Description
- Bei der Erfindung handelt es sich um neuartige antivirale Wirkstoffe mit Breitband-Wirkung gegen 3C- oder 3C-like (3CL)-Proteasen von RNA-Viren, insbesondere Coronaviren, Picornaviren (vor allem Enteroviren) und Noroviren.
- RNA-Viren, insbesondere Coronaviren, Picornaviren und Noroviren haben in den vergangenen Jahrzehnten mehrfach zu Epidemien und Pandemien geführt, so brach 2003 das SARS-Coronavirus in Südchina aus und verbreitete sich in nahezu 30 Länder der Erde. Neun Jahre später trat das MERS-Coronavirus auf und zu Beginn des Jahres 2020 das neuartige Coronavirus 2019-nCoV.
- Die 3C- oder 3C-like (3CL)-Proteasen der betreffenden RNA-Viren werden als vielversprechende Targets bei der Entwicklung antiviraler Wirkstoffe mit Breitbandwirkung gesehen.
- In den letzten Jahren ist es mit Hilfe röntgenkristallografischer Studien gelungen, die dreidimensionale Struktur der Zielsysteme zu charakterisieren und so wertvolle Hinweise für die Struktur und Beschaffenheit möglicher antiviraler Wirkstoffe zu erlangen. [
EP16233028B1 - In „Solution-phase and solid-phase synhesis of novel transition state inhibitors of coagulation enzymes incorporating a piperidinyl moiety", Bioorgaanic & Medicinal Chemistry Letters, 9 (1999), 1227-1232 zeigen A. Adang et al. 2-Amino-3-piperidin-4-yl-propionsäurehaltige Peptidomimetika als potente Proteaseinhibitoren, wenn sie mit einer geeigneten Ketothiazol- oder Ketocarbonsäurekomponente kombiniert werden. Es wird ein neuartiger P1-Rest in Faktor-Xa- und Thrombin-Inhibitoren gefunden, der zu IC50-Werten von nur 0,048 µM führt und damit um den Faktor zehn wirksamer ist als Argatroban. Ausgehend von nicht-chiralen Synthesewegen werden zudem ein neuer stereospezifischer Weg sowie eine neue Festphasenmethode gezeigt.
- Die Druckschrift
DE 698 23 178 T2 offenbart neue Serin-Protease-Inhibitoren, pharmazeutische Zusammensetzungen, die diese enthalten, und auch die Verwendung der besagen Inhibitoren für die Herstellung eines Medikaments zur Behandlung und Prävention Thrombin-bedingter Erkrankungen. - In der Druckschrift
WO 97/ 31 939 A1 - Die Wirksamkeit von α-Ketoamiden als antivirale Wirkstoffe mit dem Target 3C- oder 3C-like (3CL)-Proteasen ist unter anderem aus Kim et al. „Broad-Spectrum Antivirals against 3C or 3C-Like Proteases of Picornaviruses, Noroviruses, and Coronaviruses" Journal of Virology, Volume 86 Number 21, p. 11754-11762 (2012) bekannt.
- Kristallographie-Studien zeigen, dass die α-Ketoamid-Gruppe in der Tasche der Target-Protease (3C oder 3CL) an Position P1 interagiert, während zwischen P2 und P3 eine Amidgruppe zur Interaktion mit Proteasen der Wirtszelle befähigt ist. Es kann daher eine vielversprechende Strategie sein, die Stabilität der antiviralen Wirkstoffe zu erhöhen, indem eben diese Amidgruppe vor vorzeitigem Abbau geschützt wird.
- Die
US 2015/0133368 A1 und Groutas et al. „Structure-guided design, synthesis and evaluation of oxazolidinone-based inhibitors of norovirus 3CL protease" European Journal of Medicinal Chemistry, Volume 143, 1 January 2018, Pages 881-890 lösen das Problem über einen aliphatischen Ringschluss z.B. über ein Oxazolidinon, einen aliphatischen 5-Ring der neben dem Stickstoff und der hierzu benachbarten Carbonylgruppe noch ein weiteres Sauerstoffatom trägt oder über die aliphatischen 6-Ringe 2,6-Piperidindion oder 2,6-Diketo-1,3-Diazan. Die bisher bekannten Ergebnisse zeigen Optimierungsbedarf insbesondere im Hinblick auf die Stabilität und Bioverfügbarkeit dieser Verbindungen. - Des Weiteren ist es insbesondere auch angesichts des derzeitigen Coronavirus-Ausbruchs von Vorteil, Wirkstoffe mit Lungentropismus zu haben.
- Daher ist es Aufgabe der Erfindung, neue antivirale Wirkstoffe zur Verfügung zu stellen.
- Im besonderen ist es Aufgabe der Erfindung, neue antivirale Wirkstoffe, welche auf 3C- oder 3C-like (3CL)-Proteasen von RNA-Viren wirken, zur Verfügung zu stellen.
- Insbesondere ist es Aufgabe der Erfindung, neue antivirale Wirkstoffe mit verbesserter Bioverfügbarkeit zur Verfügung zu stellen.
- Des Weiteren ist es Aufgabe der Erfindung, neue antivirale Wirkstoffe mit Lungentropismus zur Verfügung zu stellen.

- In einer weiteren besonderen Ausführungsform wird die Aufgabe der Erfindung durch einen antiviralen Wirkstoff gelöst, der auf 3C- oder 3C-like (3CL)-Proteasen von RNA-Viren wirkt.
- In einer weiteren bevorzugten Ausführungsform wird die Aufgabe der Erfindung durch einen antiviralen Wirkstoff gelöst, der eine Halbwertszeit im Plasma von mehr als 30 Minuten, bevorzugt mehr als 40 Minuten, besonders bevorzugt mehr als 44 Minuten aufweist.
- In einer weiteren bevorzugten Ausführungsform wird die Aufgabe der Erfindung durch einen antiviralen Wirkstoff gelöst, der Lungentropismus zeigt.

- Die Leitverbindung DZL08 wurde in virus-infizierter Zellkultur getestet und zeigt hervorragende Ergebnisse gegen das Enterovirus EV-A71, das Coxsackievirus B3 sowie das SARS-Coronavirus und das MERS-Coronavirus (getestet in Huh-T7-Leberzellen). Tabelle 1 EC50-Werte für die Leitsubstanz DZL08

- Im Organismus kann diese Amidgruppe insbesondere auch von Proteasen der Wirtszelle prozessiert werden und so die Bioverfügbarkeit der gewünschten Verbindung verringern.

- Die Aktivität der erfindungsgemäßen Wirkstoffe im Vergleich zur Leitsubstanz DZL08 ist vor allem für Betacoronavirus-Proteasen (SARS-CoV Mpro, MERS-CoV Mpro, 2019-nCoV Mpro) gut. So zeigte etwa die Substanz RHCDS1c gegen diese Proteasen IC50-Werte von 0.90 uM, 0.58 uM und 0.67 uM (gemessen wurde die Inhibierung der Spaltung eines Standardsub-strats durch den Wirkstoff in einem fluoreszenz-basierten Test). Diese Werte sind leicht verbessert im Vergleich zu den mit der Leitsubstanz DZL08 erzielten.
- Die Bioverfügbarkeit der Leitsubstanz DZL08 wurde auch im Rahmen einer externen CRO-Studie eingehend charakterisiert und die Ergebnisse verifiziert.
- Pharmacokinetische Studien zeigen, dass die erfindungsgemäße Verbindung RHCDS1e gegenüber der Leitsubstanz DZL08 (t1/2 =0,33 ± 0,0 h) eine um 50% verbesserte Halbwertszeit von (t1/2 = 0,45 ± 0,1h) aufweist. Durch die erfindungsgemäße Stabilisierung der P3-P2-Amidbindung konnte ebenfalls die hohe Plasmaproteinbindung gegenüber DZL08 (99 %) auf 94 % gesenkt werden.

Nach 30 Minuten verblieben 80% der Substanz metabolisch stabil in Maus-Mikrosomen und 60% im Fall von menschlichen Mikrosomen. Keinerlei Anzeichen für Toxizität in Mäusen wurden beobachtet. Pharmacokinetische Untersuchungen nach subkutaner Injektion von RHCDS1e in CD-1-Mäuse (20 mg/kg) zeigten, dass die Verbindung nur für etwa 4 Stunden im Plasma verbleibt, aber bis zu 24 Stunden über den Urin ausgeschieden wird. - In einem weiteren Aspekt der Erfindung kann der erfindungsgemäße Wirkstoff in der Medizin zur Therapie einer Erkrankung verwendet werden. Insbesondere zur Therapie einer Erkrankung die von RNA-Viren verursacht wird. Ganz besonders zur Therapie einer Erkrankung, die von Coronaviren, Picornaviren, Enteroviren und/oder Noroviren verursacht wird.
- Der erfindungsgemäße Wirkstoff kann dabei in Form einer pharmazeutischen Zubereitung zugeführt werden.
- Solche Zubereitungen sind dem Fachmann allgemein bekannt. Insbesondere werden solche pharmazeutischen Zubereitungen und Darreichungsformen in der
US 10,189,810 B2 - Material und Methoden
- Im folgenden soll die Allgemeinheit der Lehre nicht einschränkend die exemplarische Herstellung der erfindungsgemäßen Verbindungen beschrieben werden.
- Allgemeines Vorgehen:
- Die verwendeten Reagenzien und Edukte wurden im Handel aus dem Fachmann bekannten kommerziellen Quellen bezogen und ohne weitere Vorbehandlung eingesetzt.
- Für die Dünnschichtchromatographie (DC) wurden HSGF 254 Kieselgel-Platten (Dicke 0.15 - 0.2 mm) verwendet.
- Alle Produkte wurden mit NMR und Massenspektroskopie (MS) charakterisiert.
- 1H-NMR wurde bei 300 MHz gemessen, die chemische Verschiebung (δ) ist in ppm angegeben, als Standard wurde Tetramethylsilan verwendet. Die Kopplung der Protonen wird als Singulett (s), Dublett (d), Triplett (t), Multiplett (m), und breit (br) charakterisiert.
- Für die MS wurde ein Bruker ESI ion-trap HCT Ultra verwendet.
- Die HPLC Daten wurden auf einem LC20A (Shimadzu Corporation) Säulen: GIST C18 (5 µm, 4.6x150mm) ternäres Lösungsmittelsystem (Methanol/Wasser, Methanol/ 0.1% HCOOH in Wasser oder Methanol/0.1 % Ammoniak in Wasser). Diereinheit wurde per reversed-phase HPLC bestimmt und lag bei ≥95% bei allen Proben.
- Synthese der Komponente 1
- Eine Lösung von (R)-2-Amino-3-cyclopropylpropansäure oder (R)-2-Amino-3-cyclohexylpropansäure (7.74 mmol) in 2N H2SO4 (15 mL) wird bei 0 °C gerührt. Dann werden tropfenweise NaNO2 (5.34 g, 77.4 mmol) in H2O (6 mL) hinzugefügt. Die Lösung wird 3 h bei 0°C gerührt und anschließend auf 20°C gebracht und für 16 h bei 20°C gerührt. Die Mischung wird mit MTBE (50 mL) extrahiert. Die organische Phase wird anschließend über wasserfreiem Na2SO4 unter Vakuum getrocknet. (Ausbeute an Komponente 1: 50-75%, farbloses Öl).
- Synthese der Komponente 2
- SOCl2 (0.8 mL, 11.34 mmol) wird bei 0°C tropfenweise zu einer Lösung von Komponente 1 (5.72 mmol) in MeOH (20 mL) zugegeben. Anschließend wird die Mischung 1.5 h bei 20°C gerührt. Unter Vakuum wird das Lösungsmittel entfernt und zwecks Reinigung über Kieselgel chromatographiert (PE/EA = 1/1). (Ausbeute an Komponente 2: 30-59 %, farbloses Öl).
- Methyl (R)-3-cyclopropyl-2-hydroxypropanoat 2a:
- 1H NMR (300 MHz, CDCl3) δ 4.35 (dd, J1 = 9.0 Hz, J2= 4.2 Hz, 1H), 3.80 (s, 3H), 1.81-1.72 (m, 2H), 0.92-0.68 (m, 1H), 0.50-0.43 (m, 2H), 0.15-0.05 (m, 2H).
- Methyl (R)-3-cyclohexyl-2-hydroxypropanoat 2b:
- 1H NMR (300 MHz, CDCl3) δ 4.39-4.34 (m, 1H), 3.82 (s, 3H), 1.82-1.48 (m, 8H), 1.29-1.12 (m, 4H), 1.00-0.85 (m, 2H).
- Synthese der Komponente 3
- Komponente 2 (5.32 mmol) wird in DCM (10 mL) gelöst und auf 0 °C gekühlt. Portionsweise werden 2,6-Lutidin (1.5 mL, 13.26 mmol) und Tf2O (3.3 g, 11.87 mmol) zugegeben. Die Mischung wird 30 min bei 0 °C gerührt. Die Mischung wird mit Kochsalzlösung und 1N HCl (3:1 v/v) gewaschen und anschließend mit MTBE extrahiert und mit wasserfreiem Na2SO4 unter Vakuum getrocknet. (Ausbeute an Komponente 3: 82 %, braunes Öl).
- Synthese der Komponente 5
- Tert-butyl (2-oxo-1,2-dihydropyridin-3-yl)carbamat (379 mg, 1.8 mmol) wird in THF (15 mL) gelöst. Das NaH (115 mg, 2.80 mmol, 60% in Öl) wird bei 0 °C zugegeben und dann 30 min gerührt. Komponente 3 (515 mg, 1.86 mmol) in THF (10 mL) wird zugegeben. Die Mischung wird für 20 h bei 25 °C gerührt. Unter Vakuum wird das Lösungsmittel entfernt und zwecks Reinigung über Kieselgel chromatographiert (PE/EA). (Ausbeute an Komponente 5: 56-60%, hellgelber Feststoff).
- Methyl (S)-2-(3-((tert-butoxycarbonyl)amino)-2-oxopyridin-1(2H)-yl)-3-cyclopropylpropanoat 5a:
- 1H NMR (300 MHz, DMSO-d6) δ 7.83-7.78 (m, 2H), 7.35 (dd, J1 = 7.2 Hz, J2= 1.5 Hz, 1H), 6.30 (t, J = 7.2 Hz, 1H), 5.36 (dd, J1 = 10.8 Hz, J2= 4.5 Hz, 1H), 3.57 (s, 3H), 1.81-1.62 (m, 2H), 1.48 (s, 9H), 0.55-0.48 (m, 1H), 0.34-0.29 (m, 2H), 0.15-0.12 (m, 1H), 0.04-0.01 (m, 1H). - Methyl (S)-2-(3-((tert-butoxycarbonyl)amino)-2-oxopyridin-1(2H)-yl)-3-cyclohexylpropanoat 5b:
- 1H NMR (300 MHz, DMSO-d6) δ 7.82-7.76 (m, 2H), 7.35 (dd, J1 = 7.5 Hz, J2= 1.5 Hz, 1H), 6.30 (t, J= 7.5 Hz, 1H), 5.35 (dd, J1 = 11.1 Hz, J2= 4.5 Hz, 1H), 3.56 (s, 3H), 2.10-1.88 (m, 2H), 1.78-1.72 (m, 1H), 1.65-1.44 (m, 13H), 1.14-0.82 (m, 6H). ESI-MS (m/z): 379 [M + H]+.
- Synthese der Komponente 6
- Zur Komponente 5 (1.65 mmol) in MeOH (15 mL) und H2O (3 mL) wird LiOH.H2O (139 mg, 3.31 mmol) gefügt. Die Mischung wird für 1 h bei 20 °C gerührt. Mit 1N HCl wird ein pH=6-7 eingestellt. Unter Vakuum wird das Lösungsmittel entfernt und zwecks Reinigung über Kieselgel chromatographiert (DCM/MeOH = 10/1) (Ausbeute an Komponente 6: 452 mg, 84%, hellgelber Feststoff).
- (S)-2-(3-((tert-Butoxycarbonyl)amino)-2-oxopyridin-1(2H)-yl)-3-cyclopropylpropansäure 6a:
- 1H NMR (300 MHz, DMSO-d6) δ 13.11(s, 1H), 7.81-7.77 (m, 2H), 7.36 (dd, J1 = 6.9 Hz, J2= 1.5 Hz, 1H), 6.30 (t, J = 6.9 Hz, 1H), 5.35 (dd, J1 = 10.5 Hz, J2= 4.5 Hz, 1H), 1.80-1.69 (m, 2H), 1.47 (s, 9H), 0.53-0.48 (m, 1H), 0.32-0.29 (m, 2H), 0.14-0.11 (m, 1H), 0.03-0.00 (m, 1H). - (S)-2-(3-((tert-Butoxycarbonyl)amino)-2-oxopyridin-1 (2H)-yl)-3-cyclohexylpropansäure 6b:

- Synthese der Komponente 8:
- HOBT (245 mg, 1.82 mmol) und EDCI (349 mg, 1.82 mmol) werden zu einer Lösung von Komponente 6 (1.65 mmol) in DCM (20 mL) hinzugefügt. Die Mischung wird für 1 h bei 0 °C gerührt. Anschließend wird Komponente 7 Methyl-(S)-2-amino-3-((S)-2-oxopyrrolidin-3-yl)propanoat (307 mg, 1.65 mmol) zugegeben, und mit Et3N pH = 9 eingestellt. Die Mischung wird für 24 h bei 0 °C gerührt. - Synthese der Komponente 11 (Generelle Methode):
- Essigsäure (26 mg, 0.44 mmol) and Isocyanid (0.22 mmol) werden zu einer Lösung der Komponente 10 (0.22 mmol) in DCM (15 mL) gegeben. Die Mischung wird für 24 h bei 20 °C gerührt. Unter Vakuum wird das Lösungsmittel entfernt und zwecks Reinigung über Kieselgel chromatographiert (DCM/MeOH = 20/1) (Ausbeute an Komponente 11: 57-65 %, annähernd weißer Feststoff).
- Synthese der Komponente 12 (Generelle Methode):
- Zur Komponente 11 (0.13 mmol) in MeOH (15 mL) und H2O (3 mL) wird LiOH.H20 (11 mg, 0.26 mmol) hinzugefügt. Die Mischung wird für 20 min. bei 20 °C gerührt. Mit 1N HCl wird ein pH=6-7 eingestellt. Unter Vakuum wird das Lösungsmittel entfernt und zwecks Reinigung über Kieselgel chromatographiert (DCM/MeOH = 10/1) (Ausbeute an Komponente 12: 90-95%, annähernd weißer Feststoff).
- Synthese der Komponente 13 (Generelle Methode)
- Komponente 12 (0.115 mmol) wird in DCM (15 mL) gelöst, Dess-Martin Periodinan (58 mg, 0.14 mmol) und NaHCO3 (4 mg, 0.05 mmol) werden hinzugefügt. Die Mischung wird für 1 h bei 25 °C gerührt. Unter Vakuum wird das Lösungsmittel entfernt und zwecks Reinigung über Kieselgel chromatographiert (DCM/MeOH = 10/1) (Ausbeute an Komponente 13: 69-80%, annähernd weißer Feststoff).
- Tert-butyl(1-((S)-1-(((S)-4-(benzylamino)-3,4-dioxo-1-((S)-2-oxopyrrolidin-3-yl)butan-2-yl)amino)-3-cyclopropyl-1-oxopropan-2-yl)-2-oxo-1,2-dihydropyridin-3-yl)carbamat 13a:
- 1H NMR (300 MHz, DMSO-d6) δ 9.25 (d, J = 5.4 Hz, 1H), 9.00 (dd, J1 = 14.1 Hz, J2= 7.2 Hz, 1H), 7.79-7.69 (m, 3H), 7.35-7.22 (m, 5H), 6.30-6.24 (m, 1H), 5.69-5.61 (m, 1H), 4.97 (s, br, 1H), 4.29 (s, 2H), 3.17-3.09 (m, 2H), 2.30-2.15 (m, 2H), 1.91-1.62 (m, 5H), 1.46 (s, 9H), 0.52-0.47 (m, 1H), 0.33-0.29 (m, 2H), 0.14-0.11 (m, 1H), 0.05-0.02 (m, 1H). ESI-MS (m/z): 594 [M + H]+.

- 1H NMR (300 MHz, CDCl3) δ 8.78-8.50 (m, 1H), 8.01-7.92 (m, 1H), 7.65 (d, J = 7.5 Hz, 1H), 7.10-6.90 (m, 2H), 6.35-6.14 (m, 2H), 5.85-5.75 (m, 1H), 5.25-5.10 (m, 1H), 3.24-3.13 (m, 2H), 2.75-2.71 (m, 1H), 2.49-2.20 (m, 1H), 2.10-1.81 (m, 3H) 1.80-1.52 (m, 8H), 1.49 (s, 9H), 1.25-1.01 (m, 4H), 1.00-0.73 (m, 4H), 0.56-0.51 (m, 2H). - Synthese der Komponente 14 (Generelle Methode):
- Zur Komponente 13 (0.11 mmol) wird eine Lösung von 4N HCl/EA (30 mL) zugegeben. Die Mischung wird für 1 h bei 25 °C gerührt.Anschließen wird unter vermindertem Druck das Lösungsmittel entfernt und der Rückstand unter Rühren mit Diethylether (2 mL) versetzt. Der weiße Feststoff wird ausgefällt, filtriert und der Filterkuchen wird mit Diethylether (1 mL) gewaschen. (Ausbeute an Komponente 14: 58-94 %, weißer Feststoff).
- (S)-3-((S)-2-(3-amino-2-oxopyridin-1(2H)-yl)-3-cyclopropylpropanamido)-N-benzyl-2-oxo-4-((S)-2-oxopyrrolidin-3-yl)butanamid 14a:
- 1H NMR (300 MHz, DMSO-d6) δ 9.25 (d, J = 5.4 Hz, 1H), 9.06 (dd, J1 = 14.1 Hz, J2= 7.2 Hz, 1H), 7.73-7.59 (m, 2H), 7.34-7.21 (m, 5H), 6.30 (t, J= 7.5 Hz, 1H), 5.75-5.64 (m, 1H), 5.00 (s, br, 1H), 4.30 (s, 2H), 3.17-3.09 (m, 2H), 2.26-2.12 (m, 2H), 1.99-1.63 (m, 5H), 1.46 (s, 9H), 0.49-0.46 (m, 1H), 0.32-0.30 (m, 2H), 0.15-0.14 (m, 1H), 0.05-0.00 (m, 1H). ESI-MS (m/z): 494 [M + H]+.

- Antiviraler Wirkstoff der folgenden Form:
- Antiviraler Wirkstoff nach einem oder mehreren der vorangegangenen Ansprüche zur Verwendung in der Medizin.
- Antiviraler Wirkstoff nach einem oder mehreren der vorangegangenen Ansprüche zur Verwendung für die Therapie einer Erkrankung.
- Antiviraler Wirkstoff nach dem vorangehenden Anspruch zur Verwendung für die Therapie einer Erkrankung, wobei die Erkrankung von RNA-Viren verursacht wird.
- Antiviraler Wirkstoff nach den beiden vorangehenden Ansprüchen zur Verwendung für die Therapie einer Erkrankung, wobei die Erkrankung von Coronaviren, Picornaviren, Enteroviren und/oder Noroviren verursacht wird.
