技术领域
本发明属于农业微生物技术领域。更具体地说,本发明涉及一种热带根瘤菌FM-19及其应用。
背景技术
微生物与药用植物间存在复杂的互作关系,根际微生物可直接作用于药用植物影响其产量及质量。根瘤菌是一类广泛分布于土壤中的有益固氮微生物,为革兰氏阴性细菌,可以与豆科植物形成根瘤或茎瘤共生固氮***,将氮气转化成植物需要的氮素营养,是最高效的生物固氮体系。毛鸡骨草(Abrus mollis Hance)被收录于《广西中药材标准》,因其与鸡骨草功效相似并且生长速度快,在民间常将毛鸡骨草代替鸡骨草药材入药。随着鸡骨草药材需求量不断增加,也导致了人工栽培成为常态化。但是传统农业大量使用农药化肥的种植模式也被用于中药种植,造成了药材有效成分降低、土壤肥力下降等问题。因此,为了充分发挥根瘤菌与豆科植物的共生固氮作用,减少化肥施用,提高药材产量和质量,需要筛选高效固氮菌株进行人工接种。

本发明的一个目的是解决至少上述问题,并提供至少后面将说明的优点。
本发明的一个目的是提供一种热带根瘤菌菌株FM-19,该菌株能够显著提高毛鸡骨草生物量、结瘤数、单株根瘤重量、根瘤固氮酶活性及藤茎中总黄酮的含量。
为了实现本发明的这些目的和其它优点,提供了一株热带根瘤菌(Rhizobiumtropici)FM-19,其特征在于,该菌株保藏于广东省微生物菌种保藏中心,保藏编号为GDMCCNo.63403。
一种促进毛鸡骨草生长和黄酮类活性成分积累的菌剂,包含有所述的热带根瘤菌FM-19。
一种毛鸡骨草的种植方法,毛鸡骨草种子催芽,芽苗长至1-2cm时移栽到栽培介质中,待长出真叶后,在栽培介质中接种所述热带根瘤菌FM-19。
优选的是,毛鸡骨草种子表面消毒处理后再进行催芽处理。
优选的是,消毒处理方法为:用0.1%升汞溶液浸泡5-15min进行表面消毒,然后浸泡于40-60℃的热水中,自然冷却至室温。
优选的是,每隔10-15d接种1次,接种3-6次。
优选的是,热带根瘤菌FM-19的接种OD600为0.8-1.0。

下面结合附图对本发明做进一步的详细说明,以令本领域技术人员参照说明书文字能够据以实施。

需要说明的是,下述实施方案中所述实验方法,如无特殊说明,均为常规方法,所述试剂和材料,如无特殊说明,均可从商业途径获得。
<试验内容>
一、菌株的分离纯化
2022年6月在广西玉林制药有限责任公司福绵种植基地采集毛鸡骨草根瘤。将采集的根瘤用水冲洗干净,并用滤纸吸干表面水分,在95%的乙醇中浸泡30s以消除表面张力,倒去乙醇再用0.2% HgCl2消毒5min,进行表面的消毒,最后用无菌水冲洗5-6次,用无菌滤纸吸干水分。取0.5g消毒后的根瘤置于灭菌的研钵中,加入4.5mL无菌水研磨,梯度稀释至10-3,将10-2和10-3稀释液各100μL涂布在YMA培养基,YMA培养基的配方如表1,倒置于28℃培养箱中黑暗培养,直到单菌落出现。待长出单菌落后观察,菌落形态:根瘤菌的菌落为圆形、乳白色、半透明、边缘整齐,快生型根瘤菌在YMA平板上培养3-5天菌落直径为2-4mm,其代时为2-4h;慢生型根瘤菌在YMA平板上培养5-7天内菌落直径不超过1mm,其代时为6-8h。菌体形态:挑取单菌落菌苔图片进行革兰氏染色,油镜下观察,杆状,0.5~0.9μm×1.2~6.0μm,一般含有聚-β-羟基丁酸盐颗粒,革兰氏阴性,无芽孢。

本实施例的菌株为快生型,在菌落形态上呈乳白色半透明圆形,粘质较多,在YMA平板上生长3天后直径为1.0-2.0mm(图1);最适生长条件为:30℃,转速200r/min;经革兰氏染色反应镜检为阴性(G-),呈小短杆状(图2)。
二、分子鉴定
使用试剂盒提取菌株总DNA,使用通用引物P1-P6扩增菌株16S rDNA片段,PCR扩增产物在1.0%琼脂糖凝胶电泳检测,纯化回收送到上海生工生物工程技术服务有限公司进行序列测定,测序结果如SEQ ID No.1所示;将所得到的序列在NCBI进行BLAST检索,从中选择了10个相似性高的模式菌株作为参比菌株,利用MEGA软件中NJ法进行16S rDNA基因***发育树的构建。由对比结果和***发育树(图3)可知,根瘤菌FM-1916S rDNA基因序列与热带根瘤菌(Rhizobium tropici)菌株的模式菌株Rhizobium tropici strain BF-R18的序列相似性达到99.92%,并位于同一进化分支,从而确定根瘤菌FM-19为根瘤菌属Rhizobium的新菌株系。
该菌株与2023年4月25日保藏于广东省微生物菌种保藏中心,保藏地址广州市先烈中路100号大院59号楼5楼,广东省科学院微生物研究所,保藏编号为GDMCC No.63403。

<实施例1>
一种毛鸡骨草的种植方法,包括以下步骤:
1)用0.1%升汞溶液浸泡10min进行表面消毒,然后浸泡于50℃的热水中,自然冷却至室温,种子吸胀后置于无菌发芽盒内,在25℃培养箱内催芽;
2)芽苗长至1-2cm时移栽到栽培介质中,待长出真叶后,在栽培介质中接种所述热带根瘤菌FM-19,热带根瘤菌FM-19的接种OD600为0.8,每隔10d接种1次,接种3次,持续种植,即得毛鸡骨草。
<效果试验>
毛鸡骨草种子灭菌培养:挑选饱满、大小一致的毛鸡骨草种子,用0.1%升汞溶液浸泡10min进行表面消毒,然后浸泡于50℃的热水中让其自然冷却。种子吸胀后置于无菌发芽盒内,在25℃培养箱内催芽。
菌体培养:配置YMA液体培养基分装,每瓶100mL,121℃灭菌20min;接种已纯化并达到对数生长期的根瘤菌菌液,接种量为1%,在30℃的摇床200r/min震荡培养,菌液吸光值OD600达到0.8-1.0时做接种使用。
试验于广西中医药大学温室大棚中进行,将蛭石、商品育苗基质按1:2混合均匀后高温灭菌,选取芽长1-2cm的芽苗移入盆栽中,每盆移栽5棵,生长两周后间苗,并进行接菌处理,接种量为10mL/株,每隔10d接种1次,共接种3次,并在植株生长60天后,测定各项指标,结果如图4、图5、图6所示。图中的数据是具有标准误差的三个生物学重复的平均值。。
由图4可知,接种根瘤菌FM-19对毛鸡骨草地上部和地下部生物量积累具有显著影响,即与不接种根瘤菌相比,接种处理的毛鸡骨草植株地上部鲜重和干重分别增加了157.42%和126.46%,地下鲜重和地下干重分别增加了154.32%和98.0%
由图5可见,接种根瘤菌FM-19对毛鸡骨草结瘤数、单株根瘤重和根瘤固氮酶活性具有显著影响,与不接种根瘤菌相比,接种处理的毛鸡骨草结瘤数、单株根瘤重和根瘤固氮酶活性分别增加了242.82%、980.45%和121.48%。
由图6可见,接种根瘤菌FM-19对毛鸡骨草藤茎中总黄酮积累具有显著影响,与不接种根瘤菌相比,接种处理的毛鸡骨草藤茎中总黄酮含量增加了42.70%。
本发明所述的一种热带根瘤菌(Rhizobium tropici)菌株FM-19,经试验表明:使用菌株FM-19具有明显的增产效果,毛鸡骨草的生物量、结瘤数、单株根瘤重、根瘤固氮酶活性和藤茎中总黄酮含量均显著高于不接种的植株。可见热带根瘤菌(Rhizobium tropici)菌株FM-19可以用于毛鸡骨草生产上大面积推广使用。
尽管本发明的实施方案已公开如上,但其并不仅仅限于说明书和实施方式中所列运用,它完全可以被适用于各种适合本发明的领域,对于熟悉本领域的人员而言,可容易地实现另外的修改,因此在不背离权利要求及等同范围所限定的一般概念下,本发明并不限于特定的细节和这里示出与描述的图例。
Claims (8)
1.一株热带根瘤菌(Rhizobium tropici)FM-19,其特征在于,该菌株于2023年4月25日保藏于广东省微生物菌种保藏中心,保藏编号为GDMCC No.63403。
2.一种促进毛鸡骨草生长和活性成分积累的菌剂,其特征在于,包含有权利要求1所述的热带根瘤菌FM-19。
3.一种毛鸡骨草的种植方法,其特征在于,毛鸡骨草种子催芽,芽苗长至1-2cm时移栽到栽培介质中,待长出真叶后,在栽培介质中接种权利要求1所述热带根瘤菌FM-19。
4.根据权利要求3所述的毛鸡骨草的种植方法,其特征在于,毛鸡骨草种子表面消毒处理后再进行催芽处理。
5.根据权利要求4所述的毛鸡骨草的种植方法,其特征在于,消毒处理方法为:用0.1%升汞溶液浸泡5-15min进行表面消毒,然后浸泡于40-60℃的热水中,自然冷却至室温。
6.根据权利要求3所述的毛鸡骨草的种植方法,其特征在于,每隔10-15d接种1次,接种3-6次。
7.根据权利要求3所述的毛鸡骨草的种植方法,其特征在于,热带根瘤菌FM-19的接种OD600为0.8-1.0。
8.如权利要求1所述的热带根瘤菌FM-19在提高毛鸡骨草藤茎中总黄酮含量上的应用。
