Results of the experiment
1. Animal drug effect test
1.1 antitussive test
80 mice, half of the males and females, were randomly divided into 8 groups, 7 groups for drug administration and a blank group. 7 groups of administration groups used the drugs of examples 1-7 of the invention in sequence, and administered by intragastric administration, 12.5g/kg, 1 time per day for 4 consecutive days; the blank group was gavaged with the same dose of physiological saline. After 1 hour of the last administration, an ammonia-induced cough test was performed, the mice were placed in a constant-pressure ammonia water spray device at a pressure of 140mmHg and an ammonia concentration of 28%, and were immediately taken out after receiving constant-pressure ammonia water spray for 15 seconds, and the cough latency and the number of times of cough of the mice within 2min were observed and recorded for each mouse, and the following data were the average values of each group, and the results are shown in table 1.
TABLE 1 results of the mouse antitussive test
Figure BDA0003432018360000111
As can be seen from Table 1, 1-7 groups had the effect of extending the incubation period and reducing the number of coughs in mice with ammonia-induced coughs, compared with the blank group. The 6 groups and 7 groups are different in dosage form, and have similar cough relieving effect. The 6 groups had the best antitussive effect compared to the 1-5 groups. Therefore, the medicine has obvious cough relieving effect and is helpful for treating respiratory diseases.
1.2 phlegm eliminating test
80 mice, half of the males and females, were randomly divided into 8 groups, 7 groups for drug administration and a blank group. 7 groups of administration groups used the drugs of examples 1-7 of the invention in sequence, and administered by intragastric administration, 12.5g/kg, 1 time per day for 5 consecutive days; the blank group was gavaged with the same dose of physiological saline. After 0.5h of the last administration, 0.1mL (mass fraction 0.5mg/g) of phenol red solution was injected into the vein, each mouse was sacrificed after 0.5h, the trachea was stripped off, a section of trachea from the inferior trachea of the thyroid cartilage was cut off, the trachea was placed in a 2mL test tube containing 0.9% physiological saline, 0.1mL of 0.1mol/L sodium hydroxide solution was added, OD was measured at a wavelength of 546nm to prepare a phenol red standard curve, and the phenol red content (. mu.g/mL) was calculated from the standard curve, and the following data were averaged for each group, and the results are shown in Table 2.
TABLE 2 phlegm eliminating test results
