Prostaglandine sind eine Gruppe von Naturstoffen, die aus verschiedenen
Tiergeweben isoliert wurden. In Säugern sind sie für
eine Vielzahl von pharmakologischen Wirkungen verantwortlich,
von denen hier nur die Beeinflussung der Kontraktionen der
glatten Muskulatur und des Blutdruckes erwähnt seien. Bezüglich weiterer pharmakologischer Eigenschaften siehe unter anderen

Für die katalytische Hydrierung eignen sich Metalle wie Kobalt,
Iridium, Rhodium, vorzugsweise Nickel, Palladium und Platin in feinverteilter Form, gegebenenfalls auf Trägern wie Calcium»,
Strontium- oder Bariumsulfat, Calcium-, Strontium- oder Bariumcarbonate Aluminiumoxid, Kieselgur oder Aktivkohle. Weiterhin
eignen sich für die katalytische Hydrierung Katalysatoren wie Niekelborld, Nickel- oder Kupferchromite Rutheniumoxyd
oder lösliche Metallkomplexe wie Tris-(triphenjrlphosphin)■··

PUr die Hydrierung eignen sich auch Metallsalze wie Nickelchlorid oder Kobaltchlorid zusammen mit komplexen Hydriden wie

in Tetr. Lett. I968, 6513 beschrieben.
Für die katalytische Hydrierung können, Je nach Aktivität- des
KatalysatorS., Temperaturen von 0° bis 150°, vorzugsweise 20
bis 60 und Wasserstoffdrucke von 1 bis 100 Atmosphären, vorzugsweise
Normaldruck, angewendet v/erden.
Als Lösungsmittel eignen sich solche, die unter den gewählten Bedingungen nicht reduziert werden, z.B. Kohlenwasserstoffe
wie Benzole A'ther wie Diäthyläther, Tetrahydrofuran, 1,2-Di~
methoxyäthan und Dioxan, Alkohole wie Methanol, Kthanol, Isopropanol

Bei der katalytischen Hydrierung von Verbindungen, in denen
R Tetrahydropyranyl oder 1-Alkoxyalkyl bedeutet, in protisehen
Lösungsmitteln wie Äthanol wird in Gegenwart katalytischer Mengen von Säure dieser Rest abgespalten und gegen Viasserstoff
ersetzt* Als chemische Reduktionsmittel kommt z.B. Diimin (vgl. J. org. Chem. ^Q, 5985 (1965) in Betracht.
Man arbeitet in der üblichen Weise auf und reinigt die Produkte
gegebenenfalls chromatographisch.
Die Herstellung der Ausgangsverbindungen der Formel II kann
gemäß der deutschen Patentschrift (Patentanmeldung P 2j5 31 O8I.8) erfolgen.
Die Ufflsetzung einer Verbindung der Formel IY mit einer Verbindung
der Formel V erfolgt nach an sich bekannten Methoden,
zweckmäßig bei Temperaturen zwischen Raumtemperatur und 14O°C
in inerter Atomosphäre. Besonders vorteilhaft werden sie folgendermaßen
hergestellt:
Man löst eine Verbindung der Formel IY1 in welcher R/ vorzugsweise
den Tetrahydropyranyl- oder einen niedermolekularen 1-Alkoxyalkylrest bedeutet,, in einem absoluten, aprotischen
Lösungsmittel vorzugsweise Benzol, Toluol oder Xylol und fügt 1 bis .1,5 Mol einer wasserfreien. Base, vorzugsweise Natriumäthylat
oder Ivaliumtertiärbutylat zu und rührt JO Minuten bis'
3 Stunden in inerter Gasatmosphäre bei Raumtemperatur.
Danach gibt man 1 bis 2 Mol eines Halogencarbonsäureesters der Formel V zu und rührt bei Temperaturen zwischen 20 und 14O°C
- je nach Reaktivität der eingesetzten Halogenverbindung- zwischen
1 und 20 Stunden unter SauerstoffausSchluß (siehe auch
belgische Patentschrift Nr* ?66 521). Anschließend arbeitet
man in üblicher Weise auf, wobei man die meist als öl anfal-

lenäen Verbindungen chromatographisch reinigt.

oder.. Äthyl vinyl äther oder Vinyl thio.äthern in Gegenwart eines
sauren Katalysators wie z.B. p-Toluolsulfonsäure, BF-^-Ätherat
oder eines sauren Ionenaustauschers oder an et-, ß-ungesättigte
Ketone oder Säurederivate wie Methylvinylketon, Acrylsäuremethylester oder Acrylnitril in Gegenwart eines basischen
Katalysators wie z.B. Kaliurntertiärbutylat, · Natriummethylat oder eines basischen Ionenaustauschers erhalten werden.
Verbindungen der Formel I g werden in an sich bekannter Weise durch Reduktion einer Verbindung der Formel I e oder I f mit
einem komplexen Metallhydrid, zweckmäßig einem Metallboranate
vorzugsweise Alkaliboranat, wie beispielsweise Natriumborhydrid
oder mit Zinkborhydrid in ätherischer oder alkoholischer Lösung, vorzugsweise in absolut alkoholischer Lösung hergestellt. Dabei
kann man bei Temperaturen zwischen -10 bis +600C arbeiten,

im allgemeinen hat sich der Temperaturbereich zwischen O und
■10°C bewährt.
Überraschenderweise kann man aus den Estern der Formel I g Halbester
der Formel I h erhalten, wenn man einen Ester der Formel I gin einem geeigneten Lösungsmittel wie z.B. Methanol,
Äthanol, Dioxan, Tetrahydrofuran oder 1,2-Dimethoxyäthan;gegebenenfalls
in Gegenwart von Wasser mit einer Base, wie z.B.
Natrium-, Kalium- oder Lithiumhydroxyd oder Natriumcarbonat
4 verseift. Dabei wird nur die Estergruppe mit dem Rest R der
Formel I g in' das entsprechende Salz überführt. Dieses Salz ■

Überräschenderweise haben die Verbindungen der Formel I, die
wie eingangs festgestellt wurde, strukturell den natürlichen
Prostaglandinen nahe verwandt sind, eine starke prostaglandinantagonistische
Wirkung. Gibt man beispielsweise zum isolierten Meerschweinchenileum oder -uterus eine Lösung der erfiridungsgomäßen
Verbindungen in Wasser und fügt zu dieser Lösung beispielsweise Prostaglandin Ep oder Fp<x, in Konzentrationen,

Die antagonistische Wirkung der erfindungsgemäßen Verbindungen konnte mit Hilfe der in der deutschen Patentschrift
(Patentanmeldung P 2J 5I 08I.8) beschriebenen
Versuchsanordnung nachgewiesen werden.
Die erfindungsgemäßen Verbindungen können wegen ihrer antagonistischen
Wirkung gegenüber den natürlichen Prostaglandinen als Arzneimittel verwendet werden.
Insbesondere ist die Hemmung oder Unterdrückung einer oder mehrerer der vielen pharmakologischen Eigenschaften der
Prostaglandine r- z.B. der spasmogenen Wirkung auf bestimmte
glatte Muskeln - sehr erwünscht.
Die erfindungsgemäßen Verbindungen der allgemeinen Formel I können in Form ihrer wäßrigen Lösungen oder Suspensionen
oder auch als Lösungen in phartnakologisoh unbedenklichen
organischen Lösungsmitteln wie z.B. ein- oder mehrwertigen
Alkoholen, Dimethylsulfoxid oder Dimettuylsulfoxid oder
Dimethylformamid, auch in Gegenwart von pharmakologisch unbedenklichen Pclymerträgern wie z.B. Polyvinylpyrrolidon
zur Anwendung gelangen. Als Zubereitungen kommen neben den üblichen galenischen Infusions- oder Injektionslösungen auch

Tabletten in Betracht. Bevorzugt sind Jedoch örtlich anwendbare
Zubereitungen /wie Cremes, Emulsionen, Supposit.^_orien oder

Die Verbindungen gernäß der Erfindung können für sich allein"
oder zusammen-mit anderen.-pharmakologischen Wirkstoffen wie
z.B.- Prostaglandinsynthetasehemmern wie dem Natriumsalz der
Acetylsalicylsäure zur Anwendung gelangen.
Weiterhin"· stellen die oben beschriebenen Verbindungen wertvolle
Zwischenprodukte bei der-Synthese neuer Prostaglandine
