Abstract
本发明公开了一种具有降血糖功能的产品及其制备方法,配方按照质量份数组成有:苦瓜提取物40‑80份,黄芩提取物90‑130份,桑叶提取物20‑40份,玉米须发酵物90‑130份;按照制备方法制备出的产品能够增强糖尿病病患的葡萄糖负荷糖耐量,对正常动物的空腹血糖无影响,安全无毒副作用,可长期服用。

技术领域
本发明属于药品及保健食品领域,特别是一种具有降血糖功能的产品及其制备方法。
背景技术
随着人民生活水平的日益提高,糖尿病患病率正急剧增加,已成为影响人类身体健康的主要疾病之一。糖尿病(DM)是世界范围内的流行性疾病。目前,全球DM总患病人数达2亿,预测到2025年这有一数字将超过3亿。我国DM平均患病率已达3.2%~3.6%,患病人数大于4000万,居世界第2位。
糖尿病是一种与遗传因素有关,又与多种环境因素相关的慢性全身性疾病,是由于体内胰岛素的绝对或相对分泌不足而引起的糖、脂肪、蛋白质的代谢紊乱,可分为胰岛素依赖型糖尿病(1型,即IDDM)和非胰岛素依赖型糖尿病(2型,即NIDDM),其中2型患者占糖尿病病例的80%以上。糖尿病若没有及时诊断、正规治疗可引起多种慢性并发症,导致功能障碍和衰竭,成为致残、致死的主要原因。
由于糖尿病的病因和发病机制尚未完全明了,因此目前还缺乏有效的病因治疗方法,现在临床上对糖尿病的治疗多采用胰岛素治疗和口服降糖药物治疗,然而糖尿病属于一种终身性疾病,需要长期用药,胰岛素的注射给药方法往往会给患者带来许多不便,而临床常用的口服降糖药物多属于化学药品或生物药品,长期服用也会引发一系列的不良反应。因此寻找开发具有调节血糖水平的产品,配合药物治疗,在有效地控制血糖和糖尿病的同时,降低副作用成为目前急待解决的难题。
发明内容
为解决现有技术的不足,本发明的目的在于提供一种具有降血糖功能的产品及其制备方法,本产品能够明显降低血糖,安全无毒副作用,可长期服用。
为了实现上述目标,本发明采用如下的技术方案:
一种具有降血糖功能的产品,按照质量份数组成有:苦瓜提取物40-80份,黄芩提取物90-130份,桑叶提取物20-40份,玉米须发酵物90-130份;
苦瓜提取物含苦瓜苷≥10.0%;
黄芩提取物含黄芩苷C21H18O11≥85.0%;
桑叶提取物含总黄酮以芦丁C27H30O16计≥5.0%。
前述的一种具有降血糖功能的产品,按照质量份数组成有:苦瓜提取物45-70份,黄芩提取物90-120份,桑叶提取物20-35份,玉米须发酵物90-120份。
前述的一种具有降血糖功能的产品,玉米须发酵物使用的发酵菌为婴儿双歧杆菌、保加利亚乳杆菌、嗜热链球菌以1:1:1等量混合而成的混合菌。
一种具有降血糖功能的产品的制备方法,包括如下步骤:
制备玉米须发酵物,烘干制粉;
按照配方称量准备材料,分别过筛,备用,
配方包括:苦瓜提取物40-80份,黄芩提取物90-130份,桑叶提取物20-40份,玉米须发酵物90-130份,吸附剂5-20份,防黏剂1-10份;
苦瓜提取物、黄芪提取物、桑叶提取物、玉米须发酵物、吸附剂混合搅拌10-20min,得混合粉A;
在混合粉A内加入乙醇,粘合均匀;
用筛制粒,制粒完成后,在条件为温度55-65℃,水分≤6%的环境下进行干燥,干燥时间1-3h;
干燥颗粒过筛进行整粒;
将干颗粒与防黏剂混合均匀,得总混粉;
将总混粉填充胶囊,即得本产品胶囊剂。
前述的一种具有降血糖功能的产品的制备方法,玉米须发酵物的制备方法包括如下步骤:
步骤一,在60-100℃的温度下,用蒸馏水浸渍玉米须粉,过滤,留玉米须汁,
步骤二,选取活化后的发酵菌,放入含有玉米须汁的扩大培养基内,扩大培养2-3次;
步骤三,将玉米须汁放入厌氧箱内,加入葡萄糖、硫酸镁、醋酸钠,以4-5%的接种量对玉米须汁进行发酵,发酵温度设置为40-41℃,发酵时间为25-35h;
步骤四,过滤发酵液,在条件为温度40-50℃的环境下进行干燥,干燥时间1-3h,得到玉米须发酵物粉末。
前述的一种具有降血糖功能的产品的制备方法,
步骤二,选取活化后的婴儿双歧杆菌、保加利亚乳杆菌、嗜热链球菌,分别放入含有玉米须汁的扩大培养基内,扩大培养2-3次,将各个单菌种按照1婴儿双歧杆菌:1保加利亚乳杆菌:1嗜热链球菌等量混合成混合菌。
前述的一种具有降血糖功能的产品的制备方法,按100g混合粉A加入15-17ml乙醇的比例,在混合粉A内加入乙醇,粘合均匀。
前述的一种具有降血糖功能的产品的制备方法,吸附剂为微晶纤维素。
前述的一种具有降血糖功能的产品的制备方法,防黏剂包括:硬脂酸镁,微粉硅胶。
本发明的有益之处在于:
本产品能够明显降低血糖,安全无毒副作用,可长期服用;
本产品的玉米须经过发酵,营养成分和有效成分得到较大的增加,提高降糖效果;
玉米须发酵菌采用1婴儿双歧杆菌:1保加利亚乳杆菌:1嗜热链球菌等量混合成混合菌,得到的发酵液味道没有馊味,乳酸香味强,且营养成分含量高。
具体实施方式
以下结合具体实施例对本发明作具体的介绍。
一种具有降血糖功能的产品,按照质量份数组成有:苦瓜提取物40-80份,黄芩提取物90-130份,桑叶提取物20-40份,玉米须发酵物90-130份;玉米须发酵物使用的发酵菌为婴儿双歧杆菌、保加利亚乳杆菌、嗜热链球菌以1:1:1等量混合而成的混合菌;
黄光苦瓜提取物提取自葫芦科植物苦瓜的果实,含苦瓜苷≥10.0%;
黄芩提取物的制法:取黄芩,加水煎煮,合并煎液,浓缩至适量,用盐酸调节PH值至1-2,80摄氏度保温,静置,滤过,沉淀物加适量水搅匀,用40%氢氧化钠溶液调节PH值至7,加等量乙醇,搅拌使溶解,滤过,滤液用盐酸调节PH值至1-2,60摄氏度保温,静置,滤过,沉淀依次用适量水及不同浓度的乙醇洗至PH值至7,挥尽乙醇,减至干燥,即得。黄芩提取物含黄芩苷C21H18O11≥85.0%。
桑叶提取物提取自桑科植物桑的干燥叶,桑叶提取物含总黄酮以芦丁C27H30O16计≥5.0%。
一种具有降血糖功能的产品的制备方法,包括如下步骤:
制备玉米须发酵物,烘干制粉;
玉米须发酵物的制备方法包括如下步骤:
步骤一,在60-100℃的温度下,用蒸馏水浸渍玉米须粉,过滤,留玉米须汁,
步骤二,选取活化后的婴儿双歧杆菌、保加利亚乳杆菌、嗜热链球菌,分别放入含有玉米须汁的扩大培养基内,扩大培养2-3次,将各个单菌种按照1婴儿双歧杆菌:1保加利亚乳杆菌:1嗜热链球菌等量混合成混合菌。需要说明的是,也可以是选用单菌种或是其他配合方式,婴儿双歧杆菌、保加利亚乳杆菌、嗜热链球菌等量混合只是一种优选实施例。
步骤三,将玉米须汁放入厌氧箱内,加入葡萄糖、硫酸镁、醋酸钠,以4-5%的接种量对玉米须汁进行发酵,发酵温度设置为40-41℃,发酵时间为25-35h。
步骤四,过滤发酵液,在条件为温度40-50℃的环境下进行干燥,干燥时间1-3h,得到玉米须发酵物粉末。

苦瓜提取物、黄芪提取物、桑叶提取物、玉米须发酵物、吸附剂混合搅拌10-20min,得混合粉A;
按100g混合粉A加入15-17ml乙醇的比例,在混合粉A内加入乙醇,粘合均匀。
用18目筛制粒,制粒完成后,在条件为温度55-65℃,水分≤6%的环境下进行干燥,干燥时间1-3h。
干燥颗粒过20目筛进行整粒;
将干颗粒与防黏剂混合均匀,得总混粉,作为一种实施例,防黏剂包括:硬脂酸镁,微粉硅胶;
将总混粉填充胶囊,每粒重0.35g,即得本产品胶囊剂。
以上是胶囊剂的制备方法,应用本方法还可以制成片剂、颗粒剂、散剂等,产品的类型不受限制,只要运用了本发明的配方,都在本发明的保护范围之内。

对降血糖产生效果的主要成分是:皂苷,多糖,黄酮,由上表可知,有效成分的含量的提取得到很大程度的提升。
实验验证:
通过实验验证本发明的配方能够有效降低血糖。
按照如下实施例的配方制备出产品,按照剂量为0.47g/kg给予受试小鼠,每组由12只小鼠,雌雄各半,再设置一组空白组,对比血糖量。
实施例1,配方为:苦瓜提取物40g,黄芩提取物90g,桑叶提取物20g,玉米须发酵物90g;
实施例2,配方为:苦瓜提取物45g,黄芩提取物90g,桑叶提取物20g,玉米须发酵物90g;
实施例3,配方为:苦瓜提取物50g,黄芩提取物100g,桑叶提取物25g,玉米须发酵物100g;
实施例4,配方为:苦瓜提取物60g,黄芩提取物110g,桑叶提取物30g,玉米须发酵物110g;
实施例5,配方为:苦瓜提取物70g,黄芩提取物120g,桑叶提取物35g,玉米须发酵物120g;
实施例6,配方为:苦瓜提取物80g,黄芩提取物130g,桑叶提取物40g,玉米须发酵物130g;
统一使用如下的制备方法:
(1)苦瓜提取物、黄芪提取物、桑叶提取物、玉米须发酵物、微晶纤维素、微粉硅胶分别过80目筛,称量,备用。硬脂酸镁称量备用。
(2)苦瓜提取物、黄芪提取物、桑叶提取物、玉米须发酵物、微晶纤维素混合15min,得混合粉A。
(3)按100g物料加入15~17ml乙醇粘合均匀,以手轻握成团,捏之即散为度。
(4)用18目筛制粒,制粒后进行干燥(温度60℃左右,干燥时间1~3h,水分≤6%)。
(5)干燥颗粒过20目筛进行整粒。
(6)上述干颗粒与微粉硅胶、硬脂酸镁混合均匀,得总混粉,混合时间约10min。
(7)将总混粉填充胶囊,每粒重0.35g,即得本产品胶囊剂。
制备出样品1,样品2,样品3,样品4,样品5,样品6。
样品对高血糖模型小鼠葡萄糖负荷糖耐量的影响,如表1所示;

由表1可知,给葡萄糖2h时血糖值与空白组有显著性差异,说明本发明的配方能够增强糖尿病小鼠的葡萄糖负荷糖耐量。
样品对正常小鼠空腹血糖的影响,给予受试小鼠每天服用产品30天,检测实验前血糖值,实验后血糖值如表2所示;

由表2可知,30天后,实验组血糖值与空白组没有显著性差异,说明本发明的配方对正常动物空腹血糖无影响。
总结:本发明的样品具有辅助降血糖的功能。
本发明提供一种具有降血糖功能的产品及其制备方法,本产品能够明显降低血糖,安全无毒副作用,可长期服用。

所述黄芩提取物含黄芩苷C21H18O11≥85.0%;
所述桑叶提取物含总黄酮以芦丁C27H30O16计≥5.0%。
