VEGF是一种强促***原,可诱导血管发生(初始血管发生(vasculogenesis)和再生血管发生(angiogenesis))。初始血管发生是血管通过胚胎发生期间早期内皮细胞分化的从头发育。另一方面,再生血管发生是预先存在的血管通过新组织形成新的毛细血管。因此,VEGF在损伤的组织中发现,缺氧对VEGF的诱导随后导致血管发生,这是修复机制的一部分。VEGF也与绝经前妇女子宫内膜的每月循环再生有关。
天然VEGF证明在许多种(包括人和啮齿类动物)之间具有相当高的序列同源性。一般而言,它是一种糖蛋白,通过单一基因的多种备选剪接变体的二聚作用产生。121、165、189、206个氨基酸残基的多肽链已经鉴定(Houck,K.A.等人1996;Mol.Endocrinol.5:1806-1814)。其中已经证明121个氨基酸残基的同种型VEGF121是血管发生中最有效的分子。

其中A是VEGF,A′是倒置或反转的VEGF序列。
尽管甘氨酸是优选的,但是也可以用脂肪族侧链氨基酸代替上述通式中的一个或多个Gly残基。
尽管通过计算机辅助能量最低化模建检查,通常认为本发明的肽衍生物太小,以至单独时没有最佳免疫原性,但是已经发现,当通过载体生物太小,以至单独时没有最佳免疫原性,但是已经发现,当通过载体结合部分与一种载体偶联时,这些肽衍生物引发强烈且可能具有保护性的免疫应答。它们的偶联物因此特别适合在对抗肿瘤血管发生的免疫治疗中使用。不希望被理论所束缚,可以认为肽与载体通过载体结合部分偶联产生与天然VEGF序列基本具有相同构象的衍生物。
根据本发明的新衍生物可以利用大量标准技术产生,对于肽,包括Merrifield固相法,其中向与固体基质连接的成长多肽上逐步添加氨基酸(Merrifield,R.B 1962;Fed.Proc.Amer.Soc.Biol.21:412和Merrifield,R.B.1963;Jour.Amer.Chem.Soc.85:2149),以及常规FMOC化学法。希望时,在链合成期间,成长链中的氨基酸的反应性侧链基团可以受到保护。分支结构可以用类似的技术制备。
当新衍生物是线性肽时,它们也可以利用本领域公知的技术,通过重组DNA表达制备,如Sambrook等人,《分子克隆:实验手册》,第二版,1989所述。
因此,本发明也提供编码本发明的VEGF肽衍生物的一种核酸分子,和与之序列互补的核酸分子。
根据本发明的另外一个方面,我们提供了一种表达载体,其含有本发明的核酸分子。这种载体可能适合在以下宿主中表达:微生物,可以是原核或真核生物,例如大肠杆菌或酵母,或者植物或动物,例如哺乳动物细胞。
本发明的衍生物与载体的偶联可以利用本领域公知的方法实现,例如用异双功能交联剂处理。当通过末端半胱氨酸(或N-乙酰半胱氨酸)偶联时,连接剂可以是间马来酰亚胺苯甲酰-N-羟基磺基琥珀酰胺酯;在此情况下,马来酰亚胺修饰肽载体中的一个或多个赖氨酸侧链,在末端半胱氨酸残基处形成硫醚键。也可以使用本领域公知的其它偶联试剂,例如碳二亚胺偶联。
可以使用本领域公知用于这些用途的任何载体,包括结核菌素、破伤风类毒素、白喉类毒素、匙孔血蓝蛋白、谷胱甘肽S-转移酶或其衍生物的纯化的蛋白质衍生物。
物也可以利用重组DNA方法制备,其中编码该偶联分子的核酸分子在适当的宿主细胞中表达。
本发明的新VEGF偶联物可以在一种免疫治疗方法中使用,治疗与血管生成活性和/或VEGF水平正常或升高有关的疾病,这是优于当前可用的方法的一种方法。患者顺应性应当提高,因为施用频率低于使用现有治疗的情况,且可以避免不希望的副作用。
因此根据另外一方面,本发明提供一种药物组合物,其含有根据本发明的VEGF偶联物,以及一种或多种药学可接受的载体或赋形剂。
根据另一方面,本发明提供一种根据本发明的VEGF偶联物,用于治疗或预防。
根据另外一方面,本发明提供一种治疗人或非人(例如哺乳动物)患者的肿瘤的方法,包括对该患者施用有效量的根据本发明的VEGF偶联物。
因此可以利用该方法控制形成的肿瘤的血管形成,并阻止癌症通过转移扩散。
在一个实施方案中,本发明的方法是一种针对转移免疫的方法,即不需要在患者中检测到转移。在该实施方案中,当检测到肿瘤时,例如在确定为恶性肿瘤之前,或许在开始或已经结束对检测的肿瘤的治疗(例如化学治疗、手术或放射治疗)之前,可以施用该偶联物。

这些组合物适当地配制为单位剂型,例如用于粘膜、肠胃外或经口施用。
实际治疗方案或预防方案、制剂和剂量在很大程度上取决于个体患者,可由医生根据个体情况计划。
制剂类型应当适合施用途径。例如,偶联的类似物在PBS、盐水或注射用水中的无菌水悬液,任选地与一种或多种免疫佐剂(例如氢氧化铝、皂苷、quil A、胞壁酰二肽、矿物油或植物油、基于囊泡的佐剂、非离子嵌段共聚物或DEAE葡聚糖)一起,通过皮下或肌肉内注射肠胃外施用。也可以使用其它成分,如防腐剂。
注射剂量可以是1-100μg肽相当物,施用频率可以从每3个月或6个月一次,到每年一次或每5年一次。
为了经口施用,偶联的衍生物可以配制为片剂、液体、胶囊等。剂量为1-1000μg肽相当物,根据产品的生物利用度以一定的间隔施用。
根据另外一个方面,本发明提供一种方法,用于在人或非人患者中获得最大VEGF阻断,相当于或者超过化学或放射治疗获得的,该方法包括对该患者施用有效量的一种根据本发明的VEGF偶联物。
此外,本发明的VEGF偶联物也可以对女性或雌性非人哺乳动物施用,作为一种避孕方法,或者治疗子宫内膜异位症。这些用途、方法等是本发明的另外一些方面。

实施例1:肽的产生
肽根据Fmoc固相肽合成策略用Protein Technologies,Symphony肽合成仪合成。使用的树脂是含有Rink酰胺接头的Tentagel S-NH2。对于Cys、His、Asn和Gln,使用的Fmoc氨基酸的侧链保护基是Trt,对于Tyr、Thr、Asp、Glu和Ser是tBu;对于Lys是Boc,对于Trp是吲哚N,对于Arg是Pmc。羧基的活化利用TBTU/HOBt/DIPEA实现,所有偶联都在DMF中进行。Fmoc基团的脱保护使用溶于DMF的20%哌啶完成。使用溶于TFA的5%茴香醚/5%苯甲硫醚/5%EDT/3%水/2%TES,经1小时,从树脂上切下肽。使用Waters Deltaprep 4000上的40mm×210mm Deltapak C18径向压力柱,通过RP-HPLC纯化这些肽,并且使用Kratos Maldi 3通过MALDI-TOF并且通过AAA表征。
对于树状聚合物Fmoc,用上述方法连接Lys(Fmoc)-OH,产生自由肽延伸的氨基。因此必须提高使用的Fmoc氨基酸的量。

实施例2:偶联程序
向匙孔戚血蓝蛋白(KLH)在磷酸缓冲液(PBS)中的溶液中加入150摩尔过量的S-MBS,间马来酰亚胺基苯甲酰-N-羟基磺基琥珀酰亚胺酯,在密封瓶中在20-25℃下搅拌0.5小时。
在PBS中通过层析除去过量的S-MBS交联剂(使用G-25 Sephadexcourse matrix凝胶排阻)。收集活化KLH峰,测定游离马来酰亚胺基,如下所述使用。
向获得的KLH载体蛋白溶液中加入2倍摩尔过量的VEGF衍生肽。获得的溶液在密封容器中在2-8℃下搅拌可达2小时。
如上所述通过凝胶排阻层析从游离肽中纯化该偶联物。
将终偶联物稀释为使用浓度,如希望地配制。
按照该程序,实施例1的VEGF衍生物(1)-(4)分别与活化KLH等份偶联。
实施例3:免疫研究
实施例1的4种VEGF衍生物如实施例2所述分别与KLH等份偶联,配制方法是吸附于在生理盐水(0.9%(w/v))载体中的0.4%(w/v)氢氧化铝凝胶上(铝胶,Superfos s/a,丹麦)。所有偶联物均作为25μg/ml肽相当物溶液使用。在5个处理组中使用C57BL/6小鼠,每组23只小鼠。处理组接受:
A组:铝胶/盐水安慰剂疫苗0.2ml/小鼠。
B组:N-乙酰-VEGF 6-Gly-Cys,每只小鼠5μg肽相当物/衍生物免疫治疗剂,在0.2ml疫苗中。
C组:N-乙酰-Cys-Gly-VEGF 7,每只小鼠5μg肽相当物/衍生物免疫治疗剂,在0.2ml疫苗中。
D组:N-乙酰-VEGF 8-Gly-Cys,每只小鼠5μg肽相当物/衍生物免疫治疗剂,在0.2ml疫苗中。
E组:N-乙酰-Cys-Gly-VEGF 9,每只小鼠5μg肽相当物/衍生物免疫治疗剂,在0.2ml疫苗中。
施用途径为皮下途径,在研究过程中每只小鼠接受3个独立剂量的指定的试验样品。在施用的每天和研究终止时记录每只小鼠的体重。
实验程序
在第-50天,随后在第-28天和第-7天,小鼠单次皮下施用载体,或试验样品。在研究的第-1天和+14天制备血清进行分析。
通过心脏穿刺将每种血样收集到血清试管中,使之凝结,然后在采样45分钟内离心,产生血清。
血清标本尽可能快地冷冻于大约-20℃下。

通过利用酶联免疫吸附测定(ELISA)滴定血清中存在的抗VEGF肽抗体,测定每个血清标本中由于处理引起的抗体应答产生。
该测定如下进行:
用100μl在PBS中稀释的检测底物,例如N-乙酰-VEGF6-Gly-Cys-BSA(10μg肽相当物/孔),包被96孔Nunc Maxisorp微量滴定板,20-25℃1小时。同时向不同的孔中加入100μl PBS,作为底物空白。
用200μl磷酸缓冲液(PBS)/0.1%Tween 20洗板3次。
用PBS/0.1%Tween 20稀释血清标本至适当的稀释度。稀释度一般为:
i)1/100-5μl小鼠血清+495μl PBS/0.1%Tween 20
ii)1/1000-20μl(i)+180μl PBS/0.1%Tween 20
iii)1/2000-10μl(i)+190μl PBS/0.1%Tween 20
iv)1/5000-4μl(i)+196μl PBS/0.1%Tween 20
将适当稀释的血清(100μl)加入适当的孔中,20-25℃温育1小时,使底物与抗体结合。
用200μl PBS/0.1%Tween 20洗板3次。
用PBS/0.1%Tween 20以1∶5000稀释兔抗小鼠IgG过氧化物酶偶联物,即1μl IgG过氧化物酶+5ml PBS/0.1%Tween 20。其结合小鼠血清抗体,并且使得抗体可以检测。
将100μl稀释的IgG过氧化物酶加入适当的孔中,在20-25℃下放置45分钟。
用200μl PBS洗板3次。
将250μl等份过氧化物酶底物3,3’,5,5’-四甲基联苯胺(TMB)加至含有4μl 30%过氧化氢的25ml 0.1M乙酸钠缓冲液pH5.5中。
将制备的100μl TMB底物加入适当的孔中,包括空白孔。发生显色反应,其中发生抗体/底物结合。在20-25℃下放置15分钟,然后向每孔中加入50μl 10%硫酸终止反应。
读取平板在405nm下的吸光度,这是过氧化物酶在TMB底物上的反应产生的,它与第一(抗VEGF)抗体键的量成正比。用统计学软件包(SAS研究所,1997)分析获得的吸光度读数,测定滴度。

与抗体滴度数据平行,检测所有动物在体重、呼吸困难和全身表现方面的总体生理学改变,作为毒性或有害影响的全面评价。
所有处理组均未记录到不良反应,表明VEGF处理在产生抗VEGF抗体方面有效,而在动物中没有有害的生理学效应。
实施例4:鼠抗转移研究
为了评价VEGF免疫治疗疫苗的抗转移能力,如此处所述处理实施例3所述的接种小鼠。另外,平行处理既不接受疫苗也不接受安慰剂的另外一组小鼠,作为鼠模型对照组。
实验程序
在研究的第0天,每只小鼠均静脉内注射0.1ml B16/F10黑素瘤细胞悬液。在研究的第14天杀死小鼠,从每只小鼠中取出肺。肺称重,之后在布安氏液中固定,以显示施用B16/F10细胞产生的所有黑素瘤集落。固定后,计数来自每只小鼠的肺表面存在的集落数量。

表3证明用VEGF偶联物免疫的所有组的肺重量均增加。与鼠模型对照组相比,证实疫苗处理的B、C、D组肺重量增加可达15%。同时,与既不接受疫苗也不接受安慰剂的鼠模型对照相比,免疫的安慰剂对照组显示肺重量增加极少。
表4证明,与只用疫苗安慰剂处理的安慰剂组(A组)相比,用VEGF偶联物免疫的组的肺黑素瘤集落计数降低超过三分之一。对于用含有VEGF8和VEGF9的疫苗处理的组,集落计数降低最大,表明转移得到最佳控制,也是统计学显著的。
在研究的第14天,所有VEGF疫苗处理组都有良好的抗VEGF肽IgG滴度。但是,用制剂VEGF8和VEGF9免疫后产生的IgG证明了明显控制肿瘤扩散的不同的内在能力。这强调了针对肽VEGF8和VEGF9的IgG分子可能具有最强的控制肿瘤转移的活性。这些数据证明,在这些研究使用的鼠模型上,针对肽VEGF8和VEGF9的IgG分子与天然VEGF特异性相互作用。
Claims (20)
