本发明属于茶制品技术领域,具体涉及一种提取茶叶活性物质的工艺方法。
背景技术
19世纪初,茶叶的成分才逐渐明确起来。经过现代科学的分离和鉴定,茶叶中含有机化学成分达四百五十多种,无机矿物元素达四十多种。有机化学成分主要有:茶多酚类、植物碱、蛋白质、氨基酸、维生素、果胶素、有机酸、脂多糖、糖类、酶类、色素等。无机矿物元素主要有:钾、钙、镁、钴、铁、铝、钠、锌、铜、氮、磷、氟、碘、硒等。
随着茶叶研究的不断深入以及和医学研究的交叉和渗透,已经证实茶氨酸具有抗肿瘤、降压安神、促进神经生长等医疗功效;茶多酚具有多种生理功能,它不仅是良好天然抗氧化剂,还具有增强机体抵抗力、防癌、抗肿瘤、抗辐射、抑菌、抗病毒、降血糖、降血脂、预防心血管疾病、抗衰老、防止皮肤紫外损伤等多种生理功能,在油脂、食品、医药、日用化工、轻化工等领域具有广阔应用前景;茶多糖具有多种生理作用,如防辐射、抗凝血及抗血栓、降血糖等,它还可以增强机体免疫功能,实验证明茶多糖具有降血压、减慢心率、耐缺氧、增加冠状动脉流量等作用,还可以降血脂、抗动脉硬化、抗炎和抗癌等。因此,安全有效的从茶叶中提取和分离茶氨酸、茶多酚和茶多糖等成分,具有重要的实际意义。
目前的茶叶活性物质的提取方式存在提取率较低、溶剂用量大、有效成分提取物纯度低等缺点。
发明内容
针对上述问题,本发明基于超声波提取技术、超滤膜技术和离子交换树脂技术提供一种高效节能的提取茶叶活性物质的工艺方法。
为达到上述目的,本发明所提出的技术方案为:一种提取茶叶活性物质的工艺方法,其特征在于:包括如下步骤:
1)茶叶粉末中加入10-30倍的水,在超声波作用下,60℃-80℃温度范围内,每次15-30分钟,反复浸泡提取3-5次,经过滤分离得到茶叶提取液;
2)茶叶提取液经真空浓缩,将浓缩液和无水乙醇溶液按照1:3-1:5体积比均匀混合并搅拌,过滤后分别得到多糖沉淀和溶液;将多糖沉淀用水溶解,除去蛋白后利用PH值1-3的强酸阳离子交换树脂吸附与分离,得到茶多糖;
3)将多糖提取后溶液,用酸调pH3.5-5,经超滤膜过滤,得到膜透过液和截留液,截留液经离心机过滤得到滤液和蛋白饲料固形物;将滤液与膜透过液合并,然后用阳离子交换树脂进行提取,得到吸附有茶多酚的树脂和酸性水溶液;将吸附有茶多酚的阳离子交换树脂加到无水乙醇中,于50-60℃慢速搅拌下进行分离,将乙醇分离液用反渗透膜分离与浓缩,得到膜透过液和截留液,截留液经低温真空干燥,得到含乙醇的馏出液和茶多酚;
4)将馏出液,调整pH值为1-5,然后经过阳离子交换树脂柱进行离子交换,用10%浓度氨水洗脱,收集流出液,浓缩,降温、结晶、干燥可得到茶氨酸。
进一步方案,
步骤2中所述的分离所用溶剂为无水乙醇、乙酸、丙酮中的任意一种。
进一步方案,
所述步骤3中调节PH值所用酸为盐酸、硫酸和磷酸中的任意一种。
进一步方案,
所述步骤3中的超滤膜选用孔径O.8-5微米毛细管式超滤膜,操作压力为1.2-1.6Mpa。
进一步方案,
步骤4中经过氨水洗脱的流出液浓缩至1/3-1/5。
进一步方案,
步骤2所述的除去蛋白的方法为碱性蛋白酶解法,所用碱性蛋白酶量为1600-2000U/g。
进一步方案,
所述的酶解法工艺条件为:PH值9-10,温度25-30℃,时间45-75min。
有益效果:
1)本发明的一种提取茶叶活性物质的工艺方法以水为溶剂,采用超声波提取茶叶中的有效成分,具有提取时间短、效率高、活性物质损失少等优点;
2)本发明的一种提取茶叶活性物质的工艺方法采用离子交换树脂分离茶氨酸、茶多酚及茶多糖三种活性物质,树脂材料可重复使用,具有较高的性价比;
3)本发明的一种提取茶叶活性物质的工艺方法用酶解法去除茶叶蛋白,具有高效环保的优点;
4)本发明的一种提取茶叶活性物质的工艺方法得到的提取物纯度高,茶多糖总糖含量≥72%;茶多酚的纯度≥99%;茶氨酸的纯度≥85%。
具体实施方式
为使本发明的上述目的、特征和优点能够更加明显易懂,下面对本发明的具体实施方式做详细说明。在下面的描述中阐述了很多具体细节以便于充分理解本发明。但是本发明能够以很多不同于在此描述的其它方式来实施,本领域技术人员可以在不违背本发明内涵的情况下做类似改进,因此本发明不受下面公开的具体实施方式的限制。
下面将结合实施方式对本发明作进一步说明。
实施例1
1)茶叶粉末中加入10倍的水,在超声波作用下,80℃温度范围内,每次15分钟,反复浸泡提取5次,经过滤分离得到茶叶提取液;
2)茶叶提取液经真空浓缩,将浓缩液和无水乙醇按照1:3体积比均匀混合并搅拌,过滤后分别得到多糖沉淀和溶液;将多糖沉淀用水溶解;用1600U/g碱性蛋白酶,工艺条件为PH值10,温度25℃,时间45min去除蛋白;利用PH值1的强酸阳离子交换树脂吸附与分离,得到茶多糖;
3)将多糖提取后溶液,用硫酸调pH3.5,经超滤膜过滤,得到膜透过液和截留液,截留液经离心机过滤得到滤液和蛋白饲料固形物;将滤液与膜透过液合并,然后用阳离子交换树脂进行提取,得到吸附有茶多酚的树脂和酸性水溶液;将吸附有茶多酚的阳离子交换树脂加到无水乙醇中,于50℃慢速搅拌下进行分离,将乙醇分离液用反渗透膜分离与浓缩,得到膜透过液和截留液,截留液经低温真空干燥,得到含乙醇的馏出液和茶多酚;
4)将馏出液,调整pH值为1,然后经过阳离子交换树脂柱进行离子交换,用10%浓度氨水洗脱,收集流出液,浓缩,降温、结晶、干燥可得到茶氨酸。
实施例2
1)茶叶粉末中加入30倍的水,在超声波作用下,60℃温度范围内,每次30分钟,反复浸泡提取3次,经过滤分离得到茶叶提取液;
2)茶叶提取液经真空浓缩,将浓缩液和乙酸按照1:5体积比均匀混合并搅拌,过滤后分别得到多糖沉淀和溶液;将多糖沉淀用水溶解;用2000U/g碱性蛋白酶,工艺条件为PH值9,温度30℃,时间75min去除蛋白;利用PH值3的强酸阳离子交换树脂吸附与分离,得到茶多糖;
3)将多糖提取后溶液,用磷酸调pH5,经超滤膜过滤,得到膜透过液和截留液,截留液经离心机过滤得到滤液和蛋白饲料固形物;将滤液与膜透过液合并,然后用阳离子交换树脂进行提取,得到吸附有茶多酚的树脂和酸性水溶液;将吸附有茶多酚的阳离子交换树脂加到无水乙醇中,于60℃慢速搅拌下进行分离,将乙醇分离液用反渗透膜分离与浓缩,得到膜透过液和截留液,截留液经低温真空干燥,得到含乙醇的馏出液和茶多酚;
4)将馏出液,调整pH值为5,然后经过阳离子交换树脂柱进行离子交换,用10%浓度氨水洗脱,收集流出液,浓缩,降温、结晶、干燥可得到茶氨酸。
实施例3
1)茶叶粉末中加入20倍的水,在超声波作用下,70℃温度范围内,每次25分钟,反复浸泡提取4次,经过滤分离得到茶叶提取液;
2)茶叶提取液经真空浓缩,将浓缩液和丙酮按照1:4体积比均匀混合并搅拌,过滤后分别得到多糖沉淀和溶液;将多糖沉淀用水溶解;用1800U/g碱性蛋白酶,工艺条件为PH值9.5,温度28℃,时间60min去除蛋白;利用PH值2的强酸阳离子交换树脂吸附与分离,得到茶多糖;
3)将多糖提取后溶液,用硫酸调pH4,经超滤膜过滤,得到膜透过液和截留液,截留液经离心机过滤得到滤液和蛋白饲料固形物;将滤液与膜透过液合并,然后用阳离子交换树脂进行提取,得到吸附有茶多酚的树脂和酸性水溶液;将吸附有茶多酚的阳离子交换树脂加到无水乙醇中,于55℃慢速搅拌下进行分离,将乙醇分离液用反渗透膜分离与浓缩,得到膜透过液和截留液,截留液经低温真空干燥,得到含乙醇的馏出液和茶多酚;
4)将馏出液,调整pH值为3,然后经过阳离子交换树脂柱进行离子交换,用10%浓度氨水洗脱,收集流出液,浓缩,降温、结晶、干燥可得到茶氨酸。
虽然在上文中已经参考了一些实施例对本发明进行描述,然而在不脱离本发明的范围的情况下,可以对其进行各种改进并且可以用等效无替换其中的技术点,尤其是,只要不存在技术冲突,本发明所纰漏的各种实施例中的各项特征均可通过任一方式结合起来使用,在本发明中未对这些组合的情况进行穷举行的描述仅仅是处于省略篇幅和节约资源的考虑。因此,本发明并不局限于文中公开的特定实施例,而且包括落入权利要求。
Claims (7)
1.一种提取茶叶活性物质的工艺方法,首先将茶叶粉碎至30-50目,其特征在于:方法的步骤如下:
1)茶叶粉末中加入10-30倍的水,在超声波作用下,60℃-80℃温度范围内,每次15-30分钟,反复浸泡提取3-5次,经过滤分离得到茶叶提取液;
2)茶叶提取液经真空浓缩,将浓缩液和溶剂按照1:3-1:5体积比均匀混合并搅拌,过滤后分别得到多糖沉淀和溶液;将多糖沉淀用水溶解,除去蛋白后利用PH值1-3的强酸阳离子交换树脂吸附与分离,得到茶多糖;
3)将多糖提取后溶液,用酸调pH3.5-5,经超滤膜过滤,得到膜透过液和截留液,截留液经离心机过滤得到滤液和蛋白饲料固形物;将滤液与膜透过液合并,然后用阳离子交换树脂进行提取,得到吸附有茶多酚的树脂和酸性水溶液;将吸附有茶多酚的阳离子交换树脂加到无水乙醇中,于50-60℃慢速搅拌下进行分离,将乙醇分离液用反渗透膜分离与浓缩,得到膜透过液和截留液,截留液经低温真空干燥,得到含乙醇的馏出液和茶多酚;
4)将馏出液,调整pH值为1-5,然后经过阳离子交换树脂柱进行离子交换,用10%浓度氨水洗脱,收集流出液,浓缩,降温、结晶、干燥可得到茶氨酸。
2.根据权利要求1所述的一种提取茶叶活性物质的工艺方法,其特征在于:步骤2中所述的分离所用溶剂为无水乙醇、乙酸、丙酮中的任意一种。
3.根据权利要求1所述的一种提取茶叶活性物质的工艺方法,其特征在于:所述步骤3中调节PH值所用酸为盐酸、硫酸和磷酸中的任意一种。
4.根据权利要求1所述的一种提取茶叶活性物质的工艺方法,其特征在于:所述步骤3中的超滤膜选用孔径O.8-5微米毛细管式超滤膜,操作压力为1.2-1.6Mpa。
5.根据权利要求1所述的一种提取茶叶活性物质的工艺方法,其特征在于:所述步骤4中经过氨水洗脱的流出液浓缩至1/3-1/5。
6.根据权利要求1所述的一种提取茶叶活性物质的工艺方法,其特征在于:所述的除去蛋白的方法为碱性蛋白酶解法,所用碱性蛋白酶量为1600-2000U/g。
7.根据权利要求6所述的一种提取茶叶活性物质的工艺方法,其特征在于:所述的酶解法工艺条件为:PH值9-10,温度25-30℃,时间45-75min。
