背景技术

脂肪酸合成酶抑制剂作为一种作用于肠道内的减肥药物,不进入血液,不会产生中枢神经性副作用,不影响矿物质代谢,是近年来研究的热点。而各种植物提取物作为脂肪酸合成酶抑制剂的来源,具有非常巨大的潜力,且其安全性更高、来源更广泛,必将在新型减肥药品或保健品开发中占有重要的地位。
石榴皮是水果石榴(Punica granatum L.)干燥后的果皮。石榴皮中的化学成分以鞣质类、生物碱、有机酸和黄酮类为主。研究表明,石榴皮中含有较高含量的安石榴苷、鞣花酸等多酚类物质,具有很强的脂肪酸合成酶抑制活性。丁香为桃金娘科植物丁香(Syzygium aromaticum (L.) Merr. & L. M. Perry)的干燥花蕾,其主要活性成分有丁香酚、丁香酚酯、β-石竹烯、单宁酸、水杨酸甲酯、山奈酚、鼠李黄素、桃素亭以及香草醛等。研究证实,丁香提取物具有脂肪酸合成酶抑制活性。桑叶为桑(Morus alba L.)的叶片,其性平,味甘,具有疏散风热、清肺涧燥、清肝明目的功效,是临床上运用较多的一味中药。现代化学研究表明,桑叶中含有黄酮类、酚酸类、生物碱类及多糖类等活性成分。桑叶所含多酚具有降血糖、降血脂、降血压、抗病毒、抑制脂肪酸合成酶等多种生理活性。决明子为豆科决明属植物决明(Cassia tora Linn.)的干燥成熟种子。决明子始载于《神农本草经》,列为上品。1993年,决明子被国家***列为药食同源原料。
发明内容
本发明的目的在于提供一种植物减肥功能饮料,该饮料由石榴皮、丁香、桑叶、决明子这三种药食同源的原料制成,纯天然,不含有毒有害化学物质,可用于肥胖的预防保健。
为实现上述目的,本发明采用如下技术方案:
一种植物减肥功能饮料,按重量百分数之和为100%计,其所含各成分的重量百分数为:石榴皮多酚提取物0.11~0.52%、丁香黄酮提取物0.30~0.68%、桑叶多酚提取物0.18~0.62%、决明子蒽醌提取物0.15~0.38%、蔗糖0.02~0.08%、阿斯巴甜0.01~0.05%、安赛蜜0.01~0.05%、维生素C 0.05~0.20%、柠檬酸0.015~0.03%,其余为纯化水。
所述植物减肥功能饮料的制备方法包括如下步骤:
(1)石榴皮多酚提取物的制备:收集石榴果皮,切碎,在40~60℃下低温烘干,粉碎,过200目筛;然后按料液比1:15~1:25 g/mL加入质量浓度60%的乙醇溶液,超声提取5~10min,过滤,滤渣重复提取1~2次,合并提取液,得到石榴果皮提取液,减压浓缩至无醇味,上D101大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度20%、40%、60%、80%、100%的乙醇溶液进行梯度洗脱,收集60%~80%部位的乙醇洗脱液,60℃下真空减压浓缩,得到石榴皮多酚提取物;
(2)丁香黄酮提取物的制备:收集丁香花蕾,40~60℃条件下烘干,粉碎,过120目筛;然后按料液比1:15~1:25 g/mL加入质量浓度75%的乙醇溶液,超声提取10~15 min,过滤,滤渣重复提取1~2次,合并提取液,得到丁香提取液,减压浓缩至无醇味,上D101大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度10%、30%、50%、70%、100%的乙醇溶液进行梯度洗脱,收集30%~50%部位的乙醇洗脱液,60℃下真空减压浓缩,得到丁香黄酮提取物;
(3)桑叶多酚提取物的制备:收集桑叶,40~60℃条件下烘干,粉碎,过200目筛;然后按料液比1:20~1:30 g/mL加入质量浓度70%的乙醇溶液,超声提取30~45 min,过滤,滤渣重复提取1~2次,合并提取液,得到桑叶提取液,减压浓缩至无醇味后,上AB-8大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度10%、30%、50%、70%、100%的乙醇溶液进行梯度洗脱,收集50%~70%部位的乙醇洗脱液,60℃下真空减压浓缩,得到桑叶多酚提取物;
(4)决明子蒽醌提取物的制备:决明子经40~60℃烘干,粉碎,过200目筛;然后按料液比1:10~1:20 g/mL加入质量浓度90%的乙醇溶液,超声提取30~45 min,过滤,滤渣重复提取1~2次,合并提取液,得到决明子提取液,经减压浓缩至无醇味后,上AB-8大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度20%、40%、60%、80%、100%的乙醇溶液进行梯度洗脱,收集80%部位的乙醇洗脱液,60℃下真空减压浓缩,得到决明子蒽醌提取物;
(5)复配:将所得石榴皮多酚提取物、丁香黄酮提取物、桑叶多酚提取物、决明子蒽醌提取物与蔗糖、阿斯巴甜、安赛蜜、维生素C、柠檬酸、纯化水按比例混合搅拌均匀;
(6)灌装灭菌:将配制好的饮料灌装于易拉罐瓶中,于115-120℃下灭菌15-20 min,然后冷却至室温,得到含有四种植物提取物成分的所述植物减肥功能饮料。
本发明的显著优点在于:
(1)本发明以脂肪酸合成酶作为减肥的重要靶点制作的植物减肥功能饮料主要以石榴皮以及丁香、桑叶、决明子这三种药食同源植物的提取物组成,其多成分协同作用,功效更强,且天然、安全、低毒。
(2)本发明制备工艺简单,成本低廉,且只在提取和分离的过程中使用乙醇,不使用其他毒性溶剂,不会对环境造成污染,提取物中也不存在毒性溶剂残留问题,产品品质可得到有效保证。
(3)本发明功能饮料绿色天然,口感酸甜,具有清香,经成分分析表明,其总多酚含量为505 mg/100 g、总黄酮含量为480 mg/100 g、总蒽醌含量为265 mg/100 g,能起到预防肥胖的保健功效。
具体实施方式
为了使本发明所述的内容更加便于理解,下面结合具体实施方式对本发明所述的技术方案做进一步的说明,但是本发明不仅限于此。
实施例1
(1)石榴皮多酚提取物的制备:收集石榴果皮,切碎,在40℃下低温烘干,粉碎,过200目筛;然后按料液比1:15 g/mL加入质量浓度60%的乙醇溶液,超声提取5 min,过滤,滤渣重复提取1次,合并提取液,得到石榴果皮提取液,减压浓缩至无醇味,上D101大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度20%、40%、60%、80%、100%的乙醇溶液进行梯度洗脱,收集60%部位的乙醇洗脱液,60℃下真空减压浓缩,得到石榴皮多酚提取物;
(2)丁香黄酮提取物的制备:收集丁香花蕾,40℃条件下烘干,粉碎,过120目筛;然后按料液比1:15 g/mL加入质量浓度75%的乙醇溶液,超声提取10 min,过滤,滤渣重复提取1次,合并提取液,得到丁香提取液,减压浓缩至无醇味,上D101大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度10%、30%、50%、70%、100%的乙醇溶液进行梯度洗脱,收集30%部位的乙醇洗脱液,60℃下真空减压浓缩,得到丁香黄酮提取物;
(3)桑叶多酚提取物的制备:收集桑叶,40℃条件下烘干,粉碎,过200目筛;然后按料液比1:20 g/mL加入质量浓度70%的乙醇溶液,超声提取30 min,过滤,滤渣重复提取1次,合并提取液,得到桑叶提取液,减压浓缩至无醇味后,上AB-8大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度10%、30%、50%、70%、100%的乙醇溶液进行梯度洗脱,收集50%部位的乙醇洗脱液,60℃下真空减压浓缩,得到桑叶多酚提取物;
(4)决明子蒽醌提取物的制备:决明子经40℃烘干,粉碎,过200目筛;然后按料液比1:10 g/mL加入质量浓度90%的乙醇溶液,超声提取30 min,过滤,滤渣重复提取1次,合并提取液,得到决明子提取液,经减压浓缩至无醇味后,上AB-8大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度20%、40%、60%、80%、100%的乙醇溶液进行梯度洗脱,收集80%部位的乙醇洗脱液,60℃下真空减压浓缩,得到决明子蒽醌提取物;
(5)复配:按石榴皮多酚提取物0.11%、丁香黄酮提取物0.30%、桑叶多酚提取物0.18%、决明子蒽醌提取物0.15%、蔗糖0.02%、阿斯巴甜0.01%、安赛蜜0.01%、维生素C 0.05%、柠檬酸0.015%、纯化水99.155%的比例混合搅拌均匀;
(6)灌装灭菌:将配制好的饮料灌装于易拉罐瓶中,于115℃下灭菌20 min,然后冷却至室温,得到含有四种植物提取物成分的所述植物减肥功能饮料。
将本实施例所得功能饮料以0.2 g/kg BW、0.4 g/kg BW、0.8 /kg BW的剂量经口灌胃给予肥胖模型小鼠饲喂30天,结果显示,各剂量组的体重和脂/体比明显低于模型对照组(P<0.05),并且各剂量组的摄食量不显著低于模型对照组(P>0.05),表明本发明所得植物减肥功能饮料具有辅助减肥功能。
实施例2
(1)石榴皮多酚提取物的制备:收集石榴果皮,切碎,在50℃下低温烘干,粉碎,过200目筛;然后按料液比1:20 g/mL加入质量浓度60%的乙醇溶液,超声提取8 min,过滤,滤渣重复提取1次,合并提取液,得到石榴果皮提取液,减压浓缩至无醇味,上D101大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度20%、40%、60%、80%、100%的乙醇溶液进行梯度洗脱,收集70%部位的乙醇洗脱液,60℃下真空减压浓缩,得到石榴皮多酚提取物;
(2)丁香黄酮提取物的制备:收集丁香花蕾,50℃条件下烘干,粉碎,过120目筛;然后按料液比1:20 g/mL加入质量浓度75%的乙醇溶液,超声提取12 min,过滤,滤渣重复提取1次,合并提取液,得到丁香提取液,减压浓缩至无醇味,上D101大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度10%、30%、50%、70%、100%的乙醇溶液进行梯度洗脱,收集40%部位的乙醇洗脱液,60℃下真空减压浓缩,得到丁香黄酮提取物;
(3)桑叶多酚提取物的制备:收集桑叶,50℃条件下烘干,粉碎,过200目筛;然后按料液比1:25 g/mL加入质量浓度70%的乙醇溶液,超声提取35 min,过滤,滤渣重复提取1次,合并提取液,得到桑叶提取液,减压浓缩至无醇味后,上AB-8大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度10%、30%、50%、70%、100%的乙醇溶液进行梯度洗脱,收集60%部位的乙醇洗脱液,60℃下真空减压浓缩,得到桑叶多酚提取物;
(4)决明子蒽醌提取物的制备:决明子经50℃烘干,粉碎,过200目筛;然后按料液比1:15 g/mL加入质量浓度90%的乙醇溶液,超声提取35 min,过滤,滤渣重复提取1次,合并提取液,得到决明子提取液,经减压浓缩至无醇味后,上AB-8大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度20%、40%、60%、80%、100%的乙醇溶液进行梯度洗脱,收集80%部位的乙醇洗脱液,60℃下真空减压浓缩,得到决明子蒽醌提取物;
(5)复配:按石榴皮多酚提取物0.20%、丁香黄酮提取物0.48%、桑叶多酚提取物0.52%、决明子蒽醌提取物0.28%、蔗糖0.04%、阿斯巴甜0.02%、安赛蜜0.02%、维生素C 0.1%、柠檬酸0.02%、纯化水98.32%的比例混合搅拌均匀;
(6)灌装灭菌:将配制好的饮料灌装于易拉罐瓶中,于120℃下灭菌18min,然后冷却至室温,得到含有四种植物提取物成分的所述植物减肥功能饮料。
实施例3
(1)石榴皮多酚提取物的制备:收集石榴果皮,切碎,在60℃下低温烘干,粉碎,过200目筛;然后按料液比1:25 g/mL加入质量浓度60%的乙醇溶液,超声提取10 min,过滤,滤渣重复提取2次,合并提取液,得到石榴果皮提取液,减压浓缩至无醇味,上D101大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度20%、40%、60%、80%、100%的乙醇溶液进行梯度洗脱,收集80%部位的乙醇洗脱液,60℃下真空减压浓缩,得到石榴皮多酚提取物;
(2)丁香黄酮提取物的制备:收集丁香花蕾,60℃条件下烘干,粉碎,过120目筛;然后按料液比1:25 g/mL加入质量浓度75%的乙醇溶液,超声提取15 min,过滤,滤渣重复提取2次,合并提取液,得到丁香提取液,减压浓缩至无醇味,上D101大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度10%、30%、50%、70%、100%的乙醇溶液进行梯度洗脱,收集50%部位的乙醇洗脱液,60℃下真空减压浓缩,得到丁香黄酮提取物;
(3)桑叶多酚提取物的制备:收集桑叶,60℃条件下烘干,粉碎,过200目筛;然后按料液比1:30 g/mL加入质量浓度70%的乙醇溶液,超声提取45 min,过滤,滤渣重复提取2次,合并提取液,得到桑叶提取液,减压浓缩至无醇味后,上AB-8大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度10%、30%、50%、70%、100%的乙醇溶液进行梯度洗脱,收集70%部位的乙醇洗脱液,60℃下真空减压浓缩,得到桑叶多酚提取物;
(4)决明子蒽醌提取物的制备:决明子经60℃烘干,粉碎,过200目筛;然后按料液比1:20 g/mL加入质量浓度90%的乙醇溶液,超声提取45 min,过滤,滤渣重复提取2次,合并提取液,得到决明子提取液,经减压浓缩至无醇味后,上AB-8大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后依次以质量浓度20%、40%、60%、80%、100%的乙醇溶液进行梯度洗脱,收集80%部位的乙醇洗脱液,60℃下真空减压浓缩,得到决明子蒽醌提取物;
(5)复配:按石榴皮多酚提取物0.52%、丁香黄酮提取物0.68%、桑叶多酚提取物0.62%、决明子蒽醌提取物0.38%、蔗糖0.08%、阿斯巴甜0.05%、安赛蜜0.05%、维生素C 0.20%、柠檬酸0.03%、纯化水97.39%的比例混合搅拌均匀;
(6)灌装灭菌:将配制好的饮料灌装于易拉罐瓶中,于120℃下灭菌15min,然后冷却至室温,得到含有四种植物提取物成分的所述植物减肥功能饮料。
以上所述仅为本发明的较佳实施例,凡依本发明申请专利范围所做的均等变化与修饰,皆应属本发明的涵盖范围。
Claims (2)
1. 一种植物减肥功能饮料,其特征在于:按重量百分数之和为100%计,该植物减肥功能饮料中所含各成分的重量百分数为:石榴皮多酚提取物0.11~0.52%、丁香黄酮提取物0.30~0.68%、桑叶多酚提取物0.18~0.62%、决明子蒽醌提取物0.15~0.38%、蔗糖0.02~0.08%、阿斯巴甜0.01~0.05%、安赛蜜0.01~0.05%、维生素C 0.05~0.20%、柠檬酸0.015~0.03%,其余为纯化水。
2.一种如权利要求1所述植物减肥功能饮料的制备方法,其特征在于:包括如下步骤:
(1)石榴皮多酚提取物的制备:收集石榴果皮,切碎,在40~60℃下低温烘干,粉碎,过200目筛;然后按料液比1:15~1:25 g/mL加入质量浓度60%的乙醇溶液,超声提取5~10min,过滤,滤渣重复提取1~2次,合并提取液,得到石榴果皮提取液,减压浓缩至无醇味,上D101大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后以质量浓度20%-100%的乙醇溶液进行梯度洗脱,收集60%~80%部位的乙醇洗脱液,60℃下真空减压浓缩,得到石榴皮多酚提取物;
(2)丁香黄酮提取物的制备:收集丁香花蕾,40~60℃条件下烘干,粉碎,过120目筛;然后按料液比1:15~1:25 g/mL加入质量浓度75%的乙醇溶液,超声提取10~15 min,过滤,滤渣重复提取1~2次,合并提取液,得到丁香提取液,减压浓缩至无醇味,上D101大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后以质量浓度10%-100%的乙醇溶液进行梯度洗脱,收集30%~50%部位的乙醇洗脱液,60℃下真空减压浓缩,得到丁香黄酮提取物;
(3)桑叶多酚提取物的制备:收集桑叶,40~60℃条件下烘干,粉碎,过200目筛;然后按料液比1:20~1:30 g/mL加入质量浓度70%的乙醇溶液,超声提取30~45 min,过滤,滤渣重复提取1~2次,合并提取液,得到桑叶提取液,减压浓缩至无醇味后,上AB-8大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后以质量浓度10%-100%的乙醇溶液进行梯度洗脱,收集50%~70%部位的乙醇洗脱液,60℃下真空减压浓缩,得到桑叶多酚提取物;
(4)决明子蒽醌提取物的制备:决明子经40~60℃烘干,粉碎,过200目筛;然后按料液比1:10~1:20 g/mL加入质量浓度90%的乙醇溶液,超声提取30~45 min,过滤,滤渣重复提取1~2次,合并提取液,得到决明子提取液,减压浓缩至无醇味后,上AB-8大孔吸附树脂层析柱,以水洗脱除去大分子水溶性蛋白质和多糖,然后以质量浓度20%-100%的乙醇溶液进行梯度洗脱,收集80%部位的乙醇洗脱液,60℃下真空减压浓缩,得到决明子蒽醌提取物;
(5)复配:将所得石榴皮多酚提取物、丁香黄酮提取物、桑叶多酚提取物、决明子蒽醌提取物与蔗糖、阿斯巴甜、安赛蜜、维生素C、柠檬酸、纯化水按比例混合搅拌均匀;
(6)灌装灭菌:将配制好的饮料灌装于易拉罐瓶中,于115-120℃下灭菌15-20 min,然后冷却至室温,得到所述植物减肥功能饮料。
