Description
Vorliegende Erfindung betrifft ein stabilisiertes pharmazeutisches Mittel mit einem Gehalt an einem
biologisch aktiven Prostaglandin der Ε-Reihe und einem gesättigten, aliphatischen tertiären Alkohol mit 4-10
Kohlenstoffatomen, wobei das Prostaglandin in einer Menge von 0,005 - 0,500 Gew.-% vorliegt.
Die Prostaglandine der Ε-Reibe sind diejenigen,
weiche in der 9-Steiiung eine Carbonyigruppe und der
11-Stellung eine Hydroxygruppe aufweisen. Die Prostaglandine sind ungesättigte Car Fettsäuren, welche
verschiedene physiologische Wirkungen zeigen. Beispielsweise sind die Prostaglandine der E-Reihe
wirksame Vasodilatoren [vgl. Acta Physiol. Scand. Band
64, Seite 332-333 (1965) und Life Sei. Band 6, Seiten
449—455 (1967)] und erniedrigen den systemischen arteriellen Blutdruck (Vasodepression) bei intravenöser
Verabreichung [vgl. Federation Proa Band 23, Seite 327 (1964); Acta PhysioL Scand. aa.C; Acta Med. Scand.
Band 133, Seiten 423-430 (1968) und Acta Physiol. Scand. Band 75, Seiten 161-169 (1969)]. Eine andere
gut bekannte physiologische Wirkung von PGEi und PGE2 ist die bronchodilatorische [vgL BnL Med. J. Band
4, Seiten 723-726 (1969)]. PGEi besitzt ferner die physiologische Wirkung, die Durchgängigkeit der Nase
zu erhöhen [vgL Curr. Ther. Res. Band 12, Seiten
711-717(1970)].
Eine weitere wichtige physiologische Rolle der natürlichen Prostaglandine steht im Zusammenhang mit
dem Fortpflanzungszyklus. PGE2 besitzt bekanntermaßen die Fähigkeit, Wehen einzuleiten [vgl. J. Obstet
Gynaec Brit Cwlth. Band 77, Seiten 200-210 (1970)1 den therapeutischen Abort einzuleiten [vgl. Contraception Band 4, Seite 293 (1971)] und ist brauchbar zur
Steuerung der Fruchtbarkeit [vgL Contraception Band 3, Seite 173 (1971)]. Für mehrere Prostaglandine der
Ε-Reihe als Mittel zur Einleitung von Wehen bei Säugetieren wurden Patente erteilt (BE-PS 7 54 158 und
DE-PS 20 34 641). Darüber hinaus ist eine bekannte physiologische Wirksamkeit von PGFi1 die zeitliche
Abstimmung des östrus bei Haustieren [vgl.}. Animal Sei. Band 36, Seite 1149 (1973) 1
Weitere bekannte physiologische Wirksamkeiten von
PGEi sind die Hemmung der Magensäuresekretion (vgl. Worcester Symp. on Prostaglandins, New York, Verlag
Wiley, 1968, Seiten 55-64) und femer der Blutplättchenaggregation [vgl. Brit. Med. ]. Band 2, Seilen
468-472(1967)].
Die Anwendung von Prostaglandin auf einer
Anzahl von Gebieten wurde jedoch ernsthaft durch ihre Unbeständigkeit, insbesondere in Lösung, behindert. Es
wurde bereits die biologische Wirksamkeit von Kochsalzlosungen mit einem Gehalt an lOOng/ml
PGEi, PGEi und PGFj*. welche bei verschiedenen
pH-Werten bei Raumtemperatur gelagert wurden, gemessen [vgl. Eur. J. Pharmacol. Band 4. Seite 416
(1968)} Die PGE-Verbindungen zeigten nach 60 Tagen
bei einem pH-Wert von 5—7 einen 25—40prozentigen
Verlust an biologischer Wirksamkeit, während die PGF-Vejbindungen noch nach 182 Tagen voll aktiv
waren. Ähnliche Ergebnisse wurden bei der Untersuchung der Stabilität anderer Prostaglandine erhalten.
Die Stabilität anderer Prostaglandine wurde ebenfalls untersucht [vgL J. Lipid. Res. Band 10, Seite 320 (1969)
und J, Pharm. PharmacoL Band 23, Seite 804 (1971)].
to Das Bedürfnis für ein Verfahren zur Lagerung von Lösungen der Prostaglandine der Ε-Reihe während
langen Zeiträumen liegt auf der Hand.
Dieses Bedürfnis wird durch die erfindungsgemäßen Mittel befriedigt
Aus der US-PS 36 44 638 sind Lösungen von Prostaglandin in einem Gemisch von Äthanol und
Fluorkohlenwasserstoffen bekannt und ·α der US-PS
37 49 800 werden Lösungen von PGE2 in einem wasserfreien, mit Wasser mischbaren pharmakologisch
akzeptablen Alkohol, wie Äthanol, 1,3-ButandioI,
Propyiengiykoi und Polyälhyienglykoi vorgeschlagen.
Wie jedoch bei Untersuchungen festgestellt wurde (vgl. z. B. Beispiele 15 und 16 der vorliegenden Patentanmeldung sowie die nachfolgend angegebenen Untersu-
chungsergebnisse), bewirken Äthanol oder sekundäre
von Prostaglandinen der Ε-Reihe in Lösung, sondern
eher einen gegenteiligen Effekt
gesättigte, aliphatische tertiäre Alkohole mit 4 bis 10
Kohlenstoffatomen im Vergleich zu sekundären Alkoholen und Äthanol (vgL US-PS 37 49 800, Sp. 2, Z.
19-25) eine ausgezeichnete Stabilisierungswirkung besitzen. Das wurde vom Stand der Technik, wie z. B.
der US-PS 37 49 800, nicht im geringsten nahegelegt
Erfindungsgemäß wird dasjenige Mittel bevorzugt bei dem das Prostaglandin PGEi oder PGE2 ist
insbesondere wenn das PGE2 das N-Acetyl- oder
N-Benzoyl-PGErcarboxamid der Formel

so Auch wird das Mittel bevorzugt bei dem das Prostaglandin in einer Menge von etwa 0,010-0,100

Eine andere bevorzugte Ausführungsform der erfindungsgemüBen Mittel ist diejenige, bei der auch ein
halogenierter Kohlenwasserstoff mit bis zu 2 Kohlenstoffatomen als Treibmittel vorliegt, wobei das Treibmittel in einer Menge von 1 -10 Volumenteilen pro Teil
Alkohol vorliegt

durchgeführt; die Prostaglandine der Ε-Reihe, welche durch diese Reaktionsfolge hergestellt wurden, sowie
diejenigen, welche nach anderen Reaktionsschemata
h5 hergestellt oder aus natürlichem Material isoliert
wurden, sind /ur Verwendung in den erfindungsgemäßen Mitteln geeignet. Ebenfalls sind hierfür die
l5-nied.AlkyUlcrivate der natürlichen Prostaglandine

geeignet
Besonders wichtige Beispiele für biologisch aktive Prostaglandine der Ε-Reihe, welche für die erfindungsgemäßen Mittel brauchbar sind, sind die folgenden:
N-substituierte Prostaglandincarboxamide, wie zum Beispiel N-Acetyl- und N-Benzoyl-PGErcarboxamid (vgl, BE-PS 8 00 580);
dia Tetrazolylderivate von Prostaglandinen (vgl. BE-PS 7 88 209);
die Oxaprostaglandine (vgL BE-PS 8 00 350);
die p-Biphenylester von ω-nor-substituierten Prostaglandinen, wie zum Beispiel der 16-Phenyl-13,14-dihydro-co-tetranor-PGE2-p-biphenylester (vgl.
BE-PS 8 07 047);
und die ω-substituierten Pentanorprostaglandine, wie zum Beispiel 16-p-Biphenyl-w-tetranor-PGE2,
sowie 16-(«-Thienyl)-ö-tetranor-PGE! (vgl. BE-PS
802231). Μ
Auf diese Druckschriften wird im vorliegenden Bezug genommen.
Die zur Verwendung in den erfindungsgemäßen Mitteln geeigneten Alkohole sind zahlreich; sie können
alle als gesättigte, aliphatisch.e tertiäre Alkohole mit 4-10 Kohlenstoffatomen gekennzeichnet werden.
Beispiele für derartige Alkohole sind unter anderem:
tert-Butanol,
23-Dimethyl-2-butanol, J0
3-Methyl-2-butanoI,
23-Dimethyl-3-pentanol und
3-Äthyl-3-pentanol.
Aufgrund der leichten Zugängliche it und dies geringen
Preises wird tert-Butanol bevorzugt
In manchen Fällen ist es erwünscht, daß die erfindungsgemäßen Mittel in Form eines Aerosolsprühnebels verabreicht werden. Für eine derartige Anwendung wird ein halogenierter Kohlenwasserstoff mit bis
zu 2 Kohlenstoffatomen als Treibmittel verwendet Das Treibmittel kann ein beliebiges der bei Aerosolformulierungen verwendeten herkömmlichen Treibmittel sein,
wie zum Beispiel ein Halogenkohlenwasserstoff des Fluorkohlenwasserstoffs- oder Fluorhalogenkohlenwasserstofftyps, wie Trichlormonofluormethan, Dichlordifluormethan, Dichlortetrafluoräthsm, Monochlortrifluormethan, Monochlordifluormethan oder Gemische derselben untereinander oder mit anderen
Treibmitteln. Typisch für geeignete Treibmittel sind diejenigen aliphatischen Fluorchlorverbindungen, welche in der US-PS 2868 691 beschrieben «find und im
Handel vertrieben werden, z. B. Dichlordifluormethan.

1. Zum Nachweis der stabilisierenden Wirkung von tertiären, gesättigten aliphatischen Alkoholen mit 4 — 10
Kohlenstoffatomen auf Prostaglandine der E-Reihe wurden Lösungen mit einer Konzentration an PGE2 von
0,5 mg/ml in einem Gemisch aus einem der zu testenden to nachfolgenden Alkohole und 1,1,2-Trichlortirifhioräthan
im Volumenverhältnis von 50 : 50 hergestellt:
tert.-Butylalkohol
2,3-Dimethyl-2-butanol, 2,3-Dimethyl-3-pentanol,
2-Methyl-2-pentanol und
3-Äthyl-3-pentanol.
2, Zu Vergleichszwecken wurden analoge Lösungen
mit einem der folgenden, nicht unter obige Definition fallenden Alkohole hergestellt:
Äthanol,
sek.-Butanol,
2-Hexanol,
3-Methyl-2-butanoI,
Linalool,
2-Methyl-4-penten-2-oI.
l.li-Trichlortrifluoräthan wurde deshalb für das
Lösungsmittelsystem verwendet, weil es dem üblicherweise als Treibmittel für PGErAerosolsprays benutzten
Gemisch aus 1,2-Dichlortetrafluoräthan und Chlorpentafluoräthan (im Verhältnis von 60 :40) ähnlich ist und
einen niederen Dampfdruck aufweist
3. Alle vorgenannten Lösungen wurden in 2 ml-Ampullen abgefüllt, die nach dem Verschließen 1 Woche bei
500C gelagert wurden. Das Ausmaß des Abbaus von
PGE2 wurde durch dünnschichtchromatographische Analyse ermittelt Hierbei wurden gleiche Teile mit
einem Gehalt an 25 Mikrogramm PGE2 auf Silikagelplatten (Typ GF, Analtech, Inc.) als 1,27 cm Streifen
aufgebracht, welche bis 2£4cm über den Ursprung
hinaus in absol. Methanol entwickelt, getrocknet und
sodann in einem Gemisch aus Äthylacetat und 88prozentiger HCOOH (im Verhältnis von 400 :5) ganz
entwickelt wurden. Die Abbauprodukte PGA2 und PGB2 wurden im UV-Licht von 254 nm nachgewiesen,
und PGE2, PGA2, KJB2 sowie andere, nicht identifizierte Produkte wurden sichtbar gemacht, indem die Platten
mit einer Lösung von Phosphormolybdänsäure in Äthanol besprüht und erwärmt wurden.
4. Ergebnisse
Die PGErLösungen in dem Lösungsmittelsystem 1,1,2-Trichlortrifluoräthan-ÄthanoI zeigten unter vorgenannten Versuchsbedingungen einpn etwa 50prozentigen Abbau zu PGA2 und PGB2, diejenigen im System
2-Hexanol-l,lJ2-Trichlortrifluorätha.T zeigten ein analoges Verhalten, während die Lösung im System
sek.-Butanol-1,l,2-Trichlortrifluoräthan einen etwa 25prozentigen Abbau zeigte.
Die Lösung von PGE2 in dem Lösungsmittelsystem
mit 3-MethyI-2-butanol zeigte nach 2 Wochen Lagerung bei 500C einen merklichen Abbau des PGE2.
Die tertiären Alkohole Linalool, 2-Methyl-4-penten-2-0I und 1-Methylcyclopentanol, welche keine gesättigten aliphatischen Alkohole darstellen, führten zu keiner
signifikanten Stabilisierungswirkung.
Demgegenüber zeigten die Lösungen in dem Lösungsmittelsystem, welches einen der unter (1)
genannten tert Alkohole enthielt überraschenderweise keinen Abbau von PGE2 unter den Versuchsbedingungen.
Die nachfolgenden Beispiele dienen zur näheren Erläuterung der vorliegenden Erfindung.
Beispiele 1-6
Durch Verwendung der erfindungsgemäßen pharmazeutischen Mittel kann die bronchodilatorische Wirksamkeit des Prostaglandins erfolgreich über lange
Zeiträume hinweg bei erhöhten Temperaturen aufrecht erhalten werden. Dies ist insbesondere für eine
Verwendung als Bronchodilatoren in Form von Aerosolen wichtig.
Ergebnissen geht die Wirksamkeit von tert-Butanol enthaltenden Prostaglandin Ε-Präparaten bezüglich
einer anhaltenden Wirksamkeit hinsichtlich des Schutzes von Meerschweinchen gegen Bronchospasmen,
welche durch Histamin induziert wurden, nach einer Lagerung bei erhöhten Temperaturen hervor. Durch die
erfindungsgemäßen Mittel wird die Stabilität von Prostaglandinen der Ε-Reihe bei einer Erhöhung der
nasalen Durchlässigkeit, der Einleitung von Wehen oder eines Aborts sowie der Regulierung der Fruchtbarkeit
in gleicher Weise erhöht.

2) Vorratslösungen, verdünnt auf 56-100 mcg/ml mit Äthanol oder Wasser. Diese verdünnten Lösungen wurden in eine
durchsichtige Box hinein vernebelt, und die Meerschweinchen wurden dem direkten Sprühnebei während eitler
Minute ausgesetzt, wonach man sie eine weitere Minute einatmen ließ. Die Tiere wurden sodann in eine andere
Box gebracht, in welcher eine Histaminlösung (2 mg/ml) während einer Minute vernebelt wurde. Nachdem sie
60-75 Sekunden in dieser Atmosphäre gelassen wurden, wurde eine subjektive Bewertung des Atmungsbildes de'
Tiere vorgenommen (0 = keine Wiikung, 4 = Krämpfe). Der prozentuale Schutz ergibt sich wie folgt:

N-Benzoyl-prostaglandin-Ej-carboxamid als Aerosol
N-Benzoyl-prostaglandin-Ej-carboxamid als Aerosol
N-Benzoy !-prostaglandin-Er carboxamid 10 mg
tert.-Butylalkohol 2 g
Gemisch aus Chlorpentafluoräthan
und 1,2-Dichlortetrafluoräthan
(Gewichtsverhältnis 40:60), ad 10 g
Das Prostaglandin wurde in tert.-Butylalkohol gelöst und in mit Kunststoff ausgekleidete Aerosolflaschen
abgefüllt. Das Treiomittel wurde sodann in der Kälte in
die Behälter eingefüllt, welche mit einem geeigneten Dosierventil verschlossen wurden.
Claims (1)
- Patentanspruch:Stabilisiertes pharmazeutisches Mittel mit einem Gehalt an einem biologisch aktiven Prostaglandin der Ε-Reihe und einem gesättigten aliphatischen tertiären Alkohol mit 4—10 Kohlenstoffatomen, wobei das Prostaglandin in einer Menge von 0,005 - 0,500 Gew.-% vorliegt
