The essence containing the extracts prepared in specific examples 1 to 5 and the essence containing the extracts prepared in comparative examples 1 to 3 were respectively subjected to whitening, moisture absorption and retention efficacy, SOD activity and melanin value tests.
(I) detection method
1. Whitening effect
The whitening effect of the essence is evaluated by detecting the inhibition effect of the essence on the tyrosinase activity, and the lower the residual activity of the tyrosinase is, the better the inhibition effect of the essence on the tyrosinase activity is, and the better the whitening effect is.
The test is to measure the inhibition of tyrosinase and the activity of tyrosinase by the sample by ultraviolet spectrophotometry (analysis is performed by the selective absorption of light by substances). The experiment is carried out by using tyrosinase and dopa to form oxidation product dopachrome under the catalysis of enzyme in an ultraviolet region, wherein the dopachrome has maximum absorption at 475nm, and the absorption coefficient epsilon is 3700(cm & mol/L).
Using a hand-held pipette, 1.0mL of 2 mmoL/L-dopa and 2.6mL of a phosphate buffer (pH 6.8) were pipetted into cuvettes. The temperature was maintained in a constant-temperature water bath at 30 ℃ for 10min, and then 0.2mL of the essences (samples) of examples 1 to 5 and comparative examples 1 to 4 and 0.2mL of 100ug/mL of tyrosinase solution were added to constitute a 4mL solution system. The absorbance of the solution system was measured at a wavelength of 475nm as a function of time over 8 min. The tyrosinase residual activity (U) is calculated as follows:

in the formula, OD1 refers to the absorbance of a live test system containing a substrate, tyrosinase and a sample solution; OD3 represents the absorbance of the assay system containing only the substrate and the sample solution; OD2 is the absorbance of the activity detection system containing only substrate and tyrosinase solution; OD4 refers to the absorbance of the assay containing only the substrate solution.
2. Moisture absorption and retention effects
The moisture absorption and retention efficiency of the essence is evaluated through the moisture absorption rate and the moisture retention rate of the essence, and the higher the moisture absorption rate and the moisture retention rate is, the better the moisture absorption and retention effect is.

Placing 2 dryers in a constant temperature environment with the temperature of 20 ℃, standing for 24 hours, weighing the mass of each sample, and calculating the moisture absorption rate:

m1 is the initial mass of the sample and m2 is the mass of the sample after 24 h.
0.5g of the essence solutions of examples 1 to 5 and comparative examples 1 to 4 dried to a constant weight were precisely weighed, deionized water having a sample weight of 10% was added, and the solution was placed in small weighing bottles and placed in the 2 dryers described above, respectively. Placing 2 dryers in a constant temperature environment with the temperature of 20 ℃, standing for 24 hours, weighing the mass of each sample, and calculating the moisture retention rate:
the moisture retention rate is m4/m3 x 100%
m3 is the initial moisture mass of the sample and m4 is the moisture mass of the sample after 24 h.
3. Determination of anti-aging Capacity
The essences prepared in examples 1 to 5 and the essences prepared in comparative examples 1 to 3 were subjected to anti-aging capacity measurement using SOD activity as an index.
50 Kunming clean-grade mice, half male and female, weight (25 +/-2) g, provided by the Experimental animal center of Guangdong university of medical science, were randomly divided into 5 groups (control group, sample group), and 10 mice each. The experimental interventions of the essences prepared in examples 1-5 of the present invention and in comparative examples 1-4 were performed daily by gavage of the sample groups starting from 8:30 in the first morning, and the mice were weighed 1 time every 1 day, and the gavage (10g/kg) was adjusted accordingly according to the body weight. The control group was also perfused with an equal amount of saline. After one month, the mouse cerebral cortex cells were taken to test the SOD activity.
4. Melanin value test
Randomly selecting 100 women in 20-45 years of age for voluntary test, arranging 10 experimenters in each example or comparative example (one example or the comparative example is tested in 10 groups), and carrying out skin melanin test on the same position of the cheek of each experimenter by using a German DJM skin tester to respectively test an initial value;
then, the extracts of examples 1 to 5 and comparative examples 1 to 3 were used for coating, the coating was carried out for 10 minutes, then the skin melanin value was measured at the same site, and the skin melanin value was measured after coating once a day for 1 day, 3 days, 7 days, 14 days, 21 days, and 28 days, respectively, and the average values were obtained from the data table.
(II) the result of the detection
The results of the test on the whitening and moisturizing efficacy and the SOD activity of the essences of examples 1 to 5 and comparative examples 1 to 3 are shown in table 1:

From the above results, it can be seen that the essences of examples 1 to 5 of the present invention all have good whitening and moisturizing effects, can effectively inhibit tyrosinase activity, and have good hygroscopicity and moisturizing properties. Among them, the essence in example 3 is the most excellent in whitening and moisturizing effects.
