Abstract
本发明涉及一种调节免疫力的组合物及其制备方法和应用,所述组合物包括如下原料:杜仲雄花、雪莲培养物、新鲜芦笋花、新鲜小麦胚芽、覆盆子、枸杞、黄精、桑椹、花生低聚肽、核桃低聚肽和桃胶。本发明提出的调节免疫力的组合物,以植物内源激素、多糖和植物低聚肽为主要活性成分,通过调补结合,调节内分泌,促进机体新陈代谢,改善器官机能,显著增强机体免疫力,且采用天然植物源原料加工,安全无副作用,特别适合更年期及其后的中老年人使用。

技术领域
本发明属于中药和食品技术领域,具体涉及一种调节免疫力的组合物及其制备方法和应用。
背景技术
随着生命科学和医学研究的发展,人们发现男性和女性都有更年期,这是一个人随着年龄增长进入老化的必然胜利过程。更年期的变化主要是性腺逐渐衰萎所致。性腺在女性为卵巢,男性为睾丸。更年期间卵巢***、分泌***和孕激素的能力不规则的减退,表现为***紊乱。男性就表现为性功能下降。除性腺外,其他各内分泌腺的功能均有变化,同时内脏器官慢慢老化,造成机体免疫力下降,常出现体虚多病,疲倦乏力、腰膝酸软、头晕耳鸣、失眠多梦、心慌气短、消化不良等不良症状。因此,如何有效提高免疫力一直是中老年人的重要健康诉求。
目前市面上提高免疫力产品的功效成分主要有植物多糖、皂苷、益生元、益生菌、蛋白质、大豆异黄酮等多种物质,此类功效成分虽然对更年期及其后中老年人提高免疫力也有一定的帮助,但是由于它们并没有着重针对更年期及其后中老年人激素水平下降,进而引起免疫力下降这个原理进行产品研究开发,因此,产品没有针对性,效果并不稳定和显著。
发明内容
针对上述情况,为克服现有技术上的不足,本发明的目的就是提供调节免疫力的组合物及其制备方法和应用,通过合理的提取制备方法,以植物激素调理为主、并补充多糖和多肽等进行复配,更有效的提高更年期及其后中老年人的免疫力。
本发明的增强免疫力的组合物,包括如下重量份数的原料:杜仲雄花5~25份、雪莲培养物1~10份、新鲜芦笋花8~25份、新鲜小麦胚芽1~10份、覆盆子10~30份、枸杞30~50份、黄精10~30份、桑椹10~30份、花生低聚肽10~30份、核桃低聚肽20~40份和桃胶3~18份。
本发明提出的调节免疫力的组合物,通过合理的提取方法萃取原料多种活性成分,以植物***为主,发挥其激素作用,调节内分泌***,延缓性腺退化,维持机体新陈代谢;植物多糖通过介导细胞信号而激活多种免疫细胞,植物多肽则能增强免疫功能。三者有机结合,更有效改善机体器官功能,明显提高机体免疫力。

作为本发明进一步优选的实施方式,所述的调节免疫力的组合物,还包括如下重量份数的原料:人参1~5份、百合3~15份、葛根5~10份、龙眼5~20份、灵芝1~10份、银耳3~10份、大豆20~50份和玉米低聚肽10~30份。

本发明的另一个目的在于提出所述的调节免疫力的组合物的制备方法。
所述的调节免疫力的组合物的制备方法,包括如下步骤:S101:将所述杜仲雄花、雪莲培养物、新鲜芦笋花、新鲜小麦胚芽、覆盆子、枸杞、黄精、桑椹、人参、百合、葛根、龙眼、灵芝、银耳与大豆混合,捣碎打浆,移入发酵罐内,然后调节温度为25℃~37℃,再加入质量分数为0.5%~3%的双歧杆菌和质量分数为0.5%~3%的乳杆菌混合菌液,并发酵12h~36h;S102:向所述步骤S101得到的发酵物中添加酒精并调节酒精度,然后浸泡1h~3h;S103:将所述步骤S102得到的发酵物离心分离,分别得到滤液和滤渣,首先用旋转蒸馏器蒸馏所述滤液以去除乙醇,然后进行真空冷冻干燥,得到冻干粉;然后将所述滤渣用水浸泡并搅拌1h~2h,然后依次进行离心分离、过滤、真空浓缩、喷雾干燥,得到含多糖的水溶性成分的粉末;S104:将所述冻干粉和所述含多糖的水溶性成分的粉末与所述剩余原料混合,得到增强免疫力的组合物。
进一步地,在所述步骤S102中,所述酒精度为65%~75%。
进一步地,在所述步骤S103中,蒸馏温度为35℃~40℃。
本发明的再一个目的在于提出所述的组合物在制备调节免疫力的药物和保健食品中的应用。
与现有技术相比,本发明的有益效果是:
1)特别针对更年期及其后中老年人激素下降、免疫力低下的情况,具有明显提高免疫力、改善体质的效果。
2)通过发酵萃取、冷冻分离等工艺提取多种药食两用植物活性成分,活性成分保留率高,而且对人体安全、无副作用。
3)产品口感风味好,易被中老年人接受。

下面详细描述本发明的实施例,所述实施例是示例性的,旨在用于解释本发明,而不能理解为对本发明的限制。
下述实施例中的实验方法,如无特殊说明,均为常规方法。
下述实施例中所用的试验材料,如无特殊说明,均为自常规试剂商店购买得到的。
以下实施例中的定量试验,均设置三次重复实验,数据为三次重复实验的平均值或平均值±标准差。
下述实施例中所用的中药均符合中国药典(2015年版)一部正文各药材项下的有关规定。投料前,通过鉴定,各味药材实物与名称相符,质量符合标准。
本发明实施例中所采用的菌种和其他原料的具体来源如下:
(1)双歧杆菌,哈尔滨美华生物技术股份有限公司;
(2)乳杆菌,山东向日葵生物科技有限公司。
(3)雪莲培养物,是在无菌条件下,将雪莲离体的植物器官、组织、细胞、胚胎或原生质体培养在人工配制的培养基上,在人工控制的条件下进行培养,使其组织或细胞增值进而按照需要进行培养的技术;或使其脱分化产生愈伤组织,并再逐步分化出器官并长成完整的植株而生产的培养物。
下面通过具体实施例详细描述本发明:
实施例1
实施例1的增强免疫力的组合物,包括如下重量份数的原料:杜仲雄花5份、雪莲培养物10份、新鲜芦笋花8份、新鲜小麦胚芽10份、覆盆子10份、枸杞50份、黄精10份、桑椹30份、花生低聚肽10份、核桃低聚肽40份和桃胶3份。
实施例1的增强免疫力的组合物的制备方法,包括如下步骤:
(1)将所述杜仲雄花、雪莲培养物、新鲜芦笋花、新鲜小麦胚芽、覆盆子、枸杞、黄精、桑椹混合,捣碎打浆,移入发酵罐内,然后调节温度为25℃,再加入质量分数为3%的双歧杆菌和质量分数为0.5%的乳杆菌混合菌液,并发酵36h。
(2)向所述步骤(1)得到的发酵物中添加酒精并调节酒精度为65%,然后浸泡3h。
(3)将所述步骤(2)得到的发酵物离心分离,分别得到滤液和滤渣,首先用旋转蒸馏器在35℃下蒸馏所述滤液以去除乙醇,然后进行真空冷冻干燥,得到冻干粉;然后将所述滤渣用水浸泡并搅拌2h,然后依次进行离心分离、过滤、真空浓缩、喷雾干燥,得到含多糖的水溶性成分的粉末。
(4)将所述冻干粉和所述含多糖的水溶性成分的粉末与所述剩余原料混合,得到增强免疫力的组合物。
实施例2
实施例2的增强免疫力的组合物,包括如下重量份数的原料:杜仲雄花25份、雪莲培养物1份、新鲜芦笋花25份、新鲜小麦胚芽1份、覆盆子30份、枸杞30份、黄精30份、桑椹10份、花生低聚肽30份、核桃低聚肽20份和桃胶18份。
实施例2的增强免疫力的组合物的制备方法,包括如下步骤:
(1)将所述杜仲雄花、雪莲培养物、新鲜芦笋花、新鲜小麦胚芽、覆盆子、枸杞、黄精、桑椹混合,捣碎打浆,移入发酵罐内,然后调节温度为37℃,再加入质量分数为0.5%的双歧杆菌和质量分数为3%的乳杆菌混合菌液,并发酵12h。
(2)向所述步骤(1)得到的发酵物中添加酒精并调节酒精度为75%,然后浸泡1h。
(3)将所述步骤(2)得到的发酵物离心分离,分别得到滤液和滤渣,首先用旋转蒸馏器在40℃下蒸馏所述滤液以去除乙醇,然后进行真空冷冻干燥,得到冻干粉;然后将所述滤渣用水浸泡并搅拌1h,然后依次进行离心分离、过滤、真空浓缩、喷雾干燥,得到含多糖的水溶性成分的粉末。
(4)将所述冻干粉和所述含多糖的水溶性成分的粉末与所述剩余原料混合,得到增强免疫力的组合物。
实施例3
实施例3的增强免疫力的组合物,包括如下重量份数的原料:杜仲雄花15份、雪莲培养物5份、新鲜芦笋花17份、新鲜小麦胚芽6份、覆盆子22份、枸杞38份、黄精20份、桑椹20份、花生低聚肽20份、核桃低聚肽30份和桃胶10份。
实施例3的增强免疫力的组合物的制备方法,包括如下步骤:
(1)将所述杜仲雄花、雪莲培养物、新鲜芦笋花、新鲜小麦胚芽、覆盆子、枸杞、黄精、桑椹混合,捣碎打浆,移入发酵罐内,然后调节温度为29℃,再加入质量分数为1.75%的双歧杆菌和质量分数为1.75%的乳杆菌混合菌液,并发酵24h。
(2)向所述步骤(1)得到的发酵物中添加酒精并调节酒精度为70%,然后浸泡2h。
(3)将所述步骤(2)得到的发酵物离心分离,分别得到滤液和滤渣,首先用旋转蒸馏器在37℃下蒸馏所述滤液以去除乙醇,然后进行真空冷冻干燥,得到冻干粉;然后将所述滤渣用水浸泡并搅拌1.5h,然后依次进行离心分离、过滤、真空浓缩、喷雾干燥,得到含多糖的水溶性成分的粉末。
(4)将所述冻干粉和所述含多糖的水溶性成分的粉末与所述剩余原料混合,得到增强免疫力的组合物。
实施例4
实施例4的增强免疫力的组合物,包括如下重量份数的原料:杜仲雄花5份、雪莲培养物10份、新鲜芦笋花8份、新鲜小麦胚芽10份、覆盆子10份、枸杞50份、黄精10份、桑椹30份、花生低聚肽10份、核桃低聚肽40份、桃胶3份、人参5份、百合3份、葛根10份、龙眼5份、灵芝10份、银耳3份、大豆50份和玉米低聚肽10份。
实施例4的增强免疫力的组合物的制备方法,包括如下步骤:
(1)将所述杜仲雄花、雪莲培养物、新鲜芦笋花、新鲜小麦胚芽、覆盆子、枸杞、黄精、桑椹、人参、百合、葛根、龙眼、灵芝、银耳与大豆混合,捣碎打浆,移入发酵罐内,然后调节温度为25℃,再加入质量分数为3%的双歧杆菌和质量分数为0.5%的乳杆菌混合菌液,并发酵36h。
(2)向所述步骤(1)得到的发酵物中添加酒精并调节酒精度为65%,然后浸泡3h。
(3)将所述步骤(2)得到的发酵物离心分离,分别得到滤液和滤渣,首先用旋转蒸馏器在35℃下蒸馏所述滤液以去除乙醇,然后进行真空冷冻干燥,得到冻干粉;然后将所述滤渣用水浸泡并搅拌2h,然后依次进行离心分离、过滤、真空浓缩、喷雾干燥,得到含多糖的水溶性成分的粉末。
(4)将所述冻干粉和所述含多糖的水溶性成分的粉末与所述剩余原料混合,得到增强免疫力的组合物。
实施例5
实施例5的增强免疫力的组合物,包括如下重量份数的原料:杜仲雄花25份、雪莲培养物1份、新鲜芦笋花25份、新鲜小麦胚芽1份、覆盆子30份、枸杞30份、黄精30份、桑椹10份、花生低聚肽30份、核桃低聚肽20份、桃胶18份、人参1份、百合15份、葛根5份、龙眼20份、灵芝1份、银耳10份、大豆20份和玉米低聚肽30份。
实施例5的增强免疫力的组合物的制备方法,包括如下步骤:

(2)向所述步骤(1)得到的发酵物中添加酒精并调节酒精度为75%,然后浸泡1h。
(3)将所述步骤(2)得到的发酵物离心分离,分别得到滤液和滤渣,首先用旋转蒸馏器在40℃下蒸馏所述滤液以去除乙醇,然后进行真空冷冻干燥,得到冻干粉;然后将所述滤渣用水浸泡并搅拌1h,然后依次进行离心分离、过滤、真空浓缩、喷雾干燥,得到含多糖的水溶性成分的粉末。
(4)将所述冻干粉和所述含多糖的水溶性成分的粉末与所述剩余原料混合,得到增强免疫力的组合物。
实施例6
实施例6的增强免疫力的组合物,包括如下重量份数的原料:杜仲雄花15份、雪莲培养物5份、新鲜芦笋花16份、新鲜小麦胚芽5份、覆盆子20份、枸杞40份、黄精20份、桑椹20份、花生低聚肽20份、核桃低聚肽30份、桃胶10份、人参2份、百合9份、葛根7份、龙眼12份、灵芝5份、银耳6份、大豆35份和玉米低聚肽20份。
实施例6的增强免疫力的组合物的制备方法,包括如下步骤:
(1)将所述杜仲雄花、雪莲培养物、新鲜芦笋花、新鲜小麦胚芽、覆盆子、枸杞、黄精、桑椹、人参、百合、葛根、龙眼、灵芝、银耳与大豆混合,捣碎打浆,移入发酵罐内,然后调节温度为31℃,再加入质量分数为1.75%的双歧杆菌和质量分数为1.75%的乳杆菌混合菌液,并发酵24h。
(2)向所述步骤(1)得到的发酵物中添加酒精并调节酒精度为70%,然后浸泡2h。
(3)将所述步骤(2)得到的发酵物离心分离,分别得到滤液和滤渣,首先用旋转蒸馏器在37℃下蒸馏所述滤液以去除乙醇,然后进行真空冷冻干燥,得到冻干粉;然后将所述滤渣用水浸泡并搅拌1.5h,然后依次进行离心分离、过滤、真空浓缩、喷雾干燥,得到含多糖的水溶性成分的粉末。
(4)将所述冻干粉和所述含多糖的水溶性成分的粉末与所述剩余原料混合,得到增强免疫力的组合物。

对比例1提出了一种增强免疫力的组合物,对比例1与实施例4的区别仅在于,对比例1中未向所述组合物中加入杜仲雄花、雪莲培养物、新鲜芦笋花、新鲜小麦胚芽,其余参数和步骤均相同。

对比例2提出了一种增强免疫力的组合物,对比例2与实施例5的区别仅在于,对比例2中未向所述组合物中加入覆盆子、枸杞、黄精、桑椹,其余参数和步骤均相同。

对比例3提出了一种增强免疫力的组合物,对比例3与实施例6的区别仅在于,对比例3中未向所述组合物中加入花生低聚肽、核桃低聚肽,其余参数和步骤均相同。
对比例4
对比例4为空白对照组。
一、毒性试验
随机选体重为18~22g的小白鼠450只,分为9组,每组小白鼠背部去毛露出外表皮,去毛范围2cm×2cm,分别涂抹本发明实施例1至对比例3制备得到的组合物,给药剂量为0.5g,连续给药30天。给药期间及停药后1周时,观察白鼠生长状态和活动饮食,鉴定血液学、血液生化学、脏器组织结构以及尿常规等。结果发现所有小白鼠均健康生存,无任何毒副作用;解剖观察血象、肝功能、各脏器组织状态,与正常小白鼠比较无差异。实验证明,本发明提供的组合物无毒,用药安全可靠。
二、小鼠试验
随机选体重为18~22g的12月龄雌性wistar小鼠(自然更年期模型)100只,平均分为10组,依次对9组小鼠连续灌胃30天由实施例1-对比例3制备的增强免疫力的组合物,每天0.5g,检测其对小鼠抗体生成细胞数和血清溶血素水平的影响,对对比例4的小鼠灌胃相同数量的纯净水,结果见表1。

溶血空斑试验是指将抗体生成细胞和红细胞混合于凝胶薄层中,抗体生成细胞所释出的抗体与周围红细胞结合,在补体作用下形成溶血空斑,从而进行定量计数。从表1中可以看出,实施例1-6的溶血空斑数明显比对比例1-4的溶血空斑数有明显增加,同时,实施例4-6的溶血空斑数相比于实施例1-3也略有增加。
半数溶血值是指使50的标准致敏红细胞悬浮液溶解的补体量,这样可以得出一个较精确的终点。表中可以看出,实施例1-6的半数溶血值相比于对比例1-4具有明显增加,且实施例4-6相比与实施例1-3也有一定程度的增加。
表1可以得出,实施例1-6与对比例1-4相比具有显著性差异。
连续灌胃30天后,无菌取脾,并将脾脏磨碎,过200目筛网后,置于盛有Hank’s洗液的小平皿冲洗3次,然后用完全1640培养液配成3×106个/mL细胞悬浮,再加入100μL靶细胞(YAC-1细胞,4×105个/mL),设置靶细胞自然释放孔(靶细胞100μL+1%NP-40 100μL)各3孔,放置于37℃5%CO2培养箱中培养4h,离心5min。取各孔上清液100μL置于另一96孔板中,每孔再加入100μL基质液,10min后加30μL 1mol/L HCl终止反应,再测定492nm出的O.D.值,按照公式1计算NK细胞活性率,测定结果见表2。

从表2可以看出,实施例1-6与对比例1-4相比具有显著性差异。
综上,本发明的增强免疫力的组合物,特别针对更年期及其后中老年人激素下降、免疫力低下的情况,具有明显提高免疫力、改善体质的效果。通过发酵萃取、冷冻分离等工艺提取多种药食两用植物活性成分,活性成分保留率高,而且对人体安全、无副作用,且口感风味好,易被中老年人接受。

Claims (9)
1.一种调节免疫力的组合物,其特征在于,包括如下重量份数的原料:杜仲雄花5~25份、雪莲培养物1~10份、新鲜芦笋花8~25份、新鲜小麦胚芽1~10份、覆盆子10~30份、枸杞30~50份、黄精10~30份、桑椹10~30份、花生低聚肽10~30份、核桃低聚肽20~40份和桃胶3~18份。
2.根据权利要求1所述的调节免疫力的组合物,其特征在于,还包括如下重量份数的原料:人参1~5份、百合3~15份、葛根5~10份、龙眼5~20份、灵芝1~10份、银耳3~10份、大豆20~50份和玉米低聚肽10~30份。
3.根据权利要求2所述的调节免疫力的组合物,其特征在于,包括如下重量份数的原料:杜仲雄花5份、雪莲培养物10份、新鲜芦笋花8份、新鲜小麦胚芽10份、覆盆子10份、枸杞50份、黄精10份、桑椹30份、花生低聚肽10份、核桃低聚肽40份、桃胶3份、人参5份、百合3份、葛根10份、龙眼5份、灵芝10份、银耳3份、大豆50份和玉米低聚肽10份。
4.根据权利要求2所述的调节免疫力的组合物,其特征在于,包括如下重量份数的原料:杜仲雄花25份、雪莲培养物1份、新鲜芦笋花25份、新鲜小麦胚芽1份、覆盆子30份、枸杞30份、黄精30份、桑椹10份、花生低聚肽30份、核桃低聚肽20份、桃胶18份、人参1份、百合15份、葛根5份、龙眼20份、灵芝1份、银耳10份、大豆20份和玉米低聚肽30份。
5.根据权利要求2所述的调节免疫力的组合物,其特征在于,包括如下重量份数的原料:杜仲雄花15份、雪莲培养物5份、新鲜芦笋花16份、新鲜小麦胚芽5份、覆盆子20份、枸杞40份、黄精20份、桑椹20份、花生低聚肽20份、核桃低聚肽30份、桃胶10份、人参2份、百合9份、葛根7份、龙眼12份、灵芝5份、银耳6份、大豆35份和玉米低聚肽20份。
6.权利要求1-5任一项所述的调节免疫力的组合物的制备方法,其特征在于,包括如下步骤:
S101:将所述杜仲雄花、雪莲培养物、新鲜芦笋花、新鲜小麦胚芽、覆盆子、枸杞、黄精、桑椹、人参、百合、葛根、龙眼、灵芝、银耳与大豆混合,捣碎打浆,移入发酵罐内,然后调节温度为25℃~37℃,再加入质量分数为0.5%~3%的双歧杆菌和质量分数为0.5%~3%的乳杆菌混合菌液,并发酵12h~36h;
S102:向所述步骤S101得到的发酵物中添加酒精并调节酒精度,然后浸泡1h~3h;
S103:将所述步骤S102得到的发酵物离心分离,分别得到滤液和滤渣,首先用旋转蒸馏器蒸馏所述滤液以去除乙醇,然后进行真空冷冻干燥,得到冻干粉;然后将所述滤渣用水浸泡并搅拌1h~2h,然后依次进行离心分离、过滤、真空浓缩、喷雾干燥,得到含多糖的水溶性成分的粉末;
S104:将所述冻干粉和所述含多糖的水溶性成分的粉末与所述剩余原料混合,得到增强免疫力的组合物。
7.根据权利要求6所述的调节免疫力的组合物的制备方法,其特征在于:在所述步骤S102中,所述酒精度为65%~75%。
8.根据权利要求6所述的调节免疫力的组合物的制备方法,其特征在于:在所述步骤S103中,蒸馏温度为35℃~40℃。
9.权利要求1-5任一项所述的组合物在制备调节免疫力的药物和保健食品中的应用。
