Abstract
本发明属于肿瘤靶向治疗技术领域,涉及一种PTN‑PTPRZ1通路的阻断抗体及其在胶质瘤靶向治疗的应用。本发明提供一种分泌PTPRZ1单克隆抗体的杂交瘤细胞株,所述杂交瘤细胞株的保藏编号为CCTCC NO:C2023174;本发明还提供上述杂交瘤细胞株分泌的PTPRZ1单克隆抗体,其中优选的单克隆抗体为鼠抗PTPRZ1抗体2F10。本发明还提供了上述PTPRZ1单克隆抗体在制备胶质瘤靶向治疗药以及PTN阻断试剂中的应用。根据上述应用可以得出,本发明公开的鼠抗PTPRZ1抗体(2F10)可以作为PTN阻断抗体来抑制胶质瘤干细胞生长,进而达到治疗作用。

技术领域
本发明属于肿瘤靶向治疗技术领域,涉及一种PTN-PTPRZ1通路的阻断抗体及其在胶质瘤靶向治疗的应用。
背景技术
胶质母细胞瘤(Glioblastoma,GBM)是一种致死性极高的原发恶性脑肿瘤,因具有高度异质性和侵袭性而难以有效治疗。相关研究表明,胶质瘤干细胞侵袭迁移能力和克隆形成能力的大幅增加是GBM难以彻底清除的关键因素。PTN(Pleiotrophin)是多效生长因子,可旁分泌到细胞外,通过与其受体PTPRZ1(Protein Tyrosine Phosphatase Receptor-type Zpolypeptide1)结合,调控细胞增殖、迁移运动、血管生成等多种生物学功能。研究表明,PTN-PTPRZ1信号通路的过度激活促进GBM的快速增殖、侵袭转移等恶性生物学行为,目前尚缺乏靶向PTN-PTPRZ1信号轴的治疗措施。
PTPRZ1是一种跨膜受体蛋白酪氨酸磷酸酶,在GBM中特异性高表达,在其他组织中不表达或仅微量表达。PTPRZ1以高度硫酸软骨素蛋白聚糖或蛋白聚糖修饰的形式存在。PTN可与PTPRZ1的糖胺聚糖结构通过配受体作用而结合,PTN介导的PTPRZ1二聚化导致PTPRZ1磷酸酶活性抑制,继而激活ERK磷酸化,ERK激活下游与细胞增殖、迁移运动等相关通路,促进细胞增殖和迁移。
因此,如果研发出相应靶向PTPRZ1的单克隆抗体,竞争性阻断PTN与PTPRZ1结合,有望抑制PTN介导的PTPRZ1激活,抑制PTPRZ1下游ERK信号通路的激活,最终降低GBM的增殖和侵袭能力,将为GBM提供新的治疗手段。
而目前缺乏明确抗原识别区序列的PTPRZ1靶向单克隆抗体。
发明内容
有鉴于此,本发明的目的在于提供一种PTN-PTPRZ1通路的阻断抗体及其在胶质瘤靶向治疗中的应用,具体的,本发明公开的鼠抗PTPRZ1抗体(2F10)作为PTN阻断抗体可以抑制胶质瘤干细胞生长。
为达到上述目的,本发明提供如下技术方案:
本发明提供一种分泌PTPRZ1单克隆抗体的杂交瘤细胞株,所述杂交瘤细胞株的保藏编号为CCTCC NO:C2023174。
本发明还提供上述杂交瘤细胞株分泌的PTPRZ1单克隆抗体。

首先,本发明通过胶质瘤干细胞成球实验,发现本发明的PTPRZ1单克隆抗体显著抑制PTN对胶质瘤干细胞成球能力的促进作用;其次,本发明通过胶质瘤干细胞Transwell实验,发现本发明的PTPRZ1单克隆抗体显著抑制PTN对胶质瘤干细胞迁移、侵袭能力的促进作用;另外,本发明发现PTPRZ1单克隆抗体通过抑制PTN介导的ERK信号通路活化发挥作用。因此,可将本发明PTPRZ1单克隆抗体用于制备PTN阻断试剂。
本发明的有益效果在于:
1、在胶质瘤干细胞成球实验中,本发明公开的鼠抗PTPRZ1抗体(2F10)显著抑制PTN对胶质瘤干细胞成球能力的促进作用;
2、在胶质瘤干细胞Transwell实验中,本发明公开的鼠抗PTPRZ1抗体(2F10)显著抑制PTN对胶质瘤干细胞迁移、侵袭能力的促进作用;
3、本发明公开的鼠抗PTPRZ1抗体(2F10)抑制PTN介导的ERK信号通路活化;
4、总的来说,本发明公开的鼠抗PTPRZ1抗体(2F10)可以作为PTN阻断抗体来抑制胶质瘤干细胞生长,进而达到治疗作用。

实施例1.PTPRZ1单克隆抗体的制备
一、重组PTPRZ1蛋白的表达纯化
选取人PTPRZ1基因(UniProtKB:P23471)的特定区域(选取的片段为编码人PTPRZ1的氨基酸26-300区段的基因)克隆至真核表达载体pSIN-EF-1a-puro获得真核表达质粒。将表达质粒转染HEK293细胞后进行悬浮培养5天,离心收集细胞上清,通过镍柱纯化和阴离子交换纯化得到目的蛋白(重组PTPRZ1蛋白)。
二、小鼠免疫及抗体检测
将弗氏完全佐剂与浓度为2mg/mL的抗原蛋白(重组PTPRZ1蛋白)按等体积混合并乳化,将乳化完成的抗原免疫6-8周龄SPF级雌性BALB/c小鼠,采用脚底注射的方式,每只小鼠注射100μg抗原蛋白。初次免疫完成两周后,将抗原蛋白与弗氏不完全佐剂混合乳化,再次采用脚底注射的方式,每只小鼠注射100μg抗原蛋白。两周以后通过尾静脉采血,离心收集上清并用ELISA检测血清效价,并选取血清效价>8万的小鼠开展融合前的加强免疫。
三、B细胞融合及阳性杂交瘤细胞的筛选和亚克隆
1.脾脏B淋巴细胞和SP2/0细胞悬液的制备
取用重组PTPRZ1蛋白免疫好的雌性Balb/c小鼠,摘除眼球放血处死,眼血离心后收集血清作ELISA的阳性对照,无菌操作取出小鼠腹股沟***,放入盛有10mL不完全培养基的玻璃皿中,洗涤,小心剥去周围的***和脂肪组织,换一玻璃皿,将脾脏捞出,置于200目不锈钢网中,用注射器的内芯研磨,同时不时用不完全培养基冲洗,使脾细胞穿过网孔进入溶液中,将脾细胞移至10mL玻璃离心管中,1500rpm水平离心10分钟,去上清。同法,用不完全培养基10mL洗涤细胞1次,离心收集沉淀的细胞,将细胞用10mL不完全培养基重悬混匀,细胞计数约为1×108个细胞。
将SP2/0细胞从液氮中取出,迅速放入37℃水浴中,不断摇晃,直至细胞溶液完全溶解,将细胞转移到10mL离心管中,1500rpm水平离心10分钟,弃上清,10mL完全培养液重悬沉淀,把细胞悬液转移到50mL培养瓶中,置37℃、5% CO2培养箱中培养。待细胞生长良好后用含8-AG的1640培养基筛选细胞一周;融合前2天,将1瓶细胞传至4瓶,则融合当天细胞正处于对数生长期,活力正好,细胞大小均匀,圆而透亮,融合当天,用弯头滴管将SP2/0细胞从管壁上轻轻吹下,收集于离心管中,离心,弃上清,沉淀用1640不完全培养基洗涤后,10mL不完全培养基重悬,细胞计数,约为5×107个。
2.饲养细胞的制备
取未免疫的雌性大鼠,摘眼球放血处死,70%乙醇浸泡消毒5分钟,剪开大鼠皮肤,用镊子提起腹膜,用剪刀剪一小口,弯头滴管吸取预冷的不完全培养基冲洗腹腔,将洗液吸至50mL离心管中。同法,用不完全培养基冲洗腹腔3次,收集洗液,室温下1000rpm水平离心10分钟,去上清,10mL不完全培养基重悬细胞并计数。
3.骨髓瘤细胞和脾脏B淋巴细胞融合
融合前将PEG 1450置于37℃培养箱中预温,吸取1×107个骨髓瘤细胞悬液和1×108个脾脏B淋巴细胞悬液至一个50mL无菌离心管中,补加30mL RPMI-1640培养基,充分混匀,1500rpm离心10分钟,弃上清,轻弹管底,使细胞团松散成糊状,将离心管37℃水浴,用滴管吸取0.8mL预温的50% PEG1450溶液,在离管底约2cm处沿管壁慢慢加入细胞中,边加边转动离心管,在1分钟左右加完,然后静置90秒,逐滴加入37℃预温的RPMI-1640培养基30mL终止融合,3分钟之内加完,速度先慢后快,动作轻柔,将离心管在37℃培养箱中静置5分钟,取出离心管,1500rpm离心5分钟,弃去上清,加入10mL HAT培养基重悬细胞,轻轻吹打,混匀,将融合细胞接种至已铺有饲养细胞的96孔细胞培养板,按100μL/孔,每块培养板留6孔接种SP2/0细胞,作为HAT选择的阴性对照,置37℃、5% CO2培养箱中培养。
融合后第4-5天即可在倒置显微镜下观察细胞的生长情况,并补加HAT培养基100μL,第10-12天可用间接ELISA法测杂交瘤效价,第14-15天换HAT培养基培养。
4.特异性杂交瘤细胞的筛选
初步筛选:融合后12-15天,待细胞长到满培养孔的1/4-1/2孔底时,采用游离ELISA法检测培养上清,筛选阳性克隆;以山羊抗小鼠多抗包被酶标板(0.5μg/孔),4℃过夜,洗涤缓冲液洗涤5次,每次5分钟,拍干液体,加入50μL细胞培养上清和50μL的0.2%的PBST,阳性对照选小鼠的免疫血清,阴性对照选SP2/0培养上清,空白对照用洗涤液,37℃孵育1小时;使用生物素标记重组PTPRZ1蛋白得到PTPRZ1-生物素,洗涤酶标板后加入100μLPTPRZ1-生物素稀释液(约0.5μg/mL)37℃孵育1小时。洗涤酶标板以后,再加入1:5000稀释的链霉素亲和素-HRP,37℃孵育30分钟。洗涤,拍干液体,加新鲜配制的TMB溶液100μL/孔,室温暗处反应3-5分钟,加终止液每孔50μL终止反应,酶标仪检450nm吸光度值。
5.复核筛选
采用细胞ELISA方法进行复核筛选。选取前一步骤中显色值OD450>0.5的细胞孔用于复核筛选,其余细胞孔淘汰。于96孔细胞培养板中培养胶质瘤成球细胞,约4×104个细胞/孔,贴壁培养过夜。第二天吸走培养基,用PBS清洗两遍,使用4% PFA固定处理10分钟。吸走固定液,用PBS清洗2遍。取50μL待查的细胞上清加入细胞ELISA孔,于室温条件300rpm孵育1小时。吸走上清,用洗涤缓冲液洗涤2次,拍干液体后,加入1:5000稀释的HRP标记山羊抗鼠多抗,于室温条件300rpm孵育1小时。洗涤,拍干液体,加新鲜配制的TMB溶液100μL/孔,室温暗处反应3-5分钟,加终止液每孔50μL终止反应,酶标仪检450nm吸光度值。选出显色值>0.3的细胞进行后续克隆化培养。
结果显示,以重组PTPRZ1蛋白为抗原,先后免疫雌性Balb/c小鼠6只,共融合2次,先后共筛选获得5株能分泌高亲和力的PTPRZ1抗体的杂交瘤细胞株,经3次克隆化和ELISA筛选后,得到分泌PTPRZ1单克隆抗体的杂交瘤细胞株,将这些杂交瘤细胞经数次冻存,体外传代培养3个月以上均能稳定分泌PTPRZ1单克隆抗体,最终冻存于液氮罐。
四、抗体的纯化制备
将经过筛选及克隆化所得目的杂交瘤细胞扩大培养,并接种至摇瓶进行无血清摇瓶培养发酵。按照1×106个/mL的浓度接种摇瓶,于37℃,65rpm的无菌条件连续培养4天。发酵完成后,收取培养液,于15℃,4000g离心条件离心30分钟,收取离心上清,并用0.45μm滤膜对离心上清进行过滤,收集滤液。采用蛋白G纯化柱进行亲和纯化,得到目的抗体。
五、抗体的亚型鉴定及基因序列克隆
采用Southern Biothech公司的SBA Clonotyping System-HRP试剂盒,按照说明书的操作来鉴定单克隆抗体重链和轻链的亚型。
具体操作为:用包被液(0.05M pH=9.5的碳酸盐和碳酸氢盐缓冲液)将捕获抗体稀释至1μg/mL,按照100μL/孔加入酶标板,于4℃包被过夜。用含有0.05%Tween-20的PBS缓冲液(洗板液)洗板3次。用稀释液(1%BSA,0.1%PBST)按照1:1稀释待检杂交瘤细胞的培养上清,按照100μL/孔加入酶标板,于37℃孵育30分钟。用稀释液将对应的酶标抗体(Ig-HRP、IgG1-HRP、IgG2a-HRP、IgG2b-HRP、IgG3-HRP、IgM-RP、kappa-HRP、lambda-HRP)按照1:3000稀释。用洗板液洗板3次后每孔加入100μL稀释的酶标抗体,于37℃孵育30分钟。再次洗板3次以后加入显色液,约5分钟(视反应强弱而定)之后加入2M硫酸终止反应,读取450nm吸光值。经过鉴定,本发明抗体的重链均为IgG2a,轻链均为Kappa。根据抗体亚型结果,采用成熟的技术路线克隆抗体基因序列,具体的,收集生长状态良好的杂交瘤细胞,利用Thermo公司的Trizol提取杂交瘤细胞总RNA,按照Takara公司的PrimerScript II ReverseTranscriptase说明书的操作方法将mRNA逆转录为cDNA,于-20℃冻存备用。
反转录具体操作流程如下:
首先配制10μL总体积的模板RNA/引物DNA的预混液,其中包括1μL 10μM浓度的逆转录引物引物(Oligo d(T)18Primer),4μL浓度为2.5mM的dNTP混合液,5μL RNA(总量小于5μg)。混匀之后于65℃处理5分钟,立即放于冰上。向预混液中加入4μL 5×PrimeScript II缓冲液,20单位的RNA酶抑制剂,1μL Primer Script II RTase(200单位),混匀之后于42℃反应60分钟,再于70℃处理15分钟,之后放于冰上冷却备用。
然后以cDNA为模板,采用文献(Sequencing and cloning of antigen-specificantibodies from mouse memory B cells,Nature Protocols,2016Oct;11(10):1908-1923.doi:10.1038/nprot.2016.102.)报道的抗体基因扩增引物分别独立进行扩增尝试,筛选出能够高效扩增抗体基因的引物。按南京诺唯赞公司的Phanta Max Super-FidelityDNA Polymerase说明书的操作方案进行PCR。
PCR反应体系为:25μL 2×Phanta、1μL dNTP、4μL 10uM引物对、4μL杂交瘤细胞cDNA、1μL DNA polymerase、15μL dd H2O,总反应体积为50μL。扩增条件为:预变性94℃,3分钟;变性94℃,30秒;退火56℃,30秒;延伸72℃,2分钟。按照OMEGA公司OMEGA GelExtraction Kit说明书对PCR产物进行胶回收,扩增产物链接T载体以后经过质粒测序获得抗体基因序列。
实施例2.PTPRZ1单克隆抗体亲和力SPR实验
一、捕获分子偶联
配制运行缓冲液(0.01M HEPES,0.15M NaCl,3mM EDTA,0.05% Tween-20,pH=7.4),连接运行缓冲液试剂瓶至Biacore T200(GE Healthcare)。将CM5芯片置于芯片仓,运行Primer程序,冲洗***管路。用10mM醋酸钠缓冲液(pH=5.0)将Anti-Mouse IgG稀释20倍,装入EP管,另将氨基偶联相关试剂装入EP管,置于试剂架上,放入Biacore T200样品仓,运行手动程序,依次进行芯片活化、偶联配体、封闭三个步骤,将Anti-Mouse IgG共价偶联至CM5芯片的4个通道。
二、亲和力测定
使用HEPES运行缓冲液将配体及分析物稀释至目的浓度。将配体及分析物装入EP管,置于试剂架上;将再生液10mM甘氨酸(pH=1.7),装入EP管,置于试剂架上。将试剂架放入Biacore T200样品仓,打开亲和力测定程序,芯片流路选择“2-1、3-1、4-1”通道,采用多循环动力学方法(设置参数:捕获通道2/3/4,流速10μL/分钟,捕获45秒;分析物1、2、3、4通道,流速30μL/分钟,进样120秒,解离900秒;再生液流速30μL/分钟,再生时间90秒。芯片仓与样品仓温度选择25℃,测定分析物与配体亲和力。动力学数据使用1:1结合模型分析。
通过SPR实验检测5株PTPRZ1抗体亲和力。所有抗体的KD值均在可信范围内(KD在E-08M级),其中鼠抗PTPRZ1抗体(2F10)KD值最低,为3.31E-08M,说明鼠抗PTPRZ1抗体(2F10)的抗体亲和力最佳。
鼠抗PTPRZ1抗体(2F10)的重链的核苷酸序列如SEQ ID NO:1所示。
鼠抗PTPRZ1抗体(2F10)的轻链的核苷酸序列如SEQ ID NO:2所示。
鼠抗PTPRZ1抗体(2F10)的重链的氨基酸序列如SEQ ID NO:3所示。
鼠抗PTPRZ1抗体(2F10)的轻链的氨基酸序列如SEQ ID NO:4所示。

实施例3.鼠抗PTPRZ1抗体(2F10)显著抑制PTN对胶质瘤干细胞成球能力的促进作用
1、建立胶质瘤干细胞,具体方法如下:取胶质瘤肿瘤组织与木瓜蛋白酶解离***(Worthington Biochemical)分离,分离的肿瘤细胞在干细胞培养基中培养。胶质瘤干细胞培养基为Neurobasal medium基础培养基,再加入1×B27补充剂(Gibco),表皮生长因子(20ng/ml,PeproTech)和碱性成纤维细胞生长因子(20ng/ml,PeproTech),培养6小时后。然后用异硫氰酸荧光素(FITC)缀合的抗CD15抗体(BD Biosciences)和CD133抗体(Miltenyi,130-090-854)在4℃下标记细胞30分钟,然后用流式分离胶质瘤干细胞(CD15+/CD133+)。
2、取对数生长期胶质瘤干细胞,弃掉培养基,加入PBS洗三次,加入0.25%胰蛋白酶消化2分钟,用含10%FBS的DMEM培养基终止消化。
3、离心300g,5分钟,弃上清,PBS洗三次。
4、用1mL Neurobasal medium干细胞培养基重悬,细胞计数。
5、将细胞分为四组,分别为对照组、10ng/mL重组人PTN处理组、1μg/mL鼠抗PTPRZ1抗体处理组和10ng/mL重组人PTN+1μg/mL鼠抗PTPRZ1抗体四组。每组10个复孔,每孔接种40个细胞到96孔板中,每2天进行补液。
6、10天后观察细胞成球情况,并拍照记录。
通过成球实验检测胶质瘤干细胞成球能力情况。结果如图1所示,与对照组相比,加入重组人PTN(购自北京义翘神州公司)可以促进胶质瘤干细胞成球能力,而加入鼠抗PTPRZ1抗体(2F10)则不影响胶质瘤干细胞成球能力,当同时加入重组人PTN和鼠抗PTPRZ1抗体(2F10)后,胶质瘤干细胞成球能力大幅降低,说明鼠抗PTPRZ1抗体(2F10)作为中和抗体可以有效阻断PTPRZ1以抑制PTN对胶质瘤干细胞自我更新和成球能力的促进作用。
实施例4.鼠抗PTPRZ1抗体(2F10)显著抑制PTN对胶质瘤干细胞迁移能力的促进作用
1、制备胶质瘤干细胞悬液
收集处于对数生长期的胶质瘤干细胞,消化细胞,终止消化后离心弃去培养液,用PBS洗三遍,用无血清培养基重悬。调整细胞密度至1×105/mL。取200μL细胞到1.5mL EP管中,再分别加入PBS(对照)、10ng/mL重组人PTN、1μg/mL鼠抗PTPRZ1抗体和10ng/mL重组人PTN+1μg/mL鼠抗PTPRZ1抗体,混匀后取上述细胞悬液加入Transwell小室上室中,24孔板下室加入600μL含5%FBS的DMEM培养基。
2、培养细胞
放入CO2培养箱中,培养24小时。
3、细胞染色
取出Transwell小室,PBS洗三次,4%PFA室温固定15分钟,结晶紫染色20分钟,蒸馏水洗三次。用棉签擦去Transwell上室内的细胞,37℃晾干后拍照。
通过Transwell实验检测胶质瘤干细胞迁移情况。结果如图2所示,与对照组相比,加入重组人PTN促进胶质瘤干细胞迁移能力,而加入鼠抗PTPRZ1抗体(2F10)则不影响胶质瘤干细胞迁移能力,当同时加入重组人PTN和鼠抗PTPRZ1抗体(2F10)后,胶质瘤干细胞迁移能力大幅降低,说明鼠抗PTPRZ1抗体(2F10)中和抗体可以显著抑制PTN对胶质瘤干细胞迁移能力的促进作用。
实施例5.鼠抗PTPRZ1抗体(2F10)显著抑制PTN对胶质瘤干细胞侵袭能力的促进作用
1、Transwell小室制备
将Matrigel与DMEM培养基进行1:1稀释,吸取稀释液均匀涂抹在Transwell小室底部膜的上室面,37℃晾干。
2、制备胶质瘤干细胞悬液
消化胶质瘤干细胞,终止消化后离心弃去培养液,用PBS洗三遍,用无血清培养基重悬。调整细胞密度至1×105/mL。取200μL细胞到1.5mL EP管中,对应加入重组人PTN、鼠抗PTPRZ1抗体(2F10)。取上述细胞悬液200μL加入Transwell小室上室中,24孔板下室加入600μL含5%FBS的DMEM培养基。
3、培养细胞
放入二氧化碳培养箱中,培养24小时。
4、细胞染色
取出Transwell小室,PBS洗三次,4%PFA室温固定15分钟,结晶紫染色20分钟,蒸馏水洗三次。用棉签擦去Transwell上室内的细胞,37℃晾干后拍照。
通过Transwell实验检测胶质瘤干细胞侵袭情况。结果如图3所示,与对照组相比,加入重组人PTN促进胶质瘤干细胞侵袭能力,而加入鼠抗PTPRZ1抗体(2F10)则不影响胶质瘤干细胞侵袭能力,当同时加入重组人PTN和鼠抗PTPRZ1抗体(2F10)后,胶质瘤干细胞侵袭能力大幅降低,说明鼠抗PTPRZ1抗体(2F10)中和抗体可以显著抑制PTN对胶质瘤干细胞侵袭能力的促进作用。
实施例6.鼠抗PTPRZ1抗体(2F10)抑制PTN介导的ERK信号通路活化
1、收集胶质瘤干细胞,用枪头吸除培养皿中的培养基,加PBS清洗,吸除PBS;
2、用细胞刮收集培养皿中全部贴壁细胞,装入离心管中,300g离心5分钟;
3、向细胞沉淀中加入配好的RIPA蛋白裂解液(RIPA:蛋白酶/磷酸酶抑制剂=100:1),充分混匀,置于冰上裂解30分钟,每隔5分钟充分震荡,直至无白色沉淀;
4、4℃离心机12000g,离心30分钟,吸取上清液置于离心管中,冰上保存;
5、按照BCA蛋白浓度测定试剂盒说明书测定蛋白浓度,562nm测定吸光值,计算出蛋白浓度;
6、将蛋白样品用RIPA调至等浓度后,按照3:1体积比加入4×loading buffer,用枪头混匀,在95℃金属浴锅中加热10分钟;
7、取20μg蛋白和5μL marker缓慢加入电泳槽拔掉梳子的电泳道中,恒压跑电泳,初始电压为80V,大约30分钟蛋白进入分离胶后,设置电压为120V,大约再跑60分钟;
8、电泳结束后甲醇激活PVDF膜1分钟,放入转膜液,滤纸需至少提前15分钟用转膜液浸湿,组装电转夹子(由正极至负极顺序为海绵、薄滤纸、PVDF膜、电泳凝胶、薄滤纸、海绵),三明治法压紧,并用滚轮赶走气泡,恒流150mA跑电转大约90分钟;
9、电转结束后,将PVDF膜置于配置好的封闭液中,室温震荡封闭60分钟;
10、用PBST轻柔洗去封闭液,按照说明书浓度用抗体稀释液配制好一抗溶液,4℃过夜震荡孵育;
11、将PVDF膜用PBST室温震荡洗3次,每次5分钟;
12、二抗室温震荡孵育60分钟;
13、将PVDF膜用PBST室温震荡洗3次,每次5分钟;
14、用ECL试剂盒进行显影。
通过免疫印迹实验检测胶质瘤干细胞ERK蛋白活化情况。结果如图4所示,与对照组相比,加入重组人PTN促进胶质瘤干细胞磷酸化ERK表达,而加入鼠抗PTPRZ1抗体(2F10)则不影响胶质瘤干细胞磷酸化ERK表达,当同时加入重组人PTN和鼠抗PTPRZ1抗体(2F10)后,胶质瘤干细胞磷酸化ERK表达显著降低,说明鼠抗PTPRZ1抗体(2F10)中和抗体抑制PTN介导的通过ERK信号通路活化。
最后说明的是,以上实施例仅用以说明本发明的技术方案而非限制,尽管参照较佳实施例对本发明进行了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本技术方案的宗旨和范围,其均应涵盖在本发明的权利要求范围当中。
Claims (10)
1.一种分泌PTPRZ1单克隆抗体的杂交瘤细胞株,其特征在于,所述杂交瘤细胞株的保藏编号为CCTCC NO: C2023174。
2.由权利要求1所述的杂交瘤细胞株分泌的PTPRZ1单克隆抗体。
3.根据权利要求2所述的PTPRZ1单克隆抗体,其特征在于,所述PTPRZ1单克隆抗体的重链的CDR1的序列为 DYYML、CDR2的序列为LVYPYNGGTSYNQKFKG、CDR3的序列为SDSYVDYFDY;
PTPRZ1单克隆抗体的轻链的CDR1的序列为KSSQSLLNSRTRKNYLA、CDR2的序列为WASTRES、CDR3的序列为KQSYNLYT。
4.根据权利要求2所述的PTPRZ1单克隆抗体,其特征在于,所述PTPRZ1单克隆抗体的重链的氨基酸序列如SEQ ID NO:3所示、轻链的氨基酸序列如SEQ ID NO:4所示。
5.编码权利要求2-4任一项所述的PTPRZ1单克隆抗体的基因。
6.根据权利要求5所述的基因,其特征在于,所述PTPRZ1单克隆抗体的重链的基因序列如SEQ ID NO:1所示、轻链的基因序列如SEQ ID NO:2所示。
7.含有权利要求5或6所述的基因的重组载体。
8.含有权利要求7所述的重组载体的宿主细胞。
9.权利要求2-4任一项所述的PTPRZ1单克隆抗体在制备胶质瘤靶向治疗药物中的应用。
10.一种药物,其特征在于,所述药物为胶质瘤靶向治疗药物,所述药物中含有权利要求2-4任一项所述的PTPRZ1单克隆抗体。
