Description
in der R ein WasserstoFfatom, einen Alkylrest mit 1 — 12 Kohlenstoffatomen oder einen Phenyirest, X
eine NHrGruppe, eine Piperidinogruppe, eine Morpholinogruppe oder eine NRiRrGruppe bedeuten,
wobei Ri und R2 Alkylreste mit 1—4 Kohlenstoffatomen
darstellen, dadurch gekennzeichnet,
daß man Carbonsäureamiddichloride oder -dibromide mit phosphoriger Säure umsetzt
Gegenstand der Erfindung ist ein neues Verfahren zur ;s
Herstellung von 1-Aminoalkaii-U-diphosphonsäuren,
deren Aminognippe gegebenenfalls substituiert sein kann.
Es ist bekannt, l-Aminoalkan-1,l-diphosphonsäuren durch Umsatz von Alkylnitrilen mit PBr3 oder PCIj
herzustellen, wobei befriedigende Ausbeuten jedoch nur mit ΡΒΓ3 erzielt werden. Nach diesem Verfahren ist es
nicht möglich, Verbindungen herzustellen, bei denen die Wasserstoffatome der Aminognippe beispielsweise
durch Alkylreste substituiert sind. Ebenfalls ist es Ji
bekannt, N,N-Dimethy!-1 -aminomethan-1,1 -diphosphonsäuretetraäthylester
durch Umsetzung von Dimethylformamiddiacetal mit Diäthylphosphit herzustellen.
Dieses Verfahren ist umständlich, zumal die Ester noch in die freien Säuren überführt werden müssen.
Es wurde nun gefunden, daß man auf einfache Weise 1-Aminoalkan-1,1-diphosphonsäuren der allgemeinen
Formel

in der R ein Wasserstoffatom oder einen Alkylrest mit 1—12 Kohlenstoffatomen, X eine NHi-Gruppe, eine
Piperidinogruppe, eine Morpholinogruppe oder eine NRiRrGruppe bedeuten, wobei Ri und R2 Alkylreste
mit I -4 Kohlenstoffatomen darstellen, herstellen kann. ">">
wenn man sich des nachstehend beschriebenen Verfahrens bedient. Das neue Verfahren ist dadurch gekennzeichnet,
daß man Carbonsäureamiddichloride oder -dibromide mit phosphoriger Säure umsetzt.
Die als Ausgangsprodukte verwendeten Carbonsäu- w>
reamiddichloride entsprechen den Formeln RCCIjNHj
oder RCCI2NRiR2. Sie werden nach literaturbekannten
Methoden hergestellt, die nicht Gegenstand der vorliegenden Erfindung sind. Verfahren zur Herstellung
dieser Verbindungen sind beispielsweise in den Berich- '■>
ten der Deutschen Chemischen Gesellschaft Band 64 (193J)S. 667 ff., in Liebigs Annalen der Chemie Band 594
(1955) S. 21 ff. sowie in Helvetia Chimica Acta Band 42
(1959) S. 1653 ff, und 1659 ff, besehrieben. Als geeignete
Carbonsäureamiclderivate kommen Dichloride und
Dibromide von Carbonsäureamiden in Frage, in denen der Rest R ein Wasserstoffatom, eine geradlceuige oder
verzweigte Alkylgruppe, eine cycloaliphatische, aromatische oder heterocyclische Gruppe darstellt, wobei die
genannten Gruppen weitere Substituenten beispielsweise Alkyl- und Phenylreste oder funktionelle Gruppen
enthalten können. So eignen sich als Ausgangsmaterial
für die Darstellung der Aminoalkandiphosphonsäuren unter anderem die Dichloride und Dibromide folgender
Verbindungen: Acetamid, Isobuttersäureamid, Valeriansäureamid,
Caprinsäureamid, Laurinsäureamid, Cyclohexancarbonsäureamid.
Benzamid, Phenylessigsäureamid, Hydrozimtsäureamid, Pyridincarbonsäureamide
und Carbäthoxyacetamid.
In den genannten Säureamiddichloriden oder -dibromiden
können die Wasserstoffatome der ,Aminognippe
auch durch gleiche oder verschiedene Alkylreste mit 1 bis 4 C-Atomen substituiert sein. Schließlich können
auch solche Säureamiddichloride oder -dibromide verwendet werden, bei denen die Substituenten der
Aminognippe zusammen mit dem Stickstoffatom einen heterocyclischen Ring, beispielsweise einen Piperidin-
oder Morpholinring, bilden. Als Beispiele für die zuletzt beschriebene Gruppe von Verbindungen sind die
Dichloride und Dibromide folgender Carbonsäureamide zu nennen: Dimethylformamid, Diäthylformamid, Dibutylformamid,
Piperidinoformamid, Dimethylacetamid, Diäthylacetamid, Dimethylbenzamid und Morpholinocyclohexancarbonsäureamid.
Es hat sich als zweckmäßig erwiesen, die Carbonsäureamiddihalogenide
mit phosphoriger Säure im Molverhältnis 1 :2 bis 1 :0,5 umzusetzen. Vorzugsweise wird
ein Molverhältnis von 1 :1 verwendet Die Umsetzung kann ohne Zufuhr von Wärme erfolgen. Gewünschtenfalls
kann in Gegenwart eines Lösungsmittels wie beispielsweise Tetrachlorkohlenstoff, Dimethylformamid
oder Diäthyläther oder anderen inerten Lösungsmitteln gearbeitet werden.
Die Reaktion geht unter Halogenwasserstoffentwicklung vor sich. Gleichzeitig tritt eine Viskositätszunahme
und eine Temperaturerhöhung des Reaktionsgemisches ein. Eine Kühlung ist bei geeigneter Reaktionsführung
nicht erforderlich.
Nach dem Ende der Reaktion wird das Reaktionsgemisch mit Wasser behandelt. In Wasser schwer- oder
unlösliche l-Aminoalkan-U.-diphosphonsäuren werden
abfiltriert und ausgewaschen. Die wasserlöslichen Verbindungen dieser Reihe lassen sidvaus den wäßrigen
Lösungen durch Zusatz von geeigneten Lösungsmitteln v;e Aceton, Methanol, Äthanol oder Isopropanol
ausfällen.
Die nach dem neuen Verfahren herstellbaren Aminodiphosphonsäuren können als Komplexbildner
für viele technische Zwecke Anwendung finden. Sie können beispielsweise verwendet werden für die
Wasserenthärtung, bei Reinigungsprozessen, insbesondere bei der Flaschenspülung, oder auch zur Entkrustung
von Geweben in Waschprozessen. Weiterhin sind sie geeignet als Zusatz zu Färbebädern für Textilien, um
Metallionen zu binden, die unerwünschte Farbnuancen hervorrufen.
25,6 g (0,2 Mol) Dimethylformamiddichlorid, in 100 ml
Dimethylformamid suspendiert, werden unter Rühren mit 32,6 g (0,4 Mol) phosphoriger Säure vermischt. In
stark exothermer Reaktion entwickelt sich sofort Chlorwasserstoff, Nach dem Erkalten der Reaktionsmischung
wird das Lösungsmittel abdestilliert und das zurückbleibende zähflüssige öl in Wasser gelöst. Nach
Zusatz von Aceton scheidet sich die N,N-Dimethylaminometban-i.l-dipbosphonsäure
kristallin ab,
Ausbeute: 25,0 g(57% d.Th.).

25,6 g (0,2 Mol) kristallines Dimethylformamiddichlorid
wird mit 16,4 g (0,2 Mol) kristalliner phosphoriger Säure vermischt Unter Erwärmung und Chlorwasserstoffentwicklung
verflüssigt sich das Reaktionsgemisch. Nach dem Erkalten wird in Wasser aufgenommen und
bis zur Abscheidung der Ν,Ν-Dimethylaminomethan-1,1
-diphosphonsäure mit Aceton versetzt
Ausbeute: 16,5 g (75% d. Th.).

Zu 25,6 g (0,2 Mol) kristallinem Dimethylformamiddichlorid
werden 32,8 g(0,4 Mol) kristalline phosphorige Säure zugegeben. Unter Erwärmung und Chlorwasserstoffentwicklung
verflüssigt sich das Reaktionsgemisch. Nach dem Erkalten wird in Wasser aufgenommen und
bis zur Abscheidung der Ν,Ν-Dimethylaminomethan-1,1
-diphosphonsäure mit Aceton versetzt
Ausbeute: 26 g (60% & Th.).

Zu 42 g (0,2 Mol) Phosphorpentachlorid, in 200 ml
Tetrachlorkohlenstoff suspendiert werden 16 g (0,2 Mol) Dimethylformamid unter Rühren zugetropft Das
Reaktionsgemisch wird 10 Minuten weiter gerührt Der Suspension, die jetzt aus Tetrachlorkohlenstoff, Phosphoroxychlorid
und dem Dimethylformamiddichlorid besteht, werden — ebenfalls unter Kühren — 33 g (0,4
Mol) phosphorige Säure zugesetzt Unter Erwärmung und Chlorwasserstoffentwicklung setzt sich eine ölige
Phase ab, die in 100 ml Wasser aufgenommen und abgetrennt wird. Die wäßrige Lösung wird eingedampft.
Das zurückbleibende Öl wird nochmals in wenig Wasser gelöst. Die Lösung wird bis zur Abscheidung der
kristallinen Ν,Ν-Dimethylaminomethan-1,1 -diphosphonsäure
mit Aceton versetzt.
Ausbeute: 23,5 g (54% d. Th.).

28.4 g (0,2 Mol) kristallines Dimethylacetamiddichlorid
werden mit 32,8 g (0,4 Mol) kristalliner phosphoriger Säure versetzt Unter Erwärmung und Chlorwasserstoffentwicklung
verflüssigt sich das Reaktionsgemisch. Nach dem Erkalten wird das zähe Öl in Wasser
aufgenommen und bis zur Abscheidung der N,N-Dimethylaminoäthan-l,l-diphosphonsäure
mit Aceton versetzt.
20.5 g (0,1 Mol) Dimethylbenzamiddichlorid werden mit 8 g (0,1 Mol) phosphoriger Säure versetzt. Unter
Erwärmung und Chlorwasserstoffentwicklung entsteht ein zähflüssiges Reaktionsprodukt, das nach dem
Erkalten in Wasser gelöst und bis zur Trübung mit Aceton versetzt wird. Nach einigen Stunden scheidet
sich die l-Phenyl*I-dimethylaminomethan-l,l-diphosphonsäure
kristallin ab.
Zu 42 g (0,2 Moi) Phosphorpentachlorid, in 200 ml
Tetrachlorkohlenstoff suspendiert, werden 22,5 g (0,2
Mol) Piperidinoformamid zugetropft und nach 15
Minuten Rühren 16,5 g (0,2 Mol) phosphorige Säure zugesetzt Unter Erwärmung und HCI-Entwicklung
scheidet sich eine ölige Phase ab, die in Wasser aufgenommen und abgetrennt wird. Aus der wäßrigen
Lösung fällt nach Zusatz von Aceton oder Äthanol die 1 -Piperidinomethan-1,1 -diphosphonsäure aus.
Ausbeute: 14,5 g(56% d. Th.).

114 g (1,0 Mol) Acetamiddichlorid werden bei etwa
O'C mit 82 g (1,0 Mol) phosphoriger Säure versetzt Dabei erwärmt sich das Reaktionsgemisch unter
Verflüssigung und unter HCI-Entwicklung. Nach dem Abklingen der exothermen Reaktion und dem Abkühlen
auf Raumtemperatur nimmt die Viskosität stark zu. Anschließend wird das Reaktionsprodukt in Wasser
gelöst Auf Zusatz von Aceton scheidet sich die 1 -Aminoäthan-1,1 -diphosphonsäure ab.
Ausbeute: 52 g (51 % d. Th.).

315 g (1,0 Mol) Caprinsäureamiddibromid werden mit
82 g (1,0 MoI) phosphoriger Säure vermischt. In exothermer Reaktion entwickelt sich Bromwasserstoff.
Nach dem Abklingen der Reaktion wird das Rohprodukt in Wasser gelöst und die 1-Aminodecan-U-diphosphonsäure
mit Aceton ausgefällt
Ausbeute:92 g (58% d. Th.).

176 g (1,0 Mol) Benzamiddichlorid werden mit 82 g (1,0 Mol) phosphoriger Säure vermischt Dabei erwärmt
sich das Gemisch und entwickelt Bromwasserstoff. Nach dem Abklingen der Reaktion wird das Rohprodukt
in Wasser gelöst und die 1-Aimno-l-phenylmethan-1,1-diphosphonsäure
mit Aceton ausgefällt.
Ausbeute: 58 g (43% d. Th.).

28Og(I1O Mol) Phenylacetamiddibromid werden mit
82 g (1,0 MoI) phosphoriger Säure vermischt, wobei sich das Gemisch unter Bromwasserstoffentwicklung erwärmt
Nach dem Abklingen der Reaktion wird das Rohprodukt in Wasser gelöst und die kristalline
l-Amino-2-phenyläthan-1,1-diphosphonsäure durch Zugabe
von Aceton ausgefällt.
Afsbeute: 100 g (72% d. Th.).

275 g (1,0 Mol) Carbäthoxyacetamiddibromid werden mit 82 g (1,0 MoI) phosphoriger Säure vermischt.
Nachdem die exotherme Reaktion, bei der Bromwasser-
hn stoff entsteht, abgeklungen ist, wird das Rohprodukt in
Wasser gelöst und die l-Amino-2-carboxyäthan-l,1-diphosphonsäure durch Zugabe von Aceton oder Äthanol
gefällt. Wenn die so erhaltene Säure nach dem Papierchromatogramm nicht einheitlich ist, wird sie mit
f,-, konzentrierter, wäßriger Bromwasserstoffsäure erhitzt
und anschließend wieder ausgefällt.
Ausbeute: 25-40%.
