Abstract
本发明公开了一种降血糖的中药组合物及其制备方法,所述中药组合物,包括以下重量份的组分:白子菜2‑50份、人参1‑30份、马齿苋2‑50份。所述制备方法,所述制备方法为将配方量的白子菜、人参、马齿苋、槐米/槐花、桑叶、栀子、葛根、葡萄籽和玉米须,混合、提取、浓缩、干燥,破碎过筛,即得所述中药组合物。本发明的降血糖的中药组合物通过白子菜、人参和马齿苋三种组分的协同复配,可以恢复胰腺分泌胰岛素的功能或/和消除胰岛素抵抗,从而达到有效降血糖的效果,同时,以药食同源中药为主要原料,安全性高,无毒副作用,可以长期服用,有效规避了多种中药组分共同使用可能带来肝脏损伤的风险,提高了药物的安全性和患者的依从性。

本发明属于中药组合物领域,具体涉及一种降血糖的中药组合物及其制备方法。
背景技术
随着社会经济的快速发展,人们日常生活习惯和饮食结构正发生很大的转变,主要表现在现代生活中的运动量逐渐减少的同时,饮食中高热量、高脂肪、低膳食纤维的不健康食品比例逐渐增多。不良的生活习惯和饮食结构的转变导致了高血糖、高血脂、高血压和肥胖等症状的快速增长,其中以高血糖为主要症状的糖尿病发病率及其带来的经济负担的增长最为显著。
糖尿病,是由于胰岛β细胞不能产生足够的胰岛素,或者身体细胞对产生的胰岛素抵抗,进而由一系列的代谢紊乱引发的以持续高血糖水平为特征的代谢性疾病。胰岛素是胰腺内的胰岛β细胞在内源性或外源性物质刺激下分泌的一种蛋白质激素,它是人体内唯一具有降血糖功能的激素,同时还能促进糖原、脂肪、蛋白质合成。缺乏胰岛素或对胰岛素抵抗均会导致血糖水平较高或高血糖症,进而形成糖尿病。糖尿病如果没有得到及时治疗,会引起多种并发症。急性并发症包括糖尿病酮症酸中毒、高渗高血糖状态或死亡。因此,预防和治疗高血糖极其重要。
目前全世界对于糖尿病和高血糖尚没有根治的方法,对于如此复杂的疾病,常用的西药治疗糖尿病和高血糖的药物主要包括口服降糖类药物和注射胰岛素,口服降糖类药物包括双胍类,磺脲类,膳食葡萄糖调节剂等,上述药物对糖尿病和高血糖的仅有一定的控制效果,并无治疗作用,长期服用大都有肝肾毒副作用,不适合长期服用。
中医理论认为,消渴病(糖尿病)的成病机制主要是素体阴虚、五脏柔弱、饮食不节、过食肥甘、情志失调、劳欲过度,从而导致肾阴亏虚、肺胃燥热、肝脾湿热。成病机制重点为阴虚燥热,而以阴虚为本,燥热为标,病延日久,阴损及阳,阴阳俱虚;阴虚燥热,耗津灼液,使血液黏滞,血行涩滞而成瘀,阴损及阳,阳虚寒凝,导致淤血内阻。中医对糖尿病的分型主要为阴虚燥热型、气阴两虚型、阴阳两虚型。按中医的辨证施治原则,治疗糖尿病必须针对不同的证型分别采用养阴清热法、益气养阴法、温阳育阴法,同时全程兼顾活血化瘀通络,治疗中常选用具有清热、补益、理气及活血化瘀通络功效的降血糖中药进行治疗。
CN105381304A公开了一种用于治疗糖尿病和高血糖的中药组合物,由如下重量的原料药组成:三七3-7份、决明子1-5份、紫杉叶4-8份、西瓜皮1-5份、玉米须1-5份、侧柏叶4-8份、白玉竹1-5份、杜仲1-5份、来凤藤1-5份、黄精1-5份、山茱萸1-5份、枸杞1-5份、细毛山药1-5份、辣木根3-7份,该发明利用中药各个组分之间的配伍,使得能够有效激活惰性胰岛素,降低血糖,使得惰性胰岛素恢复活性,正常参与糖代谢,提高了人体自身对血糖的利用和转化能力,达到平稳降低血糖;可保护胰岛细胞,抑制胰岛素衰竭,在可参与糖代谢的胰岛素增多的情况下,胰岛不必像以前那样分泌过多的胰岛素,胰脏负担减轻,获得必要的休整,进而起到保护胰岛,抑制胰岛衰竭的作用。
CN106266216A公开了一种用于缓解糖尿病引起的血糖升高以及预防和治疗糖尿病的中药组合物,所述的中药组合物由红菇茑、苦瓜以及陈皮组成,该中药组合物能有效缓解糖尿病所引起的血糖升高,预防和治疗糖尿病。
CN107661475A公开了一种稳定血糖和治疗糖尿病的中药药物组合物,主要是由玄参、人参、灵芝、冬虫夏草、川芎、当归、熟地、归尾、黄芪、红花、桃仁、丹参、赤芍、内金、山楂、三棱、莪术、三七、杜仲、牛膝、地龙、威灵仙等中草药或药学上可用辅料制备而成的制剂。该发明注重滋阴降火、软坚散结、软化血管、滋阴生津凉血、清热解毒,气血双补又升阳,活血化瘀,扶正又祛邪,且祛邪不伤正,不但很快稳定血糖,还能调节代谢紊乱,补肾治疗糖尿病,摆脱了对胰岛素的依赖。
目前已知的具有降血糖功能的植物和/或中药单体有很多种,如何从中筛选具有降糖协同功效的中药组合物,对降低血糖和治疗糖尿病的临床应用具有重要意义。
发明内容
为了解决上述技术问题,本发明提供了一种降血糖的中药组合物及制备方法,该中药组合物以药食同源中药为主要原料,安全性高,无毒副作用,可以长期服用,同时,通过不同组分之间的协同复配,可以恢复胰腺分泌胰岛素的功能或/和消除胰岛素抵抗,从而达到降血糖的效果。
为了实现上述目的,本发明采用以下的技术方案:
一种降血糖的中药组合物,包括以下重量份的组分:白子菜2-50份、人参1-30份、马齿苋2-50份。
优选地,所述中药组合物,包括以下重量份的组分:白子菜5-48份、人参1.5-28份、马齿苋5-45份。
白子菜,具有较髙的药用和食用价值。作为蔬菜食用,白子菜具有普通蔬菜所富含的氨基酸、蛋白质、脂肪、糖类、维生素类、纤维素类、碳水化合物和矿物质及微量元素,是一种营养丰富的天然绿色食物。同时,白子菜嫩茎叶还富含黄酮类、多糖、生物碱等活性成分,其中白子菜嫩茎叶中所含总黄酮为86.78mg/g、VC 2.52mg/g、类胡萝卜素0.09mg/g、可溶性蛋白1.42mg/g、槲皮素43.30mg/g、绿原酸6.04mg/g。因此,白子菜具有降“三高”、治疗坏血病及贫血、解毒、防癌、抗氧化、抗诱变、抗炎、抗菌、抗肿瘤、保肝利胆及调节免疫功能等生物活性。
人参,为五加科植物人参属的根茎,为多年生草本植物,是驰名中外的名贵药材和新资源食品。人参具有多种祛病养生功效,在中草药中被称为“百草之王”。近年来,越来越多的学者将目光转向人参降低血糖的作用。人参虽然不能代替胰岛素,但是能够缓解症状,起到相当有利的辅助作用。人参尚有抗消渴病气阴虚疲劳和缺氧的作用。现代医药学研究发现,人参成分复杂,有降血糖功效的成分包括人参提取物、人参皂苷、人参多糖、人参多肽等,这些成分对正常及多种糖尿病动物模型的血糖浓度均有调整作用,能使紊乱的代谢过程正常化,从而达到改善糖尿病病情的效果。
马齿苋,是一种药食同源的植物,是马齿苋科一年生草本植物马齿苋的干燥地上全草。其功能据《本草纲目》记载“散血消肿,利肠滑胎,解毒通淋,治产后虚汗”;据《本草拾遗》记载“破痃癖,止消渴,又主马恶疮虫”。马齿苋味酸、性寒,具有清热解毒、益气生津、凉血止血、散血消肿等作用,主治热痢脓血、热淋、带下、痈肿恶疮、丹毒等。现代研究表明,马齿苋具有显著的抑菌、抗病毒、抗肿瘤、降血糖、降血脂、抗动脉粥样硬化、抑制或兴奋子宫平滑肌及增强免疫力等作用。

优选地,所述的提取为水提、醇提。
本发明还提供了上述中药组合物在制备降血糖药物和/或健康食品中的应用。
本发明还提供了一种降血糖的药剂,包括上述中药物组合物和医学上可接受的辅料。
优选地,所述药剂包括但不限于颗粒剂、片剂、丸剂和胶囊剂。
与现有技术相比:
(1)本发明的降血糖的中药组合物通过白子菜、人参和马齿苋三种组分的协同复配,可以增强恢复胰腺分泌胰岛素的功能或/和消除胰岛素抵抗,从而达到有效降血糖的效果。
(2)本发明的降血糖的中药组合物,以药食同源中药为主要原料,安全性高,无毒副作用,可以长期服用,有效规避了多种中药组分共同使用可能带来肝脏损伤的风险,提高了药物的安全性和患者的依从性。
(3)本发明的制备方法工艺简单,成本低,适合规模化生产。

无特殊说明,本发明所采用的原料均为目前普通的市售产品。其中,胰岛素试剂盒购自北京伊普瑞斯科技有限公司;血糖试剂盒购自深圳迈瑞生物医疗电子股份有限公司;所述槐米/槐花为槐的花及花蕾。
基础实施例
一种降血糖的中药组合物,包括以下重量份的组分:白子菜2-50份、人参1-30份、马齿苋2-50份。
优选地,所述中药组合物,包括以下重量份的组分:白子菜5-48份、人参1.5-28份、马齿苋5-45份。
在一些优选的实施方案中,所述的中药组合物,按重量份计还包括:槐米/槐花1-30份、桑叶1-30份和栀子1-20份。
优选地,所述的桑叶和栀子的重量为1:1。
在一些优选的实施方案中,所述的中药组合物,按重量份计还包括:葛根1-20份、葡萄籽1-20份、枸杞子1-20份和玉米须1-30份。
优选地,所述的葛根、葡萄籽和玉米须的重量为1:1:2。

实施例1-14中所述的降血糖的中药组合物的制备方法为,将配方量的白子菜、人参、马齿苋、槐米、桑叶、栀子、葛根、葡萄籽和玉米须,干燥,混合,提取、浓缩、干燥、破碎过筛,即得所述中药组合物。
对比例1
本对比例与实施例6的区别在于,所述人参的用量为5重量份。
对比例2
本对比例与实施例6的区别在于,用百合代替马齿苋。
对比例3
本对比例与实施例6的区别在于,用玉竹代替白子菜。
对比例4
本对比例与实施例10的区别在于,不含桑叶。
对比例5
本对比例与实施例10的区别在于,决明子代替栀子。

本对比例与实施例14的区别在于,不含葛根。

一、急性毒性实验
1、实验动物
ICR小鼠(购自上海斯莱克实验动物有限公司),体重18-22g,雄性。
2、分组与给药方法
将ICR小鼠按体重随机分为23组,每组8只,将实施例1-14和对比例1-8制备的中药组合物,配成浓度分别为0.2g/mL的悬浮液;给药体积为0.4mL/10g,确定小鼠的给药剂量为8.0g/kg;对照组给药等体积的去离子水,全部采用灌胃法给药,每日一次。
3、实验结果
小鼠经灌胃给药后,较安静,活动正常。其它情况如饮食、活动、毛色、粪便等均无异常发现。14天后每只动物体重均有增加(见表4),未见死亡。

由上表可知,小鼠灌胃给予本发明的中药组合物的最大耐受量MTD>8.0g/kg,并未见到小鼠有明显异常。因此,本发明的中药组合物无毒副作用,安全性高,可以长期服用,有效规避了多种中药组分共同使用可能带来肝脏损伤的风险,提高了药物的安全性和患者的依从性。
二、降血糖试验
1、建立糖尿病动物模型
ICR小鼠腹腔注射200mg/kg·bw4%四氧嘧啶溶液,注射前禁食12h但不禁水,72h后眼眶静脉丛采血测定空腹血糖,血糖值大于11.1mmol/L者为实验性糖尿病动物模型。
2、分组与给药
将建模成功的糖尿病动物模型,随机分成23组,每组8只,其中,22组为实验组,每天分别灌胃实施例1-14和对比例1-8制备的降血糖的中药组合物,灌胃剂量为200mg/kg,1组模型对照组,每天灌胃等体积的生理盐水。同时,设1组正常对照组(8只正常小鼠),每天灌胃等体积的生理盐水;连续灌胃28d。
各组小鼠的空腹血糖值(FBG)每周测定一次;末次给药前禁食12h,给药2h后眼眶静脉采血,离心分离血清,按试剂盒方法测定血清胰岛素水平。

注:##P<0.01,与正常对照组相比呈极显著性差异;*P<0.5,与模型对照组相比呈显著性差异;**P<0.01,与模型对照组相比呈极显著性差异。
由表5可知,本发明的正常对照组和模型对照组小鼠的空腹血糖呈极显著性差异(P<0.01),表明,本发明的糖尿病动物模型造模成功。
灌胃本发明实施例1-6组制备的中药组合物的小鼠,其21d及28d监测的空腹血糖与模型对照组相比,呈显著性差异(P<0.5),表明,本发明制备中药组合物具有降血糖的效果。
同时发现,灌胃实施例6制备的中药组合物的小鼠,其14d、21d监测的空腹血糖与模型对照组相比,呈显著性差异(P<0.5);28d监测的空腹血糖与模型对照组相比,呈极显著性差异(P<0.01)。表明,当白子菜、人参和马齿苋的重量比为10:1:10时,制备的中药组合物的降血糖效果较佳。
根据上表5中的实施例7-10可知,灌胃实施例7-9制备的中药组合物的小鼠,其14d、21d监测的空腹血糖与模型对照组相比,呈显著性差异(P<0.5);28d监测的空腹血糖与模型对照组相比,呈极显著性差异(P<0.01);灌胃实施例10制备的中药组合物的小鼠,其21d、28d监测的空腹血糖与模型对照组相比,呈极显著性差异(P<0.01);表明,当桑叶和栀子的重量为1:1时,制备的中药组合物的降血糖效果较佳。
根据上表5中的实施例11-14可知,灌胃实施例11-13制备的中药组合物的小鼠,其7d、14d监测的空腹血糖与模型对照组相比,呈显著性差异(P<0.5);21d、28d监测的空腹血糖与模型对照组相比,呈极显著性差异(P<0.01);灌胃实施例14制备的中药组合物的小鼠,其7d监测的空腹血糖与模型对照组相比,呈显著性差异(P<0.5);14d、21d、28d监测的空腹血糖与模型对照组相比,呈极显著性差异(P<0.01);表明,当葛根、葡萄籽和玉米须的重量为1:1:2时,制备的中药组合物的降血糖效果较佳。

由表6可知,本发明模型对照组小鼠的血清胰岛素水平显著低于正常对照组,呈极显著性差异(P<0.01),而灌胃本发明实施例1-14制备的中药组合物的小鼠的胰岛素水平,较模型对照组显著升高(P<0.5),表明本发明制备的中药组合物可能具有修复胰岛β细胞、提高胰岛β细胞分泌胰岛素的能力,从而提高糖尿病小鼠血清胰岛素水平,降低糖尿病小鼠血糖。
综上所述,本发明的降血糖的中药组合物通过白子菜、人参和马齿苋三种组分的协同复配,可以恢复胰腺分泌胰岛素的功能或/和消除胰岛素抵抗,从而达到有效降血糖的效果。
本发明的降血糖的中药组合物,以药食同源中药为主要原料,安全性高,无毒副作用,可以长期服用,有效规避了多种中药组分共同使用可能带来肝脏损伤的风险,提高了药物的安全性和患者的依从性。
另外,本发明的制备方法工艺简单,成本低,适合规模化生产。
需要强调的是,本发明所述的实施例是说明性的,而不是限定性的,因此本发明包括并不限于具体实施方式中所述的实施例,凡是由本领域技术人员根据本发明的技术方案得出的其他实施方式,同样属于本发明保护的范围。
Claims (10)
1.一种降血糖的中药组合物,其特征在于,包括以下重量份的组分:白子菜2-50份、人参1-30份、马齿苋2-50份。
2.根据权利要求1所述的中药组合物,其特征在于,包括以下重量份的组分:白子菜5-48份、人参1.5-28份、马齿苋5-45份。
3.根据权利要求1所述的中药组合物,其特征在于,按重量份计还包括:槐米/槐花1-30份、桑叶1-30份和栀子1-20份。
4.根据权利要求3所述的中药组合物,其特征在于,所述的桑叶和栀子的重量为1:1。
5.根据权利要求3所述的中药组合物,其特征在于,按重量份计还包括:葛根1-20份、葡萄籽1-20份、枸杞子1-20份和玉米须1-30份。
6.根据权利要求5所述的中药组合物,其特征在于,所述的葛根、葡萄籽和玉米须的重量为1:1:2。
7.根据权利要求5所述的中药组合物,其特征在于,包括以下重量份的组分:白子菜2-50份、人参1-30份、马齿苋2-50份、槐米/槐花1-30份、桑叶1-30份、栀子1-20份、葛根1-20份、葡萄籽1-20份、枸杞子1-20份和玉米须1-30份。
8.根据权利要求7所述的中药组合物,其特征在于,按重量份计包括:白子菜20份、人参2份、马齿苋20份、槐米/槐米4份、桑叶2份、栀子2份、葛根2份、葡萄籽2份、枸杞子2份和玉米须4份。
9.根据权利要求1-8任一项所述的中药组合物的制备方法,其特征在于,所述制备方法为将配方量的白子菜、人参、马齿苋、槐米/槐花、桑叶、栀子、葛根、葡萄籽和玉米须,混合、提取、浓缩、干燥、破碎过筛,即得所述中药组合物。
10.根据权利要求1-8任一项所述的中药组合物在制备降血糖药物和/或健康食品中的应用。
