspendiert. Zu dieser Suspension wurden 19,4 g Dibutyl
phosphonat unter Rückfluß über 35 Minuten hinweg zugetropft
und die Mischung weitere 2,5 Stunden auf Rück
flußbedingungen gebracht. Danach wurden 23,63 g 1-Brom-
3-chlor-propan hinzugegeben und die Mischung weitere
7 Stunden unter Rückfluß und Rühren aufgeheizt. Nach
dem Abkühlen wurde die resultierende Mischung zweimal
mit 200 ml Wasser gewaschen, über Magnesiumsulfat getrocknet
und unter vermindertem Druck eingeengt unter
Erzielung von 21,15 g Dibutyl-3-chlor-propylphosphonat
in Form einer öligen Substanz.

150°C gerührt. Die resultierende Mischung wurde unter
vermindertem Druck eingeengt unter Erzielung von 77,7 g
Diethyl-3-brompropylphosphonat in Form einer öligen
Substanz.

in Mineralöl wurden zweimal mit 150 ml trockenem Petrolether
gewaschen und in 400 ml Tetrahydrofuran suspendiert.
Die Suspension wurde mit 55,2 g Diethylphosphonat bei
-8 bis -10°C versetzt und dann 1,5 Stunden bei Raum
temperatur gerührt, wonach 126,0 g 1-Brom-3-chlor-propan
zugesetzt und die Reaktionsmischung 4 Stunden lang bei
Zimmertemperatur gerührt wurden. Die resultierende
Mischung wurde mit 50 ml Ethanol vermischt unter Erzielung
von Ausscheidungen, die abfiltriert wurden. Das
Filtrat wurde unter vermindertem Druck eingeengt zur
Abtrennung des Lösungsmittels. Der Rückstand wurde bei
35 bis 40°C unter vermindertem Druck (12 Torr) destilliert,
um 1-Brom-3-chlor-propan abzutrennen. Danach wurde der
Rückstand erneut bei 110 bis 120°C unter vermindertem
Druck (4 Torr) destilliert unter Erzielung von 52,9 g
öligem Diäthyl-3-chlor-propylphosphonat.

- (1)-1: 15,0 g Di-tert.-Butyl-cis-1-propenyl
phosphonat wurden zu einer Lösung von Kalium-tert.-butoxid
in tert.-Butylalkohol (K: 250 mg; tert.-C₄H₉OH:
150 ml) hinzugegeben, wonach die Mischung 6 Stunden lang
bei 55 bis 60°C gerührt wurde. Die resultierende Mischung
wurde unter vermindertem Druck eingeengt und der Rückstand
mit einer Mischung von 400 ml Ethylacetat und
100 ml Eiswasser geschüttelt. Die Ethylacetatschicht
wurde abgesondert, mit 50 ml Wasser gewaschen, über
Magnesiumsulfat getrocknet und unter vermindertem Druck
zur Trockne eingedampft unter Erzielung von 13,34 g
eines öligen Rückstandes, der unter vermindertem Druck
destilliert wurde unter Erzielung von 12 g öligem Di-
tert.-Butyl-trans-1-propenylphosphonat mit einem Kp²
von 78-80°C.

succinimid und dann 1,4 g Dibenzoylperoxid hinzugegeben.
Die Mischung wurde 1 Stunde lang auf Rückflußbedingungen
erhitzt und dann 30 Minuten unter Eis-Kühlung gerührt. Die
resultierende Mischung wurde filtriert und das Filtrat
unter vermindertem Druck zur Trockne eingedampft unter
Erzielung von 17,2 g öligem Di-tert.-butyl-3-brom-trans-
1-propenylphosphonat.

1-propenylphosphonat in 320 ml Tetrachlorkohlenstoff
wurden 41,65 g N-Bromsuccinimid und 2,8 g Dibenzoylperoxid
hinzugegeben. Die Reaktionsmischung wurde 1,5 Stunden
auf Rückflußbedingungen erhitzt und 30 Minuten unter
Eis-Kühlung gerührt. Unlösliche Materialien wurden ab
filtriert und das Filtrat unter vermindertem Druck eingeengt
unter Erzielung von 63,09 g eines öligen Rückstandes,
der einer Säulenchromatographie an Silicagel
unterzogen und mit Chloroform eluiert wurde. Die Eluate
wurden unter vermindertem Druck zur Trockne eingedampft
unter Erzielung von 27,04 g öligem Diethyl-3-brom-trans-
1-propenylphosphonat.

6,81 (1 H; m) - (3) Zu einer Lösung von 6,10 g Dimethyl-cis-1-
propenylphosphonat in 60 ml Tetrachlorkohlenstoff wurden
7,97 g N-Bromsuccinimid hinzugegeben. Die Reaktionsmischung
wurde 2 Stunden lang auf Rückfluß erhitzt und
dann auf Raumtemperatur abgekühlt unter Bildung von
Ausscheidungen, die abfiltriert wurden. Die Filtrate
wurden unter vermindertem Druck eingeengt unter Gewinnung
eines öligen Rückstandes, der einer Säulenchro
matographie an Silicagel unterzogen und mit einer
8 : 2 Mischung von Chloroform und Ethylacetat eluiert wurde
unter Erzielung von 4,94 g Dimethyl-3-brom-trans-1-pro
penylphosphonsäure.

- (1)-1: Zu einer Lösung von 5,34 g Diethyl
allylphosphonat in 107 ml Tetrachlorkohlenstoff wurde
eine Lösung von 5,04 g Brom in 10 ml Tetrachlorkohlenstoff
unter Eis-Kühlung innerhalb von 15 Minuten zugetropft.
Die Reaktionsmischung wurde 2 Stunden bei
Raumtemperatur gerührt. Nach zweimaligem Waschen mit
100 ml 5%iger wäßriger Thiosulfatlösung und dann mit
100 ml Wasser wurde die resultierende Mischung über
Magnesiumsulfat getrocknet und unter vermindertem Druck
zur Trockne eingedampft unter Erzielung von 9,74 g
öligem Diethyl-2,3-dibrompropylphosphonat.

3,50-4,63 (7 H; m) - (1)-2: Zu einer Lösung von 3,34 g Diethyl- 2,3-dibrompropylphosphonat in 10 ml tert.-Butanol wurde eine Lösung von Kalium-tert.-butoxid (K: 430 mg; tert.- C₄H₉OH: 14 ml) innerhalb von 15 Minuten bei Raum temperatur zugetropft. Die Reaktionsmischung wurde 30 Minuten lang bei der gleichen Temperatur gerührt und dann unter vermindertem Druck eingeengt. Der Rückstand wurde mit einer Mischung von 50 ml Ethylacetat und 30 ml Wasser geschüttelt. Die Ethylacetatschicht wurde abgesondert, mit 30 ml Wasser gewaschen, über Magnesiumsulfat getrocknet und unter vermindertem Druck zur Trockne eingedampft unter Erzielung von 2,13 g einer öligen Mischung von isomeren Diethyl-3-brom-propenylphosphonaten. Ein aliquoter Teil davon (1,86 g) wurde durch eine Säulenchromatographie an Silicagel (mit Chloroform als Entwicklungsmittel) in 2 Fraktionen (Fraktion A und Fraktion B) aufgetrennt. Die Fraktion A wurde unter ver mindertem Druck zur Trockne eingedampft unter Gewinnung von 10 mg öligem Diethyl-3-brom-cis-1-propenylphosphonat. Die Fraktion B wurde ebenfalls unter vermindertem Druck zur Trockne eingedampft unter Gewinnung von 1,65 g einer öligen Mischung von Diethyl-3-brom-2-propenylphosphonat und Diethyl-3-brom-trans-1-propenylphosphonat (im Mol verhältnis von etwa 1 : 1).

- (1) Eine Lösung von 40 g Isobutyl-N-hydroxycarbamat
in 400 ml absolutem Ethanol wurde bei ca. 25°C
unter Rühren zu einer Lösung von Natriumethoxid in absolutem
Ethanol (Na: 6,9 g; absoluter C₂H₅OH: 500 ml) zugetropft.
Die Mischung wurde dann innerhalb von 30 Minuten
unter Rühren unterhalb von 30°C tropfenweise mit
60 g p-Methoxybenzylbromid versetzt. Nach Weiterrühren
bei Raumtemperatur für weitere 14 Stunden wurde das
Lösungsmittel unter vermindertem Druck abdestilliert. Der
ölige Rückstand wurde mit 500 ml Wasser versetzt, mit
500 ml Ethylether extrahiert, mit 0,1 n NaOH und mit
Wasser gewaschen, über Magnesiumsulfat getrocknet und
unter vermindertem Druck zur Trockne eingedampft unter
Erzielung von 62 g Öl, das einer Säulenchromatographie
an Silicagel mit einer Mischung von 100 Volumenteilen
Chloroform und 1 Volumenteil Methanol als Elutionsmittel
unterzogen wurde. Die gewünschte Verbindung enthaltende
Fraktionen wurden gesammelt und unter vermindertem Druck
eingeengt unter Erzielung von 20,0 g Isobutyl-N-(p-meth
oxybenzyloxy)carbamat in Form einer öligen Substanz.

- (1) Eine Lösung von 156,7 g Tosylchlorid in 240 ml Pyridin wurde über 2,5 Stunden hinweg unter Kühlung auf 0∼5°C zu einer Lösung von 102,3 g p-Methoxy benzyloxyamin in 210 ml Pyridin zugetropft und die Mischung über Nacht bei Raumtemperatur gerührt. Das Lösungsmittel wurde unter vermindertem Druck abdestilliert und der Rückstand in 1 l Ethylacetat gelöst. Die un löslichen Substanzen wurden abfiltriert und das Filtrat dreimal mit 2 n Salzsäure und zweimal mit Wasser gewaschen, über Magnesiumsulfat getrocknet und unter vermindertem Druck zur Trockne eingedampft unter Erzielung eines kristallinen Produkts, das aus einer Mischung von Ethylacetat und Petrolether umkristallisiert wurde unter Gewinnung von 162,2 g N-(p-Methoxybenzyloxy)-p-toluol sulfonamid in Form von Kristallen mit einem Fp von 109∼111°C.
- (2) 312,8 g Hydroxylamin-hydrochlorid wurden in einer Lösung von 558,0 g Natriumhydroxid in 3,6 l Wasser unter Eis-Kühlung und Rühren gelöst. Zu der Lösung wurden 1025,4 g Ethylchlorformiat über 1,5 Stunden hinweg unter Eis-Kühlung und Rühren zugetropft. Nach 15 Minuten Weiterrühren wurde die Reaktionsmischung zweimal mit Methylisobutylketon (3 l und 1,5 l) extrahiert. Die Extrakte wurden mit 1,5 l Wasser gewaschen und über Magnesiumsulfat getrocknet, das abfiltriert und mit 0,9 l Methylisobutylketon gewaschen wurde. Die Waschflüssigkeiten wurden mit dem wie oben erhaltenen Filtrat vereinigt und die Mischung tropfenweise mit einer Lösung von 265,5 g Kaliumhydroxid in 1,13 l Ethanol versetzt unter Ausscheidung von Kristallen bei Eis-Kühlung (0 bis 5°C) und 40 Minuten Rühren. Nach 30 Minuten Weiterrühren bei der gleichen Temperatur wurden 758,0 g kristallines Ethyl-N-ethoxycarbonyloxycarbamat-mono kaliumsalz mit einem Fp von 169,5-170°C (Zers.) erhalten.
- (1) 4,52 g Butyraldehydoxim wurden bei 5 bis 10°C
zu einer ethanolischen Natriumethoxidlösung (hergestellt
aus 1,17 g Natrium und 100 ml absolutem Ethanol)
hinzugegeben. Die Mischung wurde mit 12,69 g Diethyl-
3-brom-propylphosphonat versetzt und 22 Stunden lang
bei Raumtemperatur gerührt und dann unter vermindertem
Druck zur Trockne eingedampft. Der Rückstand wurde in
Wasser gelöst und mit 20 ml Ethylacetat gewaschen. Die
wäßrige Schicht wurde abgesondert, mit Natriumchlorid
gesättigt und fünfmal mit Chloroform extrahiert. Die
vereinigten Chloroformextrakte wurden mit gesättigter
wäßriger Natriumchloridlösung gewaschen, über Magnesiumsulfat
getrocknet und unter vermindertem Druck zur
Trockne eingedampft unter Erzielung von 7,1 g öligem
Diethyl-3-butylidenaminopropylphosphonat-N-oxid.

6,8 (1 H; t; J = 7 Hz) - (2) 20,67 g Octanaloxim wurden in einer methanolischen
Natriummethoxidlösung (hergestellt aus 2,3 g Natrium
und 100 ml absolutem Methanol) bei 5 bis 10°C gelöst.
Zu der Lösung wurden 25,9 g Diethyl-3-brompropyl
phosphonat zugetropft, wonach die Reaktionsmischung
2 Stunden lang bei Raumtemperatur gerührt und dann
unter Rühren 2 Stunden lang auf Rückfluß erhitzt wurde.
Die resultierende Mischung wurde unter vermindertem Druck
zur Trockne eingedampft und der Rest in Wasser gelöst.
Die wäßrige Lösung wurde mit Natriumchlorid gesättigt
und mit Chloroform extrahiert. Die Chloroformextrakte
wurden über Magnesiumsulfat getrocknet und unter vermindertem
Druck zur Trockne eingedampft unter Erzielung
von 38,9 g öligem Diethyl-3-octylidenaminopropylphosphonat-
N-oxid.

Eine vorgeschriebene Menge pathogener Bakterien (suspendiert in 5%iger wäßriger Mucinlösung (0,5 ml)) wurde den Versuchstieren intraperitoneal injiziert.

- a) eine Verbindung der allgemeinen Formel: worin A die bereits angegebene Bedeutung hat, Ra³ ein Esterrest ist und R⁴ Alkyliden ist, unter Erzielung einer Verbindung der allgemeinen Formel: worin A die bereits angegebene Bedeutung hat, hydrolysiert oder
- b) eine Verbindung der allgemeinen Formel: worin A und Ra³ die bereits angegebene Bedeutung haben, R¹ Wasserstoff oder Acyl bedeutet und R² Wasserstoff, Arylnie deralkyl oder Acyl bedeutet, unter Erzielung einer Verbindung der allgemeinen Formel: worin A die bereits angegebene Bedeutung hat, hydrolysiert oder
- c) eine Verbindung der allgemeinen Formel: worin A und R² die bereits angegebene Bedeutung haben, Ra¹ Acyl bedeutet und R³ Wasserstoff oder einen Esterrest bedeutet, unter Erzielung einer Verbindung der allgemeinen Formel: worin A die bereits angegebene Bedeutung hat, hydrolysiert oder
- d) Hydroxylamin oder ein Salz davon mit einer Verbindung der allgemeinen Formel worin A die bereits angegebene Bedeutung hat und X² ein Säurerest ist, unter Erzielung einer Verbindung der allgemeinen Formel: worin A die bereits angegebene Bedeutung hat, umsetzt.
