The invention relates to a method for the production of organic compounds by producing Aryllithium compounds and their implementation with suitable electrophiles, being initially an intermediate aryllithium compound by reacting halogen aromatics with lithium metal is generated (step 1 in equation I), which is then in a halogen-metal exchange reaction with sufficiently reactive aromatic halogen compounds to form the desired ones Lithium aromatics is implemented (step 2 in equation I), wherein concluding this be reacted with an appropriate electrophile (Step 3 in Equation 1).
PhLi), Lithiumamiden (z.B. LDA oder LiNSi) oder den SCHLOSSER-Superbasen (RLi/KOtBu)
- Almost every organolithium compound can be easily generated via the modern arsenal of organometallic chemistry and implemented with almost any electrophile to the desired product. Most of the lithium organyls are generated in one of the following ways:
- (1) The most important way is undoubtedly the halogen-metal exchange, in which mostly bromoaromatics are reacted with n-butyllithium or phenyllithium at low temperatures. The less reactive phenyllithium is used in particular with electron-poor substrates, for example polyhalogenated benzenes or benzotrifluoride, and / or when a particularly high selectivity is required.
- (2) A large number of Li organyls can also be prepared by reacting aromatic bromates with lithium metal
- (3) The deprotonation of sufficiently acidic organic compounds is also very important gene with lithium alkyl or aryl compounds (e.g. PhLi), lithium amides (e.g. LDA or LiNSi) or the SCHLOSSER super bases (RLi / KOtBu)
It already follows that for the most part this chemistry the use of salable Organolitinium compounds are required, mostly here n-butyllithium or the less reactive and therefore often more selective Phenyllithium can be used. The synthesis of phenyllithium and related lithium aromatics is technically complex and requires a lot of know-how, which makes phenyllithium and similar molecules only - in industrial Measured standards - very expensive Tobe offered. This is the most important, but far from it the only disadvantage of this otherwise very advantageous and wide usable reagent.
Because of the extreme sensitivity and in concentrated solutions The pyrophoric nature of such lithium aromatics must be considered in the industrial large-scale production desired orders of magnitude (Annual production quantities between 5 and 500 tons) very complex logistic systems for Transport, feeding into the dosing template and dosing set up become.
Furthermore, arise during the implementations of phenyllithium is either benzene, which is carcinogenic in humans (Deprotonation) or phenyl halides (halogen-metal exchange), which are generally considered environmentally hazardous are classified and sometimes only by elaborate processing Additional operations can be separated from the products. The Formation of benzene prevent themselves by using alternatives such as tolyllithium, which, however, are not available on the market or only in small quantities. The availability of such alternatives the further advantage that n-butyllithium, the most commonly used alkyl lithium compound, in many cases e.g. could be replaced by tolyllithium. This would eliminate the Problems associated with the use of butyllithium gaseous Butane (deprotonation) or halobutane (halogen-metal exchange with an equivalent BuLi), which have to be collected or flared in a complex manner in order to to comply with the strict immission control regulations.
Another disadvantage is the accumulation complex solvent mixtures after working up. Due to the high reactivity of aryllithium compounds across from Ethers, which are almost always solvents for the Follow-up implementations can Aryllithium compounds are usually not offered in these solvents. The manufacturers offer different aryllithium compounds Concentrations in various hydrocarbons or hydrocarbon-ether mixtures, however, for example, halogen-metal exchanges run in pure Hydrocarbons do not come off, so you inevitably in Mixtures of ethers and hydrocarbons must work. The mostly for reasons of stability Diethyl ether used is extremely flammable and forms very easily highly explosive peroxides with atmospheric oxygen, which is why a large-scale Generally prohibits application. In addition, water-containing mixtures are obtained after hydrolysis from ethers and hydrocarbons, the separation of which is expensive and in many cases not economical at all feasible is. For a large industrial production is however the repatriation of the solvents used indispensable requirement.
It would therefore be for the reasons mentioned very desirable to have a method in which the aromatic lithiation reagent to be used based on the cheap raw materials halogen aromatic and lithium metal generated in an ether and then with the one to be converted Halogen aromatics is implemented because by this procedure everyone disadvantages of the "classic" mentioned above Generation of aromatic lithium compounds could be avoided.

The lithium organyls prepared in this way can be processed using any electrophilic compounds state of the art. For example, C, C linkages can be carried out by reaction with carbon electrophiles, boronic acids can be prepared by reaction with boron compounds, and a very efficient route to organosilanes is opened up by reaction with halogen or alkoxysilanes.
All can be used as halogen aromatics (i) available or fluorine, chlorine, bromine or iodoaromatics which can be prepared be because lithium metal in ethereal solvents with all halogen aromatics light and in almost all cases reacted with quantitative yields. Chlorine or bromine aromatics because iodine compounds are often expensive, Fluorine compounds lead to the formation of LiF, which in later aqueous Refurbishments as HF can lead to material problems. In special cases, too such halides can be used advantageously. The halogen aromatics can isomerically pure or used as technical isomer mixtures, which is often even cheaper is.
Such aryl halides are preferred used after the halogen-metal exchange to be converted to such aromatics or aryl halides can be easily removed during processing. Particularly preferred are either isomerically pure or present as isomer mixtures Chloro- or bromotoluenes, the reaction products of which can easily be distilled or have it removed by azeotropic distillation. With volatile Products also higher boiling Halogen aromatics are used.
Suitable ethereal solvents are for example tetrahydrofuran, dioxane, diethyl ether, di-n-butyl ether, diisopropylether, Diethylene glycol dibutyl or methyl ether or anisole, preferred THF is used.
Aufgrund der hohen Reaktivität von Aryllithiumverbindungen, insbesondere auch gegenüber den als Lösungsmittel eingesetzten Ethern, liegen die bevorzugten Reaktionstemperaturen zwischen –120 und +25°C, besonders bevorzugt sind Temperaturen zwischen -80 und 0°C.Due to the high reactivity of aryllithium compounds, especially opposite the as a solvent used ethers, the preferred reaction temperatures between -120 and + 25 ° C, especially temperatures between -80 and 0 ° C. are preferred.
Concentrations of the aromatic intermediates are preferred (IV) from 5 to 30% by weight, in particular 12 to 25% by weight.
In the two preferred embodiments will first the aryl halide is metered into the lithium metal in the ether, whereby first the lithium aromatic (II) forms. Then the Halogen aromatics (III) to be metallized are added and after the Halogen-metal exchange of the electrophilic reactants.
Die zugesetzte Lithiummenge beträgt je Mol umzusetzenden Halogens 1,95 bis 2,5 mol, bevorzugt 1,98 bis 2,15 Mol.The lithium can in the present Process as dispersion, powder, chips, sand, granules, pieces, bars or used in another form, the size of the lithium particles not relevant to quality is, but only affects the response times. Therefore are smaller particle sizes preferred, for example granules, powders or dispersions. The added Amount of lithium 1.95 to 2.5 mol, preferably 1.98 to, per mole of halogen to be reacted 2.15 mol.
In all cases, at the stage of manufacture from ArLi by adding organic redox systems, for example biphenyl, 4,4'-di-tert-butylbiphenyl, naphthalene or anthracene, clear Increases in reaction rates are observed. The The addition of such systems proved to be particularly advantageous if the lithiation times without this catalysis> 12 h were. The concentrations of organic catalyst added are expediently between 0.01 and 1 mol%, preferably between 0.02 and 0.1 mol%.
Aromatics which can be used for halogen-metal exchange are initially all aromatic bromine and iodine compounds. It applies to chlorine compounds that those with activating, ie strongly electron-withdrawing substituents such as CF 3 radicals, can be lithiated in partially good yields.
The lithium aromatics generated according to the invention (IV) can according to the methods familiar to the expert be implemented with electrophilic compounds, in particular Carbon, boron and silicon electrophiles in terms of required Intermediates for the pharmaceutical and agrochemical industries are of interest.
